Natural antibacterial agents exhibit tremendous potential in fabricating the durable antibacterial fabrics. Herein, natural pterostilbene (PL) was grafted onto chitosan (CS) through a Mannich reaction to obtain PLCS with high antibacterial activity. Furthermore, the durable antibacterial cotton fabric was fabricated using a layer-by-layer assembled coating with PLCS as the antibacterial agent. The results showed that at a molar ratio of PL to CS of 0.2:1 and a temperature of 80 ℃, PLCS with a grafting ratio of 17.3% achieved the lowest minimum inhibitory concentrations of 33.2 and 16.6 µg/mL against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ), respectively. Compared to those of CS and PL alone, the antibacterial activity of PLCS against S. aureus improved by 67.0% and 58.5%, while the activity against E. coli improved by 94.8% and 44.7%, respectively. The antibacterial rate of the PLCS cotton fabric against S. aureus and E. coli reached up to 99.9% after three assembled layers and maintained more than 80% after 20 washings. The PLCS antibacterial fabrics exhibited good wearing safety and wearing performances.

Bacterial infections severely threaten human health; therefore, it is important to endow the matrix for tissue engineering with antibacterial efficiency. The loading of antibacterial drugs on nanomaterials provides an efficient strategy to realize synergistic antibacterial efficiency. By depositing various metal–organic frameworks, such as UIO-66, onto konjac glucomannan (KGM), composite hydrogels (KGM/UIO-66) were created. These hydrogels were used as drug carriers, enabling the development of antibacterial hydrogels with high drug loading capacities (e.g., the maximum loading amount of pterostilbene on KGM/UIO-66 reached 0.157 mg/mg) and sustained drug release. The resulting KGM/UIO-66/pterostilbene hydrogel exhibited a three-dimensional porous structure, excellent biocompatibility, antibacterial efficiency, and anti-inflammatory activity. It effectively protected cells from bacterial attacks while ensuring cell adhesion and proliferation, demonstrating great potential as a three-dimensional substrate for biomedical applications, including tissue engineering and regenerative medicine.

Caffeic acid was grafted onto starch (Cf-HAMS) via C-6 esterification. • Caffeic acid grafting simultaneously enhanced amphiphilicity and bioactivity. • Cf-HAMS solely stabilized emulsion gels via reinforced interfacial films and networks. • Emulsion gels enhanced pterostilbene stability and in vitro bioaccessibility. Caffeic acid-grafted high-amylose maize starch (Cf-HAMS) with grafting degrees of 4%, 8%, and 12% was synthesized via esterification to address the limitations of native HAMS in emulsion-gel stabilization. Nuclear magnetic resonance (NMR), Fourier transform infrared spectroscopy (FTIR), and X-ray photoelectron spectroscopy (XPS) confirmed covalent grafting at the C-6 hydroxyl groups. Due to the introduction of caffeic acid's phenolic hydroxyls and aromatic rings, Cf-HAMS exhibited markedly enhanced hydrophobicity (contact angle from 26.8° to 65.1°) together with strong antioxidant and antibacterial activities. Consequently, Cf-HAMS alone, without any added surfactants, stabilized emulsion gels with smaller droplets, reinforced interfacial films, and enhanced viscoelasticity. When applied as carriers for pterostilbene, Cf-HAMS achieved approximately 90% encapsulation efficiency, improved UV and thermal protection, suppressed lipid oxidation, and increased bioaccessibility by a factor of 2.5 compared with native HAMS. Overall, these findings demonstrate that phenolic-acid grafting effectively integrates interfacial stabilization with chemical protection for hydrophobic bioactives. Ulcerative colitis (UC) is a chronic, non-specific inflammatory disorder of the intestines whose etiology is influenced by various factors. Intestinal barrier impairment due to disturbances in the intestinal microenvironment is a key feature of UC. Current therapeutic strategies are constrained in their capacity to fully restore the intestinal barrier and achieve comprehensive resolution of inflammation in a coordinated manner. In this study, we constructed a pterostilbene (PSB)-loaded prebiotic microcapsule (PSB@MC) using a microfluidic electrospray method and characterized it using various means. Its safety, biodistribution, protective, and therapeutic effects on colitis were evaluated in various animal models. The potential mechanisms by which PSB@MC exerts its therapeutic effects were subsequently explored. The results indicated that PSB@MC exhibited favorable biocompatibility and facilitated targeted delivery of PSB to the colon. Moreover, the wrinkled morphology of PSB@MC contributed to prolonged drug retention in the colon. Oral PSB@MC administration restored intestinal microenvironment homeostasis by scavenging reactive oxygen species (ROS), decreasing pro-inflammatory cytokines, modulating gut microbiota and metabolism, and providing protective and therapeutic benefits against dextran sulfate sodium-induced colitis. Additionally, our research demonstrated that PSB@MC could activate the aryl hydrocarbon receptor/interleukin-22 (AHR/IL-22) pathway to enhance the integrity of the intestinal barrier. These results suggest that PSB@MC could be a new, secure, and efficient UC therapy option.

Pterostilbene is a demethylated resveratrol derivative with attractive anti-inflammatory, anti-tumor and anti-oxidative stress activities. However, the clinical use of pterostilbene is limited by its poor selectivity and druggability. Heart failure is a leading cause of morbidity and mortality worldwide, which is closely related to enhanced oxidative stress and inflammation. There is an urgent need for new effective therapeutic drugs that can reduce oxidative stress and inflammatory responses. Therefore, we designed and synthesized a series of novel pterostilbene chalcone and dihydropyrazole derivatives with antioxidant and anti-inflammatory activities by the molecular hybridization strategy. The preliminary anti-inflammatory activities and structure-activity relationships of these compounds were evaluated by nitric oxide (NO) inhibitory activity in lipopolysaccharide (LPS)-treated RAW264.7 cells, and compound E1 exhibited the most potent anti-inflammatory activities. Furthermore, pretreatment with compound E1 decreased reactive oxygen species (ROS) generation both in RAW264.7 and H9C2 cells by increasing the expression of nuclear factor erythroid 2-related factor 2 (Nrf2), as well as downstream antioxidant enzymes superoxide dismutase 1 (SOD1), catalase (CAT) and glutathione peroxidase 1 (GPX1). In addition, compound E1 also significantly inhibited LPS or doxorubicin (DOX)-induced inflammation in both RAW264.7 and H9C2 cells through reducing the expression of inflammatory cytokines by inhibiting nuclear factor-κB (NF-κB) signaling pathway . Moreover, we found that compound E1 improved DOX-induced heart failure by inhibiting inflammation and oxidative stress in mouse model, which is mediated by the potential of antioxidant and anti-inflammatory activities. In conclusion, this study demonstrated the novel pterostilbene dihydropyrazole derivative E1 was identified as a promising agent for heart failure treatment.

Pterostilbene (PTB) is a hydrophobic compound derived from plants in the Astragalus family. PTB possesses remarkable skin brightening and antioxidant properties. However, delivery methods for PTB require enhancement, and this can be achieved by formulating suitable carriers. In this study, we prepared ionic liquid microcapsules containing PTB by using a microencapsulation technique based on a matrine and cocinic acid ([Mat][Coc]) ionic liquid. The particle size of the microcapsules was 154.5 ± 3.42 nm, and the encapsulation efficiency was as high as 99.7 %. Further, the PTB-loaded ionic liquid microcapsules had good biocompatibility, enhanced skin retention (7.7-times higher than that of free PTB), and improved skin penetration without damaging the skin structure. Clinical testing with human subjects revealed that the microcapsule emulsion reduced fine lines and brightened skin tone. Therefore, PTB-loaded ionic liquid microcapsules have promise for use in cosmetic formulations and applications in transdermal drug delivery and efficacy enhancement.

Pterostilbene (PTE) is a natural stilbene compound abundantly found in various small berries such as blueberry and grape, and exhibits multiple health promoting potentials and excellent pharmacological activities, especially antioxidant and anti-inflammatory activities. This study explored the protective effects of PTE on pulmonary fibrogenesis and inflammation and its underlying mechanisms. TGF-β-stimulated BEAS-2B or primary lung fibroblasts (LFs) were cultured with PTE or pirfenidone (PFD). Mice were received intratracheal instillation of bleomycin (BLM) and administered PTE or PFD. IL-23/ATP6V 0 D 2 -deficient BEAS-2B were treated with TGF-β or PTE. Primary LFs were stimulated with conditioned medium from TGF-β-primed RLE-6TN (CM) and treated with PTE. In vitro , PTE inhibited TGF-β-induced BEAS-2B cell migration, extracellular matrix (ECM) deposition, epithelial-mesenchymal transition (EMT) and inflammation. In BLM-induced mice, PTE reduced lung index and IL-1β levels in BALF, and improved histopathological changes. PTE inhibited BLM-induced fibrogenesis, EMT, inflammation and immune cell infiltration through blocking IL-23 and activating ATP6V 0 D 2 . In TGF-β-induced BEAS-2B, PTE decreased IL-23, increased ATP6V 0 D 2 and ATP levels, and disrupted the interaction between IL-23 and ATP6V 0 D 2 . In IL-23-deficient BEAS-2B, PTE further elevated ATP6V 0 D 2 and decreased α-SMA, Vimentin and IL-1β expressions. ATP6V 0 D 2 deficiency attenuated the anti-fibrotic and anti-inflammatory effects of PTE in TGF-β-induced BEAS-2B. PTE also inhibited fibrogenesis, EMT, and inflammation in activated-primary LFs and blocked CM-triggered primary LFs activation by regulating IL-23-ATP6V 0 D 2 axis. In conclusion, PTE ameliorated pulmonary fibrosis targeting IL-23/ATP6V 0 D 2 pathway, especially remodeling the local microenvironment of alveolar epithelial cells and fibroblasts, which might be a novel therapeutic strategy against pulmonary fibrosis.

The emergence of polymyxin-resistant Gram-negative pathogens has severely compromised colistin (COL) efficacy in veterinary therapy, posing critical challenges to animal health and food safety. Here, we show that pterostilbene (PTE), a dietary phenolic compound abundant in blueberries and grapes, effectively restores and enhances the antibacterial activity of COL. PTE acts synergistically with COL against multidrug-resistant strains, eliminates metabolically dormant persister cells and suppresses biofilm formation. Mechanistically, PTE chelates ferrous iron to deplete intracellular iron pools, while unexpectedly inhibiting iron-driven lipid peroxidation and preserving membrane potential. This disruption of iron homeostasis stimulates ATP generation but, when combined with COL, triggers a futile metabolic cycle that leads to severe ATP leakage. Additionally, PTE compromises outer membrane integrity and increases inner membrane phospholipid susceptibility, augmenting COL-induced lethal damage. In a Galleria mellonella infection model, the PTE-COL combination significantly improves survival. Moreover, in weaned piglets, PTE supplementation promotes restoration of a balanced gut microbiota and reduces pathogen abundance. These results highlight PTE as a safe, food-compatible adjuvant that resensitizes resistant bacteria to COL via a multi-target mechanism, offering a sustainable strategy for controlling resistant infections at the food-animal interface.

Pterostilbene, a dietary polyphenol in blueberries, grapes and wine, possesses an anti-diabetic efficacy. However, the mechanism of anti-diabetic efficacy for pterostilbene is poorly understood. The purpose of the present study was to systematically investigate inhibitory effects and inhibition mechanisms of pterostilbene or its metabolite (pinostilbene) against aldose reductase and α-glucosidase. In the present study, inhibition kinetics and molecular docking were applied to investigate the effect of pterostilbene and pinostilbene on the activity of aldose reductase and α-glucosidase. The results showed that pinostilbene exhibited stronger inhibition on aldose reductase (IC50 = 55.41 ± 4.31 μM) and α-glucosidase (IC50 = 11.69 ± 0.888 μM) in noncompetitive-type manner. Whereas, pterostilbene displayed the weak effect on aldose reductase and α-glucosidase (IC50 = 1569 ± 177.858 μM) inhibition. Moreover, pinostilbene interacted with amino acid residues outside of aldose reductase and α-glucosidase active site to inhibit its activities by van der Waals, π–anion or π–alkyl interactions. The current study has provided comprehensive reference not only for the anti-diabetic mechanism of pterostilbene, but also offered a basis to develop and design adjuvant antidiabetic drugs or novel functional foods.

Sepsis remains one of the most common life-threatening illnesses that is characterized by a systemic inflammatory response syndrome (SIRS) and usually arises following severe trauma and various septic infections. It is still in urgent need of new effective therapeutic agents, and chances are great that some candidates can be identified that can attenuate oxidative stress and inflammatory responses. Pterostilbene, which exerts attractive anti-oxidative and anti-inflammatory activities, is a homologue of natural polyphenolic derivative of resveratrol. Starting from it, we have made several rounds of rational optimizations. Firstly, based on the strategy of pharmacophore combination, indanone moiety was introduced onto the pterostilbene skeleton to generate a novel series of pterostilbene derivatives (PIF_1–PIF_16) which could possess both anti-oxidative and anti-inflammatory activities for sepsis treatment. Then, all target compounds were subjected to their structure–activity relationships (SAR) screening of anti-inflammatory activity in mouse mononuclear macrophage RAW264.7 cell line, and their cytotoxicities were determined after. Finally, an optimal compound,PIF_9, was identified. It decreased the mRNA levels of lipopolysaccharide (LPS)-induced interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), inducible nitric oxide synthase (iNOS), and cyclooxygenase 2 (COX2). We also found that the anti-inflammatory effects might be contributed by its suppression on the nuclear factor-κB (NF-κB) and MAPKs signaling pathway. Moreover,PIF_9also demonstrated potent anti-oxidative activity in RAW264.7 macrophages and the sepsis mouse model. Not surprisingly, with the benefits mentioned above, it ameliorated LPS-induced sepsis in C57BL/6J mice and reduced multi-organ toxicity. Taken together,PIF_9was identified as a potential sepsis solution, targeting inflammation and oxidative stress through modulating MAPKs/NF-κB.Keywords:antioxidants;anti-inflammatory agents;indanone;pterostilbene;sepsis

Ecofriendly fabrics with antibacterial and anti-adhesion properties have been attracted an increasing attention in recent years. Herein, natural menthol modified polyacrylate (PMCA) antibacterial adhesion agent was synthesized by esterification and polymerisation while natural pterostilbene-grafted-chitosan (PGC) antibacterial agent was prepared through Mannich reaction. The antibacterial and anti-adhesion cotton fabric was fabricated through durable PMCA dip finishing and then layer-by-layer self-assembly of PGC. The results showed that the antibacterial adhesion rates and antibacterial rates of the dual-function cotton fabric against Staphylococcus aureus and Escherichia coli reached up to 99.9 %. Its antibacterial adhesion rates improved by 36.1 % and 40.1 % in comparison with those of cotton fabric treated by menthol alone. Meanwhile against S. aureus , the dual-function cotton fabrics improved the antibacterial rates by 56.7 % and 36.4 %, respectively, from those of chitosan- and pterostilbene-treated fabrics. Against E. coli , the improvements were 89.4 % and 24.8 %, respectively. After 20 household washings, the dual-function cotton fabric maintained >80 % of its original anti-adhesion and antibacterial rates against both species. The dual-function cotton fabric also possessed safe and excellent wearability.

Abstract Exosomes derived from probiotics are emerging as innovative oral nanocarriers but face challenges in targeted delivery and stability. Here, Lactobacillus helveticus-derived exosomes (LHexo) were dual-functionalized with mannose and cRGD (McRLHexo), yielding improved membrane lipid ordering, denser PEG brush layer, and distinct “cloud-like” surface nanostructure. McRLHexo exhibited enhanced stability under UV (transmittance reduction > 98%), thermal, and digestive conditions (drug retention >86%), and increased pterostilbene (PTS) loading. Functionally, McRLHexo@PTS achieved efficient epithelial translocation via cRGD-integrin pathways and macrophage targeting via Man/cRGD dual-recognition. In DSS-induced colitis mice, McRLHexo showed optimal targeting accumulation and uptake in the colon. Importantly, McRLHexo@PTS markedly alleviated colitis, restoring body weight, colon length, and epithelial integrity while reducing inflammatory cytokines and oxidative stress and enhancing antioxidant defenses. This study establishes a dual-targeted probiotic exosome platform for oral therapy and provides insights into ligand-membrane engineering for precision intestinal delivery.
