In the work, we reported an effective method for the preparation of surface molecularly imprinted polymers based on bacillus as substrate material with superparamagnetic property through miniemulsion polymerization. Then, the obtained magnetic surface molecularly imprinted polymers (MMIPs) were evaluated as adsorbents for selective recognition 2,4,6-trichlorophenol (2,4,6-TCP) from aqueous medium. The resulting MMIPs were characterized by several techniques including FT-IR, SEM, TEM, Raman, XRD, VSM and TGA. The results demonstrated rod-shaped MMIPs was covered with imprinted layer, and exhibited chemical stability and good magnetic sensitivity. Batch mode of binding studies were carried out to determine the equilibrium isotherm, kinetics, selectivity adsorption and regeneration of MMIPs toward 2,4,6-TCP. The results indicated that the selective adsorption behaviors of MMIPs were well described by the Langmuir isotherm model and the pseudo-second-order kinetics model. The specific adsorption capacity of MMIPs was 45.16 mg g −1 at 298 K, which was 2.15 times higher than that of magnetic non-imprinted polymers (MNIPs). The selective recognition studies demonstrated the outstanding affinity and selectivity toward 2,4,6-TCP in the presence of competitive phenols . The regeneration study showed excellent adsorption capacity even after five regeneration cycles. In addition, MMIPs were successfully applied to the extraction of 2,4,6-TCP from milk sample.

The bio-recyclability of polylactic acid (PLA) is hampered by deficient capacity and low yields of the depolymerases. Whether native production in original isolate Laceyella sacchari LP175 or preparation using Escherichia coli system, the poly( l -lactide) (PLLA) depolymerase, Ls PLAase was restricted for industrial applications by complex downstream purification and limited protein yields. Herein, based on secretory expression system in Bacillus subtilis , purification-free Ls PLAase without solubility tags was obtained, following fermentation optimization, directed evolution, combinatorial mutation, and high-density fermentation, a superior variant, Ls PLAase IWFI (A13I/S136W/S138F/W141I) with boosted performance, achieving protein yield up to 3.21 g/L and enzymatic activity of 6.7 U/mL in 5-L bioreactors, with significant improvement relative to the wild-type (0.9 g/L, 2.4 U/mL) and E. coli system (0.01 g/L, 0.18 U/mL). The prominent variant outperforming ProteinK with superior depolymerization performance toward diverse PLA materials, represented 70% increase in conversion efficiency compared to the wild-type. In all, this study providing a scalable solution for industrial PLA waste recycling to enable circularity.

Bacilohydrin A, a new cyclic lipopeptide composed of seven mixed D/L-amino acids (L-Ile1—L-Leu2—D-Asp3—L-Val4—L-Leu5—L-Leu6—L-Gln7) and a (R)-3-hydroxy-12-methyltetradecanoic acid (HMT) residue, was isolated from the crude culture extract of Bacillus sp. SY27F, which was obtained from the Indian Ocean hydrothermal vent. Its planar structure was elucidated by spectroscopic analyses, including 1D and 2D NMR spectroscopy, mass spectrometry (MS), and secondary ion mass spectrometry (MS/MS), whereas its absolute configuration was determined by modified Marfey's analysis. Bacilohydrin A displayed significant cytotoxicities against DU-145, MCF-7 and HepG2 cancer cell lines with IC50 values of 50.3-175.1 nM. Additionally, this is the first time to determine the absolute configuration of a D-aspartic acid residue in surfactin class.

Bacillus spp. are known for their ability to control plant diseases; however, the mechanism of disease control by Bacillus spp. is still unclear. Previously, bacterial organic acids have been implicated in the process of disease suppression. We extracted the total organic acid from Bacillus cereus AR156 culture filtrate and identified oxalic acid (OA) as the programmed cell death–inducing factor. OA strongly suppressed the lesion caused by Botrytis cinerea without significant antagonism against the fungus. Low concentration of OA produced by Bacillus spp. inhibited cell death caused by high concentrations of OA in a concentration- and time-dependent manner. Pretreatment with a low concentration of OA led to higher accumulation of active oxygen-scavenging enzymes in tomato leaves and provoked the expression of defense-related genes. The activation of gene expression relied on the jasmonic acid (JA) signaling pathway but not the salicylic acid (SA) pathway. The disease suppression capacity of OA was confirmed on wild-type tomato and its SA accumulation–deficient line, while the control effect was diminished in JA synthesis–deficient mutant, suggesting that the OA-triggered resistance relied on JA and ethylene (ET) signaling transduction. OA secretion ability was widely distributed among the tested Bacillus strains and the final environmental OA concentration was under strict regulation by a pH-sensitive degradation mechanism. This study provides the first systematic analysis on the role of low-concentration OA secreted and maintained by Bacillus spp. in suppression of gray mold disease and determines the dependence of OA-mediated resistance on the JA/ET signaling pathway.

This study aimed to evaluate the effects of dietary supplementation with Bacillus velezensis R-71003 combined with sodium gluconate on antioxidant capacity, immune response and resistance against Aeromonas hydrophila in common carp . In addition, the biocontrol potential of the secondary metabolites of B. velezensis R-71003 was also evaluated to analyze the possible mechanism of B. velezensis R-71003 against A. hydrophila . The results indicated that the antibacterial crude extract of B. velezensis R-71003 can destroy the cell wall of A. hydrophila . Moreover, the results showed that dietary B. velezensis R-71003 could promote antioxidant capacity, which significantly increased the activities of CAT and SOD and decreased the content of MDA. Additionally, B. velezensis R-71003 supplementation significantly enhanced the immunity of common carp, as measured by the mRNA expression levels of cytokine-related genes ( TNF-α , TGF-β , IL-1β and IL-10 ). In addition, dietary B. velezensis R-71003 exhibited an upregulation of IL-10 and a downregulation of IL-1β, coupled with higher survival rates when challenged with A. hydrophila compared to the positive group. Furthermore, compared to prechallenge, the mRNA expression levels of TLR-4 , MyD88 , IRAK1 , TRAF6 , TRIF and NF-κB in the head kidney of common carp were significantly increased after challenge. The fish fed the B. velezensis R-71003 diet showed lower expression of TLR-4 , MyD88 , IRAK1 , TRAF6 , TRIF and NF-κB after the challenge than those fed the control diet. Thus, this study revealed that B. velezensis R-71003 can improve the resistance of common carp to pathogenic bacteria by destroying bacterial cell walls and improving fish immunity by activating the TLR4 signaling pathway . Importantly, this study indicated that sodium gluconate has a positive effect on B. velezensis R-71003 in enhancing the anti-infection ability of common carp. The results of this study will lay the foundation for the application of B. velezensis R-71003 in combination with sodium gluconate as an alternative to antibiotics in aquaculture.

Finding new crosslinking enzymes and elucidating internal cross-linking mode are beneficial to meeting the requirement of various protein substrates and processing conditions in food industry. Transglutaminase (TG) can lead to the covalent crosslink of ε-(γ-glutamine)-lysine isopeptidic bonds in proteins through acyl-transfer reaction. This work reported the characterization of transglutaminases (TGs) from four Bacillus strains with obvious differences in amino acid sequences and enzymatic properties. Among them, TG from B. clausii (BCLTG) displayed the highest optimal temperature (70 °C) and specific activity (282 U/mg). The molecular dynamics simulation indicated that enlargement of substrate accessible tunnel in BCLTG with the increased temperature might result in the easy approach of substrate molecule to the active center. Moreover, BCLTG could cause the crosslinking of bovine serum albumin and almost all protein constituents in whey protein and soy protein isolate, and the crosslinking path of BCLTG towards three proteins was revealed. This study provides an ideal candidate for proteins crosslinking and theoretical foundation and preliminary information for applying TG in food processing.

Lonicera japonica Thunb. has attracted much attention for its treatment of bacterial and viral infectious diseases, while its active ingredients and potential mechanisms of action have not been fully elucidated. Here, we combined metabolomics, and network pharmacology to explore the molecular mechanism of Bacillus cereus ATCC14579 inhibition by Lonicera japonica Thunb. In vitro inhibition experiments showed that the Lonicera japonica Thunb.'s water extracts, ethanolic extract, luteolin, quercetin, and kaempferol strongly inhibited Bacillus cereus ATCC14579. In contrast, chlorogenic acid and macranthoidin B had no inhibitory effect on Bacillus cereus ATCC14579. Meanwhile, the minimum inhibitory concentrations of luteolin, quercetin, and kaempferol against Bacillus cereus ATCC14579 were 15.625 μg mL−1, 31.25 μg mL−1, and 15.625 μg mL−1. Based on the previous experimental basis, the metabolomic analysis showed the presence of 16 active ingredients in Lonicera japonica Thunb.'s water extracts and ethanol extracts, with differences in the luteolin, quercetin, and kaempferol contents between the water extracts and ethanol extracts. Network pharmacology studies indicated that fabZ, tig, glmU, secA, deoD, nagB, pgi, rpmB, recA, and upp were potential key targets. Active ingredients of Lonicera japonica Thunb. may exert their inhibitory effects by inhibiting ribosome assembly, the peptidoglycan biosynthesis process, and the phospholipid biosynthesis process of Bacillus cereus ATCC14579. An alkaline phosphatase activity assay, peptidoglycan concentration assay, and protein concentration assay showed that luteolin, quercetin, and kaempferol disrupted the Bacillus cereus ATCC14579 cell wall and cell membrane integrity. Transmission electron microscopy results showed significant changes in the morphology and ultrastructure of the cell wall and cell membrane of Bacillus cereus ATCC14579, further confirming the disruption of the cell wall and cell membrane integrity of Bacillus cereus ATCC14579 by luteolin, quercetin, and kaempferol. In conclusion, Lonicera japonica Thunb. can be used as a potential antibacterial agent for Bacillus cereus ATCC14579, which may exert its antibacterial activity by destroying the integrity of the cell wall and membrane.

The potentiality of PGPR and biocontrol bacteria in agriculture is steadily increased as it offers an attractive way to replace the use of chemical fertilizers, pesticides and other supplements. This study aimed to investigate the effects and potential mechanism of inoculation with two Bacillus velezensis strains on the growth and quality improvement of Anoectochilus roxburghii (Wall.) Lindl. (MRH) and Anoectochilus formosanus (Wall.) Lindl. (YYB). The control treatment was non-inoculated (CK) and the coculture treatments were inoculated with Bacillus velezensis D2WM (D2), Bacillus velezensis ZJ-11 (ZJ-11), Bacillus velezensis D2WM and Bacillus velezensis ZJ-11 (D2+ZJ-11). The fresh weight, plant length, amount of active compounds, as well as chlorophyll content were significantly increased under D2, ZJ-11, and D2+ZJ-11 treatments compared with MRH and YYB plants in CK. In particular, D2+ZJ-11 treatment resulted in the greatest growth promotion. In MRH and YYB plants, the fresh weight increased by 82.6 % and 106.6 %, the kinsenoside content increased by 9.33 % and 21.65 % per gram, and the flavonoid content increased by 44.70 % and 21.07 % per gram, respectively. D2WM and ZJ-11 were capable of secreting siderophore, phytase, indole-3-acetic acid (IAA), and zeatin to promote plant nutrient assimilation and growth. Moreover, when D2WM and ZJ-11 were both inoculated with Anoectochilus roxburghii , the rhizosphere soil enzyme activities of urease, phosphatase, and invertase were significantly higher than control, thus helped to provide more nutrients to Anoectochilus . Further, the analysis of microbial community diversity indicated the increase of abundance of beneficial microorganisms, such as nitrogen-fixing bacteria, Basidiomycota and Ascomycota both of which are known to fuel their plant partners with mineral nutrients from the soil. Additionally, the abundance of pathogens belonging to Xanthobacteraceae as well as Cladophialophora and Penicillium were reduced. Hence, coculture with beneficial microorganisms can improve the growth and quality of Anoectochilus plants via promoting nutrient assimilation and regulating rhizosphere microbial community. This technique is of great application value in promoting the efficient cultivation and production of high-quality plant materials.

d -Mannose and l -ribose are two important monosaccharides, which have attracted public attention recently because of their great application potentials in food, cosmetic and pharmaceutical industries. Sugar isomerases catalyze the sugar isomerization and therefore can be used as the biocatalysts for production of the high-value sugars from inexpensive sugars. l -arabinose isomerase catalyzes the conversion of l -arabinose to l -ribulose, while d -lyxose isomerase catalyzes l -ribulose and d -fructose to l -ribose and d -mannose, respectively. In this paper, a putative d -LI from Bacillus velezensis (BvLI) was identified, characterized and used to produce d -mannose and l -ribose from d -fructose and l -arabinose, respectively. The recombinant BvLI exhibited a maximum activity at 55 °C and pH 6.5, in the presence of 0.1 mM Co 2+ . Approximately 110.75 g/L d -mannose was obtained from 500 g/L d -fructose in 6 h by the recombinant BvLI, and approximately 105 g/L l -ribose was obtained from 500 g/L l -arabinose in 8 h by the successive biocatalysis of l -arabinose isomerase from Bacillus licheniformis (BlAI) and BvLI.

More and more antibacterial agents are used together to treat bacterial infections in diverse fields, but the overuse of antibacterial agents may cause the environmental pollution of antibiotic resistance genes (ARGs). In order to reduce the use of antimicrobial agents, the potential joint effects of quorum-sensing inhibitors (QSIs) and traditional antimicrobial agents have been proposed to be effective. In this study, the joint effects of traditional antimicrobial agents, represented by sulfonamides (SAs) and silver antibacterial agents (silver nitrate (AgNO 3 ) and nanosilver (AgNP, 5 nm)), and five potential QSIs, were investigated using B. subtilis. It was found that AgNP showed higher toxicity than AgNO 3 , whereas the joint effects on B. subtilis showed no difference between AgNO 3 and AgNP when they combined with SAs or QSIs, respectively. In general, AgNO 3 and AgNP presented synergetic and additive effects with QSIs, but additive and antagonistic effects with SAs; SAs exhibited synergetic, additive and antagonistic effects with different QSIs whether in binary or ternary mixed system. Moreover, it was found that the use of antimicrobials was reduced and the synergistic combined toxicity of antimicrobial agents on B. subtilis was increased through the addition of the QSIs. This study can offer a valuable reference for the combined medication of the different antimicrobial agents, which will benefit the environmental and human health.

Carbonyl reductase, as biocatalyst, is very important to chiral alcohols production through asymmetric reduction of carbonyl compound. A novel thermal stable carbonyl reductase from Bacillus cereus (BcCR) dependant on NADPH was obtained through a new genome mining strategy proposed in this work. By analyzing its amino acid sequence and structure, the BcCR should be a thermal stability and wide pH tolerance carbonyl reductase. Its gene was cloned by PCR with B. cereus genomic DNA as template. Its heterologous expression system, E. coli BL21 (DE3) plysS/pET28a- bccr , was constructed, and BcCR was successfully expressed. Enzymatic properties show that at 57.5 °C and pH 7.0 it can reach maximum reaction rate. Its K m and V max to ethyl 4-chloroacetoacetate is 1.85 mmol/L and 0.22 µmol/(min·mg protein ), respectively. It can catalyze the asymmetric reduction of the β-carbonyl compound, such as ethyl 4-chloroacetoacetate to ethyl S -4-chloro-3-hydroxybutyrate. This paper proposes a practical method for discovery of new carbonyl reductases, and provides a novel enzyme as biocatalyst for asymmetric reduction of β-carbonyl compound.

A simple, ultrasensitive, and specific electrochemical biosensor was designed to determine the given DNA sequence of Bacillus subtilis by coupling target-induced strand displacement and nicking endonuclease signal amplification. The target DNA (TD, the DNA sequence from the hypervarient region of 16S rDNA of Bacillus subtilis) could be detected by the differential pulse voltammetry (DPV) in a range from 0.1 fM to 20 fM with the detection limit down to 0.08 fM at the 3sblank level. This electrochemical biosensor exhibits high distinction ability to single-base mismatch, double-bases mismatch, and noncomplementary DNA sequence, which may be expected to detect single-base mismatch and single nucleotide polymorphisms (SNPs). Moreover, the applicability of the designed biosensor for detecting the given DNA sequence from Bacillus subtilis was investigated. The result obtained by electrochemical method is approximately consistent with that by a real-time quantitative polymerase chain reaction detecting system (QPCR) with SYBR Green.
