Aldehyde group pendant-grafted pectin-based injectable hydrogel

De-qiang Li,Maryamgul Tohti,Yong-sheng Fu,Yue Zhang,Zi-wei Xiong,Jun Li,Yan-Feng Guo

Periodate oxidation has been the widely accepted route for obtaining aldehyde group-functionalized polysaccharides but significantly influenced the various physicochemical properties due to the ring opening of the backbone of polysaccharides. The present study, for the first time, presents a novel method for the preparation of aldehyde group-functionalized polysaccharides that could retain the ring structure and the consequent rigidity of the backbone. Pectin was collected as the representative of polysaccharides and modified with cyclopropyl formaldehyde to obtain pectin aldehyde (AP), which was further crosslinked by DL-lysine (LYS) via the Schiff base reaction to prepare injectable hydrogel. The feasibility of the functionalization was proved by FT-IR and 1 H NMR techniques. The obtained hydrogel showed acceptable mechanical properties, self-healing ability, syringeability, and sustained-release performance. Also, as-prepared injectable hydrogel presented great biocompatibility with a cell proliferation rate of 96 %, and the drug-loaded hydrogel exhibited clear inhibition of cancer cell proliferation. Overall, the present study showed a new method for the preparation of aldehyde group-functionalized polysaccharides, and the drug-loaded hydrogel has potential in drug release applications.

Sunflower-Derived Pectin Oligogalacturonides Promote Defecation in Mice

Hongming Gu,Xian Qu,Xiaotong Sheng,Xuran Wang,Yuhan Cai,Jie Geng,Liangnan Cui,Kevin H. Mayo,Yifa Zhou,Hairong Cheng,Guihua Tai

Sunflower pectin is widely used as a food additive due to its gelling and stabilizing properties. However, its bioactive potential remains poorly understood. Here, we explored its effect on defecation. Using a diphenoxylate-induced constipation mouse model, we found that native sunflower pectins did not significantly promote defecation. Subsequently, we investigated whether they could be activated via biotransformation. For this, we enzymatically hydrolyzed the pectin and produced a number of fragments that covered a wide range of molecular weights (0.5 to 30 kDa) in two esterification states (0% and 10% methyl esterification). Interestingly, all fragments exhibited defecation-promoting effects. Moreover, the effect progressively increased as the molecular weight decreased, a trend that was observed with both esterified and non-esterified fragments. Comparative studies identified E3 as the most effective fraction. E3 also displayed a prebiotic effect in the gut of model mice. Structural analysis indicated that E3 comprises oligogalacturonides with degrees of polymerization from one to six, mainly DP3 and DP4. Overall, our study reveals an inverse correlation of molecular weight with defecation-promoting activities, and offers a simple and scalable approach to convert ineffective pectin into effective oligogalacturonides that have the potential to become functional food ingredients.

Alkylated pectin hydrogels as potential protein drug carriers

Xiao-hong He,Rui-hong Liang,Cheng-mei Liu,Wei Liu,Rui-yun Chen,Xiao-xue Hu,Jun Chen

Pectin has been used in the development of safe and effective drug delivery systems because of its unique physicochemical and biological characteristics. However, it still needs to be modified to overcome some inherent drawbacks. In the present study, pectin was alkylated by bromododecane. The C 12 -alkylated pectin (PC 12 ) can form strong hydrogels in the presence of Ca 2+ . Such hydrogels were explored to prepare bovine serum albumin (BSA)-loaded microspheres (PC 12 -MS). The BSA encapsulation yield of PC 12 -MS was significantly higher than that of microspheres prepared with the original pectin (PC-MS). Differential scanning calorimetry analysis indicated that interactions existed between BSA and pectin during the preparation of microspheres. Release tests showed that PC 12 -MS almost avoided BSA release in simulated gastric fluids, which was verified by environmental scanning electron microscopy analysis, and exhibited an effective controlled release throughout the simulated gastrointestinal tract compared to PC-MS. In addition, an MTT assay showed the biocompatibility of C 12 -alkylated pectin. Thus alkylated pectin may serve as a potential protein drug carrier. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Cross-Linked Pectin Nanofibers with Enhanced Cell Adhesion

Sainan Chen,Sisi Cui,Hui Zhang,Xuejing Pei,Junli Hu,Yifa Zhou,Yichun Liu

Polysaccharides display poor cell adhesion due to the lack of cell binding domains. This severely limits their applications in regenerative medicine. This study reports novel cross-linked pectin nanofibers with dramatically enhanced cell adhesion. The nanofibers are prepared by at first oxidizing pectin with periodate to generate aldehyde groups and then cross-linking the nanofibers with adipic acid dihydrazide to covalently connect pectin macromolecular chains with adipic acid dihydrazone linkers. The linkers may act as cell binding domains. Compared with traditional Ca2+-cross-linked pectin nanofibers, the pectin nanofibers with high oxidation/cross-linking degree exhibit much enhanced cell adhesion capability. Moreover, the cross-linked pectin nanofibers exhibit excellent mechanical strength (with Young’s modulus ∼10 MPa) and much enhanced body degradability (degrade completely in 3 weeks or longer time). The combination of excellent cell adhesion capability, mechanical strength, and body degradability suggests that the cross-linked pectin nanofibers are promising candidates for in vivo applications such as tissue engineering and wound healing. This cross-linking strategy may also be used to improve the cell adhesion capability of other polysaccharide materials.

Citrus flavonoid-pectin conjugates with enhanced emulsifying properties

Xitong Liu,Fengzhang Wang,Jirong Wang,Ting Liu,Chengying Zhao,Jinkai Zheng

Pectin and flavonoids are important components of citrus fruits, but the effects of flavonoid grafting on the emulsifying properties of citrus pectin remain unclear. In this study, a novel natural citrus flavonoid-pectin conjugate in citrus peel was identified through alkaline hydrolysis combined with ultra-high performance liquid chromatography. Subsequently, two major citrus flavonoids were enzymatically conjugated to citrus pectin at high grafting rates (hesperidin: 20.21%; naringin dihydrochalcone: 16.11%). The flavonoids increased pectin amphipathy while ensuring a tighter conformation and denser network structure. These structural changes reduced the interfacial tension (from 20.6 to 18.4 mN/m) and increased the adsorbed layer thickness (from 11.8 to 37.8 nm), facilitating rapid pectin anchoring at the oil-water interface. These led to smaller droplet size and better emulsion stability. Prepared flavonoid-pectin conjugates with high grafting rates exhibited better emulsifying properties than those with medium grafting rates; hesperidin-pectin conjugates were more effective than naringin dihydrochalcone-pectin conjugates. The prepared flavonoid-pectin conjugates aid in the structural identification of natural flavonoid-pectin conjugates; they also facilitate analyses of the properties of these conjugates and offer new insights into improving the functional properties of pectins used in the food industry.

Alkylated pectin: Synthesis, characterization, viscosity and emulsifying properties

Rui-hong Liang,Ling-hua Wang,Jun Chen,Wei Liu,Cheng-mei Liu

Recently there has been an increased interest in modification of pectins. Pectins were alkylated with alkyl chain of various lengths (hexyl, dodecyl or octadecyl) at different degrees of substitution (0.69, 2.43 or 3.60) by reaction of TBA-pectin with alkyl bromide in this study. Their structures were characterized by GC, FTIR and 1 H NMR, which confirmed the formation of ester linkages between hydrophobic alkyl segment and carboxyl group of galacturonic acid. Alkylated pectins produced interesting rheology properties. They showed higher apparent viscosities but lower intrinsic viscosities than the original one. The higher apparent viscosities may be due to intermolecular association of hydrophobic segments in the molecule. The lower intrinsic viscosities would be ascribed to more compact conformation. In addition, the modified pectins showed an enhancement of emulsifying properties. These properties were significantly influenced by the degree of substitution and alkyl chain length. Such amphiphilic derivatives open new perspectives for improving or altering viscosity and emulsifying properties of pectin.

Gelatin-crosslinked pectin nanofiber mats allowing cell infiltration

Xiaoqi Shi,Sisi Cui,Xiaoyu Song,Alex P. Rickel,Hanna J. Sanyour,Jia Zheng,Junli Hu,Zhongkui Hong,Yifa Zhou,Yichun Liu

Pectin nanofiber mats are promising tissue engineering scaffolds but suffer from poor cell infiltration. In this study, gelatin, a collagen derived cell adhesive protein, was used to crosslink the electrospun nanofibers of periodate oxidized pectin. Cell culture experiment results demonstrated that cells were able to grow into the gelatin-crosslinked pectin nanofiber mats rather than only spread on mat surface. The nanofiber mats showed moderate mechanical strength, with a maximum tensile strength of up to 2.3 MPa, an ultimate tensile strain of up to 15%, and were capable of degrading gradually over 4 weeks or even longer periods in simulated body fluids. Thus, gelatin-crosslinked pectin nanofiber mats hold a great potential for soft tissue regeneration.

Construction of a Rice Glutelin-Rhamnolipid-Pectin Nanoparticle Delivery System for Curcumin Delivery: Effect of Pectin Esterification on Characterization, Stability, and Bioaccessibility

Lei Chen,Qiongqiong Wu,Shaokang Li,Shuangli Wang,Weidong Lin,Xiaodan Zang

Riceglutelin (RG)-rhamnolipids (Rha)-high-methoxyl pectin (HMP)/medium-methoxy pectin (MMP)/low-methoxy pectin (LMP) were used to attract self-assembled nanoparticles by electrostatic attraction, and the effects of pectin esterification degree and concentration on the characteristics of nanoparticles and the bioavailability of curcumin were evaluated. The minimum particle size and the highest encapsulation efficiency of the nanoparticles were at mass ratios of RG:HMP = 2:1, RG:MMP = 4:1, and RG:LMP = 2:1. The results of Fourier transform infrared spectroscopy and circular dichroism spectroscopy show that hydrogen bonding, hydrophobic interaction, and electrostatic attraction are the main driving forces for the formation of complexes. The X-ray diffraction results verified that the curcumin in the complex was in an amorphous state. In addition, curcumin embedded with nanoparticles has good photothermal and storage stability. The results of in vitro simulated digestion showed that nanoparticles constructed from LMP had high bioaccessibility. The results of this study are expected to provide new ideas for improving the bioaccessibility of curcumin.

Self-sustaining microbattery fueled by pectin via enzymatic catalysis

Yuting Zhang,Chaohong Liang,Yuan Guan,BoTian Liu

A pectin-fueled enzymatic microbattery with methylene blue redox mediator. • Electrochemically rechargeable by reducing oxidized MB to its leuco form. • Maximum power density of 1.57 mWcm −2 , exceeding most reported EBFCs. • Retains 96.03% capacity after 100 cycles at 2.25 mA cm -2. • Decouples energy delivery from continuous fuel supply via electrochemical recharging. Bio-derived self-sustaining microbatteries are promising power sources for implantable and wearable electronics due to their excellent biocompatibility and biodegradability. However, their practical deployment is hindered by limited operational lifetime and low energy density, primarily stemming from the rapid depletion of conventional monosaccharide fuels such as glucose. Herein, we report an enzymatic microbattery fueled by pectin, a naturally abundant polysaccharide, with methylene blue (MB) immobilized on a pectinase-modified anode as a redox mediator. Unlike conventional enzymatic biofuel cells (EBFCs) that rely solely on continuous fuel supply, our hybrid EBFCs can be electrochemically “recharged” by reducing oxidized MB to its leuco form under an applied potential, enabling repeated discharge cycles without immediate fuel replenishment. As a results, the hybrid EBFCs deliver a high maximum power density of 1.57 mWcm −2 , substantially exceeding that of recently reported EBFCs. Moreover, it retains 96.03% of its initial capacity after 100 charge-discharge cycles at a current density of 2.25 mA cm −2 , demonstrating exceptional cycling stability. This pseudo-rechargeable behavior bridges the gap between enzymatic energy conversion and secondary battery functionality.

Lubricating performance of pectin: Influence from the colloidal structures

Wenxin Jiang,Fengting Li,Tianci Qiu,Wei Lu,Zhiming Gao,Yuehan Wu,Dan Yuan,Yanlei Li

This study demonstrates that the lubrication performance of pectin is markedly modulated by its colloidal structure. Motivated by the pH-altered lubricating performance of acidified pectin solutions, this work further investigates the lubricating properties of Ca 2+ -crosslinked pectin microgels (PMGs). The impact of Ca 2+ crosslinking on the physical and colloidal characteristics of PMGs suspensions is evaluated by visual appearance, microscopic morphology, and large amplitude oscillatory shear (LAOS) rheology analysis. Results show that only a low concentration of Ca 2+ (20–60 mmol/L) promotes the formation of a robust colloidal structure. The strain stiffening index (S) shows that Ca 2+ crosslinking endows pectin with greater resistance to applied strains, and notable strain stiffening behaviors are observed in PMGs suspensions. Friction curves suggest that acidified pectin solutions have lower maximum friction coefficients ( μ max ) and PMGs suspensions have narrower boundary regimes. Furthermore, the lubricating performance of PMGs suspensions can be regulated by the homogenization rate and NaCl concentration, where shorter boundary regimes are found in robust PMGs suspensions. For pectin-PMGs mixtures, friction curves further confirm that a low PMGs proportion (10 %) is sufficient to promote the rapid lubrication regime transition. By clarifying the linkage between colloidal structure and lubricating performance of pectin, this study provides new perspectives for regulating oral lubricating behavior in food products.

Citrus pectin derived silver nanoparticles and their antibacterial activity

Wenlin Zhang,Xi Juan Zhao,Yanyan Jiang,Zhiqin Zhou

Herein, a green, simple, and cost-effective synthesis method for silver nanoparticles (Ag NPs) has been established by using citrus pectin as reducing and capping agent. Importantly, no toxic chemicals were added into the synthesis process. The Ag NPs were characterized by UV-vis, TEM, XRD, and FTIR. The results indicated the Ag NPs were spherical shape with small, uniform size of 3 nm and excellent dispersity. Additionally, the Ag NPs have good antibacterial activity against Escherichia coli and Staphylococcus aureus. Using citrus pectin for preparation of Ag NPs was potentially a new path for the efficient utilization of citrus resources.

Blue-light emitting aminated pectin for detecting Cu2+ ion

Chengsheng Ni,Wenxuan Lu,Jing Zhang,Luo Peng,Deti Xie,Jiupai Ni

This research studied the chemo-sensing of low-cost aminated pectin (PE) obtained by a facile calcination under ammonia gas at temperature no higher than 175 °C without excessive use of alkaline, acid or solvents. The ammonia gas was found to replace the hydroxyl and methoxyl group, enhancing the crystallinity and solubility of the resultant pectin than those calcined in air or in 5% H 2 . Though the increase of light absorption could be attributed mainly to the dehydration during calcination which caused the formation of C C double bond or aromatic ring, the N incorporation could be important to the photoluminescence (PL) emission. The PL quenching of the blue fluorescent aminated pectin showed a good linearity with the concentration of Cu 2+ , Fe 3+ and the highest sensitivity toward Cu 2+ among the investigated metal ions. In order to further increase the PL quenching toward Cu 2+ and decrease the interference of Fe 3+ , a method involving H 2 O 2 and ultraviolet illumination was developed to catalyze the oxidation of fluorophores on the polymer. This work provides new horizon on the modification and application of pectin in chemosensing.
