The tunable synthesis of orange emissive carbon dots for sensitive and selective detection of Hg2+ via portable paper sensor

Yu Gao,Linan Zhou,Zechang Li,Peng Sun,Qijun Dai,Ruifeng Xie,Fang Chai

Creating a sensitive, accuracy and portable fluorescent probe to monitor Hg 2+ with rapid visual convenient process is highly desirable but remains challenging. Herein, orange emissive carbon dots (O-CDs) were synthesized and optimized by solvothermal method using p-phenylenediamine (p-PDA) as precursor. The O-CDs emitted an orange emission at 594 nm excited at 480 nm, with QY of 23.25% and a long lifetime of 14.43 μs. The O-CDs exhibited sensitive quenching response towards Hg 2+ based on dynamic quenching effect, which facilitated O-CDs utilizing as the fluorescent probe with the limit of detection at 37.6 nM. The O-CDs fulfilled sensing Hg 2+ in environmental water with satisfactory recoveries (90.3%−107.8%) and relative standard deviation (RSD) of less than 3.81%, manifested their excellent environmental tolerance. Notably, the O-CDs based test paper was fabricated and served as a portable sensor with a smartphone assistant for detection of Hg 2+ , which greatly elevated the sensing performance of test paper. This work contributed an innovative approach for the synthesis of O-CDs, as well as a reliable sensitive sensor based on portable test paper for an intelligent readable recognizing system, which could be regarded as promising candidate for real detection of Hg 2+ .

Analyte-enhanced photocatalytic activity of CdSe/ZnS quantum dots for paper-based colorimetric sensing of Hg2+ under visible light

Haiyu Li,Wan Wang,Zhijuan Wang,Qing Lv,Hua Bai,Qing Zhang

CdSe/ZnS quantum dots (QDs) and 3,3,5,5-tetramethylbenzidine (TMB) were incorporated into a filter paper to produce a paper-based colorimetric device for Hg 2+ detection. Under visible light, the CdSe/ZnS QDs acted as a photocatalyst to oxidize TMB, resulting in blue colorization on the paper. Moreover, Hg 2+ could enhance the photocatalytic activity of CdSe/ZnS QDs and consequently caused an increase in the color intensity of the paper. The color change of the paper-based device can be directly identified by naked-eye observation or captured by a smartphone and analyzed using image processing software for quantitative analysis. Under optimal conditions, a low detection limit of 0.09 µM was achieved. This device was applied for real samples, including tap water and cosmetics, and achieved satisfying results.

Sodium Alginate Micelle-Encapsulating Zinc Phthalocyanine Dye-Sensitized Photoelectrochemical Biosensor with CdS as the Photoelectric Material for Hg2+ Detection

Xiaoyan Yang,Mengjie Zhang,Zixuan Chen,Yuwei Bu,Xue Gao,Yongkun Sui,Yueqin Yu

A simple and selective photoelectrochemical (PEC) biosensor was constructed for Hg2+ detection based on zinc phthalocyanine (ZnPc) dye-sensitized CdS using alginate not only as a carrier but also as a binder. First, CdS as a photoactive material was in situ modified on the electrode surface using a rapid and simple electrodeposition to obtain an initial photocurrent signal. Second, ZnPc was loaded in the amphiphilic alginate micelle and then was coated onto the CdS film surface via alginate as the binder. The photocurrent was subsequently enhanced due to the favorable dye sensitization effect of ZnPc to CdS. Finally, the thymine-rich probe DNA was immobilized on the modified ITO surface via coupling reaction between the carbonyl groups of the amphiphilic polymer and the amino groups of the probe DNA. In the presence of Hg2+, the thymine–Hg2+–thymine (T–Hg2+–T) structure was formed due to the specific bond of Hg2+ with thymine, resulting in the decrease of photocurrent due to the increase of steric hindrance on the modified electrode surface. The proposed PEC biosensor for Hg2+ detection possessed a wide linear range from 10 pM to 1.0 μM with a detection limit of 5.7 pM. This biosensor provides a promising platform for detecting other biomolecules of interest.

Preparation of fluorescent bimetallic silver/copper nanoparticles and their utility of dual-mode fluorimetric and colorimetric probe for Hg2+

Jingyu Pang,Ruyan Xie,Sophie Chua,Yu Zou,Mingyu Tang,Fang Zhang,Fang Chai

A dual-mode colorimetric and fluorimetric probe was successfully established based on silver/copper bimetallic nanoparticles (AgCu-BNPs). The AgCu-BNPs were confirmed as individually bimetallic nanoparticles with a mean size of 7.7 ± 0.2 nm, as characterized by high resolution transmission electron microscopy. Intriguingly, the AgCu-BNPs possess both surface plasmon resonances (SPR) and fluorescence emission. AgCu-BNPs emanate bright blue fluorescence with optical emission centered at 442 nm with high quantum yield of 30.3%, and AgCu-BNPs were attenuated or even quenched by Hg 2+ via both static and dynamic quenching, coincidently accompanied by a visible color change, which endow AgCu-BNPs a unique utility as dual-mode colorimetric and fluorimetric probes. The detection limits as low as 89 nM and 9 nM were determined by dual-mode of AgCu-BNPs, respectively. The recovery rates in real samples were found to be 97.3–118.8%, and 89.5–112.7% by colorimetric and fluorescent methods separately, demonstrates the good environmental tolerance of the dual-mode probe.

Hydrothermal synthesis of fluorescent carbon dots from gardenia fruit for sensitive on-off-on detection of Hg2+ and cysteine

Dong Sun,Tiantian Liu,Chunfeng Wang,Lifang Yang,Shengkai Yang,Kelei Zhuo

Nitrogen and sulfur co-doped carbon dots (N/S-CDs) were prepared by a simple hydrothermal method using gardenia fruit as precursor. The N/S-CDs are nearly spherical particles with a size of 2.1 nm and possess excellent fluorescence stability in a wide pH range and high NaCl concentrations, as well as under UV light irradiation . The absolute quantum yield (QY) without any surface modification reaches up to 10.7%. Meanwhile, the N/S-CDs can be quenched by Hg 2+ (turn-off), while the quenched fluorescence can be recovered (turn-on) by introducing cysteine (Cys), with linear ranges of 2–20 μM for Hg 2+ and 0.1–2.0 μM for Cys. On the basis of the on-off-on sensing mode, the N/S-CDs can be used to detect Hg 2+ and Cys. Hence, the N/S-CDs would be a promising sensor in environmental and biological analysis.

Rapid and sensitive colorimetric sensor for H2O2 and Hg2+ detection based on homogeneous iodide with high peroxidase-mimicking activity

Yang Wang,Liujing Xu,Wen Xie

Peroxidase reaction is known as a classical reaction that is widely used in the analytical chemistry field because of not only the important peroxide analytes but also the key role of H 2 O 2 detection in various coupled enzyme systems. For this reason, exploiting appropriate enzymes with good catalytic properties, low cost, easy preparation, and simple operation for the reaction becomes extremely important for acquiring desired analytical performance. Herein, we proposed a simplified, rapid, and universal colorimetric platform based on the peroxidase-mimicking activity of I − for sensing applications. The I − species exhibited the peroxidase-like ability to catalyze the oxidation of colorless 3,3′,5,5′-tetramethylbenzidine (TMB) to blue TMBox in the presence of H 2 O 2 via the I 2 intermediate. Compared with natural bio-enzymes and emerging nanozymes , the homogeneous I − mimic provided excellent activity to trigger the color reaction faster. More importantly, commercial reagents were able to be used directly, thus avoiding the tedious preparation and purification processes required in bio-enzymes and nanozymes. As a result, the fabricated platform could be used to detect H 2 O 2 within 3 min, providing a linear range of 1.7–166.7 μM and a detection limit of 1.1 μM. Given that Hg 2+ could suppress the color reaction through formation HgI 2 , the Hg 2+ target (20–80 nM) was also determined rapidly. Practical measurements demonstrated the reliability of the colorimetric assay . It is believed that the general platform based on the H 2 O 2  + TMB + I − reaction holds broader applications in other analytical systems.

Engineered cells for selective detection and remediation of Hg2+ based on transcription factor MerR regulated cell surface displayed systems

Dan Wang,Yanan Zheng,Lina Xu,Xiaosu Fan,Na Wei,Nanhao Jin,Shan Huang,Qi Xiao,Zihua Wu

Mercury pollution has been one of the most serious environmental problems today. The detection and remediation of mercury contamination are of special concern due to its nondegradable nature, bioaccumulation effect, and significant toxicity. In this research, the whole-cell based detection and remediation systems for Hg 2+ were engineered based on the cell surface engineering technique and transcriptional activation mechanism of metalloprotein MerR. The engineered biodetection system showed high selectivity for Hg 2+ over other metal ions with the detection limit as low as 0.1 nM. It also permitted a rapid, visual, and on-site detection of bioavailable Hg 2+ for large-scale water samples with simple processes. The engineered bioremediation system, which was more efficient than the most current biosystems with limited adsorption capacities, could continuously reduce mercury contamination in wide concentration by transforming highly toxic Hg 2+ to volatile and much less deleterious Hg 0 with extraordinary selectivity. In addition, the bioremediation system could perform function in the absence of extra inductive agent and showed an increased protection of cells against Hg 2+ biotoxicity. By fully imitating and utilizing the microorganisms, the engineered biochemical systems have great potential in the practical application of detecting and remediating Hg 2+ in the contaminated aquatic systems.

An ultrasensitive competitive chemiluminescence immunosensor coupled flow injection cell modified by oxidized graphene-chitosan for the detection of Hg2+

Xinchun Zhou,Shihua Zhang,Jing Shi,Kang Zhao,Anping Deng,Jianguo Li

In this work, an ultra-sensitive and simple chemiluminescence (CL) immunosensor has been constructed to detect Hg 2+ in real water samples. It is based on the excellent properties of oxidized graphene and chitosan modified a novel flow cell to achieve satisfactory results. Oxidized graphene has good biocompatibility, can effectively immobilize antigens and construct an efficient biosensor interface. Chitosan has excellent biodegradability , excellent film-forming and biocompatibility. Hereby, oxidized graphene-chitosan composites provide a favorable microenvironment for immobilizing coating-antigen and introducing HRP-labeled secondary antibodies into the CL reaction of luminol-PIP-H 2 O 2 . With a competitive-type assay mode, the Hg 2+ in solution competed with the immobilized coating antigen for the limited binding sites of monoclonal antibody, which resulted in the CL of this system decreases because of less HRP. At the optimal conditions, the CL signal displayed a good linear relation toward Hg 2+ in the range of 0.001–300 ng mL −1 with a detection limit (3σ) of 0.33 pg mL −1 . The immunosensor possessed acceptable accuracy, high specificity and reproducibility, and was applied to examine Hg 2+ in real water samples and cream with favorable results. The successful development of this method also provides reliable prospects and technical support for the follow-up detection of similar environmental pollutants .

A highly selective fluorescent chemosensor for Hg2+ based on rhodamine B and its application as a molecular logic gate

Zhengping Dong,Xin Tian,Yuanzhe Chen,Jingran Hou,Yueping Guo,Jian Sun,Jiantai Ma

A novel and simple fluorescent chemosensor based on rhodamine (R-2) is designed and synthesized to detect Hg 2+ . Probe R-2 exhibits high selectivity and sensitivity for sensing Hg 2+ with a detection limit at 10 −8  M level, and displays a significant color change from colorless to pink color in the presence of Hg 2+ . About a 400-fold increase in fluorescence emission intensity is observed upon binding excess Hg 2+ in 50% H 2 O/CH 3 CN HEPES buffer at pH 7.00. The titration results show a 1:1 complex formation between R-2 and Hg 2+ . The reversibility of chemosensor R-2 is verified through its spectral response toward Hg 2+ and I − titration experiments. Using Hg 2+ and I − as chemical inputs and the fluorescence intensity signal as outputs, R-2 can be utilized as an INHIBIT logic gate at molecular level.

Determination of trace Hg2+ ions based on the fluorescence resonance energy transfer between fluorescent brightener and CdTe quantum dots

Huilin Tao,Xiufen Liao,Mingze Xu,Shuhuai Li,Fuxin Zhong,Zhongsheng Yi

The fluorescence resonance energy transfer between fluorescent brightener (FB, donor) and CdTe quantum dots (CdTe QDs, acceptor) was studied and a new method for detecting Hg 2+ ions in water samples was accordingly developed in this paper. It was found that the effective energy transfer occurred from FB to CdTe QDs in Tris–HCl buffer solution at pH=8.2, in which the QDs' fluorescence intensity increased greatly. Then the fluorescence quenching occurred as Hg 2+ interacted with CdTe. Under the optimized conditions, there was a linear relationship between the fluorescence quenching intensity and the concentration of Hg 2+ . Hg 2+ could be determined in the concentration range of 8.0×10 −9 –8.0×10 −7   g   L −1 with a detection limit of 6.6×10 −10   g   L −1 ( S / N =3). The method was applied to the determination of Hg 2+ in water samples via the standard addition method, and the recovery rate obtained was 97.3–105.5%.

Fluorescent probes for “off–on” highly sensitive detection of Hg2+ and L-cysteine based on nitrogen-doped carbon dots

Yi Zhang,Peipei Cui,Feng Zhang,Xiaoting Feng,Yaling Wang,Yongzhen Yang,Xuguang Liu

Fluorescent nitrogen-doped carbon dots (NCDs) were synthesized by a facile, and low-cost one-step hydrothermal strategy using citric acid as carbon source and ammonia solution as nitrogen source for the first time. The obtained NCDs show stable blue fluorescence with a high quantum yield of 35.4%, along with the fluorescence lifetime of ca. 6.75   ns. Most importantly, Hg 2+ can completely quench the fluorescence of NCDs as a result of the formation of a non-fluorescent stable NCDs–Hg 2+ complex. Static fluorescence quenching towards Hg 2+ is proved by the Stern–Volmer equation, ultraviolet–visible absorption spectra, temperature dependent quenching and fluorescence lifetime measurements. Subsequently, the fluorescence of the NCDs–Hg 2+ system is completely recovered with the addition L-cysteine (L-Cys) owing to the dissociation of NCDs–Hg 2+ complex to form a more stable Hg 2+ -L-Cys complex by Hg 2+ –S bonding. Therefore, such NCDs can be used as an effective fluorescent “turn-off” probe for rapid, rather highly selective and sensitive detection of Hg 2+ , with a limit of detection (LOD) as low as 1.48   nM and a linear detection range of 0–10   μM. Interestingly, NCDs–Hg 2+ system can be conveniently employed as a fluorescent “turn-on” sensor for highly selective and sensitive detection of L-Cys with a low LOD of 0.79   nM and a wide linear detection range of 0–50   μM. Further, the sensitivity of NCDs to Hg 2+ is preserved in tap water with a LOD of 1.65   nM and a linear detection range of 0–10   μM.

N, B-doped carbon dots as a sensitive fluorescence probe for Hg2 + ions and 2,4,6-trinitrophenol detection for bioimaging

Qianghua Ye,Fanyong Yan,Dechao Shi,Tancheng Zheng,Yinyin Wang,Xuguang Zhou,Li Chen

Nitrogen and boron co-doped carbon dots (BCNDs1–3) were prepared from three kinds of borate via a facile hydrothermal method . The as-prepared BCNDs did not shift with the change of excitation wavelength and possess good water dispersibility, strong fluorescence emission with high fluorescent quantum yield of 29.01%, 51.42%, 68.28%, respectively. Subsequently, these BCNDs were exploited as excellent Hg 2   + ion and 2,4,6-trinitrophenol (TNP) probe. The efficient selective detection of Hg 2   + can be attributed to non-radiative electron/hole recombination annihilation through an effective electron transfer process and the detection of TNP can be attributed to the fluorescence resonance energy transfer process (FRET). The results show that the BCNDs2 is the most sensitive fluorescence probe for Hg 2   + ions and TNP detection as low as Hg 2   + 7.3   nM and TNP 0.35   μM compared with BCNDs1 and BCNDs3. The as-prepared BCNDs possess the advantages of good selectivity, fast response and a broad linear detection. They were applied to sensing and imaging of human umbilical vein endothelial cells , showing low cytotoxicity and good biocompatibility .
