This study was conducted to develop a W/O/W emulsion encapsulated Lactobacillus plantarum 23 – 1 ( LP23 – 1 ) to significantly enhance the survival rate of LP23-1 under simulated digestion and storage conditions. The zein particles and pectin formed a complex through electrostatic interaction and hydrogen bonding. When the proportion of zein particles to pectin was 1:1, the emulsifying stability index (ESI) was 304.17 %. Additionally, when the proportion of the internal aqueous phase to the oil phase was 1:9, the polyglycerol polyricinoleate (PGPR) concentration was 5 %, the proportion of primary emulsion to the external aqueous phase was 5:5, the zein particles concentration was 4 %, and the proportion of zein particles to pectin was 1:1, the encapsulation rate was the highest at 96.27 %. Cryo-scanning electron microscopy and fluorescence microscopy confirmed the morphology of W/O/W emulsion and successful encapsulation of LP23–1 . Furthermore, compared with free LP23–1 , the W/O/W emulsion encapsulation significantly improved the survival rate of LP23–1 to 73.36 % after simulated gastrointestinal digestion and maintained a high survival rate of 78.42 % during the 35-day storage. The W/O/W emulsion was found to effectively improve the survival rate of LP23–1 during simulated digestion and storage, which has implications for the development of probiotic functional foods with elevated survival rates.

In order to enhance the stability and bioavailability of fat-soluble kaempferol (Kae), the zein-pectin nanoemulsions were synthesized by a novel dual-frequency pulsed ultrasound (DFPU) technology to improve the loading effect of Kae. The effects of ultrasonic operating parameters on the nanoemulsions were examined, along with the influence of DFPU treatment on the structure, stability, digestion, and intracellular uptake of the nanoemulsions in this study. The results indicated that both emulsion encapsulation and DFPU treatment significantly enhanced the protection and delivery of Kae. Compared to the unsonicated emulsion, the emulsifying activity index, stability index, Kae embedding rate, and hydroxyl radical scavenging rate increased by 133.81%, 4.18%, 19.20%, and 19.73%, respectively, while the particle size decreased by 94.43% after DFPU treatment. DFPU treatment notably improved the stability of the nanoemulsions during long-term storage, high temperature, and salt concentration conditions, as well as the retention of Kae by 15.59% in intestinal digestives. Furthermore, DFPU treatment significantly enhanced the Caco-2 intracellular absorption rate, transport rate and bioavailability of Kae by 7.67%, 9.96% and 14.67%, respectively at a dosage of 0.6 mg/mL, which was attributed to the significant downregulation of mRNA expression levels of tight junction protein Occludin and efflux proteins MDR1 and BCRP by 21.27%, 51.05%, and 62.26%, respectively. This downregulation enhanced the intracellular transport capacity of Kae while reducing its exocytosis, thereby improving cellular utilization. The mechanism of improving the intracellular bioavailability of kaempferol was elucidated. This study offers a promising nanoemulsion for the efficient oral delivery of fat-soluble active ingredients, fully facilitating its antioxidant activity.

The K + -induced pectin-based vitrimer hydrogel ink, with dynamic exchange properties, offers an innovative solution for single-channel extrusion-based 3D food printing. However, forming hydrogen-bonded cross-linked vitrimer traditionally requires over 150 mM of K + to overcome the electrostatic repulsion between enzyme-modified pectin (EMP) molecules, which raises health concerns due to the high K + content. This study developed a novel strategy to introduce Ca 2+ bridges to partially weaken the electrostatic repulsion, fabricating low-K + vitrimer gel inks. Using 67 mM K + and 0.9 mM Ca 2+ , a densely cross-linked gel (Ca-EMP-K 67 mM) was formed at 25 °C, exhibiting excellent mechanical performance (~140 g hardness). Meanwhile, the gel possessed excellent self-healing properties, with a relaxation time of 8.3 s at 80 °C, confirming its vitrimer status. Notably, the Ca-EMP-K 67 mM gel ink achieved a ~ 55 % reduction in K + doses while enabling an increase in shaping temperature from 4 °C to 25 °C, compared to the traditional EMP-K 150 mM gel ink. These advancements enabled a low-K + pectin ink for ambient 3D printing. The introduction of Ca 2+ not only reduces the K + concentration requirement but also increases the shaping temperature, broadening the gel ink's applications in the food industry.

Current therapies for ulcerative colitis (UC) remain inadequate, necessitating novel treatment strategies. This study isolated SCPII-1, a homogeneous pectin polysaccharide (Mw: 278.153 kDa, DM: 57.21 %) from Schisandra chinensis , primarily composed of →4)-α-D-GalpA-(1 → units. In dextran sulfate sodium (DSS)-induced colitis mice, SCPII-1 alleviated symptoms, repaired mucosal ultrastructure (microvilli, mitochondria), and enhanced barrier integrity. Mechanistically, SCPII-1 ameliorated gut dysbiosis in UC mice, notably through the selective enrichment of the beneficial bacterium Akkermansia muciniphila (AKK). Interestingly, SCPII-1 not only promoted the proliferation of AKK but also modulated its tryptophan metabolism, particularly enhancing the production of the tryptophan-derived metabolite indole-3-ethanol (I3E). The beneficial role of SCPII-1-induced microbiota in UC was validated using pseudo-germ-free mice and fecal microbiota transplantation experiments. Moreover, I3E was identified as a key microbiota-derived active metabolite regulated by SCPII-1, and exogenous supplementation of I3E further confirmed its therapeutic efficacy in UC. Importantly, I3E activated the aryl hydrocarbon receptor (AhR), upregulated IL-22 expression, and promoted mucosal repair and immune homeostasis, thereby confirming the protective role of the SCPII-1-AKK-tryptophan metabolism axis in colitis. These findings highlight SCPII-1 as a promising prebiotic agent for UC treatment by modulating AKK-dependent tryptophan metabolism and activating the AhR/IL-22 signaling pathway, offering a novel microbiota-based therapeutic strategy.

This study systematically investigates the structure-function relationships between Maillard-glycated (PWM) and genipin-mediated crosslinked (PWG) pectin-whey protein isolate (WPI) complexes, with particular emphasis on their differential impacts on bioadhesive performance and digestion profiles for curcumin-loaded delivery systems. Both crosslinking strategies induced structural unfolding of WPI and endogenous pectin proteins, exposing amino-active sites that facilitated their reconstitution into stable complexes. PWG developed denser and more homogeneous hydrophobic microdomain networks compared to PWM. Curcumin was encapsulated via hydrogen bonding and hydrophobic interactions within these networks. Ex vivo intestinal adhesion assays demonstrated PWG loaded curcumin (PWG-Cur) achieved 28.69 % particulate retention in colonic environments, representing 7.0-fold and 1.75-fold enhancements over free curcumin and PWM loaded curcumin (PWM-Cur), respectively. In vitro digestion models confirmed PWG-Cur's superior bioaccessibility (22.46 %), reflecting a 37 % enhancement over PWM-Cur (16.35 %). These findings provide critical design principles for engineering mucoadhesive pectin-polyphenol delivery platforms capable of precisely modulating cargo release while enhancing bioaccessibility.

Cell wall is the major component of root apoplast which is the main reservoir for iron in roots, while nitric oxide (NO) is involved in regulating the synthesis of cell wall. However, whether such regulation could influence the reutilization of iron stored in root apoplast remains unclear. In this study, we observed that iron deficiency elevated NO level in tomato ( Solanum lycopersicum ) roots. However, application of S-nitrosoglutathione, a NO donor, significantly enhanced iron retention in root apoplast of iron-deficient plants, accompanied with a decrease of iron level in xylem sap. Consequently, S-nitrosoglutathione treatment increased iron concentration in roots, but decreased it in shoots. The opposite was true for the NO scavenging treatment with 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO). Interestingly, S-nitrosoglutathione treatment increased pectin methylesterase activity and decreased degree of pectin methylation in root cell wall of both iron-deficient and iron-sufficient plants, which led to an increased iron retention in pectin fraction, thus increasing the binding capacity of iron to the extracted cell wall. Altogether, these results suggested that iron-deficiency-induced elevation of NO increases iron immobilization in root apoplast by decreasing pectin methylation in cell wall.

Two novel pectin polysaccharides were extracted from purple passion fruits ( Passiflora edulia Sims) peel by hot-water extraction (PFSP60) and ultrasonic-assisted extraction (UPFSP60), their structural characteristics, antioxidant and anti-inflammatory effects were preliminary investigated. The results revealed that the monosaccharide compositions of both polysaccharides mainly include galacturonic acid, glucose, xylose, arabinose, galactose and rhamnose. Methylation analysis indicated that the main linkage types of PFSP60 included 1,4-linked-GalA, 1,4-linked-Glc, 1,4-linked-Xyl and 1-linked-Gal in a molar ratio of 1.02:1.00:0.36:0.13. The glycosidic linkage types of UPFSP60 included 1,4-linked-GalA, 1,4-linked-Glc, 1,2-linked-Rha, 1,4-linked-Xyl and 1,3,6-linked-Gal in a molar ratio of 4.03:1.00:0.32:0.31:0.29. The DPPH radical, ABTS radical, hydroxyl radical, and superoxide anion scavenging activities of PFSP60 and UPFSP60 enhanced in a dose-dependent manner. In LPS-induced RAW 264.7 macrophages, PFSP60 and UPFSP60 could inhibit secretion amounts of pro-inflammatory factors (NO, IL-1β, TNF-α and IL-6) and down-regulated gene expressions. However, the polysaccharides extracted with the assistance of ultrasonication showed stronger antioxidant and anti-inflammatory activities. The results suggested that ultrasound treatment might be an effective approach to obtain pectin polysaccharides with lower molecular weight and stronger antioxidant and anti-inflammatory activities. These results also indicated that passion fruit peel polysaccharides have potential application prospect in functional foods and medicine industry.

The grafting of amino acid [glycine (Gly), L (+)-glutamic acid (Glu) and l -lysine (Lys)] onto pectin at the ultra-low temperature solid-liquid two-phase interface under papain catalysis was investigated, and the effects of amino acids amidation on the viscosity and gel properties of pectin were analysed. Fat-mimic characteristics were also evaluated. Fourier transform infrared spectrometer, the proton NMR and X-ray photoelectron spectroscopy were used to characterise and discuss the synthesis mechanisms of Gly-amidated pectin (LMP-Gly), Glu-amidated pectin (LMP-Glu) and Lys-amidated pectin (LMP-Lys). Results suggested that papain still exhibited synthesis function under ultra-low temperature reaction. The amino group of amino acid and the carboxyl group of pectin bound at the interface between organic and water phases to form amidated pectin under papain catalysis. The grafting rates of LMP-Glu, LMP-Gly and LMP-Lys were 13.04%, 9.32% and 11.71%, respectively. Under low calcium concentration and no sugar, the dynamic viscosity and gel properties of the amidated pectin were better than those of the low-esterified pectin, and its gel particle size met the requirements for fat mimics.

The therapeutic potential of essential oils in feed and medicine is limited by their volatility, degradation, and poor bioavailability. To overcome these challenges, a novel colon-targeted microsphere was engineered for the controlled delivery of tea tree essential oil (TEO), as illustrated in Scheme 1 . A critical component of this system is a pH-responsive coating composed of pectin and starch, which acts as a colon-specific polymeric barrier. This coating demonstrated precise pH-responsive release, with minimal leakage in simulated gastric fluid (SGF) (only 10.87 % release over 4 h) and sustained, colon-specific release in simulated intestinal fluid (SIF) (79.77 % over 14 h). The core of the system, designated TEOCMT, was fabricated through the modification of montmorillonite (MMT) with cetyltrimethylammonium bromide (CTAB), resulting in organo-montmorillonite (CMT). Tea tree essential oil was then encapsulated within the interlayer spaces of CMT, forming an effective sustained-release system (TEOCMT). Compared to raw MMT (5.63 %), the TEO loading capacity of CMT increased 8.3-fold, reaching 46.79 %. Furthermore, the antibacterial activity of PS/TEOCMT microspheres was triggered in alkaline media, where they dissolved and released the TEOCMT core. This resulted in enhanced efficacy, particularly against S. aureus (MIC = 1.25 mg mL −1 ), which was superior to that of pure TEO (MIC = 2.5 mg mL −1 ). The PS/TEOCMT microspheres also exhibited excellent biocompatibility. This work presents a robust and effective strategy for unlocking the full potential of essential oils in colon-specific applications.

Rhamnogalacturonan I (RG-I) pectin are regarded as strong galectin-3 (Gal-3) antagonist because of galactan sidechains. The present study focused on discussing the effects of more structural regions in pectin on the anti-Gal-3 activity. The water-soluble pectin (WSP) recovered from citrus canning processing water was categorized as RG-I pectin. The controlled enzymatic hydrolysis was employed to sequentially remove the α-1,5-arabinan, homogalaturonan and β-1,4-galactan in WSP. The Gal-3-binding affinity KD (kd/ka) of WSP and debranched pectins were calculated to be 0.32 μM, 0.48 μM, 0.56 μM and 1.93 μM. Moreover, based on the more sensitive cell line (MCF-7) model, the IC 30 value of WSP was lower than these of modified pectins, indicating decreased anti-Gal-3 activity. Our results suggested that the total amount of neutral sugar sidechains, the length of arabinan and cooperation between HG and RG-I played important roles in the anti-Gal-3 activity of WSP, not the Gal/Ara ratio or RG-I/HG ratio. These results provided a new insight into structure-activity relationship of citrus segment membrane RG-I as a galectin-3 antagonist and a new functional food.

This study focuses on developing a water-in-oil-in-water (W 1 /O/W 2 ) double emulsion system using high-intensity ultrasound (HIU)-treated pea protein isolate (HIU-PPI) and pectin to encapsulate Lactobacillus plantarum ( L. plantarum ). The effects of ultrasound treatment on pea protein isolate (PPI) characteristics such as solubility, particle size, emulsification, surface hydrophobicity, and surface free sulfhydryl group were examined, determining optimal HIU processing conditions was 400 W for 10 min. The developed W 1 /O/W 2 double emulsion system based on HIU-PPI demonstrated effective encapsulation and protection of L. plantarum , especially at the HIU-PPI concentration of 4 %, achieving an encapsulation efficiency of 52.65 %. Incorporating both HIU-PPI and pectin as emulsifiers increased the particle size and significantly enhanced the emulsion’s viscosity. The highest bacterial encapsulation efficiency of the emulsion, 59.94 %, was attained at a HIU to pectin concentration ratio of 3:1. These emulsions effectively encapsulate and protect L. plantarum , with the concentration of HIU-PPI being a critical factor in enhancing probiotic survival under simulated gastrointestinal digestion. However, the concurrent utilization of pectin and HIU-PPI as emulsifiers did not provide a notable advantage compared to the exclusive use of HIU-PPI in enhancing probiotic viability during in vitro simulated digestion. This research offers valuable perspectives for the food industry on harnessing environmentally friendly, plant-based proteins as emulsifiers in probiotic delivery systems. It underscores the potential of HIU-modified pea protein and pectin in developing functional food products that promote the health benefits of probiotics.

The Al-doped Fe 3 O 4 /hydroxyapatite (HAP) nanocomposites prepared in pectin matrix was employed as adsorbent that offers high adsorption capacity for fluoride, magnetic separability and good stability. Powder X-ray diffraction and transmission electron microscopy characterizations were used to determine the structure and morphology of as-prepared samples. The needle-like Al-doped Fe 3 O 4 /HAP nanocrystals were coated with pectin molecules, which not only induced the nucleation, but inhibited the further growth of nanocrystals. The results of thermogravimetric analysis and Fourier transform infrared spectra showed that the obtained products are organic–inorganic nanocomposites; and the Al doping in composites increased the amount of hydroxyl groups, which could supply more adsorption sites for fluoride. In bath experiments, the adsorption capacity of Al-doped samples was significantly higher than that of pure HAP due to the high affinity of aluminium toward fluoride. The effect of reaction parameters such as pH, calcination temperature, co-existing anions and Al/Ca molar ratio on the adsorption capacity was also investigated to determine optimum adsorption condition. The Al-doped Fe 3 O 4 /HAP nanocrystals exhibited high adsorption rate for fluoride during the first 30 min and reached equilibrium within 80 min. The adsorption kinetics and isotherms were well-fitted by the pseudo-second-order kinetic model and the Freundlich isotherm model, respectively. In addition, the adsorption mechanism of fluoride on Al-doped Fe 3 O 4 /HAP nanocomposites was investigated, which can be mainly attributed to electrostatic attraction and ion-exchange.
