Inulin as a natural polysaccharide regulates intestinal microorganisms, and improves the immune and gastrointestinal function. In order to explore the effect of inulin on pulmonary metastasis of colon cancer, we set up a CT26 injected pulmonary metastatic model. The results showed that inulin used alone did not improve pulmonary metastasis of colon cancer, while inulin combined with rifaximin significantly prolonged the survival time of mice, and inhibited pulmonary metastasis compared with model and inulin groups. Inulin treatment increased the abundance of harmful bacteria such as Proteobacteria and Actinobacteria , while combined treatment decreased their abundance and increased the abundance of beneficial bacteria containing Firmicutes and Eubacterium which belonged to the bile acid-related bacteria. The combination treatment decreased the content of primary bile acids and secondary bile acids in the feces of mice, especial for DCA and LCA which were the agonists of TGR5. Furthermore, the combination treatment reduced the mRNA expression of the TGR5, cyclin dependent kinase 4, cyclin 1 and CDK2, increased the mRNA expression of p21 in the lung, down-regulated the level of NF-κB p65, and up-regulated the level of TNF-α compared with the model group. The above may be the reason for the better use of the combination treatment.

Inulin could be converted to bio-based chemicals by an inulinase producer without external inulinase, and the production of 2,3-butanediol was less than 50 g/L. In this work, a novel inulinase producer of Klebsiella pneumoniae H3 was isolated, and inulinase catalytic properties as well as 2,3-butanediol fermentation were investigated. The enzyme was an intracellular inulinase with an optimal pH of 6 ∼ 7 and a temperature of 30 °C. The use of inulin by H3 was dependent on the degree of polymerization (DP), and the average DP of inulin in fermentation broth increased from 2.82 to 8.08 in 24-h culture of batch fermentation. Acidic pretreatment was developed to increase inulin utilization by adjusting medium pH to 3.0 prior to sterilization. In batch fermentation with optimized medium and fermentation conditions, the concentration of target product (2,3-butanediol and acetoin) was 80.4 g/L with a productivity of 2.23 g/(L⋅h), and a yield of 0.426 g/g inulin.

Pomegranate seed oil is rich of conjugated fatty acids which are highly appealing for a variety of applications in food industry. In this research, ovalbumin (OVA) and ovalbumin-inulin glycoconjugates with different Maillard reaction times were used to stabilize pomegranate seed oil emulsions and their impact on physicochemical stability and oxidative stability of the products was investigated. The OVA-inulin glycoconjugate produced on 10th day of Maillard reaction has exhibited significantly higher conjugation efficiency, lower surface hydrophobicity and lower surface tension than other glycoconjugates . The secondary conformation of OVA and conjugates determined by far-UV circular dichroism spectroscopy has remarkably changed. The reduction in intensity of Trp-fluorescence observed in glycated proteins with inulin indicated that the glycation affected partially the side chains of protein in tertiary structure through the Maillard reaction without great disruption of native structure. The emulsion stabilized by OVA-inulin glycoconjugate obtained by 10 days Maillard reaction has shown the best physicochemical stability. Compared with the OVA emulsion, the oxidative stability of the glycated OVA emulsion system was significantly improved (p < 0.05). Fatty acid profile results also confirmed that OVA-inulin glycoconjugates were able to prevent the pomegranate seed oil from oxidation . It is suggested that the inulin attached to OVA by glycation played a vital role in physicochemical stability and oxidative stability of pomegranate seed oil emulsions.

Nanovesicles are attractive for cosmetics application owing to their enhanced stability and transdermal delivery of active ingredients. Retinyl propionate (RP) is a promising anti-aging ingredient but it is usually unstable in formulations. In this work, we developed RP-loaded nanoemulsions using polymeric hydrophobically modified inulin (HMI), small-molecular polysorbate 20 (PS-20) as the main emusifiers and positively-charged behentrimonium chloride (BC) as a zeta-potential adjuster. Systematically optimizing the composition of the three emulsifiers led to small droplet size (<100 nm) and high physical stability of the nanoemulsions. Importantly, RP loaded in the nanoemulsions exhibited a high retention rate exceeding 80% after storage at 50 ℃ for 30 days. The superior RP stabilization and anti-coalescence property of the nanoemulsion could be attributed to the densely-packed, sterically-hindered and highly-charged interfacical layers composed of multiple emulsifiers at the oil-water interface. In addition, in vitro skin permeation experiments in a Franz diffusion cell suggested that the as-prepared nanoemulsions exhibited highly enhanced transdermal delivery of RP into the epidermis and dermis compared to the conventional emulsions. Therefore, the present work provides a feasible approach for more effective application of retinol esters in cosmetic formulations.

A new class of inulin derivatives possessing 1,2,3-triazolium charged units by associating “click reaction” with efficient 1,2,3-triazole quaternization were designed and synthesized. After the chemical modification, all the inulin derivatives showed good solubility in water, and exhibited excellent bioactivity compared to inulin. In the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging assay, the scavenging activity of 1,2,3-triazolium inulin derivatives (5a–5d) (IC50 0.21–0.29 mg mL−1) was found to be higher than that of inulin derivatives (4a–4d) (IC50 0.58–0.95 mg mL−1) and inulin. 5a–5d (IC50 < 0.01 mg mL−1) also exhibited stronger inhibitory activity against superoxide anion formation than 4a–4d (IC50 0.02–0.09 m g mL−1) and Vitamin C (IC50 0.02 mg mL−1). In the antifungal assays, 5a–5d also inhibited the growth of tested phytopathogens more effectively with inhibitory indices of 43.10–82.56% at 1.0 mg mL−1, compared with 4a–4d and inulin. Based on these data, it is reasonable to propose that the introduction of 1,2,3-triazolium group through chemical modification is significant to enhance the free radical-scavenging activity and antifungal activity of inulin.

Iron deficiency is the utmost critical problem of nutrition worldwide. Polysaccharide-iron chelate is a promising iron supplementation that exhibits good stability, high absorption rate, and fewer adverse reactions, but it still owns drawbacks of extra energy and lower iron content with respect to inorganic ones. Carboxylated polysaccharide-iron has been proven with enhanced iron content, however with poor aqueous solubility. Here, Fe 3+ concentrations controlled highly soluble carboxylated inulin-iron chelates were synthesized to promote the sustained release and efficient supplement of iron with no extra energy. Inulin, soluble dietary fiber, was initially reacted with succinic anhydride (SA) to get carboxylated inulin (IN-SA) consequently enhancing the complexing ability of Fe 3+ and its bioavailability. Interestingly, the aqueous solubility of the IN-SA-iron chelate (IN-SA-Fe) directly correlated with the Fe 3+ concentrations. Relatively higher or lower Fe 3+ concentrations resulted in highly soluble IN-SA-Fe, otherwise, leading to insolubility precipitates. The possible mechanism was proposed as the polymerization reaction of IN-SA in different degrees with Fe 3+ acting as the cross-linker. Moreover, IN-SA-Fe was demonstrated with properties of sustained slow-release of iron in artificial gastric juice and good in vivo biosafety. This work provides a simple, efficient, no extra energy soluble iron supplement for the treatment of iron deficiency.

Purpose Gadolinium-based contrast agents (GBCAs), which are indispensable for magnetic resonance imaging (MRI), carry a severe risk of nephrogenic systemic fibrosis (NSF) in patients with chronic kidney disease (CKD), particularly in those with advanced renal impairment. This critical safety concern necessitates the development of biocompatible alternatives that do not compromise diagnostic efficacy.Materials and Methods Inulin-based nanoparticles (Inulin@Gd/Zn NPs) were synthesized by chelating inulin with gadolinium and zinc ions, aimed at being used as contrast agents that can replace conventional GBCAs. The morphology, size, and zeta potential of the nanoparticles were determined. The anti-fibrotic ability of Inulin@Gadolinium/zinc nanoparticles (Inulin@Gd/Zn NPs) was evaluated in vitro and in vivo, as well as their MRI imaging ability and metabolic performance in normal and fibrotic kidneys. A unilateral ureteral obstruction (UUO) model was created in mice and was used to evaluate the in vivo anti-fibrotic efficacy. Results The ultra-small Inulin@Gd/Zn NPs (<10 nm) exhibited high stability and T1 relaxivity. Compared to Gadolinium diethylenetriaminepentaacetic acid (Gd-DTPA), Inulin@Gd/Zn NPs demonstrated precise kidney targeting, excellent T1-weighted MRI capabilities, and rapid renal clearance. In vitro studies showed that Inulin@Gd/Zn NPs had very low cytotoxicity for HK-2 cells up to 100 µM. Further, the NPs were able to reduce reactive oxygen species generation in hydrogen peroxide–stimulated HK-2 cells. Negligible levels of hemolysis and organ toxicity were confirmed in vivo. Further, in the UUO model, administration of Inulin@Gd/Zn NPs was found to significantly inhibit fibrosis. Both in vitro and in vivo experiments demonstrated that Inulin@Gd/Zn alleviated GBCA-induced early tubulointerstitial fibrosis by inhibiting the transforming growth factor-β1 (TGF-β1) signaling pathway. Conclusion Inulin@Gd/Zn NPs represent a promising strategy for clinical application of GBCAs. It addresses the diagnostic and therapeutic challenges in patients with CKD, offering a paradigm shift in the safe application of GBCAs in high-risk populations, indicating the need for further clinical trials to enable clinical applications.

Effective oral delivery of natural products is essential for sustained therapeutic effects for type 2 diabetes mellitus (T2DM) therapy. However, compromised bioavailability leads to undesirable effect and hypoglycemia. Herein, biomimetic nanoplatform based on mesoporous polydopamine (MPDA) and inulin gel were designed to enhance the oral delivery of 1-Deoxynojirimycin (DNJ). DNJ was adsorbed via mesopores and hydrogen bonding by MPDA and encapsulated within the inulin hydrogel. The MPDA@Inulin gel exhibited sustained-release properties that allowed DNJ to be released persistently in the small intestine, thereby enhancing its oral bioavailability. Moreover, the capacity of DNJ@MPDA@Inulin gel in controlling the blood glucose levels was evaluated in a high-fat diet induced T2DM model. Notably, after 4 weeks of daily oral administration, DNJ@MPDA@Inulin demonstrated enhanced efficacy in controlling blood glucose levels in T2DM model mice, improving insulin sensitivity. More important, this hydrogel nano-sustained release platform based DNJ delivery system could ameliorate glucolipid metabolism, enhance intestinal mucosal barrier and reshape gut microbial composition. Thus, it is anticipated that inulin-MPDA based oral delivery system have a potential for natural products delivery to improve bioavailability and alleviate T2DM.

Gellan gum (GG) is often added to gelatin (GL) to improve the gel property. GG-based or inulin (IL)-based hydrogels were developed. Rigid and brittle gels or smooth and delicate gels were prepared with GG and IL, respectively. This study aimed to explore the properties and interaction mechanisms of the mixed-gel system containing GL, GG, and IL, in which different ratios of GG-IL (0.4%) (10:0, 8:2, 6:4, 5:5, 4:6, 2:8, and 0:10) were added to GL (6%). Texture profiles, rheological properties, water mobility, intermolecular forces, circular dichroism (CD) spectra, and microstructures were analyzed. The results showed that addition of GG-IL could improve the hardness, chewiness, and cohesiveness of mixed-gel, besides maintaining appropriate springiness. Water mobility of the mixed-gel decreased while viscoelasticity increased upon the addition GG. At GG:IL = 2:8, the melting temperature of mixed-gel was far higher than that of GL gel itself. The GL-GG-IL gel showed decrease in nonspecific bonding and increase in hydrogen bonding compared with the GL gel. CD spectra indicated the promotion of GL unfolding by GG, hence suggesting the binding of GG to GL; binding ability was better at GG:IL >5:5. Cryo- SEM provided evidence for the formation of cross-linked network within GL-GG-IL. Overall, we concluded that addition of GG-IL to GL system would be most suitable for improving the properties of mixed-gel. This finding may be potentially applicable in the further development of gel food products, such as meat jellies and gummy jellies.

The therapeutic efficacy of dietary fibers is closely related to structural heterogeneity, yet the impact of chain length on physicochemical behavior and gut microbiota modulation remains unclear. Herein, three homologous linear β-(2 → 1)-linked inulin-type fructans with distinct molecular weights, hereafter referred to as CBP1–3, were isolated from Codonopsis bulleynana . All fractions spontaneously assembled into stable supramolecular nanoparticles/disks in water, a process thermodynamically driven by intermolecular hydrogen bonding according to molecular dynamics simulations. Biological evaluation revealed a striking divergence: while low-molecular-weight fractions showed superior efficacy in in vitro fermentation, the relatively high-molecular-weight CBP1 exhibited superior in vivo potency. Multi-omics analysis supports the hypothesis that this supramolecular structure, in synergy with extended chain length, may function as a structural barrier and confer sustained-release characteristics. This likely limits mucin-degrading Akkermansia proliferation via a nutrient-sparing effect, thereby maintaining metabolic homeostasis. These findings demonstrate that relatively high-molecular-weight polysaccharides possess unique ecological advantages in physiological contexts, challenging traditional in vitro screening paradigms and positioning CBP1 as a promising candidate for precision microbiome restoration.

Shale hydration dispersion and swelling are primary causes of wellbore instability in oil and gas reservoir exploration. In this study, inulin, a fructo-oligosaccharide extracted from Jerusalem artichoke roots, was modified by acylation with three acyl chlorides, and the products (C10-, C12-, and C14-inulin) were investigated for their use as novel shale hydration inhibitors. The inhibition properties were evaluated through the shale cuttings hot-rolling dispersion test, the sodium-based bentonite hydration test, and capillary suction. The three acylated inulins exhibited superb hydration-inhibiting performance at low concentrations, compared to the commonly used inhibitors of KCl and poly (ester amine). An inhibition mechanism was proposed based on surface tension measurements, contact angle measurements, Fourier-transform infrared analysis, and scanning electron microscopy. The acylated inulin reduced the water surface tension significantly, thus, retarding the invasion of water into the shale formation. Then, the acylated inulin was adsorbed onto the shale surface by hydrogen bonding to form a compact, sealed, hydrophobic membrane. Furthermore, the acylated inulins are non-toxic and biodegradable, which meet the increasingly stringent environmental regulations in this field. This method might provide a new avenue for developing high-performance and ecofriendly shale hydration inhibitors.

Berberine hydrochloride (BH), a classic drug for treating acute enteritis, has been found to possess multiple immunoregulatory functions. However, its impact on the chemotherapy of colorectal cancer (CRC) has not been revealed. In addition, a suitable oral delivery system for BH is required to reduce and slow its clearance in the intestines and to improve its bioavailability. In the present work, a BH-loaded hybrid liposome containing a prebiotic inulin derivative, named BLPN, was designed and constructed. BLPN was stable in media simulating the gastrointestinal environment. It showed a bidirectional modulatory effect on immune cells, depending on the BH concentration. After oral administration, BLPN prolonged the retention time of BH in the intestines and blood circulation. In an MC38 orthotopic CRC mouse model, BLPN substantially increased the BH accumulation inside tumors. A chemotherapeutic agent, irinotecan hydrochloride-loaded liposome (ILPS), was also prepared. The combination of BLPN with ILPS activated both innate and adaptive anti-tumor immune cells and promoted the secretion of proinflammatory cytokines, which enhanced the extravasation of ILPS from the blood vessels within the tumor. The combination with BLPN significantly improved the CRC tumor-suppressive effect of ILPS and extended the survival time of tumor-bearing mice. This study provides a potential strategy for improving CRC chemotherapy by combining it with a prebiotic oral delivery system for BH.
