Polysaccharides and fruit extracts are applied in dairy products to enhance their nutritional property, but the effects of such formulations on the functions and biological activities are yet to be explored. Therefore, this study was aimed at evaluating the effect of interactions among milk protein (beta-lactoglobulin; BLG), polysaccharides (pectin, P; chitosan, CH), and anthocyanin (pelargonidin-3- O -glucoside; P3G) in improving the bioavailability and biological activity of P3G. After gastrointestinal digestion (GID), the content of free P3G in different model solutions were as follows: P3G-alone (73.59 μg/mL), P3G-P (66.59 μg/mL), P3G-CH (36.72 μg/mL), P3G-BLG (64.92 μg/mL), P3G-P-BLG (64.92 μg/mL), and P3G-CH-BLG (39.61 μg/mL). Less amount of free P3G in model solutions indicated increased complex formation of P3G with protein and/or polysaccharides during GID. These complexes resulted in protection and progressive release of P3G in the gastrointestinal tract. Chitosan exhibited more protection to P3G compared with P and BLG. In addition, α-glucosidase inhibitory activity and ROS scavenging activities of conjugated-P3G samples were potentially augmented after GID. However, the presence of polysaccharides and protein in the model solutions did not show any negative effect on the biological activity of P3G. Thus, pure P3G can be used as a nutritional ingredient in dairy industries.

The direct conversion of CO 2 with methanol into dimethyl carbonate (DMC) has received considerable attention as the sustainable development process for conventional routes. The CeO 2 -based catalysts owing to suitable acid-base sites were regarded as the promising catalysts. However, the process has been hindered mainly due to the difficulty in simultaneously regulating the appropriate acid-base properties on CeO 2 -based catalysts, as well as thermodynamic limitations of generating by-product water. In this work, by virtue of enhancing interaction between active components and supports, the highly efficient CeO 2 /Beta catalysts (CeO 2 /Beta-E) were prepared by ethanol as impregnation solvents to DMC synthesis. Meanwhile, the synergistic promoting catalytic systems of DMC synthesis from CO 2 with methanol was proposed. On one hand, the content and electronic properties of oxygen vacancies and Ce 3 + species were regulated to achieve the balance between suitable acid and base sites by ethanol as impregnation solvents. Furthermore, the desired intermediates of bidentate carbonates derived from activated CO 2 on oxygen vacancies and consecutive methoxy species originated from methanol on Ce 3+ species could be markedly promoted by reconstructed surface acid-base properties of CeO 2 /Beta catalysts. Notably, methanol could be rapidly converted to intermediate species over CeO 2 /Beta-E catalysts, which was the crucial step for activation. On the other hand, it was revealed the nature of continuous removal of by-product water into the reaction solution phase to enhance thermodynamic driving forces over CeO 2 /Beta-E catalysts. Consequently, CeO 2 /Beta-E catalysts delivered a DMC yield of 38.4 mmol/g with > 99 % selectivity and excellent cycling stability, providing theoretical insights and practical guidance for designing efficient catalysts for DMC synthesis from CO 2 with methanol.

Endometriosis , defined as the abnormal growth of functional endometrium outside the uterus , is characterized by the abnormal phenotype of endometrial cells. This study aimed to investigate the role of latent transforming growth factor beta binding protein 2 (LTBP2), an extracellular matrix protein , in the occurrence and development of endometriosis. Elevated LTBP2 expression levels were observed in endometrial tissues and serum of endometriosis patients and their area under the ROC curve (AUC) values for distinguishing endometriosis were 0.9044 and 0.9534, respectively. Overexpressing-LTBP2 could promote proliferation, migration, and invasion, whereas suppressing apoptosis of endometrial stromal cells (ESCs). Moreover, LTBP2 downregulation induced the opposite effect. The supernatant from ESCs overexpressing LTBP2 promoted the tube formation of human umbilical vein endothelial cells (HUVECs), thus indicating an angiogenic effect. Furthermore, overexpression of LTBP2 facilitated the inflammation and might promote endometriosis progression through the NF- k B signaling pathway . Conclusively, LTBP2 might be a potential target in the diagnosis and treatment of endometriosis.

2′,2′,4,4′-tetrabromo diphenyl ether (BDE-47), one of the most abundant congeners of commercial pentaBDE utilized as flame retardants , has been phased out of production due to its potential neural toxicity and endocrine disrupting activities, and yet still present in the environment. Several alternatives to BDE-47, including tetrabromobisphenol A (TBBPA), tetrabromobisphenol S (TBBPS), tetrachlorobisphenol A (TCBPA) and decabromodiphenyl ether (BDE-209), are presently employed without restrictions and their potential toxic effects on human neural development are still unclear. In this study, we utilized a human neural stem cell (hNSC)-based system to evaluate the potential developmental neurotoxic effects of the above-mentioned five chemicals, at environment and human exposure relevant concentrations. We found that those compounds slightly altered the expression of hNSC identity markers ( SOX2 , SOX3 and NES ), without impairing cell viability or proliferation, in part by either modulating glycogen synthase kinase 3 beta (GSK3β) signaling (TBBPS, TCBPA and BDE-47), and slightly disturbing the NOTCH pathway (TBBPA, TBBPS and TCBPA). Moreover, the five chemicals seemed to alter hNSC differentiation by perturbing triiodothyronine (T3) cellular signaling. Thus, our findings suggest that the five compounds, especially TBBPS, TCBPA, and BDE-47, may affect hNSC self-renewal and differentiation abilities and potentially elicit neural developmental toxicity.

Summary Four types of carbohydrates, including Dendrobium officinale polysaccharide, Dendrobium aphyllum polysaccharide and β-glucans from yeast and barley, were examined, and their structures were found to mainly contain 1,4-linked-β- d -Glc p . Artificially simulated gastrointestinal digestion was conducted to characterise the changes of molecular weight, reducing sugars and released free monosaccharides by high-performance liquid chromatography, kits and the newly developed gas chromatography (GC)-mass spectrometry (MS)/MS analysis, which indicated that high molecular weight and complex spatial structures contributed to delayed monosaccharide release following exposure to digestive solution. The spatial structures of carbohydrates were changed during gastric digestion, but their primary structures were destroyed during intestinal digestion. Additionally, for the developed 7890A/7000 GC-TQ/MS-MS, the new analytical method was successfully used to analyse very low concentrations of monosaccharides in the simulated gastrointestinal digestive system.

Several kinds of novel lanthanide organic–inorganic mesoporous hybrid luminescent materials have been assembled by introducing the ternary lanthanide ions (Eu 3+ , Tb 3+ ) and beta-diketone (TTA, TAA) into the functionalized mesoporous hosts through the coordination bond and covalent bond by using 3-chloropropyltriethoxysilane modified 1,4,7,10-tetraazacyclododecane (Cyclen-Si) as an organic bridge molecule. These multicomponent hybrids are characterized by FTIR, TEM, XRD, thermogravimetric analysis, diffuse reflectance UV–Vis spectra and nitrogen adsorption–desorption isotherms, especially the photoluminescence properties of them. The luminescent spectra of the hybrids reveal that the MCM-41 based materials show strong emission intensities, high luminescent quantum efficiencies, long luminescent lifetimes and exhibit characteristic red and green luminescence originated from lanthanide ions (Eu 3+ or Tb 3+ ), which suggesting the Cyclen-Si bridge molecule is favorable for the assembly of lanthanide functionalized luminescent hybrids.

Solid polymer electrolytes can significantly improve the safety and energy density of sodium-ion batteries compared with the liquid electrolytes. However, the low ionic conductivity and poor mechanical properties inhibit the practical application. In this paper, poly(ethylene oxide) (PEO) solid polymer electrolytes with enhanced ion conductivity are demonstrated by introducing inorganic solid electrolyte (beta-alumina) filler. With the presence of conductive beta-alumina filler, the ion conductivity of the resultant PEO polymer electrolyte is enhanced from 2.5*10 –4 to 3.95*10 –4 S cm −1 . Applied in sodium-ion batteries (SIBs), the cell delivers an initial discharge capacity of 93.1 mAh g −1 and acceptable cycling performance (77.8 mAh g −1 after 100 cycles), which are significantly superior to that of the PEO solid polymer electrolyte without beta-alumina filler modification. The presented results prove that the ion conductivity of PEO polymer electrolyte can be enhanced by adding conductive beta-alumina, promoting its practical application in Na-ion all-solid-state batteries or other electrochemical energy storage systems.

The Pichia pastoris expression system is a favorable platform for production of pharmaceutical proteins. Treatment of strains with N-acetyl-L-cysteine (NAC) has been shown to enhance the yield of recombinant proteins, thereby contributing to a reduction in production costs. However, the specific mechanism of action of NAC remains unclear. Previous research has indicated that glutathione (GSH) and autophagy are involved in the increased production of human serum albumin and porcine follicle-stimulating hormone β (HSA-pFSHβ) by NAC. This study investigated the potential interaction between GSH and autophagy in the production of HSA-pFSHβ. The findings indicated that sulfhydryl-free antioxidants such as melatonin, vitamin C, or vitamin E did not exhibit similar effects to NAC in enhancing HSA-pFSHβ yield. Moreover, NAC was found to enhance HSA-pFSHβ production by modulating GSH metabolism to reduce GSH consumption, increase total GSH levels, as well as glutathione peroxidase (GSH-Px) and glutathione reductase (GR) activities. Additionally, inhibition of autophagy through disruption of autophagy scaffolding proteins Atg1 or Atg11 led to an increase in recombinant HSA-pFSHβ production. Furthermore, NAC significantly decreased the phosphorylation of Slt2, and the absence of the SLT2 gene influenced the effect of NAC on HSA-pFSHβ secretion by modulating mitophagy and GSH metabolism. In conclusion, these results suggest a complex interplay between GSH metabolism and autophagy in the regulation of NAC-induced HSA-pFSHβ secretion.

The swim bladder performs various functions in fish, is consumed by humans and can be processed into high-grade fish glue (traditional high-value tonic). However, little is known on the optimum amount of proline (Pro) required for promoting collagen synthesis and deposition in fish swim bladder and the regulatory mechanism. This study explored the effects and optimum amount of dietary Pro supplementation on collagen synthesis and deposition in Chu's croaker ( Nibea coibor ) swim bladder and its possible molecular mechanism. A total of 450 juvenile fish (8.64 ± 0.14 g) were randomly stocked into 18 cages (25 fish per cage) in triplicate and fed Pro supplemented diets (0, 5, 10, 15, 20 and 25 g kg −1 ) for eight weeks. Another 225 fish (11.62 ± 0.15 g) were distributed randomly into control, Pro and Pro diet plus injected with specific inhibitor of Smad3 (SIS3) in order to inhibit the transforming growth factor beta (TGF-β)/Smads pathway. Pro supplementation up-regulated significantly the expression of genes involved in TGF-β/Smads pathway such as collagen, type I, alpha 1 ( col1a1 ) and 2 ( col1a2 ), TGF-β and Smad2/3 , and increased collagen content in the Chu's croaker swim bladder ( P < 0.05). The optimum dietary Pro supplementation for deposition of collagen in Chu's croaker swim bladder was 19.36 g kg −1 . The collagen content in the swim bladder positively correlated significantly with the mRNA expression of col1a1 , col1a2 , TGF-β and Smad2/3 genes ( P < 0.05). The optimum dietary Pro supplementation for maximum synthesis of collagen in Chu's croaker swim bladder was 13.25, 13.32 and 15.94 g kg −1 based on mRNA expression of Smads2 , Smads3 and col1a2 genes, respectively. SIS3 down-regulated the expression of col1a1 , col1a2 , TGF-β and Smad2/3 genes, subsequently decreased collagen deposition in the swim bladder ( P < 0.05). These results suggest that 13.25 to 19.36 g kg −1 Pro supplementation improve collagen synthesis and deposition in Chu's croaker swim bladder. The TGF-β/Smad signaling pathway regulates collagen synthesis and deposition in Chu's croaker swim bladder. Our results provide an understanding on the molecular mechanism of collagen synthesis and deposition in fish for producing collagen required for human food and fish glue.

Ischemic strokes are associated with a high rate of disability and death globally. Cerebral ischemia/reperfusion (I/R) injury is a type of brain damage associated with oxidative stress after an ischemic stroke. Beta-boswellic acid (β-BA) reportedly exerts antioxidant and neuroprotective effects, but its role in cerebral I/R injury is unclear. The aim of this research was to investigate the neuroprotective effects, as well as the mechanisms of β-BA in cerebral I/R injury. In vivo experiments were conducted using a rat middle cerebral artery occlusion and reperfusion (MCAO/R) model, and in vitro experiments were performed using a rat neuronal oxygen–glucose deprivation and reoxygenation (OGD/R) model. Triphenyltetrazolium chloride staining, neurological function scores, terminal deoxynucleotidyl transferase deoxyuridine triphosphate nick end labeling, hematoxylin and eosin staining, and antioxidant levels in the brain were used to assess the effects of β-BA. Flow cytometry was used to detect reactive oxygen species and apoptotic cells. Western blotting and immunofluorescence staining were used to measure protein levels. The results showed that β-BA markedly improved neurological deficits and decreased infarct volume and necrotic neurons in rats. The in vitro results showed that β-BA protected neurons against OGD/R-induced injury. Additionally, β-BA significantly increased the phosphorylation of protein kinase C epsilon (PRKCE) at S729, the translocation of nuclear factor erythroid 2-like 2 (NFE2L2), and expression of heme oxygenase-1 (HMOX1). This study demonstrates that β-BA exerts neuroprotective effects against cerebral I/R via the activation of the PRKCE/NFE2L2/HMOX1 pathway and is a potential therapeutic candidate for ischemic stroke. The p53 gene has the highest mutation frequency in tumors, and its inactivation can lead to malignant transformation, such as cell cycle arrest and apoptotic inhibition. Persistent high-risk human papillomavirus (HR-HPV) infection is the leading cause of cervical cancer. P53 was inactivated by HPV oncoprotein E6, promoting abnormal cell proliferation and carcinogenesis. To study the treatment of cervical intraepithelial neoplasia (CIN) and cervical cancer by restoring p53 expression and inactivating HPV oncoprotein, and to verify the effectiveness of nano drugs based on nucleic acid delivery in cancer treatment, we developed poly (beta-amino ester)537, to form biocompatible and degradable nanoparticles with plasmids (expressing p53 and targeting E7). In vitro and in vivo experiments show that nanoparticles have low toxicity and high transfection efficiency. Nanoparticles inhibited the growth of xenograft tumors and successfully reversed HPV transgenic mice’s cervical intraepithelial neoplasia. Our work suggests that the restoration of p53 expression and the inactivation of HPV16 E7 are essential for blocking the development of cervical cancer. This study provides new insights into the precise treatment of HPV-related cervical lesions.

Background Alzheimer's disease (AD) is the most common neurodegenerative disorder without effective therapy and lack diagnosis strategy for preclinical AD patients. There is an urgent need for development of both early diagnosis and therapeutic intervention of AD. Results Herein, we developed a nanotheranostics platform consisting of Curcumin (Cur), an anti-inflammatory molecule, and superparamagnetic iron oxide (SPIO) nanoparticles encapsulated by diblock 1,2-dio-leoyl- sn -glycero-3-phosphoethanolamine- n -[poly(ethylene glycol)] (DSPE-PEG) that are modified with CRT and QSH peptides on its surface. Furthermore, we demonstrated that this multifunctional nanomaterial efficiently reduced β-amyloid plaque burden specifically in APP/PS1 transgenic mice, with the process noninvasively detected by magnetic resonance imaging (MRI) and the two-dimensional MRI images were computed into three-dimension (3D) plot. Our data demonstrated highly sensitive in vivo detection of β-amyloid plaques which more closely revealed real deposition of Aβ than previously reported and we quantified the volumes of plaques for the first time based on 3D plot. In addition, memory deficits of the mice were significantly rescued, probably related to inhibition of NLR Family Pyrin Domain Containing 3 (NLRP3) inflammasomes. Conclusions Gathered data demonstrated that this theranostic platform may have both early diagnostic and therapeutic potential in AD. Graphical Abstract
