Synthesis of functional ionic liquid modified magnetic chitosan nanoparticles for porcine pancreatic lipase immobilization

Hongbo Suo,Zhen Gao,Lili Xu,Chao Xu,Dinghua Yu,Xinran Xiang,He Huang,Yi Hu

We developed magnetic chitosan nanoparticles (CS‑Fe 3 O 4 ) with mean diameter of 15–20 nm. Subsequently, these inorganic-organic composite nanoparticles were modified using an imidazole-based functional ionic liquid (IL). The prepared support (IL‑CS‑Fe 3 O 4 ), which was used to immobilize porcine pancreatic lipase (PPL), was characterized using Fourier transform infrared (FTIR) spectroscopy, vibrating sample magnetometry (VSM), thermogravimetry (TG), transmission electron microscopy (TEM) and X-ray diffraction (XRD). Circular dichroism (CD) was used to analyze the secondary structure of immobilized PPL. The immobilized PPL (PPL‑IL‑CS‑Fe 3 O 4 ) exhibited 1.93-fold higher specific activity than PPL‑CS-Fe 3 O 4 when triacetin was used as the substrate, and showed 95 mg/g of lipase immobilization capacity and 382% of activity recovery. The residual activity of PPL‑IL‑CS‑Fe 3 O 4 was above 60% of the initial activity after incubation at 50 °C for 6 h, as was higher than that of PPL‑CS‑Fe 3 O 4 which showed 40% of the initial activity. In addition, PPL‑IL‑CS‑Fe 3 O 4 retained 84.6% of the initial activity after 10 cycles, whereas PPL‑CS‑Fe 3 O 4 retained only 75.5% activity. Furthermore, the kinetic parameters, apparent K m and V max of PPL‑IL‑CS‑Fe 3 O 4 were 2.51 mg/mL and 1.395 U/mg respectively, these results indicated that the immobilized PPL had better affinity towards the substrate, especially when the nanoparticles were modified by functional IL. Besides, the magnetic chitosan nanoparticles loaded with PPL were easily recovered. A novel, efficient, and practical method for enzyme immobilization was developed.

Affinity induced immobilization of adenylate cyclase from the crude cell lysate for ATP conversion

Qianqian Zhu,Wei Zhuang,Huanqing Niu,Lei Ge,Byron Villacorta Hernandez,Jinglan Wu,Kai Wang,Dong Liu,Yong Chen,Chenjie Zhu,Hanjie Ying

The development of an orientation immobilization technique via affinity between polyhistidine tags and metal ions aims at maintaining biocatalytic activity of the enzymes. In this work, to tackle the issue of the immobilization of adenylate cyclase (AC), a simple and effective approach of synthesizing iminodiacetic acid (IDA)-Ni 2+ particles was applied for simultaneously purifying and immobilizing his-tagged AC. We chose agarose particles as carriers, and then decorated them with IDA, leading to the formation of a coordination combination of Ni 2+ . The porous carriers with a large pore size of 50 nm and a specific surface area of 45.8 m 2 /g exhibited favorable enzymatic activity and loading capacity. The optimal pH of the immobilized enzyme increased from 8.0 to 9.0 and the optimal temperature increased from 30 °C to 35 °C, compared to the free AC. Moreover, the immobilized AC retained a residual activity of approximately 80% after storing it at 25 °C for 48 h, whereas only 40% of the activity was left in the free AC at the same conditions. Maximum yield of cyclic adenosine-3′, 5′- monophosphate (cAMP) reached up to the summit of the reaction. The immobilized AC by affinity adsorption will provide a promising route for the industrial production of cAMP.

Electrochemically enhanced antibody immobilization on polydopamine thin film for sensitive surface plasmon resonance immunoassay

Daqun Chen,Yihong Mei,Weihua Hu,Chang Ming Li

For sensitive immunoassay , it is essentially important to immobilize antibody on a surface with high density and full retention of their recognition activity. Bio-inspired polydopamine (PDA) thin film has been widely utilized as a reactive coating to immobilize antibody on various surfaces. We herein report that the antibody immobilization capacity of PDA thin film is electrochemically enhanced by applying an oxidative potential to convert the surface catechol group to reactive quinone group. Quantitative surface plasmon resonance (SPR) investigation unveils that upon proper electrochemical oxidization, the antibody loading capacity of PDA film is significantly improved (up to 27%) and is very close to the theoretically maximal capacity of a planar surface if concentrated antibody solution is used. Using prostate-specific antigen (PSA) as a model target, it is further demonstrated that the SPR immunoassay sensitivity is greatly enhanced due to the improved antibody immobilization. This work offers an efficient strategy to enhance the reactivity of PDA film towards nucleophiles, and may also facilitate its immunoassay application among others.

Immobilization of an antimicrobial peptide on silicon surface with stable activity by click chemistry

Jingcai He,Junjian Chen,Guansong Hu,Lin Wang,Jian Zheng,Jiezhao Zhan,Yuchen Zhu,Chunting Zhong,Xuetao Shi,Sa Liu,Yingjun Wang,Li Ren

Infections associated with biomedical implants and devices pose a serious clinical challenge in hospitals worldwide. Antimicrobial peptides (AMPs) have become a great prospect to inhibit this type of infection due to their broad-spectrum antimicrobial activity and low cytotoxicity. However, it is still a challenge to apply AMPs on the biomaterial surface as the activity of AMPs is sensitive to salt or enzyme. In the present study, we prepared a spacer molecule, poly[2-(methacryloyloxy)ethyl]dimethyl-(3-sulfopropyl)ammonium hydroxide (polySBMA), on a model silicon surface via surface-initiated atom transfer radical polymerization (SI-ATRP). We then modified the antimicrobial peptide HHC36 (KRWWKWWRR) with L-propargylglycine (PraAMP) to improve its salt-tolerant activity and integrated PraAMP onto the spacer molecule using click chemistry. We employed X-ray photoelectron spectroscopy (XPS), contact angle goniometry, and atomic force microscopy (AFM) to confirm the success of the immobilization process. We also characterized the antimicrobial activity and stability of the surface with an antimicrobial assay. The results reveal that the modified surface exhibits good antimicrobial activity to inhibit 98.26% of E. coli, 83.72% of S. aureus, and 81.59% of P. aeruginosa. Furthermore, as compared to the control group without the polySBMA spacer, the modified surface improved its resistance to enzymolysis. An in vitro CCK-8 assay also illustrated that this surface showed negligible cytotoxicity to mouse bone mesenchymal stem cells.

A facile strategy for enzyme immobilization with highly stable hierarchically porous metal–organic frameworks

Xiao Liu,Wei Qi,Yuefei Wang,Rongxin Su,Zhimin He

Metal–organic frameworks (MOFs) have drawn extensive research interest as candidates for enzyme immobilization owing to their tunable porosity, high surface area, and excellent chemical/thermal stability. Herein, we report a facile and universal strategy for enzyme immobilization using highly stable hierarchically porous metal–organic frameworks (HP-MOFs). The HP-MOFs were stable over a wide pH range (pH = 2–11 for HP-DUT-5) and met the catalysis conditions of most enzymes. The as-prepared hierarchical micro/mesoporous MOFs with mesoporous defects showed a superior adsorption capacity towards enzymes. The maximum adsorption capacity of HP-DUT-5 for glucose oxidase (GOx) and uricase was 208 mg g−1 and 225 mg g−1, respectively. Furthermore, we constructed two multi-enzyme biosensors for glucose and uric acid (UA) by immobilizing GOx and uricase with horseradish peroxidase (HRP) on HP-DUT-5, respectively. These sensors were efficiently applied in the colorimetric detection of glucose and UA and showed good sensitivity, selectivity, and recyclability.

DNA directed immobilization enzyme on polyamidoamine tethered magnetic composites with high reusability and stability

Jiayi Song,Ping Su,Ye Yang,Ting Wang,Yi Yang

The development of new strategies for stabilizing or improving the activities of enzymes has attracted considerable interest because of the wide range of potential applications and high cost of the native enzymes. In this study, a novel trypsin immobilization procedure in which the enzyme was immobilized using polyamidoamine (PAMAM) dendrimer modified magnetic nanoparticles as carriers through DNA-directed immobilization was developed for the first time. The optimal DNA base pairs and the optimal generation of PAMAM on the enzymatic activity were 24 bases and G3.0PAMAM, respectively. The trypsin binding capacity of the immobilized trypsin was 74.6 mg g−1, and the Km and Vmax values were 1.23 mM and 468.35 μmol min−1 mg protein−1, respectively. The immobilized trypsin reactor exhibited excellent reusability and stability properties without significant loss in enzymatic activity. Notably, the high level of enzymatic activity remained at more than 63% after 82 cycles, with only a slight decrease (above 88%) after 14 weeks of continuous use at one-week intervals. The high reversibility and reproducibility of this trypsin dynamic immobilization strategy were also investigated. The significantly improved digestion performance of the immobilized trypsin composites was further demonstrated by digesting cytochrome C, myoglobin and glycated hemoglobin, with sequence coverages of 78%, 99% and 88%, respectively, which were higher than those obtained for the free enzyme. This system therefore shows great potential in high throughput enzymatic assays and proteome analysis.

Controlled synthesis, immobilization and chiral recognition of carboxylic acid functionalized cellulose tris(3,5-dimethylphenylcarbamate)

Miaomiao Han,Xiaosa Jin,Han Yang,Xiao Liu,Yadong Liu,Shengxiang Ji

Traditional cellulose-based chiral stationary phases (CSPs) are prepared by physically coating cellulose derivatives onto substrates and only compatible with a very limited range of solvents as the mobile phase. Therefore, chemical immobilization of cellulose derivatives onto silica gel has been efficiently applied to improve the solvent compatibility of cellulose-based CSPs. Here we developed a novel approach to homogeneously modifying cellulose tris(3,5-dimethylphenylcarbamate) (CDMPC). A series of carboxylic acid functionalized CDMPCs (CC-Xs) with controlled amounts and randomly distributed carboxylic acid groups was synthesized. CC-Xs were effectively immobilized onto amine-modified silica gel to afford immobilized CSPs. Compared to coated-type CSPs, immobilized CSPs significantly improved the solvent compatibility while maintaining similar chiral recognition abilities, but the introduction of excessive functional groups led to a deteriorated performance for columns. Moreover, a commercial drug, atenolol, was also sufficiently separated on the immobilized CSPs.

Immobilization of bovine carbonic anhydrase on glycidoxypropyl-functionalized nanostructured mesoporous silicas for carbonation reaction

Xiaoyao Fei,Shaoyun Chen,Chunjie Huang,Dai Liu,Yongchun Zhang

Bovine carbonic anhydrase (BCA) has been immobilized on glycidoxypropyl-functionalized SBA-15 (GFS) for biomimetic carbonation reaction. GFS was characterized by N 2 adsorption–desorption, Fourier transform infrared analysis, 13 C and 29 Si CP MAS NMR spectroscopy. The immobilization time and material dose were optimized. The influences of pH value and temperature on the activity of free and immobilized BCA, storage stability and reusability of immobilized BCA were investigated using a para-nitrophenyl acetate (p-NPA) assay. The kinetic parameters K m and K cat / K m for free and immobilized BCA were found to be 2.4   mM and 896.4   M −1   s −1 , and 3.1   mM and 757.4   M −1   s −1 , respectively. It was observed that the immobilized BCA can retain around 91% of its initial activity up to 30 days at 4   °C, which showed higher storage stability than free BCA. Reusability studies suggested that immobilized BCA could keep high activity after 20 cycles of carbonation reaction. BCA immobilized on GFS (BCA-GFS) were used for hydration of CO 2 . The CO 2 sequestration capacity in terms of conversion CO 2 to calcium carbonate was quantified by organic elemental analysis. The amount of CaCO 3 precipitated over GFS-BCA was nearly the same as that precipitated over free BCA, 227   mg of CaCO 3 /mg of BCA-GFS as compared to CO 2 sequestration capacity of 241   mg of CaCO 3 /mg of BCA.

Alloyed Trimetallic Nanocomposite as an Efficient and Recyclable Solid Matrix for Ideonella sakaiensis Lipase Immobilization

Frank Peprah Addai,Jiacong Wu,Feng Lin,Xinnan Ma,Juan Han,Yuelin Liu,Yang Zhou,Yun Wang

In this work, a trimetallic (Ni/Co/Zn) organic framework (tMOF), synthesized by a solvothermal method, was calcinated at 400 and 600 °C and the final products were used as a support for lipase immobilization. The material annealed at 400 °C (Ni-Co-Zn@400) had an improved surface area (66.01 m2/g) and pore volume (0.194 cm3/g), which showed the highest enzyme loading capacity (301 mg/g) with a specific activity of 0.196 U/mg, and could protect the enzyme against thermal denaturation at 65 °C. The optimal pH and temperature for the lipase were 8.0 and 45 °C but could tolerate pH levels 7.0–8.0 and temperatures 40–60 °C. Moreover, the immobilized enzyme (Ni-Co-Zn@Lipase, Ni-Co-Zn@400@Lipase, or Ni-Co-Zn@600@Lipase) could be recovered and reused for over seven cycles maintaining 80, 90, and 11% of its original activity and maintained a residual activity >90% after 40 storage days. The remarkable thermostability and storage stability of the immobilized lipase suggest that the rigid structure of the support acted as a protective shield against denaturation, while the improved pH tolerance toward the alkaline range indicates a shift in the ionization state attributed to unequal partitioning of hydroxyl and hydrogen ions within the microenvironment of the active site, suggesting that acidic residues may have been involved in forming an enzyme–support bond. The high enzyme loading capacity, specific activity, encouraging stability, and high recoverability of the tMOF@Lipase indicate that a multimetallic MOF could be a better platform for efficient enzyme immobilization.

Wei Li,Shengxian Chen,Yongqi Lu,Jiangjie Yu,Shiguang Zhao

The co-immobilized laccase-mediator system (LMS) has broad development prospects in the fields of biocatalysis, biochemistry, and environmental remediation. In this study, laccase and mediator molecules were co-immobilized in functionally modified calcium alginate microspheres to improve their catalytic activity and stability and to systematically characterize the physicochemical properties and evaluate the catalytic performance of the co-immobilized laccase-mediator system. The co-immobilized LMS was used to catalyze the synthesis of theaflavins (TFs) from tea polyphenols, improve the catalytic efficiency, realize the recycling of the system, and explore the safety of the catalytic products. The results showed that the catalytic efficiency of the co-immobilized LMS was significantly improved and the relative activity remained above 50% after 10 cycles. The in vitro safety of the catalytic product was verified by Human hepatocellular carcinoma (HepG2) cell-related indexes. The results show that the co-immobilized laccase mediator system has great development potential in the industrial application of theaflavins.

Effective uranium extraction from seawater through immobilization of conjugated microporous polymers on collagen fiber membrane

Xiaoxia Ye,Juan Liu,Xueying Chen,Dongjun Chen,Zhen Qian,Jie Chen,Chunxiang Lin

Extracting uranium from seawater has become an important method to supplement the nuclear industry. Herein, we introduce a simple route to load conjugated microporous polymers onto collagen fiber membranes, resulting in a novel membrane adsorbent material (CMPA-F/CFM) for uranium capture. Its Langmuir adsorption capacity reaches 304.9 mg·g −1 (at 318 K), with fast adsorption kinetic of 60 min, and excellent adsorption selectivity with K d  = 4.69 × 10 4 mL·g −1 under the temperature of 298 K, initial concentration of 50 mg·L -1 , and pH = 7, thus allowing a satisfactory adsorption performance in a 7-day real ocean experiment with a uranium adsorption capacity of 1.25 mg·g −1 . Furthermore, in continuous dynamic adsorption experiments, CMPA-F/CFM could treat up to 19.0 L·g −1 of low-enriched uranium (LEU) solution (1 mg·L -1 ), with a uranium removal rate of up to 87.5 % (10 L of seawater). This study demonstrates broad potential application of membrane adsorption in extraction of low-enriched uranium.

Effect of PFDS on the immobilization of Cs+ by metakaolin-based geopolymers in complex environments

Shengjian Zhao,Hualong Yang,Xuehui Liu,Siqi Ma,Peigang He,Zhenlin Sun,Dechang Jia,Paolo Colombo,Yu Zhou

Metakaolin-based geopolymers are very promising materials for improving the safety of low and intermediate level radioactive waste disposal, with respect to ordinary Portland cement , due to their excellent immobilization performance for Cs + and superior chemical stability. However, their application is limited by the fact that the leaching behavior of Cs + is susceptible to the presence of other ions in the environment. Here, we propose a way to modify a geopolymer using perfluorodecyltriethoxysilane (PDFS), successfully reducing the leaching rate of Cs + in the presence of multiple competitive cations due to blocking the diffusion of water. The leachability index of the modified samples in deionized water and highly concentrated saline water reached 11.0 and 8.0, respectively. The reaction mechanism between PDFS and geopolymers was systematically investigated by characterizing the microstructure and chemical bonding of the material. This work provides a facile and successful approach to improve the immobilization of Cs ions by geopolymers in real complex environments, and it could be extended to further improve the reliability of geopolymers used in a range of applications.
