Astrocyte elevated gene-1 (AEG-1) oncogene is a notorious and evolving target in a variety of human malignancies including osteosarcoma. The RNA interference (RNAi) has been clinically proven to effectively knock down specific genes. To successfully implement RNAi in vivo, protective vectors are required not only to protect unstable siRNAs from degradation, but also to deliver siRNAs to target cells with controlled release. Here, we synthesized a Zein-poly( l -lysine) dendrons non-viral modular system that enables efficient siRNA-targeted AEG-1 gene silencing in osteosarcoma and encapsulation of antitumor drugs for controlled release. The rational design of the ZDP integrates the non-ionic and low immunogenicity of Zein and the positive charge of the poly( l -lysine) dendrons (DPLL) to encapsulate siRNA and doxorubicin (DOX) payloads via electrostatic complexes and achieve pH-controlled release in a lysosomal acidic microenvironment. Nanocomplexes-directed delivery greatly improves siRNA stability, uptake, and AEG-1 sequence-specific knockdown in 143B cells, with transfection efficiencies comparable to those of commercial lipofectamine but with lower cytotoxicity. This AEG-1-focused RNAi therapy supplemented with chemotherapy inhibited, and was effective in inhibiting the growth in of osteosarcoma xenografts mouse models. The combination therapy is an alternative or combinatorial strategy that can produce durable inhibitory responses in osteosarcoma patients.

BACKGROUND l -Lysine ( l -Lys), an essential amino acid, serves as a core indicator for evaluating food nutritional quality. Although naturally abundant in ingredients such as soybeans, its content must be precisely regulated during processing to meet nutritional standards. Moreover, improper processing and storage can induce l -Lys degradation or conversion, compromising both nutritional value and food safety. Given its pivotal role in nutritional balance and quality assurance, there is an urgent need for simple, rapid, and highly sensitive analytical methods for l -Lys detection. RESULTS An innovative ‘off–on’ fluorescent sensor tailored for food-related applications was developed, utilizing CdTe quantum dots as the fluorophore and dodecyl trimethyl ammonium bromide self-assembled citrate-capped gold nanoparticles (DTAB-AuNPs) as the quencher for the rapid detection of l -lys. During the detection process, l -lys served as a binding bridge, facilitating the aggregation of DTAB-AuNPs. This reduction inhibited fluorescence resonance energy transfer between the fluorophore and the quencher, leading to fluorescence recovery. The degree of aggregation-induced fluorescence recovery exhibited a linear correlation with the concentration of l -lys within the range of 0.5–10 μ m ( R = 0.999). This newly developed sensing method demonstrated an impressive limit of detection of 3.22 n m . It was successfully applied to detect l -lys in various food-relevant samples, including milk, soybean, plasma and compound amino acid oral solution used in food-related nutritional supplements. CONCLUSION The results highlight its great potential for practical applications in multiple fields such as quality control of dairy and agricultural products, as well as clinical diagnosis related to nutritional status assessment. © 2026 Society of Chemical Industry.

Acne is a chronic skin disorder closely associated with the over-proliferation of P. acnes within hair follicles. To avoid the overuse of antibiotics and address the low efficiency of conventional transdermal drug delivery, this study reported a chitosan oligosaccharide/gallic acid-modified ε‑poly‑L‑lysine/polyvinyl alcohol microneedles (CEGP MNs) patch for acne treatment. The patch possessed sufficient needle length (600 μm) and mechanical strength (0.8 N/needle) to penetrate the stratum corneum and dissolve rapidly in the dermis, thereby releasing the active ingredients COS and EPL‑GA. Experiments demonstrated that COS and EPL‑GA effectively kill bacteria through their enhanced antibacterial action, with an antibacterial rate exceeding 80%, thus alleviating acne‑related inflammation. Furthermore, owing to the antioxidant and anti‑inflammatory properties of GA, CEGP MNs significantly reduced the expression levels of TNF‑α, IL‑6, and IL‑1β. Meanwhile, CEGP MNs up‑regulated the expression of IL‑10 and maintained VEGF at a moderate level to create favorable conditions for the acne repair phase, helping to reduce pigmentation and prevent scar formation. Consequently, in a severe acne mice model induced by P. acnes, the biocompatible CEGP MNs exhibited promising therapeutic effects. This study proposes an effective acne treatment strategy based on the transdermal delivery of natural compounds.

Food-borne advanced glycation end products (AGEs) are highly related to various irreversible diseases, and Nε-(carboxymethyl)lysine (CML) is the typical hazardous AGE. The development of feasible strategies to monitor and reduce CML exposure has become desirable to address the problems. In this work, we proposed magnetically controlled nanorobots by integrating an optosensing platform with specific recognition and binding capability, realizing specific anchoring and accurate determination as well as efficient scavenging of CML in dairy products. The artificial antibodies offered CML imprinted cavities for highly selective absorption, and the optosensing strategy was designed based on electron transfer from red emissive self-assembling peptide dots (r-SAPDs) to CML, which was responsible for the identity, response, and loading process. The r-SAPDs overcame the interference from autofluorescence, and the limit of detection was 0.29 μg L–1, which bestowed accuracy and reliability for in situ monitoring. The selective binding process was accomplished within 20 min with an adsorption capacity of 23.2 mg g–1. Through an external magnetic field, CML-loaded nanorobots were oriented, moved, and separated from the matrix, which enabled their scavenging effects and reusability. The fast stimuli-responsive performance and recyclability of the nanorobots provided a versatility strategy for effective detection and control of hazards in food.

Poly-L-lysine (PLL) is a promising candidate for the treatment of pulmonary infection with lower occurrence of drug-resistance due to its unique antibacterial mechanisms. Dry powder inhalations (DPIs) are considered as the first choice for formulating PLL to treat pulmonary infection on account of direct delivery and satisfactory stability. However, hygroscopicity of PLL limited its therapeutic effect on pulmonary infection when PLL developed into DPIs. The hygroscopicity caused two obstacles including the low drug deposition in the lower respiratory tract and undesirable aerosolization performance deterioration. In this study, PLL was co-spray-dried with L-leucine (LL) to achieve moisture-resistance and desirable aerosolization performance. The ratio of PLL and LL was optimized to obtain particles with different morphology, hygroscopicity and aerodynamic properties. The obtained PLL DPIs were suitable for inhalation with a corrugated surface formed by hydrophobic LL. The anti-hygroscopicity, aerosolization performance and rheological properties of P 2 DPIs were optimal when PLL:LL = 85:15. The DPIs particles were stable after being stored at high relative humidity (60 ± 5%), and their superiority in treating pulmonary infections was also proved by in vitro and in vivo experiments. The established PLL DPIs were proved to be a feasible and desirable approach to treat pulmonary infections.

In this study, we aimed to investigate the influence of lysine-assisted ultrasonic treatment on the conformation, and interfacial and emulsification properties of sea bass myofibrillar proteins at the oil–water interface. It was observed that the modification treatment with lysine (1 g/L) combined with low-intensity ultrasound (200 W) enhanced the interfacial pressure and penetration rate. This enhancement could be attributed to the significant structural unfolding of myofibrillar proteins located at the oil–water interface (increase in β-sheets and reduction in α-helices), which exposed the hydrophobic groups. The rate of interfacial protein adsorption subsequently increased, owing to the strengthening of protein–protein and protein–oil phase interactions. Additionally, the interfacial swelling rheology experiment showed an enhancement in the ordered structure of interfacial proteins, aiding in the development of a protein gel network structure. Consequently, the elastic modulus of the oil–water interface was larger, and the droplet interfacial structure was tight, which facilitated uniform distribution and improved the emulsification properties of the droplets, ultimately enhancing the ability of the emulsions to be stored at room temperature.

The inhibitive effect and inhibition mechanism of l -lysine derivatives, namely α- N -benzyloxycarbonyl- l -lysine (α-NL) and ε- N -benzyloxycarbonyl- l -lysine (ε-NL), on the corrosion of carbon steel in 0.5 M H 2 SO 4 solution were investigated by electrochemical technique, weightlessness test, scanning electron microscope, atomic force microscope, infrared attenuated reflectance spectroscopy and quantum chemical calculation. The results showed that α-NL and ε-NL as moderate cathodic inhibitors mainly inhibited the cathodic reaction of carbon steel in 0.5 M H 2 SO 4 solution, and the terminal substituted ε-NL showed better corrosion inhibition than the α- N -benzyloxycarbonyl protected α-NL. Their inhibition efficiencies increased with the increase in inhibitor concentration, and the inhibition efficiency of α-NL and ε-NL with concentration of 10 mmol can reach 53.4% and 61.5%, respectively, but decreased with rising temperature. The adsorption of investigated inhibitors on the surface of carbon steel obeyed the modified Langmuir isotherm and El-Awady thermodynamic-kinetic model, and the thermodynamic and kinetic parameters were determined to discuss the mechanism of inhibition of α-NL and ε-NL. Moreover, Quantum chemical calculation gave further insight into the inhibition mechanism of α-NL and ε-NL and was employed to reveal the reasons for their differences in inhibition performance.

The effects of l -histidine (His) and l -lysine (Lys) on the emulsifying and interfacial properties of porcine myofibrillar proteins (MPs) at low/high ionic strength were investigated to improve physical stability of MPs emulsion at low ionic strength. Results showed that His and Lys increased emulsion stability index, and decreased creaming index at low ionic strength, but caused no significant changes in both indexes at high ionic strength. Additionaly, His and Lys increase d the emulsifying activity and apparent viscosity, but decreased the droplet size at both ionic strength. His and Lys also promoted the diffusion of MPs and increase d the interfacial pressure. Meanwhile, the interfacial loading of MPs was increased (p < 0.05) by 16.07% and 12.79% at low ionic strength, and by 8.85% and 8.24% at high ionic strength in the presence of Lys and His, respectively. Raman spectra revealed that the α-helix content of MPs was decreased (p < 0.05) by 5.05% and 11.79% at low ionic strength, and by 12.14% and 7.23% at high ionic strength in the presence of Lys and His, respectively. In summary, His and Lys enhanced the stability of MPs emulsion at low ionic strength via changing the structure and interfacial behaviour of MPs.

Environmentally responsive hydrogels show enormous potential in various applications, such as tissue engineering and drug delivery. The site-specific controlled drug delivery of hydrogels can improve the therapeutic outcome and minimize the negative side effects. In this work, enzymatically digestible hydrogels, which are composed of equally mixed L-glutamic acid (E) and L-lysine (K) polypeptides after being crosslinked by the coupling reaction between carboxyl groups and primary amines catalyzed by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide·HCl (EDC·HCl), were prepared to improve the biocompatibility through reducing the nonspecific protein adsorption and cell attachment. Hydrogels loaded with two model drugs, doxorubicin hydrochloride (DOX·HCl) (positively charged anti-cancer drug) and diclofenac sodium (negatively charged anti-inflammatory drug), showed accelerated complete drug release and full enzymatic degradation in the presence of trypsin, which was reported to be expressed in various carcinomas and inflammations. The drug release also responds to the pH change through tuning charge–charge interaction. These indicated that the prepared hydrogels were promising candidates for drug delivery systems.

Sorafenib (SF) is a first-line drug for the treatment of hepatocellular carcinoma (HCC) in clinical practice. However, acquired drug resistance tremendously limits the clinical efficacy of sorafenib in treating HCC, which has attracted great attention. PDL1 plays a crucial role in the drug resistance of HCC. Here, a codelivery system based on poly-SS-lysine modified chitosan (TAT-C-SS-P) was established and was applied to deliver sorafenib and PDL1-siRNA for synergetic HCC therapy. The successful synthesis of TAT-C-SS-P was confirmed by 1 H NMR. Additionally, sorafenib and PDL1-siRNA were successfully transported into the cells as the decreased expression of VEGF and PD-L1 by administrated with TAT-C-SS-P@SF@ PDL1-siRNA. Simultaneously, the expression of pro-apoptosis proteins cyt-c and Bax was prominently augmented, whereas the expression of anti-apoptosis protein Bcl-2 was decreased. The reduced expression of PDL1 resulted in the downregulation of P-GP and MRP1, which contributed to more sorafenib aggregation in tumor cells. Moreover, TAT-C-SS-P@PDL1-siRNA@SF efficiently promotes apoptosis of HepG2-SI cells, as the apoptosis rate rised to 73 %. A sorafenib-insensitive model was established to evaluate in vivo antitumor effect of TAT-C-SS-P@PDL1-siRNA@SF. TAT-C-SS-P@PDL1-siRNA@SF showed a tumor inhibition rate of 90.2 ± 3.5 % and no significant decrease in body weight. Taken together, our study provided compelling evidence that TAT-C-SS-P@PDL1-siRNA@SF has great potential application in the treatment of HCC clinically.

Redox-responsive theranostic nanoparticles based on poly-( N -ε-carbobenzyloxy- l -lysine) (PZLL) grafted hyaluronan (HA) (HA-g-SS-PZLL) copolymers were constructed for hepatocellular carcinoma diagnosis and therapy. These hyaluronan derivatives formed nanoparticles via a self-assembly process in aqueous solution at low concentration. Theranostic nanoparticles were obtained after loading hydrophobic doxorubicin (DOX) and superparamagnetic iron oxide (SPIO) into the core of the nanoparticles via a dialysis method. Theranostic nanoparticles exhibited redox triggered DOX release behavior, and faster DOX released from theranostic nanoparticles was detected under a reducing environment compared with slow DOX release under a normal physiological environment. Confocal laser scanning microscopy (CLSM), flow cytometry and Prussian blue staining against HepG2 cells demonstrated that HA-g-SS-PZLL theranostic nanoparticles were capable of delivering DOX and SPIO into the cells. The analysis of the anticancer effect revealed that the HA-g-SS-PZLL theranostic nanoparticles shown higher cytotoxicity against HepG2 cells than DOX-loaded HA-g-PZLL nanoparticles. In vitro T 2 magnetic resonance imaging (MRI) results exhibited that theranostic nanoparticles showed a good contrast enhancement effect, and the r 2 relaxivity value was approximately 231 Fe mM − 1 s − 1 . Finally, the theranostic nanoparticles acted as nanoprobes for HepG2 tumor-bearing BALB/c mice for in vivo MRI. Therefore, HA-g-SS-PZLL copolymers have great potential as theranostic nanoparticles for tumor-targeted diagnosis and treatment.

Prevention of infection and elimination of excess reactive oxygen species are the key factors to achieve wound healing. In this study, we have developed a multifunctional poly(ε-caprolactone) nanofiber membrane modified by ε-poly ( L -lysine) (ε-PL) with the assistance of mussel-inspired polydopamine . The morphology of the nanofibrous membrane were observed by scanning electron microscopy and transmission electron microscopy . The chemical structure characterization by Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy demonstrated the successful modification of polydopamine and the immobilization of ε-PL. Photothermal effect assay proved that the temperature of the nanofiber membrane after near infrared photothermal treatment could reach the temperature required for antibacterial effect . The release of ε-PL exhibited an initial burst which was beneficial to achieve rapid bacteria inhibition. By combining ε-PL with near infrared photothermal therapy, a synergistic antibacterial effect was achieved against Escherichia coli and Staphylococcus aureus . The results of antioxidant activity assessment indicated that polydopamine on the nanofiber membranes served as an antioxidant agent to scavenge reactive oxygen species. Additionally, the nanofiber membranes showed good cytocompatibility and suitable mechanical properties close to the skin. In summary, the prepared membrane possessed intrinsically multifunctional properties showing great potential to be used as promising wound dressing materials.
