The Mechanism of Pectin-Mediated Thiamethoxam and Clothianidin Residues on Eggplant Peel and Enhanced Detection via Pectinase-Assisted Extraction

Yulun Chen,Linquan Wang,Aying Wen,Hang Yu,Yahui Guo,Yuliang Cheng,Weirong Yao,Shaofeng Yuan

Matrix binding poses a challenge to accurate pesticide residue detection in fruits and vegetables. This study investigated the distribution, binding status, and detection accuracy of neonicotinoid insecticides thiamethoxam (TMX) and clothianidin (CLO) in eggplant. To address the limitations posed by matrix binding, a pectinase-assisted extraction method was developed, which significantly enhanced the detectable levels of pesticides compared to the standard method and cellulase-assisted extraction. In situ Raman mapping and scanning electron microscopy revealed a strong correlation between the pectin distribution and the pesticide residues observed near the epidermal surface. Spectroscopic, nuclear magnetic resonance, and computational chemistry analyses demonstrated that TMX formed covalent bonds with pectin, while CLO interacted through hydrogen bonding. Notably, thermal processing promoted the formation of amide bonds, reducing detectable TMX levels. In summary, the optimized pectinase-assisted extraction method increased detectable TMX and CLO levels in positive samples by 25–40%, providing an effective approach for overcoming matrix binding effects.

Pectin-chitosan conjugated nanoliposome as a promising delivery system for neohesperidin: Characterization, release behavior, cellular uptake, and antioxidant property

Mohammad Rezaul Islam Shishir,Naymul Karim,Vemana Gowd,Jiahong Xie,Xiaodong Zheng,Wei Chen

Neohesperidin (NH) is a flavanone glycoside compound possessing diverse biological activities. However, NH is unstable in physiological condition and poorly soluble in water, and thereby it may show limited bioavailability. The aim of the present study was to facilitate the controlled delivery of NH in simulated gastrointestinal (GI) tract with enhanced cellular uptake using biopolymer conjugation onto nanoliposome (NL). The carrier systems were characterized by dynamic light scattering, HPLC analysis, and Fourier transform infrared spectroscopy . The biological activity was analyzed by in vitro antioxidant activity , rhodamine (Rh123)-assisted cellular uptake, and acrylamide (AA)-induced ROS model on Caco-2 cells. The results confirmed the size of nanocarriers fall in the range of 87–225 nm, well-encapsulation of NH, and conjugation of chitosan (CH) and pectin (P). In vitro digestion study revealed that P and CH-conjugated-nanoliposomes (P-CH-NL) significantly controlled the NH release following by anomalous diffusion mechanism, and preserved around 72–78% of NH in GI condition. The ABTS antioxidant activity, DPPH free radical scavenging capacity, and ferric reducing activity indicated that conjugated-nanoliposomes can preserve greater antioxidant activity after GI digestion. In vitro cellular uptake study showed enhanced Rh123 uptake by carrier systems, particularly P-CH-NL exhibited the highest uptake of around 141% followed by CH-NL (129%) and NL (100%). Cytotoxicity results also exposed that conjugated-nanoliposomes can significantly protect the human Caco-2 cells from AA-induced ROS generation. Conclusively, this work provides a promising approach for the controlled delivery of lipophilic natural products with enhanced cellular uptake at colonic epithelial cells and greater biological activity.

Shihai Zhang,Fei He,Xia Chen,Kan Ding

Crude polysaccharides were obtained from fruits of Lycium ruthenicum Murr using hot water extraction followed by ethanol precipitation. A homogeneous polysaccharide, LRP3-S1 with a relative molecular weight of 114.8 kDa was purified by anion-exchange chromatography on DEAE Sepharose ™ Fast Flow and Sephacryl S-300 HR column. Monosaccharide composition analysis revealed that LRP3-S1 was composed of rhamnose, galacturonic acid, galactose, xylose and arabinose in a molar ratio of 14.4: 17.7: 26.6: 16.4: 24.9. LRP3-S1 contained a rhamnogalacturonan I (RG-I) backbone partially substituted at C-4 of rhamnose units by side chains, which included T-linked β-D-Gal p , 1,3-linked β-D-Gal p , 1,6-linked β-D-Gal p , 1,3,6-linked β-D-Gal p , 1,5-linked α-L-Ara f , 1,3,5-linked α-L-Ara f , T-linked α-L-Ara f and T-linked β-D-Xyl p . Biological activity tests showed that LRP3-S1 could inhibit the growth of pancreatic cancer cells. In addition, LRP3-S1 could attenuate invasion ability of BxPC-3 cells and down-regulate protein expression of p-FAK, p-AKT, p-GSK-3β and p-p38 MAP kinase.

Complex pectin metabolism by Lactobacillus and Streptococcus suggests an effective control approach for Maillard harmful products in brown fermented milk

Zibian Fan,Wei Jia,An Du,Lin Shi

Harmful Maillard reaction products (HMRPs) derived from brown fermented milk pose a potential threat to human health, but the conversion mechanism during the manufacturing process remains elusive and urgently needs to be controlled. Acrylamide (FC 2.14, adjusted p -value = 0.041), 5-hydroxymethylfurfural (FC 2.61, adjusted p -value = 0.026) and methylglyoxal (FC 2.07, adjusted p -value = 0.019) were identified as the significantly increased HMRPs after browning in this study and the analysis of proteomics integrated with untargeted metabolomics demonstrated that the degradation of HMRPs was jointly accomplished by Streptococcus thermophilus and Lactobacillus bulgaricus . The galactose oligosaccharide metabolism in Streptococcus thermophilus was identified as a key biochemical reaction for HMRPs degradation, and the hydrolysates of pectin could be utilized as prebiotics for Streptococcus thermophilus . Eighteen classes of enzymes of L. bulgaricus and Streptococcus thermophilus related to energy metabolism were upregulated in the pectin-added group, indicating that the entry of acrylamide and methylglyoxal into the tricarboxylic acid cycle was accelerated. NAD-aldehyde dehydrogenase and alanine dehydrogenase are enzymes belonging to Streptococcus thermophilus, and their downregulation accelerated the efflux of acetate, which was beneficial for the proliferation of L. bulgaricus and prevented the conversion of pyruvate to l -alanine, thus facilitating the energy metabolism. The recoveries and relative standard deviations of the intraday and interday precision experiments were 89.1%-112.5%, 1.3%-8.4% and 2.1%-9.4%, respectively, indicating that the developed approach was credible. Sensory evaluation results revealed that the brown fermented milk added with pectin had a better flavor, which was due to the fact that the supplement of polysaccharide promoted the fatty acid metabolism of lactic acid bacteria and increased the aroma substances including octoic acid and valeric acid. This study provided an insight into the formation and degradation mechanism of HMRPs in brown fermented milk, aiming to reduce the intake of advanced glycation end products in the diet.

Preparation of alginate-whey protein isolate and alginate-pectin-whey protein isolate composites for protection and delivery of Lactobacillus plantarum

Xin Luo,Siqi Fan,Zhuosui He,Fangfang Ni,Chengzhi Liu,Min Huang,Lei Cai,Gerui Ren,Xuan Zhu,Qunfang Lei,Wenjun Fang,Hujun Xie

Probiotics are sensitive to external conditions, resulting in low survival rates after being ingested or during food production, transportation and storage. In order to improve the survival rate of Lactobacillus plantarum (LP) during gastrointestinal digestion, storage, and freeze-drying, alginate-whey protein isolate (ALG-WPI) and alginate-pectin-whey protein isolate (ALG-PEC-WPI) composites were employed to encapsulate LP. The encapsulation efficiency of ALG-WPI-LP and ALG-PEC-WPI-LP beads both reached more than 99 % . Scanning electron microscopy (SEM) indicated that dense and rough aggregates were formed on the surface of both composites, and attached LP cells could be observed inside the beads. The ALG-WPI and ALG-PEC-WPI composites can protect the viability of LP in simulated gastric fluid (SGF) and release the probiotics in simulated intestinal fluid (SIF). The storage stability of LP at 4 °C was improved by about 15 % in comparison with bare LP and the survival rates of LP in ALG-WPI-LP and ALG-PEC-WPI-LP powders after freeze-drying were increased by 65.37 % and 72.06 %, respectively. The formation mechanism of ALG-WPI and ALG-PEC-WPI composites was further explored by fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and thermogravimetric analysis (TGA). The ALG-WPI and ALG-PEC-WPI composites have great potential to protect and deliver probiotics in food systems.

Self-assembling gardenia pectin gel for cholestatic liver injury: Dual mechanisms of hepatoprotection via PPARα activation and gut microbial modulation

Lanjia Ao,Yujie Li,Lingling Zheng,Xiaolin Chen,Haoyuan Zeng,Wen Miao,Shuxian Yang,Tingting Zhou,Na Li,Jian-Lin Wu

Bioactive pectin is a considerable candidate for drug delivery to improve stability and bioavailability. Geniposide (GS) is a hepatoprotective compound with poor bioavailability, limiting its application. Herein, we first designed a self-assembled Gardenia Pectin Gel (GPG) from pectin-rich Gardenia jasminoides Ellis fruit extracts (GE), spontaneously encapsulating co-extracted GS. Compared with non-gelling GE, pectin gel delivery achieved enhanced stability and delayed release in vitro and increased half-time (2.6-fold, p  < 0.05) and bioavailability (2.35-fold, p  < 0.05) of GS in vivo. Moreover, GPG provided superior hepatoprotective effects over GS and GE, including reducing hepatic injury indicators (e.g. ALP level, p  < 0.01 vs GS; p  < 0.05 vs GE), inflammation and necrosis in α-naphthylisothiocyanate-induced cholestatic liver disease (CLD) mice. Mechanistically, GPG restored hepatic glycolipid metabolic homeostasis via GS-mediated PPARα activation. Meanwhile, GPG promoted gut microbes remodeling and hepatoprotective acetate and butyrate production ( p  < 0.001), producing synergistic effects. Notably, after 7-day administration, GPG outperformed ursodeoxycholic acid, a first-line CLD medicine, in the CLD mice model, demonstrating its clinical potential. Collectively, this study developed a promising self-assembled pectin-assisted formulation that enhanced the bioavailability of GS and CLD intervention, more importantly, established a theoretical foundation for clinical application of pectin-based drug delivery systems.

Fully biodegradable self-healing hydrogel prepared based on poly(aspartic acid) and pectin for drug delivery and anti-tumor therapy

Jia Boyang,Sun Weichen,Duan Wenhao,Ma Xiangbo,Wang Yong,Zhou Chengyan,Qin Jianglei

With biocompatibility and biodegradability, self-healing hydrogels prepared from biopolymers show great advantages in bioengineering and bioscience as drug delivery carrier. In this research, the biodegradable hydrogel was constructed based on poly(amino acid) of poly(aspartic acid) (PAsp) and pectin for in vivo antitumor applications as DOX release carrier. The PAsp bearing hydrazide groups and isopropyl groups (PAPH) was synthesized by ring opening of polysuccinimide (PSI) by isopropylamine and hydrazine, and used to fabricate fully biodegradable hydrogel with oxidized pectin (OPec) through dynamic acylhydrazone bond cross-linking. The regulatable mechanical strength of the hydrogel could adapt its bioapplication, and the excellent biocompatibility ensured biosafety in biomedical areas. Furthermore, the pectin backbone and the PAsp endowed the hydrogel with biodegradability both by enzyme and in vivo. The PAPH/Opec hydrogel as DOX carrier inhibited the tumor growth with the tumor weight 80% lower than the control group and reduced the in vivo toxicity of the DOX. As a result, this biodegradable PAPH/Opec hydrogel could find show great potential as sustained drug delivery vehicle in biomedical areas.

Enhanced extraction assisted by pressure and ultrasound for targeting RG-I enriched pectin from citrus peel wastes: A mechanistic study

Weiwei Hu,Huan Cheng,Dongmei Wu,Jianle Chen,Xingqian Ye,Shiguo Chen

The rhamnogalacturonan I (RG-I) region of pectin shows health benefits and novel gel applications. Commercial pectin is extracted from citrus peel in most instances, whereas they are homogalacturonan (HG)-dominated productions and the methods to extract pectin targeting RG-I are limited. To take full advantage of citrus wastes and upgrading this pectin resources, a new extraction method based on a diluted NaOH assisted by a combination of ultrasound (US) and pressure, namely manosonication (MS), was established for RG-I enriched pectin extraction. MS significantly enhanced the pectin yield (21.47 ± 0.11%) while reducing the use of NaOH reagents (50 mmol/L) and processing time (50 min). Structurally, the MS (300 kPa) resulting products achieved nearly a 2-fold greater RG-I content (58.61 mol%) than that of commercial pectin (30.12 mol%). Moreover, for the investigation of the extraction mechanism, external pressure enhanced the cavitation effect, as the calorimetric intensity increased from 1.28 W/mL (100 kPa) to 3.3 W/mL (350 kPa). A sufficient ultrasonic mechanical action allowed for obtaining RG-I enriched products with large molecular weight (9.506 × 10 4 –2.813 × 10 5 ) and low degree of methoxylation. Transmission electron microscopy (TEM) and scanning electron microscopies (SEM) verified the increased disintegration of the cell wall tissue by MS. Raman imaging and immunofluorescence labeling suggested the synergistic effect of pressure and ultrasound on enhancing the yields and RG-I content. This new method provided alternations in functional polysaccharides extraction, and the yield and structure of citrus RG-I enriched pectin strongly indicates the potentials of MS in other RG-I rich vegetables applications.

Preparation of amphoteric double network hydrogels based on low methoxy pectin: Adsorption kinetics and removal of anionic and cationic dyes

Yufan Huang,Bihua Shen,Chenmin Zheng,Bingqing Huang,Guoguang Zhang,Peng Fei

The objective of this research was to devise a functional hydrogel was synthesized using pectin (PE), acrylic acid (AA), dimethyldiallyl ammonium chloride (DC), and polyvinyl alcohol (PVA), designed to adsorb both cationic and anionic dyes concurrently. The low methoxy pectin formed double network hydrogel through chemical and physical crosslinking with AA and PVA respectively. DC is combined into the hydrogel system through copolymerization reaction. Analysis of hydrogel's physicochemical properties was conducted using techniques such as infrared spectroscopy, texture analysis, thermogravimetry , and scanning electron microscopy. Dyes adsorption studies showed that the LP/AA/DC/PVA-2 hydrogel, prepared at the molar ratio of AA to DC of 1:2, exhibited higher adsorption efficiency for methylene blue (MB) and Congo red (CR). Kinetics and isotherms studies indicated that the adsorption behavior conformed to the pseudo-second-order kinetic model and Langmuir isotherm model. By the Langmuir isotherm fitting, the maximum adsorption capacities of MB and CR by LP/AA/DC/PVA-2 were recorded to be 222.65 mg/g and 316.46 mg/g, respectively. The adsorption mechanism is dominated by the hydrogen bonding and electrostatic interactions. Further, the adsorption and desorption experiments demonstrated that LP/AA/DC/PVA-2 hydrogel have excellent reusability.

Fabrication of phenylalanine amidated pectin using ultra-low temperature enzymatic method and its hydrogel properties in drug sustained release application

Chenmin Zheng,Wanping Huang,Yuping Zou,Wensi Huang,Peng Fei,Guoguang Zhang

In this study, pectin was modified with phenylalanine by ultra-low temperature enzymatic method to improve its gel properties. The grafting ratio of phenylalanine amidated pectin was studied under different reaction conditions. The highest value (29.21 %) was reached a reaction temperature of −5 °C and time of 12 h. Further analysis indicated that phenylalanine and high methoxyl pectin combined at the solid-liquid two phase interface under the catalysis of papain to form phenylalanine amidated pectin. Moreover, the physicochemical properties of pectin hydrogel and its feasibility as a sustained-release drug carrier were discussed. The results showed that phenylalanine amidated pectin can form hydrogel with a certain strength under acidic conditions, and there is no need to add a lot of soluble solids and divalent cations. Besides, the phenylalanine amidated pectin hydrogel as a sustained release carrier of drugs showed more sustained and complete drug release.

Green fabrication of citrus pectin-Ag@AgCl/g-C3N4 nanocomposites with enhanced photocatalytic activity for the degradation of new coccine

Dezhuang Gong,Xiquan Li,Xiuling Zhang,Wentao Zhang,Tianhao Chen,Xinhua Zhang

In this research, novel citrus-pectin-based [email protected] /graphite carbon nitride nanocomposites (CACNs) were formed by assembling citrus-pectin-based [email protected] ( [email protected] ) prepared by daylight-driven reduction onto a graphite carbon nitride (g-C 3 N 4 ) surface. The as-prepared samples were characterized by various techniques to study their morphology, structure, optical and electrochemical properties. The CACNs were found to have enhanced photoelectrochemical properties, which provided excellent photocatalytic activity to degrade the food colorant new coccine (NC) under visible light. For the NC degradation, the recommended photocatalyst offered approximately 95.7% of the degradation efficiency under visible-light irradiation for 60 min, which has superior photostability and reusability. Its degradation rate constant was approximately 5.16 and 37.6 times those of pure g-C 3 N 4 and [email protected] , respectively. The NC degradation conformed to pseudo-first-order kinetics. Thus, the developed nanocomposites provide a useful reference for the removal of hazardous food colorants and resource utilization of peel waste.

A consecutive extraction of pectin and hesperidin from Citrus aurantium L.:Process optimization, extract mechanism, characterization and bio-activity analysis

Peng Zhou,Min Zheng,Xiangzhou Li,Jun Zhou,Yaxuan Shang,ZhaoShuang Li,Limin Qu

With the proposal of the Paris Climate Agreement and double carbon policy, production enterprise is now facing green and low-carbon transformation. To promote green and low-carbon development, optimizing the production process and improving utilization rate of raw materials for the reduction of production cost is an significant technological path for the plant extract industry. Thus, the aim of this work was to develop a consecutive extraction process of pectin and hesperidin with lower cost and higher purity, as well as better properties of extract. The process parameters will be put forward through single-factor experiments, Plackett-Burman design and Box-Behnken design. The experimental results showed that the consecutive extraction process was effective and feasible. Pectin with excellent properties was obtained, and the solid-liquid ratio of extraction process parameters of hesperidin was reduced to 1:8 (g: mL). The problems of environmental pollution and high energy consumption in traditional extraction could be solved. The consecutive extraction process, an innovative, economic and environmental-friendly process, may be realized easily in industrial application. At the same time, the consecutive extraction process mechanism was revealed with the help of microscopy analysis of the plant tissue structure. The results demonstrated that preferential extraction of pectin process was helpful for the exposure and dissolution of hesperidin crystallization in cells and thus can improve extraction efficiency of hesperidin. The purity of the obtained hesperidin was up to 91.7%, which can be further purified depending on application requirements. A large amount of pectin was obtained (23.3% ± 0.5% yield), and the pectin has superior gel-property, strong antioxidant activity and certain antibacterial activity. The pectin and hesperidin obtained will have better application prospects in medicine and food because of their unique characteristics.
