Separation and determination of peptide metabolite of Bacillus licheniformis in a microbial fuel cell by high-speed capillary micellar electrokinetic chromatography

Wei Wang,Ruiguang Bai,Xiaoyu Cai,Ping Lin,Lihong Ma

A method using high-speed capillary micellar electrokinetic chromatography and a microbial fuel cell was applied to determine the metabolite of the peptides released by Bacillus licheniformis . Two peptides, l -carnosine and l -alanyl- l -glutamine were used as the substrate to feed Bacillus licheniformis in a microbial fuel cell. The metabolism process of the bacterium was monitored by analyzing the voltage outputs of the microbial fuel cell. A home-made spontaneous injection device was applied to perform high-speed capillary micellar electrokinetic chromatography. Under the optimized conditions, tryptophan, glycine, valine, tyrosine and the two peptides could be rapidly separated within 2.5 min with micellar electrokinetic chromatography mode. Then the method was applied to analyze the solutions sampled from the microbial fuel cell. After 92 h running, valine, as the metabolite, was successfully detected with concentration 3.90 × 10 −5 M. The results demonstrated that Bacillus licheniformis could convert l -carnosine and l -alanyl- l -glutamine into valine. The method employed in this work was proved to have great potential in analysis of metabolites, such as amino acids, for microorganisms.

Ultrasensitive Electrochemical Biosensor for Detection of DNA from Bacillus subtilis by Coupling Target-Induced Strand Displacement and Nicking Endonuclease Signal Amplification

Yuhua Hu,Xueqin Xu,Qionghua Liu,Ling Wang,Zhenyu Lin,Guonan Chen

A simple, ultrasensitive, and specific electrochemical biosensor was designed to determine the given DNA sequence of Bacillus subtilis by coupling target-induced strand displacement and nicking endonuclease signal amplification. The target DNA (TD, the DNA sequence from the hypervarient region of 16S rDNA of Bacillus subtilis) could be detected by the differential pulse voltammetry (DPV) in a range from 0.1 fM to 20 fM with the detection limit down to 0.08 fM at the 3sblank level. This electrochemical biosensor exhibits high distinction ability to single-base mismatch, double-bases mismatch, and noncomplementary DNA sequence, which may be expected to detect single-base mismatch and single nucleotide polymorphisms (SNPs). Moreover, the applicability of the designed biosensor for detecting the given DNA sequence from Bacillus subtilis was investigated. The result obtained by electrochemical method is approximately consistent with that by a real-time quantitative polymerase chain reaction detecting system (QPCR) with SYBR Green.

Control of post-harvest soft rot (Rhizopus stolonifer) on sweet cherry using volatile organic compounds produced by Bacillus velezensis Q-84

Wang Jian-Yu,Mao Min-Xin,Xu Chun-Yan,Zou Man,Bao Wen-Ting,Zhou Yi-Ning,Hu Shun-Qing,Jiao Ke-Xin,Lu Qing-Kun,Zhao Peng,Zhang Kun-Peng,Chen Yi-Lun,Zhang Qian

The soft rot of sweet cherries caused by Rhizopus stolonifer ( R. stolonifer ) results in significant economic losses during storage and transportation. This study demonstrated that volatile organaic compounds (VOCs) derived from the fermentation broth of Bacillus velezensis ( B. velezensis ) Q-84 significantly inhibited the growth of R. stolonifer by 54.43%. Scanning electron microscope (SEM) micrographs of VOCs-treated fungi revealed antifungal activity, evidenced by the breakage and deformation of the mycelium. Through headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC/MS) analysis, ten VOCs were identified in the fermentation broth of B. velezensis . Notably, 5-methyl-2-heptanone, 2,5-dimethylpyrazine, 2-decanone, and benzaldehyde exhibited significant antifungal activity in vitro . Furthermore, in vivo assays demonstrated the efficacy of both individual and combined VOCs in managing soft rot diseases in cherries. These treatments also resulted in increased levels of total polyphenolic content (TPC) and total flavonoid content (TFC), as well as a significant enhancement in the activities of defense-related enzymes such as peroxidase (POD), polyphenol oxidase (PPO), phenylalanine ammonia-lyase (PAL), and chitinase (CHI). Our results further our understanding of B. velezensis antifungal metabolites on the management of R. stolonifer and may help in finding less toxic compounds to control soft rot on sweet cherries.

Structural characterization of the exopolysaccharide produced by Bacillus amyloliquefaciens JM033 and evaluation of its ability to regulate immunity and intestinal flora

Hongxuan Li,Yubo Zhang,Yaping Zheng,Xuejian Li,Zimu Li,Chaoxin Man,Yu Zhang,Yujun Jiang

The probiotic strain Bacillus amyloliquefaciens JM033 ( B. amyloliquefaciens JM033), isolated from the traditional Chinese fermented food Sufu (also known as Fu-ru or fermented bean curd), is distinguished by its high production of exopolysaccharides (EPS). The EPS (BAP-1) produced by this strain was purified and its structure analyzed. BAP-1 is a novel hybrid fructan with a molecular weight of 17.6 kDa. It is composed of →6)- β -D-Fru f -(2 → and →1,6)- β -D-Fru f -(2→, which form the backbone, with a branched chain of β -D-Fru f -(2 → attached at the 1-position of residue B. In vivo studies on mice indicated that BAP-1 improves immunity in immunosuppressed mice by enhancing humoral immunity ( P  < 0.01), monocyte-macrophage phagocytosis ( P  < 0.01), and NK cell killing activity ( P  < 0.05). Additionally, BAP-1 was found to improve the composition of the intestinal microbiota and stimulate the production of short-chain fatty acids. Notably, BAP-1 exhibited a significant effect on the proliferation of Akkermansia . Therefore, BAP-1 shows promise as a prebiotic and may contribute to the development of new immunomodulatory agents.

The physicochemical properties of Cassava Starch/Carboxymethyl cellulose sodium edible film incorporated of Bacillus and its application in salmon fillet packaging

Xiqian Tan,Anqi Sun,Fangchao Cui,Qiuying Li,Dangfeng Wang,Xuepeng Li,Jianrong Li

Edible film is now a trend in the food packaging industry. In this study, edible films were prepared by adding two Bacillus spp. ( Bacillus amyloliquefaciens Y11 and Bacillus velezensis Y12) to a cassava starch and carboxymethyl cellulose sodium matrix. The structural, physicochemical, and biological characteristics of the film were analyzed, and its application in salmon preservation was explored. The film had a dense structure and no pores, indicating that its polymeric components were compatible with each other. The addition of Bacillus spp. increased the antioxidant activity of the film and its ability to eliminate hydroxyl radicals (84.57% and 91.86%, respectively). The film also showed good antibacterial activity against several pathogens and underwent complete degradation in natural soil within 12 days. The film significantly reduced the total coliform count of salmon and extended its shelf life by 3 days, demonstrating its value as a food-packaging material.

Dual-purpose Bacillus subtilis fermentation: enhanced nattokinase production via oxygen-enriched fed-batch cultivation and natto starter preparation from harvested biomass

Zheng Jiawen,Sun Yaping,Liao Yunyu,Qin Peng,Che Rongzhen,Zhao Jing-yi,Xiao Zijun

Nattokinase (NK) is one of the most important functional components in natto, but its content is low. In this study, the fermentation conditions using Bacillus subtilis JZ08-02 for high-yield NK production were investigated, and the residual bacterial pellets were used to prepare a natto starter. Batch fermentation of NK was conducted using a 5 L fermenter, and soybean milk and glucose were used as the substrates. When the stirring speed was increased from 450 to 650 rpm with air supply at 1.0 vvm, NK was increased from 4859 ± 142 to 12,294 ± 226 IU/mL. When pure oxygen was supplied, 15,013 ± 550 IU/mL of NK was obtained. When fed-batch fermentation was conducted, the titer was further elevated to 18,014 ± 112 IU/mL, which was increased by about 76% compared with the previous result. The experimental findings revealed that aeration control and nutrient feeding regimens exerted pronounced effects on NK productivity during submerged fermentation. The crude enzyme supernatant was obtained by centrifugation and the precipitate was collected. With optimized protectant, the bacterial pellets were freeze-dried with 90.1% cell survival rate. Using economical and edible feedstocks, this study achieved a significant enhancement in NK fermentation yield via oxygen-enriched fed-batch cultivation. At the same time, a natto starter was prepared as a by-product using the residual cell waste.

Optimized whole cell biocatalyst from acetoin to 2,3-butanediol through coexpression of acetoin reductase with NADH regeneration systems in engineered Bacillus subtilis

Niyomukiza Samuel,Teng Bao,Xian Zhang,Taowei Yang,Meijuan Xu,Xin Li,Irene Komera,Tuyishime Philibert,Zhiming Rao

BACKGROUND 2,3-Butanediol (2,3-BD) has a wide range of applications in chiral molecular synthesis, biofuel additives, and in food flavor additive manufacturing. Fermentation is a favorable method for 2,3-BD production. However, it requires much time and produces several NADH related byproducts which compete with 2,3-BD production. Bacillus subtilis has an excellent ability for 2,3-BD production by biocatalysis. However, its production is limited by low intracellular NADH and the reversible property of acetoin reductase (AR/2,3-BDH). The whole cell biocatalyst process with two different NADH regeneration systems was designed for efficient production of 2,3-BD in B. subtilis 168. RESULTS Formate dehydrogenase and glucose dehydrogenase for NADH regeneration were successfully co-expressed with acetoin reductase in B. subtilis 168. After optimization of biocatalyst bioconversion conditions, B. subtilis 168/pMA5- bdhA-Hpa II- fdh yielded 74.5 g L −1 of 2, 3-BD with 9.3 g L −1 h −1 productivity by fed batch and 115.4 g of 2,3-BD was achieved using same batch bacterium by three repeated batch bioconversions. On the other hand, 63.7 g L −1 of 2, 3-BD was produced with 7.92 g L −1 h −1 productivity by B. subtilis 168/pMA5- bdhA-Hpa II- gdh . To our knowledge, the volume productivity obtained here is the highest ever reported for biocatalysis. CONCLUSION A higher productivity of 2,3-BD from acetoin was achieved by whole cell biocatalysis with NADH regeneration systems in B. subtilis 168. This approach can be applied for NADH related bio-based chemicals production to improve titer, yield and productivity. © 2017 Society of Chemical Industry

XOD inhibition peptides obtained via fermentation of Salmon by Bacillus amyloliquefaciens and evaluation of their anti-hyperuricemia mechanism in HK-2 cells

Xiqian Tan,Zhiqi Xing,Bolin Zhou,Lijun You,Fangchao Cui,Xuepeng Li,Jianrong Li

Hyperuricemia (HUA) is a metabolic disorder that can trigger gout and cause renal dysfunction. In this study, microbial fermentation with Bacillus amyloliquefaciens Y11 was employed to ferment salmon dark meat. Subsequently, novel peptides with high xanthine oxidase (XOD) inhibitory activity were derived from the fermented samples, purified, and identified. The hypouricemic effect of these peptides was evaluated using an in vitro HK-2 cell HUA model. Five fractions (G1–G5) were isolated via ultrafiltration and Sephadex G15 gel chromatography, and the fractions with higher XOD inhibition rates (G1 and G5) were combined to prepare a mixed fraction. Subsequently, 4122 peptides were identified from the mixed fraction using liquid chromatography with tandem mass spectrometry. Based on predictive bioactivity, toxicity, allergenicity, and solubility scores, five peptides and seven corresponding truncated peptides were selected for synthesis. Their XOD inhibitory activity was assessed, and molecular docking analysis was performed. Ultimately, GEADFMDYGR and its truncated peptide GEADF, which showed high XOD inhibitory activity in vitro (inhibition rates of 90.72 ± 2.85% and 94.06 ± 1.30%, respectively), were screened out. In the HK-2 HUA model, both peptides significantly reduced intracellular uric acid levels. Specifically, 0.5 mg/mL GEADFMDYGR and GEADF decreased the uric acid levels in these cells by 39.70 ± 8.75 and 27.43 ± 3.31 μmol/L, respectively. Further analyses revealed that GEADFMDYGR and GEADF are reversible mixed-type XOD inhibitors. These findings provide new insights and experimental evidence guiding the development of novel therapeutic peptides for HUA.

A dual strategy of Bacillus subtilis SL-44 and Klebsiella oxytoca RS-5 promotes rapeseed growth: direct stimulation and rhizosphere microbiome remodeling

Chen Ji,Wang Yazhuo,Fan Zhongdi,Wang Conghu,Zhang Zihan,Liu Yuting,He Yanhui,Wu Zhansheng

Introduction Plant growth-promoting rhizobacteria (PGPR) colonize the rhizosphere and enhance plant growth, stress tolerance, and soil properties. To better harness these benefits, this study investigates the synergistic interaction between two PGPR strains— Bacillus subtilis SL-44 and Klebsiella oxytoca RS-5—and evaluates their combined effects on rapeseed growth through integrated direct and indirect mechanisms. Methods The plant growth-promoting traits (e.g., phosphate solubilization, siderophore and indole-3-acetic acid (IAA) production) and biofilm-forming capacity of individual strains and their consortium were characterized in vitro. Subsequently, pot experiments were conducted to evaluate their effects on rapeseed growth parameters and rhizosphere soil quality. Finally, the rhizosphere microbial communities were analyzed via high-throughput sequencing on the Illumina NextSeq platform. Results The co-culture consortium (SR) demonstrated synergistic enhancements in key plant growth-promoting traits and biofilm stability in vitro. Pot experiments confirmed that SR treatment most effectively promoted rapeseed biomass accumulation, improved physiological status, and elevated antioxidant capacity. It also optimally enhanced rhizosphere soil nutrient availability, enzyme activities, and selectively reshaped the microbial community by enriching beneficial taxa and increasing ecological network complexity. Ultimately, plant growth promotion was achieved through an integrated mechanism combining direct stimulation and microbiome-mediated soil improvement. Conclusion In summary, this study demonstrates that functional complementarity between PGPR strains drives synergistic growth promotion through combined direct stimulation and rhizosphere microbiome engineering, offering a framework for developing effective microbial consortia for sustainable agriculture. Heterologous expression of two novel deacetylases from Bacillus altitudinis JYY-02 and their applications in the enzymatic modification of chitin and xylan

Gengxian Zhou,Guicai Du,Hongzheng Tai,Lijiao Wang,Chao Wang,Ronggui Li,Qunqun Guo

Deacetylation can convert chitin into chitosan with superior properties and improve xylan's hydrolytic efficiency, broadening their applications in biomedicine, agriculture, and bio-based materials. In this study, two novel deacetylase genes encoding polysaccharide deacetylase (PdaA) and chitin oligosaccharide deacetylase (COD), were screened from Bacillus altitudinis JYY-02 and heterologously expressed in Escherichia coli Rosetta (DE3). The recombinant deacetylases were subsequently purified using Ni–NTA affinity chromatography. Comprehensive enzymatic characterization revealed that PdaA exhibited optimal activity at pH 5.0 and 30 °C, while COD performed best at pH 6.5 and 30 °C. Both deacetylases demonstrated exceptional thermal stability, retaining over 90% of their activity after incubation at 50 °C for 1 h. Kinetic studies showed that PdaA and COD displayed efficient substrate binding capabilities, with K m of 58.58 μg/mL and 72.05 μg/mL. Functional analyses revealed the two deacetylases could effectively catalyze the deacetylation of colloidal chitin with a deacetylation degree of 59.81-69.02%. Moreover, when combined with xylanases, they significantly improved xylan hydrolysis, increasing the yield of reducing sugar by 35-40% compared to conventional acid hydrolysis methods. The superior catalytic properties and stability of these deacetylases position them as promising biocatalysts for the enzymatic modification of commercial chitin and xylan.

Vancomycin-modified poly-l-lysine magnetic separation combined with multiplex polymerase chain reaction assay for efficient detection of Bacillus cereus in milk

Qianying Li,Guoyang Xie,Yutong Wang,Zoraida P. Aguilar,Hengyi Xu

ABSTRACT In this study, a new vancomycin (Van)-modified poly- l -lysine (PLL) magnetic bead (MB) technique was developed for isolation of gram-positive bacteria. The method combines magnetic separation with a multiplex PCR (mPCR) assay and gel electrophoresis for easy and rapid detection of Bacillus cereus . Vancomycin was used as a molecular ligand between the MB and the d -alanyl- d -alanine moieties on the cell wall surface of B. cereus . The PLL served as a flexible molecular tether between the MB and Van that reduced steric hindrance maintaining the biological activity of Van. The MB-PLL-Van capture nanoprobes exhibited excellent capture and isolation efficiency for B. cereus in spiked milk matrix samples without interference from the complex food matrix. The subsequent mPCR assay showed high specificity for the 4 target genes in B. cereus , the entFM , cesB , cer , and 16S rRNA genes, that were used to achieve efficient genotyping and detection. Under optimum conditions, the limit of detection reached 10 3 cfu/mL, with a dynamic range of detection at 10 3 to 10 7 cfu/mL in pure culture. Application of the MB-PLL-Van mediated mPCR assay for B. cereus in milk matrix samples achieved results similar to those of the pure culture. In addition, with a 6-h pre-enrichment of B. cereus that was spiked in milk matrix samples, the limit of detection reached 10 1 cfu/mL. The MB-PLL-Van mediated mPCR assay developed in this study could be used as a universal technology platform for the efficient enrichment and genotyping of gram-positive bacteria.

Identification and molecular mechanism of novel antioxidant peptides from okara fermentation broth with Bacillus amyloliquefaciens YP2: In silico and in vitro analysis

Xiaodi Wang,Qiaolei Zhu,Yuhan Yan,Yao Yu,Wanting Yue,Yunping Zhu,Zhou Chen

This study aimed to identify novel antioxidant peptides extracted from okara fermentation broth, and to investigate the molecular mechanisms using in silico and in vitro approaches. The results demonstrated that three novel peptides—CKLLL, FMAV and SVCC—were identified, among which CKLLL exhibited the strongest DPPH radical scavenging activity (IC 50  = 30.21 ± 4.27 μM). Simulated gastrointestinal digestion showed CKLLL had stable DPPH scavenging activity and improved ABTS and hydroxyl radical scavenging capacities. Furthermore, all three antioxidant peptides effectively protected HepG2 cells against H 2 O 2 -induced damage, with CKLLL>SVCC>FMAV ranking highest to lowest in terms of protective effect. Peptide and Keap1 protein docking results indicate that the -CDOCKER energy of CKLLL (89.13 kcal/mol) is higher than that of FMAV and SVCC (55.35 and 48.01 kcal/mol). These findings suggest that these three peptides, especially CKLLL, have the potential to serve as antioxidant peptides, and provide a more reliable screening method for developing natural plant-derived antioxidant peptides.
