On-package ratiometric fluorescent sensing label based on AIE polymers for real-time and visual detection of fish freshness

Xiuying Liu,Yu Wang,Zexin Zhang,Lijie Zhu,Xue Gao,Keli Zhong,Xiaofei Sun,Xuepeng Li,Jianrong Li

Freshness is an important parameter of fish quality. This study aims to develop a ratiometric fluorescent sensing label that is responsive to volatile amines, affording real-time and visual detection of fish freshness. For developing of the sensing label, an aggregation-induced emissive (AIE) polymer was prepared from the stimuli-responsive polymer polymethacrylic acid and the AIE molecule tetraphenylethylene and coated on to filter paper with rhodamine B as an internal reference. By exploiting the ratiometric response, the freshness of fish could be identified clearly and easily according to the color of on-package label, which changes from pink (fresh) to dark blue. The difference was linearly correlated with the total volatile basic nitrogen (TVB-N, R 2  = 0.995 and 0.994 at 25 °C and 4 °C, respectively) in the range of 15–25 mg/100 g for the salmon samples, which indicated that the sensing label feasibly and non-destructively quantified TVB-N.

Molecular analysis of type II topoisomerases of Aeromonas hydrophila isolated from fish and levofloxacin-induced resistant isolates in vitro

Hu Ruixue,Du Na,Chen Nan,Lin Li,Zhai Yanhua,Gu Zemao

The mechanisms of resistance to levofloxacin for Aeromonas hydrophila isolated from diseased fish and selected in vitro were examined in this study. Levofloxacin-resistant mutants were obtained by selection of A. hydrophila in vitro. The quinolone resistance-determining regions (QRDRs) of the gyrA and parC genes were sequenced in Lev R strains and reverse mutation strains. All Lev R strains carried a point mutation at codon 83 (Ser → Ile), and one strain (25 %) harbored a mutation at position 92 (Leu → Met) in the GyrA-QRDR. After being transferred in a levofloxacin-free medium, one strain of the mutants was successfully reversed and the reversion was related with mutations o f GyrA-QRDR at positions 81 (Gly → Asp) and 83 (Ile → Ser). No amino acid alteration was found in the ParC-QRDR. In addition, the minimum inhibitory concentration (MIC) of levofloxacin for the mutants was lower in the presence of reserpine, and all mutants were also resistant to some of the other quinolones. It was found that the mechanism of levofloxacin resistance of A. hydrophila selected in vitro was related to gyrA of type II topoisomerase, and an efflux mechanism was involved in the resistance as well.

Electrochemical detection of tryptophan in fish and pharmaceutical supplement at glassy carbon electrode modified with Fe doped ZnO nanoparticle

Toleshi Teshome,Shimeles Addisu Kitte,Abera Gure,Guta Gonfa

The co-precipitation approach was employed in this study to create Fe doped ZnO (Fe-ZnO) nanoparticles (NPs). To characterize the synthesized NPs, Ultra violet-Visible (UV-Vis) spectroscopy, X-ray diffraction (XRD), fourier transform infrared (FT-IR), scanning electron microscopy (SEM), and Brunauer-Emmett-Teller (BET) were used. Here, a novel electrochemical approach for the detection of tryptophan (Trp) using a glassy carbon electrode modified with Fe-ZnO NPs (Fe-ZnO/GCE) is demonstrated. The research findings revealed that Fe-ZnO/GCE enhanced the electron transfer rate of Trp oxidation. The sensitivity of Trp detection using Fe-ZnO/GCE in phosphate buffer solution (PBS) at pH 64.0 was significantly improved compared to bare GCE due to the electrocatalytic activity of Fe-ZnO NPs. Fe-ZnO/GCE exhibited a linear response with Trp concentrations ranging from 0.1 to 150 μM with the limit of detection (LOD) is 0.089 μM (3σ /m ) and limit of quantification (LOQ) about 0.3 μM (10σ /m ) using linear sweep voltammetry (LSV). The Fe-ZnO/GCE sensor was also effectively used to detect Trp in African catfish and multivitamin tablets. Trp analysis in real samples exhibited good recovery values of 89.60 to 99.15 %, demonstrating the accuracy and practicality of the method.

Developing and Validating a UPLC-MS Method with a StageTip-Based Extraction for the Biogenic Amines Analysis in Fish

Jingbo Zhang,Jing Hu,Shuai Wang,Xinping Lin,Huipeng Liang,Shengjie Li,Chenxu Yu,Xiuping Dong,Chaofan Ji

To effectively extract biogenic amines (BAs) from complex food matrices is an important issue. In this study, a stop-and-go-extraction tip (StageTip)-based method for simultaneous enrichment of six BAs including histamine, tyramine, putrescine, cadaverine, tryptamine, and β-phenylethylamine in fish samples was developed and optimized. This StageTip-based BAs extraction method required short extraction time, small sample size, and minimal consumption of organic solvents. In addition, an ultrahigh-performance liquid chromatography coupled with triple quadrupole mass spectrometer was used to identify and quantify BAs without derivatization. Target recovery efficiencies of BAs from spiked fish samples ranged from 84% to 110%. Linear regression models were established to determine BAs in the range of 0.4 to 3.0 mg/L ( R 2 > 0.995). Good repeatability was obtained with relative standard deviation of peak areas (1.17 to 5.61%) and retention time (0.09 to 0.20%). High sensitivity was achieved for the detection of six BAs (0.064 to 1.00 ng/L for the limit of detection, 0.10 to 5 ng/L for the limit of quantification). The proposed method had an obvious advantage in the analysis of BAs in complex food matrices such as fish tissues since it could reflect the subtle distribution of BAs from one fish fillet. Practical Application A StageTip-based BAs enrichment combined with UPLC-MS analysis method was developed and optimized, which was low - cost , environmentally friendly, and with good analytical performances. As such, it has the potential to become a powerful tool for the analysis of BAs in different complex food matrices.

Flavor enhancement of cell-cultured fish meat: regulating the generation and release of volatiles through targeted fatty acid supplementation

Huilin Zhao,Zhaoxuan Fan,Shan Shang,Donghong Liu,Lei Qin,Xuhui Huang

Replicating the authentic flavor profile of cell-cultured seafood remains a critical bottleneck, as current models predominantly based on monocultured myoblasts inherently lack the essential lipid-derived flavor precursors found in natural fish. This study constructed a cell-cultured fish meat (CM) model to investigate the flavor formation mechanisms driven by thermal interactions between the non-lipid cellular framework and exogenous fatty acids (oleic acid: OA; docosahexaenoic acid: DHA). Utilizing electronic nose and HS-SPME-GC-MS coupled with partial least-squares regression (PLS-R), the study revealed distinct flavor modulation pathways. The addition of OA significantly promoted the generation of methyl-pyrazine and alkyl furans, suggesting that OA oxidation intermediates triggered Strecker degradation with matrix proteins to construct a “lipid-derived aroma profile” backbone. Conversely, DHA specifically enriched unsaturated aldehydes and ketones (e.g., 4-heptenal, (E,E)-2,4-heptadienal) alongside 5-ethyl-2-methyl-pyridine, effectively imparting the characteristic “seafood-like aroma”. Furthermore, spatial separation in PLS-R demonstrated that the CM matrix exhibited a non-additive interaction with lipids:it specifically directed OA oxidation intermediates toward Strecker degradation to construct a lipid-derived aroma backbone, while selectively entrapping highly reactive DHA-derived unsaturated aldehydes to retain a balanced seafood-like profile. The cellular network acted as interactive microenvironments, exhibiting a modulating the release effect on highly reactive secondary oxidation products. This study elucidated the differential volatile generation pathways and controlled-release behaviors between the cellular matrix and specific lipids, providing theoretical insights into the targeted flavor modulation of thermally processed cell-cultured seafood.

Establishment and characterization of an ovarian cell line from red seabream (Pagrus major) and its application in fish toxicology

Yang Yucong,Zhang Yitong,Liu Mingyang,Wang Guixing,He Zhongwei,Liu Yufeng,Zhang Heng,Han Tian,Ren Yuqin,Hou Jilun

Fish cell lines serve as ideal in vitro models for investigating the toxicity of environmental pollutants in fish. In this study, a novel cell line (PmOVC) derived from the ovary of red seabream ( Pagrus major ) was successfully established. Using polystyrene microplastics (PS-MPs)—a typical environmental pollutant—as the test substance, the toxicity sensitivity of the PmOVC cell line was systematically assessed from the dimensions of particle size intake and cell activity. The results showed that the PmOVC cells exhibited robust growth in M199 medium supplemented with 15% FBS at 23°C and high exogenous gene transfection efficiency and showed high sensitivity to the toxic effects of microplastics. Specifically, the smaller the particle size of microplastics, the higher the internalization efficiency of cells, among which 100 nm PS-MPs could be completely endocytosed into the cytoplasm; the concentration of PS-MPs was significantly positively correlated with the apoptosis rate of cells ( R 2  ≥ 0.8). In summary, the established PmOVC cell line provides a critical in vitro tool for in-depth investigation of the toxicological responses and underlying mechanisms of fish to environmental pollutants.

Coffee shell-derived carbon dot-chlorogenic acid nanoparticles with enhanced aPDT antibacterial activity for fish preservation during cold storage

Chenxiao Chen,Yiran Yang,Maha Alharbi,Ashwag Shami,Fahad Saad Alhodieb,Saleh A. Alsanie,Haiying Cui,Lin Lin

Chlorogenic acid-loaded coffee shell-derived carbon dot nanoparticles (CGA/CSCD NPs) with enhanced antibacterial photodynamic therapy (aPDT) activity were developed, exhibiting strong fluorescence (~400 nm), a graphite-like structure, and around 3 nm size. The nanoparticles achieved efficient ROS generation and bacterial enrichment, showing superior antibacterial efficacy and biocompatibility. Applied to Carassius auratus , Megalobrama amblycephala , and Dicentrarchus labrax under 4 °C storage, the CGA/CSCD NPs-aPDT treatment inhibited 99% of bacterial growth over 7 days, maintaining counts at 10 4 –10 5  CFU/g. The treatment significantly slowed pH rise, inhibited TVB-N accumulation, and reduced lipid oxidation by 2–3 times compared to controls. Furthermore, the nanoparticles preserved fish quality by minimizing moisture loss and maintaining color stability and texture (hardness and chewiness). These results demonstrate that CGA/CSCD NPs-aPDT provides a sustainable and effective strategy for delaying spoilage and extending the shelf life of aquatic products during cold storage.

Heterojunction Engineering of POMOF and ZnO: A Promising Strategy to Boost Triethylamine Gas-Sensing Performance for Fish Freshness Evaluation

Meng Mei,Yi Zheng,Kaixin Cheng,Bo Yao,Liao Wang,Juan Wu,Xinxin Xing,Yude Wang

Triethylamine (TEA) serves as a key indicator of fish spoilage, and its accurate detection is essential for assessing fish freshness. However, existing TEA gas sensors often suffer from low sensitivity, sluggish response/recovery kinetics, and high operating temperatures. To address these limitations, this study, for the first time, integrates polyoxometalate-based metal-organic frameworks (POMOF) with ZnO via heterojunction engineering to construct a POMOF-ZnO heterojunction gas sensor. At an operating temperature of 140 °C, the sensor exhibits outstanding performance toward 10 ppm TEA, including a high response value (64.54), fast response/recovery times (49/108 s), excellent selectivity, and good long-term stability over 32 days. Material characterization reveals that the heterojunction formed between POMOF and ZnO significantly increases the number of active sites on the material surface. Furthermore, combining density functional theory (DFT) calculations, current-voltage (I-V) measurements, and in-situ Fourier transform infrared (FTIR) analysis, the underlying gas-sensing mechanism is systematically and comprehensively discussed. The practical applicability of the sensor is demonstrated by its successful detection of TEA released from crucian carp during room-temperature storage, highlighting its potential for rapid assessment of fish freshness. This work presents a novel approach for designing high-performance gas sensors based on POMOF-ZnO heterojunctions for fish freshness evaluation.

Effect of Various Thermal Treatments on Erythromycin Residues and Degradation Products in Turbot Fish Meat: Implications for Food Safety

Haonan Jiang,Rong Liu,Xuhui Huang,Lei Qin

Erythromycin, a widely utilized antibiotic in aquaculture, poses potential food safety risks through residues in fish products. However, research on the effects of thermal processing on its degradation remains limited. This study developed a sensitive detection method using UHPLC-Q-Exactive HF-X to quantify erythromycin and its degradation products, investigating influences of various thermal treatments and predicting additional degradants. Turbot meat samples spiked with erythromycin were processed via roasting, microwaving, deep frying, and boiling. Post-treatment degradants were identified, with potential metabolites forecasted through structural analysis. The results indicated that erythromycin rapidly degrades under all thermal treatments, with boiling and roasting promoting substantial formation of dehydration products (anhydroerythromycin A and erythromycin A enol ether). The content of N-demethylerythromycin A remained consistently low and stable (<6.67 ng/g). Additionally, five additional erythromycin degradants were screened out. The degradation pathways and product profiles of erythromycin varied depending on the thermal treatment, indicating that thermal processing does not eliminate erythromycin residues but rather transforms them into other substances. This study revealed the specific degradation pattern of erythromycin during thermal processing of fish meat, providing scientific evidence for identifying which harmful substances require priority monitoring in processed fish products, thereby enhancing the level of food safety assessment.

Active Chitosan Films Incorporating a β-Cyclodextrin–Curcumin Inclusion Complex: Enhanced Film Performance and Refrigerated Preservation of Fish Fillets

Anxing Wang,Hao Li,Qing Zhang,Yu Zhang,Yuhang Wu,Deshui Yu,Xing Fang,Guojun Yuan

Achieving stable dispersion and sustained antibacterial activity of natural bioactive compounds in bio-based packaging remains challenging. In this study, chitosan (CS) films incorporating a β-cyclodextrin–curcumin inclusion complex (Cur/β-CD) were developed to improve film properties and the refrigerated preservation of sea bass. The CS/Cur/β-CD films were prepared by one-step solution casting without intermediate isolation or purification. The inclusion conditions were optimized, and the resulting films were evaluated in terms of tensile strength (TS), elongation at break (EAB), and water vapor permeability (WVP). Among the tested formulations, the film prepared at a Cur:β-CD ratio of 1:1, 40 °C, and 1 h (1:1 40 °C 1 h) showed the best overall performance in TS, EAB, and WVP. It was therefore selected for subsequent structural characterization, antibacterial evaluation, and preservation testing. The 1:1 40 °C 1 h film exhibited a 156% increase in tensile strength and a 28.5% decrease in water vapor permeability compared with the neat CS film. The composite film exhibited measurable diffusion-based antibacterial activity againstEscherichia coli(E. coli) andStaphylococcus aureus(S. aureus). During the 8 d refrigerated storage period, the film suppressed total viable counts (TVC), slowed the increase in pH, and retarded total volatile basic nitrogen (TVB-N) accumulation, thereby maintaining acceptable microbiological quality throughout the observation period. Compared with the unwrapped, PE, and CS-film control groups, the treated samples showed better preservation performance over the tested storage period. Overall, the incorporation of Cur/β-CD provides a simple strategy for improving the mechanical strength, moisture barrier properties, antibacterial activity, and preservation performance of CS films during refrigerated storage, highlighting their potential for active packaging of chilled aquatic products.

Chito-oligosaccharide-reinforced thermally cross-linked fish skin gelatin films with silver nanoparticles: Enhanced properties and accelerated wound healing

Qinghao Zhen,Yuanyuan Ding,Xing Guo,Yiwen Lu,Dongjian Shi,Jinghua Chen,Xi Chen

Fish skin gelatin (FG) has been widely used in medical dressings owing to its excellent biocompatibility. Nevertheless, conventional gelatin-based dressings usually rely on chemical cross-linking agents to enhance stability, which inevitably brings potential risks of toxic residues. To address this issue, a green thermal cross-linking strategy was employed to covalently stabilize FG films, providing a safer, more economical, and environmentally friendly route without chemical cross-linkers. Synergistic incorporation of chito-oligosaccharide (COS) and silver nanoparticles (AgNPs) significantly improved mechanical strength and antibacterial activity of the materials. Thermal cross-linking at 180 °C for 20 min endowed the FG film with optimal structural stability and mechanical properties. Notably, the thermally cross-linked FG/COS film with FG: COS mass ratio of 20:1 (FG/COS-5) achieved the best tensile performance. The final composite dressing (AgNPs@FG/COS-5) showed strong antibacterial activity against Escherichia coli and Staphylococcus aureus , while demonstrating low cytotoxicity and superior biocompatibility. In a rat wound healing model, AgNPs@FG/COS-5 significantly accelerated early-stage wound closure within 3 days, outperforming Tegaderm™ and FG dressings. These results demonstrate that the green thermal cross-linking strategy combined with rational component design endows composite dressing with enhanced stability, antibacterial activity, and rapid wound repair efficacy, suggesting its great promise for clinical tissue repair applications.

Highly Stable Hydrogen Peroxide Biosensor Based on Gelatin-Hierarchical Porous Carbon Obtained from Fish Scales Modified Glassy Carbon Electrode

Yunling Ge,Yulin Wang,Mingjia Wang,Huali Wang,Yaqin Huang

Biosensors for continuous application are becoming more important, and long-life enzyme electrodes have not been commercialized due to the lack of stability of its components. A highly stable biosensor for hydrogen peroxide (H 2 O 2 ) based on gelatin-hierarchical porous carbon obtained from fish scales has been developed in this work. Catalase (Cat) immobilized on gelatin-hierarchical porous carbon was realized by the adsorption-crosslinked, here glutaraldehyde (GAD) was used as cross-linking agent. The surface morphologies of modified glass carbon electrodes(GCE) were characterized by scanning electron microscopy (SEM), and the electrochemical behaviors of the biosensor were characterized by cyclic voltammogram (CV) and electrochemical impedance spectroscopy (EIS). This biosensor exhibited a linear range from 0.2 to 10.7 mM with a correlation coefficient (R 2 ) of 0.999, a low detection limit of 0.7 μM and a sensitivity of 74.69 μAmM -1 cm -2 . Especially, the stability of the as-prepared biosensor was satisfied at room temperature, which means there was almost no change in the performance even after 100 continuous scans, and the storage stability is over 30 days with retention of 98.7% activity only.
