A highly sensitive platform is developed for the determination of microRNA-21 (miRNA-21) with inductively coupled plasma mass spectrometry (ICPMS). It includes the following operations: Hairpin structures DNA H1 and H2 are designed, and DNA H1 is bound to ultrasmall lanthanide upconversion nanoparticles (UCNPs) to produce UCNPs@DNA conjugate probes. Target miRNA triggers a chain reaction for alternating hybridization between DNA H1 (bound on UCNPs@DNA probe) and DNA H2. This leads to UCNPs accumulation and serves as an efficient amplification strategy for UCNPs. The concentration of miRNA-21 is closely correlated to the number of UCNPs; thus, the detection of 89Y by ICPMS provides a promising approach for miRNA quantification. This protocol exhibits high sensitivity to miRNA-21 within 0.1–500 fM, along with a detection limit of 41 aM, which is among the hitherto reported most sensitive procedures. It is worth mentioning that rare earth elements are scarcely present in living systems, which minimizes the background for ICPMS detection and excludes potential interferences from the coexisting species, which is most suited for biological assay.

Mesoporous silica nanospheres (MSNs) are used in a triple signal amplification chemiluminescent (CL) assay for microRNA-21. It is based on (a) the synergistic amplification via loading and controlled-release of signal reagents by MSNs, (b) target recycling amplification, and (c) the enhancement effect of graphene oxide quantum dots (GOQD). CL is generated by the bis(2,4,6-trichlorophenyl) oxalate (TCPO) and H 2 O 2 reaction in the presence of the fluorophore rhodamine B (RB). RB is firstly loaded into the pores of MSNs modified with amino groupsand coupled with ssDNA. Then, the pores are capped by GOQD. Upon the addition of microRNA-21 into the system, the designed ssDNA assumes a double stranded structure. With the aid of duplex-specific nuclease, the double strand structure is cleaved and the free microRNA-21 enters into the next cycling process to combine with other ssDNA forming double strand structures. After several cycling process, amounts of GOQDs departing from the surface of MSNs cause the opening of the pores of MSNs and the release of RB causes the CL of TCPO-H 2 O 2 reaction system. Free GOQDs can lead to a further CL enhancement. By this method, even a low amount of microRNA-21 leads to a large number of released RB molecules and triggers high-intensity CL. The method was applied in an assay where the CL signal increases linearly with the logarithm of the microRNA-21 concentration in the range of 0.005–50 pmol L −1 and the detection limit is 1.7 fmol L −1 (at 3σ).

As one of the most widely distributed microRNAs, microRNA-21 (miRNA-21) significantly regulates target genes’ expression levels and participates in many cellular and intercellular activities, and its abnormal expression is always related to some diseases, especially cancer. Hence, detecting miRNA-21, as a biomarker, at the single-cell level helps us to reveal cell heterogeneity and expression level variation during the state change of cells. In this study, we constructed a gold nanoparticles nanomembrane (AuNPs-NM)-modified plasmonic glass nanopipette (P-nanopipette) surface-enhanced Raman scattering (SERS) sensing platform to sensitively detect content variation of the intracellular miRNA-21 during the electrostimulus (ES)-induced apoptosis process. The cytoplasm-located miRNA-21 was first extracted by using the extraction DNA (HP1)-modified P-nanopipette through a hybridization chain reaction (HCR). The nanopipette was then incubated with a labeling DNA (HP2) and reporter 4-MBA-modified Raman tag. The Raman signal (collected from the tip area near the orifice within 1 μm) showed a good response to the content variation of intracellular miRNA-21 under ES, and the proposed single-cell SERS detection platform provides a simple way to study intracellular substance change and evaluate cancer treatment outcomes.

A simple and novel microRNA (miRNA) biosensor was developed using DNA-Au bio bar code (DNA-Au) and G-quadruplex-based DNAenzyme. DNA-Au increased the amount of miRNA-21 participating in hybridization. Hemin/G-quadruplex DNAenzyme significantly improved the catalysis of H2O2 by oxidation of hydroquinone, resulting in an obvious reduction current of benzoquinone for miRNA-21 indirect detection. Under optimum conditions, the linear relationship between miRNA-21 concentration and reduction response was obtained with the detection limit of 0.006 pM, which showed a good sensitivity. Besides, selectivity of the biosensor was investigated by detecting the base mismatched miRNAs. This proposed method was further applied to detect miRNA-21 extracted from human hepatocarcinoma BEL-7402 cells and human mastocarcinoma MCF-7 cells. The influence of bisphenol A (BPA) on the expression of miRNA-21 in cells was also investigated. The biosensor performs well in practical applications, which suggests it may provide a new platform for gene diagnosis.

Circulating microRNAs (miRNAs) can be regarded as reliable noninvasive biomarkers in body fluids for the early diagnosis, prognosis, and monitoring of cancers. By combining target triggered bipedal DNAzyme walker cleavage cycling amplification and planar intercalated methylene blue (MB) molecules amplification, a versatile ratiometric electrochemical biosensing system is constructed for miRNAs detection. Using the microRNA-21 (miRNA-21) as a triggered model target from breast cancer cells (MCF-7) and cervical cancer cells (HeLa), the sensitivity and feasibility of the ratiometric biosensing strategy were verified on the basis of decreased streptavidin-conjugated cupric [email protected] ( [email protected] ) nanozyme signal with cleaved Zn 2+ -dependent DNAzyme walkers as well as enhanced duplex section of MB signal, which were assisted by the modification of high electronic conductivity and specific surface area of metallic WSe 2 nanoflowers on the electrode. Hence, the introduced sensing strategy of higher cleavage activity of the bipedal DNAzyme walker cyclic amplification resulted into the remarkable sensitive measurement which had a detection limit of 0.16 fM from 1 fM to 1 nM for miRNA-21. Benefiting from the precise design of the capture Hairpin DNA , this proposed method showed excellent specificity to distinguish miRNA-21 from other miRNAs sequences, in addition to possessing good stability and reproducibility. Thus, this versatility platform can be utilized to sense various miRNAs biomarkers by simple of the redesigning the capture Hairpin DNA, hence presents a great promise in clinical application towards early cancer diagnosis, biological analysis and prognosis.

Cellulose nanocrystals (CNCs) and polydopamine (PDA) have shown great application potential in the biomedical field due to their excellent biocompatibility. Using PDA@CNC as a carrier to construct functional nanocomposites for targeted fluorescence imaging of tumor cells and effective cancer treatment can expand the applications of CNCs in tumor diagnosis and treatment. Here, we conveniently modified microRNA-21 inhibitors (miRNAi) on PDA@CNC to construct a multifunctional nanocomposite (FAM-miRNAi@PDA@CNC) for achieving in situ fluorescence imaging of tumor biomarkers and photothermal effects to induce death of breast cancer cells. The overexpressed miRNA-21 in breast cancer cells hybridizes with FAM-miRNAi, triggering green fluorescence of FAM to recognize breast cancer cells. The rod-shaped morphology of FAM-miRNAi@PDA@CNC can effectively promote the endocytosis of FAM-miRNAi@PDA@CNC by the cell membrane. In addition, CNCs can stabilize the photothermal performance of PDA, and the photothermal conversion efficiency of PDA@CNC reached up to 61.66%, which will help FAM-miRNAi@PDA@CNC to achieve 70% apoptosis rate of breast cancer cells under 808 nm laser irradiation. The design of FAM-miRNAi@PDA@CNC provides a valuable pathway for CNCs utilization in biomedicine and accurate diagnosis and treatment of breast cancer.

In this work, a target-triggered three-way junction (3-WJ) structure was combined with enzyme-powered cascade amplification strategy for ultrasensitive electrochemiluminescence (ECL) detection of microRNA via free-running DNA walker. In the presence of target microRNA-21 (miRNA-21), a three-way junction structure was formed to trigger enzyme-aided multiple DNA amplification, and thus exponentially amplified triggers were generated. Then, the trigger DNA could hybridize with the blocking DNA on the electrode to release walker DNA, which specifically combined with the quantum dots (QDs)-DNA probe, and formed a recognition site for Nt.BbvCI. The movement of DNA walker is powered by the nicking endonuclease that cleaves specific QDs-DNA probes on the track. Thus one walker DNA can automatically cleave multiple QDs probes on the electrode during the movement, resulting in significantly amplified changes of ECL signal, which could be used for the highly sensitive detection of target miRNA-21. Under the optimal conditions, the ECL signal change is linear with the concentration of miRNA-21 in the range from 10 −14 M to 10 -7 M with a detection limit of 1.5 fM. This newly established strategy could achieve rapid, isothermal, and homogeneous signal amplification for specific nucleic acids in complicated biomatrix, which hold great potential for application in early clinical diagnosis.

MicroRNA exhibits different levels of expression in cancer and can affect the transformation, metastasis, and carcinogenesis of the cancer cell. Herein, we developed a novel kind of electrochemical microRNA biosensor based on two-dimensional nanomaterial of antimonene nano-flakes (AMNFs) and carbon quantum dots (CQDs) which were used as substrating to cadmium ion (Cd 2+ ) for specific detection of breast cancer-relevant biomarker-microRNA-21. Compared to graphene, the first principle energetic calculation shows that the AMNFs have completely a stronger force interaction with single strand (ssRNA), due to the antimonene has a more delocalized 5 s/5p orbital. After the addition of complementary microRNA, due to the low adsorption affinity of double-stranded RNA (dsRNA) to antimonene, the hybridized target is easy to desorb from the antimonene interface, and the oxidation peak of metal ions is significantly reduced. Results showed the microRNA-21 concentration can be detected from 100 aM to 1 nM, the limit of detection as low as 21 aM toward microRNA-21, which is 3 times lower than those of the established microRNA biosensors. The unique combination of not be attempted before existing sensing material which has special adsorption properties represents an approach to the detection of breast cancer. And it provides a promising method for early diagnosis, monitoring, and staging of breast cancer.

Abnormal expression of microRNAs is greatly associated with the occurrence of various cancer types, revealing great potential of microRNA as biomarkers for cancer diagnosis and prognosis. Herein, a MXene-MoS 2 heterostructure enhancing electrochemical biosensor coupled with catalytic hairpin assembly (CHA) amplification approach for label-free determination of microRNA-21 (miR-21) was successfully assembled. In particular, the unique micro-nano heterostructure with large specific area and favorable electroconductivity exhibited the ability of excellent confinement effect. Thus, rendered the MXene-MoS 2 heterostructure the ability to trigger more target recycling reaction, giving new vitality to the traditional CHA amplification method. Meanwhile, thionine (Thi) and gold nanoparticles (AuNPs) were anchoring at the surface of MXene-MoS 2 heterostructure, respectively, empowered the sensor the capability of capture probes fixation and miR-21 label-free determination. When numerous electronegative double-stranded DNA generated, the electron transfer was greatly hindered, resulting in signal decrease. Accordingly, the design denoted a broad dynamic range from 100 fM to 100 nM and a detection limit of about 26 fM, comparable or lower than previous reported methods for miR-21 detection. Furthermore, the sensing platform supplied satisfactory selectivity , reproducibility and stability towards the miR-21 detection. The real sample determination also showed a promising performance under clinical circumstance. Finally, from the clinical standpoint, the proposed biosensor is a considerable platform toward early disease detection and monitoring.

MicroRNA (miRNA) upconversion nanoprobes using organic dyes as acceptors have attracted intensive attention in early cancer diagnosis, but still remains a large challenge from the unsatisfactory luminescence resonance energy transfer (LRET) efficiency. Herein, we proposed to integrate the confined emitters effect and target cycling amplification strategy into a single matrix for establishing the upconversion nanoamplicon with high LRET efficiency, allowing for accurately reporting the levels of miRNA originated from cancer cells. As a proof-of-concept, the emitting ions were confined in the shell of the core-shell upconversion nanoparticles (UCNPs), enabling an enough close distance to its surface energy acceptors. The tentacle DNAs (P-tDNA-M), phosphorylated and methylated at both ends, were immobilized onto the UCNPs surfaces for effectively capturing hairpin DNA1 labeled with BHQ1 (BHQ1-HP1) to form BHQ1-UCNPs energy donor-acceptor pair, achieving ∼ 67.9 % of LRET efficiency. Target miRNA-21 could bind with HP1 to open the loop, causing BHQ1 away from UCNPs, thereby restoring the upconversion luminescence, allowing for high sensitively detecting miRNA-21 with a detection limit of 75.3 fM. Interestingly, this naoprobe exhibited good performances for monitoring miRNA-21 in human serum samples and evaluating the expression level of miRNA-21 in various cell lines. This proposed platform gives a new insight to design the efficient upconversion nanoprobes for precisely detecting low-expressed miRNA in complex biological samples.

Background MicroRNA-21 (miRNA-21) has emerged as a promising biomarker for early cancer diagnosis, yet its accurate quantification in complex biological matrices remains a significant challenge. Conventional SERS-based biosensors are inherently susceptible to non-specific adsorption of biomolecules and probe tags, which generates substantial background interference and severely compromises the signal-to-noise ratio (SNR) and detection fidelity. While various signal amplification strategies have been extensively explored, the critical issue of actively suppressing non-specific noise at the sensing interface has been largely overlooked, necessitating innovative approaches that can simultaneously enhance specific signals and minimize background interference. Results We develop an electrokinetic SERS biosensor that actively modulates the SNR through two sequential voltage-gated steps in a sandwich-structured detection format. A positive potential preconcentrates the negatively charged miRNA-21 targets and SERS probes at the electrode interface, significantly increasing their local concentration and accelerating specific hybridization kinetics, while a subsequent reverse potential acts as an electrostatic filter to selectively desorb non-specifically adsorbed probes, effectively purifying the sensing interface. This synergistic combination of preconcentration and separation efficiently decouples specific signals from non-specific interference, achieving a wide linear detection range from 1.0 × 10 −15 to 1.0 × 10 −10 M with an ultralow detection limit of 0.48 fM. The biosensor exhibits excellent selectivity in discriminating single-base mismatches and demonstrates robust analytical performance in human serum samples, with recoveries ranging from 95.4% to 104%. Significance This work introduces a novel paradigm for high-fidelity SERS biosensing by actively tuning the SNR through precise dual-action electrokinetic control. The strategy offers a generalizable, enzyme-free solution to the challenge of non-specific adsorption, critically enhancing both detection accuracy and practical applicability in complex biological matrices. The electrokinetic approach provides a versatile platform for reliable biomolecular analysis.

Natural products were extracted from traditional Chinese herbal emerging as potential therapeutic drugs for treating cardiovascular diseases. This study examines the role and underlying mechanism of dihydromyricetin (DMY), a natural compound extracted from Ampelopsis grossedentata , in atherosclerosis. DMY treatment significantly inhibits atherosclerotic lesion formation, proinflammatory gene expression and the influx of lesional macrophages and CD4-positive T cells in the vessel wall and hepatic inflammation, whereas increases nitric oxide (NO) production and improves lipid metabolism in apolipoprotein E-deficient ( Apoe − / − ) mice. Yet, those protective effects are abrogated by using NOS inhibitor L-NAME in Apoe − / − mice received DMY. Mechanistically, DMY decreases microRNA-21 (miR-21) and increases its target gene dimethylarginine dimethylaminohydrolase-1 (DDAH1) expression, an effect that reduces asymmetric aimethlarginine (ADMA) levels, and increases endothelial NO synthase (eNOS) phosphorylation and NO production in cultured HUVECs, vascular endothelium of atherosclerotic lesions and liver. In contrast, systemic delivery of miR-21 in Apoe − / − mice or miR-21 overexpression in cultured HUVECs abrogates those DMY-mediated protective effects. These data demonstrate that endothelial miR-21-inhibited DDAH1-ADMA-eNOS-NO pathway promotes the pathogenesis of atherosclerosis which can be rescued by DMY. Thus, DMY may represent a potential therapeutic adjuvant in atherosclerosis management.
