Electrochemiluminescence (ECL) is highly recommended in chiral recognition. ECL-based chiral sensors highly desire a sensitive sensing interface for signal conversion and absolute chiral discrimination. The ECL emission based on a luminol-dissolved O 2 system received much attention due to its nontoxicity and stability. However, the drawback of weak ECL emission hinders the fast signal conversion from chiral discrimination to ECL response. Herein, the amplification strategy of ECL emission is proposed based on the electrochemical reduction enhanced O 2 reduction reaction (ORR). Cadmium sulfide decorated on carbon-nanotubes (CdS/CNTs) with easy synthesis, wide-pH operation, and suitable valence-conduction band position is employed. Upon cathodic scan, the electrons transfer from electrochemically reduced-CdS/CNTs to O 2 and H 2 O 2 , thus accelerating the generation of reactive oxide species (ROS) and furthering ECL emission. Furthermore, the chiral ECL sensing interface is well-designed by combining the chiral recognition of D-amino acid oxidase (DAAO) with the signal transduction and amplification of CdS/CNTs-enhanced ECL emission. During DAAO-catalyzed enantioselective-oxidations of alanine, the O 2 is converted to H 2 O 2 , which tunes the ROS generation. With synergetic regulations of ROS generation by nano-derived CdS/CNTs and bio-derived DAAO, alanine enantiomers are highly discriminated and the L-alanine is quantitatively detected with the most competitive detection limit so far (0.014 fM).

In this work, a series of 2,2,6,6-tetramethylpiperidine-N-oxyl (TEMPO) radicals bearing different functional groups were exploited as a simple catalyst to promote electrochemiluminescence (ECL) generation in luminol/H 2 O 2 system. These TEMPO radicals were found to facilitate the electrochemical oxidation of H 2 O 2 and luminol through different catalytic mechanisms, as well as the subsequent ECL generation of luminol/H 2 O 2 system. The electrochemical oxidation and luminol ECL generation could be tuned by the functional group on the para -position of TEMPO, for which the structure/activity relationship was revealed. Finally, with the combination of enzymatic system, luminol ECL enhancement up to 9.6-fold was obtained through the catalysis of 4-hydroxyl-TEMPO. The enhanced luminol ECL allows acquiring brighter ECL images in a single-electrochemical system (SEES) for multiplex detection of cholesterol, H 2 O 2 and glucose.

In this study, it was found that tannic acid could inhibit the chemiluminescence (CL) intensity of luminol–H2O2–cupric oxide nanoparticles significantly. Based on this inhibition effect, a novel and highly sensitive flow injection method with inhibited chemiluminescence was developed for the determination of tannic acid. Under optimum conditions, the net CL intensity was proportional to the concentration of tannic acid over the range of 10–100 nM and the detection limit was 2.6 nM. The relative standard deviation (RSD) for six repeated determinations of 80 nM tannic acid was 2.8%. Note that this method has been successfully used for the analysis of tannic acid in real Chinese gall samples.

In this study, core-shell Au@Pt nanoparticles (Au@Pt NPs) with peroxidase catalytic activity were synthesized by the seed-mediated method, and were used to catalyze the reaction of luminol–H2O2 to enhance the chemiluminescence (CL) intensity. It was found that thiocyanate (SCN−) can effectively inhibit the catalytic activity of Au@Pt NPs. Based on this phenomenon, a method to detect SCN− by using the Au@Pt NPs-catalytic luminol–H2O2 CL system was established, which has an ultra-low detection limit and an ultra-wide linear range, as well as the advantages of being simple and having low-cost and convenient operation. The research mechanism indicated that SCN− could be adsorbed on the surface of Au@Pt NPs and occupies the active sites of Pt nanostructures, which led to a decrease in the amount of Pt0 and a loss of the excellent catalytic activity of Au@Pt NPs. After optimizing the experimental conditions, this assay for detecting SCN− exhibited a good linear range from 5 to 180 nM, and the low detection limit was 2.9 nM. In addition, this approach has been successfully applied to the detection of SCN− in tap-water samples, which has practical application value and embodies good development prospects.

Luminol-reduced Au nanoparticles (AuNPs) were prepared by using a facile one-step method. Luminol-reduced AuNPs were utilized as a novel dual-readout probe to develop a qualitative/quantitative immunochromatographic test strip (ITS) for detecting pesticide residues , by adopting fenvalerate as a model analyte . Fenvalerate in sample solution could compete with its corresponding immobilized coating antigen in test line of ITS to bind the AuNPs-tagged fenvalerate monoclonal antibody. After the complete immunoreaction, the AuNPs-tagged antibody in test line was the dual-readout probe. Superior to signal readout-based ITSs, especially traditional Au nanoparticles-based ITSs, luminol-reduced AuNPs-based dual-readout ITSs could simultaneously meet the demand of qualitative analysis and quantitative detection. Colorimetric signal from luminol-reduced AuNPs could be adopted as a qualitative readout, which could increase analysis efficiency in field analysis and rapid testing. In the proposed analytical platform, fenvalerate could be semi-quantitated within 15 min. Furthermore, as for affirmed contaminated sample through colorimetric analysis , chemiluminescent (CL) signal originating from luminophor-modified AuNPs as a quantitative readout aided to improve analytical sensitivity after triggering luminol-H 2 O 2 CL reaction. Under the optimal conditions, the detection limit for fenvalerate were 0.067 ng/mL (S/N = 3). The reliability of dual-readout ITSs was demonstrated through the successful practical application on traditional Chinese medicines. Owing to high analysis efficiency, low cost, high facility and improved accuracy, the dual-readout ITSs by using only one probe showed great potential in field analysis and rapid testing for drug safety, environmental sanitation and clinical diagnosis.

A highly sensitive molecularly imprinted electrochemiluminescence (MIECL) sensor was developed for detecting rifampicin (RIF) based on luminol@Co-MOF. Co-MOF had a significant enhancement of ECL signaling in the luminol-O 2 system. Molecular imprinted polymers (MIPs) with the introduction of RIF provide new properties for the specific recognition of RIF. A noteworthy decrease in ECL intensity was observed with higher concentrations of RIF. Consequently, the ECL signal was controlled by RIF elution from and adsorption by the MIP, thus establishing a new method for RIF detection. Under optimal conditions, this sensor exhibited linear detection ranges of RIF between 1.0 × 10 −11 mol L −1 and 1.0 × 10 −6 mol L −1 , with a detection limit of 3.3 × 10 −12 mol L −1 ( S / N = 3). The recoveries ranged between 98.1 and 106.0% in fish samples. This method can be used as a sensitive and rapid method to detect RIF in real samples. Graphical

Developing simple and portable luminol
electrochemiluminescence
(ECL) systems without externally added co-reactants is important for
on-site analysis. Cobalt is a low-cost and redox-active transition
metal with favorable electrocatalytic properties toward water-oxidation-related
interfacial reactions in alkaline media, which may facilitate luminol
oxidation and the generation or activation of reactive oxygen intermediates
during ECL. In this work, for the first time, a bare cobalt electrode
was introduced into a co-reactant-free luminol ECL system and used
to construct a simple direct-current two-electrode platform, with
the ECL signal recorded by smartphone imaging. Compared to a carbon-fiber
electrode, the cobalt electrode generated a much stronger luminol
ECL signal, with an enhancement of more than 80-fold in the smartphone
imaging system, indicating its strong promotion of the electrode reactions
associated with luminol ECL. After optimization of the imaging parameters
and experimental conditions, a portable analytical method for sinapic
acid determination was established. Under the optimized conditions,
the proposed system showed a linear range of 1–100 μM
and a limit of detection of 0.26 μM. The method also showed
acceptable anti-interference capability under the tested conditions
and satisfactory recoveries in spiked white mustard seed samples.
These results demonstrate a simple, low-cost, and portable strategy
for co-reactant-free luminol ECL analysis and highlight the potential
of cobalt electrodes for proof-of-concept on-site sensing.

A simple and sensitive method was developed for the electrochemiluminescence (ECL) determination of DNA. • Au-luminol NPs addressed the issues of weak solubility and ECL signals of luminol in neutral conditions. • The target DNA induced the formation of heme and G-quadruplexes as biomimetic enzymes, thereby enhancing the electrochemical luminescence intensity of luminol. A novel signal amplification approach was developed for DNA detection, leveraging the synergistic effect of G-quadruplex/hemin DNAzyme and gold nanoparticles (Au NPs) to enhance the electrochemiluminescence (ECL) of luminol. The optimized system exhibited excellent linear response toward target DNA with a linear detection range from 10 fM-50 pM and achieved fM-level DNA detection. The spiked recoveries of serum samples ranged from 95.1% to 107.6%, demonstrating its reliable performance for practical sample analysis. Compared with recently reported analogous ECL sensors for DNA detection, this system featured a wider linear range, lower limit of detection and stronger anti-matrix interference capability, offering a new strategy for rapid and ultrasensitive detection of target analytes in complex matrices. In this study, a high sensitivity chemiluminescence enzyme immunoassay (CLEIA) based on novel enhancers was developed. Under optimal conditions, we developed an enhanced chemiluminescence reaction (ECR) catalyzed by horseradish peroxidase (HRP-C) in the presence of 3-(10'-phenothiazinyl) propane-1-sulfonate (SPTZ) and 4-morpholinopyridine (MORP) as enhancers. The limit of detection of the newly prepared chemiluminescent cocktail for HRP was 0.33 pg/well, which is lower than that of commercial Super Signal substrate. The results showed that this novel chemiluminescent cocktail can significantly increase the light output of HRP-catalyzed ECR, which can be translated into a corresponding improvement in sensitivity. Similar improvements were observed in CLEIA for the determination of chloramphenicol in milk. In addition, the ECR of N-azoles as secondary enhancer was also presented. In this work, an electrochemiluminescence (ECL) aptasensor based on silver nanoparticles (AgNPs) for detection of profenofos was presented. The aptasensor was based on the affinity of the aptamer (Apt) with the target profenofos. When the Apt contacted with the profenofos, the ECL detection signals were weakened, indicating the successful construction of the aptasensor. In order to improve the aptasensor performance, the platinum electrode (PE) was modified by AgNPs due to their good conductivity and catalytic property. Meanwhile luminol-hydrogen peroxide was selected as a luminescence system. Through the standard curve experiment and specificity experiment, it was proved that the sensor had the advantages of low detection limit and good specificity. Under the optimum conditions, the aptasensor had a good linear response to profenofos from 0.5 μg/mL to 100 μg/mL (1.34 μM to 267.6 μM ), with a low detection limit of 0.13 ng/mL and a correlation coefficient (R²) of 0.9903. The aptasensor was also successfully applied for detection of profenofos in real vegetables. Because the developed aptasensor had the following advantages of simple preparation, good stability, high selectivity and sensitivity, it would show great potential for other small molecule contamination detection.

Electrochemiluminescence (ECL) of luminol is a luminescence process that proceeds in the presence of reactive oxygen species (e.g. hydrogen peroxide (H 2 O 2 )) at a suitable electrode potential, the reaction mechanism of which is complicated and remains ambiguous. In this work, we report a visualization approach for measuring the thickness of the ECL layer (TEL) of the luminol/H 2 O 2 system to decipher the reaction process by combined use of the microtube electrode, ECL microscopy, and finite element simulations. With the increase of solution pH, the ECL image captured with the microtube electrode tends to vary from spot to ring, corresponding to the decrease of TEL from >9.1 μm to ca. 4.3 μm. We propose that different intermediates are involved in the course of ECL reaction. At a low pH (e.g. pH < 9), a relatively large TEL is most likely determined by the diffusion of oxidized and deprotonated luminol intermediate that is neutral and has a long lifetime. While at a high pH (e.g. pH in the range of 10 to 12), the ECL reaction is controlled by short-lived radical intermediates of both luminol and superoxide anion. The proposed mechanism is proved theoretically by finite element simulations and experimentally by the apparent effect of concentration ratio of luminol/H 2 O 2 .

Poly(aniline-luminol-hemin) nanocomposites are prepared on an electrode surface through electropolymerization, and a highly sensitive electrochemiluminescence (ECL) biosensor for choline is developed based on the poly(aniline-luminol-hemin) nanocomposites and an enzyme catalyzed reaction of choline oxidase (CHOD). The obtained nanocomposites are characterized by scanning electron microscopy (SEM), atomic absorption spectrometry (AAS) and ECL. The results indicate that hemin can be incorporated into the poly(aniline-luminol) nanocomposites using the facile electropolymerization method, and the poly(aniline-luminol-hemin) nanocomposites are rod shaped porous nanostructure. Moreover, the poly(aniline-luminol-hemin) nanocomposites exhibit higher ECL intensity than poly(aniline-luminol) nanocomposites in alkaline media due to the catalytic effect of hemin on the ECL of the polymerized luminol and the electron transfer ability of hemin in the nanocomposites. CHOD is immobilized on the surface of the poly(aniline-luminol-hemin) nanocomposites modified electrode with glutaraldehyde, and the ECL biosensor based on poly(aniline-luminol-hemin)/CHOD exhibits a wider linear range for the choline detection. The enhanced ECL signals are linear with the logarithm of concentration of choline over the range of 1.0×10 −11 ∼1.0×10 −7 mol L −1 with a low detection limit of 1.2×10 −12 mol L −1 . Moreover, the proposed biosensor is successfully applied to the detection of choline in milk.
