Post-traumatic ankle osteoarthritis (PTOA) lacks effective conservative therapeutic strategies and requires the simultaneous mitigation of chronic inflammation, lubrication dysfunction, and progressive cartilage degeneration. In this study, spermidine-derived carbon quantum dots (Spd-CDs, SCD) were synthesized, and self-healing lubricating microspheres were fabricated from aldehyde hyaluronic acid (AHA) and 2,2′-diselanediyldiethanamine dihydrochloride (Sel). These microspheres (AHA+Sel, ASM) possess aldehyde-functionalized surfaces that enable preferential localization to damaged cartilage, and exhibit intrinsic self-healing capability through the synergistic effects of dynamic Schiff base bonds and diselenide bonds. Highly permeable SCD was subsequently encapsulated into ASM to construct a sustained-release system (ASM@SCD). In vitro, SCD exerted pronounced anti-inflammatory and antioxidative effects, inhibited chondrocyte ferroptosis, and restored dysregulated cartilage extracellular matrix metabolism. Mechanistically, bulk RNA-sequencing combined with targeted lipidomics revealed that SCD enhanced ferroptosis resistance by promoting ACSL3-dependent incorporation of monounsaturated fatty acids into plasma membranes, displacing polyunsaturated fatty acid-containing phospholipids, and thereby reducing susceptibility to lipid peroxidation. In vivo, ASM@SCD treatment suppressed early cartilage degeneration and aberrant subchondral bone remodeling, improved joint function, preserved normal gait patterns, and significantly attenuated PTOA progression. Collectively, ASM@SCD represents a promising non-surgical therapeutic strategy for PTOA.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by recurrent episodes and an inability to achieve a complete cure. The spermidine derivative (di- p -coumaroyl-caffeoyl spermidine, SPDD), as a key alkaloid, exhibits unique health benefits. However, it has not yet been reported whether SPDD can improve dextran sulfate sodium (DSS)-induced colitis in mice. Herein, we investigated the effects and mechanisms of SPDD on DSS-induced colitis in mice. SPDD was successfully purified from rose bee pollen and was found to have a protective effect on colitis, evidenced by reduced disease activity index (DAI) scores and colonic inflammation, increased colonic length and upregulated the expression of tight junction proteins (TJs) in the model ( p < 0.05). Importantly, the IL-17 signaling pathway showed significant enrichment by RNA sequencing (RNA-seq) technology with SPDD treatment, which resulted in the downregulation of MAPK4 expression ( p < 0.05). Furthermore, SPDD weakened the interaction between MAPK4 and AKT, resulting in a decrease in the phosphorylation level of AKT, thereby reducing the expression of IL-6, IL-1β, iNOS, and COX-2, and alleviating colitis ( p < 0.05). In addition, SPDD treatment also ameliorated TNF-α-induced inflammation in Caco-2 cells. Overall, our study demonstrated that SPDD reversed colonic inflammation in colitis mice through the MAPK4/AKT pathway and might be a promising candidate for UC intervention.

Chilling stress is a major abiotic constraint that affects sweet corn seedling establishment. Spermidine (Spd) is known to participate in the cold stress response in plants, nonetheless, the molecular mechanisms are largely unknown. The present study reported that exogenous Spd not only stimulated seedling growth but also enhanced photosynthetic attributes under chilling stress. Further analysis revealed that Spd application resulted in depressed abscisic acid (ABA) content and affected the expression of ABA-responsive genes. The augmented antioxidant enzymes activities, coupled with declined reactive oxygen species (ROS) content, were observed in response to Spd treatment under chilling stress. Moreover, Spd up-regulated the transcription levels of some Ca 2+ transport-related genes. We propose that Spd contributes to cold tolerance in sweet corn seedlings via the regulation of genes involved in the ROS, ABA pathways and Ca 2+ transport.

Two wheat cultivars ( Triticum aestivum L.) were used to evaluate the effects of post-anthesis severe water deficit (SD) on starch content and granule size distribution and their relations with ethylene and spermidine (Spd). Comparison to the well-watered (WW) treatment, SD led to lower Spd and higher 1-aminocylopropane-1-carboxylic acid (ACC) concentrations and ethylene evolution rate (EER) in grains at the critical stage of forming starch granules. Application of Spd or aminoethoxyvinylglycine (AVG) significantly reduced ACC concentration and EER and increased Spd concentration, while ethephon or methylglyoxal-bis (MGBG) had an opposite impact. The volume and surface area distribution of starch granules showed a bimodal curve, while the number distribution exhibited a unimodal curve. SD caused a marked drop in grain weight, grain number and starch content, also led to a significant reduction in the proportion (both by volume and by surface area) of B-type starch granules (<10 μm), with an increase in those of A-type starch granules (>10 μm). Application of Spd or AVG increased the proportion (both by volume and by surface area) of B-type starch granules under SD. Correlation analysis suggested that ethylene and Spd showed an antagonism relation in the formation of B-type granules. These results suggested that it would be good for the formation of B-type starch granules to have the physiological traits of higher Spd and lower ACC concentrations and ethylene emission under SD.

Fluorescent cuprous iodide (CuI)-based coordination polymers (CuI-CPs) were widely used in the sensor field for high selectivity and sensitivity. Here, a new CuI-CP fluorescent probe was fabricated for the detection of spermine, spermidine and cysteine with dual-mode, which was capable of both convenient colorimetric analysis and ratiometric fluorescent detection. The fluorescence of CuI-CP solution was quenched and the solution changed from white to purple upon addition of spermine or spermidine, while the solution changed from white to yellow for cysteine. This probe exhibited selective, sensitive, rapid and linear response to spermine, spermidine and cysteine with a limit of detection of 0.57, 1.06 and 1.18 μM, respectively. Furthermore, cellulose acetate was used as the matrix to fabricate intelligent fluorescent film for real-time and visual monitoring the freshness of pork samples based on distinct fluorescence changes under UV light. This work realized a rapid, efficient, and profitable method for visually evaluating food quality.

Brain aging primarily manifests as cognitive decline. Spermidine (SPD) is known to ameliorate age-related cognitive decline, and the gut microbiota is an important source of SPD. Clostridium butyricum ( C. butyricum ), a probiotic, can alleviate cognitive decline in individuals with various neurodegenerative diseases, and C. butyricum might generate SPD. However, the role of C. butyricum in natural brain aging and the efficacy of C. butyricum and the metabolite SPD in ameliorating age-related cognitive decline remain unclear. Here, we investigated the effects of C. butyricum and its metabolite SPD on cognitive decline during natural brain aging and the mechanisms underlying these effects. Aged rats (aged 17 months, n = 12 per group) were randomly divided into normal saline (NS), C. butyricum (1 ×10 8 ∼10 9 CFU/ml, by gavage for two months) and SPD (6.5 mg/kg/d, by gavage for one month) groups. Compared with the NS group, the C. butyricum and SPD groups presented improved spatial reference and recognition memory, reduced aging marker levels by immunohistochemical staining, improved synaptic plasticity-related tissue structure by transmission electron microscopy and Golgi staining, and increased level of synaptophysin and BDNF by immunofluorescence staining and western blotting in the brain. Furthermore, treatment with C. butyricum and SPD enhanced intestinal barrier function and decreased serum proinflammatory factor levels and brain tissue damage due to oxidative stress. There were no significant differences in relevant indicators between the C. butyricum and SPD groups. In conclusion, C. butyricum and its metabolite SPD may delay the cognitive decline caused by normal brain aging by reducing oxidative stress and inflammation.

Exploring biopolymer-based antibacterial packaging materials is promising to tackle the issues caused by petroleum plastic pollution and microbial contamination. Herein, a novel packaging material with two antibacterial modes, continuous and efficient, is constructed by dispersing positively charged spermidine carbon dots (Spd-CDs) in a carrageenan/polyvinyl alcohol (CP) composite biopolymer. The obtained nanocomposite film (CP/CDs film) not only gradually releases the ultra-small Spd-CDs but also rapidly generates reactive oxygen species to inhibit the reproduction of E. coli and S. aureus . Benefiting from the complementary advantages of carrageenan and polyvinyl alcohol, as well as the addition of Spd-CDs, the CP/CDs films exhibit high transparency, good mechanical performance, water vapor barrier ability, low migration, etc. The CP/CDs film as a packaging material is validated to be effective in preventing microbial contamination of pork samples. Our prepared nanocomposite film with sustainability and efficient antibacterial properties is expected as food active packaging.

ObjectivesThis study aimed to investigate the efficacy and mechanism of Ferulic acid (FA) in a high-fat diet (HFD)-induced murine model of Metabolic dysfunction-associated steatotic liver disease (MASLD), focusing on the gut microbiota-polyamine-macrophage polarization axis.MethodsA murine MASLD model was induced with an HFD and treated with FA. We evaluated its efficacy through serological and histopathological analyses. Key mechanisms, including macrophage polarization and gut microbiota, were investigated using molecular analyses and 16S rRNA gene sequencing. The direct effect of spermidine on Kupffer cell (KC) polarization was confirmed in vitro.Key findingsFA administration attenuated HFD-induced increases in body and liver weight and improved liver pathology. Mechanistically, FA treatment ameliorated gut dysbiosis, increasing beneficial genera like Akkermansia, Enterorhabdus, and Eubacterium xylanophilum, which was associated with elevated serum spermidine. Critically, FA suppressed hepatic KC M1 polarization and enhanced M2 polarization. In vitro experiments confirmed that spermidine directly mediated these immunomodulatory effects.ConclusionsFA alleviates MASLD by modulating a novel gut microbiota-spermidine-macrophage axis. These findings suggest a new hepatoprotective mechanism for FA and highlight this axis as a potential therapeutic target for MASLD.

Heat stress has been a major environmental factor limiting the growth and development of Pinellia ternata which is an important Chinese traditional medicine. It has been reported that spermidine (SPD) and melatonin (MLT) play pivotal roles in modulating heat stress response (HSR). However, the roles of SPD and MLT in HSR of P. ternata , and the potential mechanism is still unknown. Here, exogenous SPD and MLT treatments alleviated heat-induced damages in P. ternata , which was supported by the increased chlorophyll content, OJIP curve, and relative water content, and the decreased malondialdehyde and electrolyte leakage. Then, RNA sequencing between CK (control) and Heat (1 h of heat treatment) was conducted to analyze how genes were in response to short-term heat stress in P. ternata . A total of 14,243 (7870 up- and 6373 down-regulated) unigenes were differentially expressed after 1 h of heat treatment. Bioinformatics analysis revealed heat-responsive genes mainly included heat shock proteins ( HSPs ), ribosomal proteins, ROS-scavenging enzymes, genes involved in calcium signaling, hormone signaling transduction, photosynthesis, pathogen resistance, and transcription factors such as heat stress transcription factors ( HSFs ), NACs , WRKYs , and bZIPs . Among them, PtABI5 , PtNAC042 , PtZIP17 , PtSOD1 , PtHSF30 , PtHSFB2b , PtERF095 , PtWRKY75 , PtGST1 , PtHSP23.2 , PtHSP70 , and PtLHC1 were significantly regulated by SPD or MLT treatment with same or different trends under heat stress condition, indicating that exogenous application of MLT and SPD might enhance heat tolerance in P. ternata through regulating these genes but may with different regulatory patterns. These findings contributed to the identification of potential genes involved in short-term HSR and the improved thermotolerance by MLT and SPD in P. ternata , which provided important clues for improving thermotolerance of P. ternata .

The importance of synergy has been underscored in recent medical research for augmenting the efficacy of therapeutic interventions, targeting multiple biological pathways simultaneously. Our prior research elucidated that Dendrobium officinale polysaccharide (DOP) has the potential to prolong the lifespan of Caenorhabditis elegans ( C. elegans ) via regulating gut microbiota. Concurrently, spermidine (Spd), as a mimicking caloric restriction, facilitates autophagy and exerts a pronounced anti-aging effect. To enhance the anti-aging capabilities of DOP, we conducted a comprehensive study examining the combined effects of DOP and Spd in C. elegans , incorporating metabolomics analysis to investigate the underlying mechanisms. A combination of 250 mg/L DOP and 29.0 mg/L Spd yielded the most favorable outcomes in lifespan extension, evidencing a synergistic effect with a combination index (CI) of 0.65. In oxidative and heat stress tolerance assays, the observed CIs were 0.50 and 0.33, respectively. Metabolomic analysis highlighted significant alterations in metabolites related to lipid, nucleotide and energy metabolism, notably regulating glycerol 3-phosphate, linoleoyl glycerol, docosapentaenoic acid and β-nicotinamide mononucleotide, nicotinamide adenine dinucleotide. The effects of DS on lipid metabolism were further validated using Oil Red O staining and triglyceride level in C. elegans . The results indicated that DS may primarily be via modulating lipid metabolism. To further confirm these findings, a high-fat diet-induced mouse model was employed. Consequently, it can be inferred that the synergistic anti-aging impact of DOP and Spd is likely mediated primarily through alterations in lipid metabolic processes.

Spermidine has emerged as a promising biomarker for the early-stage diagnosis of cancer in humans. A novel extractive spectrophotometric analytical method has been developed for the determination of spermidine in urine samples. This technique leverages the selective extraction of spermidine from complex matrices using a cucurbit[7]uril-truxene derivative (Q[7]-Cs-Tr) as a fluorescent extractant. The Q[7]-Cs-Tr forms a stable host-guest complex with spermidine, facilitating its extraction from the aqueous phase to the organic phase. The formation of this complex results in a decrease in the fluorescence intensity of the Q[7]-Cs-Tr in the organic phase. Key factors influencing extraction efficiency, including the type of dispersed organic solvent, extractant concentration, solution pH, extraction times, and equilibrium time, were thoroughly investigated. The method demonstrated minimal interference from various metal ions, anions, amino acids, and other biogenic amines. It also exhibited a low limit of detection (8.6 nM), excellent linearity (R 2 = 0.9972), and high repeatability (RSD = 2.4 % intra-day, RSD = 3.6 % inter-day). The proposed method was successfully applied to determine spermidine levels in urine samples, yielding satisfactory recoveries ranging from 96.3 % to 103.5 %. Compared to existing methods, this approach is simple, reliable, highly sensitive, and exhibits low interference, making it a potential robust tool for clinical diagnostics.

Diquat (DQ) is a widely used herbicide, whose residues have been detected in food products, raising concerns about potential health risks. Nevertheless, the mechanisms underlying male reproductive damage after chronic DQ exposure remain unclear. In this study, we observed that chronic DQ exposure reduces Sertoli cell numbers and triggers mitochondrial quality control (MQC) impairment, oxidative stress, apoptosis, and blood–testis barrier (BTB) disruption in testicular tissue. Mechanistically, DQ downregulates NBR1 in Sertoli cells, leading to impaired mitophagy flux, increased mitochondrial fission, and elevated oxidative stress, which ultimately promote Sertoli cell apoptosis and compromise cellular barrier function. Metabolomic analysis indicated that chronic DQ exposure lowers spermidine (SPD) levels in the mouse testes. Exogenous SPD supplementation attenuated DQ-induced MQC dysfunction, oxidative stress, apoptosis, and BTB damage. Together, these results establish NBR1 as a critical regulator of DQ-triggered Sertoli cell injury and propose SPD as a potential therapeutic candidate for alleviating DQ-induced male reproductive impairment.
