Stimulus-responsive polymeric nanoparticles have great potential as nanocarriers. Here, triple-stimuli-responsive core-crosslinked (CCL) micelles were developed that were convenient for drug delivery by taking advantage of dual-redox and UV light synergistic triggers. Amphiphilic block copolymers methoxy poly(ethylene glycol)- b -poly(3-azido-2-hydroxy-propyl methacrylate- co - ο -nitrobenzyl methacrylate) [mPEG- b -P(GMA-N 3 - co -NBM)] were synthesized via atom transfer radical polymerization and ring-opening reaction of ethylene oxide; these block copolymers can spontaneously self-assemble into noncrosslinked (NCL) micelles. Afterward, CCL micelles were prepared using an alkyne-functionalized crosslinking agent. Furthermore, the CCL micelles showed an excellent Nile Red dye molecule encapsulation, light and dual-redox-responsive performance, and increasing release rate of the dye as the concentration of glutathione and light irradiation increased. The triple-stimuli-responsive CCL micelles showed excellent stability and controllable release compared to NCL micelles. Therefore, we have developed a new smart nanocarrier, and the triple-stimuli-responsive CCL micelles could also be a potential delivery system in cancer therapy. © 2019 Wiley Periodicals, Inc. J. Poly(lactic acid-co-lysine) (PLL) was applied as a modified material to improve the hydrophilicity, reactivity, and degradation rate of poly(L-lactic acid) (PLLA). In the drug delivery system, PLLA and PLLA/PLL were selected as the carrier materials of drug-loaded fibers, and bacitracin (BAC) as the sustained release drug. Uniaxial drug-loaded fibers (PLLA/BAC, PLLA/PLL/BAC) and coaxial drug-loaded fibers ((PLLA/PLL/BAC)-PLLA, (PLLA/BAC)-(PLLA/PLL)) were prepared using electrospinning. The chemical structures and thermal properties of the electrospun drug-loaded fibers were analyzed using Infrared Spectroscopy (IR) and Differential Scanning Colorimetry (DSC) separately and an Ultraviolet spectrophotometer (UV) was used to characterize the extracorporeal drug releasing behavior. The results indicated that the main interaction between the BAC and carrier materials such as PLLA and PLLA/PLL was physical force; the uniaxial drug-loaded fibers with uniform diameters showed a higher release rate compared with the coaxial drug-loaded fibers owing to the protection of shell materials in the coaxial fibers. The drug release rate of the drug-loaded fibers increased due to the addition of hydrophilic PLL; the drug release mechanism in both the uniaxial and coaxial drug delivery systems could be interpreted by a biexponential-biphase kinetics function. Two kinds of fibers with different drug release patterns could be fabricated by uniaxial and coaxial electrospinning. Drug in uniaxial fibers could be quickly released and was suitable for antibiotic treatment while the release of drug in coaxial fibers was a kind of controlled release which could be applied for a drug delivery system of long-term and small doses.

The Fe3O4@mSiO2 nanocarrier that consisted of a magnetic Fe3O4 nanoparticle core and a mesoporous silica (mSiO2) shell was synthesized. It shows a uniform sphere morphology about 65 nm in diameter. Considering the magnetic hyperthermia of Fe3O4 under an alternating magnetic field (AMF), a thermo-sensitive polymer, poly[(ethylene glycol)-co-(L-lactide)] (P(EO-co-LLA)), was used as “gatekeeper” coating outside Fe3O4@mSiO2 to regulate the drug release behavior. The design of the nanocarrier was expected to block off the pores at low temperature and to reopen them at high temperature reversibly. The obtained hybrid nanocomposites were capable of loading the anti-cancer drug doxorubicin (DOX) and controlled drug release behavior trigged by the hyperthermia of Fe3O4 under AMF. Besides, the nanocarriers also show pH-sensitive drug release based on the slight differences between the tumor (weakly acid) and the normal tissue (weakly alkaline). What's more, the chemotherapy of DOX combined with magnetic hyperthermia can improve the cytotoxicity obviously. On the basis of the high stability and excellent controlled release performance, the multifunctional nanocarriers exhibit potential applications in targeted-control drug release and hyperthermia for cancer treatment.

A unique redox responsive system for glutathione (GSH) triggered controlled release was prepared by modifying the mesoporous SBA-15 with l -cystine ester linked mPEG chains, in which the former was served as the nanoreservoir of guest molecules and the latter was applied as gatekeepers to block the mesopores. The disulfide bond in l -cystine ester can be effectively cleaved by redox reaction under glutathione of cancer cell level, and then resulting in the release of guests encapsulated in the pores. Fluorescein dye as the model drug was loaded into the mesoporous silica nanoparticles (MSNs) and released in PBS with 10 mM GSH and without GSH respectively to test the controlled release properties of the MSNs using UV–Vis spectra as the detector. The results showed that fluorescein dye in PBS with 10 mM GSH was released much more rapidly. Compared with such a controlled release system based on MCM-41, the SBA-15 based system showed a distinctive release behavior. The MSNs synthesized from SBA-15 can be used as an effective drug controlled release system for biomedical applications, especially for long-term drug delivery.

Hollow mesoporous silica spheres (HMSS) have a hierarchical mesoporous structure composed of a hollow cavity and mesoporous shell available for high capacity drug storage. The covalent attachment of ZnO quantum dots (QDs) to the HMSS outer surface as gatekeepers via disulfide-conjugated two amide linkages encapsulated the anticancer drugs doxorubicin (DOX) within the HMSS cavities and pores, and minimized premature release of the drug. The controlled release of the drug from the ZnO-gated HMSS delivery system was realized by the dissolution of ZnO QDs upon a decrease in pH and cleavage of the disulfide bonds, which indicates that the pH- and redox-responsive controlled release of the drugs could be synergically stimulated by tumor cells with weakly acidic environments and high-expressed glutathione. The constructed ZnO-gated HMSS delivery system has promising applications in site-specific drug release for tumor chemotherapy.

Bio-based hydrogel containing cyclodextrins (CDs) is of a promising polymer material that could display many advantages including wide availability, sustainability, biocompatibility and biodegradability , especially the inherent encapsulation ability with hydrophobic substance. To obtain these, the electrostatic and host-guest interactions were introduced and a hydrogel with three-dimensional double network structures was built. For preparing a spherical biopolymer cage, hydroxyethyl cellulose (HEC) and modified chitosan (HACC) were cross-linked by a one-pot reaction. The existence of HACC in this hydrogel provides a positive charge core to attract negative host molecule of sulfobutylether- β -cyclodextrin (SEB- β -CD). The loading amount of SEB- β -CD were determined by the method of weight increment and photometric titration, respectively, and an average content of active SEB- β -CD in our prepared hydrogel is more than 50%, much higher than the grafting of CD on biopolymers materials through chemical reaction. By the host-guest interaction, hydrophobic molecule of PP could adsorb rapidly in our prepared hydrogel and sustain-release in aqueous solution. Through ion-exchange interaction, different negative ions were studied for obtaining a control release of SEB- β -CD, which is to achieve the purpose of rapid release of hydrophobic guest molecule.

In this study, the application of banana-derived extracellular vesicles (EVs) as natural carriers for hydrophobic nutraceuticals was investigated for the first time. Curcumin was solubilized in an amorphous state within the hydrophobic domains of the EVs in banana juice. The bioaccessibility of the curcumin was considerably higher in the curcumin-loaded EVs (CEVs) than for pure curcumin crystals. The retention/release behavior of the curcumin in the CEVs was regulated by incorporating the curcumin-loaded banana juice into hydrogel beads, which were assembled from sodium alginate and banana pectin. The pectin and D-galacturonic acid contents of the banana juice were 0.87 % ± 0.11 % and 0.58 % ± 0.08 %, respectively. With the introduction of sodium alginate, the sphericity, hardness, chewiness, water holding capacity, surface smoothness and other physiochemical properties of the hydrogel beads were improved. Fourier Transform infrared analysis showed that hydrogen bonding played an important role in the formation of the hydrogel beads. The hydrogel beads had a higher swelling ratio under neutral conditions than under acidic conditions. Encapsulation of the curcumin-loaded banana juice within the hydrogel beads retarded curcumin release and enhanced the therapy potential of ulcerative colitis, which indicated that these delivery systems could be designed to regulate the release profile of the CEVs. Encapsulation of the curcumin also improved its resistance to heating, exposure and storage. In summary, we have shown that encapsulation of curcumin within plant-based EVs loaded into hydrogel beads can improve its functional performance.

The freeze-drying approach was used to create inclusion complexes utilizing alkyl gallates and β-cyclodextrin, namely dodecyl gallate, octyl gallate, butyl gallate, and ethyl gallate, which are exemplary examples of phenolic esters. The everted-rat-gut-sac model demonstrated that the inclusion complexes released alkyl gallates, which were subsequently hydrolyzed to generate free gallic acid, as evidenced by HPLC-UV analysis. Both gallic acid and short-chain alkyl gallates were capable of permeating the small intestinal membrane. The transport rate of gallic acid (or alkyl gallates) exhibited an initial rise followed by a drop when the carbon-chain lengths varied. The inclusion complex groups exhibited a superior sustained-release effect compared to the comparable alkyl gallates groups, thus possibly leading to higher bioavailability and stronger bioactivity. Moreover, altering the length of the carbon chain will allow for the effortless achievement of regulated release of phenolic compounds and short-chain phenolic esters from such β-cyclodextrin inclusion complexes.

A novel pH-sensitive charge reversal and self-fluorescent polymeric micelle is designed and synthesized successfully. The smart micelle is prepared based on MPEG-polyurethane multi-block copolymer, which is synthesized by polycondensation, with 1, 4-bis (hydroxyethyl) piperazine (HEP) and hydroxyl-sulfamethazine (Hydroxyl-SM) as pH-sensitive molecules and fluorescein isothiocyanate (FITC) as fluorescent segment. The resulting MPEG-polyurethane multi-block copolymer is examined by 1 H–NMR, UV-vis spectra, fluorescence spectra and an acid-base titration. Moreover, the diameter, morphology and cytotoxicity of obtained polymer micelle are measured by dynamic light scattering (DLS), transmission electron microscopy (TEM) and MTT assay. The results indicate that the micelle has a small diameter of less than 200 nm and can remain unchanged within two weeks. Zeta-potential measurement shows that the negative charge of micelle can switch into positive charge as the pH value decreasing from 9.0 to 3.0. Subsequently, the fluorescence intensity decreases significantly with the reducing of pH values. The MTT assay shows the low cytotoxicity and good biocompatibility of the MPEG-polyurethane polymeric micelles. Finally, doxorubicin (DOX) is loaded into micelles to detect in vitro release behavior. The drug loaded micelles show a faster release behavior at pH 5.0 than that at pH 7.4. Therefore, the pH-sensitive charge reversal and self-fluorescent micelle can be a potential smart carrier for delivery and controlled release of protein drug.

In the field of pharmaceutical research and development, the development of nano drug carriers based on intercalation structures has become one of the important development directions. Hydrotalcite (LDHs) and its derivatives have the advantages of low cost, simple synthesis, high drug loading efficiency, good biocompatibility, and easy degradation, and have received widespread attention in the field of biomedicine. Based on this, we synthesized 5-aminosalicylate (5-ASA) intercalated NO 3 based and CO 3 based ZnAl-LDHs drug delivery systems using ion-exchange and chemical co-precipitation methods, respectively. The crystalline structure, morphology, thermal stability, and host-guest interaction were characterized in detail by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), Ultraviolet visible spectroscopy (UV), Scanning electron microscopy (SEM), Thermogravimetric analysis (TGA) and X-ray photoelectron spectroscopic (XPS). The drug loading, drug release behavior and release kinetic of 5-ASA intercalated ZnAl-LDHs were studied. The characterization results indicated that the intercalation mode of the drug and the strength of the host-guest interaction are related to the interlayer anion type and synthesis process of ZnAl-LDHs. The amount of 5-ASA loaded into LDHs from an ion-exchange process was 28.6 and 13.8 wt% for NO 3 − and CO 3 2− based LDHs, respectively, whereas it was 6.9 and 10.4 wt% for co-precipitation process. The modeling kinetics results indicated that the release process of 5-ASA by ZnAl-LDHs intercalated with 5-ASA conforms to the parabolic diffusion model. These results indicated that the drug delivery system was controlled by diffusion of 5-ASA ions into aqueous solution through exchange with phosphate anions.

Reconstruction of bone defects has long been a major clinical challenge. Limited by the various shortcomings of conventional treatment like autologous bone grafting and inorganic substitutes, the development of novel bone repairing strategies is on top priority. Injectable biomimetic hydrogels that deliver stem cells and growth factors in a minimally invasive manner can effectively promote bone regeneration and thus represent a promising alternative. Therefore, in this study, we designed and constructed an injectable nanocomposite hydrogel co-loaded with Laponite (Lap) and vascular endothelial growth factor (VEGF) through a simplified and convenient scheme of physical co-mixing (G@Lap/VEGF). The introduced Lap not only optimized the injectability of GelMA by the electrostatic force between the nanoparticles, but also significantly delayed the release of VEGF-A. In addition, Lap promoted high expression of osteogenic biomarkers in mesenchymal stem cells (MSCs) and enhanced the matrix mineralization. Besides, VEGF-A exerted chemotactic effects recruiting endothelial progenitor cells (EPCs) and inducing neovascularization. Histological and micro-CT results demonstrated that the critical-sized calvarial bone defect lesions in the SD rats after treated with G@Lap/VEGF exhibited significant in vivo bone repairing. In conclusion, the injectable G@Lap/VEGF nanocomposite hydrogel constructed in our study is highly promising for clinical transformation and applications, providing a convenient and simplified scheme for clinical bone repairing, and contributing to the further development of the injectable biomimetic hydrogels.

A carrier possessing simple structure and composition, but with microwave-targeted-fluorescence multifunctional properties to precisely control the delivery of the drug was prepared. Herein, we have constructed the multifunctional Fe3O4@ZnAl2O4:Eu3+@mSiO2–APTES core–shell drug-carrier via direct precipitation method and sol–gel process with surfactant-assistance approach. This carrier is a monodisperse microsphere with an average particle size of 325 nm. Fe3O4 in the core has a high saturation magnetization and provides the Fe3O4@ZnAl2O4:Eu3+@mSiO2–APTES with good drug targeting properties. The ZnAl2O4:Eu3+ interlayer has the characteristic of fluorescent luminescence and can be used to monitor the transport of drugs in the body in real time. In addition, the ZnAl2O4:Eu3+ as a dielectric loss microwave absorbing material combines with the high magnetic loss Fe3O4 to form a composite material, which improved the microwave thermal response. Over 78.2% of VP16 molecules were released under microwave trigger. In addition, mesoporous silica nanoparticles in the outer layer improve the drug loading efficiency through organic modification. The results indicated that this multifunctional drug-carrier with simple structure and composition is a potential controlled drug delivery system in cancer therapy.
