Based on ExoIII-assisted nucleic acid circulation and catalytic hairpin assembly (CHA) reaction, a dual-amplification electrochemical biosensors strategy for sensitive detection of tobramycin was presented. When the target was present, the trigger 1 (T1) was released to unfold hairpin DNA (H1), which allowed the ExoIII-assisted nucleic acid circulation to proceed smoothly and create a lot of trigger 2(T2). Following that, the CHA reaction between H2 and H3 proceeded under the catalysis of T2 and produced plenty of H2-H3 complex. Signal probe labeled with ferrocene was immobilized on the electrode ahead by adenine (PolyA). Finally, the H2-H3 complex combined with the probe and the DNA strand labeled with ferrocene (C2) fell off the electrode surface. The above process significantly reduced the electrochemical signal, so the biosensor has a low detection limit of 0.11 nM. Moreover, this biosensor has a satisfactory regenerability.

A target-induced strand displacement (TISD) electrochemical aptasensor is reported for the sensitive determination of tobramycin (TOB). A novel substrate was synthesized by loading cerium-based metal organic framework (Ce-MOF) that was modified by Pd nanoparticles (PdNPs), onto polyethyleneimine (PEI)-modified molybdenum disulfide (MoS2). The substrate (denoted by PEI-MoS2/Ce-MOF@PdNPs) was used to modify the Au electrode (AuE) to expand the electrode surface area for subsequent cDNA loading. Gold-platinum core–shell particles (Au@Pt) loaded with aptamers (Apt) were introduced as signal tags and mimetic peroxidase to hybridize with cDNA and form a double-stranded structure. When the target was present, strand displacement dominated by TISD occurred and the signal tag was released from the electrode surface, turning the signal off. Using the optimal conditions, the linear range of the sensor was from 5 × 10−11 to 1 × 10−6 mol/L and the detection limit was 1.05 × 10−12 mol/L with good stability, reproducibility, and specificity. The aptasensor provided good performance for the determination of tobramycin in milk.

Antibiotic contaminants in food are associated with detrimental health issues, and thus, the design of rapid and reliable detection methods to monitor them is in high demand. Although colorimetric aptamer sensing methods can achieve accurate and quick visual read-out analysis, they still have morphological defects that affect the detection efficiency and stability of the aptasensor. Hence, a simple and highly selective iron oxide/graphene oxide (Fe 3 O 4 /GO) nanospheres was developed and modified with an aptamer as nanozymes for colorimetric detection of tobramycin (Tob). Due to the electrostatic and π-π stacking interaction between the aptamer and the spherical Fe 3 O 4 /GOs nanozyme, the catalytic activity of the nanozyme was enhanced by the robust affinity between the aptamer and the 3,3′,5,5′-tetramethylbenzidine (TMB) substrate. However, the presence of Tob could obstruct the catalytic activity through the specific binding of aptamer to Tob and the detachment of aptamer from the surface of the Fe 3 O 4 /GO, causing a low catalytic activity. Based on this strategy, a visual and quantitative detection for Tob was achieved with a detection limit of 70.7 nM. The proposed aptasensor demonstrated high selectivity, ease of operation, and potential application in food samples with satisfactory recoveries of 98.4–108.5 %. Therefore, this proposed method has great prospects for monitoring food safety and could be applied for sensing varied antibiotics by just switching the target-specific aptamer.

: The use of multiple antibiotics in combination therapy for bacterial infections has become routine in clinical practice, whereas the increasing demand for therapeutic drug monitoring (TDM) imposes stricter requirements on analytical methods. The combined administration of vancomycin (Van) and tobramycin (TOB) offers broader antimicrobial coverage across diverse pathogenic species. However, both antibiotics possess narrow therapeutic windows, making the parallel monitoring of their blood concentrations crucial for precise and safe dosing. In this study, an extended-gate field-effect transistor (EG-FET) sensor fabricated from gold-doped, paper-based laser-induced graphene (LIG) is presented. The sensor operates via an aptamer-based, label-free response mechanism, enabling the multiplexed and rapid therapeutic drug monitoring of Van and TOB. The incorporation of paper-based substrates not only simplifies device fabrication but also enhances the overall sensing sensitivity. The EG-FET sensor exhibits excellent linear responses toward Van and TOB over concentrations ranging from 0.1 nM to 100 μM, with detection limits as low as 6.89 pM for Van and 52.9 pM for TOB. To enable point-of-care TDM applications, a portable and wireless sensing platform was assembled. Additionally, our EG-FET sensors can accurately detect Van and TOB in actual blood samples (including human blood and mice serum). This work represents the first demonstration of an EG-FET sensing platform enabling parallel and multiplexed detection of vancomycin and tobramycin, providing a promising strategy for multi-analyte therapeutic drug monitoring of antibiotics with narrow therapeutic windows.

High efficiency of in-situ cross-linking and acid triggered drug delivery is realized by introducing tobramycin into the hydrogels. Injectable and biodegradable hydrogels are prepared through two steps: First generation of reactive aldehyde groups in the sodium alginate (A-Alg) and then introduction of antibiotic tobramycin as cross-linker. Due to the formation of dynamic Schiff base bonds between the amino groups in tobramycin and aldehyde groups in A-Alg, the gelation of hydrogels can be realized immediately. Thus, tobramycin acts well as the first role cross-linker and the hydrogels containing tobramycin can be injected into the wound during the treatment. In addition, the acid from the decomposition of organic compounds by the bacteria can break the cross-linking points previously formed by tobramycin in the hydrogels. Therefore, tobramycin can be released and act as the second role model drug to kill the bacteria. Because the hydrogels network is broken, the release of tobramycin is more efficient than the traditional drug delivery from hydrogels by diffusion . Based on these unique properties, the present hydrogels containing tobramycin exhibit a good injectable and biodegradable capability. In addition, due to the existence of the reversible acid-labile linkages in the hydrogels, the hydrogels containing tobramycin are also self-healing, which additionally is favorable for the application of wound dressing. More importantly, the antibacterial hydrogels also demonstrate good biocompatibility in vitro and significantly therapeutic effects on an infected mice model in vivo. Based on the above special properties, the hydrogels cross-linked by tobramycin indicate a new approach to prepare hydrogel dressings with low-cost, non-toxicity and good anti-bacterial performance in the treatment of infectious wounds.

Tobramycin (TOB) plays a considerable role in combating milk spoilage and preventing disease in dairy cows. However, overuse of TOB can lead to nephrotoxicity, ototoxicity, neuromuscular blockade, and hypersensitivity reactions. Here, nitrogen-doped carbon dots ( N -CDs) were prepared with ethylenediamine and citric acid, then molecularly imprinted layers were obtained by imprinting of surface on the N -CDs to prepare Nitrogen-doped carbon dot-based molecularly imprinted polymers ( N -CDs@MIPs). The fluorescence emission spectrum of this probe showed a linear enhancement with the TOB concentration in the 1–12 μM. Meanwhile, a detection limit of 99.2 nM was obtained. This probe was not affected by the structural analogs of the TOB and can show high sensitivity and selectivity compared to non-imprinted polymers ( N -CDs@NIPs). Therefore, it can be successfully used for the trace analysis of TOB in milk with advantages over other reported techniques such as liquid chromatography coupled with tandem mass spectrometry or various aptamer sensors.

High-salinity textile wastewater is a pressing environmental concern, urgently requiring the appropriate treatment. Sustainable management of high-salinity textile wastewater via fractionation of the existing dyes and inorganic salts (NaCl) using selective separation membranes is a key approach to address this detrimental concern. Herein, tight composite ultrafiltration membranes with impressive antibacterial function were constructed using one-step co-deposition of dopamine and tobramycin on the porous substrate to fractionate the dyes and NaCl. Triggered by ammonia persulfate, the polydopamine/tobramycin coating layer imparts the fabricated composite ultrafiltration membranes with tight surface structure and narrow pore size distribution, enabling effective dyes/salts fractionation. Specifically, the tight composite ultrafiltration membrane with a molecular weight cutoff of 3692 Da experienced a > 98.0 % rejection against various reactive dyes (molecular weight: <1000 Da) with nearly complete salt transmission (<0.8 % NaCl rejection), demonstrating an effective fractionation of dyes and salts. Additionally, the incorporated tobramycin as a broad-spectrum antibiotic endowed the tight composite ultrafiltration membrane with a sufficient inhibition efficiency (i.e., 100 %) against E. coli bacteria in a short-time contact (i.e., 3 min). Thereby, the tight composite ultrafiltration membrane constructed by fast co-deposition of dopamine and tobramycin established an unparalleled platform technology for sustainable resource recovery (dyes or salts) from high-salinity textile wastewater.

Diabetic wounds pose a significant challenge due to their susceptibility to bacterial infection in a high-glucose environment, which impedes the wound healing process. To address this issue, there is a pressing need to develop suitable hydrogels that can promote the regeneration of diabetic wounds in clinical practice. In this study, we designed and fabricated a highly stretchable, adhesive, transparent, and antibacterial hydrogel through a one-pot radical polymerization of N -[Tris (hydroxymethyl) methyl] acrylamide (THMA) and acrylic acid (AA), and with chitosan and the antibiotic tobramycin as the dynamic physical crosslinkers. The copolymer contains a large number of carboxyl and hydroxyl groups , which can form an interpenetrating network structure with chitosan and tobramycin through multiple dynamic non-covalent bonds. This hydrogel exhibited over 1600 % elongation through an energy dissipation mechanism and strong adhesion to various surfaces without any chemical reaction. In vivo, studies conducted on a staphylococcus aureus -infected full-thickness diabetic skin wound model demonstrated that the hydrogel loaded with tobramycin as one of the crosslinkers had a long-lasting antibacterial activity and effectively accelerated wound healing. Therefore, the antibiotic-loaded adhesive hydrogel we proposed holds great promise as a treatment for bacteria-infected diabetic wounds.

TOB aptamer can be adsorbed on the AuNPs surface to form AuNPs-aptamer complexation to prevent AuNPs aggregation in high salt solution. When TOB was added to the AuNPs solution, the aptamer would bind with TOB and depart from the AuNPs surface. The amount of the AuNPs-aptamer complexation depends on the TOB concentration. Different concentration of AuNPs-aptamer can catalyze the reduction reaction of CuSO 4 to produce different size Cu 2 O particle. The resonance scattering peak intensities are correlated with the Cu 2 O size. Large size Cu 2 O particle as a resonance scattering spectroscopy probe can remarkable improve the TOB detection sensitivity. We have succeeded to detect the trace TOB in aqueous solutions. The linear range and limit of detection were 0.50–17 nM and 0.19 nM, respectively. This simple and inexpensive method exhibited high sensitivity and selectivity, which was successfully used to detect TOB in milk. The results indicated the accuracy and precision were satisfied.

Tobramycin (TOB) is a widely used aminoglycoside antibiotic for treating human and animal diseases. However, its overuse poses a threat to human health, necessitating the development of a rapid and simple detection method. In this study, polyadenine-aptamer-stabilized gold nanoclusters (A x -Apt-AuNCs) were synthesized, to investigate the impact of different polyadenine lengths on their properties. A 20 -Apt-AuNCs demonstrated efficient catalytic activity in the oxidation of o -phenylenediamine to 2,3-diaminophenazine (DAP) in the presence of hydrogen peroxide, resulting in yellow fluorescence emission. Upon binding specifically to the TOB aptamer on A 20 -Apt-AuNCs, TOB enhanced both their peroxidase-like activity and the fluorescence intensity of DAP. Based on this mechanism, a fluorescence-enhanced aptasensor was developed for TOB detection. The aptasensor exhibited a linear detection range of 10.0 nM to 1.0 μM, with a detection limit of 2.63 nM. Furthermore, its application in real sample analysis produced satisfactory results.

Two aminoglycoside antibiotics (AGs), gentamicin (GEN) and tobramycin (TOB), have good antibacterial activity against most pseudomonas aeruginosa and staphylococcus. The molecular structure of these drugs lack chromogenic groups, which brings challenges to their detection. In this project, the detecting methods for GEN and TOB utilizing surface-enhanced Raman spectroscopy (SERS) based on γ -Al 2 O 3 -modified silver nanoparticles (AgNPs) coated with bovine serum albumin (BSA) were established. The enhancement factors (EFs) of GEN and TOB were 2.44 × 10 5 and 2.67 × 10 6 , respectively. The transmission electron microscopy (TEM), X-ray diffraction (XRD), thermogravimetric analysis (TGA) and spectrophotometric techniques were used to characterize the substrate and the combination of the substance with drugs. The pH, the addition amounts for the substrate and coagulant, as well as the mixing time were optimized. On the basis of single factor experiments, a more scientific response surface model was established. The concentrations of GEN and TOB showed good linear relationships with their Raman signals in the ranges of 6.67 × 10 −8 – 2.00 × 10 −6 and 6.67 × 10 −9 – 3.00 × 10 −7 mol L −1 respectively. The limits of detection (LODs) were 11.88 and 1.26 nmol L −1 for GEN and TOB, respectively. The methods were used successfully for the samples determination of the two AGs in commercial drugs and meat products.

Background Tobramycin (TOB) is widely used in livestock and aquaculture, but its excessive use leads to residues in animal-derived foods and the environment, threatening human health. Rapid on-site detection methods are urgently needed to ensure food safety and environmental surveillance. Results We synthesized sulfur and nitrogen co-doped dual-emission carbon quantum dots (S,N-CQDs) with emission at 340 nm (carbon core) and 620 nm (surface state). A polyadenine-extended TOB aptamer selectively quenched the 620 nm emission via electrostatic interaction-induced aggregation, while the 340 nm emission served as stable internal reference. Upon TOB addition, specific aptamer–target binding restored the 620 nm fluorescence, enabling ratiometric detection. Smartphone based dual and single-light-source portable platforms were further developed for visual on-site detection via RGB analysis. The three methods exhibited linear ranges of 10.0 nM–1.0 μM, detection limits of 2.14, 8.02, and 4.87 nM, and recoveries of 95.2%–99.4% in spiked milk and river water. Significance This work presents an integrated “detection–visualization” strategy combining a ratiometric fluorescence aptasensor with smartphone-based portable platforms. The method offers several distinct advantages: (1) the ratiometric design with built-in self-calibration ensures reliable detection in complex matrices; (2) the portable platforms enable rapid, user-friendly, and equipment-free on-site analysis; (3) the entire sensing system is cost-effective and does not require sophisticated instrumentation. These features make our approach a promising tool for routine monitoring of antibiotic residues in food safety and environmental protection.
