The extensive use of tetracycline antibiotics (TCs) renders their widespread in the environment that may cause bacterial resistance. Thus, detection of residual TCs and understanding of their broader roles and impacts in the environment and soil ecology are is of critical significance. This paper describes the investigation of a Mg (II)-specific fluorescent micro-probe (PCC–Mg) based on cyclotriphosphazene (Cpz) and curcumin for classical TC•HCl sensing with a low detection limit of 29 nM and a fast response time of less than 10 s. The sensing mechanism is presumably attributed to specific Mg 2+ binding with TCs since the fluorescence enhancement is not observed in the absence of Mg 2+ and not influenced by the presence of several metal ions and anions. The binding of Mg 2+ to curcumin/TC and the formation of TC-metal-curcumin ternary complex can be concluded with NMR and mass spectrometry . Moreover, for the purpose of easy applications, test strips are made to render visual detection of TC with noticeable and selective fluorescence enhancement. The formation of the ternary complex between Cur from the popular herb Curcuma longa and TC from Streptomyces spp. mediated by metal ions also hints a potential role of microbiolochemical-phytochemical interactions in maintaining biodiversity and equilibria in soil microecology.

Introduction Alzheimer’s disease (AD), a neurodegenerative condition, stands as the most prevalent form of dementia. Its complex pathological mechanisms and the formidable blood-brain barrier (BBB) pose significant challenges to current treatment approaches. Oxidative stress is recognized as a central factor in AD, underscoring the importance of antioxidative strategies in its treatment. In this study, we developed a novel brain-targeted nanoparticle, Ce/Zr-MOF@Cur-Lf, for AD therapy. Methods Layer-by-layer self-assembly technology was used to prepare Ce/Zr-MOF@Cur-Lf. In addition, the effect on the intracellular reactive oxygen species level, the uptake effect by PC12 and bEnd.3 cells and the in vitro BBB permeation effect were investigated. Finally, the mouse AD model was established by intrahippocampal injection of Aβ1-42, and the in vivo biodistribution, AD therapeutic effect and biosafety of the nanoparticles were researched at the animal level. Results As anticipated, Ce/Zr-MOF@Cur-Lf demonstrated efficient BBB penetration and uptake by PC12 cells, leading to attenuation of H2O2-induced oxidative damage. Moreover, intravenous administration of Ce/Zr-MOF@Cur-Lf resulted in rapid brain access and improvement of various pathological features of AD, including neuronal damage, amyloid-β deposition, dysregulated central cholinergic system, oxidative stress, and neuroinflammation. Conclusion Overall, Ce/Zr-MOF@Cur-Lf represents a promising approach for precise brain targeting and multi-target mechanisms in AD therapy, potentially serving as a viable option for future clinical treatment.

Renewable polymeric microcapsules hold promise for precisely controlled release of hydrophobic drugs (curcumin) in therapeutic treatment. However, there still remains challenges to stabilize the properties of microcapsules and to enhance encapsulation efficiencies of curcumin. By using an eco-friendly ternary deep eutectic solvent here, we successfully fabricated native cellulose nanofibrils, holocellulose nanofibrils, and lignocellulose nanofibrils containing both phytates and sulfate groups. In addition, a novel type of curcumin-loaded polymeric microcapsules with diameter of 10–20 μm were prepared by cellulose nanofibrils as stabilizing agents in Polylactide Pickering emulsion. The Pickering emulsions stabilized by P/N/S-containing cellulose nanofibrils, holocellulose nanofibrils, and lignocellulose nanofibrils are more stable than the unmodified cellulose nanofibrils-stabilized emulsion. The Pickering emulsion stabilized by 0.2 wt% P/N/S-containing lignocellulose nanofibrils exhibited the highest emulsion layer height and the zeta potential reached −39.74 ± 2.88 mV. Moreover, the highest encapsulation efficiency of curcumin can be up to 94.80%. Antimicrobial test revealed that curcumin in the polymeric microcapsules exhibited an excellent inhibitory effect on S. aureus and the antibacterial efficiency reached 98.66%. These results provided important information on the comparison of different cellulose nanofibrils-stabilized Pickering emulsions and the application of controlled release of curcumin incorporated polymeric microcapsules in drug delivery system.

In the field of skin wound treatment, traditional methods are often limited by the dual challenges of effectively clearing bacterial infections and accelerating wound healing. To address these issues, this study developed an innovative bioactive hydrogel that combines photothermally responsive antimicrobial and immunomodulatory effects. The hydrogel utilized methacrylated gelatin and oxidized dextran, forming a stable network structure through radical and Schiff bonds, loaded with black phosphorus (BP) nanosheets and curcumin (Cur). Characterization of the hydrogel’s physicochemical properties revealed not only its excellent swellability, biodegradability, mechanical strength, thermal stability, and antioxidative properties but also its ability to control the release of Cur through the photothermal effect. Under 808 nm NIR irradiation, the suitable photothermal effect was observed with BP nanosheets at a concentration of 250 μg/mL under a power density of 1.5 W/cm2, and the photothermal conversion efficiency (η) was 41.34%. In vitro experiments showed that the hydrogel exhibited a bactericidal rate of over 90% against E. coli, S. aureus, and P. aeruginosa and more than 80% antibiofilm formation activity. After coculturing with HUVEC cells for 24 and 48 h, cell viability remained above 80%. Cell migration assay confirmed the hydrogel’s effectiveness in promoting cell migration (closure rate reached 97.4% within 24 h). In a rat model with infected wounds, it was shown that on day 14, wounds were nearly healed, with an antibacterial rate of over 85%, the expression levels of proinflammatory factors TNF-α and IFN-γ were reduced, the expression of anti-inflammatory factors IL-4 and TGF-β1 was increased, re-epithelialization was observed, and collagen was deposited, which facilitated the rapid healing of the wounds. The hydrogel in this study with photothermal and immunomodulatory capabilities offers an efficient strategy for treating infected wounds.

As a kind of bioactive component in the rhizome of natural plantCurcuma longaL. (turmeric), curcumin is almost insoluble in water at neutral and acidic pH, which limits its further utilization and development. At the same time, traditional extraction and separation processes typically require the use of a large number of organic solvents. Ionic liquids (ILs) are organic molten salts with melting points below 100 °C. When an ionic liquid exists in a liquid state at or near room temperature, it is referred to as a room-temperature ionic liquid (RTIL). They have a temperature range, good physical and chemical stability, and good structural designability. They have a strong solubilization enhancement effect for many organic compounds. This study first explored the molecular forms of curcumin in ionic liquid aqueous solutions and the intermolecular interactions between curcumin and ionic liquids using spectral analysis and computational chemistry methods; furthermore, using an ionic liquid aqueous solution as an extraction agent, curcumin-like substances (curcuminoids) were extracted from turmeric powders under ultrasound assisted conditions, revealing the relationship between the structure of the ionic liquid and the extraction efficiency. After that, a kinetic study was conducted for the extraction of curcuminoids from turmeric powders, using second-order kinetics fitting to obtain the rate constant and initial extraction rate during the extraction process. Finally, the comparison with a ComplexGAPI tool and antioxidant experiment was performed on the extraction by using ionic liquids and traditional solvent. The full results can provide reference for the design of IL extractants and their application for natural products.

The porous rice starch (PS) and Xanthan gum (XG) are mixed at different ratios to embed the curcumin. The morphology and physicochemical properties of the microcapsules are characterized with scanning electron microscopy (SEM), X-ray diffraction (XRD), differential scanning calorimeter (DSC), and fourier transform infrared spectroscopy (FT-IR); the release of curcumin is evaluated with a simulated gastric-small intestine model. Results indicate that all the microcapsules present high encapsulation efficiency (>90%) with an average particle size of around 3.30 µm. The curcumin loaded in PS and PS/XG microcapsules forms inclusion complexes rather than simple physical mixtures. The release mechanism of curcumin from microcapsules fits first-order ( R 2 > 0.974) and Higuchi ( R 2 > 0.969) models well in gastric and intestinal models, respectively, indicating that encapsulation of curcumin within PS/XG enables controlled release of curcumin. PS/XG wall materials show great potential for encapsulation and delivery of curcumin.

This work established a novel and simple method for quantitative determination of curcumin by developing a “turn-off” fluorescence probe based on upconvert luminescent carbon dots (p-CDs). The carbon dots were synthesized with p-aminobenzoic acid (PABA) and ethanol by solvothermal method and had specific up-conversion luminescence properties which could be applied in other sensing fields. The sensing mechanism of this fluorescent probe was based on the inter filter effect (IFE) between p-CDs and curcumin. As the concentration of curcumin increased, the fluorescence of p-CDs could be selectively quenched. Under the optimal conditions, the fluorescence quenching intensity of p-CDs had a good linear relationship with curcumin in the range of 0.4–45 µΜ and the detection limit was 0.133 µM. In addition, the fluorescent “turn-off” probe constructed with p-CDs exhibited high accuracy and recovery in the analysis of real sample curry powder, indicating that the fluorescence “turn-off” probe had potential application for the detection of curcumin in the complex matrixes.

A biodegradable photodynamic antibacterial film (PS-CF) was prepared using the casting method, with κ-Carrageenan (κ-Car) as the film-forming substrate and curcumin-β-cyclodextrin (Cur-β-CD) complex as photosensitizer. Chilled pork samples were coated with PS-CF and stored at 4 °C to investigate the effects of PS-CF combined with LED light irradiation (425 nm, 45 min) (PS+L+) on pork preservation during 10 days of storage. The total viable count (TVC) of bacteria, total volatile basic nitrogen value (TVB-N) and the pH of pork treated with PS+L+ were all lower than the control, and the water-holding capacity (WHC) was higher. Ten days later, the TVB-N value was 12.35 ± 0.57 mg/100 g and the TVC value was 5.78 ± 0.17 log CFU/g, which was within the acceptable range. Sensory evaluation determined that the color, odor, and overall acceptability of pork treated with PS+L+ were significantly better than the control. These findings suggest that PS+L+ treatment effectively extended the shelf life of chilled pork from ~4–5 to 10 days. Correlation analysis showed that the sensory quality of the chilled pork significantly correlated with total bacterial counts, TVB-N and thiobarbituric acid reactive substances (TBARS) (p< 0.05), suggesting that these biomarkers could be used as standard indicators for evaluating the freshness of chilled pork. These findings demonstrate the effectiveness of Cur-β-CD photodynamic antibacterial film for the preservation of chilled pork and provide a theoretical basis for the application of the film for the preservation of fresh food in general. Keywords:curcumin;storage;chilled pork;photodynamic inactivation

BACKGROUND The low solubility and poor dispersion of alkaline-extracted walnut protein isolate (AWPI) limit its application as a protein-based carrier for the delivery of poorly soluble nutraceuticals, including curcumin. This work investigated the physicochemical characteristics of phosphorylated walnut protein isolate (PWPI) extracted using sodium tripolyphosphate (STP) and evaluated its encapsulation ability. RESULTS The results of phosphorus determination, X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared (FTIR) spectroscopy confirmed the phosphorylation of the extracted PWPI. Circular dichroism (CD) analysis indicated that PWPI contained higher α-helix and lower β-sheet contents than AWPI. The PWPI prepared at pH 9.0 and 11.0 showed significantly improved solubility, similar surface hydrophobicity, and increased surface charges compared to the AWPI. Fluorescence quenching experiments indicated that the binding affinity of curcumin to PWPI was significantly higher than that of AWPI. When bound to PWPI, the solubility of curcumin in aqueous solution was greatly enhanced, with an 8700-fold increase at a nanocomplex concentration of 10 mg mL −1 . The complexation of curcumin with PWPI significantly improved the storage stability of curcumin. Additionally, the PWPI–curcumin nanocomplexes showed significantly increased antioxidant capacity. CONCLUSION Phosphorylated walnut protein isolate showed greatly improved solubility and strong encapsulation ability, making it a promising nanocarrier for curcumin. © 2022 Society of Chemical Industry.

The conventional mono-chemotherapy still suffers from unsatisfied potency for cancer therapy due to tumor heterogeneity and the occurrence of drug resistance. Combination chemotherapy based on the nanosized drug delivery systems (nDDSs) has been developed as a promising platform to circumvent the limitations of mono-chemotherapy. In this work, starting from cisplatin and curcumin (Cur), we prepared a dual drug backboned shattering polymeric nDDS for synergistic chemotherapy. By in situ polymerization of the Cur, platinum (IV) complex-based prodrug monomer (DHP), L-lysine diisocyanate (LDI), and then conjugation with a hydrophilic poly (ethylene glycol) monomethyl ether (mPEG) derivative, a backbone-type platinum (IV) and Cur linkage containing mPEG-poly(platinum-co-Cur)-mPEG (PCPt) copolymer was synthesized. Notably, the platinum (IV) (Pt (IV)) and Cur were incorporated into the hydrophobic segment of PCPt with the fixed drugs loading ratio and high drugs loading content. The batch-to-batch variability could be decreased. The resulting prodrug copolymer then self-assembled into nanoparticles (PCPt NPs) with an average diameter around 100 nm, to formulate a synergetic nDDS. Importantly, PCPt NPs could greatly improve the solubility and stability of Cur. In vitro drug release profiles have demonstrated that PCPt NPs were stable in PBS 7.4, rapid burst release was greatly decreased, and the Pt and Cur release could be largely enhanced under reductive conditions due to the complete dissociation of the hydrophobic main chain of PCPt. In vitro cell viability test indicated that PCPt NPs were efficient synergistic chemotherapy units. Moreover, PCPt NPs were synergistic for cisplatin-resistant cell lines A549/DDP cells, and they exhibited excellent reversal ability of tumor resistance to cisplatin. This work provides a promising strategy for the design and synthesis of nDDS for combination chemotherapy. Keywords:nanosized drug delivery system;dual drug;backboned;combination chemotherapy;polyprodrug
