Synthesis and characterization of spherical C/ZnO/polydopamine composites as carriers for 5-fluorouracil: a focus on sustained release performance

Mingli Yue,Zhikuan Feng,Long Gao,Ying Fu,Fei Ye

Drug delivery systems utilize micro-nanomaterials as drug carriers, encapsulating drugs within micro-nanoparticles and releasing them to target tissues or cells via specific pathways. This technology enhances drug stability, prolongs drug retention time in the body, and reduces damage to healthy tissues. This study successfully synthesized novel carbon-doped zinc oxide (C/ZnO) micro-nanomaterials and employed polydopamine (PDA) for surface modification, resulting in the construction of C/ZnO/PDA carriers. Experimental results demonstrate that PDA modification significantly enhances the drug delivery performance of the material. This composite material exhibits a 5-Fu loading efficiency of 37.8%, representing a 1.15-fold increase compared to unmodified C/ZnO. In vitro release tests demonstrate sustained drug release characteristics, showing a 1.62-fold increase over the unmodified system. In addition, these medicine particles significantly enhanced the indicators of antibacterial and bactericidal efficacy. In vitro cell experiments also confirmed that the drug carrier can enhance the lethality of the drug against HepG2 cells. Research on the binding energy between the carrier and the drug revealed that PDA modification promotes drug-carrier binding, resulting in more stable overall adsorption. These findings demonstrate the potential application value of micro-nanoparticle-based targeted drug delivery systems in disease prevention and treatment.

A study on a novel MIL-100(Fe)@UREA Nano-Fertilizer with high loading capacity and superior sustained-release performance

Yujing Zhou,Yizhe Yang,Yinqi Sun,Zihao Wang,Hengbo Ding,Kai Liu,Weiguo Zhang,Songke Feng

To overcome the low nitrogen use efficiency (<40%) of conventional urea, this study utilizes the metal–organic framework MIL-100(Fe)—characterized by ultrahigh surface area and tunable mesoporosity—as a promising carrier for nano slow-release fertilizers with enhanced nutrient stability. Fourier transform infrared spectroscopy, thermogravimetric analysis, and Brunauer–Emmett–Teller measurements confirmed that urea was uniformly accommodated within the MOF channels in an amorphous, physically adsorbed state without chemical bonding. High-performance liquid chromatography revealed that loading capacity was primarily determined by the urea-to-MOF mass ratio, whereas loading temperature and duration had negligible effects, enabling precise synthesis control. Orthogonal optimization identified the optimal parameters (mass ratio 1:1, 110°C, 7 h), yielding a urea loading of 59.2% and encapsulation efficiency of 51.8%. Release experiments demonstrated a 22.5-fold extension in release duration compared to pure urea, with 87.6% cumulative release over 180 h governed by Fickian diffusion. Furthermore, physical characterization and bench-scale metering tests verified the granules' mechanical adaptability, achieving stable, continuous discharge comparable to urea. This work validates MIL-100(Fe)@UREA as a high-capacity, mechanically viable platform for sustainable and precision nutrient delivery. These results not only highlight the potential of MIL-100(Fe) in advanced slow-release fertilizer systems but also provide valuable insights for designing efficient and environmentally sustainable nutrient delivery platforms.

Galactosylated chitosan-grafted multiwall carbon nanotubes for pH-dependent sustained release and hepatic tumor-targeted delivery of doxorubicin in vivo

Xiaole Qi,Yao Rui,Yuchao Fan,Haiyan Chen,Ning Ma,Zhenghong Wu

Carbon nanotubes (CNTs) are well known for their distinctive drug-loading ability that is mainly due to their large surface area, which permits covalent attachment of various target ligands or drug molecules by π–π stacking, allowing them to act as potential tumor-targeting carriers. Herein, we describe the development of galactosylated chitosan-graftedoxidized CNTs (O-CNTs-LCH) for pH-dependent sustained release and hepatic tumor-targeted delivery of doxorubicin (DOX). The in vitro release behavior in aqueous release media of different pH values (5.5, 6.5 and 7.4) verified the pH-dependent sustained release of DOX from O-CNTs-LCH-DOX. Moreover, these nanocarriers exhibited significant in vitro tumor-targeting properties, with a higher cellular uptake efficiency than that of free DOX in HepG2 cells. In addition, the good biocompatibility and low toxicity of O-CNTs-LCH-DOX was demonstrated by evaluating HepG2 cytotoxicity, vascular irritation and the maximum tolerated dose. Moreover, after intravenous administration in mice bearing the H22 tumor, O-CNTs-LCH-DOX showed higher antitumor activity and stronger fluorescent intensity in tumor tissue compared to free DOX. These results indicated the selective hepatic tumor targeting and the therapeutic effect of those nanocarriers.

Self-Sacrificial Sustained-Release Effects Synergistically Modulate Dual-Electrode Interfacial Chemistry in a Biphasic Electrolyte for Electrolytic Zn–MnO2 Batteries

Xiaoli He,Song Huang,Yueliang Chen,Jiaming Xiong,Huatai Liang,Zuyang Hu,Minghui Ye,Yufei Zhang,Zhipeng Wen,Yongchao Tang,Qi Liu,Jian Zhu,Cheng Chao Li

Electrolytic zinc–manganese batteries (ZMBs) featuring high energy density and low cost hold significant potential for large-scale energy storage. However, their development is hindered by severe zinc corrosion and poor reversibility of the Mn2+/MnO2 conversion, originating from drastic pH fluctuations and an acidic environment. Herein, we propose an organic/inorganic self-stratifying electrolyte, which synergizes with the self-sacrificial sustained-release Zn(BF4)2 to regulate the zinc interface and MnO2 conversion chemistry. The design innovatively utilizes the in situ hydrolysis of BF4– to create an HF/F– buffer and free F– ions. The former stabilizes cathode pH and promotes uniform deposition and deep conversion of MnO2. Additionally, the released F– fosters a robust ZnF2-rich SEI on the zinc anode in a water-lean environment (organic phase), effectively suppressing corrosion and side reactions. Therefore, the zinc-symmetric battery with an organic phase exhibited a lifespan of over 650 h, exceeding conventional aqueous electrolytes by 38-fold. The Zn–MnO2 batteries deliver exceptional stability for 1800 cycles at 2C with an average Coulombic efficiency of 98.7%. This biphasic buffering-interface regulation strategy provides a generalizable approach to decouple interfacial challenges, which is promising for extension to other corrosion cell systems.

A Hydrogel-Mediated Sustained-Release Platform Alters the Biodistribution of Umbilical Cord Mesenchymal Stem Cell-Derived Extracellular Vesicles in Mice

Zongbin Sun,Qiuxia Zheng,Chongyang Bai,Desheng Liu,Yue Zhang,Ping Yang,Fanghong Wang,Rui Li,Jia Yao,Xun Li

Mesenchymal stem cell-derived exosomes represent a promising therapeutic avenue for tissue regeneration, yet their clinical translation is hindered by rapid clearance following systemic administration and the lack of optimized, sustained-release delivery strategies. To address these challenges, we crafted a biomimetic hydrogel to carry and gently release MSC-derived exosomes. We engineered a biocompatible, dextran-based composite hydrogel (carboxymethyl chitosan-oxidized dextran/tannic acid-iron complex, CMC-OD/TA-Fe(III)) for the encapsulation and sustained release of human umbilical cord MSC-derived exosomes. We systematically compared the organ biodistribution of exosomes delivered via this hydrogel platform using three distinct local routes: intraperitoneal injection, oral (intragastric) administration, and rectal administration, employing in vivo imaging and tissue immunofluorescence analyses in C57BL/6J mice. Following a single intraperitoneal dose, released exosomes accumulated primarily in the liver and spleen, with minimal brain enrichment. Notably, both oral and rectal administration—non-invasive alternatives to injection—facilitated considerable exosome dissemination to multiple organs. The significant distribution observed via the oral route specifically challenges the prevailing assumption regarding the inefficacy of gastrointestinal delivery for exosome-based therapeutics. This study provides critical insights into administration-route-dependent biodistribution patterns and proposes novel, non-invasive delivery strategies to enhance the therapeutic efficacy and application scenarios of exosomes.

OSA-modification rice, debranched, and soluble starches encapsulating ferulic acid: interfacial effects and enhanced solubility, stability, bioactivity, and sustained release

Linlin Chen,Zhang Ming,Wei Li,Yingjie Zhou,Sicong Zhang,Xiyao Yang,Jiaying Xin,Na Zhang

Ferulic acid (FA) is a natural polyphenolic compound that has garnered significant attention due to its exceptional antioxidant, anti-inflammatory, and anticancer bioactivities. However, its poor chemical stability, low solubility, and insufficient bioavailability in practical applications severely limit its widespread use in the food, pharmaceutical, and cosmetics industries. To improve the stability and bioavailability of FA, this study used rice starch (RS), debranched starch (DBS) and soluble starch (SS) modified with octenyl succinic anhydride (OSA) as carriers to prepare three FA complexes, with the aim of improving their solubility and stability. The antioxidant activity, stability, and in vitro release behavior of the encapsulated FA were evaluated. Experimental results indicated that the encapsulation mechanism of ferulic acid by starch-based carriers involves hydrogen bonding, van der Waals force, and hydrophobic interactions. OSA-DBS exhibited the highest encapsulation efficiency, attributed to its compact cavity spiral structure. At the same time, OSA-SS and OSA-RS had lower encapsulation efficiencies due to their loose structure and mixed crystalline-amorphous structure, respectively. FA@OSA-DBS exhibited the strongest antioxidant activity and antibacterial ability, with significantly improved solubility and stability. In vitro release studies showed that FA@OSA-DBS exhibited a slow release rate in a simulated physiological environment, following a first-order kinetic model. The results indicate that the preparation of FA complexes using esterified starch can significantly improve the physical and chemical properties of FA by regulating the interfacial characteristics of starch. This finding lays the theoretical foundation for the development of phenolic acid-starch composite systems in the field of functional materials.

Beyond immediate effects: Microbial mechanisms and ecological feedback in year-long in situ cyanobacterial control using artemisinin sustained-release algaecides

Yushen Ma,Linyun Zhang,Mengting Yuan,Lixiao Ni,Cunhao Du,Jiahui Shi,Wenlu Sang,Chu Xu,Yiping Li,Shiyin Li

In situ cyanobacterial control has become an important complement to watershed management, but long-term efficacy and ecological impacts remain underexplored. We conducted a one-year enclosure experiment using artemisinin sustained-release algaecides (ASAs) at a dosage of 1:200 (v/v, ASAs to water column) to assess ecosystem effects and persistence, specifically on Microcystis blooms. ASAs temporarily altered carbonate chemistry and carbon pools. Combined with artemisinin's allelopathic oxidation (increases to 100 μg/L in 2–5 days), this triggered cyanobacteria death, reducing chl-a (reduced by 63.9–80.4 %) and algal density (reduced by 23.7–96.4 %) while causing brief Microcystin-LR (MC-LR) spikes (4.7–5.4 μg/L). ASAs also suppressed certain rare prokaryotes, enhanced deterministic community assembly, reduced eukaryotic α-diversity (notably Arthropoda ), marginalized cyanobacteria in microbial networks, and raised Bacillariophyta centrality. Over subsequent months, sustained bloom suppression was driven by microbial dynamics: ASAs altered carbon isotope signatures, weakening cyanobacterial fractionation; increased prokaryote and eukaryote abundances; promoted diversity recovery; and reduced cyanobacterial network centrality to prevent rebound. Network analyses revealed a keystone-taxa shift from Microcystis to zooplankton genera Eucyclops and Neodiaptomus , reflecting the long-term resistance of the microbial community to cyanobacteria. Ecological modeling showed that ASAs rewired water chemistry-microbe-cyanobacteria interactions, and random forests showed that under ASAs treatment, the temperature sensitivity of cyanobacteria decreased and microbial competition increased. These findings elucidate distinct short- and long-term mechanisms of ASAs-mediated bloom control and provide a theoretical foundation for sustainable field applications.

The inhibition mechanism and death mode of Microcystis aeruginosa induced by the continuous pressure of artemisinin sustained-release microspheres (ASMs)

Cunhao Du,Lixiao Ni,Jakpa Wizi,Wenlu Sang,Shiyi Rong,Xianglan Li,Chu Xu,Yan Li,Xuqing Chen,Shiyin Li

Artemisinin , a natural allelochemical substance extracted from Artemisia annua , has been shown to have strong inhibitory effects on harmful cyanobacteria . However, only a few studies have systematically studied the mechanism of artemisinin in inhibiting algae and the process of algae cell death. In this study, the inhibition mechanism and death mode of M. aeruginosa under artemisinin sustained-release microspheres (ASMs) were investigated using a series of morphological, physiological and biochemical characteristics. The findings revealed that M. aeruginosa was significantly inhibited immediately after exposure to the 0.54 g·L −1 ASMs, with an inhibition rate (IR) exceeding 95 %, and the synthesis of phycobiliprotein and esterase activity were disturbed. In addition, ASMs exposure significantly increased caspase-3(-like) activities of M. aeruginosa . Under the continuous stress of ASMs, M. aeruginosa cells had three cell death modes: the first was necrosis-like cell death, which was characterized by cell wall rupture and leakage of cell contents; the second was apoptosis-like cell death, which was characterized by the disintegration of the cell structure, the integrity of the cell membrane , and the appearance of apoptotic bodies; and the last was autophagy-like cell death, which was characterized by cell swelling and appearance of autophagy vacuoles. These results indicated that ASMs could inhibit photosynthesis of M. aeruginosa cells, interfere with its antioxidant mechanisms and activate caspase-3(-like) activity, resulting in programmed cell death (PCD).

Calcium Phosphate Silicate Microspheres with Soybean Lecithin as a Sustained-Release Bone Morphogenetic Protein-Delivery System for Bone Tissue Regeneration

Yaxin Xue,Yun Li,Dong Zhang,Wei Xu,Congqin Ning,Dong Han

Bone morphogenetic protein (BMP) is a growth factor that effectively promotes osteogenesis. Microsphere-based drug-delivery systems can facilitate an increase in the local concentration of BMP, thus promoting bone formation. In this study, calcium phosphate silicate (CPS) microspheres were used as drug-loading systems for BMP. Three groups─CPS, CPS + BMP, and CPS + BMP + soy lecithin (SL)─were set up, where SL was used to prolong the osteogenic effect of the microsphere system. Bone marrow mesenchymal stem cells and femoral defects in rats were used to compare the osteogenic ability of the three groups. The results indicated that CPS microspheres were good carriers of BMP, facilitating a smoother release into the cells and tissues. SL loading improved the loading rate of BMP, which promoted the osteogenic effect of the microspheres with BMP. We propose CPS microspheres as potential drug-delivery systems that can be effectively used in the treatment of bone defects.

Ultrasound‒treated soy protein fibrils: A potential vehicle for curcumin with improved water solubility, antioxidant activity and sustained‒release property

Fuyun Ji,Zijun Wang,Xiaohui Bai,Yanyan Zhao,Xiyang Zhong,Shuizhong Luo,Yizhong Shen,Shaotong Jiang,Zhi Zheng

Plant protein fibrils have recently attracted considerable attention as delivery vehicles for hydrophobic bioactive compounds . In this work, ultrasound treatment was applied to improve the structure of protein fibrils. The aim of this work was to investigate the impacts of ultrasound treatment on the structure of soy protein fibrils (SPF), and further explore the interaction between ultrasound‒treated SPF (USPF) and curcumin (Cur) to clarify the impacts of ultrasound on SPF as delivery vehicles. Thioflavin T fluorescence, SDS‒PAGE, Fourier transform infrared (FTIR) spectroscopy, and far‒UV circular dichroism spectroscopy revealed no significant changes in the primary and secondary structure of SPF after ultrasound treatment. Compared with SPF, USPF presented a more uniform particle size and higher surface hydrophobicity. Fluorescence quenching, ultraviolet–visible spectroscopy, isothermal titration calorimetry , and FTIR spectroscopy indicated USPF 10 (ultrasonication for 10 min) possessed a high affinity toward curcumin via hydrophobic interactions and hydrogen bonds. The curcumin solubility in USPF 10 -Cur (133.19 μg/mL) complex was much higher than SPI-Cur (78.26 μg/mL) and SPF-Cur (116.22 μg/mL) complexes. The USPF 10 ‒Cur complex exhibited good colloidal stability and thermal stability. The DPPH and ABTS scavenging capacities of the USPF 10 ‒Cur complex were approximately 1.4 and 2.7 folds higher than those of free curcumin, respectively. An In vitro digestion assay suggested that the USPF 10 ‒Cur complex showed a much more stable and sustainable release of curcumin and provided chances for curcumin release in the intestine. This study indicated that USPF might be promising delivery vehicles for hydrophobic bioactives as stable and functional ingredients in acidic food systems.

Promoting oral mucosal wound healing using a DCS-RuB2A2 hydrogel based on a photoreactive antibacterial and sustained release of BMSCs

Wenxin Qi,Naijun Dong,Lingling Wu,Xueqi Zhang,He Li,Hao Wu,Natalie Ward,Jian Yu,He Liu,Jiao Wang,Xiaoyong Deng,Robert Chunhua Zhao

The high occurrence rate and difficulties in symptom control are listed as the major problems of oral mucosal disease by medical professionals. Following the development of oral mucosal lesions, the oral microenvironment changes, immunity declines, and continuous bacterial stimulation causes wound infection. Traditional antibacterial drugs are ineffective for oral mucosal lesions. To overcome this problem, a light-responsive antibacterial hydrogel containing sustained-release BMSCs was inspired by the trauma environment in the oral cavity, which is different from that on the body surface since it mostly remains under dark conditions. In the absence of light, the hydrogel seals the wound to form a barrier, exerts a natural bacteriostatic effect, and prevents invasion by foreign bacteria. Simultaneously, mesenchymal stem cells are presented, and the released growth factors and other substances have excellent anti-inflammatory and angiogenic effects, which result in rapid repair of the damaged site. Under light conditions, after photo-induced shedding of the hydrogel, RuB 2 A exerts an antibacterial effect accompanied by degradation of the hydrogel. Results in a rat oral mucosal repair model demonstrate that DCS-RuB 2 A 2 -BMSCs could rapidly repair the oral mucosa within 4 days. Sequencing data provide ideas for further analysis of the intrinsic molecular mechanisms and signaling pathways. Taken together, our results suggest that this light-responsive antibacterial hydrogel loaded with BMSCs can be used for rapid wound repair and may advance the development of therapeutic strategies for the treatment of clinical oral mucosal defects.

Preparation and sustained-release properties of poly(lactic acid)/graphene oxide porous biomimetic composite scaffolds loaded with salvianolic acid B

Shuqiong Liu,Zhenyi Xu,Jiapeng Hu,Zhenzeng Wu,Yuying Zheng

Biomimetic scaffolds loaded with drugs can improve the osteogenesis and neovascularisation of scaffolds. A series of PLA/GO/Sal-B drug-loaded scaffolds was prepared by thermally induced phase separation. The addition of Sal-B increased the diameter of the fibres, but the scaffold showed a porous nanofibrous structure after drug release. X-ray diffraction results showed that the addition of Sal-B did not affect the formation of the nanofibre biomimetic structure of the scaffold. FTIR results indicated a certain interaction between Sal-B and PLA/GO. Water absorption and porosity test results revealed that the scaffolds had good hydrophilicity and appropriate porosity. The addition of Sal-B was also conducive to the formation of sediments possibly due to the good water solubility of Sal-B itself. The prepared scaffolds had good blood compatibility and cytocompatibility, and a small additional amount of Sal-B could significantly promote cell proliferation and alkaline phosphatase activity. Their sustained release performance indicated that the biomimetic scaffolds had controlled the release of Sal-B. The kinetic model showed that the PLA/GO/Sal-B drug-loaded biomimetic scaffolds followed the diffusion mechanism.
