Orientin inhibits cyclin E/cyclin A-CDK2 to induce growth arrest at senescence in human gastric cancer cells

Yang Yan,Yongfei Gu

Gastric cancer (GC) is a primary epithelial malignancy originating in the stomach. This research explored the influence and mechanism of Orientin on cellular senescence in GC. Orientin was predicted to regulate multiple cell cycle-related pathways, including cyclin-dependent protein kinase activity. Orientin docked with cyclin dependent kinase 2 (CDK2), cyclin A2 (CCNA2), cyclin E1 (CCNE1), and cyclin E2 (CCNE2) with binding energies of -9.1, -7.5, -7.2, and -8.3 kcal/mol, respectively. The IC 50 values of Orientin for AGS and HGC-27 cells were 24.33 μM and 39.28 μM, respectively. Orientin dose-dependently inhibited GC cell proliferation, promoted G0/G1 phase arrest, as well as downregulated CDK2, CCNA2, CCNE1, and CCNE2 expression. It enhanced senescence-associated β-galactosidase staining intensity, upregulated p16 and p21, and downregulated phosphorylated retinoblastoma (p-Rb). In vivo , Orientin also suppressed tumor progression, promoted p16 and p21 expression, and inhibited CCNE/CCNA-CDK2 signaling. Orientin exerts anti-cancer effects in GC through triggering cellular senescence and cell cycle arrest, likely via activating p16/p21-Rb signaling axis and inhibiting the CCNE/CCNA-CDK2 pathway. Efficient biomanufacturing of vitamin E succinate in non-aqueous media via synergistic computational evolution and immobilization of lipase

Zihan Zhang,Yu Zhang,Qingqing Liu,Wanqing Song,Yi Hu,Binbin Nian

A novel triple evolution strategy overcame the lipase activity-stability trade-off. • CM5 showed 2.3-fold higher activity and 4.8-fold solvent tolerance than WT. • MD simulations link CM5 stability to rigidity and stable hydration shell. • The VES synthesis strategy achieved the highest yield and reusability to date. • A complete chain from computational design to practical production was validated. Biocatalysts exhibit broad application prospects in industrial production. However, the inherent instability of natural enzymes in non-aqueous media severely limits their practical utilization. Using Candida rugosa lipase as a model, a synergistic evolution strategy integrating ancestral sequence reconstruction, free energy optimization and surface hydration shell modulation was developed in this study. The optimal mutant CM5 exhibited simultaneously enhanced stability and activity, showing a 4.78-fold increase in tolerance to 65% (v/v) dimethyl sulfoxide, a 1.67-fold extension in half-life at 50 ℃ and a 2.30-fold increase in activity compared to the wild type. Molecular dynamics simulations indicated that the enhanced stability of CM5 originates from improved structural rigidity and the stabilized hydration shell. Meanwhile, its increased activity is attributed to a reduced energy barrier for the lid conformational transition and an optimal enzyme substrate affinity that significantly boosts substrate turnover efficiency. Moreover, a volumetric activity of 142.71 U/mL in a 5 L bioreactor was obtained via optimization of the chassis cells, and the resulting culture was then used for the immobilization. The lipase CM5 immobilized on amino octadecyl bifunctional macroporous resin (CM5@AOMR) achieved a vitamin E succinate yield of > 99.9% and maintained an outstanding yield of 98.2% even after ten consecutive batch reactions. Overall, this study developed a comprehensive workflow from computational design and efficient fermentation to immobilization and practical applications of lipase, providing a promising new approach for developing next generation biocatalysts suited for extreme industrial environments. Highly stable thiourea-functionalized polyhedral oligomeric Silsesquioxane adsorbent for efficient and selective gold recovery from E-waste leachate

Qian Du,Qingshan Li,Hao Li,Jianming Pan

With the fast pace of technological advancement and the accelerating replacement cycle of electronic devices, the process of extracting gold from electronic waste ( E -waste) has emerged as a pivotal challenge for resource recycling and environmental sustainability. In this work, a series of thiourea-functionalized polyhedral oligomeric silsesquioxane (POSS)-based polymers (POSS@ATU-X) abundant in sulfur (S) and nitrogen (N) active sites was synthesized through a facile one-step functionalization strategy, and its performance and underlying mechanism for Au(III) adsorption were systematically investigated. POSS@ATU-4 delivered a rapid adsorption kinetic profile, with 90% of its maximum adsorption capacity attained within 3 h and equilibrium reached at 6 h. Isothermal adsorption tests revealed an ultra-high maximum adsorption capacity of 2643.64 mg g −1 at 318 K. Notably, POSS@ATU-4 exhibited outstanding selectivity for Au(III) in both multi-metal coexisting systems and actual printed circuit board (PCB) leachate, with its removal rate far exceeding those of other coexisting metal ions. After six consecutive adsorption-desorption cycles, the material retained 99% of its initial adsorption efficiency, demonstrating exceptional structural stability and reusability. Mechanistic studies elucidated that Au(III) removal was governed by the synergistic effects of electrostatic attraction and surface coordination interactions. This work provides a feasible and effective strategy for achieving highly selective and stable gold recovery from acidic E -waste streams, which holds great promise for practical industrial applications.

NAD+ capping of sibD transcripts in E. coli is mediated by its minimal promoter and enhanced by ppGpp

Liu Wuzhen,Hu Kefan,Nie Shiqi,Feng Zhang,Li Qiongfang,Zhang Hailei,Liang Shumin,Peng Jialin,Zhang Shoudong,Cai Zongwei,Xia Yiji

Recently, nicotinamide adenine dinucleotide (NAD+) and other nucleotide analogs have been identified as non-canonical RNA caps in both prokaryotes and eukaryotes. In Escherichia coli, NAD capping has been shown to be influenced by environmental conditions in a gene-specific manner, yet its regulatory mechanisms remain poorly understood. We previously reported that most transcripts produced by sibD are NAD-capped during the stationary phase. In this study, we found that the 35-bp minimal promoter of sibD is sufficient for its NAD capping. When this minimal promoter was applied to express genes not typically producing NAD-RNAs, their transcripts could also be NAD-capped. These findings strongly support that NAD capping in E. coli occurs during transcription initiation mediated by the promoter and RNA polymerase. Additionally, the bacterial alarmone ppGpp and the small protein DksA, both transcription initiation regulators, synergistically enhance transcription of both NAD-capped and uncapped RNAs from sibD and its homologous genes, sibC and sibE, in vitro. In contrast, the ppGpp⁰ mutant, deficient in ppGpp synthesis, showed a significant reduction in NAD-RNA production from these genes. Our findings elucidate the cis-elements and trans-acting factors mediating NAD capping during transcription initiation, offering novel insight into this regulatory process.

Degradation of the neonicotinoid insecticide imidacloprid by ozonation and the E-peroxone process: Mechanisms, transformation products, and toxicity

Yang Zhang,Lingwei Gao,Juhong Zhan,Erzhuo Zhao,Huijiao Wang,Chunrong Wang,Yujue Wang

In this study, degradation of the neonicotinoid insecticide imidacloprid (IMI) by ozonation and the E-peroxone process was investigated. Results show that IMI is an O 3 -refractory compound with second-order rate constants for the reaction with O 3 and • OH ( k O3,IMI and k •OH,IMI ) being 3.9 and 3.95 × 10 9 M −1 s −1 at pH 7, respectively. Due to its low O 3 reactivity, IMI was only abated by < 20 % in a selected groundwater during ozonation with typical ozone doses applied in water treatment (1–5 mg/L). In comparison, the E-peroxone process considerably enhanced IMI removal efficiency to > 80 % under similar reaction conditions. During both ozonation and the E-peroxone process, IMI was mainly abated by • OH oxidation. The reaction mechanism of IMI with • OH was investigated with quantum computations, which show that H-abstraction at H23 site was the most favorable pathway. Sixteen transformation products (TPs) generated from IMI degradation were identified using various chromatographic and mass-spectrometric techniques. The acute and chronic toxicities of IMI and its TPs to different aquatic organisms were evaluated with Microtox bioassay and the ECOSAR program, which indicate that some TPs (e.g., TP 2 –TP 5 and TP 7 –TP 9 ) are even more toxic than IMI to aquatic organisms (e.g., daphnia). The results of this study suggest that IMI along with its TPs should be sufficiently abated during water treatment, and the E-peroxone process provides an efficient option to upgrade conventional ozonation for enhanced IMI abatement.

Enhancing anti-E. coli O157:H7 activity of composite phage nanofiber film by D-phenylalanine for food packaging

Haiying Cui,Xiaojing Yang,Changzhu Li,Yong Ye,Xiaochen Chen,Lin Lin

Escherichia coli O157:H7 ( E. coli O157) is still one of the main pathogenic strains causing food poisoning. Here in, we established a strategy that can simultaneously remove free E. coli O157 and the biofilms it forms. Composite nanofiber (fP-NF) films were synthesized successfully by co-encapsulating E. coli O157 phages which isolated from domestic sewage and D -phenylalanine (D-Phe) into sodium alginate (SA)/polyethylene oxide (PEO) nanofibers by electrospinning. And dehydrated trehalose was added as a phage protectant. When SA:PEO is 3:9, the spinning solution has the optimum conductivity and viscosity, the average diameter of the obtained fP-NFs is 421.87 nm, and the mechanical strength and transparency of the fP-NF film are the best. The thermal stability of the fP-NF film was good, and the phage activity in the fiber was well preserved. The remaining population of phages in fP-NF membranes was still close to 10 6 PFU/mL after one week of placement, and the cumulative release efficiencies of phages at both room temperature (25 °C) and refrigerated storage temperature (4 °C) were above 70%. The antibacterial experiment showed that compared with the control group, the addition of D-Phe slightly enhanced the antibacterial activity of the nanofiber films against free E. coli O157, and the bactericidal rate within 8 h was 99.74%. On the other hand, D-Phe significantly enhanced the inhibitory activity of the nanofiber films against E. coli O157 biofilms, with the inhibition rate exceeding 99.99% within 72 h. The application results further confirmed that the addition of D-Phe could significantly enhance the inhibitory activity of nanofiber films on E. coli O157 proliferation and biofilm formation during short-term storage of beef and fruits and vegetables. It provides an important strategy and reference for the development of new active food packaging materials .

S/MWCNt/LLZO composite electrode with e−/S/Li+ conductive network for all-solid -state Lithium–Sulfur batteries

Fei Chen,Yiluo Zhang,Qing Hu,Shiyu Cao,Shangbin Song,Xinqi Lu,Qiang Shen

Traditional liquid lithium-sulfur batteries have two key problems: poor safety and shuttle effect . These two challenges can be overcome by using solid-state electrolytes. But poor ionic and electronic conductivity due to solid/solid contact between the electrode and the electrolyte for all-solid-state lithium-sulfur batteries (ASSLSBs). Here, we have synthesized Li 6.4 Ga 0.2 La 3 Zr 2 O 12 (LLZO) conductive ceramics with high ionic conductivity . LLZO is used as the lithium-ion conductor and multi-walled carbon nanotubes (MWCNt) are used as the electronic conductor to form S/MWCNt/LLZO by the thermal diffusion method, resulting in an e − /S/Li + conductive network. The prepared S/MWCNt/LLZO composite material, composite solid electrolyte, and lithium metal are assembled into ASSLSBs. After 50 cycles, the high capacity of 825 mAh·g −1 is maintained with 72.62% capacity retention at 0.2C and 45 °C. The higher capacity indicates the success of forming a conductive network that can improve the electrochemical performance of ASSLSBs. At 0.2C, the capacity of the S/MWCNt/LLZO composite cathode remains at 873 mAh·g −1 after 60 cycles when the operating temperature is increased to 60 °C, indicating good cycle performance.

Gold-Nanoparticle-Based Multiplexed Immunochromatographic Strip for Simultaneous Detection of Staphylococcal Enterotoxin A, B, C, D, and E

Wenbin Wang,Liqiang Liu,Liguang Xu,Hua Kuang,Jianping Zhu,Chuanlai Xu

Staphylococcal enterotoxins (SEs) in food samples can cause severe gastroenteritis even at low doses. Detection of SEs with a single assay would not only improve reliability and portability, but would also save time. In this paper, for the first time, a multiplexed gold-nanoparticle-based immunochromatographic (ICT) strip that allows the simultaneous detection of the five main SEs has been reported: SEA, SEB, SEC, SED, and SEE. Based on the sandwich immunoassay of each SE, a multiplexed ICT strip with five test lines is developed. Sensitive and specific paired monoclonal antibodies facilitate the detection, allowing for high sensitivity and good specificity. The portable biosensor platform can simultaneously detect SEA, SEB, SEC, SED, and SEE at concentrations as low as 2.5, 2.5, 2.5, 1, and 5 ng mL −1 , respectively. The entire test can be performed within 15 min and requires no sophisticated instruments except a portable strip reader. To evaluate the multiplexed strip, naturally SEA and SEC contaminated milk samples and pure milk samples spiked with the five SEs are analyzed.

The preservative potential of Amomum tsaoko essential oil against E. coil, its antibacterial property and mode of action

Na Guo,Yu-Ping Zang,Qi Cui,Qing-Yan Gai,Jiao Jiao,Wei Wang,Yuan-Gang Zu,Yu-Jie Fu

Amomum tsaoko is widely distributed in south-west China as a spice. In present study, the antibacterial activity of Amomum tsaoko essential oil (AEO) against foodborne pathogens was evaluated. The antibacterial activity was determined by measuring the zones of inhibition (ZOI), minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and the time-kill assay. Results showed the susceptibility of foodborne bacteria Escherichia coli ( E. coli ) was excellent with the lowest MIC and MBC values of 3.13 and 6.25 mg/mL, respectively. The probable antibacterial mechanism of AEO was the change in permeability and integrity of the cell membranes leading to leakage of nucleic acids and proteins. Detection of the kinetics of E. coli deactivation in situ showed that AEO has good preservative activities in foods. It is necessary to consider that AEO will become a promising antibacterial additive for food preservative.

Electrochemical detection of E. coli O157:H7 based on NaCl stress improving the bacterial electrocatalytic oxygen reduction performance

Fengyang Wang,Panpan Liu,Yin Chen,Mengjuan Li,Yunlong Liu,Yan Qi,Xiaohui Xiong,Yuanjian Liu

Firstly, through various analytical methods, the mechanism by which the electrocatalytic oxygen reduction capacity of E. coli O157:H7 was enhanced under 4% NaCl stress was revealed. The cyclic voltammetry (CV) results indicated that the oxygen reduction ability of E. coli O157: H7 was improved under NaCl stress. Microbiological analysis confirmed the intracellular ROS levels in E. coli O157: H7 under NaCl stress is related to the upregulation of SOD/CAT activity. Metabolomics identified key metabolic adaptations in E. coli O157:H7, including enhanced tricarboxylic acid (TCA) cycles and ubiquinone (Q) biosynthesis, which promoted effective respiratory electron transfer in the bacteria. Secondly, the BPE-ECL (Bipolar Electrochemiluminescence) sensor designed based on 4% NaCl stress showed 9.72 times increase in anode ECL intensity. The detection limit was extremely low at 10 CFU for E. coli O157:H7 in 1 mL sample volume. This research provides a new solution for the sensitive screening of electrochemically active bacteria.

Ultrasensitive strain sensor based on superhydrophobic microcracked conductive Ti3C2Tx MXene/paper for human-motion monitoring and E-skin

Yibing Bu,Taoyu Shen,Wenke Yang,Shiyin Yang,Ye Zhao,Hu Liu,Yanjun Zheng,Chuntai Liu,Changyu Shen

With the rapid development of wearable intelligent devices, low-cost wearable strain sensors with high sensitivity and low detection limit are urgently demanded. Meanwhile, sensing stability of sensor in wet or corrosive environments should also be considered in practical applications. Here, superhydrophobic microcracked conductive paper-based strain sensor was fabricated by coating conductive Ti 3 C 2 T x MXene on printing paper via dip-coating process and followed by depositing superhydrophobic candle soot layer on its surface. Owing to the ultrasensitive microcrack structure in the conductive coating layer induced by the mismatch of elastic modulus and thermal expansion coefficient between conductive coating layer and paper substrate during the drying process, the prepared paper-based strain sensor exhibited a high sensitivity (gauge factor, GF = 17.4) in the strain range of 0–0.6%, ultralow detection limit (0.1% strain) and good fatigue resistance over 1000 cycles towards bending deformation . Interestingly, it was also applicable for torsion deformation detection, showing excellent torsion angle dependent, repeatable and stable sensing performances. Meanwhile, it displayed brilliant waterproof, self-cleaning and corrosion-resistant properties due to the existence of micro/nano-structured and the low surface energy candle soot layer. As a result, the prepared paper-based strain sensor can effectively monitor a series of large-scale and small-scale human motions even under water environment , showing the great promising in practical harsh outdoor environments . Importantly, it also demonstrated good applicability for spatial strain distribution detection of skin upon body movement when assembled into electronic-skin (E-skin). This study will provide great guidance for the design of next generation wearable strain sensor.

Synergy between vitamin E and D-sorbitol in enhancing oxidation stability of highly crosslinked ultrahigh molecular weight polyethylene

Ri-Tong Lan,Yue Ren,Xin Wei,Li-Zhi Tang,Nouman Ali Shah,Lu Xu,Shi-Shu Huang,Rizwan M. Gul,Jia-Zhuang Xu,Zhong-Ming Li

Oxidative stability of radiation crosslinked ultrahigh molecular weight polyethylene (UHMWPE) artificial joints is significantly improved by vitamin E (VE), but there is a dilemma that VE hinders crosslinking and thus jeopardizes the wear of UHMWPE. In this effort, we proposed an efficient strategy to stabilize UHMWPE under limited antioxidant contents, where VE and D-sorbitol (DS) were used as the primary antioxidant and the secondary antioxidant respectively. For non-irradiated blends with fixed antioxidant contents, oxidative stability accessed by oxidation induction time (OIT) of VE/DS/UHMWPE blends was superior to that of VE/UHMWPE blends, while DS/UHMWPE blends showed no increase in OIT. The cooperation between DS and VE exhibited a synergistic effect on enhancing the oxidative stability of UHMWPE. Interestingly, the irradiated VE/DS/UHMWPE blends showed comparable OIT but a significantly higher crosslink density than the irradiated VE/UHMWPE blends. The crystallinity, melting point, and in vitro biocompatibility of the blends were not affected by VE and DS. The quantitative relationships of mechanical properties , oxidation stability, crystallinity and crosslink density were established to unveil the correlation of these key factors. The overall properties of VE/UHMWPE and VE/DS/UHMWPE blends were compared to elucidate the superiority of the antioxidant compounding strategy. These findings provide a paradigm to break the trade-off between oxidative stability, crosslink density and mechanical properties, which is constructive to develop UHMWPE bearings with upgraded performance for total joint replacements. Statement of significance VE-stabilized UHMWPE is the most commonly used material in total joint replacements at present. However, oxidation and wear resistance of VE/UHMWPE implants cannot be unified since VE reduces the efficiency of radiation crosslinking. It limits the use of VE. Herein, we proposed a compounding stabilization by the synergy between VE and DS. The antioxidation capability of VE was revived by DS, thus enhancing the oxidation stability of unirradiated UHMWPE. The irradiated VE/DS/UHMWPE exhibited similar oxidation stability but higher crosslink density than irradiated VE/UHMWPE, which is beneficial to combat wear of UHMWPE and to inhibit the occurrence of osteolysis . This synergistic antioxidation strategy endows the UHMWPE joint material with good overall performance, which is of clinical significance.
