Characterizing metabolites and biotransformation in lactic acid bacteria fermented Millennium cherry tomato juice using integrated analytical tools

Huixian Wang,Patthanan Sakda,Le Chen,Jingkai Xie,Jiahe Yu,Bingshu Liu,Wanwipa Vongsangnak,Ruimin Wang

Fermentation with lactic acid bacteria (LAB) enhances the nutritional value and flavor of food matrices. This study investigated the impact of Pediococcus acidilactici HNU168, Bifidobacterium animalis subsp. lactis HNU329, and their combined fermentation on Millennium cherry tomato juice (MFJ). Fermentation boosted organic acids, total flavonoid content (TFC), xanthine oxidase (XOD) inhibitory activity, and antioxidant activities. Mixed-strain fermentation significantly increased antioxidant activity and lycopene content beyond single-strain fermentation. Non-targeted metabolomics identified 745 differential metabolites, mainly enriched in purine metabolism and phenylpropanoid biosynthetic pathways, facilitating key metabolite biotransformation. Notably, lycoperodine l, 3-(4-hydroxyphenyl) lactate, indole-3-lactic acid, and dihydrocoumarin contents increased significantly. Molecular docking and XOD inhibitory activity confirmed strong interactions between key metabolites and XOD. Also, electronic tongue analysis showed that LAB-fermented MFJ had increased richness and reduced bitterness and astringency. These results illustrate that LAB fermentation improves MFJ's nutrition, bioactivities, and sensory properties, offering potential applications in functional or anti-hyperuricemia foods.

Development of biodegradable multifunctional zein-curcumin-quercetin-chondroitin sulfate nanoparticles with cold plasma-modified chitosan-based active film for tilapia preservation

Tingting Yang,Xinming Zhang,Feifei Wang,Liming Zhang,Dawei Yu,Xueying Zhang,Guanghua Xia,Jiamei Wang

Cold plasma (CP) modification, combined with nanotechnology, has demonstrated significant potential for enhancing the performance of bio-based packaging films. In this study, zein-curcumin-quercetin-chondroitin sulfate nanoparticles (ZCQC NPs) were incorporated into a chitosan film and modified by CP (0, 10, 30, 50 kV) to enhance the physicochemical properties, bioactivity, and preservation ability of the chitosan-based film. Fourier transforms infrared spectrometer (FT-IR) and X-ray diffraction (XRD) analyses confirmed the presence of hydrogen bonding and electrostatic interactions between the NPs and chitosan, as well as their good compatibility. Thermogravimetric analysis revealed that CP treatment synergistically enhanced the thermal stability of NPs-incorporated composite films. The incorporation of CP and NPs enhanced the film's mechanical strength, hydrophobicity, and barrier performance synergistically. Among them, the CP-C-NPs 30 kV film exhibited lower water vapor permeability (1.33 ± 0.15 g/m·d) and oxygen permeability (4.16 ± 0.27 × 10 −6  g/m·d·Pa), along with excellent antioxidant and antibacterial properties. More importantly, the CP-C-NPs 30 kV film significantly reduced the total viable count (13.64 % compared to the control) and the degree of lipid oxidation (TBARS value decreased by 42.86 %) in packaged fish, extending the shelf life at 4 °C to 8 days, which was twice that of ordinary packaging film. This study not only reveals the synergistic enhancement mechanism of CP modification on the performance of active packaging materials but also provides new strategies for developing efficient food preservation materials.

Quaternary-ammonium chitosan/Sericin-modified carbon nanotubes Photothermal coatings: A reusable and sustainable chemical-free seed disinfection strategy

Xianrui Chen,Lin Wang,Lihong Lins,Chen Ma,Zhenzhen Wu,Nitong Bu,Di Zhang,Wei Liu,Jie Pang,To Ngai

The increasing global demand for food and the adverse environmental impacts of excessive agrochemical use highlights the urgent need for sustainable and scalable seed treatment technologies. This paper reports a novel photothermal seed coating (QC@SCCNTs) with high biocompatibility, exceptional photothermal efficiency, and notable reusability, serving as an effective alternative to conventional chemical treatments. The coating consists of sericin-functionalized carboxylated carbon nanotubes (SCCNTs) electrostatically complexed with quaternary ammonium chitosan (QC), forming a composite film (QS film). Under 808 nm near-infrared (NIR) irradiation, QC@SCCNTs demonstrated antimicrobial efficacies exceeding 97 % against six pathogenic microorganisms, including prominent pathogens such as Aspergillus flavus and Fusarium solani. Applied to red kidney bean and pakchoi seeds, the coating effectively eliminated microbial contamination, increasing the germination rate of red kidney beans from 55.56 % to 91.67 %, and promoting seedling height from 14.48 cm to 41.01 cm. The QS film exhibited exceptionally gas barrier properties and mechanical strength, and completely degraded within just three days of soil burial. The coating retained antimicrobial performance over two reuse cycles after removal and washing. These findings demonstrate the potential of QC@SCCNTs as a biodegradable, reuseable, and scalable materials for safe seed protection and storage in sustainable agriculture.

Curcumin-based antibacterial waterborne polyurethane/gelatin composite film for pork preservation and freshness indication

Yuye Zhong,Zhenhong Zhang,Zhengjie Chen,Wenshuo Zhang,Xinghai Liu,Pingping Zhao,Tao Zhang,Houbin Li

Gelatin possesses excellent film-forming ability, biodegradability, and biocompatibility, which makes it a promising alternative to petroleum-based plastics. However, its application in food packaging is limited by its brittleness, high water sensitivity, and insufficient microbial protection. In this study, curcumin was introduced as a chain extender into castor oil-based waterborne polyurethane (CWU), which was then blended with gelatin as a functional material to create reactive food composite films (CWGL). The effects of varying CWU concentrations on the microstructure and properties of the films were thoroughly investigated. The incorporation of CWU enhanced the mechanical properties, water solubility, water vapor barrier, oxygen barrier, and UV shielding capabilities of the gelatin-based composite films. Additionally, CWU imparted antioxidant, antibacterial, and pH-responsive properties to the CWGL. Packaging application tests demonstrated that CWGL could extend the shelf life of fresh pork up to 10 days under refrigeration. Furthermore, CWGL exhibited a visible color change (∆E > 5) during pork storage, suggesting its potential use as a freshness-indicating innovative packaging. This study presents a novel strategy to advance the application of gelatin in innovative food packaging.

Tissue-like Soft Dual-Network Hydrogel Patch with Instant/Tunable Adhesion and Microenvironment Modulation for Infected Wound Healing

Yan Li,Zihan Sun,Shirong Chai,Zhuo Zhang,Yang Li,Chenhui Zhu,Daidi Fan

The disorganized microenvironment driven by bacterial infection is a key factor in delayed wound healing. Additionally, uncontrolled adhesion between dressings and wound sites inevitably occurs during long-term treatment, resulting in risks of wound exposure or secondary damage. Here, we designed a dual-network hydrogel patch (CHI(C)-F/N-A) based on an orthogonal cross-linking mechanism with instant/tunable adhesion properties and the function of synergistically modulating the microenvironment, which is designed to address these complex matters. The covalent network (N-A) consisting of N-isopropylacrylamide and acrylamide serves as the basic framework of the hydrogel, providing highly conformal and stable integration between the hydrogel and the tissue through abundant polar interactions, while the phase-transition property endows the hydrogel with the capability of weakening the adhesion strength via thermal stimulation. The noncovalent network (CHI(C)-F) formed by the coordination of caffeic acid-modified chitosan with Fe3+ integrates the outstanding antibacterial and antioxidant efficacy of alkaline polysaccharides and natural polyphenols, realizing synergistic modulation of the pathological microenvironment, thereby facilitating an overall wound healing. Notably, benefiting from the superior photothermal effect of polyphenol–metal coordination, the strategy of NIR-mediated regulation of the hydrogel adhesion strength was further extended. This hydrogel patch establishes a rational paradigm for wound dressings that couples satisfactory physical properties and bioactivities expected to surmount the difficulties of practical therapy for infected wounds.

Hydrogen Therapy and Lipopolysaccharide Trapping with MgB2@Inulin Gel for Reversing Colitis and Its Associated Psychiatric Complications via the Gut–Brain Axis

Yaoyu Hu,Pengfei Zhu,Junwei Deng,Yi Yu,Qian Chen,Haitao Wu,Zhengbao Zha,Hua Wang,Yan Ma

Individuals diagnosed with inflammatory bowel disease (IBD) frequently encounter dysbiosis of the intestinal microbiota and are predisposed to psychiatric comorbidities, such as depression and anxiety. The development of oral therapeutics that concurrently attenuate intestinal inflammation, modulate gut microbiota, and ameliorate psychiatric symptoms is imperative for effective IBD management. This study introduced an orally administered Inulin gel incorporating magnesium diboride nanosheets (MgB2@Inulin gel). The hydrolysis of MgB2 nanosheets could generate reductive hydrogen gas (H2), which is critical for scavenging excess reactive oxygen/nitrogen species (RONS), and boron ions that neutralize harmful lipopolysaccharides (LPS). Prebiotic Inulin could effectively protect MgB2 nanosheets from gastric acid degradation, ensuring their persistence in the intestinal tract, where it modulated the gut microbiota and preserved intestinal barrier integrity. Notably, MgB2@Inulin gel markedly alleviated anxiety and depressive behaviors in the model of colitis by regulating the gut–brain axis. Thus, this work underscores the pivotal role of the gut–brain axis in the therapeutic approach to IBD and its associated psychiatric manifestations, proposing a straightforward, safe, and efficacious oral hydrogen treatment strategy for IBD and concurrent mental health disorders.

Nanocarbon Supporting Porous Porphyrin-Ru-Functionalized Vascular Grafts for Antioxidative Stress, Anti-inflammation, and Prorepair of Blood Vessel Injury

Ren Jianmei,Aini Guliyaer,Lei Xuelan,Yang Heng,Guo Jiusi,Zhou Hongju,Tan Yuting,Gao Yang,Cheng Chong,Qiu Li,Ma Lang

Vascular grafts are commonly used to treat acute injuries and chronic atherosclerotic diseases of the vasculature. However, the pathological environment of injured vessels is characterized by oxidative stress and severe inflammatory flares, which usually lead to insufficient vascular regeneration and poor pathological remodeling, with far from satisfactory graft results. Here, we innovatively engineered a nanocarbon supporting porous Ru-porphyrin-based nanobiocatalyst functionalized vascular graft (SPPorRu@PCL) via electrospinning technology. Our studies demonstrate that the SPPorRu@PCL has ultrafast and broad-spectrum reactive oxygen species (ROS) scavenging ability due to the highly active π-conjugated Ru–N catalytic site, π–π stacking effect, and porous structure of loaded SPPorRu, which synergistically enhances its electron transfer ability and catalytic kinetics. Strikingly, in vitro cellular experiments demonstrate that the SPPorRu@PCL is effective in alleviating oxidative stress, reducing damage of DNA and mitochondrial, and promoting cell adhesion for human umbilical vein endothelial cells in a high-ROS environment. Implantation of SPPorRu@PCL in vascular-injured rats further demonstrates its superior biocompatibility, anti-inflammatory and provascular repair capabilities. This work provides important insights into the application of the porous nanocarbon and the π-conjugated porphyrin-based Ru–N coordination nanobiocatalyst assembled on nanocarbons in catalytically scavenging ROS and offers new strategies to design high-performance artificial antioxidant functionalized vascular grafts for the treatment of blood vessel injury diseases. β-Cyclodextrin metal-organic framework as a green carrier to improve the dissolution, bioavailability, and liver protective effect of luteolin

Dan Yang,Min Zhao,Yihe Huang,Liwen Chen,Jiqin Fang,Jiaonan Liu,Miao Wang,Chunjie Zhao

The incidence of acetaminophen-induced liver injury has increased, but effective prevention methods are limited. Although luteolin has hepatoprotective activity, its low solubility and bioavailability limit its applications. Cyclodextrin metal-organic frameworks (CD-MOFs) possess 3D-network structures and large inner cavities, which make them excellent carriers of poorly soluble drugs. In this study, we used CD-MOFs as carriers to improve the dissolution of luteolin and assessed their antioxidant activity, bioavailability, and hepatoprotective effects. Luteolin was loaded into β-CD-MOF, γ-CD-MOF, β-CD, and γ-CD, and characterized by powder X-ray diffractometry (PXRD) and thermogravimetric analysis (TGA). Our results showed that luteolin-β-CD-MOF was the most stable. The main driving forces were hydrogen bonds and van der Waals forces, as determined by molecular simulation. The loading capacity of luteolin-β-CD-MOF was 14.67 wt%. Compared to raw luteolin, luteolin-β-CD-MOF exhibited a 4.50-fold increase in dissolution and increased antioxidant activity in vitro . Luteolin-β-CD-MOF increased the bioavailability of luteolin by approximately 4.04- and 11.07-fold in healthy rats and liver injured rats induced by acetaminophen in vivo , respectively. As determined by biochemical analysis, luteolin-β-CD-MOF exhibited a better hepatoprotective effect than raw luteolin in rats with acetaminophen-induced liver injury. This study provides a new approach for preventing acetaminophen-mediated liver damage.

α-Lipoic Acid-Based Nanozyme for Treating Acute Epilepsy

Yage Sun,Xinrui Zhao,Rong Yang,Meng Xiao,Wenguang Liu

Anti-epilepsy drugs (AEDs), a common therapeutic approach for clinically treating epilepsy, suffer from low permeability toward the blood-brain barrier (BBB), side effect risks, and a failure to regulate the pathological microenvironment. These issues hinder the complete control of epilepsy and lead to a vicious cycle of inflammation-epileptogenesis. Herein, an α-lipoic acid (LA)-based nanozyme that loads phenytoin (PHT) and complexes calcium ions (LA@PHT-Ca 2+ ) is developed. This nanozyme combines the functions of delivering AEDs across the BBB and regulating undesirable microenvironments to treat epilepsy. Amphipathic LA self-assembles into stable nanoparticles in water with BBB crossing ability endowed by the transporting of LA through sodium-dependent multivitamin transporter. LA's enzyme-like and redox functions provide LA@PHT-Ca 2+ with excellent scavenging effects on several free radicals via cascade reactions, thereby protecting neurons from oxidative stress. Moreover, Ca 2+ disrupts neuronal respiration, resulting in reduced production of bioenergetic adenosine triphosphate and an improved hypoxic microenvironment. This leads to decreased expression of P-glycoprotein, a drug-efflux gating protein. This novel lipoic acid-based nanozyme, with intrinsic BBB crossing capability, prolonged intracellular drug retention time, and the ability to block the inflammation-epileptogenesis cycle, presents a satisfactory therapeutic effect in epileptic rats, resulting in attenuated symptoms and an improved microenvironment.

Wireless Strain Sensors Based on Sustainable Poly(lipoic acid) Zwitterionic Conductive Biogels with Self-Healing, High Stretchability, and Biodegradability

Li Yang,Haiyan Du,Yichang Cao,Aiqing Zhang,Hui Jia,Dangchao Sun,Huimin Yang,Ying Li

Recently, sustainable poly(lipoic acid) (poly(LA))-based biogels have attracted increasing interest and have been used in wearable sensing fields. However, the low stretchability and adhesion, poor self-healing, and wire transmission remain the major issues that limit the applications of poly(LA)-based gel sensors. It is urgent to develop multifunctional biogels with excellent comprehensive performance. In this work, multifunctional conductive poly(LA)-based zwitterionic biogels (denoted as PLLS gels) were fabricated by introducing hydrophilic sulfobetaine methacrylate (SBMA) through nucleophilic addition reactions with poly(LA). The addition of SBMA endowed the gels with conductivity due to the abundant anionic and cationic groups of the zwitterionic structure. The excellent biocompatibility of poly(LA) and SBMA provided the gels nontoxicity and harmlessness. As expected, the PLLS gels possessed high stretchability, adhesion, and self-healing due to the multiple dynamic bonds, including hydrogen bonds and electrostatic interactions. Besides, the gels exhibited excellent biodegradability, antioxidant, and antibacterial activities. The PLLS gels had found application as a wireless wearable sensor, which could monitor various human activities involving temperature changes, human joint movements, and voice recognition. This work not only provides a valuable strategy for constructing the sustainable gel sensors but also expands the applications of biogels to portable mobile monitoring of wireless wearable devices.

Widely targeted metabolomics-based analysis of the impacts of L. plantarum and L. paracasei fermentation on rosa roxburghii Tratt juice

You Luo,Ruling Tang,Han Qiu,Angxin Song

Rosa roxburghii Tratt fruits (RRT) exhibit extremely high nutritional and medicinal properties due to its unique phytochemical composition. Probiotic fermentation is a common method for processing fruits. Variations in the non-volatile metabolites and bioactivities of RRT juice caused by different lactobacilli are not well understood. Therefore, we aimed to profile the non-volatile components and investigate the impacts of L. plantarum fermentation (LP) and L. paracasei fermentation (LC) on RRT juice (the control, CG). There were both similarities and differences in the effects of LP and LC on RRT juice. Both of the two strains significantly increased the content of total phenolic, total flavonoid, and some bioactive compounds such as 2-hydroxyisocaproic acid, hydroxytyrosol and indole-3-lactic acid in RRT juice. Interestingly, compared with L. paracasei , L. plantarum showed better ability to increase the content of total phenolic and these valuable compounds, as well as certain bioactivities. The antioxidant capacity and α-glucosidase inhibitory activity of RRT juice were notably enhanced after the fermentations, whereas its cholesterol esterase inhibitory activity was reduced significantly. Moreover, a total of 1466 metabolites were identified in the unfermented and fermented RRT juices. There were 278, 251 and 134 differential metabolites in LP vs CG, LC vs CG, LC vs LP, respectively, most of which were upregulated. The key differential metabolites were classified into amino acids and their derivatives, organic acids, nucleotides and their analogues, phenolic acids and alkaloids, which can serve as potential markers for authentication and discrimination between the unfermented and lactobacilli fermented RRT juice samples. The KEGG enrichment analysis uncovered that metabolic pathways, purine metabolism, nucleotide metabolism and ABC transporters contributed mainly to the formation of unique composition of fermented RRT juice. These results provide good coverage of the metabolome of RRT juice in both unfermented and fermented forms and also provide a reference for future research on the processing of RRT or other fruits.

Whey protein nanoparticles: Enhancing solubility, environmental resistance, and bio-accessibility of pterostilbene

Xiuping Liang,David Julian McClements,Zhengyu Jin,Long Chen

Pterostilbene (Pte) is a dietary phenolic compound derived from plants that offers numerous physiological advantages. Nonetheless, its low water solubility and limited oral bioavailability restrict its bioactivity. To address these limitations, nanoparticles were prepared using anti-solvent precipitation method. Optimal nanoparticles were achieved at a 10:1 mass ratio of whey protein isolate (WPI) to Pte, resulting in a particle size of 104.54 nm, a polydispersity index of 0.42, and an impressive encapsulation efficiency of Pte at 85.02%. The water solubility of Pte in the nanoparticles was found to be 53.14 times higher than that of free Pte and remained stable under various environmental conditions. Furthermore, the DPPH and ABTS + free radical scavenging activities of the nanoparticles were maximized at 55.58% and 92.97%, respectively. Pte release from WPI-Pte nanoparticles was well-fitted to Korsmeyer-Peppas model and followed a non-Fickian mechanism controlled by both diffusion and erosion. This environmentally friendly, straightforward, and efficient approach shows promise for large-scale production of Pte products with amplified health benefits.
