Oxidative stress plays a pivotal role in the onset and progression of arthritis, in which the abnormal elevation of hydroxyl radical (•OH) has been recognized as a key molecular hallmark of early inflammation. However, effective tools capable of simultaneously visualizing •OH dynamics and tracking therapeutic efficacy remain elusive. Herein, we developed a self-calibrated near-infrared II (NIR-II) ratiometric imaging probe responsive to •OH. The probe integrates downconversion nanoparticles (DCNP) as a stable internal reference (F1550nm,980Ex) and a •OH-triggered red-shiftable dye Hydro-830 (F930nm,808Ex), enabling ratiometric imaging through the signal ratio “F930nm,808Ex/F1550nm,980Ex.” The probe exhibits high sensitivity and specificity toward •OH in vitro and effectively captures the upregulation of oxidative stress at early inflammatory stages in vivo. In a longitudinal “treatment–imaging” study, the ratiometric signal gradually decreased following methotrexate (MTX) therapy, consistent with reductions in joint swelling, inflammatory cytokines, and histopathological lesions. This study establishes a self-calibrated NIR-II ratiometric imaging strategy for precise monitoring of oxidative stress, enabling early molecular diagnosis and dynamic therapeutic evaluation of arthritis, and offering a promising paradigm for imaging other ROS-related inflammatory disorders.

Hollow mesoporous manganese (HMM)-based nanocarriers, incorporating nanoparticle cavities and mesoporous channels, enhance the photostability and photobleaching resistance of indocyanine green (ICG). The addition of H2N-c(RGDfK)-OH (RGD) ensures precise targeting, making the cRGD&PEG2000-HMM@ICG (R&P-M@ICG) nanoprobe ideal for NIR-II fluorescence image-guided surgery and photothermal therapy (PTT). This approach offers accurate targeting, high therapeutic efficacy, and reduced side effects. The inclusion of manganese (Mn) in the nanocarriers further boosts the therapeutic effects by enabling chemodynamic therapy (CDT). Mn ions catalyze the conversion of hydrogen peroxide (H 2 O 2 ) into highly reactive hydroxyl radicals (•OH) within the tumor microenvironment. The synergy between Mn-driven CDT and PTT enhances tumor ablation, as the heat from PTT accelerates ROS production from CDT, leading to more effective tumor cell damage and apoptosis. The dual action of PTT and CDT overcomes the limitations of single-modal therapies. While PTT induces hyperthermia to kill tumor cells, Mn-assisted CDT amplifies the ROS generation, further enhancing treatment efficacy. This combined effect makes the R&P-M@ICG nanoprobe a powerful tool for cancer treatment, demonstrating superior anti-tumor efficacy compared to conventional therapies. In this study, we synthesized the R&P-M@ICG nanoprobe, highlighting its potential for precise surgical navigation, and for synergistic photothermal and chemodynamic therapy in cancer treatment.

The rising incidence of multidrug-resistant (MDR) bacterial infections in deep tissues, such as subcutaneous abscesses and bacterial pneumonia, has become an urgent public health concern. Both conventional antibiotic treatments and nanomaterial-based therapies are inadequate due to their limited catalytic efficiency, poor tissue penetration, and the enhanced resistance they provoke. We report the rational design and synthesis of nitrogen-doped rhodium single-atom catalysts (Rh SACs) that integrate second near-infrared (NIR-II) photothermal conversion with multienzyme mimetic activities. This nanozyme platform employs a synergistic ‘photothermal-catalytic’ therapy to effectively eliminate MDR pathogens and disrupt biofilms. Density functional theory (DFT) calculations indicate that atomically dispersed Rh sites lower the reaction barrier and optimize the adsorption of intermediates. The presence of rhodium enhances peroxidase and oxidase-mimetic activities under NIR-II irradiation. Transcriptional analysis reveals that Rh SACs modulate inflammation signaling and the immune microenvironment at the molecular level. In mice with methicillin-resistant Staphylococcus aureus -infected abscesses and lung infections, treatment with Rh SACs resulted in nearly complete bacterial clearance, biofilm disruption, and accelerated tissue repair. Additionally, macrophage polarization shifted significantly from M1 to M2. A systematic biosafety assessment demonstrated excellent hemocompatibility and minimal systemic toxicity. This work not only presents an efficient and resistance-free strategy for treating deep tissue infections but also provides crucial structural insights for structure-activity relationship studies in single-atom nanozymes. We strongly believe that nanosystems hold substantial potential for clinical translation in catalytic nanomedicine.

Nitric oxide (NO) plays key roles as a signaling molecule and an inflammatory mediator in the pathophysiology of diseases. Real-time, precise visualization of NO dynamics in deep tissues is essential for understanding disease mechanisms and evaluating therapeutic efficacy of drugs. However, existing NO probes, particularly those operating in the first near-infrared (NIR-I) window or single-wavelength “turn-on”, have limitations such as shallow penetration depth, tissue scattering, slow response and low sensitivity. To address these limitations, we used an electron cloud density enhancement strategy to develop NO-activated second near-infrared (NIR-II) window ratiometric photoacoustic (PA) probe (TCF-BA) by replacing the cationic heptamethine cyanine scaffold with an anionic counterpart. This anion-modulated strategy significantly increases electron cloud density, thereby improving the reactivity and sensitivity toward NO. The TCF-BA probe exhibits a significant ratiometric PA response upon NO activation, enabling high-contrast, deep-tissue imaging. Leveraging the high spatiotemporal resolution of PA imaging, the TCF-BA probe was used to quantitatively monitor NO fluctuations in a mouse model of liver injury. Overall, this study provides a reliable molecular imaging tool to investigate NO-related pathological processes and offers a promising strategy for the design of activatable NIR-II ratiometric probes.

Cr4+-activated phosphors are important candidate materials for NIR-II light sources, but providing a suitable lattice coordination environment for Cr4+ and achieving long wavelength broadband emission remains a challenge. In this work, a series of Cr4+-activated ABO2 (A = Li, Na; B = Al, Ga) phosphors were successfully prepared. Due to the presence of only tetrahedral coordination structures available for Cr4+ to occupy in the matrix crystal ABO2, the valence state and luminescence stability of Cr4+ are effectively guaranteed. Through the cation substitution design of A-site (Na → Li) and B-site (Ga → Al), the [BO4] tetrahedron is distorted and expanded, which degrades the symmetry of the Cr4+ coordination crystal field. Consequently, the central wavelength of the Cr4+ emission peak is tuned from 1280 to 1430 nm, and the fwhm is significantly extended from 257 to 355 nm. Thebroadband NIR-II light sources constructed with LiAlO2: 0.03Cr4+ and NaGaO2: 0.03Cr4+ phosphors verify their important potential applications in nondestructive testing and biological imaging.

Inspired by the effective interfacial interactions between natural creatures (e.g., cockleburs and viruses) and their bio-hosts, we developed a novel gold-manganese oxide nanoparticle (Au-MnOx) nanoenzyme with a spiky surface for NIR-II light-induced synergistic antibacterial therapy. Au-MnOx with favorable photothermal performance and peroxidase-like activity could achieve combined photothermal therapy (PTT)/chemodynamic therapy (CDT) under NIR-II irradiation with a low concentration. Of special note, the spiky surface endows Au-MnOx robust bacterial adhesion, in turn favoring the following synergistic PTT/CDT through effective interaction between Au-MnOx and bacteria. In vitro results display that our combined antibacterial nanoplatforms possess the broad-spectrum antibacterial ability against both Escherichia coli (E. coli, Gram-negative) and Staphylococcus aureus (S. aureus, Gram-positive). Moreover, satisfactory in vivo bactericidal efficacy and good cytocompatibility could be obtained in the rat wound model with the mixed bacterial infections. This study puts forward a facile one-pot method to establish the well-designed Au-MnOx nanozymes with a spiky surface for NIR-II light-enhanced bacterial elimination. The meteor hammer-like Au-MnOx holds great potential in construction of a novel antibacterial treatment system to settle the overuse of antibiotics.

Background Therapy with genetically modified mesenchymal stem cells (MSCs) has clinical translation promise. Optimizing the targeting migratory ability of MSCs relies on accurate imaging of the distribution and extravasation kinetics of MSCs, and the corresponding imaging results could be used to predict therapeutic outcomes and guide the optimization of the treatment program. Among the different imaging modalities, second near-infrared (NIR-II) optical-resolution photoacoustic microscopy (OR-PAM) has merits, including a fine resolution, a deep penetration, a high sensitivity, and a large signal-to-background ratio. It would be an ideal candidate for precise monitoring of MSCs, although it has not been tested for this purpose so far. Results Penetrating peptide-decorated conjugated polymer nanoparticles (TAT-CPNPs) with strong NIR-II absorbance were used to label chemokine-receptor genetically modified MSCs, which were subsequently evaluated under intravital NIR-II OR-PAM regarding their targeting migratory ability. Based on the upregulation of chemokine (C-X-C motif) ligand 10 in the inflamed ears of contact hypersensitivity mice, MSCs with overexpression of corresponding receptor, chemokine (C-X-C motif) receptor 3 (Cxcr3) were successfully generated (MSC Cxcr3 ). TAT-CPNPs labeling enabled NIR-II photoacoustic imaging to discern MSC Cxcr3 covered by 1.2 cm of chicken breast tissue. Longitudinal OR-PAM imaging revealed enhanced inflammation-targeting migration of MSC Cxcr3 over time attributed to Cxcr3 gene modification, which was further validated by histological analysis. Conclusions TAT-CPNPs-assisted NIR-II PA imaging is promising for monitoring distribution and extravasation kinetics of MSCs, which would greatly facilitate optimizing MSC-based therapy. Ultrathin two-dimensional (2D) nanosheets (NSs) have been widely explored as nanoagents for multimodal cancer therapies, such as photothermal therapy (PTT) combined with chemodynamic therapy (CDT). However, it is still a challenge to prepare 2D NSs with good performance in both CDT and NIR-II PTT. Here, we report the preparation of ultrathin 2D CuFe 2 S 3 NSs as an efficient nanoagent for synergistic CDT and NIR-II PTT to ablate cancer cells/tumors. Ultrathin 2D CuFe 2 S 3 NSs are first prepared by sulfurization of ultrathin CuFe-LDH NSs via a simple hydrothermal treatment. After modification with polyethylene glycol (PEG), the PEG-modified CuFe 2 S 3 NSs (CuFe 2 S 3 -PEG) show broadband NIR-II absorption and excellent photothermal conversion efficiency (~55.86%) at 1064 nm. Moreover, the overproduced glutathione in the tumor microenvironment can react with CuFe 2 S 3 -PEG NSs to release Fe 2+ and Cu + , thus activating Fenton reaction to efficiently produce hydroxyl radicals (·OH). Importantly, the photothermal effect of CuFe 2 S 3 -PEG could further synergistically improve the Fenton reaction by increasing the local temperature. In vitro and in vivo results reveal that the CuFe 2 S 3 -PEG has remarkable synergistic CDT/PTT antitumor activity. This study has proven that the metal chalcogenide NSs of CuFe 2 S 3 -PEG can be used as an efficient nanoagent for multimodal cancer therapies, which could also be promising in other applications like biosensors, drug delivery and antibacterial.

D2O plays important roles in a variety of fields (such as the nuclear industry and bioorganic analysis), and thus its isotopic purity (H2O contents) is highly concerned. Due to its highly similar physical properties to H2O and large excess amounts of H2O over D2O, it is challenging to distinguish D2O from H2O. On the basis of the characteristic NIR-II phosphorescence of singlet oxygen (1O2), and the fact that H2O is a more efficient quencher for 1O2 than D2O, here, we proposed to simply use the 1275 nm emission of 1O2 for the analysis of the isotopic purity of D2O. In normal cases (a xenon lamp for excitation), such steady-state emission is extremely weak for valid analytical applications, we thus employed laser excitation for intensification. To this goal, a series of photosensitizers were screened, and eventually polythiophene PT10 was selected with high singlet oxygen quantum yield (ΦΔ = 0.51), high H2O/D2O contrast, and excellent photostability. Upon excitation with a 445 nm laser, a limit of detection (LOD, 3σ) of 0.1% for H2O in D2O was achieved. The accuracy of the proposed method was verified by the analysis of the isotopic purity of several D2O samples (with randomly added H2O). More interestingly, the hygroscopicity of D2O was sensitively monitored with the proposed probe in a real-time manner; the results of which are important for strengthening the care of D2O storage and the importance of humidity control during related investigations. Besides D2O isotopic purity evaluation, this work also indicated the potential usefulness of the NIR-II emission of singlet oxygen in future analytical detection.

Malignant glioblastoma (GBM) is prone to relapse due to the inevitable tumor cells residue by surgery. During the tumor resection surgery in brain, addressing bleeding and superbug infections is also full of challenges. Currently, no method or material in clinical craniotomy can simultaneously solve these three problems. Herein, Chitosan composite nanofibers embedded with CuSe nanoparticles were prepared by green electrospinning method, in which the CuSe nanoparticles have strong absorption in the second near-infrared (NIR-II) window. Immediately after removing the tumor in craniotomy, nanofibers were electrospun and deposited directly onto the resection site with high precision (> 90%) to achieve rapid hemostasis (< 8 s). Moreover, evidenced by the deeper penetration depth of NIR-II light (1064 nm) both in the scalp and skull than NIR-I light (808 nm), photothermal and photodynamic therapy induced by NIR-II exhibits efficient superbug-killing rate (> 99%) and effectively induces cell apoptosis of residual tumor thereby to inhibit tumor recurrence. Only using the same material, a trilogy of intracranial hemostasis, killing superbug and residual cancer cells is simultaneously achieved. The short operation time reduces the risk of craniotomy. This electrospinning strategy could combine with craniotomy and minimally invasive surgery, which may provide novel perspectives in clinical operation besides craniotomy. Graphical abstract

Focused ultrasound (FUS)-induced blood–brain barrier (BBB) opening is crucial for enhancing glioblastoma (GBM) therapies. However, an in vivo imaging approach with a high spatial–temporal resolution to monitor the BBB opening process in situ and synchronously is still lacking. Herein, we report the use of indocyanine green (ICG)-dopped microbubbles (MBs-ICG) for visualizing the FUS-induced BBB opening and enhancing the photothermal therapy (PTT) against GBM. The MBs-ICG show bright fluorescence in the second near-infrared window (NIR-II), ultrasound contrast, and ultrasound-induced size transformation properties. By virtue of complementary contrast properties, MBs-ICG can be successfully applied for cerebral vascular imaging with NIR-II fluorescence resolution of ∼168.9 μm and ultrasound penetration depth of ∼7 mm. We further demonstrate that MBs-ICG can be combined with FUS for in situ and synchronous visualization of the BBB opening with a NIR-II fluorescence signal-to-background ratio of 6.2 ± 1.2. Finally, our data show that the MBs-ICG transform into lipid-ICG nanoparticles under FUS irradiation, which then rapidly penetrate the tumor tissues within 10 min and enhance PTT in orthotopic GBM-bearing mice. The multifunctional MBs-ICG approach provides a novel paradigm for monitoring BBB opening and enhancing GBM therapy.
