Multifunctional Poly(acrylic acid)/Chitosan nanoparticle network hydrogels with tunable mechanics

Xuelian Wei,Caikun Liu,Zhengxiang Gu,Kui Luo,Junxiao Yang,Shiyong Zhang

Nanoparticle network hydrogels (NNHs) employ nanoparticles as key building blocks of hydrogel networks to provide excellent mechanical properties and multifunctionality. Herein, poly(acrylic acid)/chitosan nanoparticle network hydrogels (PAAc/CS NNHs) with prominent mechanical properties were fabricated using catechol functional methacryloyl chitosan-silver nanoparticles (CFMC-Ag NPs) as multifunctional nanoparticle crosslinkers. The PAAc/CS NNHs were composed of hybrid networks derived from CFMC-Ag NPs crosslinked PAAc and entangled chitosan (CS) chains. The mechanical properties of PAAc/CS NNHs were highly impacted by the concentration of CFMC-Ag NPs. A maximum tensile strength of 2.22 MPa, a compressive strength of 10.61 MPa, and a toughness of 5.57 MJ·m −3 were obtained at a nanoparticle concentration of 10 mg/mL. In addition, PAAc/CS NNHs exhibited great self-recovery performance and excellent fatigue resistance. It was demonstrated that PAAc/CS NNHs possessed antibacterial activity, electrical conductivity and anti-freezing capability. This strategy could be applied for the preparation of other multifunctional hydrogels with significant mechanical properties.

An injectable multifunctional thermo-sensitive chitosan-based hydrogel for periodontitis therapy

Shiqing Ma,Xuemei Lu,Xinying Yu,Yaqi Du,Shendan Xu,Minting Li,Cheng Peng,Zihao Liu,Jiayin Deng

Bacteria are recognized as the driving factors of periodontitis. However, excessive reactive oxygen species (ROS) can harm periodontal tissue while also causing an uncontrolled inflammatory response. Hence, eliminating excessive ROS and blocking ROS-induced abnormal inflammatory response by antioxidants are achieving remarkable results in periodontitis therapy. Moreover, influenced by the deep and irregular periodontal pockets, injectable thermo-sensitive chitosan-based hydrogels have attracted a lot of attention. This study aimed to formulate an antibacterial and antioxidant therapeutic regimen by incorporating antimicrobial peptides (Nal-P-113) and/or antioxidants (polydopamine nanoparticles, PDNPs) into chitosan-based hydrogels. The hydrogel was characterized in vitro and finally examined in rats using the experimental periodontitis model. The release kinetics showed that the hydrogel could stably release Nal-P-113 and PDNPs for up to 13 days. The scavenging activity of the hydrogel against DPPH was about 80 % and the antibacterial ratio against Streptococcus gordonii ( S. gordonii ), Fusobacterium nucleatum ( F. nucleatum ) and Porphyromonas gingivalis ( P. gingivalis ) was about 99 %. Importantly, it was examined that the hydrogel had the ability to prevent periodontal tissue damage. Thus, chitosan-based hydrogels may provide a basis for designing multifunctional local drug delivery biomaterials for the treatment of periodontitis.

Water-in-water emulsions stabilized by self-assembled chitosan colloidal particles

Wanying Cui,Chunmiao Xia,Sheng Xu,Xinke Ye,Yihao Wu,Shukai Cheng,Rongli Zhang,Cuige Zhang,Zongcheng Miao

Dextran (Dex) and poly(ethylene glycol) (PEG)-based aqueous emulsions were stabilized using the self-assembled chitosan colloidal particles (CS CPs). Besides, the effects of pH, CS CPs concentration, polymer concentration, volume ratio of PEG solution to Dex solution, temperature, homogenizing speed and homogenizing time on the property of the W/W emulsions were investigated, respectively. In order to enhance the stability of the PEG-Dex emulsion, sodium tripolyphosphate was used to cross-link the CS CPs at the interface of emulsion droplets, which resulted in the stability duration for >1 year. Finally, the CS CPs were used as a support to immobilize urease and bovine serum albumin and a stabilizer to prepare W/W emulsion, which were then adopted as a catalysis system and as a spinning solution to fabricate drug-loaded nanofiber . This strategy potentially provides a new opportunity to encapsulate the active molecules at the water-water interface, and enrich the types of usable active molecules in the encapsulation in the W/W emulsions.

A new chitosan-based thermosensitive nanoplatform for combined photothermal and chemotherapy

Yanyan Zhang,Gareth R. Williams,Jiadong Lou,Wanting Li,Cuiwei Bai,Tong Wang,Shiwei Niu,Chun Feng,Li-Min Zhu

Targeting the delivery of anti-cancer drugs to a tumor site is essential for effective treatment and to ensure minimal damage to healthy cells and tissues. In this work, a chitosan-based nanoplatform was constructed for combined photothermal therapy and chemotherapy of breast cancer. The pH-sensitive and biocompatible biopolymer chitosan (CS) was grafted with N -vinylcaprolactam (NVCL) and modified with biotin (Bio), imparting it with temperature sensitive property and also the ability for active targeting. The polymer self-assembled to give nanoparticles (NPs) loaded with indocyanine green (ICG) and doxorubicin (DOX). When the NPs are exposed to near-infrared (NIR) laser irradiation, ICG converts the light to heat, inducing a significant phase transition in the NPs and facilitating the release of the drug cargo. In addition, the solubility of chitosan is increased in the slightly acidic microenvironment of the tumor site, which also promotes drug release. A detailed analysis of the NPs both in vitro and in vivo showed that the carrier system is biocompatible, while the drug-loaded NPs are selectively taken up by cancer cells. Particularly when augmented with NIR irradiation, this leads to potent cell death in vitro and also in an in vivo murine xenograft model of breast cancer.

Water-in-oil Pickering emulsions stabilized by phytosterol/chitosan complex particles

Chenglei Wang,Hang Jiang,Yunxing Li

The development of colloidal particles for the preparation of water-in-oil (w/o) Pickering emulsions is highly desirable for food, pharmaceuticals, and cosmetics. However, the lack of bio-based hydrophobic particulate stabilizers makes it remain a great challenge. Herein a water-in-cyclopentasiloxane (w/o) Pickering emulsion was prepared using phytosterol/chitosan (PS/CS) complex particles as the stabilizer. Adsorption of CS onto the surface of PS particles via electrostatic interaction changed the particle wettability and interfacial activity, leading both PS particles and CS to be absorbed at the w/o interface. By varying the pH and ionic strength of aqueous phase, the stabilizing mechanism was verified and the results suggested that the formation of PS/CS complex particles was critical in the preparation of w/o Pickering emulsions with small and uniform droplets. The PS/CS complex particles can emulsify a broad range of water volume fraction and even be used to prepare a high internal phase w/o Pickering emulsion. Meanwhile, the as-prepared w/o Pickering emulsions had good long-term stability, especially at ambient or lower temperature. Finally, we demonstrated that the prepared w/o Pickering emulsion could effectively protect encapsulated epigallocatechin gallate from heat and pH shock.

Kaolin-loaded carboxymethyl chitosan/sodium alginate composite sponges for rapid hemostasis

Yannan Song,Shuo Li,Huifang Chen,Xinyi Han,Gregory J. Duns,Wubliker Dessie,Wufei Tang,Yimin Tan,Zuodong Qin,Xiaofang Luo

There are several factors that contribute to the mortality of people who suffer from unmanageable bleeding. Therefore, the development of rapid hemostatic materials is necessary. Herein, novel rapid hemostatic composite sponges were developed by incorporation of kaolin (K) into carboxymethyl chitosan (CMCS)/sodium alginate (SA) via a combination of methods that includes ionic crosslinking, polyelectrolyte action, and freeze-drying. The CMCS/SA-K composite sponges were cross-linked with calcium ions provided by a sustained-release system consisting of D-gluconolactone (GDL) and Ca-EDTA, and the hemostatic ability of the sponges was enhanced by loading the inorganic hemostatic agent-kaolin (K). It was demonstrated that the CMCS/SA-K composite sponges had a good porous structure and water absorption properties, excellent mechanical properties, outstanding biodegradability , and biocompatibility. Simultaneously, they exhibited rapid hemostatic properties, both in vitro and in vivo . Significantly, the hemostatic time of the CMCS/SA-K 60 sponge was improved by 82.76 %, 191.82 %, and 153.05 %, compared with those of commercially available gelatin sponges in the rat tail amputation, femoral vein, and liver injury hemorrhage models respectively, indicating that its hemostatic ability was superior to that of commercially available hemostatic materials. Therefore, CMCS/SA-K composite sponges show great promise for rapid hemostasis .

Selective removal of Au(III) from wastewater by pyridine-modified chitosan

Lingyuan Chen,Jiali Tang,Suoque Wu,Shixing Wang,Zhaogang Ren

A pyridine-modified chitosan ( PMCS ) adsorbent with excellent adsorption performance and specific selectivity for Au(III) in wastewater was prepared. FTIR, SEM, XPS and XRD were used to study its adsorption performance, and various models were used to fit and analyze the experimental adsorption data. The maximum adsorption capacity of PMCS for Au(III) is 549.41 mg/g (298 K, pH = 4). The data analysis results prove that the PSO and Langmuir models best describe the adsorption process. Adsorption experiments in the presence of multiple ions proved that PMCS is selective for the adsorption of Au(III). The adsorption and desorption experiments showed that the adsorption rate of PMCS could still reach 94.77% after three cycles. In summary, PMCS was demonstrated to be a high-quality material for the adsorption of Au(III) from wastewater due to its excellent adsorption performance, specific selectivity and reusability.

Zhiyuan Yang,Tong Ye,Fei Ma,Xinhong Zhao,Lei Yang,Guifang Dou,Hui Gan,Zhuona Wu,Xiaoxia Zhu,Ruolan Gu,Zhiyun Meng

Uncontrolled hemorrhage from trauma or surgery can lead to death. In this study, chitosan/kaolin (CSK) and chitosan/montmorillonite (CSMMT) composites were prepared from chitosan (CS), kaolin (K), and montmorillonite (MMT) as raw materials to control bleeding. The physiochemical properties and surface morphology of CSK and CSMMT composites were analyzed by Fourier transform infrared spectrometry (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM), zeta potentials, and X-ray fluorescence (XRF). The hemostatic mechanism was measured in vitro by activated partial thromboplastin time (APTT), prothrombin time (PT), in vitro clotting time, erythrocyte aggregation, and thromboelastogram (TEG). The hemostasis ability was further verified by using tail amputation and arteriovenous injury models in rats. The biocompatibility of CSK and CSMMT was evaluated by in vitro hemolysis, cytotoxicity assays, as well as acute toxicity test and skin irritation tests. The results show that CSK and CSMMT are promising composite materials with excellent biocompatibility and hemostatic properties that can effectively control bleeding. Keywords:chitosan;kaolin;montmorillonite;hemostatic material;hemorrhage control

Effect of chitosan oligosaccharide glycosylation on the emulsifying property of lactoferrin

Wen-Duo Wang,Chao Li,Chun Chen,Xiong Fu,Rui Hai Liu

There is fast increasing interest in the development of alimentary protein stabilized emulsions due to their potential applications in functional food fields. This work studied the effect of glycation degree with chitosan oligosaccharide (COS) on the emulsifying properties of lactoferrin (LF) through Maillard reaction. In the present study, SDS-PAGE and FT-IR were used to confirm LF and COS covalently binding together successfully. Intrinsic fluorescence showed that glycation with COS led more hydrophobic groups exposed to the surface of the structure and particle size increase of LF. Emulsions with 50% ( v /v) oil phase and protein concentration of 2% ( w / v ) was fabricated through one-step shear method. Compared with native LF, emulsions stabilized by LF-COS conjugates showed smaller droplet size and lower creaming index (CI). Among these samples, LF-COS conjugates under 4 h had the best emulsifying efficiency and stability, the emulsion droplet size and the CI of which decreased 39.66% and 28.55% compared with LF, respectively. Furthermore, glycation with COS enhanced the interfacial activity of LF leading to more adsorbing amount and forming thicker layer on the droplets and gel network in the emulsions. This finding would make sense to further understand the modification of emulsifying properties of alimentary proteins through glycosylation with saccharides and develop novel protein-based emulsifiers.

Structural characteristics and rheological properties of hydroxypropyl trimethyl ammonium chloride chitosan

Ling Ding,Zhao Liu,Shen Song,Mohamed Aamer Abubaker,Lele Chen,Jipeng Shi,Zengjie Fan,Ji Zhang

Hydroxypropyl trimethyl ammonium chloride chitosan (HACC) was synthesized by reacting chitosan with glycidyl trimethylammonium chloride. Atomic force microscopy showed that HACC exhibited disorderly coils in dilute solution and formed a three-dimensional network. Flow, thixotropy , and dynamic viscoelasticity tests were conducted using an MCR301 rheometer. The HACC solution was a non-Newtonian pseudoplastic fluid, and the shear behavior of different concentrations (2–6 %, w/v) was evaluated by the Williamson model fitting. Furthermore, the thixotropy was highly dependent on concentration changes: the high-concentration solution structure was difficult to recover in a short time. The dynamic viscoelasticity test indicated that the viscoelasticity of the HACC solution not only exhibited a viscous behavior similar to that of a fluid, but also exhibited elastic properties of weak gel. HACC exhibited high-strength solid-like gel characteristics at high temperature.

Design of Biocompatible Chitosan/Polyaniline/Laponite Hydrogel with Photothermal Conversion Capability

Liying Zhang,Gao He,Yang Yu,Yu Zhang,Xiang Li,Shige Wang

In recent years, multifunctional hydrogels have received a great deal of attention because they are biocompatible and can mimic the extracellular matrix. Herein, we prepared hydrogels of biocompatible cross-linked networks with photothermal properties. In this study, a chitosan/polyaniline/laponite (COL) hydrogel with photothermal conversion capability was designed. Polyaniline was firstly grafted onto chitosan and its solution was mixed with oxidized dextran, which was then cross-linked into a hydrogel via a Schiff base reaction. Furthermore, an aluminosilicate clay material, laponite (LAP), was incorporated into the hydrogel. The swelling ratio of the COL hydrogel in various solutions was greater than 580%, and it showed good degradation ability (the mass–loss ratio was over 45% after 28 days). This composite hydrogel was demonstrated to have good photothermal conversion properties and biocompatibility at both the cell (cell viability was over 97%) and animal levels. The COL hydrogel showed a photothermal conversion efficiency of 23.7% under the irradiation of a near-infrared laser. Coupled with the osteogenic differentiation-inducing potential of LAP, the COL hydrogel has the potential to kill tumors via hyperthermia or serve as scaffolds for bone tissue regeneration. Keywords:hydrogel;chitosan;polyaniline;photothermal;biocompatibility

Preparation and performance of chitosan/cyclodextrin-g-glutamic acid thermosensitive hydrogel

Hui Yun Zhou,Jia Nan Tong,Li Jun Ren,Pei Yan Hao,Hui Jie Zheng,Xu Ming Guo,Ya Wei Chen,Jun Bo Li,Hyun Jin Park

Chitosan hydrogel had been widely used in cell culture and tissue engineering, but its long gel time, weak mechanical strength, and weak antibacterial performance limited its application. In this study, the cyclodextrin grafted glutamic acid , a water-soluble cyclodextrin derivatives , was prepared by glutamic acid (Glu) and mono-(6-OTs)-β-CD under alkaline conditions and confirmed by FTIR and 1 H NMR. Then chitosan (CS) and cyclodextrin grafted glutamic acid (CD-g-Glu) were used to prepare chitosan/cyclodextrin derivatives thermosensitive hydrogel (CS/CD-g-Glu) to improve the properties of chitosan-based thermosensitive hydrogels. The CS/CD-g-Glu hydrogel had temperature sensitivity and stable three-dimensional network structure. In addition, the hydrogel loaded with berberine hydrochloride (BBH) showed optimal sustained-release effect, and the cumulative release rate was 57% in 24 h. More importantly, the CS/CD-g-Glu hydrogel loaded with BBH showed prefect antibacterial activity against E. coli and S. aureus . Therefore, the CS/CD-g-Glu hydrogel has great promising applications in drug delivery, tissue engineering, biotechnological, and biomedical fields.
