This study aims to elucidate the synergistic antibacterial mechanism of benzyl isothiocyanate (BITC) and resveratrol (RES) on Staphylococcus aureus ( S. aureus ) at the transcriptional level. Compared with the individuals, the combination of BITC and RES (BITC_RES) reduced S. aureus growth, inhibited biofilm formation, and increased cell membrane disruption. The transcriptomic results showed that the BITC_RES group presented 245 and 1150 more DEGs than the BITC group and the RES group, respectively. In addition, some other key genes in the BITC_RES group, including serine protease ( splA , splE ), Sae regulatory system ( saeR , saeS , tsaE , sau300 ), accessory gene regulator protein C ( agrC ), cysteine protease ( sspB ), glutamyl endopeptidase ( sspA ), and hemolysin toxin family-related genes ( hly , lukDv , lukEv ), and the relative expression of these 12 genes was downregulated by 2.2–259.8-fold, 0.8–259.8-fold and 1.2–158.2-fold greater than those in the BITC group and the RES group, respectively. Finally, a synergistic antimicrobial effect of this combination was also observed in fresh lean beef at 4 °C and 25 °C. These findings provide information for future studies on the synergistic antimicrobial effects of BITC and RES on S. aureus .

Staphylococcus aureus (S. aureus), a bacterium ubiquitously distributed in natural environments and commonly colonizing human skin and mucous membranes, is a highly pathogenic agent that poses significant threats to human health. Consequently, the development of accurate and efficient detection methods for S. aureus is of utmost importance in clinical and public health contexts. Flow chromatography is widely used for the detection of S. aureus because of its short detection time and low demand for labor and material resources. Herein, we fabricated test strip sensors with dual recognition of IgG and cefradine (CE) for S. aureus detection, achieving a high degree of specificity and a low detection limit (102 CFU·mL⁻¹). Meanwhile, by capturing images of the T-line with a smartphone to quantify its fluorescence intensity, this method enabled both qualitative and quantitative detection of S. aureus. The detection process was simple, which required no personnel training, and takes only 40 minutes, offering fast and convenient analysis. To prove the practicability, we applied the test strip sensors to orange juice , milk and beef, and they were highly sensitive (orange juice: 102 CFU·mL-1, milk: 104 CFU·mL-1, beef: 104 CFU·mL-1), reproducible (RSD<5.4%) .Therefore, we believe the test strip sensors have a bright application prospect in the field of rapid detection of S. aureus.

Staphylococcus aureus (S. aureus) is globally recognized as a foodborne pathogen that causes food poisoning by secreting enterotoxins. In this study, a novel dual-recognition strategy for detecting S. aureus was developed, employing vancomycin-functionalized magnetic beads (Van-MBs) for target capture and canine immunoglobulin G (IgG) to selectively and sensitively bind to staphylococcal protein A (SPA) on the bacterial surface. Dual-recognition strategies based on vancomycin and porcine IgG have previously been developed for the detection of S. aureus. However, porcine IgG can also bind to protein G, which poses the risk of generating false-positive signals from protein G-producing Streptococci. In contrast, canine IgG demonstrates a higher affinity for protein A than for protein G. When S. aureus is present in the sample matrix, the Van-MBs and biotin-labeled canine IgG successfully form a sandwich complex. Then, streptavidin-conjugated horseradish peroxidase (HRP) is added, which binds to the sandwich structure. HRP then catalyzes the oxidation of 3,3’,5,5’-tetramethylbenzidine (TMB), resulting in the formation of a colorimetric signal that was proportional to the concentration of S. aureus within the concentration range of 6.3 × 103–4.0 × 105 CFU/mL with a detection limit of 1.9 × 103 CFU/mL. By substituting porcine IgG with canine IgG, this method eliminates cross-reactivity with protein G-expressing Streptococci, achieving specificity over non-target species. This approach highlights its potential for application in diverse sample matrices, including clinical and biological specimens.

Ultrasensitive quantitative detection of Staphylococcus aureus ( S. aureus ) is critically important for preventing food-borne illnesses caused by this pathogen. In this study, an electrochemical biosensor for accurate detection of S. aureus was developed for the first time by in situ ring-opening metathesis polymerization (ROMP) of bacterial surface proteins. In brief, the glassy carbon electrode (GCE) was functionalized via electrochemical reduction of aryl diazonium salts to provide binding sites for specific capture probe-aptamer. 2,2,6,6-tetramethylpiperidin-4-ylamine (TEMPO) was modified on the ROMP polymer by post-functionalization method to achieve signal amplification. In the concentration range of 1.0 × 10 1 CFU/mL ∼ 1.0 × 10 8 CFU/mL, the detection limit of the electrochemical biosensor for S. aureus was 2.2 CFU/mL. In addition, the sensor has excellent reproducibility and recognition ability, and can accurately detect S. aureus in the presence of a variety of bacteria. To validate practical utility, the biosensor was successfully applied to detect S. aureus in real food samples—including milk and orange juice—yielding high recovery rates and reliable quantification. The sensor adopts a mild and environmentally friendly ROMP signal amplification strategy and has excellent analytical performance, which has important potential in the field of accurate identification of food-borne pathogens.

A colorimetric aptasensor based on the dual recognition of magnetic carbon dots (M-CDs) and copper-based metal–organic frameworks (Cu-MOFs) was constructed for the visual detection of Staphylococcus aureus (S. aureus). Aptamer-modified Cu-MOFs could specifically identify target bacteria and exhibited peroxidase-like activity by catalyzing the colorimetric reaction of the 3,3′,5,5′-tetramethylbenzidine–H2O2 (TMB–H2O2) system. M-CDs electrostatically attracted bacteria because of their strong positive electrical properties and offered better capturing performance compared with magnetic beads owing to their smaller size. Visual detection of the target was achieved using the color change of the reaction catalyzed by the resuspension of the sandwich-structured complex. The bacteria were sensitively detected with a limit of detection (LOD) of 1 CFU mL−1 using the peroxidase catalytic activity of Cu-MOFs. Non-target bacteria produced negative results, demonstrating the excellent selectivity of the proposed aptasensor. The strategy showed good recovery in real sample detection, showing great potential for visual monitoring of pathogenic bacteria, and could provide a reference for further development of novel sensing platforms for bacterial detection.

Nanomaterial-based synergistic antibacterial agents are considered as promising tools to combat infections caused by antibiotic-resistant bacteria. Herein, multifunctional mesoporous silica nanoparticle (MSN)-based nanocomposites were fabricated for synergistic photothermal/photodynamic/chemodynamic therapy against methicillin-resistant Staphylococcus aureus (MRSA). MSN loaded with indocyanine green (ICG) as a core, while Prussian blue (PB) nanostructure was decorated on MSN surface via in situ growth method to form a core-shell nanohybrid (MSN-ICG@PB). Upon a near infrared (NIR) laser excitation, MSN-ICG@PB (200 μg mL −1 ) exhibited highly efficient singlet oxygen ( 1 O 2 ) generation and hyperthermia effect (48.7℃). In the presence of exogenous H 2 O 2 , PB with peroxidase-like activity promoted the generation of toxic hydroxyl radicals (•OH) to achieve chemodynamic therapy (CDT). PTT can greatly increase the permeability of bacterial lipid membrane , facilitating the generated 1 O 2 and •OH to kill bacteria more efficiently. Under NIR irradiation and exogenous H 2 O 2 , MSN-ICG@PB (200 μg mL −1 ) with good biocompatibility exhibited a synergistic antibacterial effect against MRSA with high bacterial killing efficiency (>98 %). Moreover, due to the synergistic bactericidal mechanism, MSN-ICG@PB with satisfactory biosafety makes it a promising antimicrobial agent to fight against MRSA.

The global spread of Staphylococcus aureus ( S. aureus ) not only causes significant economic losses but also poses a serious threat to public health. Consequently, there is an urgent need for multidimensional strategies to develop antimicrobial dressings to combat bacterial infections. In response, we have developed a plant polysaccharide antibacterial hydrogel formed through the self-assembly of edible Kudzu powder (KP) and the natural star molecule berberine (BBR). Rheological tests show that natural polysaccharide KP-BBR hydrogel (BBR@KP) exhibits excellent injectability and adhesion. And the degradation of the hydrogel exceeded 90 % within 3 days. The synergistic effect of these two ingredients enhances the antibacterial activity of BBR and can increase the MIC of BBR to 0.05 mM. Specifically, KP promotes the affinity of the Phosphoenolpyruvate Phosphotransferase System (PTS) of S. aureus , enabling KP, with its bioadhesive properties, to adhere to the bacterial surface and continuously release BBR. Subsequently, BBR effectively exerts its antibacterial effect by inhibiting the synthesis of histidine and isoleucine. Furthermore, the BBR@KP hydrogel exhibits negligible cytotoxicity and hemolytic toxicity, underscoring its favorable biosafety profile. This synergistic natural antibacterial hydrogel, formulated through a green and straightforward methodology, not only holds promise for broad clinical applications but also provides novel perspectives for the utilization and development of plant polysaccharides in the biomedical field.

Bacterial infection may lead to organ damage and even death. Copper ions have attracted much attention in bacterial-infected wound treatment due to their ability to kill bacteria and promote neovascularization. However, the unsatisfied copper delivery for efficient bacteria removal and unexpected side effects due to copper accumulation in normal tissues limit its application. Here, we developed a copper–gold alloy nanorod (CuAu NR) as a photothermal agent to treat chronic wounds and keratitis with Methicillin-resistant Staphylococcus aureus (MRSA) infection. The CuAu NRs demonstrated a similar surface plasmon resonance effect as gold nanorods (Au NRs), a rod-like nanoparticle with unique plasmonic properties, and could be effectively heated up under near-infrared laser (808 nm) irradiation. The pH and temperature dual modalities-triggered copper release improved the healing effect with reduced biosafety concerns. In both MRSA-infected wounds in the mice model and MRSA-infected keratitis in the rabbit model, the CuAu NRs showed significant antibacterial activity, enhanced angiogenesis, and improved healing under laser irradiation. Together with excellent biocompatibility, our CuAu NRs can serve as a potential antibacterial material for clinical use.

A direct and ultra-sensitive surface-enhanced Raman scattering (SERS) immunoassay method is introduced for the detection of Escherichia coli and Staphylococcus aureus . This methodology is based on a sandwich-structured complex probe (SCP) mechanism, combined with target-induced strand displacement. Moreover, by leveraging the amplified SERS signal from gold nanoparticles (AuNPs) corresponding to an increase in bacterial count, we achieve quantitative determination. The SCP demonstrates remarkable specificity, sensitivity, and anti-interference capability in bacterial detection. The detection limits for both bacterial strains are as low as 10 CFU/mL. In our selectivity tests, all peak intensities had standard deviations ( n = 3) below 6%. Recoveries in normal human serum were 101–110% for E. coli and 96–101% for S. aureus . In milk, the recoveries were 102–105% for E. coli and 100–105% for S. aureus , respectively, demonstrating a high level of accuracy and resistance to interference. In addition, the SCP offers a dual-detection capability, enabling simultaneous diagnosis of multiple targets, which greatly simplifies the testing procedure. The findings underscore that this immunoassay platform fulfills the demand for rapid and precise pathogenic bacterial diagnosis, holding substantial potential for practical applications. The increasing number of infections caused by pathogenic bacteria has severely affected human society. More and more deaths were originated from Gram-positive methicillin-resistant Staphylococcus aureus (MRSA) infection each year. The potential and excellent bacteriostatic activity and resistance to biofilm formation of pillar[5]arene with different functional groups attract important attention to further study the relationship between antimicrobial activity and cytotoxicity by varying the length of the hydrophobic chain, the number of positive charges, and the hydrophobic/hydrophilic balance of the molecule. In this work, four pyridinium-based cationic pillar[5]arene (PPs) with linear aliphatic chains of different lengths were synthesized. After systematic characterization, their inhibition activities against S. aureus were investigated. It revealed that PP6 (six methylenes in each linker) exhibited excellent inhibition activity against S. aureus (ATCC 6538) with a minimum inhibitory concentration (MIC) of 3.91 μg/mL and a minimum bactericidal concentration (MBC) of 62.50 μg/mL. As expected, PP6 exhibited the strongest antibiofilm ability and negligible antimicrobial resistance even after the 20th passage. A study of the action mechanism of selected PPs on the bacterial membrane depolarization and permeability by transmission electron microscopy (TEM) disclosed that the cationic pyridine groups of PPs inserted into the negatively charged bacterial membranes, thereby leading to membranolysis, cytoplasmic content leakage, and cell death. Importantly, PPs all showed very low toxicity to mammalian cells (L929 and HBZY-1), which provided a significant reference for the construction of hypotoxic antibacterial biomaterials for multiple drug-resistant bacteria based on pyridinium-grafted cationic macrocycles with controllable hydrophobic chain lengths.

Surgical site infection (SSI) with instrumentation is an undesired and troublesome complication in orthopaedics. Surgical debridement is currently the main treatment option. However, the poor prognosis remains the major problem and over 40% of patients needed additional surgery to remove the implant. To eliminate SSI, antibacterial and biocompatible implants are urgently needed. Herein, a novel biocompatible polydopamine (PDA)-IR820-Daptomycin (DAP) coating with triple therapy functions was successfully fabricated to enhance the anti-infection ability and osteogenic property of titanium implants. In which, PDA served as a biocompatible binder to immobilize IR820 and DAP, and possessed photothermal and osteogenic properties. The releasing of DAP inhibited the growth of Staphylococcus aureus and damaged the bacterial membrane. Meanwhile, after 808 nm laser irradiation, the combination of hyperthermia generated by PDA and singlet oxygen ( 1 O 2 ) produced by IR820 eradicated the biofilm noninvasively. Furthermore, this coating successfully accelerated glutathione (GSH) oxidation and destroyed the bacterial membrane, reaching an anti-bacterial efficiency of 97.2% in vitro . Additionally, the coating significantly stimulated the proliferation and osteogenic differentiation of rat bone marrow stem cells (rBMSCs). In vivo, Ti-PDA-IR820-DAP reached an anti-bacterial efficiency of 97.9% in a S. aureus biofilm-infected rat model with 15 min of irradiation at 50 °C and drastically promoted the bone-implant osseointegration in a rat femur condyle defect model. The present study demonstrates that combining antibiotics and phototherapy represents a feasible way to treat bone-implant-associated infections.

Staphylococcus aureus ( S. aureus )-infected osteomyelitis is a deep tissue infection that cannot be effectively treated with antibiotics. Microwave (MW) thermal therapy (MTT) and MW dynamic therapy (MDT) based on MW-responsive materials are promising for the therapy of bacteria-infected osteomyelitis occurring in deep tissues that cannot be effectively treated with antibiotics. In this work, the MW-responsive system of carbon nanotubes (CNTs)/Prussian blue (PB)/puerarin (Pue) with stable network-like structures is constructed. The PB is grown in situ on the CNTs, and its introduction not only reduces the aggregation of the network-like structures of the CNTs, but the large specific surface area and mesoporous structure can also provide many active sites for the adsorption of oxygen and polar molecules. Pue is a natural anti-inflammatory material that reduces inflammation at the infection site. The composite of the CNTs and PB avoids the skin effect and thus can improve dielectric and reflection losses. The MW thermal response of CNTs/PB/Pue is mainly due to the occurrence of reflection loss, dielectric loss, multiple reflections and scattering, interface polarization, and dipole polarization. In addition, under MW irradiation, the CNTs/PB/Pue can produce reactive oxygen species (ROS), such as singlet oxygen ( 1 O 2 ), hydroxyl radical (·OH).
