Nebulized riclinoctaose mitigates ovalbumin-induced allergic asthma by attenuating mast cell activation

Longwei Yang,Wenhao Ge,Xi Lin,Ning Yu,Xiaodong Xu,Jianfa Zhang

Allergic asthma is an inflammatory airway disease in which mast cells play a key role in its pathogenesis. Riclinoctaose (Rios), an octasaccharide composed of glucose and galactose, has been reported to modulate macrophage polarization in renal ischemia-reperfusion injury. This study investigates the effects of nebulized Rios in an ovalbumin (OVA)-induced allergic asthma model. Nebulized Rios significantly reduced airway hyperresponsiveness (AHR), inflammatory cell infiltration, pulmonary fibrosis, and mucus production in OVA-treated mice. Additionally, Rios suppressed lung inflammatory cytokine production and protected against OVA-induced oxidative injury. In bronchoalveolar lavage fluid (BALF), Rios decreased the number of neutrophils and macrophages, as well as serum immunoglobulin E (IgE) and interleukin-4 (IL-4) levels. Metabolomics analysis using 1 H NMR showed significant changes in the lung metabolic profile of OVA-induced asthma mice, which were partially reversed by Rios treatment. Confocal laser microscopy (CLM) revealed that fluorescently labeled Rios specifically binds to mast cell membranes. In vitro, Rios reduced degranulation and inflammatory responses in mast cells RBL-2H3 following anti-Dinitrophenyl-Immunoglobulin E (anti-DNP-IgE) sensitization and anti-Dinitrophenyl-Bovine Serum Albumin (anti-DNP-BSA) stimulation. These results suggest that Rios has therapeutic potential for allergic asthma, likely through the modulation of mast cell degranulation and mediator release.

Modulating ovalbumin-lipid bilayer interactions via fatty acid modification: A physicochemical characterization

Yibang Zhang,Dong Chen,Yang Liu,Aisha Labaran Ammani,Atena Ahmadian,Aftab Ullah

Protein-lipid interactions are pivotal in various biological processes, including membrane trafficking, signal transduction, and cellular compartmentalization. This study investigates how fatty acid modification alters the physicochemical properties of ovalbumin (OVA) and its interactions with lipid bilayers, with implications for controlled release applications. We hypothesize that modifying OVA with different fatty acids will significantly alter its surface hydrophobicity and membrane affinity, thereby influencing its potential for targeted drug delivery. OVA was chemically modified with four fatty acids (myristic, palmitic, stearic, and oleic acids) using N-hydroxysuccinimide (NHS) esters. Surface hydrophobicity was assessed via 8-Anilino-1-naphthalenesulfonic acid ammonium (ANS) fluorescence assays, revealing that modifications increased hydrophobicity, particularly with longer fatty acid chains. Interactions with liposomes were characterized using dynamic light scattering (DLS) and zeta potential measurements, indicating enhanced liposome binding and altered bilayer properties due to fatty acid modifications. Quartz crystal microbalance with dissipation (QCM-D) analysis confirmed the insertion of fatty acid chains into the lipid bilayer. In vitro release experiments demonstrated that fatty acid modification significantly affected the release kinetics of OVA, with oleic acid-modified OVA showing the lowest cumulative release. Our findings highlight the role of fatty acid structure in modulating protein-lipid interactions and underscore the potential of fatty acid-modified OVA for developing targeted drug delivery systems. Future work will explore the underlying mechanisms and evaluate the therapeutic potential of these modified proteins in vivo.

Modulation of ovalbumin gel properties via liposome incorporation: Structural and mechanistic insights

Ying Liu,Haoyu Yu,Chenyang Fan,Min-hsiung Pan,Yi-shiou Chiou,Baomiao Ding

The study investigated the enhancement of ovalbumin (OVA) gel properties through the incorporation of liposomes (Lip) and clarified the mechanisms responsible for these improvements, with the aim of addressing limitations in water-holding capacity (WHC), gel strength, and stability commonly observed in native OVA systems. Lip was incorporated into OVA gels at various Lip-to-OVA ratios, and Lip-OVA gel properties were evaluated and analyzed. Results demonstrated that Lip-OVA gel WHC, strength and freeze-thaw stability were significantly improved with the Lip-to-OVA ratio increasing. SEM micrographs revealed a denser and more homogenous network structure in Lip-OVA gels compared to native OVA gels. FTIR and fluorescence spectroscopy indicated that hydrogen bonding, hydrophobic interactions, and protein conformational rearrangements were pivotal to gel enhancement. Surface hydrophobicity and contact angle measurements further confirmed increased exposure of hydrophobic groups and supported conformational rearrangement. The increase in free sulfhydryl content and the change in CD spectra indicated that OVA structure shifted towards a decrease in α-helix content and an increase in β-sheet content, leading to the unfolding of OVA and promoting intermolecular cross-linkage. The primary forces contributing to the improved gel structure were identified as hydrophobic interactions and disulfide bonding, with ionic and hydrogen bonds playing a supportive role. Furthermore, it was observed that the gel stabilization shifted toward hydrophobic interactions over disulfide bonding after incorporation with Lip. These findings provide mechanistic insight into how Lips modulate protein gel structure and function, offering a strategy to design protein-based gels with enhanced texture, stability, and potential for advanced food applications.

Ginsenoside Rh1 attenuates ovalbumin-induced asthma by regulating Th1/Th2 cytokines balance

Li Qiang,Zhai Chunmiao,Wang Guodong,Zhou Jia,Li Weiguang,Xie Liquan,Shi Zhanli

Ginsenoside Rh1 (Rh1) has anti-inflammatory effects in asthma mice, but the underlying mechanism remains unclear. BALB/c mice were sensitized and challenged with ovalbumin (OVA) to construct asthma model. Mice received Rh1 or tiotropium bromide 0.5 h before OVA challenge. Airway morphology and airway remodeling were assessed by HE staining and Masson's trichrome staining, respectively. Th1/Th2 cytokines in serum or broncho alveolar lavage fluid (BALF) were measured by ELISA kits. Rh1 significantly alleviated the lung resistance and airway resistance, and reduced the number of total inflammation cells, eosinophils, neutrophils, and lymphocytes in BALF of the asthmatic mice. The morphological changes and collagen deposition of airway were also reduced by Rh1 in asthmatic mice. The increase of Eotaxin, IL-4, IL-5, IL-13, and IL-33 and the decrease of IL-12 and IFN-γ in both BALF and serum of OVA exposed mice were reversed by Rh1. Rh1 attenuates OVA-induced asthma in the mice model by regulating Th1/Th2 cytokines balance.

Improved oxidative stability of fish oil emulsion by grafted ovalbumin-catechin conjugates

Jin Feng,He Cai,Hua Wang,Chunyang Li,Songbai Liu

Conjugates of ovalbumin (OVA) with (+)-catechin (C), (−)-epigallocatechin (EGC) or (−)-epigallocatechin gallate (EGCG) were synthesized through a free-radical grafting approach. The covalent binding between OVA and catechins was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). OVA-catechin conjugates were further characterized by a combination of steady-state fluorescence spectroscopy, differential scanning calorimetry (DSC), Fourier transform infrared (FTIR) spectroscopy and circular dichroism (CD) analyses. Results suggested that the conjugates contained less α-helix but more β-sheet secondary structure than OVA. The catechin-grafting reaction led to improved surface hydrophobicity and decreased tertiary conformation stability of OVA. Compared with that emulsified by OVA, fish oil emulsion coated by conjugates demonstrated smaller droplet size, better storage stability and less viscosity. Besides, conjugates inhibited the lipid oxidation in fish oil emulsion more effectively than OVA because of their higher antioxidant activity and interfacial accumulation.

Poria cocos polysaccharide induced Th1-type immune responses to ovalbumin in mice

Xiaoxiao Dong,Boye Li,Boyang Yu,Tian Chen,Qin Hu,Bo Peng,Wang Sheng

In the present study, we evaluated adjuvant potential of Poria cocos polysaccharide (PCP) on the Th1-type immune responses of C57/BL6 mice against ovalbumin (OVA). We first determined the effect of PCP on maturation of murine bone marrow derived dendritic cells (BMDCs), PCP significantly upregulated surface expression of MHCII, CD40, CD80, CD86 and enhanced production of IL-6 and IL-12p40. In addition, PCP affected receptor-mediated endocytosis, but not pinocytosis in BMDCs. Furthermore, OVA + PCP immunization induced specific cytotoxic CD8 + T cell killing of OVA (257–264) peptide pulsed cell. When mice were immunized subcutaneously in a week interval with OVA + PCP. Serum were collected for measuring OVA-specific antibody and splenocytes were harvested for analyzing CD69, IFN-γ ELISpot and cytokines production. The result indicated that OVA-specific IgG, IgG2a and IgG1 antibody levels in serum were significantly elevated by PCP compared with control. PCP increased OVA-specific IFN-γ-secreting CD8 + , CD4 + T cells, promoted CD8 + T cell proliferation and up-regulated Th-1 type (IFN-γ, IL-2) cytokine production. In conclusion, data suggest that PCP enhanced cellular immune response and possess potential as a vaccine adjuvant for Th1 immune response.

Comparative study of the interactions between ovalbumin and three alkaloids by spectrofluorimetry

Wang Rui-qiang,Yin Yu-jing,Li Hua,Wang Yi,Pu Juan-Juan,Wang Rui,Dou Huan-jing,Song Chuan-jun,Wang Rui-yong

The interaction between ovalbumin (OVA) and three purine alkaloids (caffeine, theophylline and diprophylline) was investigated by the aid of intrinsic and synchronous fluorescence, ultraviolet-vis absorbance, resonance light-scattering spectra and three-dimensional fluorescence spectra techniques. Results showed that the formation of complexes gave rise to the fluorescence quenching of OVA by caffeine, theophylline, and diprophylline. Static quenching was confirmed to results in the fluorescence quenching. The binding site number n, apparent binding constant K A and corresponding thermodynamic parameters were measured at different temperatures. The binding process was spontaneous molecular interaction procedures in which both enthalpy and Gibbs free energy decreased. Van der Waals forces and hydrogen bond played a major role in stabilizing the complex. The comparison between caffeine, theophylline, and diprophylline was made, and thermodynamic results showed that diprophylline was the strongest quencher and bound to OVA with the highest affinity among three compounds. The influence of molecular structure on the binding aspects was reported.

Improved bioavailability of curcumin in ovalbumin-dextran nanogels prepared by Maillard reaction

Jin Feng,Shanshan Wu,Hua Wang,Songbai Liu

In this work, ovalbumin-dextran nanogels were fabricated via the Maillard reaction followed by a heat-gelation process and their potential to improve curcumin bioavailability was investigated using an in vitro gastrointestinal tract. The result of electrophoresis, conjugation efficiency and yield, and far-UV circular dichroism verified the covalent attachment of dextran to ovalbumin by the Maillard reaction. The conjugates were then heated at the isoelectric point of ovalbumin to give stable nanogels. The transmission electron microscopy observation suggested that nanogels displayed a spherical structure. Curcumin was loaded into nanogels by a pH-driven method and the incorporation of curcumin affected the morphology of nanogels marginally. The transformation of curcumin in nanogels was significantly higher than that in ovalbumin nanoparticle. On the other hand, the bioaccessibility was similar for curcumin in nanogels and nanoparticle. The ovalbumin-dextran nanogels hold the potential as delivery systems for curcumin to fortify functional foods and beverages.

Thermally induced gelation behavior and fractal analysis of ovalbumin-carboxymethylcellulose electrostatic complexes

Wenfei Xiong,Cong Ren,Xiaoying Xu,Jing Li,Lifeng Wang,Bin Li

The main purpose of this work was to investigate the effect of electrostatic combination between ovalbumin (OVA) and carboxymethylcellulose (CMC) with different substitution degree (CMC0.7 and CMC1.2, the higher substitution degree means greater charge density) on the thermally induced gelation behavior of OVA. It has been found that electrostatic combination with CMC greatly inhibited OVA thermal gelation, and CMC 1.2 with higher charge density exhibited stronger inhibition than CMC 0.7. Rheological fractal analysis showed that the thermogels of OVA/CMC 0.7 and OVA/CMC 1.2 electrostatic complexes were in the transition ragime. Furthermore, as the OVA-CMC ratio increased from 10:1 to 40:1, the fractal dimensions of the OVA/CMC 0.7 and OVA/CMC 1.2 thermally induced gels increased from 1.83 and 1.75 to 2.17 and 2.08, respectively. This result indicated that an increasing in the proportion of protein resulted in a more compact fractal aggregate structure, allowing the gel to exhibit a higher elasticity modulus (G′). On the other hand, the charge shielding effect of salt ions enhanced the thermogel strength of the OVA/CMC complexes. However, when salt ions (100 mM) completely dissociate the protein molecules on the polysaccharide chain, the apparent viscosity of the polysaccharide has a strong influence on the thermal gelation of OVA. At this time, CMC 0.7 having a lower charge density exhibited a stronger anti-gelation ability of OVA than CMC 1.2 because of its higher apparent viscosity. This kind of novel OVA/CMC electrostatic complexes thermogel could provide a potential new approach to the design and regulation of food texture.

Interaction Mechanism of Mangiferin and Ovalbumin Based on Spectrofluorimetry and Molecular Docking

Si Chen,Shiyu Zhen

Mangiferin (MAG) is a kind of polyphenol with many bioactivities. However, its application in medicines and functional foods is restricted because of its poor aqueous solubility and stability. The construction of a MAG/protein complex is an effective way to solve this bottleneck. In this study, the interaction of MAG and ovalbumin (OVA) was systematically investigated by spectrofluorimetry, and their binding mode was clarified based on molecular docking. The results suggested that MAG could cause the static fluorescence quenching of OVA with the quenching constant (Kq) of >2 × 1010 L/(mol·s). Their binding performance increased with increasing temperature, and the binding-site number (n) was close to 1. The thermodynamic analysis indicated that the binding was a spontaneous process, which was mainly driven by hydrophobic force. During this process, there was no apparent change in the microenvironment surrounding the tyrosine and tryptophan residues of OVA. The molecular docking results demonstrated the hydrophobic interaction and hydrogen bonding in the complex, which well-confirmed the results of the fluorescence experiments.

Enhancing Stability and Antioxidant Activity of Resveratrol-Loaded Emulsions by Ovalbumin–Dextran Conjugates

Wen Zhang,Lingli Meng,Xinyi Lv,Limin Wang,Pei Zhao,Jinrong Wang,Xinping Zhang,Jinyu Chen,Zijian Wu

A reliable strategy for improving the stability and shelf life of protein-stabilized systems is by covalently attaching the protein onto a polysaccharide. In this study, ovalbumin (OVA) was modified with dextran (DEX) of different molecular weights by the Maillard reaction, and was used to enhance the stability of emulsions loaded with resveratrol. The surface hydrophobicity, thermal stability, and FT-IR spectroscopy of the OVA–DEX conjugates were evaluated. The results showed that the surface hydrophobicity of OVA decreased, while the thermal stability of OVA was significantly improved after DEX covalent modification. The OVA–DEX1k-stabilized emulsion exhibited high encapsulation efficiency of resveratrol, with the value of 89.0%. In addition, OVA–DEX was considerably more effective in droplet stabilization against different environmental stresses (heat, pH, and ionic strength). After 28 days of storage at 25 °C, the OVA-stabilized emulsion showed faster decomposition of resveratrol, whereas the OVA–DEX-conjugate-stabilized emulsion had approximately 73% retention of resveratrol. Moreover, the antioxidant activity of resveratrol-loaded emulsions stabilized by OVA–DEX was higher during storage under different temperatures. These results proved that the OVA–DEX conjugates had the potential to form stable, food-grade emulsion-based delivery systems against environmental stresses, which strongly supports their potential in the field of food and biomedical applications.

Gut Microbiota-Derived Tryptophan Metabolites Alleviate Allergic Asthma Inflammation in Ovalbumin-Induced Mice

Hongchao Wang,Yuan He,Danting Dang,Yurong Zhao,Jianxin Zhao,Wenwei Lu

Asthma is a prevalent respiratory disease. The present study is designed to determine whether gut microbiota-derived tryptophan metabolites alleviate allergic asthma inflammation in ovalbumin (OVA)-induced mice and explore the effect and potential mechanism therein. Asthma model mice were constructed by OVA treatment, and kynurenine (KYN), indole-3-lactic acid (ILA), in-dole-3-carbaldehyde (I3C), and indole acetic acid (IAA) were administered by intraperitoneal injection. The percent survival, weight and asthma symptom score of mice were recorded. The total immunoglobulin E and OVA-specific (s)IgE in the serum and the inflammatory cytokines in the bronchoalveolar lavage fluid (BALF) were detected by the corresponding ELISA kits. The composition of the gut microbiota and tryptophan-targeted metabolism in mouse feces were analyzed using 16S rRNA gene sequencing and targeted metabolomics, respectively. The four tryptophan metabolites improved the percent survival, weight and asthma symptoms of mice, and reduced the inflammatory cells in lung tissues, especially I3C. I3C and IAA significantly (p< 0.05) downregulated the levels of OVA-IgE and inflammatory cytokines. KYN was observed to help restore gut microbiota diversity. Additionally, I3C, KYN, and ILA increased the relative abundance ofAnaeroplasma,Akkermansia, andRuminococcus_1, respectively, which were connected with tryptophan metabolic pathways. IAA also enhanced capability of tryptophan metabolism by the gut microbiota, restoring tryptophan metabolism and increasing production of other tryptophan metabolites. These findings suggest that tryptophan metabolites may modulate asthma through the gut microbiota, offering potential benefits for clinical asthma management.
