Identification and molecular mechanism of novel antioxidant peptide from fish sauce: A combined quantum chemistry and molecular simulation

Chaonan Sun,Yingnan Liu,Shiyang Liu,Ying Bu,Wenhui Zhu,Xuepeng Li

Fish sauce, derived from fermented fish, exhibits a notable antioxidant effect after a six-month fermentation process, and we propose that potential antioxidant peptides were present in the fish sauce. We isolated, purified, and identified potential bioactive antioxidant peptides by using fish sauce fermented for 6 months. Additionally, molecular simulation was employed to investigate the antioxidant action mechanism of these bioactive peptides. The molecular docking results revealed that FS4-1 (MHQLSKK), FS4-2 (VLDNSPER), FS4-3 (MNPPAASIK), FS6-1(VLKQAAAGR), and FS6-2 (SPDVSPRR), could dock with the Keap1 receptor. The primary force (Van der Waals' force and hydrogen bonds) and key sites (GLY509 and ALA510) of Keap1 binding to peptides were determined. The active center was located in the side chain of amino acid Met at positions C7H78 and C7H79. We here identified antioxidant peptides in fish sauce and revealed the antioxidant mechanism through molecular simulations.

In vitro crystallization of single crystalline guanine microplates mediated by proteins extracted from the carp fish Cyprinus carpio

Yujing Ren,Xiubin Hou,Ying Zhang,Jingyan Hao,Juan Gao,Xinbing Song,Yurong Ma

Anhydrous guanine (AG) crystals are widely applied in organisms due to their excellent optical properties. However, the influence of biomolecules on the biomineralization process of guanine crystals in organisms remains unexplored. In this study, mixed proteins from the silver film of carp fish scales were extracted and were used as a model system to elucidate the effect of biomolecules on the guanine crystallization for the first time. The mixed proteins were enriched with amino acids such as Asp, Glu, Leu and Lys. Elongated hexagonal microplates of α-form AG (α-AG) mixed with a small amount of β-form AG (β-AG) mainly exposing (100) plane were obtained in the presence of the mixed proteins under mild condition while irregular particles were formed in the absence of proteins, indicating that the extracted mixed proteins have a strong influence on the formation of AG microplates exposing (100) plane. It is found that the proteins not only adsorb on the surface but also occluded into the AG crystals by using confocal laser scanning microscopy analysis. We also find that guanine analogues have profound synergetic effects on the morphology and polymorphs of guanine in the in-vitro synthetic system. The obtained α-AG exhibit superior optical properties such as high reflectivity, strong depolarization and pearlescent luster. This in-vitro synthesis work shows clearly the strong influence of biomolecules for the biomineralization process of guanine crystals in the organisms.

Michael-Addition-Mediated Photonic Crystals Allow Pretreatment-Free and Label-Free Sensoring of Ciprofloxacin in Fish Farming Water

Yanqiu Song,Jialei Bai,Rong Zhang,Houluo He,Chao Li,Jiang Wang,Shuang Li,Yuan Peng,Baoan Ning,Minglin Wang,Zhixian Gao

Abuse of antibiotics results in a large number of antibiotics residues in the environment and even causes the problem of “super bacteria”. Therefore, it is crucial to develop a powerful analytic method to monitor antibiotics quickly and simply. Photonic crystal (PC), as a sensing material, has promising application prospects. Herein, we try to use PC to realize pretreatment-free and label-free detection of Ciprofloxacin (CF) through Michael addition reaction. The recognition process is carried out by the Michael addition reaction between the piperazine group of CF and the o-benzoquinone group on the PC. The monodisperse microspheres with o-benzoquinone groups are prepared by polymerization and oxidation and then stacked to form PC. During the detection, the peak intensity of the PC decreases with the increasing CF concentration, and the linear range is from 2 to 512 μg/L. The limit of detection (LOD) is 0.76 μg/L. Furthermore, the PC retains 97% of the initial response after storage in a Petri dish at room temperature for 1 month, which shows that it has good stability. Moreover, CF in fish farming water can be detected directly without any pretreatment and label, and the results are in good accordance with the LC-MS-MS results. This Michael-addition-mediated PC is accurate, easily prepared, cost-efficient, and long-term stable. In addition, it is environmentally friendly, because little organic solvent is needed during both the preparation and the detection.

Development of a monoclonal antibody-based indirect competitive chemiluminescence immunoassay for nitrofurantoin metabolite AHD residue in fish samples

Ruimin Lu,Yan Zhang,Daoping Zeng,Qihua Xu,Dan Wang,Peng Dai,Weijie Deng,Lin Kang,Xiaoxiao Ke,Luying Duan,Yanping Hong,Wuying Yang

An ultrasensitive indirect competitive chemiluminescence immunoassay (icCLEIA) based on monoclonal antibodies (mAb) for the effective detection of nitrofurantoin metabolite 1-aminohydantoin was developed under optimal conditions in our study. 1-aminohydantoin (AHD) was derivatized with m-formylphenoxyacetic acid (MCEP) to obtain a hapten MCEPAHD. The developed icCLEIA based on anti-MCEPAHD mAb showed high sensitivity to competing drugs 2-nitrophenyl derivative of AHD (2NPAHD) with a limit of detection (LOD) 0.04 ng/mL and a 50 % inhibitory concentration (IC 50 ) 0.27 ng/mL, and the icCLEIA had an ultra-high specificity for 2NPAHD, with little cross-reactivity with structural analogues other than parent drug nitrofurantoin. The mean recovery of AHD in fish samples was found to be 97.0 % to 118.5 %, with a coefficient of variation of less than 7.0 %. A comparison of the results of ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) with those of icCLEIA demonstrated good linearity. These assay results indicated that the prepared mAb against MCEPAHD and the developed icCLEIA in our research can be used for simple, fast and sensitive detection of AHD residues in animal tissues.

Effects of starters on the quality of fermented fish (Zaoyu): Key microorganisms for coloring, softening, and improving flavor

Jinggui Nie,Sunting Zhu,Xiao Zhang,Dan Wu,Xuxu Li,Qilin Huang

This study aimed to improve the quality of fermented fish Zaoyu by inoculating different starters (Chuzhai starter, Aroma starter and Sweetness starter), and examining the role of key microorganisms. High-throughput sequencing showed the microbial composition of Aroma starter was similar to Sweetness starter, but both were different from Chuzhai starter. Compared to traditional fermentation, inoculated fermentation with Aroma starter and Sweetness starter strengthened the color of Zaoyu by Maillard reaction and softened the muscle by degrading muscle fibers, endomysium and epicardium. Additionally, the taste and odor of Zaoyu were obviously improved, because the proportion of sweet, umami and aroma compounds increased by 2.51 %, 7.09 %, and 22.94 %, respectively. Correlation analysis combined with metabolic functions showed key microorganisms, such as Trichococcus , Rhizopus , Saccharomycopsis , Saccharomyces , etc., improved the quality mainly by promoting sugar production and conversion, and protein degradation. Overall, Aroma starter and Sweetness starter were superior in improving the quality of Zaoyu.

Ke Zhang,Zhihua Li,Wanying Zhao,Jing Guo,Sulafa B.H. Hashim,Suliman Khan,Jiyong Shi,Xiaowei Huang,Xiaobo Zou

A novel colorimetric aerogel was developed by the complexation of carboxymethyl cellulose (CMC), sodium alginate (SA), and black goji anthocyanin (BGA) followed by freeze-drying for monitoring fish (Coho salmon) freshness during storage at 4 °C and 25 °C. The various aerogels (C/S/B3:1, C/S/B2:1, C/S/B1:1, C/S/B1:2, and C/S/B1:3) externally and internally were characterized using SEM, FTIR, XRD, DSC, and TGA. Among them, the aerogel composite C/S/B1:2 exhibited the most uniform pore size, largest specific surface area, rapid color changes in various alkaline vapors (5 μM and 50 μM), and better mechanical strength. Furthermore, the colorimetric aerogel became dark blue from light purple during fish storage at temperatures of 4 °C and 25 °C when it reached pH 7.49 and 7.33, TVC 8.9 × 10 7  CFU/g and 8.5 × 10 7  CFU/g, and TVB-N 33.8 mg/100 g and 26.12 mg/100 g, respectively, indicating fish completely deteriorated. Taken together, the colorimetric aerogel composite C/S/B1:2 was promising for determining fish freshness, which could be utilized as a non-destructive and useful intelligent sensor in monitoring various fish and meat freshness and/or quality.

Active packaging coating based on agarose grafted with benzoic acid derivatives: Preparation, characterization and application in fish preservation

Liling Fu,Yi Ru,Jinming Ye,Qinglin Hong,Huifen Weng,Yonghui Zhang,Jun Chen,Anfeng Xiao,Qiong Xiao

Agarose based on gel strength can not meet the needs of diverse applications, and the structural modification of agarose is helpful to improve its functional properties and promote the economic benefits of the agarose industry chain. This study focuses on making agarose as an active packaging material with specific functions. Agarose was modified with p -hydroxybenzoic acid (Ph) and 3, 4-dihydroxybenzoic acid (Da) to produce agarose derivatives with grafting rates of 4% and 12% (Ph-Ag@4%, Ph-Ag@12%, Da-Ag@4%, and Da-Ag@12%). Characterization techniques confirmed the successful introduction of Ph and Da onto agarose, with grafting occurring at the C-6 position in d -galactose. Both Ph-Ag and Da-Ag exhibited significantly enhanced antioxidant and antibacterial properties. Da-Ag@12% showed remarkable improvements in DPPH and ABTS radical scavenging activities from 0% to 66.01% and 2.49%–84.73%, respectively. Ph-Ag@12% exhibited inhibition rates of 100% against Escherichia coli and Staphylococcus aureus . After modification, Ph-Ag and Da-Ag maintained good gelling and film-forming properties. Fish preservation experiments revealed both Ph-Ag and Da-Ag coatings effectively suppressed microbial growth, slowed down lipid oxidation, minimized water loss, and maintained the texture and appearance of fish meat. These coatings improved fish meat quality and extended its shelf life, with Da-Ag@12% showing the most promising results.

Preparation and characterization of lactoferrin-polyphenol conjugate with stabilizing effects on fish oil high internal phase Pickering emulsions

Ying Sun,Mantong Zhao,Zhongyuan Liu,Haohao Shi,Xueying Zhang,Yongqiang Zhao,Zhenhua Ma,Gang Yu,Guanghua Xia,Xuanri Shen

The combination of protein and polyphenol is an effective approach to improve the stability of protein emulsions. The lactoferrin (LF)-(−)-epigallocatechin-3-gallate (EGCG) covalent complex (LF-EGCG) was first prepared by alkali-induced reaction, then the structure and physicochemical properties between LF-EGCG and non-covalent complex (LF + EGCG) were compared, and finally the stability of complexes to fish oil high internal Pickering emulsions (HIPPEs) was tested. Results showed that LF-EGCG had stronger antioxidant activity, higher thermal stability, and better surface wettability than LF + EGCG. Meanwhile, the complexes showed no cytotoxicity within the tested concentration range (12.5–200 μg/mL). The HIPPEs stabilized with LF-EGCG possessed smaller droplet size, higher ζ-potential, and more uniform oil/water proton distribution. Covalent treatment also enhanced the storage, thermal, freeze-thaw and physical stability of LF HIPPEs. Furthermore, due to the higher antioxidant activity and denser microstructure, LF-EGCG HIPPE can more effectively inhibit the oxidation of fish oil.

Edible Films Based on Fish Gelatin and Soluble Soybean Polysaccharide Enriched with Tea Polyphenol for Active Food Packaging

Jie Liu,Zhongfeng Song,Yiwei Wang,Ying Pei,Keyong Tang

The increasing demand for environmentally friendly alternatives to conventional plastic packaging has driven interest in the development of biodegradable edible films with functional properties. In this work, edible blend films were developed based on fish gelatin (FG), soluble soybean polysaccharide (SSPS), and tea polyphenol (TP) for active food packaging applications. The FG/SSPS/TP films were prepared by solvent casting and characterized in terms of their structural, mechanical, optical, thermal, and barrier properties. FTIR, SEM, and XRD analyses revealed TP-induced morphological and structure changes in the biopolymer matrix. The incorporation of TP significantly enhanced the antioxidant activity and UV-shielding properties of the films, while also modifying their flexibility and surface hydrophilicity. The packaging performance of FG/SSPS/TP films was evaluated using beef tallow as a model food product. Compared to neat FG/SSPS and polyethylene films, the FG/SSPS/TP films effectively suppressed lipid oxidation of the samples during storage. The results demonstrated that the prepared FG/SSPS/TP films possess strong potential for use as edible and active packaging materials for food products.

Control of flavor formation in fish sauce by a rationally designed sunlight-mimicking system on regulating microbial communities

Xiaoru Feng,Xue Wang,Qiyue Zhao,Xianbing Xu,Ming Du,Ling Zhang

Traditional fish sauce fermentation depends heavily on solar radiation, which is subject to fluctuations in climate, geography, and weather, making precise control challenging. Therefore, this study developed a sunlight-mimicking system integrating temperature control and artificial light regulation to ensure the consistent quality of fish sauce and meet the requirements of the modern food industry. Compared with the no-light group, the light-treated group exhibited significantly lower pH levels, higher amino acid nitrogen (AAN) content, and enhanced formation of volatile flavor compounds. Furthermore, Pearson correlation analysis between 33 key volatile compounds and dominant microbial genera revealed that light exposure stimulated the growth of Pantoea , Amylomyces , Kosakonia , and Penicillium , which were positively associated with desirable flavor development. Conversely, light exposure inhibited the growth of Enterococcus , Mucor , and Enterobacter , thereby reducing the production of undesirable flavor compounds and improving the quality of fish sauce. These findings demonstrate that artificial sunlight simulation provides a reliable and controllable alternative to traditional sun-dependent fermentation, supporting the standardization and industrialization of fish sauce fermentation of fish sauce fermentation.

Photoelectrocatalytic Coupling of Chlorine Radicals Enhances Sulfonamide Antibiotic Degradation in Saline-Alkaline Waters in Cold-Water Fish Aquaculture

Qikai Liu,Yang Liu,Yaqi Mao,Ru Li,Yujie Jiao,Jiali Lei,Fenzhen Zhang

The degradation of organic matter using TiO2nanotube photocatalytic technology is limited by the short lifetime and diffusion radius of the generated hydroxyl radicals, decreasing the removal efficiency. This study developed a chlorine radical-coupled photoelectrocatalytic system, significantly enhancing the performance of TiO2nanotube arrays in removing sulfonamide antibiotics (SAs) from cold-water aquaculture systems. The highest degradation rates were observed at 5 mM NaCl and 15 mM NaNO3. When SA concentrations were 0.1–10 mg/L, degradation efficiency decreased with higher initial concentrations. The best degradation was achieved at an initial pH of 3 for SA. Humic acid and sodium acetate, natural organic matter in the water column, served as low-concentration promoters and high-concentration inhibitors. In our study, three degradation intermediates were identified, and hydrolysis and nitration reactions are proposed as the primary pathways for SA degradation. We confirmed that oxygen radicals play a major role in this system. Furthermore, toxicology experiments revealed the weakening of the toxicity of the degraded products. This study provides an efficient method for treating organic matter in cold-water fish culture water in chloride-containing saline and alkaline waters.

Preparation of ionic porous polymers for extraction of four phenolic endocrine disrupting chemicals from fish and water samples

Chengcheng Fan,Chenhuan Wang,Bin Zhao,Zixuan Cai,Qianqian Wang,Lin Hao,Zhi Wang,Qiuhua Wu,Chun Wang

In this paper, series of ionic polymers were synthesized by crosslinking alkyl quaternary ammonium salts with 1,4-bis(chloromethyl)benzene. Among them, hyper-crosslinked polymer fabricated with dodecyl dimethyl benzyl ammonium chloride (HCP-DD) as monomer delivered superior adsorption performance for endocrine disrupting chemicals (EDCs). The adsorption mechanism mainly includes π-π stacking, hydrophobic and electrostatic interaction . With HCP-DD as solid phase extraction sorbent , a high performance liquid chromatography-diode array detection method was developed for the detection of four phenolic EDCs in water and fish samples. The detection limits of the method were 0.005–0.02 ng mL −1 for water samples and 3–30 ng g −1 for fish samples. The recoveries of EDCs in water samples and fish samples were 80–119% and 81.3–117% (relative standard deviations <4.4%), respectively. The study not only provides a route for preparation ionic porous polymers , but also highlights the applications of ionic polymers as efficient adsorbent to enrich organic pollutants .
