Novel polyurethane-degrading cutinase BaCut1 from Blastobotrys sp. G-9 with potential role in plastic bio-recycling

Zhitong Jiang,Xue Chen,Huizhen Xue,Zhoukun Li,Jinhui Lei,Muming Yu,Xin Yan,Hui Cao,Jie Zhou,Jiawei Liu,Mingna Zheng,Weiliang Dong,Yanwei Li,Zhongli Cui

Environmental pollution caused by plastic waste has become global problem that needs to be considered urgently. In the pursuit of a circular plastic economy, biodegradation provides an attractive strategy for managing plastic wastes, whereas effective plastic-degrading microbes and enzymes are required. In this study, we report that Blastobotrys sp. G-9 isolated from discarded plastic in landfills is capable of depolymerizing polyurethanes (PU) and poly (butylene adipate-co-terephthalate) (PBAT). Strain G-9 degrades up to 60% of PU foam after 21 days of incubation at 28 ℃ by breaking down carbonyl groups via secretory hydrolase as confirmed by structural characterization of plastics and degradation products identification. Within the supernatant of strain G-9, we identify a novel cutinase BaCut1, belonging to the esterase family, that can reproduce the same effect. BaCut1 demonstrates efficient degradation toward commercial polyester plastics PU foam (0.5 mg enzyme/25 mg plastic) and agricultural film PBAT (0.5 mg enzyme/10 mg plastic) with 50% and 18% weight loss at 37 ℃ for 48 h, respectively. BaCut1 hydrolyzes PU into adipic acid as a major end-product with 42.9% recovery via ester bond cleavage , and visible biodegradation is also identified from PBAT, which is a beneficial feature for future recycling economy. Molecular docking , along with products distribution, elucidates a special substrate-binding modes of BaCut1 with plastic substrate analogue . BaCut1-mediated polyester plastic degradation offers an alternative approach for managing PU plastic wastes through possible bio-recycling.

Fabrication of CNT/Cu composites with enhanced strength and ductility by SP combined with optimized SPS method

Liang Liu,Rui Bao,Jianhong Yi,Dong Fang

Mono-dispersed and homogeneous carbon nanotube (CNT)/copper (Cu) composite powders were fabricated by spraying pyrolysis (SP) method successfully. Subsequently, CNT/Cu composites were obtained by spark plasma sintering (SPS) at sintering temperature ranged from 1023 to 1223 K (70–90% melting point of pure Cu). The mechanical properties , micro-morphology and interface microstructure of CNT/Cu composites were measured and characterized. The results reveal that Cu nano-particles (NPs) coated on the CNT surface are conducive to improving the wettability of CNT within matrix and reducing the interface energy between matrix and CNT, and finally stable interface structure is established. The traditional trade-off tendency between strength and ductility is balanced in the CNT/Cu composites, which are sintered at 1223 K. The ultimate tensile strength (UTS) shows 344 MPa and the elongation is 19.6%, which owing to the improvement of CNT–Cu interface bonding. At last, the role of interfacial bonding extent played in determining the load transfer efficiency of CNT is discussed.

Characterization of an epilactose-producing cellobiose 2-epimerase from Clostridium sp. TW13 and reutilization of waste milk

Zhou Chen,Yanyin Song,Yuhan Yan,Wei Chen,Tong Ren,Aijin Ma,Siting Li,Yingmin Jia

A novel cellobiose 2-epimerase gene ( CsCEase1 ) from Clostridium sp. TW13 was successfully overexpressed. Cs CEase1, with a molecular mass of 45.0 kDa, exhibited a specific activity of 254.75 U/mg and showed optimal activity at 40 °C and pH 7.5. Using Cs CEase1, 44.30 % of lactose was converted into epilactose, yielding a production level of 26.58 g/L. An efficient purification method was developed, incorporating crystallization and β-galactosidase treatment to remove the unconverted lactose, while Saccharomyces cerevisiae was used to consume the produced glucose. Final purification with activated carbon resulted in epilactose with 98 % purity and a recovery rate of 66.47 %. Furthermore, Cs CEase1 was incubated with whey power and expired milk, achieving epilactose conversion rates of 29.65 % and 32.69 %, respectively. These findings highlight the potential of Cs CEase1 for the cost-effective and environmentally sustainable production of epilactose from dairy waste.

Rehmannia alcohol extract inhibits neuropeptide secretion and alleviates osteoarthritis pain through cartilage protection

Yanfeng Huang,Qing Lin,Xue Tan,Liangliang Jia,Hui Li,Zaishi Zhu,Changlong Fu,Lili Wang,Linlong Liu,Min Mao,Zhouping Yi,Dezun Ma,Xihai Li

Osteoarthritis (OA) is a common joint disease characterized by chronic pain, and the perception of pain is closely associated with brain function and neuropeptide regulation. Rehmannia is common plant herb with anti-inflammatory and analgesic properties that is used to treat OA. However, it is unclear whether Rehmannia alleviates OA-related pain via regulation of neuropeptides and brain function. We examined the pain relief regulatory pathway in OA after treatment with Rehmannia by verifying the therapeutic effect of Rehmannia alcohol extract in vivo and vitro and exploring of the potential mechanism underlying the analgesic effect of Rahmanian using functional magnetic resonance imaging and measuring neuropeptide secretion. Our results showed that Rehmanni a alcohol extract and the related active ingredient, Rehmannioside D , can delay cartilage degradation and alleviate inflammation in OA rats. The Rehmanni a alcohol extract can also relieve OA pain, reduce the secretion of calcitonin gene-related peptide (CGRP) and substance P (SP), and reverse the pathological changes in the cerebral cortex and hippocampus. Our research results demonstrate that Rehmannia alleviates OA pain by protecting cartilage, preventing the stimulation of inflammatory factors on neuropeptide secretion, and influencing the relevant functional areas of the brain.

Effects of stress metabolism on physiological and biochemical reaction and polycyclic aromatic hydrocarbons degradation ability of Bacillus. sp

Bo Zhang,Lei Wang,Zhenjun Diwu,Maiqian Nie,Hongyun Nie

A strain of Bacillus licheniformis T5 was isolated from soil contaminated with crude oil due to its efficient degradation of polycyclic aromatic hydrocarbons (PAHs). When subjected to stress metabolism using phenanthrene as a carbon source, significant changes were observed in T5 cells. Infrared spectrum analysis revealed the presence of -C=C- and Ph-O-C (aromatic) groups on the bacterial surface, facilitating the adsorption of PAHs on the phospholipid layer and causing damage to the cell membrane. Scanning electron microscope (SEM) analysis showed the changes of cell morphology, including a large number of folds on the lower surface and the folding of cell membrane. Transmission electron microscope (TEM) observation showed that non-stressed bacteria with adequate nutritional conditions accumulated more lipids. However, the stress group contained more protein. It was found that stress metabolism led to the increase of protein content in T5 cells by 16.4% and the activity of oxidoreductase more than doubled. These physiological and biochemical changes enhance the ability of stressed bacteria to degrade PAHs efficiently, thereby reducing the degradation cycle. The findings offer valuable insights for the remediation of PAHs pollution.

Zhang Di,Feng Yao,Li Huifang,Zhang Zhizhen

Background: Accumulated investigations have demonstrated that the Mariana Trench is enriched in microorganisms. However, the diversity of structures and bioactivities of the secondary metabolites produced by the Mariana Trench-associated microorganisms is poorly known, which needs to be intensively investigated. Objective: This study aimed to investigate the bioactive secondary metabolites produced by a Mariana Trench-derived actinomycete Streptomyces sp. SY1414 that was cultured in BY medium, which was chosen from four different media based on the diversity of secondary metabolites. Methods: A combination of different column chromatographs and HPLC was applied for the separation and purification of the secondary metabolites. The structures of the isolated compounds were determined mainly based on their NMR data, optical rotation values, and the comparison with the reference data, and the Sulforhodamine B (SRB) method was used to evaluate their anti-glioma activities. Results: Four different types of compounds were isolated from the large culture of strain Streptomyces sp. SY1414 in BY medium, including a benzoquinoline alkaloid, actinophenanthroline C (1), a benzamide, (2E,4E)-5-(3-hydroxyphenyl)-penta-2,4-dienamide (2), a cyclopeptide, valinomycin (3), and four macrolides of bafilomycin D (4), bafilomycin A2 (5), bafilomycin W (6), and C(19), C(21)- O-methyl-bafilomycin A1 (7). Actinophenanthroline C (1), bafilomycin D (4), bafilomycin A2 (5), and bafilomycin W (6) displayed significant anti-glioma activities with IC50 values ranging from 1.62 to 8.20 μM for U87MG cells and 2.45 to 3.89 μM for U251. The anti-gliomas of actinophenanthroline C (1) was reported for the first time. Conclusion: The hadal actinomycete Streptomyces sp. SY1414 in BY medium produced four different types of secondary metabolites with significant anti-glioma activity, which enriched the diversity of structures and bioactivities of the Mariana Trench-associated natural products.

Effect of metal ions on the enzymatic hydrolysis of hemp seed oil by lipase Candida sp. 99–125

Jike Lu,Pei Wang,Zhaodi Ke,Xin Liu,Qiaozhen Kang,Limin Hao

In order to study the effect of metal ions on the enzymatic hydrolysis of hemp seed oil by Candida sp. 99–125, the spectroscopy, stability and hydrolytic activity of the biocatalyst were investigated in presence of Ca 2+ , Mg 2+ , Fe 2+ , Fe 3+ , Cu 2+ , Sn 2+ , Pb 2+ , Zn 2+ and Ba 2+ metal ions, respectively. The UV spectroscopy showed that all the metal ions enhanced the absorbance but the decrease of fluorescence intensity was observed. All the metal ions could improve the lipase thermal stability except Cu 2+ and Ba 2+ . Hydrolysis of hemp seed oil proved that Ca 2+ , Fe 3+ , Pb 2+ and Ba 2+ could significantly improve the hydrolytic rate, and metal ions could influence lipase selectivity. The study revealed that metal ions could improve lipase stability, hydrolysis activity in the hydrolytic process of hemp seed oil by Candida sp. 99–125.

Transcriptome analysis on fructose as the sole carbon source enhancing perylenequinones production of endophytic fungus Shiraia sp. Slf14

Liu Zhengying,Bao Jianying,Yang Huilin,Zhang Zhibin,Yan Riming,Zhu Du

Perylenequinones (PQ), a class of naturally occurring polypeptides, are widely used as a clinical drug for treating skin diseases and as a photodynamic therapy against cancers and viruses. In this study, the effects of different carbon sources on PQ biosynthesis by Shiraia sp. Slf14 were compared, and the underlying molecular mechanism of fructose as the sole carbon to enhance PQ production was investigated by transcriptome analysis. The results indicated that fructose enhanced PQ yield to 1753.64 mg/L, which was 1.73-fold higher than that obtained with glucose. Comparative transcriptome analysis demonstrated that most of the upregulated genes were related to transport systems, energy and central carbon metabolism in Shiraia sp. Slf14 cultured in fructose. The genes involved in glycolysis and pentose phosphate pathways, and encoding citrate synthase, ATP-citrate lyase, and acetyl-CoA carboxylase were substantially upregulated, resulting in increased overall acetyl-CoA and malonyl-CoA production. However, genes involved in gluconeogenesis, glyoxylate cycle pathway, and fatty acid synthesis were significantly downregulated, resulting in higher acetyl-CoA influx for PQ formation. In particular, the putative PQ biosynthetic cluster was upregulated in Shiraia sp. Slf14 cultured in fructose, leading to a significant increase in PQ production. The results of real-time qRT-PCR and related enzyme activities were also consistent with those of transcriptome analysis. These findings provide a remarkable insight into the underlying mechanism of PQ biosynthesis and pave the way for improvements in PQ production by Shiraia sp. Slf14.

Biotransformation of 2-Aminobenzothiazole by a newly isolated Rhodococcus sp. YQ-4 eliminates its toxicity to sensitive organisms

Fuyu Song,Jiaxi Liu,Qi Wu,Aiqin Diao,Yinhu Jiang,Xin Yan,Na Li,Jian He,Jiguo Qiu

An efficient Rhodococcus sp. YQ-4 strain degraded 98% of 2-ABT within 24 h. • A new intermediate metabolite (2-hydroxybenzothiazole) was identified. • A novel 2-ABT biotransformation pathway was proposed. • Biodegradation of 2-ABT was a detoxification process. 2-Aminobenzothiazole (2-ABT), a representative heterocyclic compound originating from tire wear particles, was widely detected in diverse environmental matrices. Due to its persistence and potential toxicity, the removal of 2-ABT from contaminated environments has become an increasing environmental concern. However, studies on the efficient biodegradation of 2-ABT by pure cultures remain limited. In this study, the abundance of Rhodococcus increased significantly following enrichment and acclimation. Subsequently, a 2-ABT-degrading pure culture, designated Rhodococcus sp. YQ-4, was isolated. Under the optimal conditions of pH 7.0, 1% NaCl, and 30 °C, strain YQ-4 achieved a removal rate of 98% for 50 μM 2-ABT within 24 h. A key metabolic intermediate, 2-hydroxybenzothiazole (2-OBT), was identified by liquid chromatography-time-of-flight mass spectrometry (LC-TOF-MS). Based on these results, a possible initial biotransformation pathway involving deamination and hydroxylation at the C2 position of the thiazole ring was proposed. Furthermore, toxicity bioassays involving the green alga Chlamydomonas ellipsoidea and mung bean ( Vigna radiata ) demonstrated that the biodegradation of 2-ABT by strain YQ-4 significantly alleviated its phytotoxicity, thereby restoring the growth and physiological vigor of the test species. Collectively, this study provided a novel bacterial resource for the bioremediation of 2-ABT-contaminated environments and offered new insights into the catabolic pathway associated with emerging pollutants.

Gut bacterial-mediated adaptation of Streltzoviella insularis to a constitutive defense compound in an ash tree (Fraxinus sp.)

Qingjie Cao,Can Wang,Tuuli-Marjaana Koski,Yingqi Zhang,Zenghe Bu,Limin Zhang,Jianghua Sun

Gut microbiota play a key role in an insect's ability to overcome host tree defenses. Streltzoviella insularis (Lepidoptera: Cossidae) is a generalist wood-boring pest in China, and Fraxinus chinensis var. rhynchophylla is its preferred host in Xiong'an New Area. Previous mechanistic studies have demonstrated that S. insularis can tolerate and degrade host defense compounds via its gut microbes. However, whether hydroxytyrosol (a constitutive phenolic compound potentially involved in browning reactions) functions as a key defense of Fraxinus chinensis var. rhynchophylla against woodborers, and whether the gut microbiota of attacking S. insularis can degrade hydroxytyrosol, remains unclear. We determined the hydroxytyrosol content in S. insularis -infested and uninfested Fraxinus chinensis var. rhynchophylla . We further evaluated the effects of hydroxytyrosol on larval survival, tested the ability of larval gut bacteria to degrade hydroxytyrosol, and identified the resulting degradation products. Although hydroxytyrosol was toxic to S. insularis , its concentrations were lower in larval galleries than in uninfested trees, suggesting that S. insularis can degrade this compound. Gut bacteria from S. insularis degraded hydroxytyrosol both in vivo and in vitro , and Serratia sp. showed the strongest hydroxytyrosol degradation activity. Metabolomics further indicated that gut bacteria may degrade hydroxytyrosol via the tricarboxylic acid cycle pathway. Because this pathway yields ATP, the bacterial degradation products of hydroxytyrosol may provide an energy source for the insect hosts or their gut microorganisms.

Mechanism and application potential of short-chain dehydrogenase in Rhodococcus sp. for the efficient degradation of 17α-ethynylestradiol

Kejian Tian,Jinming Gu,Lang Xie,Dandan Zhou,Fanxing Meng,Pengfei Hao,Yue Yu,Fan Li,Hongliang Huo,Shi Xie

17α-Ethynylestradiol (EE2) is a ubiquitous synthetic estrogen of global concern. Due to its recalcitrance to biodegradation, EE2 is often insufficiently removed during wastewater treatment and consequently released with effluents, posing significant ecological risks and potential threats to human health. In this study, Rhodococcus equi DSSKP-R-001 ( R. equi -001) was identified as one of the most potent EE2-degrading bacterium, achieving complete degradation of EE2 at low concentrations. Transcriptomic analysis revealed substantial upregulation and high functional similarity of genes encoding short-chain dehydrogenase/reductase (SDR) during EE2 degradation. Among these candidates, sdr R was functionally validated as the most catalytically efficient, achieving 97.88 % EE2 removal. Molecular docking analyses demonstrated that the sdr R -mediated conversion of EE2 to estrone (E1) is facilitated by multiple key amino acid residues through substrate binding, hydrogen bonding, and hydrophobic interactions. Furthermore, the sdr R -harboring genetically engineered bacterium (GEB) exhibited superior biodegradation potential, achieving up to 96.48 % EE2 removal individually and enabling complete elimination when applied for bioaugmentation. Concurrently, bioaugmentation with GEBs reshaped the sludge microbial community by optimizing interspecies interactions and increasing the abundance of denitrifying and phosphorus-accumulating microorganisms, thereby improving overall pollutant removal performance. Overall, this study expands the microbial and enzymatic repertoire for EE2 degradation and demonstrate the potential of genetic engineering strategies for improving the removal of recalcitrant micropollutants in biological wastewater treatment.

Integrated multi-omics analysis reveals the mechanism of birnessite-enhanced phenanthrene degradation by Novosphingobium sp. HDJX-2 bacteria

Junxin Jia,Yaqi Jiao,Jiaying Gu,Chunnan Zhou,Qin Zhou,Hui Li,Sa Wang,Hongtao Shi,Li Li

Phenanthrene (PHE) is a typical polycyclic aromatic hydrocarbon (PAH) pollutant characterized by biological toxicity and environmental persistence. To enhance its degradation efficiency, a composite system combining birnessite with Novosphingobium sp. HDJX-2 was established to investigate the synergistic mechanism between minerals and functional bacteria. Gas chromatography-mass spectrometry (GC–MS), coupled with transcriptomic and metabolomic analyses, was employed to elucidate the molecular regulation and metabolic pathway reconstruction involved in PHE degradation in the composite system. The results demonstrated that PHE was metabolized by the strain through a series of reactions, including aromatic ring dioxygenation, ring cleavage, and decarboxylation, leading to the formation of acetyl-CoA via the phthalic acid pathway and ultimately achieving complete mineralization through the tricarboxylic acid (TCA) cycle. This degradation process was accompanied by extensive transcriptional regulation and metabolic pathway enrichment, with 967 differentially expressed genes and 125 significantly altered metabolites identified. Further analysis revealed the upregulation of genes associated with biofilm formation ( rcsA , 2.31-fold upregulation) and energy metabolism ( Rbs , 2.47-fold upregulation), along with enhanced pathways, such as arachidonic acid metabolism (1.85-fold upregulation) and riboflavin metabolism (1.13-fold upregulation). These changes contributed to improved membrane transport, oxidative stress response, and energy production, thereby significantly promoting PHE degradation. Notably, pathways related to amino acid metabolism and carbon fixation were significantly enriched, further enhancing the system's degradation capacity. This study established an efficient and stable mineral–microbial synergistic system and revealed its multilayered regulatory mechanisms in PAH bioremediation, offering theoretical insights and practical strategies for ecological restoration.
