Enhanced Cd extraction of oilseed rape (Brassica napus) by plant growth-promoting bacteria isolated from Cd hyperaccumulator Sedum alfredii Hance

Fengshan Pan,Qian Meng,Sha Luo,Jing Shen,Bao Chen,Kiran Yasmin Khan,Jan Japenga,Xiaoxiao Ma,Xiaoe Yang,Ying Feng

Four plant growth-promoting bacteria (PGPB) were used as study materials, among them two heavy metal-tolerant rhizosphere strains SrN1 (Arthrobacter sp.) and SrN9 (Bacillus altitudinis) were isolated from rhizosphere soil, while two endophytic strains SaN1 (Bacillus megaterium) and SaMR12 (Sphingomonas) were identified from roots of the cadmium (Cd)/zinc (Zn) hyperaccumulator Sedum alfredii Hance. A pot experiment was carried out to investigate the effects of these PGPB on plant growth and Cd accumulation of oilseed rape (Brassica napus) plants grown on aged Cd-spiked soil. The results showed that the four PGPB significantly boosted oilseed rape shoot biomass production, improved soil and plant analyzer development (SPAD) value, enhanced Cd uptake of plant and Cd translocation to the leaves. By fluorescent in situ hybridization (FISH) and green fluorescent protein (GFP), we demonstrated the studied S. alfredii endophytic bacterium SaMR12 were able to colonize successfully in the B. napus roots. However, all four PGPB could increase seed Cd accumulation. Due to its potential to enhance Cd uptake by the plant and to restrict Cd accumulation in the seeds, SaMR12 was selected as the most promising microbial partner of B. napus when setting up a plant–microbe fortified remediation system.

One-Step Extraction and Hydrolysis of Flavonoid Glycosides in Rape Bee Pollen Based on Soxhlet-Assisted Matrix Solid Phase Dispersion

Xijuan Tu,Shuangqin Ma,Zhaosheng Gao,Jing Wang,Shaokang Huang,Wenbin Chen

Introduction Flavonoids are frequently found as glycosylated derivatives in plant materials. To determine contents of flavonoid aglycones in these matrices, procedures for the extraction and hydrolysis of flavonoid glycosides are required. The current sample preparation method is both labour and time consuming. Objective Develop a modified matrix solid phase dispersion (MSPD) procedure as an alternative methodology for the one-step extraction and hydrolysis of flavonoid glycosides. Method HPLC-DAD was applied for demonstrating the one-step extraction and hydrolysis of flavonoids in rape bee pollen. The obtained contents of flavonoid aglycones (quercetin, kaempferol, isorhamnetin) were used for the optimisation and validation of the method. Results The extraction and hydrolysis were accomplished in one step. The procedure completes in 2 h with silica gel as dispersant, a 1:2 ratio of sample to dispersant, and 60% aqueous ethanol with 0.3 M hydrochloric acid as the extraction solution. The relative standard deviations (RSDs) of repeatability were less than 5%, and the recoveries at two fortified levels were between 88.3 and 104.8%. Conclusion The proposed methodology is simple and highly efficient, with good repeatability and recovery. Compared with currently available methods, the present work has advantages of using less time and labour, higher extraction efficiency, and less consumption of the acid catalyst. This method may have applications for the one-step extraction and hydrolysis of bioactive compounds from plant materials. Copyright © 2017 John Wiley & Sons, Ltd.

Removal Performance and Mechanism of Emerging Pollutant Chloroquine Phosphate from Water by Iron and Magnesium Co-Modified Rape Straw Biochar

Hongwei Sun,Jinjin He,Yucan Liu,Xianguo Ji,Gang Wang,Xiaoyong Yang,Yanxiang Zhang

Chloroquine phosphate (CQP) is effective in treating coronavirus disease 2019 (COVID-19); thus, its usage is rapidly increasing, which may pose a potential hazard to the environment and living organisms. However, there are limited findings on the removal of CQP in water. Herein, iron and magnesium co-modified rape straw biochar (Fe/Mg-RSB) was prepared to remove CQP from the aqueous solution. The results showed that Fe and Mg co-modification enhanced the adsorption efficiency of rape straw biochar (RSB) for CQP with the maximum adsorption capacity of 42.93 mg/g (at 308 K), which was about two times higher than that of RSB. The adsorption kinetics and isotherms analysis, as well as the physicochemical characterization analysis, demonstrated that the adsorption of CQP onto Fe/Mg-RSB was caused by the synergistic effect of pore filling, π-π interaction, hydrogen bonding, surface complexation, and electrostatic interaction. In addition, although solution pH and ionic strength affected the adsorption performance of CQP, Fe/Mg-RSB still had a high adsorption capability for CQP. Column adsorption experiments revealed that the Yoon–Nelson model better described the dynamic adsorption behavior of Fe/Mg-RSB. Furthermore, Fe/Mg-RSB had the potential for repeated use. Therefore, Fe and Mg co-modified biochar could be used for the remediation of CQP from contaminated water.

Light-triggered catalase like nanozyme mitigates frost damage of winter oilseed rape via carbon dot-mediated membrane homeostasis and metabolic reprogramming

Yuan Li,Qingbo Xiao,Zixuan Ding,Emad M. Hafez,Wenlong Xu,Zhiyang Zhang

A light-triggered nanozyme (YTzymes) with catalase-like activity and yellow-light emission mitigates frost stress in winter oilseed rape. • YTzymes synergistically scavenge ROS and use emitted light to reprogram linoleic acid and TCA metabolism, maintaining membrane and redox homeostasis. • Foliar application (600 mg/L) enhances frost recovery, increasing shoot/root dry weight by 21.56%/11.15% and improving Ca²⁺/K⁺ homeostasis. Winter oilseed rape is a vital industrial crop, primarily cultivated for the production of vegetable oil, biodiesel, and industrial lubricants. Mitigation of frost damage in winter oilseed rape is critical for ensuring stable yields and quality. To address this challenge, we engineered a multifunctional light-triggered catalase like nanozyme (denoted as YTzymes) integrating catalase-like activity with yellow-light emission, specifically designed to counteract frost-induced adversity in winter oilseed rape. The dual-action mechanism of YTzymes underpins its efficacy:1) Intrinsic catalase-like activity directly scavenges excess reactive oxygen species (ROS) that accumulate rapidly in plant cells under frost stress, thereby alleviating oxidative damage to crop cells; 2) Yellow light emission from the nanozyme triggers targeted metabolic reprogramming, modulating linoleic acid metabolism (a key component of membrane lipids) to maintain integrity of cell membranes, while simultaneously enhancing the tricarboxylic acid (TCA) cycle flux to boost energy supply and redox homeostasis. Foliar application of 600 mg/L YTzymes solution significantly promoted frost recovery in winter oilseed rape. Compared to untreated frost-stressed plants, the dry weight of shoots and roots in the nanozyme-treated group increased by 21.56% and 11.15%, respectively, accompanied by improved cell membrane integrity. Meanwhile, the reduction of Ca 2 + concentration and increment of K + concentration could further reduce the osmotic pressure of the cell, reducing the cell dehydration in freezing conditions. This light-triggered nanozyme approach offers an efficient solution to mitigate frost damage, thereby contributing to the development of sustainable crop production under changing climate conditions. Download: Download high-res image (136KB) Download: Download full-size image YTzymes, integrating catalase-like activity and yellow-light emission, enhances frost resilience in winter oilseed rape by scavenging ROS to reduce oxidative stress and reprogramming metabolism to reinforce membrane integrity (through linoleic acid regulation) and energy supply (via TCA cycle enhancement). Foliar application of 600 mg/L YTzymes elevated shoot and root dry weights by 21.56% and 11.15%, improved ion homeostasis (Ca²⁺ reduction, K⁺ elevation), and alleviated cellular dehydration.

Molecularly imprinted polymers based on calcined rape pollen and deep eutectic solvents for efficient sinapic acid extraction from rapeseed meal extract

Yanhua Sun,Yange Zhang,Yujiao Hou,Hui Gong,Yifei Pang,Xiaoxiao Ge,Ming Li

Substances that possess hierarchical and interconnected porous features are ideal choices for acting as skeletons to synthesize surface molecularly imprinted polymers (MIPs). In this work, rape pollen, a waste of biological resources, was calcined and a porous mesh material with a high specific surface area was obtained. The cellular material was adopted as a supporting skeleton to synthesize high-performance MIPs (CRPD-MIPs). The CRPD-MIPs presented an ultrathin imprinted layered structure, with an enhanced adsorption capacity for sinapic acid (154 mg g −1 ) relative to the non-imprinted polymers. The CRPD-MIPs also exhibited good selectivity (IF = 3.24) and a fast kinetic adsorption equilibrium (60 min). This method exhibited a good linear relationship (R2 = 0.9918) from 0.9440 to 29.26 μg mL −1 , and the relative recoveries were 87.1–92.3%. The proposed CRPD-MIPs based on hierarchical and interconnected porous calcined rape pollen may be a valid program for the selective extraction of a particular ingredient from complicated actual samples.

Identification and mechanism of effective components from rape (Brassica napus L.) bee pollen on serum uric acid level and xanthine oxidase activity

Rui-dan Wang,Guan-hua Su,Lin Wang,Qing Xia,Rui Liu,Qun Lu,Jiu-liang Zhang

This study was aimed at evaluating the hypouricemic effect of rape bee pollen from Qinghai province in China. Rape bee pollen crude extracts (RPCE) was obtained by n-butanol extraction and purified by polyamide resin and AB-8 resin respectively to generate five fractions (fraction 1–5). In vitro , fraction 5 had strongest inhibition on xanthine oxidase (XO) (IC 50  = 0.21 ± 0.02 mg mL −1 ) among five fractions and had reversible and mixed inhibitory effect on XO. Fraction 5 was further purified and then generated fraction 5′. In vivo , fraction 5′ had lower serum UA level and could reduce serum BUN, Cr levels and hepatic XO activity more effectively than RPCE in potassium oxonate-induced hyperuricemic mice . Moreover, fraction 5′ had higher CAT activity and content of GSH in hyperuricemic mice. Six compounds including quercetin-3-O-(2″-O-glucopyranosyl)-glucopyranoside (peak 1), quercetin-3-O-(2″-O-glucopyranosyl)-rhamnopyranoside (peak 2), kaempferol-3-O-(2″-O-glucopyranosyl)-glucopyranoside (peak 3) and N′, N″-di- p -coumaroylspermidine (peak 4–6) in RPCE were identified by using HPLC-ESI-QTOF-MS/MS, whereas only peak 4 to 6 were found in fraction 5′. This study shows that rape bee pollen has potential clinical application value as an effective anti-hyperuricemia bioactive drug or functional food.

Synthetic HZSM-5/MCM-41 catalyst by Direct Non-thermal Plasma for promoted production of aromatic hydrocarbons by catalytic pyrolysis of rape straw

Xiaohua Li,Jinlong Yan,Shanshan Shao,Shengnan Ge,Zhichao Lv,Yixi Cai

A method of template removal by direct non-thermal plasma (DNTP) system for the synthesis of HZSM-5/MCM-41 meso-micropore composite catalyst was proposed for the production of aromatic hydrocarbons by catalytic pyrolysis of rape straw. It was revealed most of the template on the catalyst was removed by oxidizing substances such as ozone produced by DNTP. During DNTP treatment, the removal effect of the template was greatly affected by the oxygen flow rate . Also, the catalyst structure was bombarded by high-energy electrons obtained by DNTP treatment, and the physicochemical property of the synthetic composite catalysts was treated quite different from that obtained by calcination method. Under the treatment condition of 22 kV-2 L/min, the BET surface area was boosted significantly by 29.91% and the amount of acid sites was boosted by 37.71%, the yield of MAHs reached 10.57 wt%. Generally, not only the template can be effectively removed by DNTP treatment, but also the physicochemical property of meso-micropore composite catalyst can be improved, and thus promoted the catalytic performance for monocyclic aromatic hydrocarbons. Therefore, DNTP is an effective technology for removing template and the catalytic performance can be efficiently improved.

Separation and Characterization of Phenolamines and Flavonoids from Rape Bee Pollen, and Comparison of Their Antioxidant Activities and Protective Effects Against Oxidative Stress

Huifang Zhang,Rui Liu,Qun Lu

Phenolamines and flavonoids are two important components in bee pollen. There are many reports on the bioactivity of flavonoids in bee pollen, but few on phenolamines. This study aims to separate and characterize the flavonoids and phenolamines from rape bee pollen, and compare their antioxidant activities and protective effects against oxidative stress. The rape bee pollen was separated to obtain 35% and 50% fractions, which were characterized by HPLC-ESI-QTOF-MS/MS. The results showed that the compounds in 35% fraction were quercetin and kaempferol glycosides, while the compounds in 50% fraction were phenolamines, including di-p-coumaroyl spermidine, p-coumaroyl caffeoyl hydroxyferuloyl spermine, di-p-coumaroyl hydroxyferuloyl spermine, and tri-p-coumaroyl spermidine. The antioxidant activities of phenolamines and flavonoids were evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS), and ferric reducing antioxidant power (FRAP) assays. It was found that the antioxidant activity of phenolamines was significantly higher than that of flavonoids. Moreover, phenolamines showed better protective effects than flavonoids on HepG2 cells injured by AAPH. Furthermore, phenolamines could significantly reduce the reactive oxygen species (ROS), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels, and increase the superoxide dismutase (SOD) and glutathione (GSH) levels. This study lays a foundation for the further understanding of phenolamines in rape bee pollen. Keywords:pollen;antioxidant;oxidative stress;hydroxycinnamic acid amides

Catalytic pyrolysis of rape straw to produce light aromatic hydrocarbons over HZSM-5 loaded by metal ions from spent lithium batteries extracted with citric acid

Shanshan Shao,Yu Cao,Jiaxin Jiang,Xiaohua Li,Yongbo Yu

In this study, metal-loaded HZSM-5 was prepared with metal ion extracted from the cathode materials of spent lithium batteries (lithium iron phosphate, LFP; lithium nickel-cobalt manganate, NCM) with citric acid for the improved production of light aromatic hydrocarbons by catalytic pyrolysis of rape straw. Because of the dealuminization effect of citric acid on HZSM-5, mesopores were introduced, and the diffusion and conversion of pyrolysis gas over acid sites were improved. After metal loading with acid-leaching solution, the amount of weak and total acids in metal-loaded HZSM-5 increased a lot, which was conducive to the dehydration, deoxygenation, decarbonylation and aromatization of pyrolysis, and the production of monocyclic aromatic hydrocarbons (MAHs). With 0.1 mol/L citric acid for LFP and NCM treatment, the metal-loaded catalyst showed a higher yield of benzene, toluene and xylene (BTX) of 11.10 wt% during catalytic pyrolysis of rape straw. Compared with 0.1-NCM-H5, a higher yield of MAHs was obtained over 0.1-LFP-H5 with a more favorable pore structure and higher amounts of weak and total acid sites. In general, the high-valued utilization of agricultural wastes and spent lithium batteries was realized in this study to produce clean energy.

Chitosan and β-Cyclodextrin-epichlorohydrin Polymer Composite Film as a Plant Healthcare Material for Carbendazim-Controlled Release to Protect Rape against Sclerotinia sclerotiorum (Lib.) de Bary

Delong Wang,Mingchen Jia,Lanying Wang,Shuang Song,Juntao Feng,Xing Zhang

The influence of β-cyclodextrin-epichlorohydrin (β-CD-EP) polymers on the improvement of the solubility and antifungal activity of carbendazim has been investigated. Meanwhile, the potential of the chitosan and β-CD-EP composite film used as a plant healthcare material for carbendazim-controlled release to protect rape againstSclerotinia sclerotiorum(Lib.) de Bary has been evaluated. β-CD-EP-1 and 2 (β-CD content, 750 mg/g and 440 mg/g, respectively) were found to significantly improve the solubility of the guest molecule carbendazim (17.9 and 18.5 times, respectively) and the 1:1 stoichiometry of the host-guest was confirmed by the Job’s plot. A slight synergism was observed for the β-CD-EP/carbendazim complex againstS. sclerotiorum(Lib.) de Bary, indicating an enhancement to the bioavailability of carbendazim. The in vitro release studies revealed that β-CD-EP polymers could efficiently modulate carbendazim release behaviors, such as the release retard and rate. The in vivo efficacy experiments demonstrated that the β-CD-EP/carbendazim and chitosan composite film could significantly prolong the effective duration of carbendazim at a concentration of 100 μg/mL compared with spraying carbendazim at 500 μg/mL. Thereby, a highly useful and strategic concept in plant disease control by a plant healthcare material—the chitosan and polymeric β-CD-EP composite film—is provided, which could also serve as a concept for related plant diseases.Keywords:β-cyclodextrin-epichlorohydrin;chitosan film;antifungal activity;controlled release;Sclerotinia sclerotiorum(Lib.) de Bary

Discovery of Highly Efficient Novel Potential Succinate Dehydrogenase Inhibitors Containing a Stilbene Group for Management of Sheath Blight of Rice and Sclerotinia Stem Rot of Oilseed Rape

Bo Luo,Mengxing Liu,Rongrong Li,Mengnan Zhou,Fengjia Zhang,Miao Wang,Yuqing Guo,Qingsi Zhang,Xinyi Liu,Yanbing Wu,Kun Fang,Rong Wang

In the search for novel succinate dehydrogenase inhibitors (SDHIs) fungicides for managing rice sheath blight (RSB) and sclerotinia stem rot (SSR), 28 pyrazole-4-carboxamides incorporating stilbene or diphenylacetylene scaffolds were synthesized and evaluated for antifungal activities. The results showed that compound 7m exhibited the most promising antifungal efficacy against Rhizoctonia solani and Sclerotinia sclerotiorum with EC50 (half maximal effective concentration) values of 0.004 and 0.028 μg/mL, outperforming Boscalid and Fluxapyroxad. Furthermore, the detached leaf assay showed that 7m showed remarkable protective and curative efficacies against R. solani and significant inhibition of S. sclerotiorum growth on oilseed rape leaves, indicating that it had a promising application prospect for RSB and SSR management. Further SDH enzymatic inhibition assay, scanning electron microscopy (SEM), molecular docking studies, and molecular dynamics (MD) simulations revealed that compound 7m shared an antifungal mechanism similar to that of Fluxapyroxad, which was further validated by density functional theory (DFT) and electrostatic potential (ESP) calculations. These findings suggested that 7m held significant promise as a novel SDHI for the effective management of RSB and SSR, warranting further investigation.

Preparation of activated carbon with heavy fraction of bio-oil from rape straw pyrolysis as carbon source and its performance in the aldol condensation for aviation fuel as carrier

Shanshan Shao,Laixin Ma,Xiaohua Li,Huiyan Zhang,Rui Xiao

Heavy bio-oil based carbon material (HBC) was prepared via carbonization and activation, and the basic Mg-Al-O/HBC using HBC as the carrier was synthesized for the first time for the aldol condensation of cyclopentanone to produce aviation fuel precursor. It revealed that a higher carbonization temperature and heating rate led to the decreasing production of biochar. HBC presented the highest specific surface area (2046.92 m 2 /g) and total pore volume (1.2964 cm 3 /g) at the carbonization temperature of 700 °C, the heating rate of 15 °C/min, A/C ratio of 3:1, and the activation temperature of 800 °C, in which the largest proportion of micropores was 87.19%. Compared with Mg-Al-O, a higher strength of weak and moderate basic sites was found in Mg-Al-O/HBC, which was beneficial for the aldol condensation. A higher yield of targeted condensates in the range of C9∼C16 was achieved at 87.28% in the aldol condensation of cyclopentanone over Mg-Al-O/HBC catalyst. However, the condensation performance of the composite catalysts decreased seriously in the 5th run, which was related closely to the appearance of boehmite AlO(OH) and the pore blockage of HBC. To match the basic catalyst and HBC carrier well from the viewpoint of pore property, Al ions were substituted with Zr ions, and the yield of the target condensates over Mg-Zr-O/HBC was kept stably at 72.07% at the fifth use. This study provides a unique, viable and effective method to produce porous carbon from heavy fraction of bio-oil as the carbon source, and also its good application in the production of high-valued fuels.
