Photodynamic inactivation (PDI), as a new non-thermal decontamination and preservation technology, exhibits the advantages of low cost, free of residue and non-microbial resistance. Curcumin (CUR) as a natural photosensitizer could effectively inactivate food spoilage bacteria at an absorbance wavelength (420–470 nm) light illumination. In this study, the in vitro influence of PDI process parameters on the inactivation of Staphylococcus saprophyticus and the antimicrobial mechanism was analyzed. Approximate 5 log CFU/ml reduction was obtained when treated with 25 μM CUR and 4.32 J/cm 2 of blue LED. Obvious cell wrinkles, cell membrane disruption, intracellular contents leakage and increased membrane permeability of S. saprophyticus cell were observed after CUR-PDI treatment. Further, generation of intracellular reactive oxygen species, along with decreased intracellular enzyme activities and increased degree of cell membrane depolarization verified cell damage. The in vivo photo-antibacterial effect (1.04 log CFU/g and 0.81 log CFU/g) was validated on fresh millet dough sheet at two initial bacterial loads (5 log CFU/g and 3 log CFU/g, respectively). These findings illustrated that CUR mediated PDI could be applied to inhibit S. saprophyticus growth and possibly prevent spoilage of fresh dough products.

BACKGROUND Due to its high glutenin content, walnut protein isolate (WPI) exhibits low solubility, limiting its applications in the food industry. RESULTS WPI was modified with succinic anhydride (SA), and the potential of the resulting curcumin-loaded emulsions was evaluated. The degree of succinylation of WPI was measured as 89.52 ± 0.66% when 40% SA was used. Succinylation enhanced the surface charge of WPI, reduced its surface hydrophobicity and disulfide bond content, and involved both lysine residues and free sulfhydryl in the acylation reaction. Structural analyses, including Fourier transform infrared spectroscopy, intrinsic fluorescence, and scanning electron microscopy, revealed that SA modification induced significant conformational changes in WPI. Notably, at 20% SA addition, WPI exhibited optimal functional properties, achieving solubility as high as 98.24%. In vitro digestion assays demonstrated that succinylation decreased the release of free fatty acids from emulsions and improved the bioavailability of curcumin to 86.81%. CONCLUSION This study demonstrated that SA modification is an effective way for enhancing the structural and functional properties of WPI. The SA-modified WPI shows potential as a carrier for improving the solubility and bioavailability of hydrophobic bioactive compounds. Moreover, this work provides new insights into the significance of WPI modification and offers practical guidance for its application in the food industry. © 2026 Society of Chemical Industry.

Background The process of healing wounds in diabetic patients is intricate and is notably obstructed by a disordered wound microenvironment, characterized by chronic inflammation and elevated blood glucose. The combination of stem cell therapy and drug treatment is seen as a promising application in future. However, the limited proliferative capacity of stem cells and inadequate drug availability present significant challenges for achieving optimal therapeutic outcomes. Methods In this study, open porous poly (lactic‒coglycolic acid) (PLGA) microspheres were designed and synthesized via gas-assisted volatilization microemulsion technology. These microspheres encapsulate curcumin and allow its slow release, thereby enhancing wound repair. The large pores in the microspheres provide ample support for bone marrow stem cells (BMSCs), enabling continuous drug release over a period of 35 days. Results The sustained release of curcumin promoted stem cell proliferation and maintained stem cell activity. Additionally, it facilitates remodeling of the wound immune microenvironment. Additionally, the microspheres can activate mitochondrial autophagy in cells, effectively alleviating wound inflammation. Conclusions The combined actions of curcumin and stem cells aid in regenerating blood vessels and revitalizing the collagen network where the injury occurred, thus improving wound healing capabilities. Consequently, integrating drugs with stem cells and microspheres holds significant potential for diabetic wound treatment.

Porous microneedles (PMNs) can efficiently load drugs via capillary action within their porous structure and enable controlled drug release in deep wound layers, thereby significantly promoting wound healing. However, increasing the porosity of PMNs typically compromises mechanical strength, leading to needle tip deformation during skin penetration. To overcome this limitation, a mechanically robust porous microneedle (PMN) was fabricated by lyophilizing a chemically and physically dual-crosslinked hyaluronic acid/polyvinyl alcohol hydrogel formed in situ within a microneedle mold. The resulting PMN exhibited a single-needle bearing force exceeding the minimum threshold for skin penetration (>0.1 N) while maintaining a relatively high porosity (28.7%). Furthermore, a curcumin-loaded PMN (PMN@Cur) was easily prepared by adsorbing a curcumin/β-cyclodextrin complex solution into the abundant pores of PMN. The PMN@Cur showed sustained drug release, along with superior antioxidant and antibacterial activities. In infected wounds models, the PMN@Cur attained a wound contraction rate of up to 87.2% by day 14, demonstrating its high therapeutic efficacy in wound healing. These findings suggest that PMN@Cur, which combines high porosity with favorable mechanical properties, holds significant promise as a drug delivery system for advanced wound care.

Pulmonary hypertension (PH) is a severe pulmonary vascular disease that ultimately leads to right heart failure, which involves multiple core mechanisms, including oxidative stress, inflammation, and pathological cellular proliferation. However, current therapies are limited in simultaneously modulating multiple core mechanisms, leading to poor clinical outcomes and high mortality. Herein, we introduce tetrahedral framework nucleic acids-Curcumin (tFNAs-Cur) as a novel therapeutic for PH, targeting multiple synergistic pathways including TGF β1, NF-κB, and PI3K-Akt. The proposed tFNAs-Cur featured the advantages of high drug loading capacity, good water solubility, strong stability, and excellent biocompatibility. Cellular experiments demonstrate that tFNAs-Cur is efficiently internalized by pulmonary artery smooth muscle cells, alleviates oxidative stress, and inhibits their proliferation and migration. In the monocrotaline-induced PH rat model, tFNAs-Cur significantly improves pulmonary artery acceleration time, reduces vascular wall thickness, and medial pulmonary artery diameter. Cardiac assessments show improved exercise capacity, reduced Fulton index, and alleviated right ventricular fibrosis. Further transcriptomic and biochemical analyses reveal that the therapeutic effects of tFNAs-Cur were mediated via the ROS/TGF β1 antioxidant, NF-κB anti-inflammatory, and PI3K-Akt anti-proliferative pathways. The proposed tFNAs-Cur provides an efficient and safe method with potential for clinical translation that simultaneously utilizes multiple synergistic mechanisms for the treatment of PH.

Objective: Inhibition of tumor angiogenesis has become a new targeted tumor therapy. In this study, we established a micellar carrier with a tumor neovascularization-targeting effect modified by the neovascularization-targeting peptide NGR. Methods: The targeted polymer poly(ethylene glycol)-b-poly(lactide-co-glycolide) (PEG-PLGA) modified with Asn–Gly–Arg (NGR) peptide was prepared and characterized by 1 H nuclear magnetic resonance and Fourier-transform infrared spectrometry. NGR-PEG-PLGA was used to construct curcumin (Cur)-loaded micelles by the solvent evaporation method. The physicochemical properties of the micelles were also investigated. Additionally, we evaluated the antitumor efficacy of the polymer micelles (PM) using in vitro cytology experiments and in vivo animal studies. Results: The particle size of Cur-NGR-PM was 139.70 ± 2.51 nm, and the drug-loading capacity was 14.37 ± 0.06%. In vitro cytological evaluation showed that NGR-modified micelles showed higher cellular uptake through receptor-mediated endocytosis pathways than did unmodified micelles, leading to the apoptosis of tumor cells. Then, in vivo antitumor experiments showed that the modified micelles significantly inhibited tumor growth and were safe. Conclusions: NGR-modified micelles significantly optimized the therapeutic efficacy of Cur. This strategy offers a viable avenue for cancer treatment. Export

To prevent food spoilage caused by microbial infection, the development of an environmentally friendly antimicrobial preservation material is crucial. Here, the microporous γ-CD-MOFs was utilized to encapsulate the hydrophobic active substance curcumin, resulting in the preparation of a non-toxic antimicrobial material (Cur-CD-MOFs). The results revealed that curcumin encapsulation in Cur-CD-MOFs occurred primarily in the carbonyl group, benzene ring, and enolic side ring of curcumin. The Cur-CD-MOFs had a 100% bactericidal effect on Escherichia coli and Staphylococcus aureus at 4 h and 8 h, and a strong inhibitory effect on aerial mycelium of Penicillium expansum and Botrytis cinerea . Furthermore, the incorporation of Cur-CD-MOFs improved the Pul/Tre film barrier and mechanical properties. The effectiveness of Cur-CD-MOFs-Pul/Tre in retaining fruit freshness was validated using Centennial Seedless grapes. This study confirmed that Cur-CD-MOFs is a promising antibacterial material, and Cur-CD-MOFs-Pul/Tre will be a potent candidate for food preservation.

Foodborne spoilage bacteria are the key factor causing food deterioration, seriously restricting the shelf life of food and threatening consumer safety. In this study, we investigated the synergistic bactericidal effect of curcumin-mediated photodynamic inactivation technology (Cur-PDI) combined with eugenol (Eug) on two typical f foodborne spoilage bacteria Leuconostoc mesenteroides and Carnobacterium maltaromaticum , and further evaluated its application in bacon preservation. The results indicated that under the combined treatment conditions (20 μΜ Cur, 25 μg/mL Eug, 20 min of light exposure), the viable bacterial counts of two bacteria decreased significantly by 7.11 log CFU/mL and 7.12 log CFU/mL compared with the blank control. As both Cur and Eug are lipophilic, the combination exhibited a stronger bactericidal effect in a lipid system than the water-soluble photosensitizer erythrosine (ERY). Mechanistic studies revealed that, compared with PDI alone, the combined treatment more significantly disrupted bacterial membrane integrity, induced higher levels of reactive oxygen species (ROS), and severely interfered with antioxidant defense and energy metabolism. In the bacon preservation experiment, the combined treatment effectively reduced the initial colony count of the product and continuously inhibited the growth of microorganisms during storage for 25 days. This study systematically clarified the synergistic antibacterial mechanism of Cur-PDI and Eug, and verified its potential in the preservation of bacon, providing a theoretical basis for the development of natural and efficient preservation strategies for foods rich in fat.

Surface fouling and bacterial contamination present significant challenges to daily life and industrial production, motivating the development of multifunctional surfaces with antifouling and antimicrobial properties. In this context, the development of multifunctional surfaces based on environmentally friendly green materials, which possess both superhydrophobic anti-fouling and highly efficient antibacterial properties, has become one of the approaches to address the aforementioned issues. In this study, superhydrophobic antimicrobial multifunctional coatings were successfully prepared by using environmentally friendly materials chitosan (CS) and curcumin (Cur) as antimicrobial agents, shellac and rosin instead of the traditional fluoride as low surface energy substances and adhesives, and nano‑calcium carbonate (Nano-CaCO 3 ) as hydrophobic particles. The multifunctional coating exhibits a contact angle exceeding 150° for both water and other liquids, demonstrating exceptional liquid-repellent properties and superior antifouling performance. Under an 80 g load, the coating withstands 45 cycles of sandpaper abrasion testing and 50 cycles of adhesive tape peeling testing. Furthermore, the composite coating samples show a 28% increase in tear strength and a 24% enhancement in puncture resistance compared to the pristine counterpart. The coating exhibits significant antibacterial activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), achieving antibacterial efficiency rates of 99.1% and 99.3%, respectively. In strawberry preservation tests, the shelf life of the samples was extended to the sixth day. Composed of non-toxic and sustainable chemicals, the coating integrates superhydrophobicity, antifouling properties, and antimicrobial functionalities, thereby demonstrating broad applicability across diverse sectors, including paper protection, packaging, biomedical applications, and the food industry.

The repair of irregular bone defects poses a significant challenge for orthopedic surgeons. This study presents a customizable, novel bone tissue engineering scaffold capable of promoting bone regeneration, which is composed of curcumin-quercetin-piperine-loaded poly(lactic-co-glycolic acid) nanoparticles (Cur-Que-Pip-PLGA NPs) loaded onto 3D-printed porous tantalum (3DPTa). The Cur-Que-Pip-PLGA NPs were prepared via the emulsification-solvent evaporation method, and subsequently loaded onto the 3DPTa scaffold via the physical impregnation method. The morphology and structure of the composite scaffold were characterized by scanning electron microscopy (SEM), and its drug release profile was evaluated via high-performance liquid chromatography (HPLC). The biocompatibility and osteogenic potential of the scaffold were evaluated. Cell proliferation assays and live/dead cell staining demonstrated that the composite scaffold exhibited excellent biocompatibility. Both in vitro and in vivo osteogenic experiments demonstrated that the composite scaffold significantly promoted the osteogenic differentiation of MC3T3-E1 cells and new bone formation. These results indicate that this novel Cur-Que-Pip-PLGA NPs 3DPTa scaffold has excellent biocompatibility and osteogenic potential, thus representing a promising candidate material for bone defect repair.

Background Ulcerative colitis (UC) is a common type of inflammatory bowel disease, where the vicious cycle of inflammation and oxidative stress poses a major challenge in its treatment, and existing therapies have limitations. Berberine (BBR) and curcumin (CUR) have the potential for synergistic treatment of UC, but this potential has not been verified in UC. Additionally, both BBR and CUR suffer from poor water solubility and low bioavailability. This study aims to construct prebiotic microcapsules (BBR/CUR@MC) for the co-delivery of BBR and CUR and explore their therapeutic mechanism in UC. Methods Network pharmacology was used to predict the targets and pathways of BBR and CUR in UC. BBR/CUR@MC was prepared using microfluidic electrospray technology, and its colon targeting and biocompatibility were evaluated through in vivo experiments. In a Dextran sulfate sodium (DSS)-induced UC mouse model, the therapeutic effect was assessed using multiple indicators, and the mechanism of action was explored by transcriptome analysis. Results Network pharmacology showed that BBR and CUR can exert therapeutic effects on UC through synergistic regulation of TNF and AGE-RAGE signaling pathways. The successfully constructed BBR/CUR@MC had good colon targeting and biocompatibility. In the mouse colitis model, oral administration of BBR/CUR@MC inhibited ADAM17 in the TNF signaling pathway and MAPK11/13, COL4A1, and COL1A1 in the AGE-RAGE signaling pathway, thereby downregulating pro-inflammatory cytokines such as TNF-α and IL-1α, upregulating IL-10, scavenging ROS, significantly alleviating colonic inflammation and repairing the intestinal barrier in mice, with a therapeutic effect superior to that of single-drug microcapsules and 5-ASA. Conclusion BBR/CUR@MC can exert synergistic anti-inflammatory and antioxidant effects in UC treatment by regulating TNF and AGE-RAGE signaling pathways, providing a new multi-mechanism therapeutic strategy for UC.

The low bioavailability of nutraceuticals, due to chemical instability and non-targeted release, remains a major delivery challenge. To address this, biodegradable core-shell hydrogel microspheres with controllable diameters ranging from 800 to 2850 μm were fabricated via an extrusion method for the co-delivery of hydrophilic and hydrophobic nutraceuticals. These microspheres employ Fe 2 O 3 -loaded montmorillonite (Fe Mt) as the reinforcing scaffold, sodium carboxymethyl cellulose (CMC) as the core, and chitosan (CS) as the shell. They were co-loaded with hydrophilic l -glutamine (L-Gln) and hydrophobic curcumin (Cur) as model nutraceuticals. The incorporation of Fe Mt overcomes the low encapsulation efficiency and mechanical weakness of polymer microspheres. Meanwhile, at Fe Mt contents of 6.00 wt% (for L-Gln) and 5.00 wt% (for Cur) relative to the CMC weight, the encapsulation efficiencies reach up to 75.42% and 63.16%, respectively. The CS shell thickness increases from 10.83 to 16.05 μm as the CS concentration rises from 10.0 to 25.0 g/L. This thickness governs the release kinetics and pH-responsive behavior, wherein the microspheres contract in acidic environments and swell under neutral/alkaline conditions to facilitate targeted delivery in the gastrointestinal tract. In simulated digestion, cumulative release of nutraceuticals from the microspheres was 93.02% for L-Gln and 88.37% for Cur within 12 h. Additionally, the microspheres exhibited good storage stability (ca. 1.03% loss after 45 days). This work presents a highly promising and versatile platform for the co-delivery of hydrophilic and hydrophobic nutraceuticals with controlled release profiles.
