Luteolin differs as a radical scavenger dramatically from apigenin in response to Cu(II) coordination despite a minor structural difference. Coordination of Cu(II) increases the radical scavenging efficiency of luteolin, especially at low pH, while decreases the efficiency of apigenin at both low and higher pH as studied by ABTS•+ radical scavenging. Luteolin forms a 1:1 complex with Cu(II) binding to 4-carbonyl and 5-phenol for pH <6 and to 3′,4′-catechol for pH >6. Apigenin forms a 1:2 complex independent of pH coordinated to 4-carbonyl and 5-hydroxylyl. Cu(II) coordinated to luteolin, as studied by pH jump stopped-flow, translocates with rate constants of 11.1 ± 0.3 s–1 from 4,5 to 3′,4′ sites and 1.0 ± 0.1 s–1 from 3′,4′ to 4,5 sites independent of Cu(II) concentration, pointing toward the dissociation of Cu(II) from an intermediate with two Cu(II) coordination as rate determining. 3′,4′-Catechol is suggested to be a switch for Cu(II) translocation with deprotonation initiating 4,5 to 3′,4′ translocation and protonation initiating 3′,4′ to 4,5 translocation. For dicoordinated apigenin, the coordination symmetry balances an electron withdrawal effect of Cu(II) resulting in a decrease of phenol acidity and less radical scavenging efficiency compared to parent apigenin. Compared to that of parent luteolin, the radical scavenging rate of both 4,5 and 3′,4′ Cu(II)-coordinated luteolin is enhanced through increased phenol acidity by electron withdrawal by Cu(II), as confirmed by density functional theory (DFT) calculations. Coordination and translocation of Cu(II) accordingly increases the antioxidant activity of luteolin at pH approaching the physiological level and is discovered as a novel class of natural molecular machinery derived from plant polyphenols, which seems to be of importance for protection against oxidative stress.

Background and Purpose Antioxidants provide a promising therapeutic effect for the cardiovascular disease. Luteolin, a polyphenolic bioflavonoid, is known to confer cardioprotection, although the underlying mechanisms, especially the role of luteolin on the antioxidant enzymes, such as the peroxiredoxin family, remain unknown. Experimental Approach We measured the effects of luteolin on myocardial ischaemia/reperfusion (MI/R) injury in vivo (Sprague-Dawley rats) and in vitro , together with the underlying mechanisms, with a focus on signalling by peroxiredoxins. H9c2 cells were used to assess the changes in peroxiredoxins and the other antioxidant enzymes. Oxidative stress, cardiac function, LDH release, ROS and infarct size were also assayed. Key Results Luteolin exerted significant cardioprotective effects in vivo and in vitro via improving cardiac function, increasing the expression of anti-apoptotic protein Bcl-2 and decreasing the pro-apoptotic protein Bax and active caspases 3 and 9, associated with MI/R. Mechanistically, luteolin markedly enhanced expression of peroxiredoxin II, without significant effects on other forms of peroxiredoxin, catalase or SOD1. Molecular docking showed that luteolin could indeed bind to the enzymic active pocket of peroxiredoxin II. Furthermore, down-regulation of peroxiredoxin II by peroxiredoxin II-antisense, administered by adenovirus infection of H9c2 cardiomyocytes, and inhibition of peroxiredoxin II in vivo significantly reversed the cardioprotective effects of luteolin. Conclusions and Implications Our findings, for the first time, demonstrate that luteolin protects against MI/R injury through promoting signalling through the endogenous antioxidant enzyme, peroxiredoxin II, indicating the important beneficial role of this antioxidant system in the heart.

Ionic liquids (ILs)-bonded silica adsorbents were prepared by chemical modification of the silica surface using N -alkylimidazolium-based ILs with Cl − , BF 4
− , PF 6
− or NO 3
− anion, and applied to selective separation of the model mixture of luteolin and aloe-emodin. Among the investigated ILs-bonded silica materials, the silica grafted with N -octyllimidazolium hexafluorophosphate ([C 8 mim]PF 6 @SiO 2 ) exhibited higher adsorption capacity and selectivity of luteolin from the model mixed solution of luteolin and aloe-emodin. The isotherm data of luteolin on [C 8 mim]PF 6 @SiO 2 correlated better to the Freundlich model than the Langmuir model, and the calculated thermodynamic parameters indicated that the adsorption of luteolin was spontaneous and exothermic. The dynamic adsorption and elution experiments demonstrated that the high adsorption capacity and good desorption efficiency of luteolin on fixed bed packed with the [C 8 mim]PF 6 @SiO 2 . Moreover, the results from batch adsorption and dynamic adsorption showed well selective separation towards the model mixture of luteolin and aloe-emodin.

Luteolin is mainly metabolized by phase II enzymes in animals and humans with glucuronidation and sulfation as the two known metabolic pathways. Although methylation of luteolin was reported previously, the structure of the methylated metabolites and the enzymes involved in the process have not been clarified. In our study, two methylated metabolites, M1 (chrysoeriol) and M2 (diosmetin), were identified in the urine after intravenous administration of luteolin to rats, and the data suggested that the methylation was mediated by catechol- O -methyltransferase (COMT). When luteolin was coadministered with a specific COMT inhibitor , entacapone , the formation of M1 and M2 was significantly reduced, whereas the plasma concentration of luteolin increased. Methylation of luteolin was also studied in vitro using rat tissue homogenates. The apparent kinetic parameters associated with the formation of M1 and M2 in vitro were estimated, and regioselectivity of methylation of luteolin was observed. In the in vitro experiment, there was a preference for the formation of M2 over M1. In contrast, accumulation of M1 was preferred in vivo in both rat plasma and urine after an intravenous dose of luteolin. In conclusion, COMT played a crucial role in the disposition of luteolin in rats. Our results indicated that the methylation pathway in rats was significantly reduced when luteolin was coadministered with a specific COMT inhibitor. Therefore, COMT-associated drug-drug interactions need be considered in the future in luteolin clinical trials because the plasma concentrations and related therapeutic effects may be altered in vivo in the presence of a COMT inhibitor.

To improve the performance of biodegradable materials and endow them with functionality, luteolin (Lc) was modified by esterification with acetyl chloride (Ac), iso butyryl chloride (Ic), benzoyl chloride (Bc) to improve its hydrophilicity and hydrophobicity, and Ac-L, Ic-L and Bc-L esters were obtained. The three esterified compounds were characterized and blended with PBS to prepare PBS/Ac-L, PBS/Ic-L and PBS/Bc-L composites. Studied the interface interaction mechanism, crystallization performance, thermodynamic performance, hydrophilic properties, and antibacterial performance between composite materials. The results showed that the majority of Ac-L, Ic-L, and Bc-L were dispersed in the PBS matrix, with a tighter two-phase interface and higher dispersibility. There were electrostatic adsorption, hydrogen bond and other interactions in the composite system, and the Intermolecular force became stronger and the compatibility increased. The introduction of Ac-L, Ic-L, and Bc-L did not change the crystal structure of PBS, but the crystallization rate and crystallinity of the composite material increased. The thermal decomposition temperature of composite materials increased, and the thermal stability and mechanical properties were further enhanced. With the increase of the amount of Ac-L, Ic-L, and Bc-L added, the antibacterial and hydrophobic properties gradually enhanced, making it suitable for use as a preservation and antibacterial material.

Myocardial infarction (MI) triggers a pathological cascade involving inflammation, oxidative stress, and electrophysiological dysfunction, collectively leading to adverse cardiac remodeling. To address these interconnected pathological challenges, we developed an injectable conductive hydrogel (FLCM). Constructed from a thermo-/photo-cross-linkable Pluronic F127 diacrylate matrix functionalized with luteolin, recombinant human collagen type I, and MXene nanosheets, FLCM exhibits excellent injectability, electrical conductivity, mechanical robustness, cell affinity, and sustained drug release capability. In a rat MI model, FLCM implantation effectively attenuated the inflammatory response by reducing pro-inflammatory cytokines (tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6)) while promoting M2 macrophage polarization. Concurrently, it alleviated oxidative stress, preserved connexin 43 expression to enhance myocardial electrical coupling, and significantly lowered the incidence of ventricular arrhythmias. Through these synergistic actions, FLCM improved the infarct microenvironment, thereby suppressing excessive fibrosis, mitigating adverse cardiac remodeling, and restoring left ventricular function. Integrated transcriptomic profiling and protein-level validation revealed that FLCM synchronously modulated extracellular matrix reorganization and coordinated the regulation of the PI3K-Akt and NF-κB signaling pathways, while simultaneously activating the Nrf2/HO-1 antioxidant defense system. Therefore, this study presents an integrated hydrogel system that simultaneously targets multiple post-MI pathological processes, offering a translatable strategy for comprehensive cardiac repair and functional recovery.

N -[3-(Trimethoxysilyl)propyl]ethylenediamine, tetraethyl silicate, nickel acetate, and sodium hydroxide solution were reacted through using anhydrous ethanol as the solvent and heated in a water bath to synthesize nickel-doped amino-modified silica adsorbent material (NDAMS). The structure of NDAMS was characterized by utilizing XRD, BET, FTIR, and XPS. The thermodynamics, kinetics and mechanism of luteolin adsorption on NDAMS were examined. The results exhibit that NDAMS achieves a 94.69% adsorption efficiency for luteolin at 298 K, demonstrating excellent adsorption performance. As the temperature increases, the adsorption amount rises, which conforms to the Langmuir model. The adsorption process aligns with the pseudo-second-order kinetic model, with an enthalpy (Δ H ) of 30.68 kJ/mol, entropy (Δ S ) of 198.70 J/(mol K), and Gibbs free energy (Δ G ) of –28.44 kJ/mol at 298 K. NDAMS may adsorb luteolin through a spontaneous, endothermic and monolayer chemisorption process, in which a bond is formed between the doped nickel atoms and the oxygen atoms in the luteolin’s phenolic hydroxyl group.

Di-(2-ethylhexyl) phthalate (DEHP), an environmental pollutant, has been widely detected in both environmental and clinical samples, representing a serious threat to the homeostasis of the endocrine system. The accumulation of DEHP is notably pronounced in the liver and can lead to liver damage. The lack of effective high-throughput screening system retards the discovery of such drugs that can specifically target and eliminate the detrimental impact of DEHP. Here, by developing a Cy5-modified single-strand DNA-aptamer-based approach targeting DEHP, we have identified luteolin as a potential drug, which showcasing robust efficacy in detoxifying the DEHP by facilitating the expulsion of DEHP in both mouse primary hepatocytes and livers. Mechanistically, luteolin enhances the protein degradation of hepatic urocanate hydratase 1 (Uroc1) by targeting its Ala270 and Val272 sites. More importantly, trans-urocanic acid (trans-UCA), as the substrate of Uroc1, possesses properties similar to luteolin by regulating the lysosomal exocytosis through the inhibition of the ERK1/2 signal cascade. In summary, luteolin serves as a potent therapeutic agent in efficiently detoxifying DEHP in the liver by regulating the UCA/Uroc1 axis.

Despite the broad anticancer activity, whereas the clinical application of luteolin is hindered by unsatisfactory water solubility and non-targeting. Herein, targeted inhibitory effects of luteolin-loading HER2 nanospheres (Her-2-NPs) were successfully prepared by thin film ultrasonic
method. In comparison with the non-targeted nanospheres, Her-2 nanospheres could significantly boost the intake of luteolin in SK-BR-3 cells. The proliferation and apoptosis of breast cancer cells were detected by MTT testing and flow cytometry examination, respectively. Consequently, the
expressions of FOXO1 mRNA level was detected using qPCR assay and protein level was detected using Westernblot. We discovered that Luteolin-loading Her-2 nanospheres could significantly hinder the proliferation of breast cancer cells, down-regulation their migration, and up-regulation FOXO1
expression at mRNA and protein levels, reveal a mechanism whereby luteolin interferes with breast cancer. Collectively, these results suggest Her-2-modified nanospheres increases the efficiency of luteolin uptake and thus improves the treatment benefit of breast cancer.

In this work, the fluorescent copper nanoclusters (Cu NCs) were firstly adopted to detect luteolin with excellent performance. The blue-emitting Cu NCs was successfully prepared through a facile one-pot approach by protection of tannic acid (TA) and chemical reduction of ascorbic acid (AA). The water-soluble nanoclusters possessed uniform size and displayed good stability. The TA-Cu NCs showed maximum luminescence at 434 nm when excited at 366 nm. Based on the static quenching and inner filter effect (IFE) mechanism, the TA-Cu NCs was efficiently and selectively quenched by luteolin. The detection limit was 0.12 μM and linear relationship existed in the range of 0.2–100 μM. Moreover, the TA-Cu NCs probe was successfully employed to detect luteolin in bovine serum samples with satisfactory recoveries. This novel platform was expected to expand the possible detection method based on fluorescence properties.

Biological applications of poly(vinyl alcohol) hydrogels (PVA) are limited because the pure polymer has low hardness and poor mechanical properties. The quality of most PVA hydrogels can be enhanced by addition of molecules that form strong covalent or noncovalent cross-links between the PVA chains. Natural materials as cross-linking agents are attracting interest because of their degradability, cell compatibility, and environmental friendliness. Herein, we report the use of hydrophobic luteolin (LUT) in the facile fabrication of a type of hybrid PVA hydrogel. Even at low LUT concentration (0.04 wt %), the strong hydrogen bonding formed between PVA and LUT caused gelation at 25 °C after the reagents were mixed at high temperature. The effects of LUT concentration, freezing and thawing, and pH on hydrogels mechanics were evaluated, and a possible mechanism of hydrogel formation was deduced. Notably, after freezing and thawing, the elongation (600%) and tensile strength (0.812 MPa) of the PVA/LUT hydrogels were superior to those of common PVA hydrogels. Our study reveals a hydrophobic biocrosslinker for PVA hydrogels that has potential applications in medicine. We expect that this contribution will promote screening of other biocrosslinkers for PVA hydrogels.

Luteolin (3′,4′,5,7-tetrahydroxyflavone, LTL) is a kind of hydrophobic flavonoid, which has a number of pharmacological actions. It has a number of pharmacological actions, including anti-inflammatory, antioxidant, anticancer, and antihepatic fibrosis properties. High-dose LTL, on the other hand, can cause oxidative DNA damage and hydroxyl radical generation. It is critical to create easy, sensitive, and dependable LTL detection technologies and delivery methods for LTL quality control, clinical drug monitoring and enhancing drug bioavailability. In this work, a novel phenylboronic acid derived quantum dots (P-QDs) were synthesized for LTL sensitive detection and pH-responsive delivery. It showed remarkable selectivity and a brilliant fluorescence with a quantum yield of 43.62 %. In vitro experiments reveal negligible cytotoxicity. P-QDs and LTL coupling (P-QDs/LTL) showed powerful proliferation inhibition and migration restriction effects on HepG2 cells. In addition, P-QDs/LTL could time-dependently release it to restore fluorescence in the microacidic environment of HepG2 cells, thus demonstrating satisfactory fluorescence imaging capability. This strategy demonstrated a favourable linear relationship between LTL concentration and the fluorescence intensity of P-QDs in the range of 0.02–200 μM ( R 2 = 0.9952), with a detection limit of 1.16 nM (S/N=3). The recoveries in different real samples were in the range of 85.23 %-108.04 % with RSDs ≤6.1 %. The material is both a new attempt at LTL detection methodology and has significant promise for providing pH-responsive intracellular release of drug monitoring and cancer therapies from naked QDs without the requirement for sophisticated modified grafting.
