Development of natamycin-loaded zein-casein composite nanoparticles by a pH-driven method and application to postharvest fungal control on peach against Monilinia fructicola

Xuejiao Xu,Xinlin Peng,Chen Huan,Jie Chen,Yuecheng Meng,Sheng Fang

This work fabricated natamycin-loaded zein-casein nanoparticles (N-Z/C NPs) by a pH-driven approach and applied to control postharvest peach brown rot caused by Monilinia fructicola . When casein and phosphoric acid were used as a stabilizer and neutralizing acid, respectively, NPs with mean particle sizes < 100 nm and zeta-potentials < −30 mV could be obtained. The NPs could increase the aqueous dispersibility of natamycin and showed high stability against environmental changes, which could be attributed to both hydrophobic stacking and hydrogen bonds between natamycin and zein. Besides, the effects of N-Z/C NPs on the storage of peach were assessed in vitro and in vivo . Nanoencapsulation did not affect the antifungal activities of natamycin. The NPs with 20 and 80 µg/mL of natamycin could basically inhibit the spore germination and mycelial growth of M. fructicola , respectively. The N-Z/C NPs coatings exhibited better results than natamycin in controlling of peach brown rot.

Comparison of the enhanced attachment and proliferation of the human mesenchymal stem cells on the biomimetic nanopatterned surfaces of zein, silk fibroin, and gelatin

Xueying Huang,Jie Zeng,Yi Wang

Natural proteins have been reported to positively affect the attachment and proliferation of cells. For the first time, zein, a plant protein, was utilized to make patterned surface mimicking the extracellular matrix to assist the attachment and proliferation of stem cells. Zein would promote the attachment and proliferation of the stem cells more than 10 times of that of gelatin and silk fibroin, respectively, which are popular protein selections for the formation of the biomaterial scaffolds. The more the surface was covered by zein, the more the stem cell grown. It was revealed that the stem cells would grow and stretch in the direction of the patterns, and the stem cells preferred to grow in the grooves in the size of 8 μm, that was similar to the size of the stem cells, rather than the size larger or smaller than that of the cells, such as 50 and 2 μm. It was concluded that zein is a better choice than silk fibroin and gelatin with highly potential for the formation of patterned surface and structure as the biomaterial scaffolds for stem cell therapy.

Preparation and characterization of zein-based core-shell nanoparticles for encapsulation and delivery of hydrophobic nutrient molecules: Enhancing environmental stress resistance and antioxidant activity

Zhiheng Zhang,Yaxu Meng,Jinpeng Wang,Chao Qiu,Wenbo Miao,Qianzhu Lin,Xiaojing Li,Shangyuan Sang,David Julian McClements,Aiquan Jiao,Zhengyu Jin

A simple reverse solvent co-precipitation method was designed to prepare zein/debranched oxidized starch core-shell composite nanoparticles . Studies have shown that the composite nanoparticles have good stability and the ability to protect resveratrol from chemical degradation during long-term storage, pasteurization , and ultraviolet irradiation. The encapsulation rate of resveratrol in composite nanoparticles (81.15%) was significantly higher than that of pure protein nanoparticles (47.15%). The results of FT-IR (fourier transform infrared spectroscopy), circular dichroism , and fluorescence spectra show that hydrogen bonding, hydrophobic interaction, and electrostatic interaction exist in the composite nanoparticles. Under pasteurization conditions, ultraviolet radiation, and two weeks of room-temperature storage, the composite nanoparticles can better maintain the biological activity of nutrient molecules. Moreover, the particles can maintain stability in different pH and ionic strength environments and gradually degrade and release active molecules in the intestinal tract in vitro in the simulated gastrointestinal environment. These results show that zein/debranched oxidized starch nanoparticles (ZDNPs) have great potential for application in the fields of functional drinks , food , and biomedicine-related delivery.

Loading curcumin on hyperbranched polymers functionalized Zein via the phenol-yne click reaction as pH-responsive drug delivery system for chemotherapy and photodynamic therapy

Meiying Liu,Tongsheng Huang,Yiping Liu,Lihua Chen,Shijie Liao,Xinxin Gong,Mubarak G. Bello,Weifeng Zhu,Shaorong Huang,Xiaoyong Zhang

Zein and its complexes have been considered as promising carriers for encapsulating and delivering various biological active ingredients, however, there still have some issues about Zein-based drug delivery systems should be considered, including poor colloidal stability, low drug encapsulation efficiency as well as rapid initial drug release, and uncontrollable release. In this work, we reported for the first time that hyperbranched polymers (HPG) functionalized Zein with terminal alkyne (Zein-HPG-PA) can be used for loading anticancer agent curcumin (CUR) via a facile phenol-yne click reaction. The resultant product (Zein-HPG-PA@CUR) displays high drug loading capacity, small particle size and excellent water dispersibility. More importantly, almost no CUR was released from Zein-HPG-PA@CUR under pH 7.4 and the cargo will be gradually released under acidic environment. As compared with free CUR, Zein-HPG-PA@CUR shows considerable cytotoxicity towards MDA-MB-231 cells under dark environment, while the cytotoxicity was significantly enhanced upon light-irradiation, implying great potential of Zein-HPG-PA@CUR for cancer treatment. Considered the above aspects, we believe that this work should be of significant impact on the biomedical applications of Zein, especially for fabrication of Zein-based responsive drug delivery systems.

Preparation and characterization of thymol-loaded ZIF-8/κ-carrangeenan/Zein composite film as active food packaging with enhanced functional properties for blueberry preservation

Huihui Li,Shunchang Wang,Jin Chen,Mengchu Sun,Hongjin Tang

Polysaccharide-protein-based composite films are promising for food packaging, but interfacial incompatibility between hydrophilic biopolymers and hydrophobic proteins limits their use. In this study, a new thymol-loaded zeolitic imidazolate framework-8/κ-carrageenan/Zein composite film was developed for blueberry preservation. The film exhibited excellent physical properties with a thickness of 0.052 ± 0.0001 mm, a high tensile strength of 40.631 ± 1.527 MPa, an elongation at break of 21.223 ± 1.018 %, and a reduced water vapor permeability of (0.764 ± 0.166) × 10 −8  kg·m·m −2 ·s −1 ·Pa −1 . The incorporation of thymol and ZIF-8 improved the film's microstructure and interfacial compatibility between κ-carrageenan and Zein. Thymol release was pH-dependent and sustained with a cumulative release of 91.959 ± 0.454 % over 36 h at pH 4.5. Biological testing revealed significant antibacterial properties with survival rates of 0.002 ± 0.002 % against Staphylococcus aureus and 5.973 ± 0.307 % against Escherichia coli , and antioxidant capacity with DPPH and ABTS radical scavenging rates of 75.60 ± 0.53 % and 61.49 ± 1.19 %, respectively. The film showed good biodegradability and extended blueberry shelf life by 9 days. This composite film offers enhanced mechanical properties, controlled release, and bioactivity, showing great potential as active food packaging for fruit preservation.

Improvement of gel quality and emulsion stability of high-dose gallic acid-modified myofibrillar protein by zein nanosystems: emphasizing the role of anionic polysaccharides

Jinyu Chen,Siyang Wang,Beibei Jia,Zhirui Ji,Ximing Li,Dengyong Liu,Zijian Wu

The binding of high-dose polyphenols can induce aggregation of myofibrillar protein (MP), which compromises its gelling and emulsifying functionalities. Herein, zein/anionic polysaccharide nanoparticles were fabricated for efficient encapsulation of gallic acid (GA), to inhibit the excessive modification of MP by GA (180 μM/g). The GA encapsulation hindered the aggregation of MP, facilitating more proteins to participate in the construction of a denser and strengthened gel network with more entrapped water, and promoting the diffusion of the proteins to the oil surface, resulting in a lower interfacial tension and a more uniform droplet distribution. The electronegativity, hydrophilicity and viscosity of these polysaccharides also helped to improve the gel quality and emulsion stability. Overall, the presence of sodium alginate showed the best improvement effect, followed by gum arabic and xanthan gum. This work provides a strategy to mitigate phenolic antioxidant-induced damage to functionalities of meat proteins.

Co-delivery of siAEG-1 and doxorubicin to treat osteosarcoma via nanomicelles for azide−alkyne “click” conjugation of poly(l-lysine) dendrons onto Zein

Jiadong Pang,Leilei Huang,Yating Lian,Zhijie Yuan,Fen Wang,Li-Ming Zhang

Astrocyte elevated gene-1 (AEG-1) oncogene is a notorious and evolving target in a variety of human malignancies including osteosarcoma. The RNA interference (RNAi) has been clinically proven to effectively knock down specific genes. To successfully implement RNAi in vivo, protective vectors are required not only to protect unstable siRNAs from degradation, but also to deliver siRNAs to target cells with controlled release. Here, we synthesized a Zein-poly( l -lysine) dendrons non-viral modular system that enables efficient siRNA-targeted AEG-1 gene silencing in osteosarcoma and encapsulation of antitumor drugs for controlled release. The rational design of the ZDP integrates the non-ionic and low immunogenicity of Zein and the positive charge of the poly( l -lysine) dendrons (DPLL) to encapsulate siRNA and doxorubicin (DOX) payloads via electrostatic complexes and achieve pH-controlled release in a lysosomal acidic microenvironment. Nanocomplexes-directed delivery greatly improves siRNA stability, uptake, and AEG-1 sequence-specific knockdown in 143B cells, with transfection efficiencies comparable to those of commercial lipofectamine but with lower cytotoxicity. This AEG-1-focused RNAi therapy supplemented with chemotherapy inhibited, and was effective in inhibiting the growth in of osteosarcoma xenografts mouse models. The combination therapy is an alternative or combinatorial strategy that can produce durable inhibitory responses in osteosarcoma patients.

Fabrication of novel pullulan/carboxymethyl chitosan-based edible film incorporated with ultrasonically equipped aqueous zein/turmeric essential oil nanoemulsion for effective preservation of mango fruits

Jawad Ashraf,Nadia Ismail,Tabussam Tufail,Jiyao Zhang,Muhammad Awais,Qianqian Zhang,Zahoor Ahmed,Yajing Qi,Shuyi Liu,Bin Xu

This study aimed to develop ultrasonically-assisted, alcohol-free, and noncorrosive aqueous zein/turmeric essential oil (TEO)-loaded nanoemulsions (NEs) to stimulate pullulan/carboxymethyl chitosan (P/CMCS)-based edible films for mango fruit preservation. The influence of innovative sonicated zein/TEO-based NEs (ZTNEs) as nanofillers on the physico-mechanical characteristics of the resulting P/CMCS edible films was investigated. A stable and well-dispersed ZTNE was achieved using 20 % zein with 10 min of ultrasound treatment, leading to a reduced droplet size (194.23 ± 0.41 nm) and high ζ-potential (−48.72 ± 0.74 mV). Furthermore, the homogeneity of the ZTNE dispersion, evaluated via confocal laser microscopy (CLSM) analysis, indicated that it can be uniformly incorporated into P/CMCS edible films. In addition, the incorporation of ZTNE into P/CMCS edible films improved barrier properties, mechanical strength, oxidative stability, and antimicrobial properties compared with P/CMCS edible films without ZTNE, possibly due to hydrogen bonding in the film matrix, as confirmed by scanning electron microscopy (SEM) micrographs as well as Fourier transform infrared spectroscopy and X-ray diffraction. Interestingly, the results indicated that P/CMCS/ZTNE/US 10 min and P/CMCS/ZTNE 4 % films demonstrated substantial antibacterial activity against Escherichia coli (11.54 ± 0.72 and 16.62 ± 1.13 mm, respectively) and Staphylococcus aureus (12.81 ± 1.03 and 14.13 ± 0.69 mm), respectively. Moreover, P/CMCS/ZTNE/US 10 min and P/CMCS/ZTNE 4 % films were found to be effective in mango preservation over a 12-day storage period at 25 °C ± 1 °C, as assessed by fruit firmness, weight loss, total soluble solid content, total phenolic content, lipoxygenase activity, and electronic nose analysis. Consequently, these findings indicate that P/CMCS/ZTNE edible films may function as an effective biodegradable packaging solution for fruit preservation within the food industry.

Fabrication and characterization of zein-tea polyphenols-pectin ternary complex nanoparticles as an effective hyperoside delivery system: Formation mechanism, physicochemical stability, and in vitro release property

Xiaojing Wang,Mei Li,Fuguo Liu,Fei Peng,Feng Li,Xinman Lou,Yu Jin,Jun Wang,Huaide Xu

Hyperoside (HYP) has various potential benefits, however, its low water-solubility and poor bioavailability have restricted its application. Here, HYP-loaded zein-tea polyphenols (TP)-pectin ternary complex nanoparticles (Z/TP/P-HYP) were prepared by the antisolvent precipitation method for HYP delivery. The formed Z/TP/P-HYP are negatively charged spherical particles with a size of 246 nm, and have the highest HYP encapsulation efficiency (94.2%) at TP was 0.25 mg/mL. Fourier transform infrared spectroscopy revealed that hydrogen bonding, electrostatic interactions, and hydrophobic effects were major interactions to Z/TP/P-HYP formation. Differential scanning calorimetry confirmed that encapsulated HYP was in an amorphous state. Freeze-dried Z/TP/P-HYP displayed good water-redispersibility and high particle yield (95.2%). Z/TP/P-HYP exhibited improved pH (2.0–8.0) and ionic (0–500 mM) stability. Furthermore, Z/TP/P-HYP demonstrated stronger antioxidant properties than free HYP and provided HYP sustained release under simulated gastrointestinal conditions. Therefore, Z/TP/P-HYP have great potential as an effective HYP delivery system for applications in foods.

Tailored NaOH/phytic acid-driven method for fabricating carvacrol-loaded zein/trehalolipid nanoparticles with higher processing adaptability: Utilizing dual critical pH of wall and core materials

Jiyao Zhang,Shuyi Liu,Yajing Qi,Zhongwei Chen,Bin Xu

Encapsulating within zein/trehalolipid nanoparticles (Z/TL) via NaOH/phytic acid-driven method (PAPD) can improve water solubility and stability of carvacrol (CAR). However, this technique still has limitations including empirically determined parameters and potential risk of core material degradation. In this respect, we introduced a strategy of positioning dual critical pH based on specific attributes of wall and core materials. The finest alkalization endpoint for Z/TL was determined by monitoring conformational changes across pH 7–12. Meanwhile, the suitable pH of deprotonation was identified by assessing pH-dependent water solubility and chemical stability of CAR. Obtained information was applied to modify PAPD into PAPD’. The colloidal properties, encapsulation efficiency (EE), loading capability (LC), environmental stress resistance, and in vitro release behavior of nanoparticles prepared using PAPD’ and PAPD were systematically compared. The optimal alkalinization endpoint for Z/TL was determined to be pH 11.25. where the degree of hydrolysis would not increase further, but the acceleration of increase in aqueous solubility would reach its maximum. The optimal pH of deprotonation for CAR was pH 11, balancing high solubility with controllable degradation. PAPD’-nanoparticles exhibited a smaller size (99.36 nm), higher ζ-potential (−43.91 mV), and improved EE/LC (97.95 %/29.39 %). Enhanced stability, along with a sustained release profile were demonstrated. Consumption of NaOH/PA was also reduced while maintaining required efficacy. Overall, customized PAPD’ not only improves the processing adaptability and flexibility of fabricating protein-based nanoparticles but also provides a theoretical framework and practical paradigm for the rational design of nanocarriers for bioactive compounds.

Label-free electrochemical immunosensor for detection of insulin-like growth factor-1 (IGF-1) using a specific monoclonal receptor on electrospun Zein-based nanofibers/rGO-modified electrode

Fatemeh Maleki,Mohammad-Reza Rashidi,Habib Razmi,Marjan Ghorbani

A novel electrochemical immunosensor was developed for ultrasensitive determination of the hormone insulin-like growth factor 1 (IGF-1) based on immobilization of a specific monoclonal antibody on the electrospun nanofibers of Polyacrylonitrile (PAN)/Zein-reduced graphene oxide (rGO) nanoparticle . The nanofibers deposited on glassy carbon electrode (GCE) showed good electrochemical behaviors with synergistic effects between PAN, Zein, and rGO. PAN/Zein nanofibers were used due to flexibility, high porosity, good mechanical strength, high specific surface area, and flexible structures, while rGO nanoparticles were used to improve the detection sensitivity and anti-IGF-1 immobilizing. Different characterization techniques were applied consisting of FE-SEM, FT-IR, and EDS for the investigation of morphological features and nanofiber size. The redox reactions of [Fe(CN) 6 ] 4−/3− on the modified electrode surface were probed for studying the immobilization and determination processes, using differential pulse voltammetry (DPV) and cyclic voltammetry (CV). Under optimal conditions, LOD (limit of detection) and LOQ (limit of quantification) were obtained as 55.72 fg/mL and 185.73 fg/mL respectively, and sensitivity was acquired 136.29 μA/cm 2 .dec. Moreover, a wide linear range was obtained ranging from 1 pg/mL to 10 ng/mL for IGF-1. Furthermore, the proposed method was applied for the analysis of IGF-1 in several human plasma samples with acceptable results, and it also exhibited high selectivity, stability, and reproducibility.

Two different fabrication method of high-hydrophobic Bi-layer indicators based on sodium alginate-polyvinyl alcohol/chitosan-zein incorporated with alizarin@ZIF-8 for beef freshness visualization

Jianing Zhang,Junjun Zhang,Lei Zhou,Nurmymeti Yasen,Abudulekemu Abudinaibi,Xiaowei Huang,Xiaodong Zhai,Zhihua Li,Jiyong Shi,Remah Sobhy,Ibrahim Khalifa,Xiaobo Zou

Intelligent films are essential for real-time monitoring of food spoilage. However, the application is often limited by challenges such as hydrophilicity and color instability. In this study, two high hydrophobic bilayer indicators based on sodium alginate-polyvinyl alcohol/chitosan-zein with alizarin grafted onto ZIF-8 carrier (AL@ZIF-8) were fabricated using the casting method and electrospinning, respectively. Molecular docking simulations confirmed the successful grafting of AL@ZIF-8 onto the film substrates. The performance of films was compared and the effectiveness in monitoring freshness indicators was evaluated. The cast film had a precisely controlled thickness due to self-assembly performance, resulting in superior mechanical properties (TS = 10.64 MPa, EB = 43.764 %). In comparison, the nanofiber films demonstrated higher UV-blocking ability (ΔE < 4), faster spoilage response, and greater safety (Cell viability >80 %). Importantly, the nanofiber films exhibited higher sensitivity in monitoring beef freshness, with a color change pattern strongly correlating with spoilage (R 2  = 0.8976).
