Polycystic ovary syndrome (PCOS) is a complicated endocrinopathy affecting women at reproductive age. Increasing evidence has shown the anti-PCOS effect of electroacupuncture (EA), a modified approach of traditional Chinese medical therapy “acupuncture”. However, the underlying mechanism of EA-alleviated PCOS waits further explored. In this study, experimental PCOS were induced in rats by dehydroepiandrosterone (DHEA) injection. Testosterone (T)-induced human ovarian granulosa cell (GC) line KGN was used to mimic PCOS in vitro . EA significantly alleviated histological changes and hormone disruption in PCOS rats. Besides, EA inhibited cell apoptosis, autophagy and the activation of endoplasmic reticulum (ER) stress-related PERK/eIF2α/ATF4/CHOP signaling in ovaries of PCOS rats. More interestingly, intermedin (IMD), a member of calcitonin gene-related peptide (CGRP), was evidently up-regulated in ovarian GCs after EA treatment, and its main bioactive form IMD 1-53 suppressed cell apoptosis, autophagy and PERK/eIF2α/ATF4/CHOP signaling in T-induced KGN cells. Consistent with IMD 1-53 , ER stress inhibitor 4-PBA exerted an inhibitory effect on T-induced cell apoptosis and autophagy in KGN cells. Collectively, this study validates the protective effect of EA on DHEA-induced PCOS, and proposes that IMD relieved apoptosis and autophagy in T-induced granulosa cells via inhibiting ER stress.

Rare earth (RE 3+ )-doped phosphors generally suffer from thermal quenching, in which their photoluminescence (PL) intensities decrease at high temperatures. Herein, we report a class of unique two-dimensional negative-thermal-expansion phosphor of Sc 2 (MoO 4 ) 3 :Yb/Er. By virtue of the reduced distances between sensitizers and emitters as well as confined energy migration with increasing the temperature, a 45-fold enhancement of green upconversion (UC) luminescence and a 450-fold enhancement of near-infrared downshifting (DS) luminescence of Er 3+ are achieved upon raising the temperature from 298 to 773 K. The thermally boosted UC and DS luminescence mechanism is systematically investigated through in situ temperature-dependent Raman spectroscopy, synchrotron X-ray diffraction and PL dynamics. Moreover, the luminescence lifetime of 4 I 13/2 of Er 3+ in Sc 2 (MoO 4 ) 3 :Yb/Er displays a strong temperature dependence, enabling luminescence thermometry with the highest relative sensitivity of 12.3%/K at 298 K and low temperature uncertainty of 0.11 K at 623 K. These findings may gain a vital insight into the design of negative-thermal-expansion RE 3+ -doped phosphors for versatile applications.

Background Parkinson's disease (PD) is an age-related neurodegenerative disorder without effective treatments. Mesencephalic astrocyte-derived neurotrophic factor (MANF) has been suggested to be capable of protecting against PD by inhibiting endoplasmic reticulum (ER) stress-mediated neuronal apoptosis. Purpose This study was aimed to evaluate the antiparkinsonian effect of dendrobine and reveal its underlying mechanisms from the perspective of MANF-mediated ER stress suppression. Methods Behavioral assessments of PD mice as well as LDH/CCK-8 assay in SH-SY5Y cells and primary midbrain neurons were carried out to detect the antiparkinsonian effect of dendrobine. Immunofluorescence, western blot, flow cytometry and shRNA-mediated MANF knockdown were used to determine the apoptosis of dopaminergic neurons and the expressions of ER stress-related proteins for investigating the underlying mechanism of dendrobine. Results Dendrobine significantly ameliorated the motor performance of PD mice and attenuated the injuries of dopaminergic neurons. Dendrobine could also relieve neuronal apoptosis, up-regulate MANF expression and inhibit ER stress, which were largely abolished by shRNA-mediated MANF knockdown in PD model. Conclusion Dendrobine might protect against PD by inhibiting dopaminergic neuron apoptosis, which was achieved by facilitating MANF-mediated ER stress suppression. Our study suggested that dendrobine could act as a MANF up-regulator to protect against PD, and provided a potential candidate for exploring etiological agents of PD.

Rare-earth-ion-doped nanocrystals have low quantum conversion efficiency due to their high surface-to-volume ratios and many surface defects, which induce fluorescence quenching. We propose a laser annealing strategy that uses a 974.5 nm laser with a high power density to heat Er3+-doped Y2O3 nanocrystals for several seconds. As a result, the upconversion of Er3+-doped Y2O3 nanocrystals is enhanced obviously after laser annealing: the green emission is enhanced by 90 times and red emission by 24 times. The main reason for the upconversion enhancement is the ultrahigh temperature caused by laser annealing, which increases the grain size and reduces the surface defects. It is an encouraging result, and laser annealing is suitable for various rare-earth-ion-doped oxide nanoparticles. Then, the strong upconversion is applied to measure temperature. After laser annealing, the highly improved fluorescence efficiency decreases the required excitation power density. The thermal effect of excitation light is negligible when the excitation power density is less than 0.13 W/mm2. The thermally coupled levels of I538/I563 and nonthermally coupled levels of I661/I800 are studied for thermometry. The experimental results show that I661/I800 has high absolute sensitivity than I538/I563 at a temperature below 400 K. Rate equations and multiphonon transitions are applied to explain the relationship between the nonthermally coupled fluorescence intensity ratio and temperature. It is instructive for the design of new temperature sensors.

Chlorpyrifos (CPF) has caused large-scale pollution worldwide and posed a threat to non-target organisms. Baicalein (BAI) is a flavonoid extract with anti-oxidant and anti-inflammatory activities. The gills are the mucosal immune organ and the first physical barrier of fish. However, it is not clear whether BAI counteracts organophosphorus pesticide CPF exposure-caused gill damage. Therefore, we established the CPF exposure and BAI intervention models by adding 23.2 μg/L CPF in water and/or 0.15 g/kg BAI in feed for 30 days. The results showed that CPF exposure could cause gill histopathology lesions. Moreover, CPF exposure led to endoplasmic reticulum (ER) stress, caused oxidative stress and Nrf2 pathway activation, and triggered NF-κB-mediated inflammation reaction and necroptosis in carp gills. BAI adding effectively relieved the pathological changes, and lighten inflammation and necroptosis involving in the elF2α/ATF4 and ATF6 pathways through binding to GRP78 protein. Moreover, BAI could ease oxidative stress, but did not affect Nrf2 pathway in carp gills under CPF exposure. These results suggested that BAI feeding could alleviate necroptosis and inflammation against chlorpyrifos toxicity through elF2α/ATF4 and ATF6 axis. The results partially explained the poisoning effect of CPF, and showed BAI could be act as an antidote for organophosphorus pesticides.

BackgroundAlthough exposure to individual metal does exhibit its toxicity, combined exposures provide a more effective representation of the toxic effects of different heavy metal exposures on public health as well as ecosystems. Furthermore, there are few studies on composite exposure to low concentrations of heavy metals, which is more consistent with real-life exposure. The purpose of this study was to explore the neurotoxicity induced by combined exposure to low concentrations of Lead (Pb) and cadmium (Cd) and the potential interaction of their mixture in vitro.MethodsPC12 cells were incubation with the corresponding concentration of cadmium chloride and/or lead acetate. Viability of PC12 cells was measured by CCK8 assay after 12, 24 and 48h incubation. Next, We measured the ROS, mitochondrial membrane potential (MMP) and apoptosis produced by different treated cells using ROS assay kit, JC-1 MMP assay kit and annexin V-FITC/propidium iodide (PI) apoptosis assay kit, respectively. Expression of proteins related to PI3K/AKT and endoplasmic reticulum (ER) stress in PC12 cells were tested by western blotting. Our study was the first to analyze the interaction between Pb and Cd using a 3 × 3 factorial design approach to observe neurotoxicity.ResultsThe results showed that the combined exposure of them was more cytotoxic than the single metal. The activation of PI3K/AKT signaling pathway and several parameters related to oxidative stress and ER stress were significantly altered in combined exposure to low concentrations of Pb and Cd compared with the Pb or Cd. Regarding apoptosis and ER stress, a synergistic interaction between Pb and Cd was evident. Moreover, evoked ER stress as a mechanism involved in the apoptosis of PC12 cells by the combined exposure to Pb and Cd.ConclusionThe present study provides a theoretical basis used for the toxicological assessment of metal mixtures induced neurotoxicity of concern in terms of public health, and more effective control measures should be taken for the environmental pollution caused by various mixed heavy metals discharged from industry and agriculture.

Herein, a series of Y2W3O12:10%Yb3+/x%RE3+ (RE = Er/Ho/Tm) phosphors is prepared via a solid-state reaction. The upconversion and downshift luminescence properties of the phosphors were investigated under an excitation of 980 nm. The bright blue light emission from Tm3+ ion and the green and red light emissions from Ho3+(Er3+) ions were observed. The near-infrared light intensity of NIR-I (Tm3+, ∼850 nm), NIR-II (Er3+: ∼1550 nm; Tm3+: ∼1783 nm) and NIR-III (Ho3+: ∼2050 nm) were analyzed. In particular, the dramatic thermal enhancement phenomenon in visible and NIR regions was exhibited by the Y2W3O12:10%Yb3+/x%RE3+ (RE = Er/Ho/Tm) phosphors. Among them, the green light intensity of Er3+ ions increased 26.77 times, from 303 to 573 K. The NIR-II emission band (∼1783 nm) intensity of Tm3+ ions at 533 K increased 168.7 times compared to that at 313 K. The possible thermal enhancement mechanism is illustrated by the negative thermal expansion (NTE) and Frenkel defect of the Y2W3O12 host. Finally, the optical temperature sensing performances of Y2W3O12:10%Yb3+/x%RE3+ (RE = Er/Ho/Tm) samples are investigated according to the luminescence intensity dependence relationship on temperature. The maximum value of SR reached 4.24% K−1 at 353 K for Y2W3O12:10%Yb3+/0.6%Ho3+ phosphor. The results indicate that the Y2W3O12:10%Yb3+/x%RE3+ (RE = Er/Ho/Tm) phosphors possess anti-thermal quenching properties and are suitable for developing optical temperature sensors.

Size-tunable β-NaYF 4 : Yb/Er (50–400 nm) up-converting nanoparticles (UCNPs) with a strong green emission were successfully prepared by a facile thermal decomposition method in a short period of time. The effects of the reaction time, the oleic acid (OA) content, and the doped Gd 3+ concentration on the crystal structure, the size evolution mechanism and the luminescence mechanism of the β-NaYF 4 : Yb/Er nanoparticles were discussed in detail. Characterization was completed using X-ray diffraction (XRD), transmission electron microscopy (TEM), high-resolution (HR-TEM), scanning electron microscopy (SEM), and photoluminescence (PL). The results reveal that the phase transition was completed at 15 min and the morphology of the NaYF 4 : Yb/Er UCNPs changed from spherical to large hexagonal disks (200–400 nm) in non-doped Gd 3+ . An appropriate decrease in the volume ratio of oleic acid (OA) to 1-octadecene (ODE) promoted NaF production, resulting in the formation of the hexagonal phase crystals and the subsequent increase of luminous intensity by 30–40 times. More importantly, the size of the β-NaYF 4 : Yb/Er decreased with Gd 3+ doping, and the UCNPs ultimately transformed into small hexagonal prisms (50 nm) with high monodispersity when the doping concentration of Gd 3+ was 60 mmoL. The PL spectra confirmed that the luminous intensity of the UCNPs with doped Gd 3+ was much higher than that of the undoped UCNPs. The results show that this route for UCNP fabrication with high luminous efficiency and a small size could be extended to other UCNP systems for multifunctional biomedical applications.

Due to the low abundance of miRNAs and the coordinated regulation of multiple miRNAs toward some biological processes, a method that can sensitively and simultaneously detect multiple miRNAs is needed. Here, a strand displacement dual amplification (SDDA) strategy was developed for multiple miRNAs analyses using a unique single strand-double strand-single strand DNA (sdsDNA), which generated by the target recognition probe hybridizing with the site region probe. During the process of SDDA, miRNA hybridized to the target recognition domain of sdsDNA, thereby releasing the site, which triggered the first strand displacement reaction. This strand displacement event resulted in the release of the other site, triggering the second strand displacement reaction. Both two strand displacement reactions in each assembly circle were integrated for amplified signals, which were tested by a fluorescence resonance energy transfer (FRET) assay. NaYF 4 :Yb,Tm/Er UCNPs and Ti 3 C 2 nanosheets were used as the energy donor-acceptor pair. By employing 1:2 ratio of donor-acceptor, the energy transfer efficiency between NaYF 4 :Yb,Tm/Er UCNPs and Ti 3 C 2 nanosheets could reach 90%, which enhanced signal-to-background ratio. Combining the benefits of dual amplification and efficient energy transfer on miRNAs detection, this assay exhibited a linear range from 5 fM to 100 pM for miRNA-21 and miRNA-10b. Moreover, this assay was applied to estimate the expression of miRNA-21 and miRNA-10b in human hepatoma and hepatocyte cell lines. Therefore, this FRET-based SDDA strategy has been used for highly sensitive detection of multiple miRNAs in cell lysate and facilitates the development of efficient biosensor platform.

We develop a facile layer-by-layer deposition process to create bi-functional Fe3O4@SiO2@Gd2O3:Yb,Er nanostructures composed of magnetic Fe3O4 cores, variable SiO2 mid-layers, and up-converting Gd2O3 shells. The synthetic process is well-controlled and the obtained Fe3O4@SiO2@Gd2O3:Yb,Er nanoparticles show relative monodispersity and exhibit tunable magnetic and up-conversion luminescence depending on the thickness of SiO2 mid-layers. The influences of SiO2 mid-buffer-layers on morphology, magnetism, and up-conversion luminescence are well addressed. The obtained bi-functional Fe3O4@SiO2@Gd2O3:Yb,Er nanoparticles may be potentially applicable in magnetic, fluorescent, and biological applications. The synthetic route may be employed for fabricating other multifunctional nanostructures.

A new turn-on luminescence energy transfer (LET) system has been designed for the detection of prostate specific antigen (PSA, a cancer marker) that utilizes Mn 2+ -enhanced long wavelength luminescence NaYF 4 : Yb, Er upconversion nanorods as the donor and gold nanorods as the acceptor. The Mn 2+ -doped NaYF 4 :Yb,Er upconversion luminescence nanorods with an emission peak located in the red region were synthesized. The presence of Mn 2+ markedly increased the luminescence intensity over that of the NaYF 4 :Yb, Er upconversion nanomaterials (excited by a 980 nm continuous wavelength laser). The surfaces of Mn 2+ -doped NaYF 4 :Yb, Er upconversion nanorods were modified with poly(acrylic acid). Antibodies against prostate specific antigen were bound to the surface of the carboxyl-functionalized upconversion nanorods, which acted as the energy donor in this LET system. Gold nanorods with an absorption band at ~666 nm were synthesized by the seed growth method, acted as the energy acceptor. The emission band of the upconversion nanorods overlapped well with the absorption band of the gold nanorods. The luminescence was quenched because of the electrostatic interactions that shortened the distance between the donor (negatively charged) and the accepter (positively charged). When the PSA antigen was added into the system, the energy acceptor and the energy donors were separated because the binding affinity between PSA and anti-PSA was greater than the electrostatic interactions, and thereby the luminescence was recovered. The linear range of detecting PSA was from 0.1172 to 18.75 ng/mL ( R =0.995), with a limit of detection for PSA as low as 0.1129 ng/mL. The method was successfully applied to the sensing of PSA in human serum samples.

Ethnopharmacological relevance Sambucus williamsii Hance (SWH), which belongs to the Caprifoliaceae family distributed in various regions of China, Korea and Japan, has been used as a folk medicine for treatment of bone and joint diseases in China for thousands of years. In previous studies, SWH was shown to possess anti-osteoporosis, healing fracture, anti-inflammatory and analgesic activities. Our previous studies showed that SWH extract effectively suppressed ovariectomy-induced increase in bone turnover and improved bone mineral density and bone biomechanical strength in rats as well as in mice. An 8- O -4′ norlignan, (7 R ,8 S )-1-(4-hydroxy-3-methoxyphenyl)-2-[4-(3-hydroxypropyl)-2-methoxyphenoxy]-1,3-propanediol (PPD) was previously isolated and identified as the bioactive ingredient in SWH. The present study aimed to characterize the bone protective effects as well as its mechanism of actions in osteoblasts. Materials and methods Bone protective actions of PPD on different cells were determined by proliferation assay, alkaline phosphatase (ALP) activity assay, calcium deposition as well as real-time reverse transcriptase-polymerase chain reaction (RT-PCR). In addition, estrogen receptor (ER) antagonist ICI182,780 and mitogen-activated protein kinase kinase (MEK) inhibitor U0126 blocking assays, competitive ER radioligand binding assay, ERE-dependent luciferase reporter assay and immunoblotting were used to determine if PPD activated ER and if the effects of PPD on osteoblastic functions were ER dependent. Results PPD exerted anabolic effects in osteoblasts and its effects were abolished by co-incubation with ICI182,780 or U0126. PPD induced mRNA expressions of Runx2, ALP, osteocalcin, and increased the ratio of osteoprotegerin/receptor activator of nuclear factor κB (OPG/RANKL). PPD failed to bind to either ERα or ERβ and did not activate ERE-luciferase activity via ER. PPD induced the phosphorylation of extracellular regulated kinases (ERK) and its effect was completely abolished by U0126. It also induced the phosphorylation of ERα at serine 118. Conclusion These data show that PPD is a bioactive compound in SWH that exerts oestrogen-like actions in osteoblast-like cells via ligand-independent, estrogen response element (ERE)-independent and mitogen-activated protein (MAP) Kinase-mediated rapid nongenomic ER signaling pathway.
