Ambient mass spectrometry (MS) has revolutionized the way of MS analysis and broadened its application in various fields. This paper describes the use of microfluidic techniques to simplify the setup and improve the functions of ambient MS by integrating the sampling probe, electrospray emitter probe, and online mixer on a single glass microchip. Two types of sampling probes, including a parallel-channel probe and a U-shaped channel probe, were designed for dry-spot and liquid-phase droplet samples, respectively. We demonstrated that the microfabrication techniques not only enhanced the capability of ambient MS methods in analysis of dry-spot samples on various surfaces, but also enabled new applications in the analysis of nanoliter-scale chemical reactions in an array of droplets. The versatility of the microchip-based ambient MS method was demonstrated in multiple different applications including evaluation of residual pesticide on fruit surfaces, sensitive analysis of low-ionizable analytes using postsampling derivatization, and high-throughput screening of Ugi-type multicomponent reactions.

In this work, we developed a novel strategy using fluorescent quantum dots (QDs) combined with molecularly imprinted polymers (MIPs) on three-dimensional (3D) origami paper-based microfluidic devices for specific recognition and sensitive detection of phycocyanin. This method can realize the liquid phase of QDs@MIPs being transferred to the solid-phase paper base and achieve easy portability for the analysis. Under optimal conditions, we successfully demonstrated the proposed paper@QDs@MIPs 3D microfluidic chip for the sensitive and selective detection of phycocyanin protein target in a simple and robust manner. Our results revealed that the method exhibited a dynamic response to phycocyanin in the range of 10–50 mg/L with a limit of detection of 2 mg/L. Importantly, this device could provide quantitative information very conveniently and show great potential to be further extended to the detection of other proteins or biomarkers for environmental and food safety research.

Monolithically integrated emitters have been increasingly applied to microfluidic devices that are coupled to mass spectrometers (MS) as electrospray ionization sources (ESI). A new method was developed to fabricate a duplicable structure which integrated the emitter into a poly(dimethylsiloxane) chip corner. Two photoresist layers containing a raised base which guaranteed the precise integration of the electrospray tip emitter and ensured that the cutting out of the tip exerted no influence even during repeated prototyping were used to ease the operation of the process. Highly stable ESI-MS performance was obtained and the results were compared with those of a commercial fused-silica capillary source. Furthermore, chip-to-chip and run-to-run results indicated both reliability and reproducibility during repeated fabrication. These results reveal that the proposed chip can provide an ideal ion source for MS across many applications, especially with the perspective to be widely used in portable MS during on-site analysis. Keywords:microfluidic chip;multi-layer soft lithography;electrospray ionization

Tetramethylammonium hydroxide (TMAH), an extensively utilized photoresist developer, is frequently present in ammonium-rich wastewater from semiconductor manufacturing, and its substantial ecotoxicity should not be underestimated. This study systematically investigated the effects of TMAH on the anammox granular sludge (AnGS) system and elucidated its inhibitory mechanisms. The results demonstrated that the median inhibitory concentration of TMAH for anammox was 84.85 mg/L. The nitrogen removal performance of the system was significantly decreased after long-term exposure to TMAH (0 - 200 mg/L) for 30 days ( p < 0.05), but it showed adaptability to certain concentrations (≤ 50 mg/L). Concurrently, the stability of the granules decreased dramatically, resulting in the breakdown of AnGS. Further investigations indicated that TMAH exposure increased the secretion of extracellular polymeric substances but weakened their defense function. The increase in reactive oxygen species resulted in damage to the cell membrane. Reduced activity of anammox bacteria, impeded electron transfer, and changes in enzyme activity suggested that TMAH affected the metabolic activity. Microbiological analysis revealed that TMAH caused a decrease in the abundance of anammox bacteria and a weakening of symbiotic interactions within the microbial community. These results provide valuable guidance for the AnGS system application in chip wastewater treatment.

This work designed a DNA conformation-regulated hemin switch for rapid chemiluminescent (CL) detection of a monoclonal antibodies. This switch was performed with an affinity probe and an inhibition probe, which were conveniently prepared by hybridizing hemin-labeled DNA1 with KHL peptide-labeled DNA2 and binding biotin-labeled DNA3 to streptavidin, respectively. In the absence of the target antibody, streptavidin-DNA3 could hybridize with hemin-DNA1/KHL-DNA2 to release KHL-DNA2, which led to the loss of hemin activity due to the affinity hindrance of streptavidin-DNA3. After the KHL peptide was recognized by the target antibody, the strand replacement hybridization could be inhibited by the bound antibody, which retained the high catalytic activity of hemin overhung on the antibody-bound affinity probe for a CL reaction, leading to a “signal-on” process for CL antibody detection. Using a KHL-specific antibody, anti-proprotein convertase subtilisin/kexin type 9 antibody (PCSK9-Ab), as a target model and common L012–1,2,4-triazole–H2O2 CL system, the designed switch showed a detection range of 10 ng mL–1 to 1 μg mL–1 with a detection limit of 4.16 ng mL–1 (56.2 pM) and a short analytical time of 6.5 min. The proposed quick method could simply be used for lab-on-chip CL detection of PCSK9-Ab in situ-secreted from PCSK9-6E3 hybridoma cells, which showed an accuracy of 90.2% compared with the statistical results from general fluorescence imaging, providing a potential technique for screening specific hybridoma cells.

The emergence of “precision medicine” marks a notable shift in cancer treatment, moving from a tumor type–oriented approach to a more targeted, gene-oriented approach. Detecting low-abundance mutant genes in blood is challenging but crucial for personalized treatment plans. Herein, a novel platform combining catalytic hairpin self-assembly (CHA)-mediated self-calibrating surface-enhanced Raman spectroscopy (SERS) with a high-throughput Raman system (CCSPS) was designed. This platform enables ultrasensitive and rapid genotype analysis of gene mutations. The development of CCSPS specifically targets EGFR mutations, which serve as crucial therapeutic targets for precision therapy in lung cancer. This system shows excellent sensitivity and selectivity, capable of detecting multiple EGFR mutations ( Del-19 , L858R , and T790M ) with a detection limit as low as attomolar levels. Additionally, precise genotyping analysis was successfully conducted on 42 clinical samples using the CCSPS, yielding results consistent with those obtained through next-generation sequencing. These results underscore the efficacy of the CCSPS in noninvasively identifying circulating tumor DNA (ctDNA) mutations, facilitating immediate therapeutic decision making at the bedside. In summary, the CCSPS is a fast, accurate, versatile, and compact testing system capable of precisely screening individuals who stand to benefit from targeted therapy, thus promoting personalized and precise healthcare.

A multifunctional microfluidic chip integrated with perfusion cell culture and in situ SERS detection of cell secretion was designed and developed for the detection of IL-6 secretion from LPS-stimulation of A549 cells in this paper. Researching works were focused on A549 cell activity and secretion in the constructed LPS-stimulated A549 cells model. On the designed microchip, a bubble trap chamber was designed to remove the bubbles in the culture medium which could also be simultaneously preheated by a split hot plate. Then, a long-time perfusion culture process of 549 cells could be realized. Under the optimized conditions the A549 cells could be cultured and kept in good activity for more than 36 h. Subsequently, the model of interaction between LPS and A549 cells was established on the designed microchip. When LPS-stimulated A549 cells, the IL-6 which was one of the secretions formed in this process was detected quantitatively by SERS spectral technique. The silver-coated gold nano-stars were prepared and taken as a sensitive enhancing probe for the SERS detection of IL-6 secreted from LPS-stimulated A549 cells. The immunomagnetic beads, IL-6 antigen, and SERS probes were mixed and incubated in the microchip and form a sandwich structure which was captured by the permanent magnet in the detection zone for SERS detection. The reference material of IL-6 was used to establish the calibration curve, and the linear range and detection limit were 1–10000 pg/mL and 0.75 pg/mL, respectively. Then, the IL-6 secretion from LPS-stimulated A549 cells was detected hourly for 7 h by this established method. The process of LPS stimulation of A594 cells did not lead to a sustained increase in the SERS spectral signature of IL-6. Instead, IL6 secretion initially increased sharply, then decreased and eventually stabilized. It could be due to a potential mechanism that the cells self-regulated to mitigate the inflammatory effects in response to sustained stimulation. The proposed multifunctional microfluidic chip, characterized by high sensitivity and the ability to perform continuous hourly detection, exhibited significant application prospects in the study of external stimulation on cells.

Drying preservation of mammalian cells presents a promising alternative to cryopreservation, with intracellular vitrification being the central challenge. Trehalose, a critical desiccation protectant, is essential for achieving this vitrification, but effective intracellular delivery and concentration control remain major hurdles. Current microfluidic methods for generating Giant Unilamellar Vesicles (GUVs, cell models) rely heavily on glycerol. This glycerol dependence fundamentally impedes the study and application of trehalose-mediated vitrification, as glycerol’s low glass transition temperature and difficulty of removal interfere with trehalose’s protective function. This study develops a glycerol-free microfluidic strategy that directly utilizes trehalose as the core component to enable stable generation of monodisperse water-in-octanol-in-water (W/O/W) double emulsions (GUV precursors) with precisely programmable intra- and extra-vesicular trehalose concentrations. Using a PDMS microfluidic chip with a six-way junction, the effects of replacing glycerol with varying trehalose concentrations (0–1.2 M) on double emulsion generation were investigated. Results demonstrate that trehalose effectively compensates for the destabilization caused by glycerol removal by increasing inner aqueous phase viscosity and reducing inner interfacial tension, significantly broadening the stable dripping regime. Droplet size exhibited non-monotonic variation with inner trehalose concentration, while increasing outer aqueous trehalose consistently reduced droplet size. This work is the first to demonstrate that trehalose can fully replace glycerol for generating double emulsions with spatially controlled trehalose loading. This glycerol-free approach overcomes the limitations of conventional methods, providing a vital experimental platform for investigating trehalose-mediated intracellular vitrification mechanisms and advancing trehalose-based anhydrous cell preservation technologies.

Surface-enhanced Raman scattering (SERS) substrates hold great potential for rapid food monitoring, but the inability to reconcile high enhancement, uniformity, and stability presents a critical trilemma for substrate design and practical applications. Here, inspired by the hierarchical structure of lychee, we developed an innovative "integrated balanced enhancement-protection" strategy, fabricating a lychee-like Au@Ag/Au nanoarray chip via a simple, uniform oil/water/oil (O/W/O) self-assembly technique. This design mimics the architecture of lychee: the outer Au layer, modeled after lychee’s peel, provides chemical protection; the inner Au@Ag layer, mirroring lychee’s seed and flesh, delivers strong plasmonic enhancement. The unique bilayer nanoarchitecture, featuring dense and vertically coupled plasmonic hotspots, ensures highly efficient electromagnetic (EM) field by Au-Ag synergistic effects. It demonstrates an analytical enhancement factor (AEF) of 2.30 × 10 6 , uniformity with a relative standard deviation (RSD) of 6.55 %, and stability with 93.26 % signal retention over 60 days. When coupled with a universal pretreatment protocol to minimize matrix interference, the platform enables sensitive and reliable quantitative detection of three representative food additives, including potassium sorbate (PS), sodium benzoate (SB), and sodium saccharin (SS) in complex fruit matrices. The limit of detection (LOD) for all analytes was below 75 mg/kg, substantially surpassing the national regulatory standards of China. Importantly, the chip demonstrated high recoveries (86.62–102.49 %) and low RSD (< 9 %) across multiple fruit types. Beyond food safety, this scalable, low-cost approach not only overcomes SERS substrate bottlenecks but also provides a viable field-deployable tool with broader implications for SERS-based analysis.

Screening of pathogenic bacteria is a key to avoid food poisoning. The major drawbacks of existing assays for foodborne bacteria detection include long time for culture, complex DNA extraction for the polymerase chain reaction (PCR), and low sensitivity for enzyme-linked immunosorbent assay (ELISA), greatly limiting their practical applications. Here, we developed a sensitive optical biosensor based on porous gold@platinum nanocatalysts (Au@PtNCs) and a passive three-dimensional (3D) micromixer for fast detection of Salmonella typhimurium. The target Salmonella cells were first separated using immunomagnetic nanoparticles and the passive 3D micromixer. Then, immune Au@PtNCs were labeled onto the target cells as signal output to catalyze hydrogen peroxide–3,3′,5,5′-tetramethylbenzidine. Finally, the absorbance was measured at 652 nm to calculate the bacterial amount. This optical biosensor could detect Salmonella at concentrations from 1.8 × 101 to 1.8 × 107 CFU/mL in 1 h. Its detection limit was calculated to be 17 CFU/mL. Besides, this passive 3D micromixer could magnetically separate 99% of target bacteria from the sample in 10 min. This biosensor has the potential to be extended to detect other bacteria by changing the antibodies.

Antibiotic resistance, the ability of some bacteria to resist antibiotic drugs, has been a major global health burden due to the extensive use of antibiotic agents. Antibiotic resistance is encoded via particular genes; hence the specific detection of these genes is necessary for diagnosis and treatment of antibiotic resistant cases. Conventional methods for monitoring antibiotic resistance genes require the sample to be transported to a central laboratory for tedious and sophisticated tests, which is grueling and time-consuming. We developed a paper-based chip, integrated with loop-mediated isothermal amplification (LAMP) and the “light switch” molecule [Ru(phen)2dppz]2+, to conduct turn-on fluorescent detection of antibiotic resistance genes. In this assay, the amplification reagents can be embedded into test spots of the chip in advance, thus simplifying the detection procedure. [Ru(phen)2dppz]2+ was applied to intercalate into amplicons for product analysis, enabling this assay to be operated in a wash-free format. The paper-based detection device exhibited a limit of detection (LOD) as few as 100 copies for antibiotic resistance genes. Meanwhile, it could detect antibiotic resistance genes from various bacteria. Noticeably, the approach can be applied to other genes besides antibiotic resistance genes by simply changing the LAMP primers. Therefore, this paper-based chip has the potential for point-of-care (POC) applications to detect various gene samples, especially in resource-limited conditions.

s Perfluorinated compounds (PFCs), due to their persistence, toxicity, and bioaccumulation, pose significant environmental and public health concerns. However, their low concentrations and the complexity of environmental matrices present major analytical challenges. In this study, we developed a carbon quantum dot-based fluorescent sensing array integrated with membrane enrichment technology for the sensitive and selective detection of three representative PFCs: PFOA, PFOS, and PPVE. Three types of carbon quantum dots with distinct surface functionalities were synthesized, enabling differential fluorescence quenching upon interaction with target analytes. To improve detection sensitivity and reduce matrix effects, a membrane-based enrichment strategy was employed, allowing selective preconcentration and visible signal generation on the membrane surface. The platform achieved detection limits as low as 0.33, 0.35, and 0.40 μM for PFOA, PFOS, and PPVE, respectively. Multivariate analysis using linear discriminant analysis (LDA) and hierarchical agglomerative clustering (HAC) enabled clear discrimination of the three PFCs. The method was successfully applied to real water samples, achieving identification accuracies of 95.82%, 93.23%, and 90.48% in tap, river, and lake water, respectively.
