Colorimetric aptasensor based on peroxidase-mimetic metal–organic framework nanoparticles and magnetic carbon dots for visual detection of Staphylococcus aureus

Shanglin Li,Zhaomeng Sun,Ziyang He,Mei Liu

A colorimetric aptasensor based on the dual recognition of magnetic carbon dots (M-CDs) and copper-based metal–organic frameworks (Cu-MOFs) was constructed for the visual detection of Staphylococcus aureus (S. aureus). Aptamer-modified Cu-MOFs could specifically identify target bacteria and exhibited peroxidase-like activity by catalyzing the colorimetric reaction of the 3,3′,5,5′-tetramethylbenzidine–H2O2 (TMB–H2O2) system. M-CDs electrostatically attracted bacteria because of their strong positive electrical properties and offered better capturing performance compared with magnetic beads owing to their smaller size. Visual detection of the target was achieved using the color change of the reaction catalyzed by the resuspension of the sandwich-structured complex. The bacteria were sensitively detected with a limit of detection (LOD) of 1 CFU mL−1 using the peroxidase catalytic activity of Cu-MOFs. Non-target bacteria produced negative results, demonstrating the excellent selectivity of the proposed aptasensor. The strategy showed good recovery in real sample detection, showing great potential for visual monitoring of pathogenic bacteria, and could provide a reference for further development of novel sensing platforms for bacterial detection.

Light-Activatable Ionic and Photothermal Therapy by CuAu Nanorods for Treating Wounds and Keratitis Caused by Methicillin-Resistant Staphylococcus aureus

Lili Chen,Xiaojing Wu,Tiancheng Qi,Kai Feng,Cheng Pei

Bacterial infection may lead to organ damage and even death. Copper ions have attracted much attention in bacterial-infected wound treatment due to their ability to kill bacteria and promote neovascularization. However, the unsatisfied copper delivery for efficient bacteria removal and unexpected side effects due to copper accumulation in normal tissues limit its application. Here, we developed a copper–gold alloy nanorod (CuAu NR) as a photothermal agent to treat chronic wounds and keratitis with Methicillin-resistant Staphylococcus aureus (MRSA) infection. The CuAu NRs demonstrated a similar surface plasmon resonance effect as gold nanorods (Au NRs), a rod-like nanoparticle with unique plasmonic properties, and could be effectively heated up under near-infrared laser (808 nm) irradiation. The pH and temperature dual modalities-triggered copper release improved the healing effect with reduced biosafety concerns. In both MRSA-infected wounds in the mice model and MRSA-infected keratitis in the rabbit model, the CuAu NRs showed significant antibacterial activity, enhanced angiogenesis, and improved healing under laser irradiation. Together with excellent biocompatibility, our CuAu NRs can serve as a potential antibacterial material for clinical use.

A Bacterial Capturing, Neural Network-Like Carbon Nanotubes/Prussian Blue/Puerarin Nanocomposite for Microwave Treatment of Staphylococcus Aureus-Infected Osteomyelitis

Liguo Jin,Hanpeng Liu,Chaofeng Wang,Xiangmei Liu,Congyang Mao,Yu Zhang,Zhaoyang Li,Shengli Zhu,Hui Jiang,Zhenduo Cui,Yufeng Zheng,Shuilin Wu

Staphylococcus aureus ( S. aureus )-infected osteomyelitis is a deep tissue infection that cannot be effectively treated with antibiotics. Microwave (MW) thermal therapy (MTT) and MW dynamic therapy (MDT) based on MW-responsive materials are promising for the therapy of bacteria-infected osteomyelitis occurring in deep tissues that cannot be effectively treated with antibiotics. In this work, the MW-responsive system of carbon nanotubes (CNTs)/Prussian blue (PB)/puerarin (Pue) with stable network-like structures is constructed. The PB is grown in situ on the CNTs, and its introduction not only reduces the aggregation of the network-like structures of the CNTs, but the large specific surface area and mesoporous structure can also provide many active sites for the adsorption of oxygen and polar molecules. Pue is a natural anti-inflammatory material that reduces inflammation at the infection site. The composite of the CNTs and PB avoids the skin effect and thus can improve dielectric and reflection losses. The MW thermal response of CNTs/PB/Pue is mainly due to the occurrence of reflection loss, dielectric loss, multiple reflections and scattering, interface polarization, and dipole polarization. In addition, under MW irradiation, the CNTs/PB/Pue can produce reactive oxygen species (ROS), such as singlet oxygen ( 1 O 2 ), hydroxyl radical (·OH).

Natural polysaccharide hydrogel with bioadhesion characters to synergistically enhance berberine's antibacterial effect by regulating the PTS system of Staphylococcus aureus

Yaozhi Zhang,Zhijia Wang,ShuChang Yao,Xiaoyu Lin,Xiang Zhang,Xinru Tan,Liuyang Zhang,Ran Xu,Yihang Zhao,Chen Zhao,Fuhao Chu,Wenguang Jing,Xuemei Huang,Penglong Wang

The global spread of Staphylococcus aureus ( S. aureus ) not only causes significant economic losses but also poses a serious threat to public health. Consequently, there is an urgent need for multidimensional strategies to develop antimicrobial dressings to combat bacterial infections. In response, we have developed a plant polysaccharide antibacterial hydrogel formed through the self-assembly of edible Kudzu powder (KP) and the natural star molecule berberine (BBR). Rheological tests show that natural polysaccharide KP-BBR hydrogel (BBR@KP) exhibits excellent injectability and adhesion. And the degradation of the hydrogel exceeded 90 % within 3 days. The synergistic effect of these two ingredients enhances the antibacterial activity of BBR and can increase the MIC of BBR to 0.05 mM. Specifically, KP promotes the affinity of the Phosphoenolpyruvate Phosphotransferase System (PTS) of S. aureus , enabling KP, with its bioadhesive properties, to adhere to the bacterial surface and continuously release BBR. Subsequently, BBR effectively exerts its antibacterial effect by inhibiting the synthesis of histidine and isoleucine. Furthermore, the BBR@KP hydrogel exhibits negligible cytotoxicity and hemolytic toxicity, underscoring its favorable biosafety profile. This synergistic natural antibacterial hydrogel, formulated through a green and straightforward methodology, not only holds promise for broad clinical applications but also provides novel perspectives for the utilization and development of plant polysaccharides in the biomedical field.

Prussian blue-decorated indocyanine green-loaded mesoporous silica nanohybrid for synergistic photothermal-photodynamic-chemodynamic therapy against methicillin-resistant Staphylococcus aureus

Chenyang Qi,Guitao Tan,Haonan Hu,Yipin Zhang,Jie Chen,Qinqin Zhang,Jing Tu

Nanomaterial-based synergistic antibacterial agents are considered as promising tools to combat infections caused by antibiotic-resistant bacteria. Herein, multifunctional mesoporous silica nanoparticle (MSN)-based nanocomposites were fabricated for synergistic photothermal/photodynamic/chemodynamic therapy against methicillin-resistant Staphylococcus aureus (MRSA). MSN loaded with indocyanine green (ICG) as a core, while Prussian blue (PB) nanostructure was decorated on MSN surface via in situ growth method to form a core-shell nanohybrid (MSN-ICG@PB). Upon a near infrared (NIR) laser excitation, MSN-ICG@PB (200 μg mL −1 ) exhibited highly efficient singlet oxygen ( 1 O 2 ) generation and hyperthermia effect (48.7℃). In the presence of exogenous H 2 O 2 , PB with peroxidase-like activity promoted the generation of toxic hydroxyl radicals (•OH) to achieve chemodynamic therapy (CDT). PTT can greatly increase the permeability of bacterial lipid membrane , facilitating the generated 1 O 2 and •OH to kill bacteria more efficiently. Under NIR irradiation and exogenous H 2 O 2 , MSN-ICG@PB (200 μg mL −1 ) with good biocompatibility exhibited a synergistic antibacterial effect against MRSA with high bacterial killing efficiency (>98 %). Moreover, due to the synergistic bactericidal mechanism, MSN-ICG@PB with satisfactory biosafety makes it a promising antimicrobial agent to fight against MRSA.

A study on the antibacterial activity and antimicrobial resistance of pyridinium cationic pillar[5]arene against Staphylococcus aureus and Escherichia coli

Yang Hua,Zhao Dengqi,Wang Shuping,Yang Lijun,Huang Jianying,Zhang Zibin,Li Shijun

An increasing number of infections caused by multidrug-resistant (MDR) Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ) have severely affected human society. Thus, it is essential to develop an alternative type of antibacterial agents that has a different bacterial resistance mechanism from that of traditional antibiotics. After the synthesis and structural characterization of a cationic pillar[5]arene with pyridinium groups (PP5), the antibacterial and antibiofilm activities as well as its microbial resistance were systematically investigated. In-depth evaluation of biological studies revealed that PP5 was an active antibacterial agent, with surprising antibiofilm formation ability against E. coli and S. aureus. From the results of differential scanning calorimetry and transmission electron microscopy, it was concluded that the microbicidal activity of PP5 was due to the physical disruption of the pathogen’s membrane and the subsequent leakage of cytoplasmic components, which could greatly reduce the rapid generation of resistance. It was presented that the easily available PP5 has high activity to inhibit Gram-positive and Gram-negative bacteria and/or their biofilms with low cytotoxicity. This pillar[5]arene derivative can be used as a good candidate for controlling drug-resistant pathogenic bacterial infections and treating MDR bacteria.

Synergistic Inhibitory Effect and Mechanism of Action of Cinnamaldehyde‐Carvacrol Nanoemulsion on the Staphylococcus aureus and Escherichia coli Mixed Biofilms Formation

Siqi He,Yuxin Wang,Wenli Wang,Yanhua Li,Zhiyun Zhang

The formation of Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ) mixed biofilms challenges the treatment of bacterial infections. In this study, the synergistic effect of plant essential oils cinnamaldehyde (CA) and carvacrol (CV) on inhibiting S. aureus and E. coli mixed biofilms formation was investigated. The Q value of CA and CV combination on inhibiting mixed biofilms formation was 2.28, suggesting their synergistic effect. Furthermore, CA/CV nanoemulsion was prepared, which further enhanced the inhibitory effect. CA/CV nanoemulsion could significantly inhibit the mixed biofilms formation at 64 μg/mL by reduction of bacterial adhesion and motility, polysaccharide intercellular adhesin (PIA) synthesis, and the inhibition of LuxS/AI‐2 expression. In the S. aureus and E. coli infected implant model of mice, CA/CV NEs demonstrated prominent inhibitory effect on the formation of S. aureus and E. coli biofilms, and simultaneously reduced the bacterial burden. The study prepared Cinnamaldehyde‐Carvacrol nanoemulsions (CA/CV NEs) through high‐energy emulsification. In a mouse infection model, CA/CV NEs exhibited a synergistic inhibitory effect on the formation of Staphylococcus aureus–Escherichia coli mixed biofilms, reducing PIA, LuxS/AI‐2, adhesion, athletic ability, and extracellular protein.

Transcriptomic insights into species-specific responses of Staphylococcus aureus and Vibrio parahaemolyticus biofilms to acidic electrolyzed water on blueberries and oysters

Xin Xia,Xinjie Ji,Penglu Liu,Yi Xia,Nan Cheng

AEW 30 s kills high-load S. aureus & V. para without resistance in 10 generations. • S. aureus enriches carbon metabolism & CAMP resistance; V. para enriches QS & ABC. • AEW acts via ROS accumulation, not membrane rupture. • AEW disrupts two-component & QS of S. aureus / V. para to remove biofilm. • Blueberries require 1 min AEW; oysters need ≥3 min due to rough surfaces. Foodborne pathogens and their biofilms seriously endanger global fresh food safety, and conventional non-thermal disinfection is limited by poor biofilm removal capacity. However, the species-specific mechanisms against biofilm-embedded pathogens and the matrix-dependent efficacy on real food surfaces have not been systematically investigated for AEW. Herein, the disinfection efficiency and molecular mechanisms of residue-free acidic electrolyzed water (AEW) were explored against Staphylococcus aureus (Gram-positive) and Vibrio parahaemolyticus (Gram-negative). AEW achieved full bacterial inactivation within 30 s and showed no evidence of inducing stable inherited resistance over 10 successive generations. In practical food matrices, AEW eliminated pathogens on blueberries in 1 min, whereas over 3 min was needed for rough-shelled oysters, and mature biofilms, especially of S. aureus displayed stronger tolerance, which was further quantified via the Weibull kinetic model. Transcriptomic changes in quorum sensing, two-component, and metabolic pathways were consistent with increased reactive oxygen species (ROS) levels and biofilm disruption observed physiologically and ultrastructurally, suggesting that AEW may impair biofilm integrity and contribute to metabolic suppression in a species-dependent manner, without evident membrane lysis. With its fast disinfection and zero-residue features, AEW shows application potential for on-site decontamination of fruits and seafood in food industry. A hybridization-gated ZIF-8 ratiometric nanoprobe for hydrogel-based portable detection of Staphylococcus aureus via ATP-triggered DNA duplex signaling

Shulan Chen,Jiang Huang,Wenyue Chen,Hongwen Wu,Hongliang Tan

Hybridization gating reduces fluorophore ratio-control constraints. • Post-activation dye recruitment limits background from pre-integrated signal dyes. • Immunomagnetic enrichment provides target selectivity for ATP-based sensing. • Chitosan-modified hydrogel limits acid-induced background in portable readout. Rapid bacterial detection in dairy food safety monitoring requires portable platforms that combine analytical sensitivity with target selectivity. Here, we report a hybridization-gated zeolitic imidazolate framework-8 (ZIF-8) ratiometric nanoprobe, designated RSZ, for portable detection of Staphylococcus aureus ( S. aureus ) through adenosine triphosphate (ATP)-triggered DNA duplex signaling. The RSZ probe adopts a dual-zone architecture with Ru(bpy) 3 2+ and ssDNA1 embedded within the ZIF-8 particle interior and ssDNA2 adsorbed on the exterior surface. This architecture separates the stable reference channel from the formation of the responsive fluorescence channel, thereby reducing dependence on precise dual-fluorophore loading and limiting background associated with a pre-integrated responsive fluorophore. Upon ATP-induced framework disruption, ssDNA1 becomes accessible for hybridization with ssDNA2, and the resulting duplex recruits SYBR Green I from solution to generate the responsive fluorescence channel relative to the stable Ru(bpy) 3 2+ reference. To enable field-deployable detection, the nanoprobe is embedded in a chitosan-modified agarose hydrogel in which chitosan maintains a near-neutral microenvironment and limits acid-induced nonspecific activation. Fe 3 O 4 nanoparticles conjugated with porcine immunoglobulin G (Fe 3 O 4 -pIgG) are integrated upstream to selectively capture S. aureus via protein A-pIgG recognition, followed by smartphone red-green-blue analysis without specialized instrumentation. The integrated platform achieves a detection limit of 6 CFU/mL and recoveries of 98.8–101.7% in spiked milk. Therefore, this study demonstrates a hybridization-gated strategy for translating stimulus-induced ZIF-8 disruption into controlled ratiometric signal generation for portable bacterial sensing in food analysis. Ascorbate–ferrous iron complex loaded into sodium hyaluronate-based eye drops for the treatment of methicillin-resistant Staphylococcus aureus bacterial keratitis

LiLi Zhao,Hongbo Li,Jiajia Yuan,Zhen Wang,Dan Xu,Zhenbin Liu,Lishan Yao,Junjian Ran,Quanyu Dai,Liangbin Hu

Bacterial keratitis is one of the most common causes of visual impairment and blindness worldwide, and suffers the risk of drug-resistant infections due to the abuse of antibiotics. Despite its ability to counteract antibiotic resistance by inducing ferroptosis, the application of Fe 2+ is limited by oxidative instability and a solution pH often incompatible with ophthalmic use. Thus, this study designed and synthesized an ascorbate–ferrous iron complex (SA-Fe(II)) using ferrous lactate and sodium ascorbate, followed by comprehensive structural characterization. Results confirmed the formation of coordination bonds between Fe 2+ and sodium ascorbate. The molecular ion peak and characteristic decarboxylated fragment, along with broad and diffuse diffraction peaks in X-ray diffraction patterns, verified the formation of an amorphous metal chelate structure stabilized by dynamic coordination bonds. The SA-Fe(II) complex exhibited potent antibacterial activity, achieving more than 99.99 % killing of Staphylococcus aureus ( S. aureus ) and methicillin-resistant Staphylococcus aureus (MRSA) at 100 μM within 3 h. When incorporated into a sodium hyaluronate eye-drop matrix (SA-Fe(II)-SH), the formulation exhibited excellent ocular biocompatibility, as evidenced by hemolysis rates less than 5 %, more than 95 % viability of human corneal epithelial cells (HCE-T) corneal epithelial cells over 3 days, and no observable tissue inflammation in vivo. Importantly, in a mice model of MRSA keratitis, SA-Fe(II)-SH significantly reduced bacterial load, suppressed the production of inflammatory cytokines (IL-1β, IL-6, TNF-α), promoted corneal repair, and prevented systemic dissemination. This non-antibiotic strategy integrates antibacterial, anti-inflammatory, and pro-healing functions, offering a safe and effective therapeutic approach for the treatment of drug-resistant bacterial keratitis.

Antimicrobial activity and mechanism of tea pigments derived from the discarded tea plant branches and leaves against methicillin-resistant Staphylococcus aureus

Xueluan Liu,Xiaolin Feng,Chongxin Shi,Ying Wen,Xianchun Hu,Shuhong Lin,Yong Ye

Microbial contamination in food can cause foodborne illnesses, particularly methicillin-resistant Staphylococcus aureus (MRSA), which poses a serious threat to human health due to its ability to produce heat-resistant enterotoxins and express the broad-spectrum antibiotic resistance protein penicillin-binding protein 2a (PBP2a). In this study, tea pigments with potent anti- MRSA activity were prepared from discarded tea plant branches and leaves via in vitro enzymatic oxidation. Optimal process conditions obtained through response surface methodology (RSM) were: enzyme substrate ratio of 0.6 U/mg, reaction temperature of 43 °C, and reaction time of 22 h, yielding a production rate of 7.88%. Theaflavins (TFs), thearubigins (TRs), and theabrownins (TBs) isolated and purified via organic solvent extraction and macroporous resin D101 exhibited similar phenolic compound spectral characteristics, with average molecular weights (Mw) of 208, 368, and 16,064 Da, respectively. The minimum inhibitory concentration (MIC) of TFs, TRs, and TBs against MRSA were 156.25, 312.5, and 1250 μg/mL, respectively, demonstrating excellent anti- MRSA activity that was significantly stronger than their inhibitory activity against Escherichia coli . Furthermore, tea pigments exert antibacterial effects through mechanisms including cell wall disruption, alteration of cell membrane permeability, inhibition of biofilm formation, reduction of respiratory chain dehydrogenase activity, and increased levels of intracellular reactive oxygen species in bacteria. Additionally, molecular docking indicates that theaflavins effectively binds to PBP2a, thereby interfering with MRSA growth. Tea pigments as anti- MRSA natural products have great prospects in food industry.

A convenient electrochemical biosensor mediated by filtration-assisted pretreatment and multi-TEs amplification system for Staphylococcus aureus detection in complex matrices

Shiting Chen,Haojie Jia,Qinjie Wu,Panzhu Qin,Lin Huang,Chao Yan

Rapid detection of foodborne pathogens plays a more and more significant role in the field of public health. In this research, we developed a convenient electrochemical biosensor mediated by filtration-assisted pretreatment and multiple thermostatic enzymes (Multi-TEs) amplification system for the detection of Staphylococcus aureus ( S. aureus ) in complex matrices. Firstly, the designed filtration-assisted pretreatment could simultaneously collect and enrich target bacterium from various actual samples within 5 min. Then, the target bacterial genome template was reacted at 37 °C in Multi-TEs amplification system to obtain specific double stranded products. Finally, the specific double stranded products were fixed on the surface of the electrode and the electrochemical indicators (Methylene blue, MB) was combined for the quantitative one-step electrochemical analysis of S. aureus . This assay has great linear calibration curves with the concentration ranging from 10 1 CFU/mL to 10 6 CFU/mL. The limit of detections (LOD) was defined as 1.28 CFU/mL, which performance is superior to some commercial ELISA kits. Meanwhile, amazingly, it has excellent specificity for S. aureus against other foodborne pathogens and could be applied for S. aureus analysis in human serum samples with the great recoveries and associated relative standard deviations (RSDs). These features do make the ideal facile electrochemical analytical platform for on-site detection of S. aureus.
