Seed-assisted hydrothermal synthesis of Sn-Beta for conversion of glucose to methyl lactate: effects of the H2O amount in the gel and crystallization time

Xiaomei Yang,Liuyong Wang,Tianliang Lu,Beibei Gao,Yunlai Su,Lipeng Zhou

Hydrothermally synthesized Sn-Beta is an excellent catalyst for the conversion of sugars to alkyl lactates. However, its catalytic performance is highly dependent on the synthesis parameters. In this work, the effects of the synthesis parameters such as the water amount of the synthesis gel and the crystallization time on the synthesis and properties and catalytic performance of Sn-Beta in the conversion of glucose to methyl lactate (MLA) were investigated. The crystallization rate was accelerated remarkably by reducing the water amount of the synthesis gel. With the aid of the Si-Beta seed, Sn-Beta was almost fully-crystallized after one day with nH2O/nSiO2 of 4.5, but seven days were needed with nH2O/nSiO2 of 7.5. However, Sn-Beta synthesized with nH2O/nSiO2 of 4.5 has more extraframework SnO2 due to the fast formation of the *BEA structure and the slow incorporation of Sn4+ into the framework sites, which resulted in the lower activity of MLA formation. More Sn4+ species were incorporated into the framework sites and thus more Lewis acid sites were generated with increasing crystallization time. At the same time, the ratio of open to closed Sn sites (RO/C) increased, which led to the decrease in the activity of MLA formation and due to that, the open Sn site with a proximal silanol is unfavorable for the conversion of glucose to MLA. During recycling study, it is surprisingly found that the yield of MLA increased gradually over the fully-crystallized Sn-Beta catalyst. Characterization results of the recovered catalyst from the eighth run indicate that the structure was preserved well, but the value of RO/C reduced due to the change of the state and microenvironment of Sn species.

Magnolol alleviates Alzheimer's disease-like pathology in transgenic C. elegans by promoting microglia phagocytosis and the degradation of beta-amyloid through activation of PPAR-γ

Zhishen Xie,Jianping Zhao,Hui Wang,Yali Jiang,Qiaoling Yang,Yu Fu,Huahui Zeng,Christian Hölscher,Jiangyan Xu,Zhenqiang Zhang

This study aims to investigate whether magnolol (MG), a natural neolignane compound, can prevent AD induced by beta-amyloid (Aβ) and the possible mechanisms involved. MG dose-dependently reduces Aβ deposition, toxicity and memory impairment caused by Aβ in transgenic C. elegans . More importantly, these effects are reversed by GW9662, a selective peroxisome proliferator-activated receptor-γ (PPAR-γ) antagonist. MG is more effective in enhancing PPAR-γ luciferase levels than honokiol (HK). Meanwhile, MG has the potential to bind with the ligand binding domain of PPAR-γ (PPAR-γ-LBD). As expected, MG inhibited the luciferase activity of NF-κB and its target genes of inflammatory cytokines, and this effect was blocked by GW9662. The luciferase activity of Nrf2-ARE expression can be activated by MG and decreased Aβ-induced reactive oxygen species (ROS). The target gene LXR of PPAR-γ is activated by MG, which upregulates ApoE and promotes microglia phagocytosis and the degradation of Aβ, and these effects were also reversed by GW9662. In summary, MG can attenuate Aβ-induced AD and the underlying mechanism is the reduction of inflammation and promotion of phagocytosis and degradation of Aβ, which is dependent on PPAR-γ.

Highly sensitive Curcumin-conjugated nanotheranostic platform for detecting amyloid-beta plaques by magnetic resonance imaging and reversing cognitive deficits of Alzheimer's disease via NLRP3-inhibition

Ruan Yuting,Xiong Ying,Fang Wenli,Yu Qun,Mai Yingren,Cao Zhiyu,Wang Kexi,Lei Ming,Xu Jiaxin,Liu Yan,Zhang Xingcai,Liao Wang,Liu Jun

Background Alzheimer's disease (AD) is the most common neurodegenerative disorder without effective therapy and lack diagnosis strategy for preclinical AD patients. There is an urgent need for development of both early diagnosis and therapeutic intervention of AD. Results Herein, we developed a nanotheranostics platform consisting of Curcumin (Cur), an anti-inflammatory molecule, and superparamagnetic iron oxide (SPIO) nanoparticles encapsulated by diblock 1,2-dio-leoyl- sn -glycero-3-phosphoethanolamine- n -[poly(ethylene glycol)] (DSPE-PEG) that are modified with CRT and QSH peptides on its surface. Furthermore, we demonstrated that this multifunctional nanomaterial efficiently reduced β-amyloid plaque burden specifically in APP/PS1 transgenic mice, with the process noninvasively detected by magnetic resonance imaging (MRI) and the two-dimensional MRI images were computed into three-dimension (3D) plot. Our data demonstrated highly sensitive in vivo detection of β-amyloid plaques which more closely revealed real deposition of Aβ than previously reported and we quantified the volumes of plaques for the first time based on 3D plot. In addition, memory deficits of the mice were significantly rescued, probably related to inhibition of NLR Family Pyrin Domain Containing 3 (NLRP3) inflammasomes. Conclusions Gathered data demonstrated that this theranostic platform may have both early diagnostic and therapeutic potential in AD. Graphical Abstract

Rapid and sensitive tapered-capillary microextraction combined to on-line sample stacking-capillary electrophoresis for extraction and quantification of two beta-blockers in human urine

Shucheng Sun,Yujia Wang,Xin Liu,Rao Fu,Li Yang

A tapered-capillary microextraction ( t Cap-μEx) combining with field-amplified stacking (FASI) method for CE analysis was developed. The t Cap- μ Ex method is based on the construction of a micro solid phase extraction (SPE) column by narrowing the end of a silica capillary from 530   µm (inner diameter) to 20   µm, enabling the packing of 45   µm sorbent particles without a frit. Various parameters that may affect the microextraction and FASI-CE analysis have been investigated and optimized. This study shows that microextraction exhibits advantages of small sample and sorbent volumes (less than 200   μL sample and 2   μL sorbent) and fast extraction time of 6   min. The method was successfully applied for efficient determination of atenolol and metoprolol in human urine samples, with recovery of 93.7–105.5% and RSD (n=3) lower than 8.5%. Twenty-one-fold and nineteen-fold average enhancement of detection sensitivity was achieved for atenolol and metoprolol, respectively, versus the CE method without t Cap- μ Ex and FASI. The method is environmentally friendly and allows reuse of the sorbent at least 8 times without an obvious loss in performance. The results indicate that the proposed method could be potentially applied in a wide range of doping control, clinical, forensic toxicology, food analysis and environmental analyses.

N-3 polyunsaturated fatty acids attenuate amyloid-beta-induced toxicity in AD transgenic Caenorhabditis elegans via promotion of proteasomal activity and activation of PPAR-gamma

Yanqing Wang,Huanying Zhang,Feng Ding,Jianhua Li,Lianyu Li,Zhong Xu,Yan Zhao

Alzheimer's disease (AD) is a common neurodegenerative disease that causes progressive cognitive decline. A major pathological characteristic of AD brain is the presence of senile plaques composed of β-amyloid (Aβ), the accumulation of which induces toxic cascades leading to synaptic dysfunction, neuronal apoptosis, and eventually cognitive decline. Dietary n-3 polyunsaturated fatty acids (PUFAs), such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are beneficial for patients with early-stage AD; however, the mechanisms are not completely understood. In this study, we investigated the effects of n-3 PUFAs on Aβ-induced toxicity in a transgenic AD Caenorhabditis elegans (C. elegans) model. The results showed that EPA and DHA significantly inhibited Aβ-induced paralytic phenotype and decreased the production of reactive oxygen species while reducing the levels of Aβ in the AD worms. Further studies revealed that EPA and DHA might reduce the accumulation of Aβ by restoring the activity of proteasome. Moreover, treating worms with peroxisome proliferator-activated receptor (PPAR)-γ inhibitor GW9662 prevented the inhibitory effects of n-3 PUFAs on Aβ-induced paralytic phenotype and diminished the elevation of proteasomal activity by n-3 PUFAs, suggesting that PPARγ-mediated signals play important role in the protective effects of n-3 PUFAs against Aβ-induced toxicity.

A Novel Nanoscale Phase-Change Contrast Agent Evaluates the Hepatic Fibrosis Through Targeting Hepatic Stellate Cell Platelet-Derived Factor Beta Receptor by Ultrasound in Vitro

Han-mei Li,Lin-li Feng,Qiong Jiang,You Yang,Ju-ying Zhang,Xia Luo,Xing Yang,Bo Ren,Li-tao Ye,Zheng-ju Hou,Yang Li,Jin-hong Yu

Objective As a reversible condition at its early stages, liver fibrosis can progress to cirrhosis and hepatocellular carcinoma, underscoring the importance of early detection for preventing severe outcomes and improving prognosis. To address this issue, we developed a platelet-derived growth factor receptor β (PDGFRβ)-targeted nanoscale phase-change contrast agent to target activated hepatic stellate cells (aHSC) and enable ultrasound imaging as a foundation for the early evaluation of liver fibrosis. Methods PDGFR-β antibody-modified phase-change contrast agents (PPCAs) were synthesized utilizing film hydration and ultrasonic emulsification with perfluoropentane (PFP) encapsulated. PPCAs were specifically conjugated to aHSC with high PDGFR-β expression, whose targeting ability was evaluated using fluorescence confocal microscopy and flow cytometry. Phase transition at different temperatures and mechanical indices (MIs), as well as contrast-enhanced ultrasound imaging were analyzed. Results PPCAs had an average diameter of 283.6 ± 11.3 nm with good dispersibility and relative stability, and the echo intensity increased correspondingly with increasing MIs. PPCAs exhibited both excellent biocompatibility and imaging ability when excited by high-frequency ultrasound set to an MI of 1.0 at 37°C, and simultaneously showed strong specific targeting ability to aHSC, with cellular uptake reaching 56.67 ± 5.96%. Conclusion As a new imaging avenue, PPCAs have the potential to enhance ultrasound imaging and establish the basis for diagnosis by targeting aHSC specifically with good biocompatibility and stability.

Characterization of a novel beta-cypermethrin-degrading strain of Lactobacillus pentosus 3-27 and its effects on bioremediation and the bacterial community of contaminated alfalfa silage

Fang Liu,Jie Bai,Wenkang Huang,Fuhou Li,Wencan Ke,Yixin Zhang,Dongmei Xie,Bo Zhang,Xusheng Guo

In this study, a novel beta-cypermethrin (beta-cyp)-degrading strain Lactobacillus pentosus 3–27 (LP3–27) was screened from beta-cyp-contaminated silage. The strain could degrade 96% of beta-cyp (50 mg/L) in MSM medium after 4 d of culture, while the strain lost its degradation ability when the beta-cyp concentration reached 250 mg/L. The effects of LP 3–27 on fermentation, bacterial community, and bioremediation of contaminated alfalfa silage at two dry matter (DM) contents were studied. The results showed that inoculation with LP3–27 not only degraded beta-cyp, but also improved the fermentation quality of alfalfa silage after 60 d of ensiling. Meanwhile, L. pentosus dominated the bacterial community during ensiling in LP3–27 inoculated silages, whereas Pediococcus acidilactici was the dominant species in the control silage. LP3–27 inoculation also simplified the bacterial interaction networks of ensiled alfalfa. Beta-cyp degradation was positively correlated with L. pentosus in LP- inoculated silages, which confirmed the function of beta-cyp degradation by L. pentosus . In addition, higher beta-cyp degradation was observed in silage with 35% versus 43% DM. In summary, strain LP3–27 could be used as a candidate inoculum for bioremediation of beta-cyp-contaminated silage and to produce safe silage for animal production.

Blue Light and Methyl Jasmonate Synergistically Enhance Betalain Accumulation, Antioxidant Enzyme Activity, and Osmotic Adjustment in Sugar Beet (Beta vulgaris L.) Seedlings: A Time-Course Analysis

Hui Wang,Chao Yang,Yanling Yu,Dayou Cheng,Cuihong Dai,Chengfei Luo

Betalains are natural antioxidant pigments valued as food colorants (E162), yet their combined responses to light quality and methyl jasmonate (MeJA) during long-term growth remain poorly understood. In this study, the interactive effects of blue light and MeJA on growth, betacyanin, betaxanthin, antioxidant enzyme activities [peroxidase (POD), superoxide dismutase (SOD), catalase (CAT)], malondialdehyde (MDA), and proline (PRO) were investigated at weeks 3, 10, and 13 of sugar beet seedling growth. Four treatments were established: white light (W, control), white light + MeJA (WM), white light + blue light (WB), and white light + blue light + MeJA (WBM). The results showed that WB rapidly induced betaxanthin accumulation and enhanced SOD and POD activities while reducing MDA at week 3. MeJA alone triggered an explosive increase in betacyanin (45.74 mg·g−1FW) at week 10, accompanied by elevated activities of POD, SOD, and CAT. The combined treatment (WBM) maintained the highest betacyanin (36.48 mg·g−1FW) and betaxanthin (8.97 mg·g−1FW) contents, the lowest MDA level (17.17 nmol·g−1FW), and a high proline level (528.39 μg·g−1FW) at week 13, providing sustained pigment maintenance at the late stage. The three antioxidant enzymes exhibited a temporal division of labor: high SOD activity at the early stage, while POD and CAT activities continuously increased during the middle and late stages. Notably, the correlation between proline and MDA shifted from positive at week 10 to negative at week 13, suggesting a temporal transition in the protective role of proline against membrane lipid peroxidation during late-stage development. In conclusion, blue light and MeJA enhance the antioxidant capacity of sugar beet seedlings through a temporally synergistic pattern of “priming by blue light, burst by MeJA, and maintenance by combined treatment,” offering a potential preharvest regulation strategy for the production of natural pigments and functional food ingredients.

Abrus cantoniensis α-glucan-like polysaccharide alleviates influenza via gut microbial acetate to activate free fatty acid receptor 2/ mitochondrial antiviral signaling protein/interferon-beta pathway

Yuting Yi,Dongdong Li,Yuan Li,Haotong Wang,Daxin Yang,Simin Yang,Shangping Xing,Shanshan Wei,Jie Yang,Hongwei Guo,Zhuo Luo

ACP-A1, a novel an α-glucan-like Polysaccharide from Abrus cantoniensis , is structurally resolved for the first time. • ACP-A1 reshapes gut microbiota and triggers the FFAR2/MAVS/IFN-β antiviral circuit. • Microbiota-driven IFN-β signaling underpins ACP-A1 protection against H1N1 influenza. Background The gut microbiota is critical for host defense against influenza. Polysaccharides are known for their microbiota-modulating and immunomodulatory activities; however, the anti-influenza efficacy of homogeneous Abrus cantoniensis polysaccharides (ACP) remains unexplored. Purpose The present study seeks to clarify the protective role of ACP in influenza and explore its underlying molecular mechanisms. Methods Initially, crude polysaccharides were extracted via ethanol precipitation and subsequently purified by gel chromatography. Systematic structural characterization of ACP was then performed using carbohydrate chemistry techniques, including scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), ultraviolet (UV) spectroscopy, and nuclear magnetic resonance (NMR). The therapeutic efficacy of ACP was assessed by monitoring various indicators such as body weight, survival rate, viral load, and pulmonary pathological changes in mouse models. Furthermore, to elucidate the biological mechanism underlying ACP’s anti-influenza activity via regulation of pulmonary interferon-beta (IFN-β) immune networks by intestinal acetate-producing microbiota, multi-omics analyses integrating metagenomics, metabolomics, gene knockout, immunofluorescence, and Western blot were conducted. Finally, the potential anti-influenza effects of ACP via the gut-lung axis were evaluated based on in vivo and in vitro detection of protein expression of IFN-β, free fatty acid receptor 2 (FFAR2), and mitochondrial antiviral signaling protein (MAVS), as well as antiviral interferon-stimulated genes (ISGs). Results In this study, we purified a novel polysaccharide, ACP-A1, with a backbone of→4)-α-D-Glc p -(1→,→4)-β-D-Gal p -(1→, and →4,6)-α-D-Glc p -(1→ linkages and α-D-Glc p -(1→ branches at O-6. In H1N1-infected mice, oral ACP-A1 alleviated weight loss, increased survival, and reduced lung inflammation and viral load. Metagenomic and targeted metabolomic analyses showed that ACP-A1 enriched Limosilactobacillus reuteri and elevated acetate levels. Fecal microbiota transplantation, FFAR2 inhibition, and MAVS knockout experiments demonstrated that ACP-A1 enhances the FFAR2/MAVS/IFN-β antiviral pathway via microbial-derived acetate. Conclusion Collectively, our findings elucidate that ACP mitigates influenza virus-induced lung dysfunction by promoting the proliferation of acetate-producing gut microbiota, particularly Limosilactobacillus reuteri , and activating the FFAR2/MAVS/IFN-β antiviral axis in pulmonary immune cells. These findings establish ACP-A1 as a natural polysaccharide regulating IFN-β homeostasis, highlighting its potential for influenza prevention. Enhanced antibacterial efficacy and mechanism of octyl gallate/beta-cyclodextrins against Pseudomonas fluorescens and Vibrio parahaemolyticus and incorporated electrospun nanofibers for Chinese giant salamander fillets preservation

Yu-gang Shi,Dong-hui Li,Yi-ming Kong,Run-run Zhang,Qing Gu,Meng-xin Hu,Shi-yi Tian,Wen-gang Jin

A series of alkyl gallates were evaluated for the antibacterial activity against two common Gram-negative foodborne bacteria ( Pseudomonas fluorescens and Vibrio parahaemolyticus ) associated with seafood. The length of the alkyl chain plays a pivotal role in eliciting their antibacterial activities and octyl gallate (OG) exerted an excellent inhibitory efficacy. To extend the aqueous solubility, stability, and bactericidal properties of octyl gallate (OG), an inclusion complex between OG and β -cyclodextrin ( β CD), OG/ β CD, was prepared and identified with various methods including X-ray diffraction (XRD), differential scanning calorimeter (DSC) and Fourier transform infrared spectroscopy (FTIR). Furthermore, the enhanced inhibitory effect and potential antibacterial mechanism of OG/ β CD against two Gram-negative and Gram-positive foodborne bacteria were comprehensively investigated. The results show that OG/ β CD could function against bacteria through effectively damaging the membrane, permeating into cells, and then disturbing the activity of the respiratory electron transport chain to cause the production of high-level intracellular hydroxyl radicals. Moreover, the reinforced OG/ β CD-incorporated polylactic acid (PLA) nanofibers were fabricated using the electrospinning technique as food packaging to extend the Chinese giant salamander fillet's shelf life at 4 °C. This research highlights the antibacterial effectiveness of OG/ β CD in aqueous media, which can be used as a safe multi-functionalized food additive combined with the benefits of electrospun nanofibers to extend the Chinese giant salamander fillets shelf life by 15 d at 4 °C.

Production of Natural Penicillin-Binding Protein 2 and Development of a Signal-Amplified Fluorescence Polarization Assay for the Determination of 28 Beta-Lactam Antibiotics in Milk

Jia Jia Hu,Jian Ping Wang

In this study, two magnetic activity-based protein profiling probes based on cephradine and amoxicillin were first synthesized that were used to produce the natural penicillin-binding protein 2 of Escherichia coli. After characterization by using LC-ESI-MS/MS, it was found that the obtained proteins by using the two probes were the same. The molecular docking for 28 beta-lactam antibiotics showed that the key amino acids were Ser330 and Ser387, the main intermolecular forces were hydrogen bond and hydrophobic interaction, and the main binding sites in their molecules were on the beta-lactam ring. Then this protein was combined with streptavidin-labeled tracer and biotinylated fluorescein isothiocyanate to establish a signal-amplified fluorescence polarization assay to determine the 28 drugs in milk. The limits of detection ranged from 0.07 to 2.21 ng/mL, and the sensitivities for the 28 drugs were improved 4 to 48-fold in comparison with the use of a fluorescein isothiocyanate-labeled fluorescent tracer. Therefore, this method could be used for rapid multiscreening of the 28 beta-lactam antibiotics in milk.

9R, the cholinesterase and amyloid beta aggregation dual inhibitor, as a multifunctional agent to improve cognitive deficit and neuropathology in the triple-transgenic Alzheimer's disease mouse model

Yaojun Ju,Kin Yip Tam

Alzheimer's disease (AD) is the most common kind of dementia in the aging population leading to great social and financial burdens in many countries around the world. For decades, disease-modifying drug developed using the “one target, one drug” strategy failed to conquer this disease. Recently, we have designed and synthesized 9R , which exhibited dual inhibition of cholinesterase and amyloid beta (Aβ) aggregation in vitro . Herein, we evaluated the in vivo efficacy of 9R in a triple transgenic AD (3xTg-AD) mouse model. 3xTg-AD mice (10-month-old) were dosed intraperitoneally with 9R (daily 3, 10 or 30 mg/kg) for a month. Known cholinesterase inhibitor donepezil (0.3 mg/kg) and Aβ aggregation inhibitor tramiprosate (30 mg/kg) were used as positive controls. Cognitive performance of the mice was then evaluated by using Morris Water Maze (MWM), Y-maze tasks and Open Field test. The acetylcholine level, degree of Aβ deposition, amyloid precursor protein (APP) processing, neuroinflammation , tau deposition and tau hyperphosphorylation in the brains of the 3xTg-AD mice were examined. We have observed that one-month treatment with 9R significantly improved cognitive deficits in 3xTg-AD mice. Moreover, 9R treatment enhanced the brain acetylcholine level and mitigated the amyloid burden, tau hyperphosphorylation and neuroinflammation in the mouse brains. The effects of 9R on APP processing, neuroinflammation, tau hyperphosphorylation and Cdk-p25 action demonstrated its multifunctional role in 3xTg-AD mouse model. Our results suggested that the use of multi-target compound could be a potential approach to treat AD.
