Atherosclerosis (AS) is a chronic cardiovascular disease characterized by the accumulation of excess lipids within arterial walls. Since oxidative stress is one of the key drivers in its pathogenesis, antioxidative therapy serves as a promising strategy for AS treatment. Despite the potent antioxidant properties of polyphenolic salvianic acid A (SAA) and salvianolic acid B (SAB), clinical application is limited by their short half-life, insufficient target selectivity, and poor chemical stability. To overcome these limitations, we developed a biomimetic platelet membrane-coated polyphenol–cerium dioxide nanozyme complex (SS-CeO2@PLTM) as a broad-spectrum antioxidative system for comprehensive AS treatment. The SS-CeO2@PLTM was constructed by conjugating SAA and SAB on the surface of cerium dioxide via a one-pot method, followed by surface coating with a biomimetic platelet membrane. Systematic characterization using UV–vis spectroscopy, Fourier transform infrared (FT-IR), X-ray photoelectron spectroscopy (XPS), transmission electron microscopy-energy dispersive X-ray spectroscopy (TEM-EDS) elemental mapping analysis, Forster resonance energy transfer (FRET), molecular simulations, and pH-responsive release assays validated the successful synthesis and pH-responsive drug release behavior of SS-CeO2@PLTM. It was also found that SS-CeO2@PLTM exhibited broad-spectrum antioxidant activities, effectively scavenging superoxide anions, DPPH, H2O2, and intracellular reactive oxygen species (ROS) while enhancing hepatic SOD and GSH activities. Further cellular uptake and in vivo targeting assays confirmed their specific targeting for atherosclerotic plaques. SS-CeO2@PLTM demonstrated great efficiency in attenuating oxidative stress, inhibiting lipid uptake and lipid accumulation, mediating cholesterol efflux, and inhibiting inflammatory factor (IL-1β, IL-6, and TNF-α) secretion in RAW264.7 cells. We also found that SS-CeO2@PLTM exhibited improved lipid metabolism and diminished plaque area in apoE–/– mice via attenuating oxidative stress and NF-κB-mediated inflammation. Furthermore, biosafety was verified via hemolysis and in vivo safety tests. This study is the first to reveal the superior efficacy of the PLTM-based nanozyme complex in targeted AS therapy, offering a paradigm shift for multifactorial cardiovascular disease management.

Atherosclerosis (AS) is a prevalent chronic inflammatory disease characterized by excessive accumulation of reactive oxygen species (ROS) and persistent inflammation. Polyphenolic natural antioxidants possess strong ROS-scavenging properties. However, the poor targeting ability and rapid metabolism greatly limit their further clinical applications. To this end, a multifunctional biomimetic nanoplatform (CCR2@NPs) is developed by engineering macrophage membranes with the overexpressed C-C chemokine receptor 2 (CCR2) and subsequently enhances the target delivery to inflammatory lesions of AS via the C-C motif ligand 2 (CCL2)/CCR2 chemotactic signaling pathway for significantly improving drug bioavailability. In pathological local lesions, CCR2@NPs can effectively scavenge the excessive ROS, alleviate vascular injury, and finally inhibit AS progression. This strategy enables precise, synergistic therapy and offers new insights into the potential treatment approaches for AS.

Oxidative stress is the key contributor to the onset of numerous diseases. Herein, we develop tea polyphenol–zinc (Tp-Zn) using a metal–polyphenol coordination strategy through a simple hybrid approach. The product is characterized by methods such as dynamic light scattering (DLS), ultraviolet–visible spectroscopy (UV–vis) and transmission electron microscopy (TEM) to evaluate the particle size and potential of Tp-Zn. Oxidative stress was induced in mice by administering diquat (25 mg/kg body weight) followed by pre-treatment with 210 mg/kg body weight tea polyphenols (TPs), 280 mg/kg body weight Tp-Zn, and 70 mg/kg body weight ZnSO4for 7 days. Results showed that Tp-Zn treatment significantly improved intestinal barrier function by preventing the diquat-induced down-regulation of tight junction proteins Zonula Occludens protein 1 (ZO-1) and occludin. It also mitigated liver inflammation and damage, as evidenced by reduced serum levels of Aspartate aminotransferase (AST), Alanine aminotransferase (ALT), and Alkaline phosphatase (AKP). Furthermore, Tp-Zn enhanced the antioxidant response in both the intestine and liver by up-regulating the mRNA expression of antioxidant enzymes and reducing the levels of Malondialdehyde (MDA) and reactive oxygen species (ROS) compared to the diquat group (DIQ group). Also, the detection of ROS in the small intestine confirmed Tp-Zn markedly increased intestinal Nuclear factor erythroid 2-related factor 2 (Nrf2) expression compared to the control group. This study aims to clarify that metal–polyphenol coordination with multifaceted regulation of the inflammatory microenvironment could be a novel approach for preventing or treating oxidative stress-related diseases.

The mechanistic effects ofZanthoxylum bungeanumon the textural properties of Tibetan pig sausages remain inadequately elucidated. We conducted a dose–response analysis usingZ. bungeanumaqueous extraction (ZBAE) containing 36.97 mg GAE/g polyphenols, applied at concentrations from 0.125% to 1.20% in the meat paste. Optimal textural enhancement was achieved at 0.25% ZBAE, as proved by significantly improving water holding capacity (1.77% increase), hardness (139.87% increase), and gel strength (46.04% increase) relative to the control group (p< 0.05). Specifically, this concentration: (i) promoted protein molecular rearrangement of by enhancing hydrophobic interactions (33.33% increase) and hydrogen bonding (287.99% increase); (ii) induced conformational transitions fromα-helix (42.62% decrease) toβ-sheet formations (21.11% increase); and (iii) generated a homogeneous three-dimensional protein network characterized by a fractal dimension of 2.769 ± 0.006 and a porosity of 38.350 ± 0.333%. The addition of natural polyphenols from Z.bungeanummay optimize the textural quality of processed meat products.

Pyroptosis triggered by pore-forming Gasdermin proteins in cancer cells facilitates anti-tumor immune activation by releasing pro-inflammatory cytokines and immunogenic contents following cellular rupture. However, selectively triggering pyroptosis in tumors still remain limited in clinical applications. Here, it is reported a microfluidic-assisted bacterial delivery system using attenuated Salmonella typhimurium VNP20009 encapsulated with metal-phenolic networks composed of ferric ions (Fe 3 + ) and tannic acid (TA) to enhance intracellular gasdermin D (GSDMD) expression through targeted CRISPR/dCas9 delivery, thereby inducing robust tumor pyroptosis. Mechanistically, this system achieves cascade amplification of pyroptotic cell death through coordinated multi-modal mechanisms. Following systemic administration, VNP20009 specifically accumulates in hypoxic tumor regions while the coated Fe 3 + –TA nanofilm undergoes pH-responsive dissolution in the acidic tumor microenvironment (TME), simultaneously generating ROS through Fenton reaction and releasing CRISPR/dCas9 system to upregulate GSDMD expression. Concurrently, the abundant flagella of VNP activate caspase-1, which in turn cleaves the overexpressed GSDMD proteins into its active form, thereby triggering robust pyroptosis in tumor cells. Taken together, by coupling bacterial adjuvanticity with ROS-mediated stress and CRISPR-driven GSDMD upregulation, this strategy achieves efficient amplification of pyroptosis and promotes antitumor immune activation.

Glycation promotes the generation of advanced glycation end products (AGEs), which in turn exacerbate inflammation, oxidative stress, and chronic metabolic disorders. Given the potential side effects associated with synthetic antiglycation agents, a natural polyphenol-based antiglycation agent was explored from the Tamarindus indica L. shell. In this study, Tamarindus indica L. shell polyphenols (TSP) were obtained via ultrasound-assisted extraction combined with AB-8 macroporous resin purification, which yielded the highest levels of phenolics and flavonoids. UPLC-Q-Orbitrap-MS² analysis identified 23 phenolic compounds, with flavonoids constituting the predominant class, including (+)-catechin, luteolin, and apigenin. TSP exhibited strong antiglycation activity, with dose-dependent inhibition of Amadori intermediates and AGEs. Mechanistic investigations revealed that TSP effectively trapped reactive carbonyl species (RCS), reduced protein oxidation and cross-linking, and maintained protein secondary structure. Spectroscopic analyses and molecular docking further confirmed stable noncovalent interactions between TSP constituents and key glycation-prone residues (arginine and lysine) in bovine serum albumin. Among the identified monomers, (+)-catechin, luteolin, apigenin, diosmetin, 4-hydroxybenzoic acid, procyanidin A2, and eriodictyol-7-O-glucoside exhibited the greatest antiglycation effects. Overall, this study demonstrated that TSP effectively inhibited glycation by trapping RCS, suppressing protein oxidation and structural alteration, and establishing specific interactions with key amino acid residues in proteins. The innovation of this study lies in systematically developing TSP as a novel natural antiglycation agent, demonstrating its multi-mechanistic efficacy and identifying key active constituents, thereby offering a sustainable strategy to valorize agricultural by-products.

The oral delivery of bioactive compounds for obesity management is severely hampered by their low bioavailability and lack of targeted release. To address this, we engineered a V-type single-helix starch (HAMS) via guest-directed molecular assembly as an advanced oral delivery platform. This system features well-defined nanocavities (~1.2 nm) that achieved 90.7% encapsulation efficiency for hydrophobic polyphenols (e.g., quercetin) and provided robust protection against gastrointestinal degradation. The crystalline architecture enabled enzyme-responsive dissociation, yielding a sustained polyphenol release profile and a 4.3-fold enhancement in systemic bioavailability compared to conventional carriers. In a diet-induced obesity mouse model, the HAMS-polyphenol co-assembly (HAMS-QP) elicited multifaceted therapeutic effects: it significantly mitigated body weight gain, dyslipidemia, glucose intolerance, and hepatic steatosis. Crucially, HAMS-QP restored gut barrier integrity, as evidenced by enhanced expression of tight junction proteins (ZO-1, occludin) and mucin-2, and alleviated chronic inflammation. Mechanistic studies revealed that these benefits were mediated through the suppression of PI3K-AKT pathway overactivation. Furthermore, HAMS-QP functioned as a potent prebiotic, selectively enriching beneficial gut microbiota (e.g., Lactobacillus , Akkermansia ) and boosting the production of protective short-chain fatty acids, particularly butyrate. Collectively, our work presents HAMS as a versatile and efficacious biomaterial that integrates precise encapsulation, controlled release, and host-microbe crosstalk into a single therapeutic strategy for metabolic disorders via the gut-metabolism axis.

This paper is focused on the effects of radio frequency (RF) heating on the relative activity of polyphenol oxidase (PPO), weight loss, texture, color, and microstructure of potatoes. The results showed that pure mushroom PPO was almost completely inactivated at 80 °C by RF heating. The relative activity of potato PPO reduced to less than 10% with increasing temperature (25–85 °C). Enzyme extract showed the lowest PPO relative activity at 85 °C after RF treatment, followed by the potato cuboids and mashed potato, about 0.19 ± 0.017%, 3.24 ± 0.19%, and 3.54 ± 0.04%, respectively. Circular dichroism analysis indicated that RF heating changed the secondary structure of PPO, as α-helix content decreased. Both electrode gap and temperature had significant effect ( P < .05) on weight loss, color, and texture of the potato cuboids. Microstructure analysis showed the changes of potato cell and starch during RF heating.

Rapid and accurate measurement of polyphenol oxidase (PPO) activity is important in the food industry as PPOs play a vital role in catalyzing enzymatic reactions. The aim of this study was to develop surface-enhanced Raman scattering (SERS) approach for accurate determination of PPO activity in fruit and vegetables using the reduction in SERS intensity of catechol in reaction medium. Within a certain catechol concentration, when a purified PPO solution was analyzed, the reduction in SERS intensity (ΔI) was linear to PPO activity (Ec) in a wide range of 500–50 000 U/L, and a linear regression equation of log ΔI/Δt = 0.6223 log Ec + 0.8072, with a correlation coefficient of 0.9689 and a limit of detection of 224.65 U/L, was obtained. The method was used for detecting PPO activity in apple and potato samples, and the results were compared with those obtained from colorimetric assay, which demonstrated that the proposed method could be successfully used for detecting PPO activity in food samples.

This study investigates the effects of high-pressure freezing (HPF) at 0.1–400 MPa and -20oC for 30 min on polyphenol oxidase (PPO) in both solid crystal (S-PPO) and liquid forms at high and low concentrations (HL/LL-PPO). The effects were ranked according to enzyme activity and structural changes in the following order: physical state > concentration > pressure > crystallization path. Notably, LL-PPO exhibited greater stability than both HL-PPO and S-PPO, under HPF conditions, low temperatures provide a protective effect against the pressure-induced denaturation of S-PPO. Additionally, the sulfhydryl content and the high concentration state are crucial for maintaining the structural stability of L-PPO. By altering the physical state of PPO (Liquid and Solid), the enzyme's activity can be effectively regulated through activation and inhibition. Furthermore, the back propagation artificial neural network model demonstrates a precise fit for the correlation between spatial structure and enzyme activity.

Multi-layer graphene oxide (GO) membranes show great perspective in vast fields due to special two-dimensional structure. However, pure GO membranes generally face main obstacles of low flux, weak interlayer interaction and poor stability, which hinder their wide application in separation. Introduction of nano-porous crystals into interlayer of GO nanosheets to form 3D/2D structure is a potential strategy to overcome these issues. In this study, novel polyphenol etched hollow MOF/GO composite membrane was first synthesized employing modified hollow metal organic frameworks with strong hydrophilicity and hollow structure by one-step etching-aided cross-linking method. MOF nanoparticles with hollow structure expanded interlayer spacing of GO nanosheets and brought about formation of additional water channels, significantly reducing water transfer resistance. Meanwhile, nano-porous crystal MOFs modified by phenolic materials were able to cross-link adjacent GO nanosheets via π–π interfacial interactions to achieve stable structure and increase anti-swelling property of membranes. The pores with abundant hydroxyl groups provided strong free energy barrier for ions at medium and low concentrations of ionic strength , therefore not only greatly improved membrane permeaselectivity, but also enhanced anti-fouling performance. The prepared eZIF-8/GO nanofiltration membrane exhibited excellent permeate flux of 16.4 L m −2 h −1 bar −1 , 382% higher than pure GO membrane, and competitive Na 2 SO 4 rejection of 86.3%. By simply adjusting GO loading to further reduce membrane thickness, the permeate flux reached up to 45.4 L m −2 h −1 bar −1 with high rejection more than 99% for organic molecules. These results indicate that novel MOF/GO composite membranes have broad application prospects in desalination and water treatment.

Dietary fiber (DF) is a crucial part of a healthy diet. However, the impacts of bound polyphenols (BPs) on the diversity of gut microbiota and the production of metabolites—such as short-chain fatty acids (SCFAs)—as regulated by DF during in vitro digestion and fermentation remain largely unclear. Here, we evaluated the effects of BP removal on the in vitro digestion and fermentation of DF derived from Rosa roxburghii Tratt. (RRT)—a medicinal and edible fruit. Using DF rich in BPs (RRT-DF) and dephenolic DF (RRT-dP-DF), changes in polyphenol content (PC) and antioxidant activity during digestion and fermentation were evaluated alongside fermentation parameters (pH and SCFAs), gut microbiota structure, and metabolome profiles. Polyphenol release from RRT-DF was limited during gastrointestinal digestion (4–6%) but increased significantly during fermentation (25–27%). The fermentation broth of RRT-DF exhibited a significantly higher antioxidant capacity than that of RRT-dP-DF. RRT-DF also displayed favorable prebiotic properties; BPs enhanced DF-mediated SCFA production, modulated gut bacterial composition, and promoted the metabolism of lipids/lipid molecules and organic acids and their derivatives. Overall, these findings advance understanding of the potential role of BPs in regulating the in vitro digestion and fermentation of RRT-DF.
