A novel AIE-active dye for fluorescent nanoparticles by one-pot combination of Hantzsch reaction and RAFT polymerization: synthesis, molecular structure and application in cell imaging

Yali Chen,Zengfang Huang,Xiaobo Liu,Liucheng Mao,Jinying Yuan,Xiaoyong Zhang,Lei Tao,Yen Wei

In recent years, amphiphilic AIE-active fluorescent organic materials with aggregation-induced emission (AIE) properties have been extensively investigated due to their excellent properties. This study describes the synthesis of a novel AIE-active dye of tetraphenylethylene diphenylaldehyde (TPDA). As compared with the reported fluorescent dye TPB, the fluorescence intensity of TPDA is significantly enhanced with the distinct red shift of emission wavelength. Subsequently, the corresponding novel polymers PEG-TPD were obtained through the one-pot combination of Hantzsch reaction and RAFT polymerization. The structure of PEG-TPD1 by the two-step process was similar with that of PEG-TPD2 by the one-pot method at the same feeding ratio of TPDA and PEGMA. The molecular weights (Mn) of the polymers PEG-TPD1 and PEG-TPD2 were respectively 52 000 and 28 000 with narrow polydispersity index (PDI), and their molar fractions of TPDA were respectively about 9.5% and 14.3%, indicating that the degree of Hantzsch reaction in the one-pot process was more complete. Subsequently, the effect of feed ratio of TPDA and PEGMA on polymer structure was further studied. It can be seen that the Mn of the polymers gradually increases as the proportion of TPDA increases. In aqueous solution, these amphiphilic PEG-TPD polymers tended to self-assemble into corresponding fluorescent polymer nanoparticles (FPNs). The diameter of PEG-TPD2 FPNs ranged from 200 to 300 nm, and their fluorescence emission spectra have maximum emission peak at 509 nm. The PEG-TPD FPNs have significant advantages such as good fluorescence intensity, high water dispersibility, good biocompatibility and easy absorption by cells, which can be attractively used in the field of bioimaging.

Multi-responsive fluorescent probe based on AIE for the determination of Fe3+, total inorganic iron, and CN- in aqueous medium and its application in logic gates

Xiaoye Wen,Li Yan,Zhefeng Fan

A novel and simple probe (denoted LN-1) with AIE property based on malononitrile and 4-diethylaminobenzaldehyde was designed and synthesized. The use of two different mechanisms for the independent detection of Fe 3+ and CN − is more convenient and efficient than the continuous detection of anions and cations. The detection mechanism was verified by mass spectrometry (MS), nuclear magnetic resonance (NMR) spectrum, and particle size distribution. Under optimized experimental conditions, excellent calibration curves were obtained for the LN-1-Fe 3+ and the LN-1-CN − with detection limits of 0.17 and 0.39 μM, respectively. This method was successfully applied to detect the content of Fe 3+ and total inorganic iron in actual samples. Owing to its low cytotoxicity, good biocompatibility, and enhanced orange emission in aqueous solution, the LN-1 was successfully applied for CN − imaging in live cells. In addition, Fe 3+ and EDTA incorporation into the LN-1 changed the fluorescent intensity, which is beneficial in constructing ultrasensitive INHIBIT logic gate.

A high-throughput fluorescence immunoassay based on conformational locking strategy of MOFs to enhance AIE effect of CuNCs-CS for bisphenol S analysis in food samples

Nuanfei Zhu,Kuijing Yuan,Dinghui Xiong,Fengxiang Ai,Kun Zeng,Biying Zhao,Zhen Zhang,Hongjun Zhao

Dietary intake is an important exposure pathway of environmental bisphenols including bisphenol S (BPS, a typical emerging organic pollutant). However, available data on the occurrence and risk assessment of BPS in food samples are limited due to the lack of high-throughput analytical methods. Herein, based on H 2 O 2 inducing fluorescence turn-off signals, a sensitive and high-throughput fluorescence enzyme-linked immunosorbent assay (FELISA) was developed for BPS investigation in beverages. In this system, the fluorescence of copper nanoclusters in ZIF-8 (CuNCs-CS@ZIF-8) could be quenched with the presence of H 2 O 2 owing to the strong oxidation, being served as signal amplification in FELISA. Meanwhile, the aggregation-induced emission (AIE) fluorescence enhancement of CuNCs-CS through the conformational locking of ZIF-8 provided an effective strategy to overcome the intrinsic limitations of FELISA including high background, serious fluorescence quenching in the detection of real samples. Thus, the integration of AIE building blocks into porous ZIF-8 showed obvious advantages in enhancing the analytical performance of FELISA. Under optimized conditions, satisfactory results with good accuracy were achieved in this proposed FELISA, which was applied for the systematic investigation of BPS in foods and further health risk assessments. The results after 100 beverages analysis displayed an average detection frequency of 55%, and the detected concentration of BPS ranged from below the limit of quantification (LOQ) to 12.08 ng/mL with a mean value of 1.717 ng/mL. Besides, the estimated daily intake (EDI) of BPS was calculated to be 0.05449–4.483 ng/kg-bw/day, in which milk–tea drinks contributed to the majority of BPS daily intake (mean EDI = 4.483 ng/kg-bw/day). And the hazard quotient (HQ) according to EDI values indicated that BPS exposure in beverages might not make a considerable impact on human health.

Sequential determination of cerium (IV) ion and ascorbic acid via a novel organic framework: A subtle interplay between intramolecular charge transfer (ICT) and aggregated-induced-emission (AIE)

Xiangqian Li,Zhijun Lai,Jiapei Gu,Wanqiang Liu,Jinwei Gao,Qianming Wang

Construction of systems with variable electron distribution is a relatively new method in design of responsive fluorophores. Herein, 4,4′,4″,4‴-(ethene-1,1,2,2-trayltetrakis (benzene-4,1-diyl))tetrakis(1-carboxymethyl-pyridin-1-ium)bromide ( LG556 ) is developed and its specific spectral behavior has been verified based on perturbation involving intramolecular charge transfer (ICT) via the connected electron donor and acceptor. In the presence of Ce 4+ ions, the fluorescence intensity of LG556 can be efficiently quenched due to the disassembly of LG556 nanoaggregates and inner filter effect (IFE). The sensitivity is dependent on the concentration of Ce 4+ with an excellent linear range 1–150 μM and a low detection limit of 0.62 μM. At this stage, the working principle behind the aggregated-induced-emission (AIE) has been employed and the difficulty lying in aggregate formation is overcome. The incorporated ascorbic acid causes the reduction of Ce 4+ to Ce 3+ and the re-assembly of LG556 has been realized. The detection limit has been evaluated to be 39.2 nM, which is lower than most of the reported results. Therefore, the “on-off-on” signal conversion leads to a reversible IMPLICATION logic gate and the molecular probe has been applied for the analysis of Ce 4+ and ascorbic acid in pure aqueous solution with satisfactory results.

AIE-active polysulfates via a sulfur(VI) fluoride exchange (SuFEx) click reaction and investigation of their two-photon fluorescence and cyanide detection in water and in living cells

Haibo Wan,Shiyuan Zhou,Peiyang Gu,Feng Zhou,Da Lyu,Qinghua Xu,Anna Wang,Haibin Shi,Qingfeng Xu,Jianmei Lu

Three polysulfates P1, P2 and P3 containing two representative AIE-active groups, tetraphenyl ethylene and naphthylamide, were successfully synthesized based on a sulfur(VI) fluoride exchange (SuFEx) click reaction. Their solid fluorescence was studied in detail and liquid fluorescence emission could be tuned by choosing solvents with different polarities. P1 with a branched structure showed higher fluorescence emission than P2 and P3 both in the solid state and in organic solutions because of the more crowded local environment of the tetraphenyl ethylene group in the main chain. Moreover, all the polymers exhibited typical aggregation induced emission (AIE) effects in the solid state and in DMF/water solutions. Furthermore, their two-photon fluorescence behaviours were similar to their one-photon fluorescence behaviours in different organic solvents and in 90% DMF/water solutions. Interestingly, they could selectively detect cyanide ions with lower detection limits in water on account of the anion–π interactions between the naphthylamide groups and the cyanide ions. The detection limits of P1 (0.137 μM) and P2 (0.131 μM) are lower than P3 (0.145 μM), which may be attributed to the more exposed active sites of the P1 and P2 nanoparticles (NPs). All the results indicate that these polymers have higher sensitivity and better applicability for cyanide ion detection in water and in living cells.

Three-in-One: Molecular Engineering of D–A–π–A Featured Type I and Type II Near-Infrared AIE Photosensitizers for Efficient Photodynamic Cancer Therapy and Bacteria Killing

Yun-Yun Quan,Tingting Pan,Zhongda Zhang,Shihua Wang,Guiyun Wang,Lichao Yu,Ye Wang,Xu-Feng Zang,Fangjun Zhang,Xiaoxia Ye,Xuebo Pan,Zu-Sheng Huang

Bacterial infections are closely correlated with the genesis and progression of cancer, and the elimination of cancer-related bacteria may improve the efficacy of cancer treatment. However, the combinatorial therapy that utilizes two or more chemodrugs will increase potential adverse effects. Image-guided photodynamic therapy is a highly precise and potential therapy to treat tumor and microbial infections. Herein, four donor–acceptor–π-bridge–acceptor (D–A–π–A) featured near-infrared (NIR) aggregation-induced emission luminogens (AIEgens) ( TQTPy , TPQTPy , TQTC , and TPQTC ) with type I and type II reaction oxygen species (ROS) generation capabilities are synthesized. Notably, TQTPy shows mitochondria targeted capacity, the best ROS production efficiency, long-term tumor retention capacity, and more importantly, the three-in-one fluorescence imaging guided therapy against both tumor and microbial infections. Both in vitro and in vivo results validate that TQTPy performs well in practical biomedical application in terms of NIR-fluorescence imaging-guided photodynamic cancer diagnosis and treatment. Moreover, the amphiphilic and positively charged TQTPy is able to specific and ultrafast discrimination and elimination of Gram-positive (G+) Staphylococcus aureus from Gram-negative (G−) Escherichia coli and normal cells. This investigation provides an instructive way for the construction of three-in-one treatment for image-guided photodynamic cancer therapy and bacteria elimination.

Aggregation-Induced Emission Supramolecular Organic Framework (AIE SOF) Gels Constructed from Supramolecular Polymer Networks Based on Tripodal Pillar[5]arene for Fluorescence Detection and Efficient Removal of Various Analytes

Juan Liu,Yan-Qing Fan,Shan-Shan Song,Guan-Fei Gong,Jiao Wang,Xiao-Wen Guan,Hong Yao,You-Ming Zhang,Tai-Bao Wei,Qi Lin

Herein, an aggregation-induced emission (AIE) supramolecular organic framework gel (SOF-TPN-G) was successfully constructed from novel supramolecular polymer networks based on tripodal pillar[5]arene. The SOF-TPN-G shows fluorescent response for multiple metal ions. Interestingly, by introducing Fe3+, Cu2+, Cr3+, Ag+, Tb3+, and Eu3+ into the SOF-TPN-G, respectively, a series of metallogels (SOF-Ms) with multicolor fluorescence was obtained. Moreover, based on these gels, an eight-unit sensor array was successfully developed, which exhibits a highly sensitive fluorescent response for Fe3+, Cu2+, Cr3+, Ag+, Tb3+, Eu3+, F–, CN–, HSO4–, His, Ser, and Cys. Among them, the detection for Fe3+, Cr3+, Tb3+, Eu3+, CN–, F– and Ser reached an ultrasensitive level: the lowest limits of detection (LODs) for these ions and molecules are 7.90 × 10–9, 9.20 × 10–9, 6.60 × 10–9, 6.80 × 10–10, 3.83 × 10–9, 3.26 × 10–9, and 3.37 × 10–9 M, respectively. More importantly, the xerogel of SOF-TPN-G exhibits nice adsorption capacity for multimetal ions (Fe3+, Ag+, Cr3+, Tb3+, Cu2+, and Eu3+), the absorption percentages for them were 98.30%, 99.57%, 98.80%, 98.30%, 99.34% and 98.40%, respectively. These AIE organogel/metallogels are novel multiresponsive fluorescent materials for high-efficiency detection and separation of multianalytes.

A phenanthro[9,10-d]imidazole-based AIE active fluorescence probe for sequential detection of Ag+/AgNPs and SCN− in water and saliva samples and its application in living cells

Fanqiang Bu,Bing Zhao,Wei Kan,Liyan Wang,Bo Song,Jianxin Wang,Zhe Zhang,Qigang Deng,Guangming Yin

Ag + and SCN − play extremely important roles in the fields of the physiology and environment. In this work, on the basis of phenanthro[9,10- d ]imidazole derivative ( DIPIP ) which can exhibit the aggregation-induced emission (AIE) properties in aqueous solution, we achieved a sequential on-off-on switch for Ag + and SCN − with high selectivity and sensitivity. A remarkable fluorescence quenching effect of Ag + on the probe DIPIP was observed with 1:2 stoichiometry , Subsequently, the fluorescence intensity of in situ generated DIPIP -Ag + ensemble was easily switched on after the interaction between Ag + and SCN − , which was attributed to the stronger affinity of SCN − to capture Ag + . In particular, the extreme limits of detection (LOD) for Ag + and SCN − in standard solutions were as low as to be 74.5 nM and 7.8 nM, respectively. Furthermore, the probe DIPIP and the DIPIP -Ag + ensemble could be used to detect Ag + in the real water and SCN − in smoker saliva samples, respectively. In addition, the sequential “on-off-on” fluorescence mode of DIPIP to Ag + and SCN − were also successfully applied in living HeLa cells.

An aggregation-induced emission fluorogen/DNA probe carrying an endosome escaping pass for tracking reduced thiol compounds in cells

Hu Yinhua,Cao Xiuping,Guo Yingshu,Zheng Xiaofei,Li Dongjiao,Chen Si-Kai,Chen Guang,You Jinmao

The fluorescent nanoprobes for reduced thiol compounds (represented by glutathione, GSH) are constructed based on the aggregation-induced emission (AIE) luminescence mechanism and endosome escape technology. First, a DNA sequence was designed with the decoration of biotin at the 5′-end, disulfide bound in the internal portion, and amino at the 3′-end. The aptamer of the MCF-7 cell was also one of the most important structures in our DNA sequence for the selectivity of MCF-7 cells. We modified streptavidin-modified magnetic beads (MB) with biotin-modified influenza virus hemagglutinin peptide (HA) and biotin-DNA-amino to form MB/DNA/HA. Carboxyl-modified tetraphenylethylene (TPE), an iconic AIE fluorogen, was bonded with amino-modified DNA by covalent interactions (TPE/DNA). Then, the TPE molecule was attached on the outer layer of MB via biotin-modified TPE/DNA to form MB/DNA/HA/TPE. Compared with traditional AIE/biomolecule conjugates, the nanoprobe had an enhanced endosome escape function, due to the assembly of HA. This construction made the intracellular fluorescence response more accurate. In the presence of reduced thiol compounds (take GSH, for example), the disulfide bond on the DNA was reduced by thiol-disulfide exchange reactions and the TPE molecule was released into the solution. The shedding TPE molecule was more hydrophobic than TPE/DNA and the conversion of TPE/DNA to shedding TPE could lead to the aggregation of the TPE fluorogen. Thus, its fluorescence was enhanced. Under the optimized condition, the fluorescence intensity increased with the increase in concentration of GSH‚ ranging from 1.0 × 10 −9  M to 1.0 × 10 −5  M‚ and the detection limit was 1.0 × 10 −9  M. The relative standard deviation (RSD) was calculated to be 3.6%. The recovery in cell homogenate was from 94.5 to 102.7%. The nanoprobe provided a way for the detection of reduced thiol compounds in MCF-7 cells. We envision that, in the near future, our strategy of DNA-instructed AIE could be widely applied for biosensing and bioimaging in vitro and even in vivo with dramatically enhanced sensitivity.
