NIR-triggered hydrogel with dynamic stiffness via ion chelation to modulate macrophage phenotypes

Peiqi Yuan,Yilun Luo,Lie Ma

The immune system plays a central role in tissue repair and regeneration. Macrophages have emerged as a primary target because of their critical roles in regulating multiple phases of tissue repair through their unique plasticity and ability to rapidly shift phenotypes. Smart biomaterials, which able to harness macrophage phenotypes on demand during the healing process, have become a promising strategy. Here, a novel integrated hydrogel with near-infrared (NIR) responsive dynamic stiffness was conducted, and found sequentially polarizing bone marrow-derived macrophages from pro-inflammatory to anti-inflammatory phenotype in situ. Under NIR irradiation, ethylene diamine tetraacetic acid released from the IR780-mixed phase change material into the hydrogel, resulting in the decrease of hydrogel stiffness in situ by chelating the calcium ion (Ca 2+ ) in the calcium alginate hydrogel. The up-regulation of arginase-1 and interleukin-10 were quantified by immunostaining and enzyme-linked immune sorbent assay, respectively. The result indicated the transformation of macrophage from the pro-inflammatory to anti-inflammatory phenotype by dynamic stiffness drop. The modulation of macrophage phenotypes by stiffness-degradation without the stimuli of cytokines offers an effective and noninvasive strategy to manipulate inflammatory or tissue-regenerative immune responses to achieve optimized healing or therapeutical outcomes. Lanthanide/Cu2–xSe Nanoparticles for Bacteria-Activated NIR-II Fluorescence Imaging of Infection

Leilei Sun,Sisi Shi,Zeming Wu,Yao Huang,Chenhui Ji,Craig A. Grimes,Xinxin Feng,Qingyun Cai

A material system enabling specific NIR-II fluorescence imaging of Gram-positive bacteria is described. The material system is based on the electrostatic binding of Cu2–xSe and vancomycin-modified NaGdF4:Nd,Yb@NaGdF4 downconversion nanoparticles (DCNPs), the fluorescence of which is weak owing to the spectral overlap of Cu2–xSe absorption with the DCNP NIR emission. The presence of Gram-positive bacteria precisely disconnects the bond between vancomycin-modified DCNPs and Cu2–xSe, thus enabling a strong fluorescent signal. In vivo studies show that the material system can be specifically activated at the site of Gram-positive bacterial infection but is essentially nonfluorescent in the area of Gram-negative bacterial infection.

Oxygen-Abundant and pH/NIR Dual-Responsive Nanocarriers for Tumor Hypoxia Reduction Therapy

Jiafeng Zhuang,Wei Zhang,Qize Xuan,Tonghao Ma,Qi Zhang,Chao Chen,Ping Wang

Pre-existing tumor hypoxia and oxygen consumption by photodynamic therapy (PDT) always generate an inadequate oxygen supply, which often contributes to other therapy resistance, especially for aerobic treatments such as chemotherapy. In order to overcome this problem, we synthesized a chitosan (CS)-capped pH-/NIR-dual responsive and perfluorocarbon-based oxygen self-enriching photodynamic therapy nanosystem which could codeliver oxygen, photosensitizer, and chemotherapeutic drug. Simultaneously, in vitro results revealed that DOX release was stimulated by the acidic pH value with CS as a pH-responsive layer and rising temperature caused by the photothermal effect with an 808 nm laser, which could effectively reduce the nonspecific toxicity caused by premature drug release. Furthermore, due to the higher oxygen-carrying capability of perfluorocarbon, the decreased expression of the hypoxia-inducible factor-1α (HIF-1α) of tumors, as well as the significantly intense photodynamic effect and longer singlet (1O2) lifetime of the loaded photosensitizer, the elevated cytotoxicity was improved. More importantly, both in vitro and in vivo results showed that drug-loaded nanoparticles with oxygen-abundant properties achieved a similar anticancer therapy in both normoxic and hypoxic conditions against the difficulty led by deficient oxygen, as well as a good cellular internalization. Summarily, all these results pointed out that such a pH-/NIR-dual responsive drug carrier could enhance the synergetic anticancer therapy of chemotherapy and PDT by alleviation of tumor hypoxia, which provided a tempting controllable and multifunctional therapeutic application.

NIR responsive AuNR/pNIPAM/PEGDA inverse opal hydrogel microcarriers for controllable drug delivery

L. Z. Liu,X. Y. Sun,Z. Y. Yan,B. F. Ye

Particle-based delivery systems have emerged as a powerful platform in controlled drug release. In the present study, we developed a new inverse opal hydrogel microcarrier system composed of gold nanorods (AuNRs) for near-infrared light (NIR) activated drug delivery. AuNRs with a desired aspect ratio were prepared by a seed-mediated growth method. The inverse opal hydrogel microcarriers composited with AuNRs were synthesized by negatively replicating silica colloidal crystal beads (SCCBs). The NIR-thermo response of the microcarriers was due to the NIR photothermal heating effect of AuNRs and the thermo responsive volume transition of poly(N-isopropylacrylamide) (pNIPAM) hydrogel. It was found that the NIR-controlled reversible shrinking response of the microcarriers was ideal for the controllable drug release with real-time self-reporting color indication.

Iodine Immobilized Metal–Organic Framework for NIR-Triggered Antibacterial Therapy on Orthopedic Implants

Wangsiyuan Teng,Zengjie Zhang,Yikai Wang,Yuxiao Ye,Eloy Yinwang,An Liu,Xingzhi Zhou,Jianxiang Xu,Chengwei Zhou,Hangxiang Sun,Fangqian Wang,Lingling Zhang,Chongguang Cheng,Peng Lin,Yan Wu,Zhongru Gou,Xiaohua Yu,Zhaoming Ye

Iodine has been known as an effective disinfectant with broad-spectrum antimicrobial potency yet without drug resistance risk when used in clinic. However, the exploration of iodine for antibacterial therapy in orthopedics remains sparse due to its volatile nature and poor solubility. Herein, leveraging the superior absorption capability of metal–organic frameworks (MOFs) and their inherent photocatalytic properties, iodine-loaded MOF surface is presented to realize responsive iodine release along with intracellular reactive oxygen species(ROS) oxidation under near-infrared (NIR) exposure to achieve synergistic antibacterial effect. Iodine is successfully loaded using vapor deposition process onto zeolitic imidazolate framework-8(ZIF-8), which is immobilized onto micro arc oxidized titanium via a hydrothermal approach. The combination of NIR-triggered iodine release and ZIF-8 mediated ROS oxidative stress substantially augments the antibacterial efficacy of this approach both in vitro and in vivo. Furthermore, this composite coating also supported osteogenic differentiation of bone marrow stromal cells, as well as improved osseointegration of coated implants using an intramedullary rat model, suggesting improvement of antibacterial efficacy does not impair osteogenic potential of the implants. Altogether, immobilization of iodine via MOF on orthopedic implants with synergistic antibacterial effect can be a promising strategy to combat bacterial infections.

A lysosomal targeted NIR photosensitizer for photodynamic therapy and two-photon fluorescence imaging

Yuping Zhou,Di Zhang,Genghan He,Chuang Liu,Yinuo Tu,Xiang Li,Qianbing Zhang,Xu Wu,Ruiyuan Liu

The design and synthesis of near-infrared (NIR) emissive fluorophores for the imaging of organelles and photodynamic therapy have received enormous attention. Hence, the development of NIR emissive fluorophores for high-fidelity lysosome targeting, two-photon fluorescence imaging, and the inducing of photo-triggered cancer-cell apoptosis is highly desirable. In this study, a novel lysosome-targeting two-photon fluorescent photosensitizer (TTRh-CN) is prepared and comprehensively investigated. TTRh-CN demonstrates near-infrared (NIR) emission, good biocompatibility, and superior photostability, and it can act as a two-photon fluorescent agent for the clear visualization of living cells and the vascular system within tissue, with deep-tissue penetration abilities. Furthermore, TTRh-CN can efficiently produce ROS in conjunction with lysosomes in situ upon light irradiation, which can damage lysosomes, up-regulate LC3 and Beclin1, increase BAX release, and induce cell apoptosis. The efficacy of TTRh-CN as a photosensitizer is explored in vivo. All these results confirm that TTRh-CN can serve as a potential platform for the two-photon fluorescence imaging of cells/tissue and for organelle-specific photodynamic therapy.

Sequential Catalytic, Magnetic Targeting Nanoplatform for Synergistic Photothermal and NIR-Enhanced Chemodynamic Therapy

Shuming Dong,Yushan Dong,Tao Jia,Fangmei Zhang,Zhao Wang,Lili Feng,Qianqian Sun,Shili Gai,Piaoping Yang

Tailored to the peculiar tumor microenvironment, Fenton reaction-based chemodynamic therapy (CDT) has attracted considerable attention for tumor treatment. However, the efficacy of CDT is highly limited by both H2O2 overproduction and the low activity of catalysts at the tumor site. Herein, a novel magnetic targeting nanoplatform (γ-Fe2O3-GOx-DMSN) has been designed by simply depositing ultrasmall γ-Fe2O3 nanoparticles and natural glucose oxidase (GOx) into the large mesopores (∼13 nm) of dendritic mesoporous silica (DMSN) spheres for near-infrared (NIR) light-enhanced CDT efficacy. In this structure, GOx can effectively consume glucose in the tumor cells to induce a decrease in the pH value and generate a considerable amount of H2O2, both of which promote subsequent Fenton reaction. These ultrasmall γ-Fe2O3 nanoparticles not only serve as an efficacious Fenton catalyst for degradation of the increased H2O2 within the tumor to produce highly toxic hydroxyl radicals (•OH) but also exhibit high photothermal therapy (PTT) efficiency upon irradiation with 808 nm light. Importantly, the generated hypothermia can significantly accelerate the Fenton process, thereby enabling a synergetic PTT/hypothermia-enhanced CDT effect. Our work manifests a proof of concept of H2O2-evolving and NIR-enhanced CDT, providing a new perspective for cancer therapy.

Pb-Doped Ag2Se Quantum Dots with Enhanced Photoluminescence in the NIR-II Window

Mengxuan Yu,Xiaohu Yang,Yejun Zhang,Hongchao Yang,Haoying Huang,Zan Wang,Jinyi Dong,Rong Zhang,Ziqiang Sun,Chunyan Li,Qiangbin Wang

Ag 2 Se quantum dots (QDs) as an effective biological probe in the second near-infrared window (NIR-II, 1000–1700 nm) have been widely applied in bioimaging with high tissue penetration depth and high spatiotemporal resolution. However, the ions deficiency and crystal defects caused by the high Ag + mobility in Ag 2 Se crystals are mainly responsible for the inefficient photoluminescence (PL) of Ag 2 Se QDs. Herein, a tailored route is reported to achieve controllable doping of Ag 2 Se QDs in which Ag is exchanged by Pb via cation exchange (CE), which is unattainable by direct synthetic methods. The Pb-doped Ag 2 Se QDs (denoted as Pb:Ag 2 Se QDs) present fire-new optical features with significantly enhanced PL intensity of 4.2 folds. Photoelectron spectroscopy confirms that Pb acts as an n-type dopant for Ag 2 Se QDs and therefore the electronic impurities provide additional carriers to fill the traps. Moreover, the general validity of this method is demonstrated to convert different sized Ag 2 Se into Pb:Ag 2 Se QDs, so that a wide range of NIR-II PL with high intensity is obtained. The bright NIR-II emission of Pb:Ag 2 Se QDs is further successfully performed in lymphatic system mapping.

NIR Stimulus-Responsive PdPt Bimetallic Nanoparticles for Drug Delivery and Chemo-Photothermal Therapy

Chun Chu,Zhihong Bao,Meng Sun,Xiaowei Wang,Hongyan Zhang,Weiguo Chen,Yang Sui,Ji Li,Yuanyuan Zhuang,Dongkai Wang

The combination of chemotherapy and phototherapy has attracted increasing attention for cancer treatment in recent years. In the current study, porous PdPt bimetallic nanoparticles (NPs) were synthesized and used as delivery carriers for the anti-cancer drug doxorubicin (DOX).[email protected]NPs were modified with thiol functionalized hyaluronic acid (HA-SH) to generate[email protected]@HA NPs with an average size of 105.2 ± 6.7 nm. Characterization and in vivo and in vitro assessment of anti-tumor effects of[email protected]@HA NPs were further performed. The prepared[email protected]@HA NPs presented a high photothermal conversion efficiency of 49.1% under the irradiation of a single 808 nm near-infrared (NIR) laser. Moreover, NIR laser irradiation-induced photothermal effect triggered the release of DOX from[email protected]@HA NPs. The combined chemo-photothermal treatment of NIR-irradiated[email protected]@HA NPs exerted a stronger inhibitory effect on cell viability than that of DOX or NIR-irradiated[email protected]NPs in mouse mammary carcinoma 4T1 cells in vitro. Further, the in vivo combination therapy, which used NIR-irradiated[email protected]@HA NPs in a mouse tumor model established by subcutaneous inoculation of 4T1 cells, was demonstrated to achieve a remarkable tumor-growth inhibition in comparison with chemotherapy or photothermal therapy alone. Results of immunohistochemical staining for caspase-3 and Ki-67 indicated the increased apoptosis and decreased proliferation of tumor cells contributed to the anti-tumor effect of chemo-photothermal treatment. In addition,[email protected]@HA NPs induced negligible toxicity in vivo. Hence, the developed nanoplatform demonstrates great potential for applications in photothermal therapy, drug delivery and controlled release. Keywords:PdPt bimetallic nanoparticles;drug delivery;chemo-photothermal therapy;anti-tumor effect

NIR-II-Activated Yolk–Shell Nanostructures as an Intelligent Platform for Parkinsonian Therapy

Yao Liu,Daoming Zhu,Jingshan Luo,Xiaojia Chen,Liqian Gao,Wei Liu,Tongkai Chen

Despite the relative severity of Parkinson’s disease (PD), to date, there has been only limited success in preventing or treating this condition owing to the low permeability of the blood–brain barrier (BBB), which makes the cerebral delivery of pharmaceutical agents very challenging. In the present study, we explored an approach to increasing BBB permeability via the use of mesoporous silica-encapsulated gold nanorods (MSNs-AuNRs) that could reliably achieve a robust photothermal effect in response to second near-infrared (NIR-II) irradiation. To test the potential anti-Parkinsonian activity, we loaded MSNs-AuNRs with the frequently utilized anti-PD agent quercetin (QCT) to yield MSNs-AuNRs@QCT. Following NIR-II irradiation, we observed a dramatical increase in QCT transfer through the BBB, indicating that MSNs-AuNRs may enhance BBB permeability via the photothermal effect. These findings were further supported by rat pharmacokinetic studies, which clearly revealed that the combination of MSNs-AuNRs and NIR-II irradiation was associated with significantly improved brain accumulation. In a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced murine model of PD, we also found that intravenous MSNs-AuNRs@QCT delivery and subsequent NIR-II irradiation was sufficient to markedly reduce the neurological abnormalities in these mice. Together, this combination offers promising potential as a nanoplatform for neuroprotective drug delivery and treatment of PD.

A thermosensitive nanoplatform for photoacoustic imaging and NIR light triggered chemo-photothermal therapy

Zikang Chen,Yinuo Tu,Di Zhang,Chuang Liu,Yuping Zhou,Xiang Li,Xu Wu,Ruiyuan Liu

Temperature-sensitive liposomes (TSLs) have received constant attention due to the release of contents around the physiological temperature, which holds great potential in the treatment of tumors. However, the development of TSLs is limited by the long-term biotoxicity and low photothermal conversion efficiency of heat-generation materials. In this study, we develop a new D–A (donor–acceptor) structure NIR absorbing dye (C720) which exhibits a high photothermal conversion efficiency (62%), good photostability, and photothermal reproducibility. Then C720 is doped in the lipid bilayer and DOX (doxorubicin) is wrapped in the core of temperature sensitive liposomes to construct a C720 and DOX-loaded thermosensitive nanoplatform (CDTSL). The as-prepared CDTSL achieves NIR light controlled drug release and can be applied for photoacoustic imaging. In vitro and in vivo studies confirm that CDTSL exhibits high tumor suppressive efficiency by synergistic photothermal therapy and chemotherapy. Our research provides fundamental insights for the rational design and preparation of a promising nanoplatform for photoacoustic imaging and chemo-photothermal therapy.

Cellulose Nanocrystal Films with NIR-II Circularly Polarized Light for Cancer Detection Applications

Di Lu,Mengfei Li,Xiaoqing Gao,Xiao Yu,Lihong Wei,Shoujun Zhu,Yan Xu

Near-infrared circularly polarized light is attractive for wide-ranging applications. However, high-performance near-infrared circularly polarized light is challenging to realize. Here, we show that left-handed chiral photonic cellulose nanocrystal (CNC) films produced from ultrasonicated suspensions enable right-handed circularly polarized luminescence with a dissymmetry factor of −0.330 in the second near-infrared window (NIR-II). We present a theoretical analysis of the adverse effect of structural defects and luminescence intensity heterogeneity on the right-handed circularly polarized luminescence glum inside the bandgap and the occurrence of left-handed circularly polarized luminescence at the band edges. We demonstrate the potential of the chiral photonic CNC films with NIR-II circularly polarized light for cancer cell discrimination. The present work identifies key scientific questions in CNC-based circularly polarized luminescence materials research.
