Biofilm-Induced Corrosion Inhibition of Q235 Carbon Steel by Tenacibaculum mesophilum D-6 and Bacillus sp. Y-6

Xiaoxi Ruan,Linlin Yang,Yan Wang,Yizhe Dong,Dake Xu,Mingxing Zhang

The corrosion of carbon steel causes dramatic economic losses each year. Since conventional corrosion prevention approaches may cause pollution problems to the environment, ecofriendly and effective corrosion approaches are desired. Microbiologically influenced corrosion inhibition (MICI) has been reported as a sustainable corrosion prevention method. This work aims to evaluate the corrosion inhibition effect of two bacterial strains,Tenacibaculum mesophilumD-6 andBacillussp. Y-6 by choosing Q235 carbon steel as a model system. Scanning electron microscopy (SEM), confocal laser scanning microscopy (CLSM) and a series of electrochemical techniques were applied to study the corrosion prevention effect. The electrochemical and pitting results indicated thatT. mesophilumD-6 displayed a better corrosion protection effect.T. mesophilumD-6 formed a denser and thicker biofilm on the Q235 surface thanBacillussp. Y-6. The maximum thickness of theT. mesophilumD-6 biofilms was 11.6 ± 0.7 μm, which is about twice as thick than that ofBacillussp. Y-6. The corrosion prevention mechanism was ascribed to the formation of biofilms as a barrier to block corrosive agents such as O2. This study provides a theoretical foundation for the application of biofilms as green and effective corrosion inhibitors for carbon steel.Keywords:Q235 carbon steel;Tenacibaculum mesophilumD-6;Bacillussp. The ecotoxicological effects of chromium (III) oxide nanoparticles to Chlorella sp.: perspective from the physiological and transcriptional responses

Liu Huan-Huan,Yang Lei,Li Xiao-Tong,Shi Hui,Guo Lin-Kai,Tu Li-Xin,Wang Jia,Li Yan-Li

Extensive application of nanomaterials enlarges its concentrations in the aquatic environments and poses a threat to algae. This study comprehensively analyzed the physiological and transcriptional responses of Chlorella sp. after being exposed to chromium (III) oxide nanoparticles (nCr 2 O 3 ). The nCr 2 O 3 at 0–100 mg/L presented adverse effects on cell growth (96 h EC 50  = 16.3 mg/L), decreasing the photosynthetic pigment concentrations and photosynthetic activity. Moreover, more extracellular polymeric substances (EPS), especially polysaccharides in soluble EPS, were produced in algae cell, which mitigated the damage of nCr 2 O 3 to cells. However, with the increase of nCr 2 O 3 doses, the EPS protective responses were exhausted, accompanied by toxicity in the form of organelle damage and metabolic disturbance. The enhanced acute toxicity was closely related to the physical contact of nCr 2 O 3 with cells, oxidative stress, and genotoxicity. Firstly, large amounts of nCr 2 O 3 aggregated around and were attached to cells, causing physical damage. Then, the intracellular reactive oxygen species and malondialdehyde levels were significantly increased that led to lipid peroxidation, especially at 50–100 mg/L nCr 2 O 3 . Finally, the transcriptomic analysis further revealed that the transcription of ribosome, glutamine, and thiamine metabolism-related genes were impaired under 20 mg/L nCr 2 O 3 , suggesting nCr 2 O 3 inhibited algal cell growth through metabolism, cell defense, and repair, etc. Graphical Abstract

Enhancing Biodegradation of Pyridine with Trehalose Lipid in Rhodococcus pyridinivorans sp. Strain HR-1-Inoculated Microbial Fuel Cell

Peng Cheng,Muhammad Usman,Muhammad Arslan,Huanqing Sun,Li Zhou,Mohamed Gamal El-Din

A Gram-positive exoelectrogenRhodococcus pyridinivoranssp. strain HR-1 was cultivated from leachate-fed microbial fuel cell (MFC) to study the biodegradation effect of pyridine. In the comparison with mixed cultured MFC, HR-1 presented a remarkable electrical capacity with a maximum output of 4.33 W/m3under 30 °C in neutral anolyte with 1 g/L acetate as a substrate. Further, HR-1 demonstrated the environmental resistance as a Gram-positive strain. Microbial metabolism was evident at pH between 5–9 and temperature in the range of 20–40 °C, whereas optimal condition for pyridine degradation was observed at 30 °C. This is the first study reporting the degradation of pyridine in the bio-electrochemical system that achieved a 42% ± 5% degradation rate in a full operation cycle at 2 g/L of the concentration. Considering the nonnegligible internal resistance of HR-1-inoculated MFC, trehalose lipid was also introduced as a bio-surfactant to reduce the charge transfer obstacle between the microbes and the electrodes. The surface morphology illustrated that the strain had a plump shape with a high specific area. Accordingly, bio-surfactant addition promoted the anode biomass (1.2 ± 0.1 mg/cm2to 1.7 ± 0.2 mg/cm2) and achieved a higher degradation rate (68% ± 4%). The feasibility of electrochemical disposal on pyridine and eminent adaptability of strain sp. HR-1 as a Gram-positive exoelectrogen makes MFC a practical approach for real application. Keywords:microbial fuel cells;gram-positive exoelectrogen;pyridine;trehalose lipid;Rhodococcus pyridinivorans

Efficient degradation and decolorization of triphenylmethane dyes by Serratia sp. WKD under extreme environmental conditions and the mechanism

Linna Du,Huan Wu,Gang Li,Yi Wei,Fei Wang,Ligen Xu,Xinjiao Dong

Microbial degradation of dyes is fundamental and essential for understanding their environmental fate . In this study, a new bacterial strain with efficient degrading capability of triphenylmethane (TPM) dyes named WKD was isolated and identified as the genus Serratia sp. by 16S rDNA analysis . The decolorization of TPM dyes by the strain especially under extreme environmental conditions were studied. The results showed that the strain could completely decolorize Malachite Green (MG) and Crystal Violet (CV) at pH values ranging from 3.0 to 10.0 and at temperatures ranging from 20 °C to 45 °C in a 12 h of incubation. The strain still showed an excellent decolorizing capability even at low (5 °C) and high (60 °C) temperatures. Moreover, the enzymatic analysis indicated that MnP, NADH-DCIP reductase, MG reductase, and CV reductase might be involved in TPM dyes biodegradation. Besides, the biodegradation pathways were proposed based on UV–visible, FTIR , and LC-Q-TOF-MS analyses. Results indicated that degradation of MG by the strain was mainly regulated by N-demethylation and hydroxylation pathways, while degradation of CV was only by the N-demethylation pathway. Therefore, Serratia sp. strain WKD is expected to have a broad application prospect for the bioremediation of TPM dyes, especially in extreme environments.

Characterization of a new chlorimuron-ethyl-degrading strain Cedecea sp. LAM2020 and biodegradation pathway revealed by multiomics analysis

Qingyun Ma,Xiaoyan Han,Jinlong Song,Jie Wang,Qingqing Li,Rebecca E. Parales,Lin Li,Zhiyong Ruan

The widespread use of the herbicide chlorimuron-methyl is hazard to rotational crops and causes soil degradation problems. Biodegradation is considered a promising way for removing herbicide residues from the environment. Here, a new isolated strain, Cedecea sp . LAM2020, enabled complete degradation of 100 mg/L chlorimuron-methyl within five days. Transcriptome analysis revealed that ABC transporters, atrazine degradation and purine metabolism were enriched in the KEGG pathway. Integrating GO and KEGG classification with related reports, we predict that carboxylesterases are involved in the biodegradation of chlorimuron-methyl by LAM2020. Heterologous expression of the carboxylesterase gene carH showed 26.67% degradation of 50 mg/L chlorimuron-methyl within 6 h. The intracellular potential biological response and extracellular degradation process of chlorimuron-ethyl were analyzed by the nontarget metabolomic and mass spectrometry respectively, and the biodegradation characteristics and complete mineralization pathway was revealed. The cleavage of the sulfonylurea bridge and the ester bond achieved the first step in the degradation of chlorimuron-methyl. Together, these results reveal the presence of acidolysis and enzymatic degradation of chlorimuron-methyl by strain LAM2020. Hydroponic corn experiment showed that the addition of strain LAM2020 alleviated the toxic effects of chlorimuron-ethyl on the plants. Collectively, strain LAM2020 may be a promising microbial agent for plants chlorimuron-ethyl detoxification and soil biofertilizer .

High PCBs mineralization capability of a resuscitated strain Bacillus sp. LS1 and its survival in PCB-contaminated soil

Zhen Han,Qihua Lin,Shusheng Zhang,Xinru Zhou,Si Li,Faqian Sun,Chaofeng Shen,Xiaomei Su

Polychlorinated biphenyl (PCB)-degrading strains resuscitated by resuscitation promoting factor (Rpf) enlarged pure degraders to screen effective bio-inoculants for soil bioaugmentation . In this study, whole-genome analysis and PCB-degrading performance of a resuscitated strain LS1 were investigated. Importantly, the persistence and the physiological response of soil-inoculated LS1 were checked. The results indicate that the Bacillus sp. strain LS1 possessed the potential to degrade polycyclic aromatic compounds. LS1 exhibited better performance in degrading PCBs 18 and 52, but lower PCB 77 degradation capability. At PCBs concentration of 10 mg/L, the degradation efficiencies of PCBs 18, 52 and 77 within 96 h were 62.8 %, 59.6 % and 39.8 %, respectively. Combined the bph genes and metabolites detected, as well as the genes found in the genome, the abilities of LS1 for oxidative dehalogenation and mineralization of PCBs via HOPDA-benzoate-protocatechuate- β -ketoadipate pathway were determined. Notably, LS1 can still maintain survival and culturable state after inoculation into PCB-contaminated soil for 70 days. This is the first report to demonstrate the fate of resuscitated strain when used as soil bio-inoculant, which revealed the necessity and feasibility of using resuscitated strains to enhance bioremediation of PCB-contaminated soils.

Reclassification of Enterobacter sp. FY-07 as Kosakonia oryzendophytica FY-07 and Its Potential to Promote Plant Growth

Ge Gao,Yan Zhang,Shaofang Niu,Yu Chen,Shaojing Wang,Nusratgul Anwar,Shuai Chen,Guoqiang Li,Ting Ma

Precise classification of bacteria facilitates prediction of their ecological niche. The genusEnterobacterincludes pathogens of plants and animals but also beneficial bacteria that may require reclassification. Here, we propose reclassification ofEnterobacterFY-07 (FY-07), a strain that has many plant-growth-promoting traits and produces bacterial cellulose (BC), to theKosakoniagenera. To re-examine the taxonomic position of FY-07, a polyphasic approach including 16S rRNA gene sequence analysis, ATP synthase β subunit (atpD) gene sequence analysis, DNA gyrase (gyrB) gene sequence analysis, initiation translation factor 2 (infB) gene sequence analysis, RNA polymerase β subunit (rpoB) gene sequence analysis, determination of DNA G + C content, average nucleotide identity based on BLAST, in silico DNA–DNA hybridization and analysis of phenotypic features was applied. This polyphasic analysis suggested thatEnterobactersp. FY-07 should be reclassified asKosakonia oryzendophyticaFY-07. In addition, the potential of FY-07 to promote plant growth was also investigated by detecting related traits and the colonization of FY-07 in rice roots.Keywords:Enterobacter;Kosakonia;reclassification;FY-07;promote plant growth

Antifungal activity of volatile compounds generated by endophytic fungi Sarocladium brachiariae HND5 against Fusarium oxysporum f. sp. cubense

Yang Yang,Yipeng Chen,Jimiao Cai,Xianbao Liu,Guixiu Huang

The soil-born filamentous fungal pathogen Fusarium oxysporum f. sp. cubense (FOC), which causes vascular wilt disease in banana plants, is one of the most economically important Fusarium species. Biocontrol using endophytic microorganisms is among the most effective methods for controlling banana Fusarium wilt. In this study, volatile organic compounds (VOCs) showed strong antifungal activity against FOC. Seventeen compounds were identified from the VOCs produced by endophytic fungi Sarocladium brachiariae HND5, and three (2-methoxy-4-vinylphenol, 3,4-dimethoxystyrol and caryophyllene) showed antifungal activity against FOC with 50% effective concentrations of 36, 60 and 2900 μL/L headspace, respectively. Transmission electron microscopy (TEM) and double fluorescence staining revealed that 2-methoxy-4-vinylphenol and 3,4-dimethoxystyrol damaged the plasma membranes, resulting in cell death. 3,4-dimethoxystyrol also could induce expression of chitin synthases genes and altered the cell walls of FOC hyphae. Dichloro-dihydro-fluorescein diacetate staining indicated the caryophyllene induced accumulation of reactive oxygen species (ROS) in FOC hyphae. FOC secondary metabolism also responded to active VOC challenge by producing less fusaric acid and expressions of genes related to fusaric acid production were interrupted at sublethal concentrations. These findings indicate the potential of S . brachiariae HND5 as a biocontrol agent against FOC and the antifungal VOCs as fumigants.

Efficient production of biosurfactant surfactin by a newly isolated Bacillus subtilis (sp.) 50499 strain from oil-contaminated soil

Yulu Du,Yue Wang,Tianyou Cui,Lianpeng Ge,Fan Yu,Mengqian Zhao,Qihang Xu,Mingjie Jin,Bo Chen,Xuwei Long

Biosurfactant surfactin has a high market value and broad application prospects in the food industry and various industrial fields. However, its production efficiency limits its suitability for real applications. In this study, a new Bacillus subtilis (sp.) 50499 strain was isolated from crude oil-contaminated soil and used to efficiently produce surfactin. The initial surfactin titer was 2.25 g/L after three days of fermentation. Adding 2 g/L of L -Leu significantly improved the titer to over 5.5 g/L after optimizing the culture medium at 37 °C. However, adding D -Leu had the opposite effect on surfactin production , and the surfactin titer dropped to below 0.2 g/L. Furthermore, the product of this strain was analyzed to confirm the identity of surfactin. The surfactin obtained in the present study reduced the surface tension of water to 28.6 mN/m at a critical micelle concentration of 20 mg/L. The emulsification index against n -hexane, corn , and crude oil was as high as over 90%. Hence, the present study provides a newly over-producing strain for surfactin production.

Enantioselective resolution of (±)-1-phenylethyl acetate using the immobilized extracellular proteases from deep-sea Bacillus sp. DL-1

Lu Dong,Shujuan Qi,Jianwei Jia,Yun Zhang,Yunfeng Hu

Bacillus sp. DL-1 was isolated from the deep sea of the Western Pacific Ocean and behaved very good resistance to NaCl. The extracellular proteases of Bacillus sp. DL-1 were found to exhibit excellent enantioselectivity for the kinetic resolution of (±)-1-phenylethyl acetate. To improve the stability of the enzyme, the immobilized extracellular proteases were preparated by using 60 g kieselguhr and 1-L crude fermentation broth containing extracellular proteases of Bacillus sp. DL-1, shaking at 25 °C, 200 r/min for 10 h. Every gram of kieselguhr adsorbed 25.7 mg extracellular protease and the enzymatic activity recovery was 79.86%. The immobilized proteases preserved about 35.8% of its activity after 6 repeated uses, which were also used as biocatalyst to asymmetrically hydrolyse (±)-1-phenylethyl acetate for the preparation of (R)-1-phenylethanol and (S)-1-phenylethyl acetate with high optical purities. The effects of pH, temperature, enzyme concentration, substrate concentration, reaction time and additives (metal ions/surfactants) on the resolution were investigated by single factor experiments. Under the optimal reaction conditions (10 mM (±)-1-phenylethyl acetate, 40 mg/mL immobilized extracellular proteases, pH 7.5 (Tris-HCl), 5% (v/v) methanol and 45 °C for 2 h), (R)-1-phenylethanol was generated with the e.e.p being > 97%, and the yield being 53%, respectively. Analogously, under the optimal reaction conditions (10-mM (±)-1-phenylethyl acetate, 360 mg/mL immobilized extracellular proteases, pH 6.0 (PB), 5% (v/v) DMSO and 35 °C for 1.5 h), (S)-1-phenylethyl acetate was generated with the e.e.s being over 99% and the yield being 79%, respectively. Compared with the extracellular proteases from Bacillus sp. DL-2, the immobilized extracellular proteases from Bacillus sp. DL-1 exhibited higher hydrolytic activity and could asymmetrically hydrolyse (±)-1-phenylethyl acetate by using higher substrate concentrations, shorter reaction times to obtain higher yields. Notably, the extracellular proteases of Bacillus sp. DL-1 were demonstrated to behave the same enantio-preference as those of most other reported esterases/lipases. Proteases from deep-sea Bacillus sp. DL-1 are promising biocatalysts for the synthesis of valuable chiral chemicals.

Impacts of surface wettability and roughness of styrene-acrylic resin films on adhesion behavior of microalgae Chlorella sp

Jing Tang,Bin Liu,Lili Gao,Wenqing Wang,Tianzhong Liu,Ge Su

Biofilm attached cultivation is a promising method for efficient production of microalgae. Determining the surface property index to select an appropriate substrate benefiting the algae adhesion and biofilm formation is very important for the cultivation method. This work focused on elucidating and quantifying the influence of surface wettability and roughness of substrate on Chlorella vulgaris adhesion. Firstly, surface modified styrene-acrylic (SA) resin films by adding different dosage of perfluoroalkyl ethyl acrylate (FM) were prepared. Property characterization shows that the surface contact angle in water, formamide and diiodomethane of FM modified SA films is significantly associated with the FM dosage, while the other surface properties including zeta potential, surface potential and surface roughness have insignificant difference. The calculated surface free energy parameters show that the SA films belong to the non-polar substrata. A well quantitative correlation that the adhesion capacity of C. vulgaris linearly declines with the increase of water contact angle was obtained. And a near linear relationship between the adhesion capacity and the surface free energy (γ), or the cohesion free energy ( Δ G c o h ) ΔGcoh) was also observed. Secondly, the surface roughness solely changed SA films were prepared by replicating the morphology of stainless steel sieves through the PDMS template method. The patterned SA films have alternately arranged rectangular “valleys” and “ridges”. A well linear correlation between the microalgae adhesion capacity and the surface roughness was also obtained.

Characterization of a New Chitosanase from a Marine Bacillus sp. and the Anti-Oxidant Activity of Its Hydrolysate

Chunrui Ma,Xiao Li,Kun Yang,Shangyong Li

Chitooligosaccharide (COS) has been recognized to exhibit efficient anti-oxidant activity. Enzymatic hydrolysis using chitosanases can retain all the amino and hydroxyl groups of chitosan, which are necessary for its activity. In this study, a new chitosanase encoding gene, csnQ, was cloned from the marineBacillussp. Q1098 and expressed inEscherichia coli. The recombinant chitosanase, CsnQ, showed maximal activity at pH 5.31 and 60 °C. Determination of CsnQ pH-stability showed that CsnQ could retain more than 50% of its activity over a wide pH, from 3.60 to 9.80. CsnQ is an endo-type chitosanase, yielding chitodisaccharide as the main product. Additionally, in vitro and in vivo analyses indicated that chitodisaccharide possesses much more effective anti-oxidant activity than glucosamine and low molecular weight chitosan (LMW-CS) (~5 kDa). Notably, to our knowledge, this is the first evidence that chitodisaccharide is the minimal COS fragment required for free radical scavenging.Keywords:chitosanase;pH-stability;Bacillussp.
