The low water solubility and stability of hesperidin (HP) as a bioactive substance greatly limit its application. In this paper, the anti-solvent precipitation and pH-shifting methods were used to prepare soybean isolate protein (SPI) and HP complexes. The complex prepared by the pH-shifting method (PSPI-HP) showed a higher loading efficiency of HP than the complex obtained by the anti-solvent precipitation method (ASPI-HP) (90.88 ± 1.27% versus 71.22 ± 0.68%). The stability of PSPI-HP was better than that in ASPI-HP when exposed to environmental factors. Multi-spectral evaluations and molecular docking results indicated that HP formed stable complexes with SPI through hydrogen bonding and hydrophobic interactions. Circular dichroism results demonstrated a decrease in α-helix and an increase in β- sheet and β-turn content upon SPI binding to HP. Compared with SPI and ASPI-HP, PSPI-HP exhibited better antioxidant activity and emulsifying activity. The results offer novel perspectives on the interaction between SPI and HP and broaden the potential applications of HP in the food and pharmaceutical industries. Graphical abstract

The incidence rate and mortality rate of breast cancer remain high, and there is an urgent need for safe and effective drugs. The excellent biological activity of hesperidin (HE) is a potential drug for the treatment of breast cancer. In this study, silk fibroin peptides (SFP) were used as delivery carriers and HE loaded SFP nanofibers (SFP/HE NFs) was prepared. The in vitro results showed that SFP/HE NFs significantly inhibited the proliferation and migration of breast cancer cell MDA-MB-231 compared with free HE. The mechanism results demonstrated that SFP/HE NFs induced apoptosis and DNA double stranded damage (DSBs) and further activated the cyclic monophosphate guanosine adenosine monophosphate synthase- stimulator of interferon gene (cGAS-STING) pathway. The in vivo studies showed that SFP/HE NFs treatment significantly inhibited the growth of breast cancer, with an inhibition rate of 65.9 % (100 mg/kg). In vivo mechanism studies also demonstrated that the anti-tumor activity of SFP/HE NFs was related to the activation of the cGAS-STING pathway. Interestingly, we found that the combination of SFP/HE NFs and cisplatin not only enhanced the anti-tumor activity of cisplatin, but also alleviated cisplatin induced nephrotoxicity. In conclusion, our results demonstrate the benefits of activating the cGAS-STING pathway in the treatment of breast cancer, which is expected to provide potential candidates for combined treatment of breast cancer.

Hesperidin hydrolysates (HHS) was produced by the hydrolysis of hesperidin (HDN) in previous studies. The potential components in HHS were identified by LC-MS, and minor components (MCS) in HHS were isolated. Antioxidant activities by radical-scavenging capacities, reducing capacity and β -carotene-linoleate assay, anti-inflammatory effects by inhibiting NO production of RAW 264.7 cells, and α-glucosidase inhibitory effects of HDN, HHS, MCS and henperetin (HTN) were investigated in present study. HHS showed higher radical scavenging activities, higher reducing capacity, and higher inhibitory activity in the β -carotene-linoleate assay than HDN. HHS inhibited the production of NO and pro-inflammatory cytokines of RAW 264.7 cells more strongly than HDN. HHS also intensively inhibited α-glucosidase activity whereas HDN showed little activity. In addition, the effects of MCS on above activities showed it play a synergistic part with HTN. This work suggested that hydrolyzation of HDN enhance the activities, and provided valuable information on effective utilization of HDN.

This research aimed to investigate the changes in the structure and functional properties of glutenin (Glu) induced by hesperetin (Ht) or hesperidin (Hd). Both Ht and Hd quenched the fluorescence of Glu through a static process and formed a Ht/Hd–Glu complex in one binding site. The binding constants between Glu and Ht and Hd were 4.20 × 10 4 L mol −1 and 3.59 × 10 4 L mol −1 at 25 °C, respectively. The computer simulations showed their bindings were dominated by hydrogen bonds and hydrophobic forces, and also provided the changes in particle motion and macroscopic properties of Glu induced by Ht/Hd. The presence of Ht/Hd promoted the exchange of sulfhydryl and disulfide bonds, resulting in the structural changes of Glu and the free sulfhydryl groups more exposed. Meanwhile, the increase in β-sheet content of Glu might cause the contraction of the carbon chain, thereby increasing the surface hydrophobicity. Due to the increase in rigid clusters and the decrease in flexibility, thermal stability of Glu was enhanced. Moreover, the emulsifying activity of Glu was significantly improved. This research lays the foundation for the application of Ht/Hd and Glu in healthy food or the optimization of Glu-based products.

Simple SummaryWhey protein is a widely utilized food ingredient known for its favorable nutritional profile and functional properties. Nevertheless, its inherent antioxidant capacity is relatively limited, which restricts its application as a natural antioxidant in the food industry. This study aims to investigate an environmentally friendly method to enhance the antioxidant activity of whey protein. In this process, a natural flavonoid derivative, methyl hesperidin, was introduced into a whey protein solution. Through physical mixing, it was observed that the two components could spontaneously bind to form a non-covalent complex. Furthermore, the microenvironment of amino acid residues in whey protein was altered with changed conformation, leading to enhancement in its antioxidant capacity. Additionally, molecular docking studies revealed that methyl hesperidin inserted itself into the hydrophobic cavity of the whey protein. These findings not only propose a novel approach for developing healthier antioxidants but also contribute to a deeper understanding of the interaction mechanisms between proteins and flavonoids. In this work, the possibility of enhancing the antioxidant capacity of whey protein (WP) through non-covalent interaction with methyl hesperidin (MH, a hesperidin derivative) was assessed. The underlying mechanism was analyzed in terms of multi-spectroscopy methods, thermodynamic analysis, and molecular docking simulation. The data indicated that MH could spontaneously bind to WP and form a non-fluorescent complex when physically mixed together. The presence of MH statically quenched the intrinsic fluorescence of WP, changed the microenvironment of amino acid residue, and altered the secondary and tertiary structure of WP, which in turn enhanced the antioxidant capacity of WP. The underlying mechanism may be assigned to hydrophobic interactions, which promoted MH inserting itself into the hydrophobic cavity in WP. The methoxy group on the B ring of MH may form hydrogen bonds with amino acids, which enhances the freedom of the phenyl hydroxyl group, resulting in higher antioxidant capacity than other hesperidin structural analogs. This research would enrich the theoretical basis about the interaction between protein and hesperidin-based derivatives, and it may supply valuable information for its application in the food and medicine fields.

Hesperidin (HD) is predominantly found in citrus fruits, and has been shown to possess various biological properties, such as anti-inflammatory, antioxidant, and anti-carcinogenic. However, its application is limited by poor solubility. In this study, a new solid dispersion (SD) of hesperidin was prepared by ball milling using PVPK30 as the carrier, and the in vitro and in vivo studies and the therapeutic effects about colitis in rats were evaluated. In vitro analysis revealed that the solid dispersion showed a better release effect. The cumulative release of HD-SD reached 48.24% at 120 min, which was 5.9 times that of pure HD. In vivo studies demonstrated that Cmaxand AUC0–24were significantly higher in HD-SD compared with pure HD (p< 0.01), which were 2.67 and 1.50 times that of HD, respectively. Furthermore, treatment with HD-SD significantly alleviates symptoms and histological features in acetic acid (AA)-induced colitis in rats. Furthermore, HD-SD treatment significantly ameliorated the disease severity of acetic acid (AA)-induced colitis in rats, as evidenced by improved clinical signs, attenuated histological damage, and decreased levels of inflammatory factors (TNF-α, IL-6, and IL-1β). Moreover, the structure and relative abundance of the gut microbiota were modulated. Specifically, the relative abundance ofErysipelotrichaceaewas decreased and the relative abundance ofBacteroidota,Lachnospiraceae, and[Eubacterium]_coprostanoligenes_groupwere increased. These results suggest that HD-SD could serve as a gut-health-promoting functional ingredient, potentially contributing to the dietary management of colitis through microbiota modulation.

Understanding drug cocrystal formation and its competitive mechanism is important for improving the physicochemical properties of drugs. In this article, the cocrystal formation of polyhydroxy natural products and their competitive mechanisms are discussed through experimental screening and theoretical calculations. The formation conditions and types of cocrystals were determined based on efficient screening experiments of polyhydroxy natural product cocrystals. To investigate the difficulty and stability of cocrystal formation, competitive cocrystal experiments of polyhydroxy natural products were carried out simultaneously. By simulation of the electrostatic potential surface of molecules and the Hirshfeld surface of crystals, the mode of hydrogen bonding within the crystals was determined. The strength of hydrogen bonding was assessed based on the intermolecular interactions and energy framework within the crystal structure. A “substitution model” was constructed based on experimental results and theoretical calculations, which is of great significance for the prediction and design of natural product cocrystals and provides a feasible solution for improving the physicochemical properties of drugs.

Bilayer film incorporating hesperidin was made for antioxidant food preservation. • Corona treatment improved the structural stability of the bilayer films. • Hesperidin enhanced the film's antioxidant activity, particularly at 3 wt%. • P-PCH3 film excelled in mechanical, barrier, and antioxidant properties. • P-PCH3 film well-preserved the arils, cutting oxidative stress and quality loss. As concerns about environmental sustainability and food safety rise, the demand for biodegradable antioxidant packaging solutions has increased. This study develops a biodegradable bilayer composite film for antioxidant food packaging, consisting of a commercial polylactic acid (PLA) substrate coated with a polyvinyl alcohol/chitosan (PVA/CS) layer enriched with hesperidin. Bilayer films were prepared using solution casting and corona treatment, improving adhesion and structural integrity. PLA layer provides water vapor barrier and UV resistance, while the PVA/CS coating loaded with hesperidin offers mechanical strength, oxygen barrier property, and antioxidant properties. Hesperidin formed an intercalation structure in the matrix, promoting the development of hydrogen, ester, and amide bonds and enhancing corresponding film crystallinity and thermostability. Bilayer films with hesperidin exhibited improved mechanical properties, hydrophobicity, thermostability, visible light transmittance, and barrier properties (moisture, oxygen, UV), particularly the film containing 3 wt% hesperidin (P-PCH3). P-PCH3 film demonstrated significant antioxidant activity, evidenced by 89.3% DPPH scavenging and 90.2% ABTS +· scavenging. In a storage experiment, the P-PCH3 film effectively preserved the quality of pomegranate arils by mitigating oxidative stress, enzymatic browning, and weight loss. This biodegradable antioxidant packaging offers a scalable, eco-friendly solution for food preservation, addressing both food safety and environmental challenges in the packaging industry.

This study synthesized nitrogen-doped mesoporous carbon (NMC) using Zn-MOF-8 as a template, followed by the preparation of ZIF-08/NMC composites via π-π adsorption interactions between ZIF-08 and NMC. The composite was characterized using scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDS), X-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectroscopy (FT-IR), and electrochemical methods. The resulting ZIF-08/NMC/GCE electrode demonstrated stable and sensitive detection of hesperidin. The modified electrode exhibited a broad linear range from 0.8 to 120 μM with a detection limit of 0.16 μM (signal-to-noise ratio ≥3). Furthermore, the sensor demonstrated excellent stability, repeatability, and interference resistance, and was successfully applied for hesperidin detection in real samples.

Kawasaki Disease (KD) is an acute and self-limiting vasculitis of unknown etiology that mainly occurs in infancy and can lead to vascular endothelial injury. Hesperidin (HES) is an economical dietary biological flavonoid with anti-oxidant, anti-inflammatory, and anti-apoptotic pharmacological effects. The main objective of this study was to investigate the protective effects of HES on KD, and try to elucidate the underlying mechanism. The Candida albicans water-soluble fraction (CAWS) was used to induce coronary arteritis of KD mouse model in vivo , and tumor necrosis factor α (TNF-α) was employed to induce human umbilical vein endothelial cell (HUVEC) injury of KD cell model in vitro to investigate the anti-inflammatory and anti-apoptotic effects of HES on KD. Our in vivo results showed that HES significantly reduced coronary artery injury in KD mice by alleviating pericoronary inflammatory infiltration and tissue fibrosis, inhibiting inflammatory cytokines and chemokine expressions, and decreasing vascular endothelial cell apoptosis. Our in vitro study confirmed that HES had the opposite ability of the NF-κB agonist NF-ĸB activator 1 (ACT1) to significantly alleviate the inflammatory response, CellROX level, and apoptosis by decreasing BAX/BCL-2 and Cleaved Caspase-3 levels as well as reducing TUNEL positive cells and the ratio of flow cytometric apoptotic cells in TNF-α induced HUVECs. The further mechanism study based on bioinformatics analysis and western blotting demonstrated that HES could protect against vascular inflammation and cell apoptosis of KD through inhibiting the TLR4/IĸBα/NF-ĸB pathway, suggesting that HES may be a promising therapeutic candidate for KD.

Endometritis is a prevalent condition of the reproductive system that leads to significant tissue damage in the uterus, thereby seriously impacting the health and quality of life of these patients. Hesperidin (HES), a natural flavonoid glycoside, displays various biological activities. Nonetheless, the protective effect of HES treatment against endometritis remains uncertain. In this study, we examined the protective effects of HES on endometritis induced by lipopolysaccharide (LPS) both in vivo and in vitro . In vitro experimental results showed that HES reduced the levels of inflammatory factors, improved mitochondrial morphology, increased mitochondrial activity and inhibited ROS levels; moreover, it activated the AMPK/PGC-1 α signalling pathway in LPS-induced human endometrial endothelial cells (HEECs). AMPK served as a potential target of HES, which was further validated using molecular docking, molecular dynamic simulation, CETSA and SPR. Moreover, in vivo experimental results indicated that HES significantly mitigated the histopathological alterations in the uterus caused by LPS, as well as reduced the levels of inflammatory factors, MPO activity, the expression levels of CD38, and CD138 and activated the AMPK/PGC-1α signalling pathway. Overall, HES mitigated mitochondrial oxidative stress and inflammatory response primarily through activation of the AMPK/PGC-1α signalling pathway, thereby exerting protective effects against endometritis.

In this study, we investigated how different proportions blends of Rhamnogalacturonan-I pectic polysaccharides and hesperidin impact the gut microbiota and metabolites using an in vitro simulated digestion and fermentation model. The results indicated that both of them could modulate the gut microbiota and produce beneficial metabolites. However, their blends in particular proportions (such as 1:1) exhibited remarkable synergistic effects on modulating the intestinal microenvironment, surpassing the effects observed with individual components. Specifically, these blends could benefit the host by increasing short-chain fatty acids production (such as acetate), improving hesperidin bioavailability, producing more metabolites (such as hesperetin, phenolic acids), and promoting the growth of beneficial bacteria. This synergistic and additive effect was inseparable from the role of gut microbiota. Certain beneficial bacteria, such as Blautia , Faecalibacterium , and Prevotella , exhibited strong preferences for those blends, thereby contributing to host health through participating in carbohydrate and flavonoid metabolism.
