Acidification at 90 °C prevented whey separation in acidified milk. • Acidification at 90 °C enhanced the gel strength and viscosity of acidified milk. • Fibrils appeared in the acidified milk obtained at 90 °C. • When casein micelles were acidified at 90 °C, fibrillation occurred. • Fibrillation of casein micelles improved the acidified milk stability. Whey separation usually occurs to traditional acidified milk beverages. Therefore, this work aimed to improve stability of acidified milk beverages through high-temperature acidification, because high-temperature acidification might induce fibrillation of milk protein. The stability was enhanced when milk was acidified at 90 °C. Atomic force microscopy analysis revealed that the traditional acidified milk gel was particulate, whereas fibrils appeared in acidified milk obtained at 90 °C. Moreover, when the whey protein solution and the casein micelle suspension were acidified at 90 °C, respectively, no fibrils were observed in the whey protein solution, while casein micelles became fibrils. These results suggested acidification at 90 °C mainly induced casein fibrillation and thus improved stability of acidified milk. Thioflavin T fluorescence showed that longer treatment time enhanced casein fibrillation, which aligned with improved stability of acidified milk products at 90 °C through extended acid-heat treatment. This correlation also suggested that casein fibrillation enhanced acidified milk stability. Acidification at 90 °C for ≥3 h induced fibrillation of casein micelles and improved the acidified milk stability. Download: Download high-res image (105KB) Download: Download full-size image

Chitosan (CS)-based drug delivery systems often face challenges in obtaining clinical approval. Enhancing the binding affinity of CS-based carriers to target cells, such as tumor cells, is crucial for their translation from bench to bedside. Strategies like crosslinking (NHS-disulfide-NHS) and utilizing materials like albumin have proven effective in strengthening interactions between CS-based nanoparticles and tumor cells, thereby enhancing cellular uptake (cellular avidity). This study presents a novel approach using crosslinked CS nanoparticles loaded with doxorubicin, featuring reduction-responsive drug release and albumin coating. The albumin-coated crosslinked chitosan (ACCC) was assembled via reaction between amino of CS/albumin and NHS-disulfide-NHS. The resulting ACCC nanoparticle micelles demonstrate enhanced stability of the micelles. The hybrid crosslinked network ensures stability while providing ample space for drug loading and achieves reduction-responsive drug release through disulfide bond crosslinking. Moreover, the proposed system possesses excellent biocompatibility. Compared to uncoated crosslinked chitosan nanoparticle micelles, ACCC nanoparticle micelles exhibit potent cytotoxicity against tumor cells, particularly in reducing environment. Fluorescence imaging and flow cytometry analyses confirm the enhanced cellular affinity conferred by albumin coating. These findings underscore the potential of ACCC nanoparticles in improving tumor cell adhesion and anti-tumor efficacy, thus offering promising prospects for translational medicine in CS-based drug delivery.

Branched copolymer self-assembled micelles, based on amphiphilic branched poly(styrene-alt-maleic anhydride) (BPSMA) synthesized by a one-pot method, are prepared and employed as particulate emulsifiers to stabilize the white oil-in-water Pickering emulsions. The influences of micelle concentration, branching degree, salinity, and oil type on the emulsifying performance of self-assembled micelles are comprehensively investigated, and the distinction between BPSMA micelles and linear poly(styrene-alt-maleic anhydride) (LPSMA) self-assembled micelles on the micellar structure and emulsifying performance is also studied. The results show that the branched structure of the copolymer plays an important role in emulsification. The emulsifying performance of BPSMA micelles is superior to that of LPSMA micelles, reflected in the emulsifying efficiency and the long-term stability of emulsions. It is attributed to the much better structural stability of BPSMA micelles compared to LPSMA micelles, which is probably due to the postponing of hydrolyzing of maleic anhydride and ionization of carboxyl to some extent caused by the introduction of the branched structure in copolymers. It is also worth noting that, salinity has a great effect on the emulsifying performance of LPSMA micelles, but little impact on that of BPSMA micelles. Additionally, BPSMA micelles can also well stabilize the oil-in-water emulsions with some other types of natural oils. Thus the findings are not only of theoretical interest but also of great practical application in the cosmetics area.

Canine mammary tumors (CMT) can severely compromise the life quality of the affected dogs through local recurrence, distant metastases and ultimately succumb to death. Recently, more attention has been given to the potential antimetastatic effect of maduramicin (MAD) on breast cancer. However, its poor aqueous solubility and toxicity to normal tissues limit its clinical application. Therefore, to address the drawbacks of MAD and enhance its anticancer and antimetastatic effects, MAD-loaded TPGS polymeric micelles (MAD-TPGS) were prepared by a thin-film hydration technique. The optimized MAD-TPGS exhibited excellent size distribution, stability and improved water solubility. Cellular uptake assays showed that TPGS polymer micelles could enhance drug internalization. Moreover, TPGS synergistically improved the cytotoxicity of MAD by targeting mitochondrial organelles, improving reactive oxygen species levels and reducing the mitochondrial transmembrane potential. More importantly, MAD-TPGS significantly impeded the metastasis of tumor cells. In vivo results further confirmed that, in addition to exhibiting excellent biocompatibility, MAD-TPGS exhibited greater antitumor efficacy than free MAD. Interestingly, MAD-TPGS displayed superior suppression of CMT metastasis via tail vein injection compared to oral administration, indicating its suitability for intravenous delivery. Overall, MAD-TPGS could be applied as a potential antimetastatic cancer agent for CMT.

Natural polysaccharides , represented by dextran , chitosan, and hyaluronic acid , are widely approved for use as pharmaceutical excipients and are important carrier materials for the design of advanced drug delivery systems, particularly in the field of anticancer drug delivery. The combination of stimuli-activable prodrug based chemotherapy and photodynamic therapy (PDT) has attracted increasing attention. Recent studies have verified the effectiveness of this strategy in the treatment of multiple aggressive cancers. However, in such combination, the stimuli-responsive chemotherapy and PDT have their own problems that need to be overcome. The uneven distribution of endogenous stimuli within tumor tissues makes it difficult for prodrug to be completely activated. And the inadequate tissue penetration depth of external light results in low efficiency of PDT. Aiming at these two bottlenecks, we designed a biocompatible dextran based - multi-component nanomedicine (PCL-NPs) that integrate a chemiluminescence agent luminol, a photosensitizer chlorine e6 (Ce6), and a reactive oxygen species (ROS)-activable thioketal-based paclitaxel (PTX) prodrug. The presence of overexpressed hydrogen peroxide (H 2 O 2 ) inside tumor oxidizes the luminol moiety to generate in-situ light for PDT through chemiluminescence resonance energy transfer (CRET). The singlet oxygen ( 1 O 2 ) produced in this process not only directly kills tumor cells but also amplifies oxidative stress to accelerate the activation of PTX prodrug. We propose that the PCL-NPs have great therapeutic potential by simultaneously enhancing chemotherapy and PDT in a combination therapy.

In this study, an amphiphilic polymer mPEG-HA(SA)-DNs was designed and synthesized to fabricate a multifunctional micellar system to enhance the therapeutic efficacy and reduce the toxic effect of paclitaxel (PTX). The polymer was prepared by introducing mPEG, stearic acid (SA) and 2,4-dinitrobenzenesulfonic acid (DNs) to the backbone of hyaluronic acid (HA). With above modifications, the fabricated micelles could encapsulate PTX in the core with high drug loading. The optimized PTX-loaded micelles had a mean size of 158.3 nm. Upon the effect of mPEG, the mPEG-HA(SA)-DNs micelles reduced the non-specific protein adsorption. In vitro drug release study revealed the excellent glutathione (GSH)-triggered PTX release behavior of the micelles. Moreover, GSH could trigger the detachment of DNs segment from mPEG-HA(SA)-DNs, and result in the release of SO 2 . In vitro and in vivo antitumor efficacy studies demonstrated that the PTX-loaded mPEG-HA(SA)-DNs micelles exhibited outstanding tumor suppression effect. The micelles would be potential carriers for combination cancer therapy by SO 2 and PTX.

In recent years, a polymer-free fracturing fluid is under research to minimize the conductivity damage in formation and fracture during and after fracturing. Through extensive experiments, we prepared CO2-sensitive wormlike micelles (WLMs) based fracturing fluid system using anionic surfactant NaOA (Sodium Oleate-C18H33NaO2) and TMTAD (2,6,10-trimethyl-2,6,10-triazaundecane-C11H27N3). Rheological results from viscosity variation with steady shear rate using CO2/pH/time-response after CO2 bubbling confirm that the 3NaOA-TMTAD (3:1) was found as the best one in the properties of WLMs structure and the intermolecular interactions when comparing to 1NaOA-TMTAD and 2NaOA-TMTAD. Further, the surface tension after CO2 bubbling shows a good capability to decrease about 27.01 mN·m−1 at a CMC of 0.543 mM. The cryo-TEM test justifies the occurrence of spherical and WLMs in the fluid, before and after CO2 injection. The fluid system presents an outstanding sand suspension and gel breaking property, which are the positive features during the fracturing and flow back period; respectively. Good response with the alternative changes in CO2 pressures possesses an effective influence on shear viscosity using different temperatures (25 and 85 °C) and shear rates (20, 170, and 200 s−1). Also, the results of a linear relationship and low filtration coefficients verify that the fluid system has good filtration property. Such an easy and environmentally benign fluid system may be involved as a potential candidate in fracturing in unconventional hydrocarbon reservoirs, to reduce the formation damage which is usually very common in the polymer-based fracturing fluid.

A protein–polymer conjugate combines the chemical properties of a synthetic polymer chain with the biological properties of a protein. In this study, the initiator terminated with furan-protected maleimide was first synthesized through three steps. Then, a series of zwitterionic poly[3-dimethyl(methacryloyloxyethyl)ammonium propanesulfonate] (PDMAPS) was synthesized via atom transfer radical polymerization (ATRP) and optimized. Subsequently, well-controlled PDMAPS was conjugated with keratin via thiol–maleimide Michael addition. The keratin–PDMAPS conjugate (KP) could self-assemble in an aqueous solution to form micelles with low critical micelle concentration (CMC) values and good blood compatibility. The drug-loaded micelles exhibited triple responsiveness to pH, glutathione (GSH), and trypsin under tumor microenvironments. In addition, these micelles showed high toxicity against A549 cells while low toxicity on normal cells. Furthermore, these micelles performed prolonged blood circulation.

A copolymeric micelle formulation of itraconazole (ITR-M) was prepared using linear-dendritic monoallyloxy poly (ethylene glycol)-b-poly (ε-caprolactone) (APEG-PCL) as drug carrier materials. DL and EE values of ITR-M were 5.70 ± 0.12% and 91.30 ± 1.90%, respectively. The micelle formulation enhanced the ITR solubility up to 30.42 μg/mL. In vitro release of ITR from the ITR-M was mainly drug diffusion process followed by the copolymer’s degradation. ITR-M showed similar anti-Candida albicans activity to that of crude ITR although its release of ITR was slow and continuous. The in vivo pharmacokinetic study demonstrated that the ITR-M could improve tissue distribution of ITR. In conclusion, APEG-PCL could be a potential carrier in the development of antifungal drug delivery system.

Sol-gel process was modified by reverse micelles (RMs) for the preparation of γ-Al 2 O 3 ultrafiltration (UF) membranes and their applications in the efficient rejection of methyl blue and bovine serum albumin were explored. The optimized boehmite sol with a narrower particle size distribution and better sphericity was synthesized by using cetyltrimethyl ammonium bromide RMs as microreactors and was dip-coated on α-Al 2 O 3 microfiltration substrates to prepare γ-Al 2 O 3 UF membranes. It was found that not only the sol particle size, it's distribution and morphology had also an important effect on the pore size, pore size distribution , porosity and pore volume of the membrane. Additionally, the as-prepared membranes with a MWCO of 8 kDa exhibited a narrow pore size distribution (4.8–6.8 nm), relatively high pure-water flux (30.4 L/(h m 2 bar)) and excellent rejection for bovine serum albumin and methyl blue with the maximum rejection rate of 96.2% and 96.8% respectively.

Polymeric micelles (PMs) have been applied widely to transport hydrophobic drugs to tumor sites for cancer treatment. However, the low load efficiency of the drug in the PMs significantly reduces the therapeutic efficiency. We report here that disulfide-linked camptothecin (CPT) as a kind of dimeric drug can be effectively embedded in the core of poly(ε-caprolactone)–poly(ethylene glycol)–poly(ε-caprolactone) (PCL–PEG–PCL) PMs for improving drug-loading efficiency, and PEG can be used as a hydrophilic shell. Moreover, the dimeric CPT-loaded PCL–PEG–PCL PMs exhibited excellent solubility in phosphate-buffered saline (PBS) media and significant cytotoxicity to cancer cells. Keywords:dimeric drug;camptothecin;micelles;cancer therapy
