Electrocatalytic oxidation of NADH at graphene-modified electrodes based on electropolymerized poly(thionine-methylene blue) films from nature deep eutectic solvents

Minling Ding,Tongtong Hou,Huizhe Niu,Nan Zhang,Ping Guan,Xiaoling Hu

In this paper, a sensing platform for the electrocatalytic oxidation of nicotinamide adenine dinucleotide (NADH) was proposed. It was prepared by the electropolymerization of poly(thionine-methylene blue) (PTH-MB) in ternary nature deep eutectic solvent (NADES) on a glassy carbon electrode (GCE) that was modified with electrochemically reduced graphene oxide (ERG). The modified electrode was denoted as PTH-MB NADES -ERG/GCE. Cyclic voltammetry (CV), chronocoulometry (CC), electrochemical impedance spectroscopy (EIS), scanning electron microscope (SEM), Raman spectroscopy, and Ultra Violet-Visible Spectroscopy (UV–Vis) were used to characterize the preparation processes. The electrochemical performances of the electrode in NADH oxidation were studied via CV and current-time methods. The NADH oxidation overpotential was lower on PTH-MB NADES -ERG/GCE compared to that on GCE, ERG/GCE, and PTH-MB NADES /GCE. The PTH-MB NADES -ERG/GCE showed excellent electrocatalytic activity for NADH oxidation with a linear range of 0.51–3333.33 µM (R 2  = 0.9534) and a detection limit of 0.159 nM. The sensor was further tested for NADH determination in artificial urine samples, showing promising biomedical applications.

An electrochemical biosensor for T4 polynucleotide kinase activity assay based on host–guest recognition between phosphate pillar[5]arene@MWCNTs and thionine

Aiwen Su,Dan Luo,Shixuan Li,Yanli Zhang,Hongbin Wang,Lijuan Yang,Wenrong Yang,Pengfei Pang

T4 polynucleotide kinase helps with DNA recombination and repair. In this study, an electrochemical biosensor was developed for a T4 polynucleotide kinase activity assay and inhibitor screening based on phosphate pillar[5]arene and multi-walled carbon nanotube nanocomposites. The water-soluble pillar[5]arene was employed as the host to complex thionine guest molecules. The substrate DNA with a 5′-hydroxyl group initially self-assembled on the gold electrode surface through chemical adsorption of the thiol group, which was phosphorylated in the presence of T4 polynucleotide kinase. Titanium dioxide nanoparticles served as a bridge to link phosphorylated DNA and phosphate pillar[5]arene and multi-walled carbon nanotube composite due to strong phosphate-Ti4+-phosphate chemistry. Through supramolecular host–guest recognition, thionine molecules were able to penetrate the pillar[5]arene cavity, resulting in an enhanced electrochemical response signal. The electrochemical signal is proportional to the T4 polynucleotide kinase concentration in the range of 10−5 to 15 U mL−1 with a detection limit of 5 × 10−6 U mL−1. It was also effective in measuring HeLa cell lysate-related T4 polynucleotide kinase activity and inhibitor screening. The proposed method offers a unique sensing platform for kinase activity measurement, holding great potential in nucleotide kinase-target drug development, clinical diagnostics, and inhibitor screening.

Selective aptasensor of deoxynivalenol based on dual signal enhancement of thionine electrochemistry using silver nanoparticle-loaded label at gold nanoparticle-loaded electrodes

Xiaoying Cui,Ke Zhao,Zhengquan Qu,Xipeng Chao,Lingling Xie,Hanyu Chen,Baoshan He,Baozhong Zhang

In this work, an efficient sensing platform deoxynivalenol (DON) detection was constructed through monitoring the current change of a competitive mechanism triggered by DON, leading the signal label detached from the electrode surface by square-wave voltammetry using thionine (Thi) as a redox indicator. The complementary strand of aptamer (cDNA) and Thi were loaded onto Fe/Ni bimetallic metal–organic framework loaded with sliver nanoparticles (AgNPs@FeNi-MOF) to construct AgNPs@FeNi-MOF/cDNA/Thi signal probes. In the presence of DON, the aptamer sequence was more predisposed to form an aptamer-DON complex, resulting in the displacement of the cDNA. The signal probe was subsequently released, leading to a decrease in the signal intensity of Thi. Notably, AgNPs@FeNi-MOF has a larger electroactive specific surface area and is able to load more cDNA and thi, which can amplify the signal. Under the optimal experimental conditions, the developed sensor exhibits a good linear response in the range of 1 × 10 −2 to 1 × 10 4 pg/mL, with a limit of detection (LOD) of 5.68 fg/mL and has good selectivity, reproducibility and stability.

A bimetallic nanoparticle/graphene oxide/thionine composite-modified glassy carbon electrode used as a facile ratiometric electrochemical sensor for sensitive uric acid determination

Xiaohui Gao,Rijun Gui,Kendrick Qizhou Xu,Huijun Guo,Hui Jin,Zonghua Wang

A novel and facile ratiometric electrochemical sensor was developed for the sensitive determination of uric acid (UA). Using one-step co-reduction of HAuCl4 and AgNO3 (as precursors) under cyclic voltammetry scanning, gold–silver bimetallic nanoparticles (Au–Ag NPs) were electrodeposited on the surface of a glassy carbon electrode (GCE). Through electrostatic interactions, self-assembly of graphene oxide (GO) and thionine (TH) occurred to form a GO–TH complex. The complex was drop-coated on Au–Ag NPs to prepare the Au–Ag NPs/GO/TH@GCE sensing platform through interactions among different components. The electrochemical signal responses of the sensing platform were recorded by square wave voltammetry measurements. With the increase of UA concentration [UA], the redox current peak intensity of UA (IUA at 0.46 V) increased regularly and that of TH (ITH at −0.28 V) was almost unchanged. The relationship between IUA/ITH and [UA] (1–100 μM) was linearly plotted (R2 = 0.9929), with a low detection limit of 0.3 μM. A facile sensor was fabricated based on the sensing platform and enabled sensitive ratiometric electrochemical sensing of UA in real human serum and urine fluids, over potential interferents. The experimental results confirmed high detection stability and recoveries of this sensor, indicating its high feasibility for UA detection in biological fluids.

Reduced Graphene Oxide-Zirconium Dioxide–Thionine Nanocomposite Integrating Recognition, Amplification, and Signaling for an Electrochemical Assay of Protein Kinase Activity and Inhibitor Screening

Zhiqiang Chen,Ying Liu,Lijie Hao,Zhencai Zhu,Fang Li,Shufeng Liu

Protein kinase activity analysis is essential and important for elucidation of many fundamental biological processes, disease diagnosis, and drug discovery. Herein, a novel electrochemical biosensing method for protein kinase (PKA) activity was demonstrated by the use of a reduced graphene oxide-zirconium dioxide–thionine (rGO-ZrO2–Thi) nanocomposite, which interestingly served as an integral phosphopeptide-recognizing, signal amplifying and reporting platform. The ZrO2 nanoparticle-decorated reduced graphene oxide (rGO-ZrO2) was first prepared by a hydrothermal reaction route, and then the thionine was conjugated onto the rGO surface via π–π stacking as an excellent electrochemical probe. The prepared rGO-ZrO2–Thi nanocomposites were well-characterized by various techniques. With the full advantage of specific recognition of ZrO2 nanoparticles for the phosphate group, signal amplification, and transduction of abundant thionines onto the rGO surface, and excellent conductivity of rGO, the rGO-ZrO2–Thi nanocomposite endowed a label-free and one-step electrochemical analysis of kemptide phosphorylation catalyzed by PKA. The detection limit for PKA activity was experimentally achieved as 0.005 U/mL, which was evidently lower than most of the reported methods. The proposed sensing strategy could be also applied for an efficient inhibitor evaluation. Therefore, it offered an excellent pathway for a generic and sensitive electrochemical assay of PKA activity and inhibitor.

A Simple Ratiometric Electrochemical Aptasensor Based on the Thionine–Graphene Nanocomposite for Ultrasensitive Detection of Aflatoxin B2 in Peanut and Peanut Oil

Fan Jia,Yuye Li,Qingfa Gong,Dong Liu,Shuyun Meng,Chengxi Zhu,Tianyan You

The accurate and reliable analysis of aflatoxin B2 (AFB2) is widely required in food and agricultural industries. In the present work, we report the first use of a ratiometric electrochemical aptasensor for AFB2 detection with high selectivity and reliability. The working principle relies on the conformation change of the aptamer induced by its specific recognition of AFB2 to vary the ratiometric signal. Based on this principle, the proposed aptasensor collects currents generated by thionine–graphene composites (ITHI) and ferrocene-labeled aptamers (IFc) to output the ratiometric signal of ITHI/IFc. In analysis, the value of ITHIremained stable while that of IFcincreased with higher AFB2 concentration, thus offering a “signal-off” aptasensor by using ITHI/IFcas a yardstick. The fabricated aptasensor showed a linear range of 0.001–10 ng mL−1with a detection limit of 0.19 pg mL−1for AFB2 detection. Furthermore, its applicability was validated by using it to detect AFB2 in peanut and peanut oil samples with high rates of recovery. The developed ratiometric aptasensor shows the merits of simple fabrication and high accuracy, and it can be extended to detect other mycotoxins in agricultural products. Keywords:ratiometry;electrochemical aptasensor;thionine;aflatoxin B2

Modified exfoliated graphene functionalized with carboxylic acid-group and thionine on a screen-printed carbon electrode as a platform for an electrochemical enzyme immunosensor

Wang Jing,Zhang Liang,Yan Guanrong,Cheng Linfeng,Zhang Fanglin,Wu Jialin,Lei Yingfeng,An Qunxing,Qi Honglan,Zhang Chengxiao,Gao Qiang

An enzyme immunoassay was developed based on the coulometric measurement of immunoglobulin M (IgM) against Hantaan viruses (HTNV) by using virus-like particles (VLPs) as recognition molecules. The surface functionalization of screen-printed carbon electrodes (SPCEs) was achieved through paste-exfoliated graphene that was modified with a COOH group and a thionine mediator through supramolecular-covalent scaffolds, on SPCEs by using the binder contained in the ink. After the covalent immobilization of the antibody, the sensor was used for the sandwich enzyme immunoassay of IgM against HTNV. By using HTNV VLPs as the second recognization molecules, the resulting sensor efficiently monitored the reaction of IgM against HTNV and anti-IgM antibody with high specificity. By attaching HTNV nucleocapsid protein antibody conjugate with horseradish peroxidase (HRP) onto VLPs, the signal response of the assay was derived from the coulometric measurement of H 2 O 2 reduction mediated by thionine on the electrode surface after the application of a potential (− 0.2 V vs. Ag/AgCl). The ratio of charges measured before or after H 2 O 2 addition was used to quantify IgM because these charges could be used as background charges or total charges, respectively. The ratio exhibited good agreement with IgM concentration within a range 0.1 to 1000 pg mL −1 , and a detection limit of 0.06 pg mL −1 was obtained. The assay demonstrated high sensitivity and specificity toward HTNV-specific IgM in serum. Graphical abstract

Electrochemical aptasensor based on aptamer-complimentary strand conjugate and thionine for sensitive detection of tetracycline with multi-walled carbon nanotubes and gold nanoparticles amplification

Bao-Shan He,Sa-sa Yan

Herein, a novel aptasensor with thionine as the electroactive probe was fabricated for the sensitive detection of tetracycline (TET). Carboxyl functionalized multi-walled carbon nanotubes and gold nanoparticles composites (COOH-MWCNTs/AuNPs) with high surface area and excellent conductivity were employed as the carrier of the complementary strands (CS). In addition, thionine was added to enhance the sensitivity of the developed aptasensor. After the immobilization of the COOH-MWCNTs/AuNPs/CS on the aptamer modified gold electrode (Apt/AuE), thionine was added on the surface of the electrode to complete the construction of the aptasensor. The change of the peak currents (ΔI) of thionine, evaluated by differential pulse voltammetry before and after the incubation of TET solution, was used for TET detection. Under optimal conditions, the proposed aptasensor showed a wide linearity from 0.1 nM to 1 μM and a low detection limit of 0.06 nM. In addition, the aptasensor displayed good selectivity, reproducibility and stability.

Electrochemical biosensor based on enzyme substrate as a linker: Application for aldolase activity with pectin-thionine complex as recognization element and signal amplification probe

Xiaonan Wang,Meiwen Wang,Yuanyuan Zhang,Xiaocao Miao,Yuanyuan Huang,Juan Zhang,Lizhou Sun

A new strategy to fabricate electrochemical biosensor is reported based on the linkage of enzyme substrate , thereby an electrochemical method to detect aldolase activity is established using pectin-thionine complex (PTC) as recognization element and signal probe . The linkage effect of fructose-1,6-bisphosphate (FBP), the substrate of aldolase, can be achieved via its strong binding to magnetic nanoparticles (MNPs)/aminophenylboronic acid (APBA) and the formation of phosphoramidate bond derived from its reaction with p -phenylenediamine (PDA) on the surface of electrode. Aldolase can reversibly catalyze the substrates into the products which have no binding capacity with MNPs/APBA, resulting in the exposure of the corresponding binding sites and its subsequent recognization on signal probe. Meanwhile, signal amplification can be accomplished by using the firstly prepared PTC which can bind with MNPs/APBA, and accuracy can be strengthened through magnetic separation. With good precision and accuracy, the established sensor may be extended to other proteins with reversible catalyzed ability.

An amperometric zearalenone aptasensor based on signal amplification by using a composite prepared from porous platinum nanotubes, gold nanoparticles and thionine-labelled graphene oxide

He Baoshan,Yan Xiaohai

The authors constructed a voltammetric zearalenone (ZEN) aptasensor based on use of porous platinum nanotubes, gold nanoparticles (p-PtNTs/AuNPs) and thionine (Thi) labelled graphene oxide (GO). The p-PtNTs were synthesized in-situ based on tellurium nanowires as sacrificial templates. Subsequently, thiol-modified aptamers were self-assembled on the AuNPs that had been electrodeposited on the surface of the modified electrode. The presence of p-PtNTs on the electrode increases the loading with AuNPs and aptamers. It also warrants that the Thi-labelled GO can be assembled onto the aptamer via π interactions. In the presence of ZEN, it will be bound by the aptamer. The GO/Thi conjugate will be released from the aptamer, and this causes a decrease in Thi current. Under the optimal conditions and at a typical working potential of −0.22 V (vs. Ag/AgCl), the method has a linear range that covers the 0.5 pg·mL −1 to 0.5 μg·mL −1 ZEN concentraion range and a lower detection limit of 0.17 pg·mL −1 . Voltammetric zearalenone aptasensor based on use of porous platinum nanotubes/gold nanoparticles and thionine labelled graphene oxide was fabricated for the detection of zearalenone.

Dual mode competitive electrochemical immunoassay for dibutyl phthalate detection based on PEI functionalized nitrogen doped graphene-CoSe2/gold nanowires and thionine-Au@Pt core-shell

Jiawen Li,Huali Jin,Min Wei,Wenjie Ren,Jinshui Wang,Yurong Zhang,Ligen Wu,Baoshan He

In this work, a sensitive dual mode competitive electrochemical immunosensor based on differential pulse voltammetry (DPV) and square wave voltammetry (SWV) response modes for quantitative determination of dibutyl phthalate (DBP) was successfully established. According to diethylenetriamine (DETA) assisted hydrothermal strategy, a novel N-doped graphene/CoSe 2 nanobelt composite was prepared. Afterwards, PEI functionalized NG-CoSe 2 and gold nanowires (AuNWs) were further compounded as a preliminary signal amplification platform for immunosensor. Thionine (Thi)-loaded [email protected] and secondary antibody (Ab 2 ) connected as a signal label. The immobilized coating antigen DBP-BSA and the DBP compete for limited antibody binding sites. The concentration of DBP can be determined by DPV detect the Thi signal loaded on the [email protected] 2 label. It can also be determined by SWV mode to detect the current generated by the reaction system of H 2 O 2 and o-phenylenediamine (o-PD) catalyzed by [email protected] with peroxidase like properties. Under optimum conditions, the linear ranges of the DPV and SWV modes were 1 pg mL −1 ∼1 μg mL −1 . And LOD were 0.486 pg mL -1 (DPV) and 0.711 pg mL -1 (SWV), respectively. Besides, it exhibited outstanding selectivity, reproducibility, stability and can be used for sensitive detection of DBP in real samples.

A novel ratiometric electrochemical sensor for the selective detection of citrinin based on molecularly imprinted poly(thionine) on ionic liquid decorated boron and nitrogen co-doped hierarchical porous carbon

Xiaopeng Hu,Yiwei Liu,Yide Xia,Faqiong Zhao,Baizhao Zeng

Citrinin can cause serious human diseases, thus its detection in foods is necessary. Herein, a molecularly imprinted polymer-based ratiometric electrochemical sensor (MIP-RECS) was presented for citrinin detection. The sensor was fabricated by electropolymerization, using thionine as monomer and citrinin as template. The ionic liquid decorated boron and nitrogen co-doped hierarchical porous carbon (BN-HPC) as supporter, provided large surface for anchoring thionine and citrinin. Poly(thionine) not only acted as MIP, but also acted as reference probe. When [Fe(CN) 6 ] 3-/4- was adopted as indicating probe, the resulting sensor demonstrated a wide linear detection range (i.e. 1 × 10 -3 –10 ng mL −1 ) and a low detection limit (i.e. 1 × 10 -4 ng mL −1 ).The sensor was applied to the detection of spiked citrinin in real samples, and satisfactory recovery (i.e. 97% – 110%) was obtained. Hence, it was promising for citrinin detection.
