Diffusive gradients in thin films based on MOF-derived porous carbon binding gel for in-situ measurement of antibiotics in waters

Suyu Ren,Jing Tao,Feng Tan,Ying Cui,Xiaona Li,Jingwen Chen,Xin He,Yi Wang

Diffusive gradients in thin films (DGT) can achieve the time-weighted-average (TWA) concentrations of analytes in the period of deployment. Here we developed an approach based on DGT with a novel binding gel using porous carbon material (PCM) derived from metal-organic framework (MOF) for in-situ measurement of antibiotics in water samples. The diffusion coefficients of 20 antibiotics (nine sulfonamides, ten fluoroquinolones, and trimethoprim) in agarose diffusive gel were 1.41 × 10 −6  cm 2 /s to 4.75 × 10 −6  cm 2 /s at 25 °C. The capacity of PCM binding gel toward the antibiotics was ~100 μg per gel disc. The masses of the antibiotics accumulated by PCM-based DGT increased linearly with time, and they were almost independent of pH (4.2–8.4) and ionic strength (1–500 mM). The performance of the novel DGT was evaluated using freshwater and synthetic seawater spiked with the antibiotics in laboratory. The results showed good agreement with the theoretical predications except sulfacetamide. The detection limits achieved by the DGT with three devices together for deploying up to seven days were 1.0–18.0 ng/L. Field deployment in municipal wastewater treatment plant effluent and seawater showed that the TWA concentrations of antibiotics were 3.8–1342.5 ng/L and 5.6–43.3 ng/L, respectively, which were comparable to those measured by grab sampling. The proposed DGT was an efficient tool for the measurement of antibiotics in environmental waters.

Changes in activation energy and kinetics of heat-activated persulfate oxidation of phenol in response to changes in pH and temperature

Jie Ma,Haiyan Li,Liping Chi,Hongkun Chen,Changzhao Chen

Persulfate (peroxydisulfate, S 2 O 8 2− ) is the newest oxidant used for the in situ chemical oxidation (ISCO) remediation of soil and groundwater. The present study investigated impacts of solution pH, temperature, and persulfate concentration on the reaction rate constant ( k 1 ), activation energy ( E a ), and reaction order of the heat-activated persulfate process. Phenol was chosen as the model organic contaminant . As temperature increased from 30 °C to 70 °C, k 1 exhibited a significant increase from 0.003 h −1 ∼0.962 h −1 (pH 1.3–13.9) to 1.184 h −1 ∼9.91 h −1 (pH 1.3–13.9), which corroborated with the activation of persulfate using heat. As pH increased from 1.3 to 13.9, k 1 exhibited a 4.3-fold increase at 70 °C and a 320-fold increase at 30 °C, thereby suggesting that: 1) the phenol oxidation rate increased under alkaline conditions, and 2) the enhancement of reaction rate due to alkaline activation was more pronounced at a lower temperature. Increasing pH significantly reduced E a from 139.7 ± 1.3 kJ/mol at pH 1.3 to 52.0 ± 3.3 kJ/mol at pH 13.9. In contrast to changing pH, increasing persulfate concentration from 20 to 320 mM significantly increased k 1 but did not affect E a . Changes in E a suggest that persulfate oxidation of phenol experienced different reaction pathways or elementary reaction sequences as the pH changed from 1.3 to 13.9. In addition, the k 1 and E a data also suggest that a minimal pH threshold of ∼11 was required for the effective alkaline activation of persulfate.

Plant N-acylethanolamines play a crucial role in defense and its variation in response to elevated CO2 and temperature in tomato

Hu Zhangjian,Shi Junying,Feng Shuxian,Wu Xiaodan,Shao Shujun,Shi Kai

The ubiquitous lipid-derived molecules N-acylethanolamines (NAEs) have multiple immune functions in mammals, but their roles and mechanisms in plant defense response during changing environment remain largely unclear. Here, we found that exogenous NAE18:0 and NAE18:2 promoted defense against the necrotrophic pathogen Botrytis cinerea but suppressed defense to the hemi-biotrophic pathogen Pseudomonas syringae pv. tomato (Pst) DC3000 in tomato. The knocking-down and overexpression function analysis of the pathogen-responsive NAE synthetic gene PHOSPHOLIPASE Dγ (PLDγ) and hydrolytic gene FATTY ACID AMID HYDROLASE 1 (FAAH1) revealed that the NAE pathway is crucial for plant defense response. Using exogenous applications and SA-abolished NahG plants, we unveiled the antagonistic relationship between NAE and SA in plant defense response. Elevated CO2 and temperature significantly changed the NAE pathway in response to pathogens, while inhibition of the NAE pathway led to the alternation of environment-mediated defense variations against Pst DC3000 in tomato, indicating that NAE pathway is associated with plant defense variations in response to elevated CO2 and temperature. The results herein reveal a new function of NAE in plant defense, and its involvement in environment-mediated defense variation in tomato. These findings shed light on the NAE-based plant defense, which may have relevance to crop disease management in future changing climate.

Collagen Triple Helix Repeat Containing 1 Deficiency Protects Against Airway Remodeling and Inflammation in Asthma Models In Vivo and In Vitro

Feng Yong,Hu Jiapeng,Liu Fen,Shang Yunxiao

Asthma is a chronic inflammatory disease characterized by airway remodeling and lung inflammation. Collagen triple helix repeat containing 1 (CTHRC1), a glycoprotein, is involved in multiple pathological processes, including inflammation and fibrosis. However, the function of CTHRC1 in asthma remains unclear. In the present study, the mouse asthma model was successfully generated by sensitizing and challenging mice with ovalbumin (OVA). CTHRC1 expression at both RNA and protein levels was significantly upregulated in lung tissues of asthmatic mice. Asthmatic mice exhibited significant airway remodeling as evidenced by increased bronchial wall and smooth muscle cell layer thickness, goblet cell hyperplasia and collagen deposition, and epithelial-mesenchymal transition (EMT), but those characteristics were reversed by CTHRC1 silencing. The cell model with transforming growth factor-β1 (TGF-β1) induction in bronchial epithelial cells (BEAS-2B) was conducted to verify the effects of CTHRC1 on EMT, a classic mechanism that mediates airway remodeling. The results showed that TGF-β1 stimulation increased CTHRC1 expression, and CTHRC1 knockdown inhibited TGF-β1-induced EMT. OVA-treated mice also showed increased inflammatory cell infiltration and the production of OVA-specific immunoglobulin E (IgE), interleukin (IL)-4, IL-5, and IL-13, which were decreased by CTHRC1 downregulation. The effects of CTHRC1 on OVA-induced airway inflammation were further determined by treating BEAS-2B cells with IL-13, in which CTHRC1 knockdown reduced the IL-13-induced secretion of pro-inflammatory factors, including IL-4 and IL-5. In conclusion, these results indicate that CTHRC1 silencing attenuates asthmatic airway remodeling and inflammation in vivo and in vitro, suggesting that CTHRC1 may be a potential target for asthma treatment. Pre-mating nitenpyram exposure in male mice leads to depression-like behavior in offspring by affecting tryptophan metabolism in gut microbiota

Sen Yan,Wei Sun,Sinuo Tian,Zhiyuan Meng,Jinling Diao,Zhiqiang Zhou,Li Li,Wentao Zhu

Several studies have confirmed that the health status of the paternal affects the health of the offspring , however, it remains unknown whether paternal exposure to pesticides affect the offspring health. Here, we used untargeted metabolomics and 16S rRNA sequencing technology, combined with tail suspension test and RT-qPCR to explore the effects of paternal exposure to nitenpyram on the neurotoxicity of offspring. Our results found that the paternal exposure to nitenpyram led to the offspring's depressive-like behaviors, accompanied by the reduction of tryptophan content and the disorder of microbial abundance in the gut of the offspring. Further, we determined the expression of tryptophan metabolism-related genes tryptophanase ( tnaA ) and tryptophan hydroxylase 1 ( TpH1 ) in gut bacteria and colonic tissues. We found that tryptophan is metabolized to indoles rather than being absorbed into colonocytes, which coursed the reduce of tryptophan availability after nitenpyram exposure. In conclusion, our study deepens our understanding of the intergenerational toxic effects of pesticides.

Evaluation and Application of a Novel Diffusive Gradients in Thin-Films Technique for In Situ Monitoring of Glucocorticoids in Natural Waters

Liying Yan,Qiuyu Rong,Hao Zhang,Kevin C. Jones,Yanying Li,Jun Luo

The potential environmental risks of glucocorticoids (GCs) have attracted attention in the past few decades. In this study, a novel diffusive gradients in thin-films (DGT) device and analytical technique based on the second generation of polar enhanced phase (PEP-2), PEP-2-DGT, were developed for sampling and quantifying natural and synthetic GCs in aquatic systems. The capacity of PEP-2 gels for accumulating all target compounds was >600 μg per disc, sufficient for long-term passive sampling of selected GCs, even in wastewaters. Systematic tests were carried out to verify the application of DGT in natural waters and wastewaters. The performance of PEP-2-DGT devices was independent (CDGT/Csoln was in the acceptable range of 0.9–1.1) of a wide range of environmental conditions: ionic strength (0.001–0.5 mol L–1), dissolved organic matter (0–20 mg L–1), and pH (3.06–9.02). It was tested for various diffusive layer thicknesses (0.565–2.065 mm) and different deployment times (10–168 h). Diffusion coefficients (D) of selected GCs through an agarose-based diffusive gel were determined for the first time (3.80–4.85 × 10–6 cm–2 s –1 at 25 °C). Linear correlations between D and log Kow were established for three groups of target GCs (R2 = 0.96–0.99). This could enable prediction of D values for other GCs with similar structures in the future, which will help for rapid screening and emergency monitoring. Concentrations and distribution patterns of analytes obtained by PEP-2-DGT devices in five rivers after 7- and 14-day deployments were in accordance with those measured from grab samples, with total GC concentrations ranging from 7 to 27 ng L–1 at all sampling sites, confirming the reliability and robustness of the DGT devices for monitoring GCs in natural waters. The development of the new DGT technique will help improve understanding of the behavior and fate of these compounds in the aquatic environments.

Berberine inhibits tumour growth in vivo and in vitro through suppressing the lincROR-Wnt/β-catenin regulatory axis in colorectal cancer

Li Shi-ying,Shi Chuan-jian,Fu Wei-ming,Zhang Jin-fang

BackgroundBerberine, a non-prescription medicine clinically applied for diarrhoea and gastroenteritis. Recent studies have demonstrated that it possesses anti-tumour properties in colorectal cancer, but the exact molecular mechanism remains obscure.ObjectivesTo elucidate the underly molecular mechanisms of berberine in colorectal cancer from a perspective of epigenetics, and tried to explore the role of lincROR-Wnt/β-catenin molecular axis in the berberine induced the anti-tumour activity in colorectal cancer.MethodsThe effects of berberine on cell growth, cell cycle and apoptosis were examined in CRC cells. The in vivo effect of berberine on tumour growth was investigated using a xenograft mice model. Moreover, lincROR and Wnt/β-catenin signalling were detected by luciferase activity, qRT-PCR and western blotting assays.Key findingsBerberine suppressed cell growth in vitro via inducing cell cycle arrest and apoptosis in CRC cell, and inhibited tumourigenesis in vivo. LincROR was significantly down-regulated by berberine, inducing the inactivation of the canonical Wnt/β-catenin signalling, meanwhile, the overexpression of lincROR partially reversed the suppressive effects on tumour growth and Wnt/β-catenin signalling induced by berberine.ConclusionsBerberine inhibits tumour growth partially via regulating the lincROR-Wnt/β-catenin regulatory axis, which provides a strategy for the design of anti-tumour drugs for CRC patients after our advanced validation.

Identification of Cytochrome P450 Isozymes Involved in Enantioselective Metabolism of Fipronil in Fish Liver: In Vitro Metabolic Kinetics and Molecular Modeling

Siqi Wu,Huizhen Li,Jing You

Fipronil has been frequently detected in waterways worldwide at concentrations that threaten aquatic organisms, yet the metabolic behavior of fipronil enantiomers in aquatic organisms is largely unknown, which is of significance in enantioselective toxicity evaluation. We quantitatively identified the specific cytochrome P450 (CYP) isozymes involved in metabolizing fipronil enantiomers in tilapia by combining in vitro metabolic kinetic assays and molecular docking. Inhibition studies suggested that CYP1A enzyme was the main isoform catalyzing metabolism of fipronil and that CYP3A contributed in a limited way to the metabolism in fish liver S9. Both the dissipation rate constant and the maximum metabolic velocity of R -(−)-fipronil were greater than those of S -(+)-fipronil in tilapia liver S9, suggesting that tilapia selectively metabolized R -(−)-fipronil. The CYP1A1 isozyme exhibited the highest binding capacity to R -(−)-fipronil and S -(+)-fipronil (binding energy ‒9.39 and ‒9.17 kcal/mol, respectively), followed by CYP1A2 (‒7.30 and ‒6.94 kcal/mol, respectively) and CYP3A4 (‒7.16 and ‒6.91 kcal/mol, respectively). The results of in vitro metabolic assays and molecular docking were consistent, that is, CYP1A, specifically CYP1A1, exhibited a higher metabolic capacity to fipronil than CYP3A, and fish liver S9 selectively metabolized R -(−)-fipronil. The present study provides insight into the enantioselective metabolic behavior and toxicological implications of the in vitro metabolic kinetics of fipronil in fish. Development and application of diffusive gradients in thin-films for in situ sampling of the bitterest chemical – denatonium benzoate in waters

Sisi Liu,Shibao Chen,Xiaohao Li,Yubo Yue,Jinling Li,Paul N. Williams,Zhanyun Wang,Cailin Li,Yuanyuan Yang,Guang-Guo Ying,Chang-Er Chen

Denatonium benzoate (DB), a commonly used bitter agent in numerous products, has recently been recognized as a waterborne contaminant due to concern about its potential persistence, mobility and toxicity (PMT). However, its occurrence, levels and fate in global aquatic environments are largely unknown. In this study, a new sampling method, based on diffusive gradients in thin films (DGT) with mixed-mode cation exchange (MCX) as the binding agent, was developed for measuring DB in waters. MCX shows a rapid adsorption and high capacity for DB. DB is linearly accumulated by MCX-DGT. pH (6–8), ionic strength (0.01–0.5 M), or DOM (0–10 M) do not show any significant effect on the MCX-DGT performance, confirming its reliability. The DGT measurements in a wastewater treatment plant (WWTP) are comparable to those by paralleled grab sampling. The field results suggest DB is persistent in WWTPs and could be a potential domestic wastewater indicator. Therefore, MCX-DGT is a promising technique for understanding the environmental occurrence, levels and fate of DB. This is a first report of using DGT for DB monitoring and of DB occurrence in Chinese environments. Further exploration of DGT as a reliable passive monitoring tool for a wide range of PMT substances in different applications is warranted.

Accumulation of 2-tert-Butyl-1,4-benzoquinone in fried food rich in protein and its conversion in oil during deep-frying

Zeyi Wu,Qin Ye,Jinshuang Wang,Ningxiang Yu,Xiaohua Nie,Chaosheng Xia,Xianghe Meng

Chicken breast, a food rich in protein, was deep-fried in soybean oil (SBO) at 140, 175 and 190 °C for 20 continuous batches, and the profiles of acid value (AV), peroxide value (PV), 2-tert-Butyl-1,4-dihydrobenzoquinone (TBHQ), and 2-tert-Butyl-1,4-benzoquinone (TBBQ) in SBO and fried chicken breast were determined. The results suggested that frying at 140 °C produced increased conversion of TBHQ to TBBQ and oxidation of the frying oil than at 175 °C, which was close to that found at 190 °C. The highest TBBQ peak concentration of 2.89 ± 0.12 and 13.88 ± 0.27 mg/kg for fried chicken breast and frying oil corresponded to the 7th and 6th frying batch at 140 °C, respectively. The experimental results on determination of the AV, PV, moisture and the content of oxygen in frying oil suggested that more moisture and oxygen were retained in oil when frying at 140 °C than that at 175 °C, which may explain the over-conversion of TBHQ to TBBQ when frying at low temperature. The results on aeration frying and water injection frying experiments indicated that moisture and oxygen had a great impact on the oxidation of TBHQ in frying oil.

Fabrication of PLGA in situ forming implants and study on their correlation of in vitro release profiles with in vivo performances

Song Yang,Mengya Hu,Wenqing Liu,Nuohan Hou,Kaidi Yin,Chen Shen,Qing Shang

In this study, a novel PLGA in situ forming implants (ISFIs) were fabricated and methods for testing the in vitro release profiles were also developed. The correlations between in vitro release profiles and in vivo performances (in vitro–in vivo correlation, IVIVC) were also studied. PLGA with different molecular weights were selected as the polymeric matrix. Biocompatible N-methy1-2-pyrrolidone (NMP) or glyceryl triacetate (GTA) were used as the solvents with the ratios of NMP/GTA from 60/40 (vol/vol) to 20/80 (vol/vol). Eprinomectin (EPR) was chosen as the model therapeutic. In vitro release profiles of the EPR-loaded PLGA ISFIs were investigated using various methods (i.e. ‘tubule’ sample-and-separate and dialysis method). Sprague–Dawley rats were used to study the in vivo pharmacokinetics of EPR-loaded PLGA ISFIs. The release data obtained via ‘tubule’ sample-separate method had a good IVIVC (Level A, R2 > 0.99). These results showed that the ‘tubule’ sample-separate method was capable of discriminating the EPR-loaded ISFIs which were equivalent in formulation composition with manufacturing differences. Meanwhile, this method could be used to predict the in vivo performances of ISFIs in the investigated animal model.

Amygdalin inhibits TGFβ1-induced activation of hepatic stellate cells (HSCs) in vitro and CCl4-induced hepatic fibrosis in rats in vivo

Ruoyu Wang,Dong Zhang,Dan Tang,Kewei Sun,Jianping Peng,Wenfang Zhu,Sihan Yin,Yunan Wu

The activation of hepatic stellate cells (HSCs) has been considered one of the major events in hepatic fibrosis. Amygdalin has been used to treat cancers and alleviate pain; however, its role and mechanism in HSC activation and hepatic fibrosis remain unclear. In the present study, transforming growth factor-beta 1 (TGF-β1) stimulated the activation of HSCs, as indicated by significantly increased alpha-smooth muscle actin (α-SMA), desmin, collagen I, and tissue inhibitor of metalloproteinase-1 (TIMP-1) protein levels. Amygdalin treatment dramatically suppressed TGF-β1-induced HSC proliferation and activation. Moreover, amygdalin treatment also reduced the TGF-β1-induced secretion of cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin 6 (IL-6), platelet-derived growth factor (PDGF), and chemokine (C-C motif) ligand 2 (CCL2), as well as the phosphorylation of Smad2, Smad3, and p65. In the CCl 4 -stimulated liver fibrosis rat model, amygdalin treatment improved liver fibrosis and liver damage by reducing focal necrosis, collagen fiber accumulation, and the protein levels of α-SMA, desmin, collagen I, and TIMP-1 in hepatic tissue samples and reducing serum alanine transaminase (ALT) and aspartate transaminase (AST) levels. In conclusion, we demonstrated the suppressive effects of amygdalin in TGF-β1-induced HSC activation through modulating proliferation, fibrogenesis, and inflammation signaling in vitro and the antifibrotic effects of amygdalin in CCl 4 -stimulated hepatic fibrosis in rats in vivo .
