Hydrazine and its derivatives are well-known environmental hazards and biological carcinogens; therefore, there is a great need for a powerful workflow solution for protecting the public from unexpected exposure to toxic contaminants. Recently, functional surface-enhanced Raman scattering (SERS) exhibits enormous benefits in sensing trace biochemical substances due to its fingerprint-like identification of individual molecules, making it an ideal method for detecting and quantifying hydrazine. Herein, for the first time, we integrated the orthogonal chemical reporter strategy with SERS to build an intelligent hydrazine detection platform (orthogonal chemical SERS, ocSERS), in which 4-mercaptobenzaldehyde was incorporated on a nanoimprinted gold nanopillar array, which acted as an orthogonal coupling partner of hydrazine to form Raman active benzaldehyde hydrazone, allowing for sensitively detecting hydrazine with a detection limit of 10–13 M in complex circumstances. Particularly, ocSERS could effectively identify the carcinogen N-nitrosodimethylamine (NDMA) after its reduction to dimethylhydrazine (UDMH), enabling ultrasensitive detection of UDMH (10–13 M). Importantly, ocSERS could not only monitor elevated levels of NDMA in ranitidine due to improper storage but also quantify NDMA in urine and blood after oral administration of NDMA-containing drugs, thereby preventing NDMA overexposure. Therefore, ocSERS represents the first click SERS sensor and may open up a new analytical field.

A green and reliable seed-mediated electroless deposition method using the Sargentgloryvine stem extract as reducing and capping agent is explored to prepare silver nanofilms (Ag NFs) as surface-enhanced Raman scattering (SERS) active substrates. The average particle size of Ag NFs can be facilely controlled only by adjusting the amount of AgNO 3 in the growth solution. Furthermore, the SERS properties of Ag NFs are established experimentally and their Raman enhancement mechanism is revealed by means of the scanning electron microscopy and the finite-difference time-domain simulation. It is found that high surface coverage and slight surface agglomeration of Ag NFs are beneficial for the amplification of Raman signal. Optimized Ag NFs display superior sensitivity towards Rhodamine 6G molecules and good reproducibility, indicating potential applications in SERS-based detection and analysis. Meanwhile, the proposed method to obtain Ag NFs provide a promising route for the preparation of other metal nanofilm.

For real application, it is an urgent demand to fabricate stable and flexible surface-enhanced Raman scattering (SERS) substrates with high enhancement factors in a large-scale and facile way. Herein, by using the electrospinning technique, a hydrophobic and flexible poly(styrene-co-butadiene) (SB) fibrous membrane is obtained, which is beneficial for modification of silver nanoparticles (Ag NPs) colloid in a small region and then formation of more “hot spots” by drying; the final SERS substrate is designated as Ag/SB. Hydrophobic Ag/SB can efficiently capture heterocyclic molecules into the vicinity of hot spots of Ag NPs. Such Ag/SB films are used to quantitatively detect trace triazophos residue on fruit peels or in the juice, and the limit of detection (LOD) of 2.5 × 10−8M is achieved. Ag/SB films possess a capability to resist heat. As a case, 6-mercaptopurine (6MP) that just barely dissolves in 90 °C water is picked for conducting Ag/SB-film-based experiments. Keywords:electrospun;Ag NPs/poly(styrene-co-butadiene);SERS;triazophos

A new class of flowerlike gold mesostructure in high yield is successfully synthesized through a facile one-step route using ascorbic acid as a reducing agent of gold salt with cetyltrimethylammonium chloride (CTAC) as surfactant. The as-prepared Au particles have spherical profiles with an averaged diameter of 770 ± 50 nm, but showing a highly rough surface consisting of many irregular and randomly arranged protrusions. The Au mesoflowers exhibit strong surface-enhanced effects and near-infrared absorption which were utilized in the design of efficient surface-enhanced Raman scattering (SERS) tags as immunosensors for immunoassay with improved sensitivity. The experimental results indicate that a good linear relationship is found between the peak intensity at 1071 cm–1 and the logarithm of H-IgG concentration in the range between 1 ng/mL and 1 fg/mL, and the limit of detection (LOD) is 1 fg/mL.

Liquid interfacial plasmonic platform is emerging for new sensors, catalysis, and tunable optical devices, but also promises an alternative for practical applications of surface-enhanced Raman spectroscopy (SERS). Here we show that vigorous mixing of chloroform with citrate-capped gold nanorod sols triggers the rapid self-assembly of three-dimensional plasmonic arrays at the chloroform/water ( O / W ) interface and produces a self-healing metal liquid-like brilliant golden droplet. The O phase itself generates stable SERS fingerprints and is a good homogeneous internal standard for quantitative analysis. This platform presents reversible O / W encasing in a common cuvette determined just by surface wettability of the container. Both O -in- W and W -in- O platforms exhibit excellent SERS sensitivity and reproducibility for different analytes by the use of a portable Raman device. It paves the way toward a practical and quantitative liquid-state SERS analyzer, likened to a simple UV–Vis spectrometer, that is far superior to typical solid substrate-based or nanoparticle sol-based analysis.

Background Reliable quantification of nitrite in complex food matrices using surface-enhanced Raman scattering (SERS) remains challenging due to signal drift and SERS substrate variability. Conventional internal standard (IS) strategies often occupy plasmonic hot spots, limiting analyte accessibility and compromising detection performance. Results Herein, we report a size-complementary modular self-calibrated co-assembled (SCCA) SERS platform that integrates large gold nanoparticles (LAuNPs) with Au@Prussian blue (PB) core-shell nanospheres via interfacial self-assembly. The Au@PB nanospheres physically occupy the interparticle gaps of LAuNP arrays, providing both structural uniformity and a built-in Raman reference band at 2128 cm −1 for ratiometric self-calibration without competing for plasmonic hot spots and compromising SERS sensitivity. The resulting SCCA substrate exhibits dense electromagnetic hot spots and good reproducibility, achieving quantitative nitrite detection through an acid-promoted S-nitrosation reaction with 2-thiobarbituric acid that generates a characteristic Raman band at 685 cm −1 . The method delivers a linear range of 0.15–2 mg/L, a limit of detection of 0.0368 mg/L, and excellent anti-interference tolerance in real food samples including cured fish, sausage, and pickled cucumber. The substrate retains 91.51% signal intensity after 60 days, confirming outstanding stability. Significance and novelty This work presents a universal strategy for constructing self-calibrated SERS substrates by spatially decoupling calibration elements from plasmonic hotspots. The SCCA design eliminates the intrinsic conflict between sensitivity and quantitative reliability, offering a modular and scalable approach for ratiometric SERS analysis.

A significant hurdle for the clinical translation of in vivo surface-enhanced Raman scattering (SERS) bioimaging is the scarcity of high-brightness near-infrared (NIR) contrast agents. To address this challenge, we develop such agents through the strategic optimization of gold nanorods (AuNRs) as plasmonic substrates and the selection of resonant Raman reporters. We quantitatively characterize the effective differential SERS cross-section of nanoparticles comprising AuNRs spectrally encoded with various Raman reporters. Our findings establish that the spectral overlap between the AuNR plasmon resonance, reporter absorption, and laser excitation wavelength is a critical parameter for achieving high brightness. Guided by this principle, we engineer NIR-I and NIR-II SERS agents using optimized AuNRs and selected resonant reporters, encapsulated within cell membranes derived from 4T1 breast cancer cells. These agents exhibit superior brightness and specific homotypic targeting toward 4T1 cells. In vivo studies in mice demonstrate their capability for precise tumor margin delineation and sensitive detection of microtumors (<100 µm) via NIR-I and NIR-II SERS imaging. Furthermore, the agents enable image-guided surgical resection of orthotopic 4T1 tumors, achieving complete tumor removal and significantly improving surgical outcomes. This work provides a foundational strategy for designing high-brightness NIR SERS contrast agents for advanced in vivo molecular imaging applications.

An Au–AgCl Janus Nanoparticle (JNP) is an emerging plasmonic nanostructure that has attracted attention in the field of surface-enhanced Raman scattering (SERS). However, because of the large lattice mismatch between Au nanoparticles (NPs) and AgCl NPs, the instability of the AgCl NPs leads to very complex nucleation and growth kinetics, and the synthesis of Au–AgCl JNPs has challenges. In this work, we report a method for the synthesis of Au–AgCl JNPs by using Phenolic compounds and photochemical reduction. Phenolic compounds can reduce the adsorption energy between AgCl NPs and Au NPs during the growth process, resulting in the asymmetric deposition of Au atoms on the AgCl NPs surface. By utilizing blue-light irradiation to modulate the Ag atom sites on the AgCl NPs surface, the number of Au domains within the Au–AgCl JNPs can controllably change. The SERS detection limit for single Au–AgCl JNPs is determined to be as low as 10–10 M for Rhodamine 6G (R6G) molecules, outperforming pure Au NPs by 2–3 orders of magnitude. The enhancement is attributed to a synergistic mechanism involving interparticle gap effects that generate intense electromagnetic “hot spots” and efficient electron transfer between Au NPs and AgCl NPs. This work not only demonstrates a scalable strategy for fabricating lattice-mismatched Janus systems but also offers a framework for the rational design of heteronanostructures for SERS.

The rapid fabrication of low-cost surface-enhanced Raman scattering (SERS) substrates is highly desirable for chemical and biological sensing. Existing customized SERS substrates, such as Au or Ag nanostructures produced by physical deposition, frequently involve complex fabrication routes, which limits the scalability of SERS devices. Here, we present the hexagonal close-packed plasmonic superlattices as an efficient, low-cost and applicable SERS platform, fabricated by scalable seed-mediated growth and interfacial self-assembly methods. We systematically compared Ag, Au, and Au@Ag nanospheres (NSs) of different sizes and demonstrated that the plasmonic superlattices made by 55 nm Au@Ag NSs exhibit the strongest Raman response, highest sensitivity, lowest detection limit, good spatial uniformity, and broad applicability. Simulations and Raman mapping experiments further confirm that Au@Ag NSs achieve an optimal balance between hotspot density and plasmonic field intensity, allowing for direct identification and quantification of diverse biochemical targets.

Bisphenol A (BPA), a pervasive endocrine disruptor in plastics, poses serious risks to human health and the environment. Surface-enhanced Raman spectroscopy (SERS) provides high sensitivity and label-free detection. However, conventional substrates often exhibit heterogeneous hotspot distribution and lack molecular selectivity, limiting their reliability in complex matrices. Herein, combining the molecular imprinting specific recognition capability with the electrospun nanofiber large surface area, we present a molecularly imprinted nanofiber SERS substrate for the rapid, sensitive, and selective BPA detection. Using BPA as the template, the substrate achieved rapid adsorption equilibrium within 5 min, high capacity, and strong selectivity. Response surface optimization identified the optimal fabrication parameters, yielding a normalized enhancement factor of 4.89. Coupled with partial least squares regression, the system enabled accurate quantification of BPA over the range of 2 × 10 − 8 -6 × 10 − 7 mol/L, with excellent linearity (R² = 0.997) and prediction error below 2 mg/L. This strategy establishes a robust and generalizable platform for highly sensitive, selective, and on-site detection of BPA, offering broad potential in food safety, environmental monitoring, and public health.

This study addresses critical concerns in epilepsy management regarding generic drug substitution and the bioequivalence risks associated with switching antiepileptic drugs (AEDs). For patients undergoing long-term treatment, precise brand identification of levetiracetam (LEV) holds significant clinical value to ensure therapeutic consistency and safety. To meet this need, we developed a qualitative brand discrimination method based on surface-enhanced Raman spectroscopy (SERS) combined with machine learning. Silver sol was used as the SERS substrate, with potassium iodide as an aggregating agent and Rhodamine 6G (R6G) as an optimized probe. After validating LEV's characteristic Raman peaks through integrated theoretical and experimental approaches, spectra from three commercial LEV brands were analyzed using principal component analysis (PCA) and support vector machine (SVM) classification. The model achieved 100% accuracy, precision, and sensitivity in 10-fold stratified cross-validation, demonstrating robust brand-discrimination capability. Unlike conventional quantitative assays that verify API content but cannot distinguish between different manufacturers, this SERS-based approach detects formulation-specific differencessuch as variations in crystal form and preparation processthrough unique spectral fingerprints. It thus provides a rapid, label-independent tool for brand authentication, batch consistency evaluation, and on-site pharmaceutical screening, effectively complementing pharmacopeial methods. This work supports medication safety and offers a reliable strategy for drug quality control and market surveillance, particularly in preventing counterfeit drugs and ensuring formulation stability in long-term epilepsy therapy.
