An aptasensor with colorimetric and electrochemical dual-outputs for malathion detection utilizing peroxidase-like activity of Fe-MOF

Xu Yiwei,Jia Xupeng,Yang Sennan,Cao Mengrui,He Baoshan,Ren Wenjie,Suo Zhiguang

An aptasensor with dual-outputs was developed for malathion detection. Fe-MOF was synthesized to design favorable signal probes for catalytic amplification. Owing to the excellent peroxidase-like activity of Fe-MOF, the redox reaction was catalyzed to produce the dual-outputs of colorimetric and electrochemical. In this sensing strategy, malathion was captured by the aptamer on sensing interface, leading to the release of signal probe. Thanks to the catalytic amplification of Fe-MOF and the high capture effect of aptamer, the aptasensor produced a sensitive response for malathion. Based on the dual-signals of absorbance and current, the detection method for malathion was developed ranging from 10 ng/mL to 500 ng/mL. The detection limit of malathion was 5.8 ng/mL for colorimetric output and 4.6 ng/mL for electrochemical output. Furthermore, the aptasensor exhibited high specificity and good repeatability in malathion detection. Finally, the aptasensor was applied to detect malathion in fruit and vegetable samples with satisfactory recovery.

Bioenzyme–nanoenzyme–chromogen all-in-one test strip for convenient and sensitive detection of malathion in water environment

Hanwen Zhang,Jiabin Jin,Xini Wang,Wei Han,Lei Qin,Lin Chen,Xiaoyan Mao,Zhenjiang Liu,Xingwei Xiang

The presence of pesticide residues in aquatic environments poses a significant threat to both aquatic ecosystems and human health. The presence of these residues can result in significant harm to aquatic ecosystems and can negatively impact the health of aquatic organisms. Consequently, this issue requires urgent attention and effective measures to mitigate its impact. However, developing sensitive and rapid detection methods remains a challenge. In this study, an all-in-one test strip, which integrated bioenzymes, nanoenzymes, and a chromogen, was developed in combination with an enzyme labeling instrument for a highly sensitive and convenient sensing of malathion residues. The oxidase activity of heme chloride (Hemin) in the strip can catalyze the oxidation of H 2 O 2 and 3,3′,5,5′-tetramethylbenzidine (TMB) to produce a blue-colored oxide. Simultaneously, the alkaline phosphatase (ALP) present in the strip can break down l -ascorbic acid-2-phosphate to produce ascorbic acid (AA). This AA then acts to reduce the oxidized form of TMB, turning it into a colorless substance and leading to the disappearance of its fluorescent signal. In the presence of a pesticide, the activity of ALP is inhibited and formation of AA is blocked, thereby preventing the reduction of oxidized TMB and producing a colored signal. According to this principle, the integrated test strip detected the target pesticide with high performance as per the optical density value determined via an enzyme marker. The detection limit of the test strip was 0.209 ng/mL with good sensitivity. The method was used for detecting malathion in actual river water samples, and the recoveries were in the range of 93.53 %–96.87 %. The newly devised technique effectively identified malathion in samples of natural water. This research has introduced a novel approach for the precise and convenient surveillance of pesticide remnants. Additionally, these discoveries could inspire the advancement of proficient multi-enzyme detection systems.

Organophosphate Insecticide Malathion Induces Alzheimer’s Disease-Like Cognitive Impairment in Mice: Evidence of the Microbiota-Gut-Brain Axis

Jingna Cui,Shouchun Xiao,Yue Cao,Yaru Zhang,Jiaxing Yang,Li Zheng,Fanrong Zhao,Xueke Liu,Donghui Liu,Zhiqiang Zhou,Peng Wang

Evidence suggests that exposure to organophosphate pesticides increases the risk of neurodegenerative diseases, but the mechanisms remain unclear. This study investigated the effects of malathion on Alzheimer’s disease (AD)-like symptoms at environmentally relevant concentrations using wild-type (WT) and APP/PS1 transgenic mouse models. Results showed that malathion exposure induced AD-like cognitive impairment, amyloid-β (Aβ) accumulation, and neuroinflammation in WT mice, with worsened symptoms in APP/PS1 mice. Mechanistic studies revealed that malathion induced AD-like gut microbiota dysbiosis (reduced Lactobacillus and Akkermansia, and increased Dubosiella), causing gut barrier impairment and tryptophan metabolism disruptions. This resulted in a significant increase in indole derivatives and activation of the colonic aryl hydrocarbon receptor (AhR), promoting the kynurenine (KYN) pathway while inhibiting the serotonin (5-HT) pathway. Increased neurotoxic KYN metabolites (3-hydroxykynurenine and quinolinic acid) triggered gut and systemic inflammation, upregulating hippocampal IL-6 and IL-1β mRNA levels and thereby causing neuroinflammation. Gut tryptophan metabolism disruptions caused hippocampal neurotransmitter imbalances, reducing the levels of 5-HT and its derivatives. These effects promoted AD progression in both WT and APP/PS1 mice. This study highlights the crucial role of the microbiota-gut-brain axis in AD-like cognitive impairment induced by malathion exposure, providing insights into the neurodegenerative disease risks posed by organophosphate pesticides.

Electrochemiluminescence Sensor Based on CTS-MoS2 and AB@CTS with Functionalized Luminol for Detection of Malathion Pesticide Residues

Zhiping Yu,Chengqiang Li,Jiashuai Sun,Xia Sun,Guodong Hu

The accumulation of pesticide residues poses a significant threat to the health of people and the surrounding ecological systems. However, traditional methods are not only costly but require expertise in analysis. An electrochemiluminescence (ECL) aptasensor was developed using chitosan and molybdenum disulfide (CTS-MoS2), along with acetylene black (AB@CTS) for the rapid detection of malathion residues. Due to the weak interaction force, simple composite may lead to uneven dispersion; MoS2and AB were dissolved in CTS solution, respectively, and utilized the biocompatibility of CTS to interact with each other on the electrode. The MoS2nanosheets provided a large specific surface area, enhancing the utilization rate of catalytic materials, while AB exhibited excellent conductivity. Additionally, the dendritic polylysine (PLL) contained numerous amino groups to load abundant luminol to catalyze hydrogen peroxide (H2O2) and generate reactive oxygen species (ROS). The proposed ECL aptasensor obtained a low detection limit of 2.75 × 10−3ng/mL (S/N = 3) with a good detection range from 1.0 × 10−2ng/mL to 1.0 × 103ng/mL, demonstrating excellent specificity, repeatability, and stability. Moreover, the ECL aptasensor was successfully applied for detecting malathion pesticide residues in authentic samples with recovery rates ranging from 94.21% to 99.63% (RSD < 2.52%). This work offers valuable insights for advancing ECL sensor technology in future applications.Keywords:electrochemiluminescence;aptamer;CTS-MoS2;AB@CTS;PLL-luminol;nano gold

Enzyme-free multicolor sensor for visual detection of malathion based on a bimetallic magnetic nanozyme-mediated gold nanorod etching

Yuanying Chen,Tianran Lin,Shuangshuang He,Qi Jiao,Shulin Zhao,Li Hou

Malathion plays an important role in agricultural production, and the accurate detection of malathion residues is essential to ensure food safety. A novel enzyme-free sensing method was proposed by employing bimetallic magnetic nanozymes for both colorimetric quantification and multicolor semi-quantitative detection of malathion. The nanozyme exhibits peroxidase-like activity, catalyzing H 2 O 2 -mediated oxidation of 3,3′,5,5′-Tetramethylbenzidine dihydrochloride (TMB). Malathion inhibits this activity via Zr O P coordination, reducing oxidized TMB (oxTMB) generation. Under acidic conditions, oxTMB is further converted to TMB 2+ , which etches gold nanorods, inducing aspect-ratio variations accompanied by distinct multicolor changes. This method demonstrates low detection limits of 0.68 ng/mL. It does not require biological enzymes and can semi-quantify the malathion concentration through color changes. Thus, it can achieve convenient and accurate quantitative analysis without instruments, meeting on-site detection requirements. This work contributes to food safety and the development of efficient and reliable methods for malathion detection.

Facilely Fabricating Bimetallic Ir/Co Nanozymes with High Oxidize-like Activities for Sensitive Detection of Malathion with Portable Swabs

Menghan Liu,Ruyu Zhang,Jie Wang,Huiting Zhang,Jingying Tan,Qingzhi Liu,Lei Han,Lihua Lu

Bimetallic nanozymes not only have strong synergistic catalysis ability but also may improve the catalytic specificity of the nanozymes. Herein, the oxidase (OXD)-mimic Ir/Co bimetallic nanozymes were prepared through a coordination reaction between the Ir(III) complex with the 2-methylimidazole (MeIm) ligands in ZIF-67 under mild conditions without the pyrolysis and physical doping procedures. It is found that the trace amount of Ir-modified ZIF-67 (Ir@ZIF-67) possesses 18.17-fold that of catalytic ability and 7.86-fold that of the reaction rate as those of pristine ZIF-67. On the other hand, compared with ZIF-67, the OXD-like specificity of Ir@ZIF-67 has been increased 9.7%. The theoretical simulation shows that the high-performance of Ir@ZIF-67 comes from its higher O2 adsorption ability. In virtue of the high performing OXD-like catalytic property, Ir/Co bimetallic nanozymes are employed to establish a simple and sensitive colorimetric pesticides detection platform without deleterious H2O2, which offers excellent “switch-on” malathion (MAL) detection in the linear range of 0.05–25 μM with a limit of detection (LOD) of 11.8 nM. Moreover, Ir/Co bimetallic nanozymes are successfully fabricated into the MAL test swabs. Assisted by a color-reading APP, the test swabs can detect MAL with the LOD as low as 18.5 nM. Surprisingly, such a simple test swab can achieve such high sensitivity for MAL detection. The real sample analysis verifies the high potential of the test swabs in practical application. The excellent performance of the fabricated test swabs can promote the formation of the situation where pesticides detection can be tested at any time and any place and can be carried out by anyone. This work not only provides a novel approach to designing polymetallic nanozymes but also displays the promising application of Ir/Co bimetallic nanozymes in the field of colorimetric sensing.

Adolescent exposure to organophosphate insecticide malathion induces spermatogenesis dysfunction in mice by activating the HIF-1/MAPK/PI3K pathway

Shouchun Xiao,Jingna Cui,Yue Cao,Yaru Zhang,Jiaxing Yang,Li Zheng,Fanrong Zhao,Xueke Liu,Zhiqiang Zhou,Donghui Liu,Peng Wang

Chemical-caused reproductive dysfunction has emerged as a global public health concern. This study investigated the adverse effects of the organophosphorus pesticide malathion on reproductive function in adolescent male mice at environmentally relevant concentrations. The results indicated that eight-week malathion exposure reduced testis weight, caused sex and thyroid hormone disorders, and induced testicular spermatogenic epithelium damage and oxidative stress. Testicular RNA sequencing indicated that malathion significantly affected testicular energy metabolism, hypoxia-inducible factor 1 (HIF-1) signaling, and steroid hormone biosynthesis pathways. Malathion significantly increased the gene and protein expression of HIF-1α by upregulating key genes in the mitogen-activated protein kinase (MAPK) pathway ( Map2k2 , Mapk3 , and Eif4e2 ) and the phosphatidylinositol 3-kinase (PI3K) pathway ( Pik3r2 and Akt1 ). Furthermore, malathion downregulated HIF-1α degradation-regulating genes while upregulating anaerobic metabolism and inflammation-related genes, thereby inhibiting normoxia and promoting hypoxia processes. Testicular hypoxia subsequently induced steroid hormone biosynthesis disorders and spermatogenesis dysfunction. Molecular docking verified that malathion interfered with HIF-1α and steroid hormone synthases (CYP11A1, CYP17A1 and CYP19A1) by forming hydrogen bonds and hydrophobic interactions with these proteins. This study presents the first evidence that malathion triggers spermatogenesis dysfunction in mice through activating the HIF-1/MAPK/PI3K pathway, providing a comprehensive understanding of the reproductive toxicity risks associated with organophosphorus pesticides.

Multiple gRNAs-assisted CRISPR/Cas12a-based portable aptasensor enabling glucometer readout for amplification-free and quantitative detection of malathion

Yi Tian,Jiaxuan Chen,Fengzheng Chen,Junru Xu,Lixiang Huang,Liai Peng,Honglei Li,Kai Shi

Background The threat of toxic malathion residues to human health has always been a serious food safety issue. The CRISPR/Cas system represents an innovative detection technology for pesticide residues, but its application to malathion detection has not been reported yet. In addition, the multiple-guide RNA (gRNA) powered-CRISPR/Cas biosensor has the advantages of being fast, sensitive and does not require pre-amplification. However, the reported multiple-gRNA CRISPR/Cas-based biosensors are largely only used for the detection of nucleic acid targets, and there are still certain challenges in detecting non-nucleic acid targets. Results In this work, a multiplex-gRNA-assisted CRISPR/Cas12a-based portable aptasensor (MgCPA) is developed for amplification-free and quantitative detection of malathion using a glucometer. When target malathion is present in the MgCPA strategy, it specifically binds with aptamer and then activates the trans -cleavage activity of the multiplex-gRNA CRISPR/Cas12a. The activated multiple Cas12a/gRNA complexes cut invertase-HP probes on the electrode surface to obtain glucose signals with glucometer assistance. Under optimal conditions, the developed MgCPA strategy achieves satisfactory portable quantitative and sensitive detection of malathion down to 300 fM (S/N = 3) without pre-amplification. Moreover, the satisfactory selectivity, high reproducibility, and good stability of the proposed strategy are also obtained. Due to its excellent and robust shelf life, our developed MgCPA strategy can be practically applied in detecting malathion in orange, apple, cabbage, and spinach samples. Significance Amplification-free, sensitive, portable quantitative and selective detection of malathion in food samples is achieved by employing our developed MgCPA strategy. This strategy not only opens up a new path for the non-nucleic-acid target detection using amplification-free methods based on multiple-gRNA-assisted CRISPR/Cas12a, but also has broad application prospects in ensuring food safety.

Analyte-triggered oxidase-mimetic activity loss of Ag3PO4/UiO-66 enables colorimetric detection of malathion completely free from bioenzymes

Peng Liu,Xin Li,Xuechao Xu,Xiangheng Niu,Mengzhu Wang,Hengjia Zhu,Jianming Pan

Detection of pesticide residues is important because it is related to food and environmental safety. Although lots of sensors have been developed and applied for pesticide analysis, most of them rely on the use of vulnerable bioenzymes, weakening the highlights of these sensors. Here we recommend a bioenzyme-free colorimetric assay of malathion, a typical organophosphorus pesticide, based on the analyte inducing the oxidase-mimicking activity loss of Ag 3 PO 4 /UiO-66. Compared to counterparts, the Ag 3 PO 4 /UiO-66 composite can provide enhanced oxidase-mimetic catalytic activity to trigger the 3,3′-tetramethylbenzidine chromogenic reaction with the participation of dissolved O 2 . When malathion exists, it rapidly adsorbs onto the composite surface via their unique interactions and significantly shields the oxidase-like activity of Ag 3 PO 4 /UiO-66, thus affecting the catalytic chromogenic reaction. With such a principle, sensing of malathion in a wide linear range (0.0083–5.333 μg/mL) was realized with a detection limit 7.5 ng/mL. Given no biological elements were involved in the proposed assay, it exhibited robust and reliable performance in determining malathion in practical samples. Furthermore, a chromogenic hydrogel-based smartphone sensing system was developed for instrument-free on-site analysis of malathion. With good analytical performance and facile operation, our assay and device will find promising applications in phosphothionate pesticide determination.

Sensitive and label-free chemiluminescence detection of malathion using exonuclease-assisted dual signal amplification and G-quadruplex/hemin DNAzyme

Hao Wu,Jun Wu,Hongyong Wang,Yaling Liu,Guoqing Han,Pei Zou

Malathion is one of the most commonly used organophosphorus pesticides that can cause serious harm to the ecological environment and human health. Herein, we demonstrated a label-free chemiluminescent aptasensor for the sensitive detection of malathion based on exonuclease-assisted dual signal amplification and G-quadruplex/hemin DNAzyme. Upon the addition of malathion, the aptamer probe specifically bound to the target to form a complex malathion-S3, leaving a duplex S1-S2. The complex malathion-S3 was digested by exonuclease I and the target was released. The released target was recycled to perform exonuclease I-assisted signal amplification. Furthermore, after treatment with exonuclease III, the duplex S1-S2 was converted into the secondary target ST. The secondary target ST interacted with the hairpin H1 to form a complex H1-ST, which was further digested by exonuclease III and released the secondary target. The released secondary target was recycled to perform exonuclease III-assisted signal amplification. After complete amplification, large numbers of G-quadruplex/hemin DNAzymes were generated. Under the optimal experimental conditions, the prepared aptasensor showed an excellent linear response to malathion with a detection limit of 0.47 pM. The relative standard deviations were in the range of 4.2–6.9%. Moreover, the aptasensor was successfully applied to detect malathion in spiked food and traditional Chinese medicine samples.

Novel electrochemical biosensor based on core-shell nanostructured composite of hollow carbon spheres and polyaniline for sensitively detecting malathion

Linghao He,Bingbing Cui,Jiameng Liu,Yingpan Song,Minghua Wang,Donglai Peng,Zhihong Zhang

A novel electrochemical biosensor was constructed by anchoring acetylcholine esterase (AChE) onto the core-shell nanostructured composite of hollow carbon spheres coated with needle-like polyaniline (denoted by HCS@PANI), and used for rapid determination of malathion, which was established based on the chemisorption/desorption process of AChE used as an indicator. The uniformly distributed HCS was wrapped with the needle-like PANI nanowires to form the HCS@PANI nanocomposite, which not only exhibits high specific surface area and chemical functionality but also possesses strong electrochemical activity. After AChE was adsorbed onto the HCS@PANI nanocomposite, the electrochemical catalysis of AChE/HCS@PANI-based biosensor towards acetylthiocholine chloride (ATCl) was inhibited in the presence of malathion, which was applied to detect malathion, and the biosensor exhibited a low detection limit of 0.16   ng   mL −1 within a linear range from 1.0   ng   mL −1 to 10   μg   mL −1 of the malathion concentration. Also, the results suggested that the developed electrochemical biosensor showed high selectivity, good reproducibility, and applicability for malathion detection, indicating that the proposed method using the HCS@PANI nanocomposite as the sensitive layer for the AChE immobilization and catalysis of ATCl has potential application in high-sensitive pesticide determinations.

A ratiometric fluorescence imprinted sensor based on N-CDs and metal–organic frameworks for visual smart detection of malathion

Lidong Yang,Wei Hu,Fubin Pei,Zhiwei Liu,Jiang Wang,Zhaoyang Tong,Xihui Mu,Bin Du,Mingzhu Xia,Fengyun Wang,Bing Liu

Sensitive and rapid detection of pesticide residues in food is essential for human safety. A ratiometric imprinted fluorescence sensor N -CDs@Eu-MOF@MIP (BR@MIP) was constructed to sensitively detect malathion (Mal). Europium-based metal organic frameworks (Eu-MOF) were used as supporters to improve the sensitivity of the BR@MIP. N -doped carbon dots ( N -CDs) were used as fluorescent source to produce fluorescent signal. A linear relationship between the concentration of Mal and the fluorescence response of the sensor was found in the Mal concentration range of 1–10 μM with a limit of detection (LOD) of 0.05 μM. Furthermore, the sensor was successfully applied for the detection of Mal in lettuce, tap water, and soil samples, with recoveries in the range of 93.0 % − 99.3 %. Additionally, smartphone-based sensors were used to detect Mal in simulated real samples. Thus, the construction of ratiometric imprinted fluorescence sensor has provided a good strategy for the detection of Mal.
