Accurate and quantitative analysis of breast cancer biomarkers is a particularly requisite in the early diagnosis of breast cancers, especially in real samples. Here, we develop a surface enhanced Raman scattering (SERS) immunoassay biosensor based on a microfluidic chip for the simultaneous detection of multiple breast cancer biomarkers in real samples. In such an immunoassay, immuno-Ag aggregates are labeled with different Raman reporters to detect multiple breast cancer biomarkers in real samples. Moreover, to better enhance the Raman signals, self-assembled silver nanoparticles are introduced on the bottom of the microfluidic channels to obtain the SERS active immune-substrate. In order to avoid the crosstalk of SERS signals, we designed a spatially separated detection biosensor by using multi-channels in a microfluidic chip. In the experiments, CA153, CA125 and CEA are selected as the models of breast cancer biomarkers, which can be simultaneously detected of in human serum with an excellently specificity and a high sensitivity. Furthermore, the proposed SERS microfluidic biosensor can be applied to detect the biomarkers in real samples using a linear regression method for the statistical analysis. Our detection results are in agreement with those obtained using commercial ELISA kits, which proves the reliability of our SERS-based microfluidic immunoassay.

Breast cancer cell-derived exosomes have high potential as biomarkers for continuous biopsies and longitudinal monitoring in breast cancer. However, it is extremely difficult to separate exosomes with high recovery and high purity from complex media, such as urine, plasma, saliva and cell culture supernatants. Here, we designed a flexible and simple microfluidic chip for exosome separation. The capture zone of the chip is a three-dimensional structure of interlaced cylinders doped with nickel powder. Exosomes were separated from cell culture supernatant by the immunomagnetic separation method in continuous flow mode and were detected by fluorescence imaging with high sensitivity. The chip achieved a high exosome recovery rate (> 74%) and purity (> 67%) at an injection rate of 3.6 mL/h. Thus, this chip was demonstrated to be a cutting-edge platform for the separation and detection of exosomes. It could also be applied to separate and detect other types of exosomes, microbubbles and cells.

Precise understanding of interfacial metal−hydrogen interactions, especially under in operando conditions, is crucial to advancing the application of metal catalysts in clean energy technologies. To this end, while Pd-based catalysts are widely utilized for electrochemical hydrogen production and hydrogenation, the interaction of Pd with hydrogen during active electrochemical processes is complex, distinct from most other metals, and yet to be clarified. In this report, the hydrogen surface adsorption and sub-surface absorption (phase transition) features of Pd and its alloy nanocatalysts are identified and quantified under operando electrocatalytic conditions via on-chip electrical transport measurements, and the competitive relationship between electrochemical carbon dioxide reduction (CO 2 RR) and hydrogen sorption kinetics is investigated. Systematic dynamic and steady-state evaluations reveal the key impacts of local electrolyte environment (such as proton donors with different p K a ) on the hydrogen sorption kinetics during CO 2 RR, which offer additional insights into the electrochemical interfaces and optimization of the catalytic systems.

Keratinocytes and fibroblasts, derived from hiPSCs, were used to construct the human epidermal model by a culture patch made by monolayer poly-(lactic-co-glycolic acid) (PLGA) nanofibers and a human skin-on-a-chip device. Unlike the conventional culture dish method, two different epidermal cells are successfully adhered to the front and back sides of the patch, which produces a three-dimensional nanofibrous scaffold similar to a natural extracellular matrix before the patch was cultured in the skin-on-a-chip device to mimic the physiological conditions of human skin. As expected, the differentiated hiPSCs show the expression of keratinocyte- and fibroblast- specific proteins on the patch, and the layering is found between these two kinds of cells, indicating that this approach creates a powerful in vitro system for modeling skin development and diseases.

Existing single-signal-response fluorescent materials for biogenic amine detection have unsatisfactory detection performance. Herein, we developed a simple pH-sensitive aggregation-induced emission-active fluorogen (AIEgen) and demonstrated its potential for ratiometric monitoring of biogenic amines. This AIEgen, namely 4-(dimethylamino)styryl)quinoxalin-2(1 H )-one (ASQ), shows a typical donor (D)-acceptor (A) structure with nonmonotonic color and fluorescence responses to pH stimulation by its protonation/deprotonation on the D/A moieties. Subsequently, the H + ASQ-labeled paper chips were successfully prepared by physically depositing ASQ on qualitative filter paper, followed by the acidification by hydrochloric acid. Upon the exposure to ammonia vapour, the obtained H + ASQ-labeled paper chips exhibited obvious color and ratiometric fluorescence responses owing to the deprotonation of H + ASQ indicators, indicating that the proposed paper chips can be used for the detection of biological amines by bimodal colorimetry and ratiometric fluorescence. Under the optimal conditions, H + ASQ-labeled paper chip achieved the real-time, non-destructive, and visual monitoring of meat and seafood freshness by simultaneously quantify the biogenic amine index with high performance liquid chromatography (HPLC). The results of this work suggest that ratiometric AIEgen can serve as a promising alternative strategy to ensure accurate monitoring of biogenic amines and food freshness.

In this work, zinc-doped carbon dots (Zn-CDs) were anchored on a three-dimensional wheel type paper-based microfluidic chip, and were decorated with 6-mercaptonicotinic acid (MNA) and L-cysteine (L-Cys) for highly sensitive and rapid fluorescence detection of Cu2+. Zn-CDs were first anchored on paper through the amide bonds between the carboxyl groups of the Zn-CDs and the amino groups of the paper. Afterwards, Zn-CDs were decorated with MNA and L-Cys, effectively preventing the Zn-CDs from aggregation. The nitrogen atom on the pyridine ring and the carboxylic acid groups in MNA and L-Cys coordinated with Cu2+ to form a nonfluorescent ground-state complex, causing the fluorescence quenching of the Zn-CDs. The three-dimensional rotary design could simplify the operation process and achieve simultaneous analysis of multiple samples with different concentrations. Under optimal conditions, the fluorescent sensor exhibits linear response for the determination of Cu2+ in the range from 0.1 to 60 μg L−1 with the detection limit (LOD) of 0.018 μg L−1. The proposed strategy provides a novel way for the highly sensitive detection of Cu2+ in a complex water environment.

The purpose of this study was to construct a delivery system using a microfluidic chip to protect procyanidins (PCs) and to achieve their pH-controlled release in simulated gastrointestinal fluid. The microfluidic chip was designed and fabricated to generate water-in-water-in-oil (W/W/O) templates for the preparation of sodium alginate/chitosan microparticles with a uniform size and core–shell structure, using an internal–external gelation method. Compared with free PCs, the stability of PCs embedded in microparticles was improved and a pH stimulus-responsive release of PCs from microparticles was observed under neutral pH conditions. The delivery system of microparticles was nontoxic and showed an inhibitory effect on the decrease of mitochondrial membrane potential in Caco-2 cells caused by H2O2 and acrylamide. This work provided a method for fabricating compact microfluidic chips to prepare a pH stimulus-responsive PCs delivery system with improved stability, which may have potential applications in the delivery of other nutrients.

The detection of phospholipids oxidation is important for meat control and disease prevention. In this paper, a photoelectrochemical sensor based on printable mesoscopic chip (PMC) for fast and real-time monitoring phospholipids oxidation was designed and fabricated. TiO 2 , ZrO 2 and carbon films of PMC were screen-printed onto the FTO glass layer by layer. The PMC and the feasibility for determination of phospholipids oxidation were investigated by scanning electron microscope (SEM), UV– vis spectroscopy, cyclic voltammograms (CVs) and electrochemical impedance spectroscopy (EIS), etc. The short circuit current (J sc ) was used as a signal current, which would decrease if phospholipids in PMC were undergoing oxidation for the change of electrical properties. Compared with other methods, phospholipids in PMC did not require pretreatment, and the process was nondestructive and real-time. Meanwhile, this method showed high sensitivity and good selectivity. The fabricating process of PMC is simple, and the costs are low, relatively.

Liquid crystal (LC) droplets have been widely used in the detection of chemical and biological species due to their large surface area-to-volume ratio and stimuli-responsive configuration transitions. However, the wide size distribution and high mobility of LC droplets in current LC droplet-based platforms have limited their real-time and quantitative detection applications. Here, we report a novel monodisperse LC droplet-based polydimethylsiloxane (PDMS) microchip, in which monodisperse LC droplets are in situ generated and trapped on a single microfluidic device. A flow-focusing element and a hydrodynamic-based microstructure are designed for the generation and entrapment of monodisperse LC droplets, respectively. This miniatured monodisperse LC droplet-based microchip containing a fixed number of monodisperse LC droplets allows us to quantitatively and rapidly (<4 min) detect bile acids in real time by monitoring the collapse of LC droplets under a polarized optical microscope with small sample consumption (~1.5 μL). The detection limits for cholic acid and deoxycholic acid are 10 μM and 1 μM, respectively. This study provides a way to develop simple and portable LC droplet-based sensing devices for the quantitative, rapid and real-time detection applications.

The effects of organic and inorganic additives on crystal morphology have been extensively studied in various materials. However, the basic fundamentals behind the morphology control are still under debate due to the differences in experimental observations and lack of technical means to control the microenvironments where the crystal grows. Microfluidic technology with convenient operation and precise control of related parameters provides a unique tool for the investigation of crystallization processes on the micrometer and even nanometer scale. A concentration-gradient microfluidic chip was designed to screen the ion compositions to investigate the factors influencing the crystal morphology, and calcium carbonate was chosen for this study. Unexpectedly, the CO32–:Ca2+ ratio is discovered to induce a calcite morphology transition from regular rhombohedral to irregular rhombohedral and nanoparticle aggregations. In contrast, the morphology transition is believed to be caused by variations of the Mg:Ca ratio and supersaturation. The incorporation of different mole ratios of magnesium ions is also found to give different morphologies. The experimentally observed crystal morphology transition indicates that the crystal morphology is influenced by the CO32–:Ca2+ ratio when magnesium ions are present in the system. The proposed microfluidic method, providing an exquisite control of microenvironments during crystallization, can be applied in other materials to analyze the origin of unique morphologies. In addition, the discovery extends our understanding of the morphology control of biominerals.

A new method for fishing antitumor ingredients by G-quadruplex recognition from Macleaya cordata seeds extracts was established using a three-phase-laminar-flow-chip (TPL chip). The TPL chip integrated the separation of drugs from the complex ingredients and organic solvent extraction, simplifying pretreatment processes and reducing reagents and time. In addition, the chip method showed a lower false negative result, owing to the gentle membrane-free filtration process based on diffusion separation in microchannel. Four ligands with high content in alkaloids of Macleaya cordata seeds were selected, those are chelerythrine (CHE), sanguinarine (SAN), protopine (PRO), and allocryptopine (ALL), which demonstrated affinity with G-quadruplex and were potential for antitumor.

Herein, we developed a novel nanosenor to detect trace amount of melamine (M) in the milk using aptamer-modified surface-enhanced Raman scattering (SERS) nanosensor and oligonucleotide chip. The SERS nanosensor was prepared by attaching Raman tag and ploy-thymine (T) aptamer to the gold nanoparticles, respectively. SERS nanosensor can be absorbed to the oligonucleotide chip with a high specificity where a “T–M–T” structure via multi-hydrogen-bond could be formed between the melamine molecule and the thymine molecule. In the measurement, the Raman signal intensities obtained from the chip increase when the concentrations of melamine in solutions increase. The proposed method allows us to detect melamine with a limit of 1.0 ppt (1.0 pg mL −1 ), which is fall far below the strictest safety limit. Compared with the previous methods, the sensitivity of this sensor shows about 100-fold improvement. The advantages of the present sensor are its low detection cost and the simplified sample pretreatment. Furthermore, the reliable and enough accurate results have been obtained by the using of the proposed nanosensor in the assay of trace melamine in real milk sample.
