Identification of pine wood nematode (Bursaphelenchus xylophilus) loading response genes in Japanese pine sawyer (Monochamus alternatus) through comparative genomics and transcriptomics

Xiangying Zou,Jiayu Jia,Tengfei Zhu,Shouping Cai,Yiman He,Shunde Su,Yu Fang,Jian Li,Guifang Lin,Jun Su

BACKGROUND Pine wood nematode (PWN; Bursaphelenchus xylophilus ) is the causative agent of pine wilt disease (PWD), which is considered the most dangerous biohazard to conifer trees globally. The transmission of PWN relies on insect vectors, particularly the Japanese pine sawyer (JPS; Monochamus alternatus ). However, the molecular mechanism underlying PWN-JPS assembly remains largely unknown. RESULTS Here, we found that both geographical and gender could significantly affect the PCA (PWN carrying amount) of JPS; thus, JPS transcriptomes from diverse locations and genders were explored regard to PWN loading. Due to the shortage of genomes, we developed a full-length reference transcriptome for analyzing next-generation sequencing data. A comparative genomic study was performed, and 11 248 potential PWN-carrying associate genes (β) were nominated in JPS by using the reported genomes of PWN and non-PWN carrier insect species. Then, 151 differentially expressed transcripts (DETs), 28 of them overlapped with β, correlated with the PCA of JPS were nominated by RNA-Seq, and found that fatty acid β-oxidation might be the key factor that affected the PCA of JPS. Furthermore, JPS fatty acid β-oxidation rates were experimentally decreased using the inhibitor Etomoxir, leading to an increased PCA of JPS. Meanwhile, silencing MaCPT1 in JPS by RNA interference led to a decreased fatty acid β-oxidation rate and increased PCA of JPS. CONCLUSIONS In conclusion, MaCPT1 was able to decrease the PWN-JPS assembly formation through the fatty acid β-oxidation of JPS. These results provide new insights for exploring the impact of PWN invasion on JPS. © 2024 Society of Chemical Industry.

Investigation of the mechanisms involved in the biocontrol activities of natural products from a marine soil bacterium against rice blast

Liwang Fei,Rahila Hafeez,Junliang Zhang,Shiquan Fu,Ying Xu,Lingyun Hao

BACKGROUND Rice blast, caused by Pyricularia oryzae , is a devastating fungal disease threatening global rice production. Overreliance on chemical fungicides has raised environmental concerns and led to resistant strains, necessitating the development of sustainable alternatives. This study integrated marine microbiology and natural antifungal compounds to create eco-friendly alternatives to chemical fungicides for disease management. RESULTS We identified Pseudomonas aeruginosa R64 with broad-spectrum antimicrobial activity from mangrove soil in the Mai Po Nature Reserve. The R64 fermentation extract (RFE) exhibited multifaceted inhibition of P. oryzae , suppressing mycelial growth, conidiation, conidial germination and appressorial formation, while disturbing cell wall and membrane function. It also attenuated virulence by impairing appressorial penetration and invasive growth. Further chemical analysis identified phenazines and quinolines as the primary compounds in RFE, corroborated by PCR detection of corresponding phenazine biosynthetic gene clusters. Comparative bioassays with two main bioactive components of RFE, phenazine-1-carboxamide (PCN) and phenazine-1-carboxylic acid (PCA), against P. oryzae implicated PCN as the principal antifungal effector. RFE and PCN had higher efficacy than tricyclazole in P. oryzae growth inhibition, but were less effective than isoprothiolane. Furthermore, RFE and PCN displayed lower acute ecotoxicity to an environmental indicator organism than isoprothiolane, suggesting their potential as sustainable biopesticides for rice blast management. CONCLUSION Natural products from mangrove soil bacterium P. aeruginosa R64 inhibited key developmental and infection processes of P. oryzae , effectively reducing rice blast development. The promising disease inhibition and low ecotoxicity of mangrove-associated bacteria highlight their untapped potential for innovative, eco-friendly fungicide mining for sustainable agriculture. © 2025 The Author(s). Impacts of the cyromazine emergence inhibition procedure of Bactrocera dorsalis on Fopius arisanus: implications for the biological control of diptera pest

Jia Lin,Guoqing Yue,Jun Chen,Kang Xiao,Qing'e Ji

Background Fopius arisanus (Sonan) is an important egg–pupal endoparasitic wasp of Bactrocera dorsalis (Hendel). As traditional method of sorting nonparasitic B. dorsalis from parasitic wasps is labor-intensive, requires specific equipment and poses the risk of spreading fertile hosts, the development of a more convenient, economical and safe sorting procedure is important. Results The optimal cyromazine emergence inhibition procedure (CEIP) involved facilitating the pupation of B. dorsalis mature larvae (Bdml) in 3 mg kg −1 cyromazine sand substrate (CSS) for 48 h. When the Bdml that had been exposed to F. arisanus during the egg stage were treated with 3–7 mg kg −1 CSS for 48 h, no negative effects on the emergence parameters of parasitoids were observed. Treatment with 3–4 mg kg −1 CSS had insignificant effects on the biological and behavioral parameters of F. arisanus . However, treatment with 5–6 mg kg −1 CSS adversely affected the fecundity and antennating activity of the wasps; specifically, 6 mg kg −1 CSS negatively affected the lifespan and flight ability of wasps. Fortunately, no transgenerational effects on these parameters were observed in the progeny. Regarding the nutrient reserves of both sexes of F. arisanus , significant dose-dependent effects were observed. Moreover, 5–6 mg kg −1 CSS significantly reduced the protein and carbohydrate content in F. arisanus ; in particular, 6 mg kg −1 CSS notably reduced the lipid content. Conclusion CEIP provides a more flexible, economical and safe mass-release program for F. arisanus . In addition, it has profound implications for the biological control of other dipteran pests. © 2024 Society of Chemical Industry.

Layer-by-layer assembled decomposable nanocapsules for light responsive release of pesticide imidacloprid on Aphis craccivora Koch

Yiyun Gao,Wenjun Xiao,Chenhua Gu,Sheng Yu,Shengke Li,Aihua Zou

Background Conventional pesticide formulations are often griped about inefficiency due to the low biological uptake after spraying. Controlled release nanopesticides can precisely release pesticides in response to specific stimuli, thereby killing pests and pathogens with the least effective concentration. This study aims to develop nanocapsules-based photo decomposable nanopesticides for efficient pesticide control. Results The target nanopesticides were successfully fabricated by using layer-by-layer assembly of azobenzene grafted hyaluronic acid (Azo-HA) and PolyDADMAC, confirmed by UV–vis, DLS, Zeta potential and TEM measurements. Particle size and Zeta potential of the fabricated nanocapsules were 220 nm and + 46.1 mV, respectively, and the nanocapsules were found to maintain stable for up to 30 days. The optimized drug loading and encapsulation ratio of Imidacloprid (IMI) in IMI/Azo-HA@PolyDADMAC were 21.5% and 91.3%, respectively. Cumulative release of IMI from the nanopesticides was increased from ~50% to ~95% upon UV-light irradiation (365 nm). LC 50 value of the nanopesticides towards Aphis craccivora Koch was decreased from 2.22 mg/L to 0.55 mg/L upon UV-light irradiation. Conclusion The trans to cis transformation of Azo group in HA will decompose IMI/Azo-HA@PolyDADMAC nanopesticides upon UV irradiation, thus facilitated the IMI release, resulting in decreased concentration of pesticides for efficient pesticide control. Our work demonstrated the great potential of light-responsive nanocapsules as controlled release nanocarrier for efficient and eco-friendly pesticide-control in sustainable agriculture. Insecticidal activities of three recombinant venom proteins of the predatory stink bug, Arma custos

Xinyi Wang,Wenhong Li,Xiang Yang,Mingwei Yang,Yucheng Gu,Zhao Du,Jingyi Yang,Mingxia Wen,Yoonseong Park,Chunyang Huang,Yueping He

BACKGROUND Widespread resistance of insect pests to insecticides and transgenic crops in the field is a significant challenge for sustainable agriculture, and calls for the development of novel alternative strategies to control insect pests. One potential resource for the discovery of novel insecticidal molecules is natural toxins, particularly those derived from the venoms of insect predators. RESULTS In this study, we identified three insecticidal proteinaceous toxins from the venom glands (VGs) of the predatory stink bug, Arma custos (Hemiptera: Asopinae). Transcriptomic analysis of A. custos VGs revealed 151 potentially secreted VG-rich venom proteins. Three VG-rich venom proteins (designated AcVP1 ~ 3) were produced by overexpression in Escherichia coli . Injection of the recombinant proteins into tobacco cutworm ( Spodoptera litura ) larvae showed that all of the three recombinant proteins caused paralysis, liquefaction and death. Injection of recombinant proteins into rice brown planthopper ( Nilaparvata lugens ) nymphs showed higher insecticidal activities, among which a trypsin (AcVP2) caused 100% mortality postinjection at 1.27 pmol mg −1 body weight. CONCLUSION A natural toolkit for the discovery of insecticidal toxins from predatory insects has been revealed by the present study. © 2024 Society of Chemical Industry.

Integrating pyriproxyfen into the incompatible insect technique enhances mosquito population suppression efficiency and eliminates the risk of population replacement

Yongjun Li,Jiameng Peng,Haiying Li,Ruiqi Zhang,Jiexia Chen,Xiuying Hou,Guang Yang

BACKGROUND The incompatible insect technique (IIT) has been used for Aedes mosquito population suppression to curb the transmission of dengue. However, its wide application is limited owing to the low output of male mosquitoes and the risk of population replacement from the release of fertile Wolbachia -infected females. This study aims to improve IIT efficiency for broader adoption. RESULTS We assessed the impact of 10% pyriproxyfen (PPF) sticky powder exposure on Wolbachia (from Culex molestus )-transinfected Aedes albopictus Guangzhou line (GUA line) (GC) mosquitoes. We found that the exposure caused chronic toxicity in adult mosquitoes without affecting the cytoplasmic incompatibility (CI)-inducing capability of males. The PPF-contaminated GC females exhibited significant sterilization and the ability to disseminate lethal doses of PPF to breeding sites. Subsequently, we conducted a field trial combining PPF with IIT aiming to suppress the Ae. albopictus population. This combined approach, termed boosted IIT (BIIT), showed a notable enhancement in population suppression efficiency. The improved efficacy of BIIT was attributed to the dispersion of PPF particles in the field via the released PPF-contaminated male mosquitoes. During the BIIT field trial, no Wolbachia w Pip-positive Ae. albopictus larvae were detected, indicating the effective elimination of the risk of Wolbachia -induced population replacement. Additionally, the field trial of BIIT against Ae. albopictus resulted in the suppression of the nontarget mosquito species Culex quinquefasciatus . CONCLUSION Our results highlight the remarkable efficiency and feasibility of combining IIT with PPF in suppressing mosquito populations, facilitating the widespread implementation of IIT-based management of mosquito-borne diseases. © 2024 Society of Chemical Industry.

Chlorbenzuron downregulated HcLCP-17 expression by depressing two 20E-responsive transcription factors Br-C and βFTZ-F1 in Hyphantria cunea (Lepidoptera: Erebidae) larvae

Yuecheng Zhao,Lu Zhang,Chuanshan Zou,Huilin Han,Chengde Li,Xingpeng Li,Liwen Song

BACKGROUND Cuticular proteins (CPs) play essential roles in forming cuticular structures in insects. However, the specific functions and regulatory mechanisms of CPs remain largely unexplored. In this study, the Larval cuticular protein 17 ( HcLCP-17 ) gene was identified from Hyphantria cunea , a highly destructive and polyphagous forest pest. To investigate the role of HcLCP-17 in cuticular function and transcriptional regulation mediated by 20E-responsive transcription factors (ERTFs), we employed RNA interference (RNAi) and yeast one-hybrid assay techniques. Additionally, we examined the molecular mechanism by which chlorbenzuron, a type of benzoylphenylurea (BPU) that functions as a chitin synthesis inhibitor (CSI), affects the 20E signaling pathway and ultimately regulates HcLCP-17 expression. RESULTS HcLCP-17 encodes a polypeptide consisting of 393 amino acids, which includes a chitin-binding domain. Silencing HcLCP-17 resulted in a disturbance in the structural organization of the larval cuticle and a notable reduction in chitin levels. HcLCP-17 expression was controlled by the interaction between Broad-Complex (Br-C) and beta Fushi Tarazu Factor-1 (βFTZ-F1) with its promoter fragment. Furthermore, the inhibitory effect of chlorbenzuron on HcLCP-17 expression was found to be potentially mediated by Br-C and βFTZ-F1. CONCLUSION The study presents a novel mode of action for the 20E signaling pathway in regulating the expression of CPs and reveals the potential mode-of-action of BPUs in insect cuticles. These findings provide a theoretical basis for future utilization of LCP-17 as a pesticide target making a significant contribution to the development of effective pest management strategies. © 2024 Society of Chemical Industry.

CRISPR/Cas9-mediated knockout of a male accessory glands-specific gene takeout1 decreases the fecundity of Zeugodacus cucurbitae female

Yun Wang,Hui-Qian Xu,Hong-Liang Han,Dong Chen,Hongbo Jiang,Guy Smagghe,Jin-Jun Wang,Dong Wei

BACKGROUND The melon fly, Zeugodacus cucurbitae (Coquillett), is an invasive Tephritidae pest with robust fertility. The male accessory glands (MAGs) form a vital organ that ensures insect reproductive efficiency. Most of the secreted proteins by MAGs exhibit a male bias expression. Takeout, one of these proteins, is abundantly present in the MAGs of many insects. RESULTS In this study, we identified 32 takeout genes in Z. cucurbitae . The phylogenetic analysis and multiple sequence alignment results showed that Zctakeout1 is the most related homolog to the MAGs-specific takeout in Tephritidae. The real-time quantitative PCR results showed that Zctakeout1 was exclusively expressed in the male adult stage, and its expression level gradually increased with the increase in age and then remained stable at the sexually matured stage. The distribution among tissues demonstrated the specific expression of Zctakeout1 in the MAGs, and fluorescence immunohistochemical results confirmed the presence of Zctakeout1 in close proximity to binuclear cells of the mesoderm epidermal MAGs. In continuation, CRISPR/Cas9-mediated genome editing was employed, resulting in successfully generating a homozygous strain with an +8 bp insertion. The mating experiments with the Zctakeout1 −/− males resulted in significant reductions in both the mating rate and egg production of females. CONCLUSION These findings prove that the MAGs-specific Zctakeout1 is essential in regulating fecundity in female Z. cucurbitae fruit flies. Our data suggests its utilization in future essential insect-specific gene-directed sterility insect technique (SIT) by the genetic manipulation to keep these important Tephritidae populations under control. © 2024 Society of Chemical Industry.

Core reinforcement strategy enhances the foliar stability and efficacy of electrostatic self-assembled microcapsules

Xing Chen,Junpeng Yang,Lingmin Xia,Rui Yang,Chaoyang Ding,Xueping Huang,Yu Chen,Jian Luo

BACKGROUND At present, it is vital to develop a stable and efficient pesticide delivery system to optimize pesticide foliar utilization, which could improve control efficacy, enhance resistance to adverse climates, and prolong foliar retention. In this study, reaction monomers methylene diphenyl diisocyanate (MDI) and polycaprolactone diol (PCL) were used to synthesize a polymer network structure for loading the organic phase of pesticides in a micron-reactor, then the shell was formed by sodium lignosulfonate (SL) and didecyl dimethyl ammonium chloride (DDAC) through electrostatic self-assembly, resulting in self-assembled microcapsules and efficient pesticide loading, and the stability and efficacy were discussed. RESULTS Self-assembled microcapsules Pyr@MCs-C and Pyr@MCs-V with cores of different mechanical strength and morphological characteristics are realized by regulating the reaction ratio of MDI and PCL. Compared with conventional self-assembled microcapsule Pyr@MCs-S, Pyr@MCs-C and Pyr@MCs-V exhibit stable and unruptured morphology in dehydrated environment. Moreover, self-assembled microcapsules provide similar fungicidal activity as emulsifiable concentrate. Notably, the washout resistance property of Pyr@MCs-C and Pyr@MCs-V increased by 3.20 and 3.51 times, respectively, and ultraviolet (UV) resistances of the two microcapsules increased by 5.72 and 5.02 times, respectively, which promote the control efficiency and prolong the duration. CONCLUSION In summary, this system has simple preparation process and stable foliar performances, making it a promising precise pesticide delivery platform. © 2025 Society of Chemical Industry.

Dipicolinic acid reduces Epicoccum sorghinum symptoms on maize and inhibits tenuazonic acid biosynthesis

Xin-Yu Ji,Bing-Yi Wang,Yi-Feng Zhang,Yu-Jing Zhang,Ya-Jie Lai,Yang Yang,Xiang-Chen Wang,Su-Yan Wang,Pedro Laborda,Xin-Chi Shi

BACKGROUND Epicoccum sorghinum is a pathogenic fungus that causes leaf spot in a wide range of plants, including maize, and synthesizes the mycotoxin tenuazonic acid (TEA), which is carcinogenic. Despite the relevant economic and yield losses caused by E. sorghinum worldwide, methods for the control of this pathogen are lacking. RESULTS In this work, the efficacy of Bacillus -produced dipicolinic acid (DPA) for control of E. sorghinum was evaluated using in vitro and in vivo assays, and compared with the efficacy of three commercial fungicides, including carbendazim, prochloraz, and thiram. DPA inhibited E. sorghinum mycelial growth, and conidia germination, and produced important alterations in E. sorghinum hyphae. Interestingly, 10 mM DPA reduced TEA biosynthesis by 86.6%. Although DPA rapidly degraded on maize leaves, 10 mM DPA showed higher preventive (67.4% lesion length inhibition) and inhibitory (89.5% lesion length inhibition) efficacies for the control of E. sorghinum on maize leaves compared to the commercial fungicides. CONCLUSIONS Collectively, this study presents the first method for the control of E. sorghinum on maize and demonstrates that DPA application is a suitable approach to inhibit E. sorghinum symptoms in plants and TEA biosynthesis. © 2024 Society of Chemical Industry.

Electrospun nanofibers as controlled release systems for the combined pheromones of Grapholita molesta and Cydia pomonella

Hui-Ming Xiang,Xiao-Hong Wei,Yi-Ran Li,Bo-Jun Zhang,Min Li,Rui-Yan Ma

BACKGROUND As sex pheromones are environmentally friendly and specific, they are often used to monitor and control oriental fruit moths (OFMs). Currently, non-biodegradable polymers are commonly employed as carriers to prepare controlled sex pheromone release systems for plant protection. Electrospinning is a relatively simple technique for preparing biodegradable nanofibers that allows for the controlled release of sex pheromones. This study aimed to develop a biodegradable controlled release system for the combination of OFM pheromone and codlemone via electrospinning, in which codlemone was used as a synergist for OFM pheromone. RESULTS New systems for the controlled release of combined pheromones from OFM and colding moths were developed using electrospun nanofibers from polycaprolactone (PCL), cellulose acetate (CA), and polylactic acid (PLA). In the indoor experiments, the load ratio and release stability of CA nanofibers loaded with combined pheromones (CA-SP) were superior to those of the other two nanofiber types. In the field experiments, among all the treatments, 10 mg of CA-SP was the most attractive to OFMs, and its effective duration was approximately 6 weeks. The optimum storage temperature for all nanofibers was −20 °C. CONCLUSION Electrospun CA-SP nanofibers were biodegradable and environmentally friendly, with stable release. This study presents a new technique that could be beneficial for the monitoring and control of OFMs. © 2025 Society of Chemical Industry.

Bio-based two-dimensional amphiphile with hierarchical self-assembly for enhancing pesticide utilization and reducing environmental risks

Yanling Wu,Songhao Zhang,Zhenping Bao,Rui Liu,Fengzhe Sun,Yue Wang,Haiqi Huang,Yuxia Gao,Yongqiang Ma,Chenhui Zhang,Fengpei Du

BACKGROUND Biotic and abiotic stresses threaten crop growth and yield. Agrochemicals are an important way to mitigate biotic stress, while frequent low utilization and potential environmental risk affect their sustainable use. In order to improve pesticide utilization, it is common practice to add tank-mix adjuvants by reducing surface tension or forming spherical self-assembly. However, there is a lack of quantitative indicators to screen suitable molecules for sustainable application. In this work, critical factors based on physicochemical properties, and kinetic and thermodynamic parameters are applied to analyze regulatory mechanisms in dynamic processes, and ultimately to establish an integrated strategy for the management of stresses. RESULTS Compared with traditional one-dimensional linear amphiphilic molecules, two-dimensional bio-based amphiphilic molecules, especially sodium deoxycholate (NaDC), form self-assembly and could significantly promote the deposition of agrochemical droplets due to maximum energy dissipation. Meanwhile, NaDC increased the inhibition rate of pyraclostrobin against Rhizoctonia solani from 24.4% to about 100.0%, which was beneficial for pesticide resistance to biotic stress. In addition, NaDC could significantly mitigate the harmful effects of salt stress on Oryza sativa by increasing the germination rate of salt-stressed seeds by about 30%, and reducing the environmental risk of pesticides to soil microbial communities for eco-friendly crop protection. CONCLUSION Herein, this work demonstrates a sustainable strategy for crop management that enhances the effects of agrochemicals on biotic stresses, mitigates abiotic stresses, and significantly reduces environmental risks. © 2025 Society of Chemical Industry.
