Bacterial keratitis is one of the most common causes of visual impairment and blindness worldwide, and suffers the risk of drug-resistant infections due to the abuse of antibiotics. Despite its ability to counteract antibiotic resistance by inducing ferroptosis, the application of Fe 2+ is limited by oxidative instability and a solution pH often incompatible with ophthalmic use. Thus, this study designed and synthesized an ascorbate–ferrous iron complex (SA-Fe(II)) using ferrous lactate and sodium ascorbate, followed by comprehensive structural characterization. Results confirmed the formation of coordination bonds between Fe 2+ and sodium ascorbate. The molecular ion peak and characteristic decarboxylated fragment, along with broad and diffuse diffraction peaks in X-ray diffraction patterns, verified the formation of an amorphous metal chelate structure stabilized by dynamic coordination bonds. The SA-Fe(II) complex exhibited potent antibacterial activity, achieving more than 99.99 % killing of Staphylococcus aureus ( S. aureus ) and methicillin-resistant Staphylococcus aureus (MRSA) at 100 μM within 3 h. When incorporated into a sodium hyaluronate eye-drop matrix (SA-Fe(II)-SH), the formulation exhibited excellent ocular biocompatibility, as evidenced by hemolysis rates less than 5 %, more than 95 % viability of human corneal epithelial cells (HCE-T) corneal epithelial cells over 3 days, and no observable tissue inflammation in vivo. Importantly, in a mice model of MRSA keratitis, SA-Fe(II)-SH significantly reduced bacterial load, suppressed the production of inflammatory cytokines (IL-1β, IL-6, TNF-α), promoted corneal repair, and prevented systemic dissemination. This non-antibiotic strategy integrates antibacterial, anti-inflammatory, and pro-healing functions, offering a safe and effective therapeutic approach for the treatment of drug-resistant bacterial keratitis.

The escalation of multidrug-resistant bacterial infections, especially infections caused by methicillin-resistant Staphylococcus aureus (MRSA), underscores the urgent need for novel antimicrobial drugs. Here, we synthesized a series of amphiphilic 2-phenyl-1H-phenanthro[9,10-d]imidazole-antimicrobial peptide (AMP) mimic conjugates (III1–30). Among them, compound III13 exhibited excellent antibacterial activity against G+ bacteria and clinical MRSA isolates (MIC = 0.5–2 μg/mL), high membrane selectivity, and low toxicity. Additionally, compared with traditional clinical antibiotics, III13 demonstrated rapid bactericidal efficacy and was less susceptible to causing bacterial resistance. Mechanistic studies revealed that III13 targets phosphatidylglycerol (PG) on bacterial membranes to disrupt membrane integrity, leading to an increase in intracellular ROS and leakage of proteins and DNA, ultimately causing bacterial cell death. Furthermore, III13 possessed good fluorescence properties with potential for further dynamic monitoring of the antimicrobial process. Notably, III13 showed better in vivo efficacy against MRSA compared to vancomycin, suggesting its potential as a promising candidate for anti-MRSA medication.

This study introduces hyaluronic acid-based (HA) hydrogel microspheres loaded with zinc oxide nanoparticles (ZnO-NPs) for the treatment of infectious bone defects. The microspheres were fabricated using a 3D-printing process, with a formulation consisting of 6 wt% HAD (methacrylated HA), 3 wt% AOHA (AMP-conjugated oxidized HA), 1 % BOHA (phenylboric acid-conjugated HA), 0.5 % photoinitiator, and 0.05 % ZnO-NPs. In vitro , the hydrogel microspheres demonstrated significant antibacterial activity against Staphylococcus aureus , with colony counts and biofilm inhibition assays showing a marked reduction in bacterial growth after 12 and 24 h. The release of antimicrobial peptides (AMPs) was enhanced in acidic conditions and in the presence of hyaluronidase. The microspheres also promoted osteogenic differentiation of bone marrow stromal cells (BMSCs), as evidenced by increased expression of osteogenic markers (ALP, OCN, OPN, and COL-1). In vivo , the hydrogel microspheres were tested in a rat skull defect model, showing significant bone regeneration, improved angiogenesis, and an anti-inflammatory response. These results indicate that ABOHA@ZnO hydrogel microspheres provide a promising strategy for treating infectious bone defects by combining antimicrobial, osteogenic.

Microbial contamination in food can cause foodborne illnesses, particularly methicillin-resistant Staphylococcus aureus (MRSA), which poses a serious threat to human health due to its ability to produce heat-resistant enterotoxins and express the broad-spectrum antibiotic resistance protein penicillin-binding protein 2a (PBP2a). In this study, tea pigments with potent anti- MRSA activity were prepared from discarded tea plant branches and leaves via in vitro enzymatic oxidation. Optimal process conditions obtained through response surface methodology (RSM) were: enzyme substrate ratio of 0.6 U/mg, reaction temperature of 43 °C, and reaction time of 22 h, yielding a production rate of 7.88%. Theaflavins (TFs), thearubigins (TRs), and theabrownins (TBs) isolated and purified via organic solvent extraction and macroporous resin D101 exhibited similar phenolic compound spectral characteristics, with average molecular weights (Mw) of 208, 368, and 16,064 Da, respectively. The minimum inhibitory concentration (MIC) of TFs, TRs, and TBs against MRSA were 156.25, 312.5, and 1250 μg/mL, respectively, demonstrating excellent anti- MRSA activity that was significantly stronger than their inhibitory activity against Escherichia coli . Furthermore, tea pigments exert antibacterial effects through mechanisms including cell wall disruption, alteration of cell membrane permeability, inhibition of biofilm formation, reduction of respiratory chain dehydrogenase activity, and increased levels of intracellular reactive oxygen species in bacteria. Additionally, molecular docking indicates that theaflavins effectively binds to PBP2a, thereby interfering with MRSA growth. Tea pigments as anti- MRSA natural products have great prospects in food industry.

Staphylococcus aureus ( S. aureus ) is a prevalent bacterium that affects patient recovery post-surgery and can cause postoperative complications . Consequently, detecting S. aureus in clinical settings is critical for ensuring patient well-being. In our work, a “visual-colorimetric-photothermal” three-in-one multimodal lateral flow immunoassays (LFIA) was developed for S. aureus detection to meet the growing demand for point-of-care testing (POCT) of bacteria in the field. The sensors relied on Pd/Pt@Ab1 NPs nanoprobes , created by decorating the surface of bimetallic nanoprobes (Pd/Pt NPs) with S. aureus antibodies (Ab1). A visual detection mode was realized by the Pd/Pt@Ab1 NPs being captured on the surface of test line (T line) using the sandwich method, offering a limit of detection (LOD) of 10 3 CFU/mL. Subsequently, Pd/Pt@Ab1 NPs catalyzed the TMB system into oxTMB to achieve colorimetric analysis , covering a detection range from 10 2 to 10 7 CFU/mL. Remarkably, the oxTMB exhibited the photothermal conversion efficiency under near-infrared (NIR) light, leading to temperature variations under 808 nm irradiation, correlating with different bacteria concentrations, with an LOD as low as 4 CFU/mL. The methodology enabled quantitative analysis of bacteria in clinical samples with satisfactory results. The diversity, complementarity, and synergy of the integrated output signals in this multimode LFIA improved the flexibility, utility, and accuracy of the assay, performing well as a POCT platform for various application scenarios.

Rapid detection of foodborne pathogens plays a more and more significant role in the field of public health. In this research, we developed a convenient electrochemical biosensor mediated by filtration-assisted pretreatment and multiple thermostatic enzymes (Multi-TEs) amplification system for the detection of Staphylococcus aureus ( S. aureus ) in complex matrices. Firstly, the designed filtration-assisted pretreatment could simultaneously collect and enrich target bacterium from various actual samples within 5 min. Then, the target bacterial genome template was reacted at 37 °C in Multi-TEs amplification system to obtain specific double stranded products. Finally, the specific double stranded products were fixed on the surface of the electrode and the electrochemical indicators (Methylene blue, MB) was combined for the quantitative one-step electrochemical analysis of S. aureus . This assay has great linear calibration curves with the concentration ranging from 10 1 CFU/mL to 10 6 CFU/mL. The limit of detections (LOD) was defined as 1.28 CFU/mL, which performance is superior to some commercial ELISA kits. Meanwhile, amazingly, it has excellent specificity for S. aureus against other foodborne pathogens and could be applied for S. aureus analysis in human serum samples with the great recoveries and associated relative standard deviations (RSDs). These features do make the ideal facile electrochemical analytical platform for on-site detection of S. aureus.

Pathogenic microorganisms in water pose a potentially serious threat to human health. As an advanced oxidation technology, plasma sterilization has been widely used to kill pathogens in water. However, few studies on the effects of various substances in natural water bodies, such as common ions (Ca 2+ , Mg 2+ , Cl − , SO 4 2− , Fe 2+ ), as well as natural organic matter (humic acid, HA) and turbidity (diatomite), on plasma sterilization. This study used a dielectric barrier discharge (DBD) plasma jet to inactivate Staphylococcus aureus ( S. aureus ) in pure water and simulated water containing the above substances. We systematically investigated how experimental parameters and water constituents affected S. aureus inactivation. Results showed that Ca 2+ , Mg 2+ , Cl − , natural organic matter (humic acid, HA), and turbidity (diatomite) inhibited inactivation, while Fe 2+ and SO 4 2− promoted it. Additionally, the MDA content, cell metabolic capacity, and cell integrity of S. aureus in pure water and simulated water system were detected before and after plasma treatment. The results showed that RS produced by plasma attacked the cell membrane of S. aureus , causing a rupture. RS also accelerated oxidative stress and disrupted cell metabolism, ultimately leading to cell death. Long-lived reactive species (RS) in water induced by plasma, such as (H 2 O 2 , NO 3 − , NO 2 − ), had no significant effect in plasma inactivation. The positive role of short-lived reactive species ( O 2 − , 1 O 2 and OH) in the inactivation of S. aureus was verified by ESR and quenching experiments, especially •O 2 − .

A facile label-free electrochemiluminescent (ECL) biosensor was developed for detection of Staphylococcus aureus ( S. aureus ) based on the specific binding between Fc region of immunoglobulin G (IgG) and S. aureus protein A (SPA) in the cell wall. Carboxyl graphene, with large surface and excellent electron transfer ability, was used as the carrier of IgG for fabrication of ECL biosensor. This biosensor was constructed by depositing carboxyl graphene/porcin IgG composite on the surface of a glassy carbon electrode. The specific reaction between SPA and IgG resulted in a decrease of ECL signal because the bound S. aureus interrupted the interfacial electron transfer and hindered the diffusion of the ECL active substances. The ECL intensity decreased linearly with S. aureus concentrations in the range of 1.0×10 3 –1.0×10 9 colony-forming units (CFU) mL −1 , with a detection limit of 3.1×10 2 CFU mL −1 . The whole assay could be accomplished within 70 min when a ready-for-use biosensor was applied. The recovery test for food, environmental and biological samples showed recoveries between 75.0% and 116.7%. This developed biosensor displayed ideal specificity, high sensitivity, facile manipulation, simple fabrication and short assay time, thus provided a new pathway for pathogenic bacteria rapid screening.

Antimicrobial peptides and their analogues have become substitutes for antibiotics in recent years. The antimicrobial peptide analogue SAMP-A4-C8 ( n -octanoic-VRLLRRRI) with high antimicrobial activity was found in our lab. We speculate that it may kill pathogens by some lethal mechanism of action. In the present investigation, the microbicidal activities of SAMP-A4-C8 and its mechanism of action were investigated. The results demonstrated that SAMP-A4-C8 had lethal activities against Staphylococcus aureus and Candida albicans by cell disruption. Based on its microbicidal activities, we believe that it is worth further research for its potential as drug candidate. The results showed that SAMP-A4-C8, with low propensity to induce the resistance of S. aureus and C. albicans , could kill the persister cells of S. aureus and C. albicans , exhibited biofilm forming inhibition activity and preformed biofilm eradication ability against S. aureus and C. albicans , and displayed therapeutic potential on pneumonia in S. aureus -infected mice by reducing lung inflammation. The present study provided a promising drug candidate in the war against multidrug resistance.

In this study, the antibacterial activities of thyme essential oil nanoemulsions (TEON) prepared by ultrasonication (US) and high-pressure homogenization (HPH) were evaluated via minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and time-killing curve analyses. TEON-US presented superior bacteriostatic and bactericidal effects , which was found to be correlated with its better physicochemical characteristics (including smaller droplet size and higher zeta potential) compared to TEON-HPH. The underlying antibacterial mechanisms were specifically investigated targeting cell membranes of Escherichia coli O157:H7 (Gram-negative bacterium) and Staphylococcus aureus (Gram-positive bacterium). Thereinto, membrane disintegrations were observed followed by massive leakage of cytoplasmic inclusions . Furthermore, the membrane dysfunctions (involving membrane potential depolarization and fluidity reduction) were likely associated with the modifications seen in the fatty acid composition upon TEONs exposure, and ultimately causing bacterial death. These results deciphered the mode of antibacterial action of TEONs, providing a critical theoretical basis for their future application in food matrix.

Ethanopharmacological relevance Potentilla kleiniana Wight et Arn is a wide-spread wild plant in the mountainous areas in southern China. The whole herb has been used as a traditional herbal medicine to treat fever, arthritis , malaria, insect and snake bites , hepatitis, and traumatic injury. In vitro studies have reported the antibacterial activity use of the plant in traditional medicinal systems. Aim of the study The aim of this study was to investigate the inhibitory activity of total flavonoid from Potentilla kleiniana Wight et Arn (TFP) on methicillin-resistant Staphylococcus aureus (MRSA) in planktonic state and biofilm state. Materials and methods Antibacterial activities of TFP on planktonic MRSA were determined by agar diffusion method, microtiter plate assay and time-kill curve assay. Electrical conductivity, membrane permeability, membrane potential and autoaggregation were analyzed to study TFP effects on planktonic MRSA growth. Crystal violet (CV) staining and confocal laser scanning microscopy (CLSM) were analyzed to study TFP effects on aggregation and maturation of MRSA biofilm. After TFP treatment, extracellular polymeric substances (EPS) production were examined. Morphological changes in planktonic and MRSA biofilm following TFP treatment were determined with scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Moreover, α-Toxin protein expression and adhesion-related gene expression were also determined. Results The minimum inhibitory concentration (MIC) of TFP against MRSA was 20 μg/mL. The agar diffusion method and time-kill curve assay results indicated that TFP inhibited planktonic MRSA growth. TFP treatment significantly inhibited planktonic MRSA growth by inhibiting autoaggregation, α-hemolysin activity, α-Toxin protein expression, but increasing electrolyte leakage, membrane permeability and membrane potential and impacting cell structure. Moreover, TFP treatment significantly inhibited aggregation and maturation on MRSA biofilm by decreasing surface hydrophobicity , EPS production and adhesion-related gene expression. Conclusion The results of this trial provide scientific experimental data on the traditional use of Potentilla Kleiniana Wight et Arn for traumatic injury treatment and further demonstrate the potential of TFP to be developed as a novel anti-biofilm drug.
