The low bioavailability of nutraceuticals, due to chemical instability and non-targeted release, remains a major delivery challenge. To address this, biodegradable core-shell hydrogel microspheres with controllable diameters ranging from 800 to 2850 μm were fabricated via an extrusion method for the co-delivery of hydrophilic and hydrophobic nutraceuticals. These microspheres employ Fe 2 O 3 -loaded montmorillonite (Fe Mt) as the reinforcing scaffold, sodium carboxymethyl cellulose (CMC) as the core, and chitosan (CS) as the shell. They were co-loaded with hydrophilic l -glutamine (L-Gln) and hydrophobic curcumin (Cur) as model nutraceuticals. The incorporation of Fe Mt overcomes the low encapsulation efficiency and mechanical weakness of polymer microspheres. Meanwhile, at Fe Mt contents of 6.00 wt% (for L-Gln) and 5.00 wt% (for Cur) relative to the CMC weight, the encapsulation efficiencies reach up to 75.42% and 63.16%, respectively. The CS shell thickness increases from 10.83 to 16.05 μm as the CS concentration rises from 10.0 to 25.0 g/L. This thickness governs the release kinetics and pH-responsive behavior, wherein the microspheres contract in acidic environments and swell under neutral/alkaline conditions to facilitate targeted delivery in the gastrointestinal tract. In simulated digestion, cumulative release of nutraceuticals from the microspheres was 93.02% for L-Gln and 88.37% for Cur within 12 h. Additionally, the microspheres exhibited good storage stability (ca. 1.03% loss after 45 days). This work presents a highly promising and versatile platform for the co-delivery of hydrophilic and hydrophobic nutraceuticals with controlled release profiles.

Curcumin-gelatin blend best balances safety and flavor in roasted beef. • Synergistically inhibits harmful HAAs and AGEs by over 50–68%. • Preserves key meaty aroma compounds better than other hydrocolloids. • Offers a clean-label strategy for healthier thermally processed meats. • Highlights the safety–flavor trade-off in functional meat formulation. This study investigated curcumin combined with ten food-grade hydrocolloids in air-fried beef patties, focusing on the trade-off between hazard reduction and flavor quality. Curcumin alone reduced total HAs by 29% and total AGEs by 39%, with individual HA inhibition varying considerably (e.g., 40% for MeIQx, 34% for PhIP, but only 12% for Harmane). Combinations with chitosan and pectin achieved the highest inhibition (77–89% for both total HAs and total AGEs) but substantially altered the volatile profile. In contrast, gelatin provided a compromise: moderate hazard reduction (54% HAs, 68% AGEs) with minimal volatile changes, positioning it as a practical balance between safety and flavor quality. Although flavor assessments were based on instrumental analysis (GC-MS and electronic nose), further sensory evaluation is needed to confirm consumer perception. These findings highlight the inherent safety-flavor trade-off in hydrocolloid-curcumin formulations and identify gelatin as a candidate for achieving a practical compromise.

Targeted nanoliposomes constitute a critical platform for improving therapeutic outcomes in oncology. However, their cellular recognition and uptake efficiency are frequently compromised by the heterogeneity of receptor expression on tumor cells and the inherent limitations of single-ligand targeting strategies. To overcome these challenges, we developed a dual-targeted liposomes system comodified with folic acid and transferrin (FA/Tf-F127-Lps) for curcumin delivery, leveraging dual-receptor synergy to enhance targeting and cellular internalization. The FA/Tf-F127 conjugate was synthesized via chemical coupling and used to prepare curcumin-loaded liposomes through thin-film hydration combined with dynamic high-pressure microfluidics, achieving a drug encapsulation efficiency of 86.69%. Cellular experiments demonstrated that the dual-targeted system exhibited the strongest cytotoxicity against C6 glioma cells, with IC50 values of 1.297 mg/L at 48 h and 1.175 mg/L at 72 h, and its cellular uptake was significantly higher than that of single-targeted and nontargeted controls. Uptake mechanistic studies revealed that internalization was energy-dependent and involved clathrin-mediated, caveolae-mediated, and macropinocytic pathways, and could be competitively inhibited by free folic acid and transferrin. This dual-targeted system shows substantial promise for treating receptor-overexpressing tumors such as glioma by achieving enhanced specificity and intracellular delivery. The strategy provides a feasible experimental basis for addressing receptor heterogeneity and improving targeting precision in glioma therapy, while also offering an approach for developing next-generation personalized nanomedicines.

BACKGROUND Due to its high glutenin content, walnut protein isolate (WPI) exhibits low solubility, limiting its applications in the food industry. RESULTS WPI was modified with succinic anhydride (SA), and the potential of the resulting curcumin-loaded emulsions was evaluated. The degree of succinylation of WPI was measured as 89.52 ± 0.66% when 40% SA was used. Succinylation enhanced the surface charge of WPI, reduced its surface hydrophobicity and disulfide bond content, and involved both lysine residues and free sulfhydryl in the acylation reaction. Structural analyses, including Fourier transform infrared spectroscopy, intrinsic fluorescence, and scanning electron microscopy, revealed that SA modification induced significant conformational changes in WPI. Notably, at 20% SA addition, WPI exhibited optimal functional properties, achieving solubility as high as 98.24%. In vitro digestion assays demonstrated that succinylation decreased the release of free fatty acids from emulsions and improved the bioavailability of curcumin to 86.81%. CONCLUSION This study demonstrated that SA modification is an effective way for enhancing the structural and functional properties of WPI. The SA-modified WPI shows potential as a carrier for improving the solubility and bioavailability of hydrophobic bioactive compounds. Moreover, this work provides new insights into the significance of WPI modification and offers practical guidance for its application in the food industry. © 2026 Society of Chemical Industry.

Background The process of healing wounds in diabetic patients is intricate and is notably obstructed by a disordered wound microenvironment, characterized by chronic inflammation and elevated blood glucose. The combination of stem cell therapy and drug treatment is seen as a promising application in future. However, the limited proliferative capacity of stem cells and inadequate drug availability present significant challenges for achieving optimal therapeutic outcomes. Methods In this study, open porous poly (lactic‒coglycolic acid) (PLGA) microspheres were designed and synthesized via gas-assisted volatilization microemulsion technology. These microspheres encapsulate curcumin and allow its slow release, thereby enhancing wound repair. The large pores in the microspheres provide ample support for bone marrow stem cells (BMSCs), enabling continuous drug release over a period of 35 days. Results The sustained release of curcumin promoted stem cell proliferation and maintained stem cell activity. Additionally, it facilitates remodeling of the wound immune microenvironment. Additionally, the microspheres can activate mitochondrial autophagy in cells, effectively alleviating wound inflammation. Conclusions The combined actions of curcumin and stem cells aid in regenerating blood vessels and revitalizing the collagen network where the injury occurred, thus improving wound healing capabilities. Consequently, integrating drugs with stem cells and microspheres holds significant potential for diabetic wound treatment.

Food spoilage poses a significant threat to human health and contributes to substantial global food waste. To address this issue, advancements in biopolymer-based packaging materials have garnered increasing research attention. A novel zein-curcumin nanoparticles/sodium alginate (U-ZcNPs/SA) composite films was fabricated by encapsulating curcumin (Cur) within zein nanoparticles via ultrasonic-assisted self-assembly, followed by integration into an SA matrix. Ultrasonic treatment induced structural reorganization of zein, enhancing electrostatic interactions and hydrogen bonding within the film matrix, which improved nanoparticle dispersion, mechanical properties (tensile strength: 40.51 N/mm 2 ; elongation at break: 72.23 %), and barrier performance (oxygen permeability: 2.44 g/hm 2 ). Characterization techniques (SEM, FTIR, XRD, TGA) confirmed superior interfacial compatibility, thermal stability, and amorphous structure in U-ZcNPs@SA films. Furthermore, the films demonstrated robust antibacterial activity against both Escherichia coli and Staphylococcus aureus , with inhibition zones measuring up to 11.09 cm and 10.78 cm, respectively. In the preservation experiment, the U-ZcNPs@SA film effectively extended the shelf life of grapes and strawberries by maintaining their visual quality. Notably, it significantly reduced weight loss in strawberries while suppressing their respiration rate and ethylene production. These results demonstrated that ultrasonic modification optimized the functional performance of biodegradable films, offering a sustainable strategy for active food packaging.

Hepatocellular carcinoma (HCC) is a serious concern worldwide. The published reports showed that aberrant CD95 receptor expression plays a critical role in apoptosis in liver cancer. While curcumin has shown promise in inducing apoptosis in liver cancer cells, its direct effects on CD95 expression during this process have not been thoroughly investigated. This study aims to quantitatively assess the expression of the CD95 receptor in HepG2 cells treated with different concentrations of curcumin using techniques such as fluorescence staining, single-molecule force spectroscopy (SMFS), and single-molecule recognition imaging (SMRI). Fluorescence staining results indicate a significant increase in CD95 expression following curcumin treatment. For the first time, SMFS and SMRI techniques were used to directly reveal the binding sites of CD95 on the cell membrane, with the number of binding sites increasing as the curcumin concentration increased. Additionally, the binding force between an antibody-modified probe and CD95 was strengthened with curcumin treatment, suggesting that curcumin enhances both the quantity and affinity of CD95 binding sites. This study provides new insights into curcumin-induced CD95-mediated apoptosis in liver cancer cells and highlights the potential of AFM techniques for investigating drug mechanisms. Overall, these findings may inform innovative therapeutic strategies for liver cancer and improve drug design processes.

Visceral adiposity is an important characteristic of metabolic dysfunction-associated steatohepatitis (MASH) that affects its development. Curcumin (Cur) attenuates MASH through microbial biotransformation. However, antibiotics (Abx) do not weaken but rather enhance the resistance to weight gain of Cur on MASH rats. The resistance to weight gain mechanism of Cur is different from its anti-MASH effect, which may relate to alleviate visceral adiposity. Here we investigated the mechanism of Cur against visceral adiposity in high-fat diet-induced MASH rats. The results demonstrated that Cur and Abx reduced body, liver and visceral fat weights of MASH rats. Unlike Abx, which induces resistance to weight gain by reducing appetite, Cur mainly reduced the weight of visceral fat, especially that of perirenal fat. Intriguingly, the reduction in perirenal fat caused by Cur was attributed to its specific inhibition of gastric inhibitory polypeptide (GIP) release. The Cur-induced decrease in intestinal GIP release inhibited the activation of GIP receptors to alleviate adipogenesis and inflammation in perirenal adipose tissue. Moreover, Cur alleviated intestinal epithelium and vascular barrier disruption-mediated hypoxia to inhibit GIP release. In summary, the pharmacological effects of Cur on visceral adiposity were mainly contributed by inhibiting gut barrier disruption-mediated hypoxia to attenuate GIP release.

Chitosan-coated nanostructured lipid carriers (CS-NLCs) were engineered to overcome the gastrointestinal instability and limited absorption of glycerol distearate (GDS)/medium-chain triglyceride (MCT)-based NLCs. Using a dropwise coating method, we systematically evaluated the impact of chitosan coating on digestive stability and intestinal permeability through in vitro digestion models, Ussing chamber assays, and confocal laser scanning microscopy (CLSM). The results indicate that Chitosan coating improved NLCs’ storage stability (except G2M8 formulation). CS-NLCs exhibited more complete digestion (the maximum fatty acid released within 20 minutes versus 30 minutes for uncoated NLCs), with Turbiscan analysis demonstrating an increase in initial digestion rate compared to uncoated NLCs, though chitosan-induced flocculation reduced small intestinal permeability (52% lower apparent permeability coefficient in Ussing chambers). CLSM imaging identified a critical solid lipid threshold (>50%) for chitosan-mediated adhesion enhancement, with 80% solid lipid CS-NLCs showing 1.6-fold higher mucus adhesion due to incomplete chitosan degradation and increased particle size (from 747 nm to 16μm). Pharmacokinetic studies in rats confirmed that CS-NLCs with >50% solid lipid achieved 1.8-fold higher curcumin bioavailability versus uncoated counterparts. This study highlights the critical role of chitosan coating in optimizing the bioavailability of lipid-based nutraceuticals in the gastrointestinal tract.

Lactobacillus plantarum (LP) and curcumin (CUR) were co-encapsulated in sodium alginate– Lactobacillus plantarum– curcumin–gelatin (ALG–LP–CUR–GE) hydrogel beads to evaluate the synergistic function of probiotics and prebiotics. The encapsulation efficiency of LP and CUR peaked at 84.6 % and 94.7 %, respectively. Results of scanning electron microscope, Fourier transform infrared spectrometry, and X-ray diffraction confirmed the encapsulation of LP and CUR in the hydrogel beads. In vitro digestion suggested that both CUR and LP can achieve successful release in the simulated intestinal fluid. The release kinetics of CUR showed that it exhibited predominantly Fickian release and non-Fickian release in gastric fluid and intestinal fluid, respectively. The application of co-encapsulated hydrogel beads in mimic milk tea illustrated that the viable count of LP remained above the minimum recommended threshold of 10 7 CFU/mL, offering a promising avenue for the delivery of synergistic probiotic and prebiotic functionalities.

This study aimed to explore the potential of soybean protein isolate hydrolysates (SPIH) prepared via Alcalase as delivery carriers and develop novel SPIH-Cur nanoparticles. Hydrolysis caused the varying degrees degradation in the 7S and 11S subunits, significantly enhancing SPI's antioxidant activity. The reduction in particle size and the exposure of hydrophobic groups in SPIH contributed to the formation of stable SPIH-Cur nanoparticles, due to their well binding capacity to curcumin (Cur). The 30 min SPIH-Cur sample exhibited the highest encapsulation efficiency (83.09 %), owing to its high binding affinity (Ka = 9.56 × 10 3 M −1 ). Encapsulation by SPIH also significantly improved Cur's thermal and light stability. Moreover, FTIR, fluorescence spectra, and molecular docking analyses revealed that the formation of SPIH-Cur were primarily driven by hydrophobic forces and hydrogen bonds. Above results provide a foundation for fabricating nanoparticles that deliver lipophilic bioactive compounds with high encapsulation efficiency and stability derived from SPIH.

Osteopontin (OPN), highly expressed in foam cells, serves as a hallmark of atherosclerotic plaques and a potential target for site-specific drug delivery. In this study, we developed OPN-targeted liposomes (OPN LIP) co-loaded with curcumin and perfluoro-n-pentane (PFP) to enhance therapeutic efficacy against atherosclerosis. The liposomes exhibited uniform particle size, colloidal stability, favorable biocompatibility, and minimal cytotoxicity. In vitro experiments demonstrated efficient and time-dependent cellular uptake by ox-LDL–stimulated RAW264.7 foam cells. Upon exposure to low-intensity focused ultrasound (LIFU), the nanocarriers underwent acoustic cavitation-induced rupture, leading to rapid curcumin release and significant induction of apoptosis. OPN LIP treatment notably reduced IL-1α and TNF-α levels and aggravated apoptosis in foam cells. Transcriptomic analysis revealed modulation of inflammatory and apoptotic pathways, including TLR, NF-κB, and Mapk3 signaling. These findings highlight the potential of OPN LIP as an ultrasound-responsive, targeted nanoplatform capable of suppressing inflammation and foam cell viability, providing a promising approach for the treatment of atherosclerosis through combined physical and pharmacological strategies.
