Subcellular targeted therapy has recently emerged as a significant approach in cancer treatment. Mitochondria, as crucial organelles that not only provide cellular energy but also play roles in cell differentiation, signal transmission, reactive oxygen species production, apoptosis, and calcium homeostasis, are considered potential targets for cancer therapy. Silver sulfide quantum dots (Ag2S QDs) have demonstrated exceptional capabilities in near-infrared fluorescence and photoacoustic imaging as well as photothermal treatment. Therefore, the development of Ag2S QDs with mitochondrial-targeting ability is of particular significance. Herein, we developed a simple method for synthesizing aqueous-phase Ag2S QDs using lipoic acid, an enzymatic substrate involved in cellular respiration, as a ligand and mitochondrial-targeting molecule. These Ag2S QDs, which exhibit excellent water solubility and biocompatibility, can be produced in a single step and act as a mitochondrial-targeted theranostics. By adsorbing doxorubicin (DOX), we successfully created multifunctional nanoprobes called Ag2S-DOX. In vitro and in vivo experiments demonstrated that Ag2S-DOX was suitable for fluorescence and photoacoustic dual-mode imaging as well as synergistic mitochondrial-targeted chemotherapy and photothermal treatment, offering a promising strategy for tumor therapy.

Cr3+-activated near-infrared (NIR) phosphors are the most promising materials for commercialization. However, the emission of most Cr3+-activated phosphors is typically located in the NIR-I region and achieving luminescence towards the NIR-II region (>900 nm) via Cr3+ doping remains challenging. This study reports a novel Cr3+-activated NaBaScSi2O7 phosphor that exhibits broadband NIR emission centred at 974 nm with a full width at half maximum of 140 nm under 493 nm blue light excitation. The emission wavelength exceeds that of most Cr3+-activated NIR phosphors. The underlying mechanism for the encouraging phenomenon is attributed to the strong charge polarisation arising from the intrinsic local coordination environment of the Cr3+ ions, as confirmed by charge differential density (CDD) and the electron localization function (ELF) analyses. The photoluminescence properties of phosphors were systematically investigated, and the performance of the fabricated NIR pc-LED device was evaluated for night vision and non-invasive imaging. Notably, the NaBaScSi2O7 phosphor displayed a bright green colour with excellent colour properties and its potential as a stable green ceramic pigment was explored.

Breast reconstruction after mastectomy aims to restore breast appearance and function. Current scaffolds often fail to meet the complex clinical demands of post-mastectomy breast reconstruction, which include personalized shaping, prevention of tumor recurrence and metastasis, and promotion of adipose tissue regeneration. This study aims to address these challenges by combining a readily processable biopolymer, polycaprolactone (PCL), with a multifunctional bioactive ceramic, cobalt orthosilicate (Co 2 SiO 4 , CoSi) to form a multi-functional CoSi/PCL composite scaffold through 3D printing technology. The scaffold exhibits a controllable shape with designed macroporous architecture. Its photothermal performances are well presented in both near-infrared (NIR) I and NIR-II regions, enabling effective tumor ablation with minimal side effects. Additionally, the release of bioactive silicate and cobalt ions promotes adipogenesis and angiogenesis, thereby enhancing the integration of the scaffold with surrounding tissues. This approach offers a promising strategy for combining tumor therapy with breast reconstruction, providing a clinically relevant solution for post-mastectomy patients.

Near-infrared II (NIR-II) photoacoustic (PA) imaging offers enhanced performance through reduced photon scattering, minimized background interference, and improved resolution at greater depths, thereby driving urgent demand for NIR-II PA nanoprobes in translational biomedicine. In this study, we developed a smart stannous sulfide@cuprous oxide (SnS@Cu 2 O) nanoprobe for colorectal cancer PA imaging within NIR-II window. Under the existence of HS − , the SnS@Cu 2 O nanoprobe exhibits a broad absorption peak within the 1000–1300 nm range, enabling concentration-dependent NIR-II PA signal amplification in colorectal cancer tissues with overexpression of endogenous H 2 S. The in vitro PA intensity was linearly correlated to HS − concentration in the range of 0–1.2 mM. Intratumoral injection of SnS@Cu 2 O nanoprobe (10 mg/kg) induced time-dependent PA signal amplification at the tumor site, exhibited rapid intensification, peaking at 4-h post-administration. Additionally, the SnS@Cu 2 O can catalyze the Cu + with H 2 O 2 in the tumor tissue yielding ·OH via Fenton-like reactions for photothermal-enhanced chemodynamic therapy, enabling NIR-II PA imaging-guided therapy against deep-tissue colorectal tumor. This paper reports on the synthesis and application of a smart SnS@Cu 2 O nanoprobe that was activated by colorectal tumor microenvironment.

Triple-negative breast cancer (TNBC) is a highly aggressive and uncommon subtype of breast cancer with a poor prognosis. It is crucial to prioritise the creation of a nanotherapeutic method that is highly selective and actively targeting TNBC. This study explores a new nanosystem, Cu 9 S 8 -SNAP@PM (C-S@P), composed of Cu 9 S 8 -SNAP coated with a platelet membrane (PM). The purpose of this nanosystem is to cure TNBC using multimodal therapy. The utilisation of PM-coated nanoparticles (NPs) enables active targeting, leading to the efficient accumulation of C-S@P within the tumour. The Cu 9 S 8 component within these NPs serves the potential to exert photothermal therapy (PTT) and chemodynamic therapy (CDT). Simultaneously, the S-Nitroso-N-Acetylvanicillamine (SNAP) component enables nitric oxide (NO) gas therapy (GT). Furthermore, when exposed to NIR-II laser light, Cu 9 S 8 not only increases the temperature of the tumour area for PTT, but also boosts CDT and stimulates the release of NO through thermal reactions to improve the effectiveness of GT. Both in vitro and in vivo experimental results validate that C-S@P exhibits minimal side effects and represents a multifunctional nano-drug targeted at tumors for efficient treatment. This approach promises significant potential for TNBC therapy and broader applications in oncology.

Background Osteosarcoma (OS) is a malignant tumor originating from primitive mesenchymal cells, characterized by rapid metastasis, high invasiveness, and significant mortality. The primary challenges in OS management include the effective elimination of residual tumor cells to prevent recurrence and the repair of extensive bone defects caused by surgical intervention. Objective This study aims to develop an innovative biomimetic 3D-printed bioactive glass ceramic (BGC) scaffold modified with two-dimensional nanosheets to address both tumor ablation and bone tissue repair. Materials and methods The nanosheets were constructed via ellagic acid (EA) and ruthenium (Ru) coordination, leveraging the non-topological adhesion properties of catechol in EA to deposit the nanosheets onto the BGC scaffold (EARu-BGC). The therapeutic effects of EARu-BGC were evaluated in vitro and in vivo . Results EARu-BGC sequentially responds to the local microenvironment during OS treatment. During the tumor ablation phase, EARu-BGC induced ferroptosis through the synergistic effects of photothermal and chemodynamic therapy, achieving over 90 % tumor cell ablation and significantly inhibiting tumor volume and weight. In the bone tissue repair phase, EARu-BGC exhibited adaptive ROS scavenging and facilitated a pro-healing microenvironment, promoting osteogenic differentiation. The gradual degradation of the BGC scaffold provided essential minerals and space for new bone formation. In vivo experiments demonstrated that EARu-BGC significantly enhanced osteogenesis, increasing the trabecular number to 1.51 ± 0.15/mm and reducing trabecular separation to 1.50 ± 0.04 mm. Conclusion The EARu-BGC scaffold presents a promising multifunctional platform for OS treatment by effectively balancing antitumor efficacy with bone repair capabilities.

Bacterial infections pose a significant threat to the healing of human skin wounds, as the reactive oxygen species (ROS) they produce can significantly delay the wound healing process, ultimately leading to the formation of chronic wounds and even endangering human life. Therefore, this study successfully constructed a dynamically reversible hydrogel network through dual crosslinking of Schiff base bonds and hydrogen bonds using oxidized hyaluronic acid (OHA), carboxymethyl chitosan (CMCS), and tannic acid (TA). By loading Cu@PDA, the OCTC2 hydrogel wound dressing was prepared, exhibiting synergistic antibacterial and antioxidant properties through TA/Cu 2+ release and NIR-II photothermal effects. We used FTIR, XRD, SEM, and TEM to thoroughly analyse the specific composition and morphology of the Cu@PDA and hydrogel. We measured the injectability, adhesion, NIR-II photothermal, and antibacterial properties of each group of hydrogels. The results showed that TA and Cu@PDA endowed the hydrogel with excellent antioxidant properties, enabling effective scavenging of various free radicals. Additionally, OCTC2, which possesses sustained-release TA/Cu 2+ capability, achieves antibacterial rates of over 94 % against S. aureus and E. coli . Under the combined effect of NIR-II photothermal therapy, it can achieve highly efficient bacterial killing (99.8 %), effectively promoting the healing of bacterial infection wounds. OCTC2 holds promise as a potential ideal hydrogel dressing for treating bacterial-infected wounds.

Photothermal therapy based on conjugated polymers represents a promising antibacterial strategy but still possesses notable limitations. Herein, degradable pseudo conjugated polymers (PCPs) containing photothermal molecular backbones and reactive oxygen species (ROS)-sensitive thioketal bonds are designed. Triphenylphosphine (PPh 3 ) is introduced into PCPs to generate phosphonium-based PCPs (pPCPs), which further assembled with hyaluronic acid into pPCP nanoparticles (pPCP-NPs). pPCP-NPs with quaternary phosphonium cations selectively anchor on and destroy bacterial cell membranes through electrostatic action. Under 1064 nm laser irradiation, pPCP-NPs (pPCP-NPs/+L) produce near-infrared-II (NIR-II) photothermal antibacterial effect, thereby killing bacteria in a sustained manner. pPCP-NPs are readily degraded upon ROS abundant at infection sites, therefore exhibiting enough biosafety. pPCP-NPs/+L display an almost 100% bacterial inhibition rate in vitro and resultin a nearly complete recovery of bacteria-induced mouse wounds. A further metabolomics analysis denotes that pPCP-NPs/+L work in a concerted way to induce bacterial DNA damage, inhibit bacterial carbon/nitrogen utilization and amino acid/nucleotide synthesis. Taken together, degradable pPCP-NPs with both NIR-II photothermal effect and cationic phosphonium structural bacteriostasis provide a new avenue for antibiotics-alternative anti-infection therapy.

Early diagnosis and effective treatment of hepatocellular carcinoma (HCC) is quite critical for improving patients’ prognosis. The combination of second near-infrared window photoacoustic imaging (NIR-II PAI) and T2-magnetic resonance imaging (T2-MRI) is promising for achieving omnibearing information on HCC diagnosis due to the complementary advantages of outstanding optical contrast, high temporospatial resolution and soft-tissue resolution. Thus, the rational design of a multifunctional targeted nanoplatform with outstanding performance in dual-modal NIR-II PAI/T2-MRI is particularly valuable for precise diagnosis and imaging-guided non-invasive photothermal therapy (PTT) of early-stage HCC. Herein, a versatile targeted organic–inorganic hybrid nanoprobe was synthesized as a HCC-specific contrast agent for sensitive and efficient theranostics. The developed multifunctional targeted nanoprobe yielded superior HCC specificity, reliable stability and biocompatibility, high imaging contrast in both NIR-II PAI and T2-MRI, and an excellent photothermal conversion efficiency (74.6%). Furthermore, the theranostic efficiency of the targeted nanoprobe was systematically investigated using the orthotopic early HCC-bearing mice model. The NIR-II PAI exhibited sensitive detection of ultra-small HCCs (diameter less than 1.8 mm) and long-term real-time monitoring of the tumor and nanoprobe targeting process in deep tissues. The T2-MRI demonstrated clear imaging contrast and a spatial relationship between micro-HCC and adjacent structures for a comprehensive description of the tumor. Moreover, when using the targeted nanoprobe, the non-invasively targeted PTT of orthotopic early HCC was carried out under reliable dual-modal imaging guidance with remarkable anti-tumor efficiency and biosafety. This study provides an insight for constructing a multifunctional targeted nanoplatform for precise and comprehensive theranostics of early-stage HCC, which would greatly benefit the patients in the era of precision medicine.

Blue-light-excitable NIR-II/III emission. • Ni 2+ occupies ScO 6 octahedral sites. • Sc substitution enables 1570–1690 nm tuning. • Zr 4+ charge compensation boosts emission 5-fold. • NIR-LED for night-vision and bioimaging. Long-wavelength NIR luminescent materials provide essential support for optoelectronic devices, yet realizing blue-light-pumped broadband emission across the NIR-II/III windows remain challenging. Herein, we successfully synthesized a series of Sr 2 Ga 1- x Sc x SbO 6 :Ni 2+ phosphors with tunable emission from 1570 to 1690 nm under 435–465 nm excitation. Corroborating evidence from DFT calculations, XANES spectroscopy, and structural refinement consistently confirms that the doped Ni 2+ ions occupy Ga/ScO 6 octahedron rather than SbO 6 sites. Furthermore, the introduction of Zr 4+ as a charge compensator effectively mitigates lattice defects induced by aliovalent substitution, leading to a remarkable enhancement in luminescence intensity across the entire composition series. The NIR pc-LED fabricated by integrating the optimized Sr 2 ScSbO 6 :Ni 2+ , Zr 4+ phosphor with commercial GaN chip exhibits promising performance for applications in night-vision imaging and chemical composition analysis. This work proposes a lattice-site engineering strategy to achieve ultralong-wavelength NIR emission and optimize luminescence performance, thereby establishing a new design paradigm for next-generation NIR phosphors. Oxidative stress plays a pivotal role in the onset and progression of arthritis, in which the abnormal elevation of hydroxyl radical (•OH) has been recognized as a key molecular hallmark of early inflammation. However, effective tools capable of simultaneously visualizing •OH dynamics and tracking therapeutic efficacy remain elusive. Herein, we developed a self-calibrated near-infrared II (NIR-II) ratiometric imaging probe responsive to •OH. The probe integrates downconversion nanoparticles (DCNP) as a stable internal reference (F1550nm,980Ex) and a •OH-triggered red-shiftable dye Hydro-830 (F930nm,808Ex), enabling ratiometric imaging through the signal ratio “F930nm,808Ex/F1550nm,980Ex.” The probe exhibits high sensitivity and specificity toward •OH in vitro and effectively captures the upregulation of oxidative stress at early inflammatory stages in vivo. In a longitudinal “treatment–imaging” study, the ratiometric signal gradually decreased following methotrexate (MTX) therapy, consistent with reductions in joint swelling, inflammatory cytokines, and histopathological lesions. This study establishes a self-calibrated NIR-II ratiometric imaging strategy for precise monitoring of oxidative stress, enabling early molecular diagnosis and dynamic therapeutic evaluation of arthritis, and offering a promising paradigm for imaging other ROS-related inflammatory disorders.

The clinical management of osteosarcoma faces critical challenges, including postoperative recurrence and metastasis of residual tumor cells, chemotherapy resistance, and impaired self-repair capacity due to extensive bone defects following tumor resection. Herein, we develop a novel functionalized 3D-printed scaffold with NIR-II responsiveness (BGS/I-LDH@MgO 2 ), designed to simultaneously address the dual needs of inhibiting osteosarcoma recurrence and promoting bone regeneration. This scaffold consists of 3D-printed bioactive glass scaffold (BGS) and MgO 2 -modified ZnAl-layered double hydroxides (ZnAl-LDHs) intercalated with 5-iodo-isophthalic acid (I-IPA). Under NIR-II irradiation, the scaffold effectively triggers a photodynamic therapy (PDT) effect to eliminate osteosarcoma cells. Notably, the incorporation of MgO 2 enables oxygen release within the tumor microenvironment, alleviating hypoxia and enhancing PDT efficacy for superior antitumor performance. Furthermore, the degradation of ZnAl-LDHs and MgO 2 releases Mg 2+ and Zn 2+ ions and generates a mildly alkaline microenvironment, which collectively facilitate the osteogenic differentiation of bone marrow mesenchymal stem cells and accelerate the process of bone healing. This functionalized 3D-printed scaffold demonstrates excellent anti-tumor and osteogenic properties, showing great promise for the treatment of osteosarcoma-associated bone defects.
